Inflammation is a complex physiological process triggered in response to harmful stimuli1. It involves cells of the immune system capable of clearing sources of injury and damaged tissues. Excessive inflammation can occur as a result of infection and is a hallmark of several diseases2–4. The molecular bases underlying inflammatory responses are not fully understood. Here we show that the cell surface glycoprotein CD44, which marks the acquisition of distinct cell phenotypes in the context of development, immunity and cancer progression, mediates the uptake of metals including copper. We identify a pool of chemically reactive copper(ii) in mitochondria of inflammatory macrophages that catalyses NAD(H) redox cycling by activating hydrogen peroxide. Maintenance of NAD+ enables metabolic and epigenetic programming towards the inflammatory state. Targeting mitochondrial copper(ii) with supformin (LCC-12), a rationally designed dimer of metformin, induces a reduction of the NAD(H) pool, leading to metabolic and epigenetic states that oppose macrophage activation. LCC-12 interferes with cell plasticity in other settings and reduces inflammation in mouse models of bacterial and viral infections. Our work highlights the central role of copper as a regulator of cell plasticity and unveils a therapeutic strategy based on metabolic reprogramming and the control of epigenetic cell states.

Persister cancer cells represent rare populations of cells resistant to therapy. Cancer cells can exploit epithelial-mesenchymal plasticity to adopt a drug-tolerant state that does not depend on genetic alterations. Small molecules that can interfere with cell plasticity or kill cells in a cell state-dependent manner are highly sought after. Salinomycin has been shown to kill cancer cells in the mesenchymal state by sequestering iron in lysosomes, taking advantage of the iron addiction of this cell state. Here, we report the chemo- and stereoselective synthesis of a series of structurally complex small molecule chimeras of salinomycin derivatives and the iron-reactive dihydroartemisinin. We show that these chimeras accumulate in lysosomes and can react with iron to release bioactive species, thereby inducing ferroptosis in drug-tolerant pancreatic cancer cells and biopsy-derived organoids of pancreatic ductal adenocarcinoma. This work paves the way toward the development of new cancer medicines acting through active ferroptosis.

Interferons (IFN) are cytokines which, upon binding to cell surface receptors, trigger a series of downstream biochemical events including Janus Kinase (JAK) activation, phosphorylation of Signal Transducer and Activator of Transcription protein (STAT), translocation of pSTAT to the nucleus and transcriptional activation. Dysregulated IFN signalling has been linked to cancer progression and auto-immune diseases. Here, we report the serendipitous discovery of a small molecule that blocks IFNγ activation of JAK-STAT signalling. Further lead optimisation gave rise to a potent and more selective analogue that exerts its activity by a mechanism consistent with direct IFNγ targeting in vitro, which reduces bleeding in model of haemorrhagic colitis in vivo. This first-in-class small molecule also inhibits type I and III IFN-induced STAT phosphorylation in vitro. Our work provides the basis for the development of pan-IFN inhibitory drugs.

Salinomycin (1) exhibits a large spectrum of biological activities including the capacity to selectively eradicate cancer stem cells (CSC), making it and its derivatives promising candidates for the development of drug leads against CSC. It has been previously shown that salinomycin and its C20-propargylamine derivative (Ironomycin (2)) accumulate in lysosomes and sequester iron in this organelle. Herein, a library of salinomycin derivatives is reported, including products of C20-amination, C1-esterification, C9-oxidation, and C28-dehydration. The biological activity of these compounds is evaluated against transformed human mammary epithelial HMLER CD24low /CD44high cells, a well-established model of breast CSC, and HMLER CD24high /CD44low cells deprived of CSC properties. Unlike other structural alterations, derivative 4, which displays a cyclopropylamine at position C20, showed a strikingly low IC50 value of 23 nm against HMLER CD24low /CD44high cells. This study provides highly selective molecules to target the CSC niche, a potential interesting advance for drug development to prevent cancer resistance.

Cancer stem cells (CSC) constitute a cell subpopulation in solid tumors that is responsible for resistance to conventional chemotherapy, metastasis and cancer relapse. The natural product Salinomycin can selectively target this cell niche by directly interacting with lysosomal iron, taking advantage of upregulated iron homeostasis in CSC. Here, inhibitors of the divalent metal transporter 1 (DMT1) have been identified that selectively target CSC by blocking lysosomal iron translocation. This leads to lysosomal iron accumulation, production of reactive oxygen species and cell death with features of ferroptosis. DMT1 inhibitors selectively target CSC in primary cancer cells and circulating tumor cells, demonstrating the physiological relevance of this strategy. Taken together, this opens up opportunities to tackle unmet needs in anti-cancer therapy.

Chemical Biology is the science of designing chemical tools to dissect and manipulate biology at different scales. It provides the fertile ground from which to address important problems of our society, such as human health and environment.

New bridged silsesquioxanes derived from isosorbide were synthesized, according to the sol–gel process, by acidic or basic hydrolysis and condensation of a bis-trimethoxysilyl precursor. The latter was prepared by thiol-ene photoaddition of 3-(trimethoxysilyl)-1-propane thiol on diallyl isosorbide ether synthesized by a phase-transfer-catalyzed reaction of allyl bromide with isosorbide. The structure of those hybrid networks prepared according to a sol–gel process was investigated by 29Si solid-state NMR and X-ray diffraction experiments. Acid hydrolysis leads to the formation of randomly branched structures while basic catalysis promotes the creation of clusters. The anisotropic character of the hybrid networks was evidenced by cross polarized optical light observations (birefringence), demonstrating an organization at the micrometric-scale. These hybrid networks derived from isosorbide showed a thermal stability up to 360 °C.

Macroporous cross-linked organic polymers, based on styrene and divinylbenzene (PS-DVB), are prepared as monolithic stationary phases for capillary electrochromatographic applications. The synthetic strategy, which relies on the semi-interpenetrating polymer network (semi-IPN) approach, is performed through UV-initiated free radical copolymerization of the comonomers in the presence of poly(ϵ-caprolactone) (PCL) within the confines of fused silica capillaries. The characterization of PS-DVB monoliths is carried out at the different stages of the synthesis using a combination of experimental techniques, thus providing information on the chemical and porous structures. All experimental results evidence the sole role of the PCL oligomer as a porogen.

Relationships between microstructure and mechanical properties in SPS-Zn/PEG blends are investigated. Ion/dipole interactions induce the miscibility of both blend components for PEG content up to 10 wt.-% as evidenced by DSC and TEM observations. Elemental mapping by EFTEM was used to investigate the ionic cluster microstructure and the phase structure in the blends. PEG chains do not hinder the formation of ionic multiplets but modify the microstructure of the clusters. Moreover, a PEG-rich layer is observed in the vicinity of the aggregates surface. As a consequence, no cluster glass transition is observed in DMA experiments for these blend systems and besides, the absence of ionic plateau is assigned to the decrease in the ionic clusters' stability.

Addition of commercially available 2-isocyanatoethyl methacrylate to a water solution of potassium bisulfite unexpectedly yields an anionically charged hydrogel. Structural analysis shows the presence of sulfonato-blocked isocyanates (with no detectable residual isocyanates), a full polymerization of the vinyl groups, and the appearance of urea functional groups. A mechanism is proposed to explain the gel formation, based on a combination of bisulfite-initiated free-radical polymerization, crosslinking by urea bridge formation, and addition of bisulfite ions to isocyanate groups. Some basic physical properties (TGA, swelling) of the gel are presented.

The structure and rheological behavior of atactic poly(vinyl alcohol) (a-PVA) hydrogels prepared by freeze/thaw cycles were investigated as a function of polymer concentration and number of freeze/thaw cycles. The presence of phases with different mobilities was observed using 13 C CP/MAS and DP/MAS NMR experiments. The degree of crystallinity of the a-PVA-rich phase was determined by 1 H NMR free decay experiments. Measurements of the shear storage and loss modulus were performed at a fixed frequency of 1 Hz and a strain value of 0.1%, i.e. under conditions where the deformation imposed on the gel structure is entirely reversible. Results thus obtained showed that an increase in the number of freeze/thaw cycles induces an increase in the degree of crystallinity in the polymer-rich phase together with an increase in the storage modulus. The a-PVA hydrogels became more fragile as the number of freeze/thaw cycles was increased. Moreover, both the percentage of protons in a rigid environment measured by 1 H NMR and the storage modulus values tended to a limiting value after six freeze/thaw cycles. These results show that the first five or six freeze/thaw cycles play a very important role in determining the hydrogel structure and rheological properties. A more detailed comparison of NMR and rheological data led to the conclusion that the storage modulus is mainly controlled by the a-PVA crystallinity while the hydrogen bond interactions have a much smaller contribution.

The new class of antibiotics called ketolides is endowed with remarkable antibacterial activity against macrolide-resistant strains. Further modifications of the 3 keto-macrolactone backbone led to 11,12-hydrazonocarbamate ketolides with an imidazolyl–pyridine chain: the file-leader of ketolide class, HMR 3647 (telithromycin), and its N-bis-demethyl-derivative, RU 72366. The potency of HMR 3647 is higher than that of RU 72366. Stereospecific 1H and 13C resonance assignments of HMR 3647 and RU 72366 have been determined and have allowed a detailed quantitative conformational analysis of the uncomplexed form of the molecules. The comparative conformation of HMR 3647 in solution and its N-bis-demethyl-derivative in D2O has been carried out using different heteronuclear correlation experiments in conjunction with nuclear Overhauser effect experiments and in particular long-range 3JCH coupling constants and using molecular dynamics (MD) methods. The study of ketolide–ribosome interaction has been investigated using two-dimensional transferred nuclear Overhauser effect spectroscopy (TRNOESY). The database of ribosome-bound ketolide structures has been used to compare the structure(s) of ketolide in ribosome–ketolide complexes with the conformational preferences of free ketolides and to highlight the significant differences between HMR 3647 and RU 72366. A comparison of the conformations bound to ribosome was made with those of other previously studied ketolide (RU 004) and macrolides and would explain the remarkable potencies of HMR 3647 in inhibiting protein synthesis.

Variants of the HSQC and HMBC experiments are described. They allow the restriction of the heteronuclear chemical shift domain without causing spectral folding. Selectivity is introduced in the HSQC experiment by means of excitation sculpting. The selective element of the pulse sequence is a double pulsed field gradient spin echo. It may be used either split by the t 1 evolution period, or not. The selectivity profile depends on the scheme used as well as on the number of protons attached to the heteronucleus. The selective HMBC experiment requires only a single echo sequence as no strict control of the signal phase is required. A complex glycoconjugate is used as a test compound for the new pulse sequences.
