Abstract Changes in upstream land-use have significantly transformed downstream coastal ecosystems around the globe. Restoration of coastal ecosystems often focuses on local-scale processes, thereby overlooking landscape-scale interactions that can ultimately determine restoration outcomes. Here we use an idealized bio-morphodynamic model, based on estuaries in New Zealand, to investigate the effects of both increased sediment inputs caused by upstream deforestation following European settlement and mangrove removal on estuarine morphology. Our results show that coastal mangrove removal initiatives, guided by knowledge on local-scale bio-morphodynamic feedbacks, cannot mitigate estuarine mud-infilling and restore antecedent sandy ecosystems. Unexpectedly, removal of mangroves enhances estuary-scale sediment trapping due to altered sedimentation patterns. Only reductions in upstream sediment supply can limit estuarine muddification. Our study demonstrates that bio-morphodynamic feedbacks can have contrasting effects at local and estuary scales. Consequently, human interventions like vegetation removal can lead to counterintuitive responses in estuarine landscape behavior that impede restoration efforts, highlighting that more holistic management approaches are needed.

To evaluate potential pathomechanisms in the induction of infective endocarditis (IE), 34 Staphylococcus aureus (S. aureus) isolates, collected from patients with S. aureus endocarditis and from healthy individuals were investigated both in vitro and in vivo. S. aureus isolates were tested in vitro for their cytotoxicity, invasion and the association with platelets. Virulence factor expression profiles and cellular response were additionally investigated and tested for correlation with the ability of S. aureus to induce vegetations on the aortic valves in vivo. In an animal model of IE valvular conspicuity was assessed by in vivo magnetic resonance imaging at 9.4 T, histology and enrichment gene expression analysis. All S. aureus isolates tested in vivo caused a reliable infection and inflammation of the aortic valves, but could not be differentiated and categorized according to the measured in vitro virulence profiles and cytotoxicity. Results from in vitro assays did not correlate with the severity of IE. However, the isolates differed substantially in the activation and inhibition of pathways connected to the extracellular matrix and inflammatory response. Thus, comprehensive approaches of host-pathogen interactions and corresponding immune pathways are needed for the evaluation of the pathogenic capacity of bacteria. An improved understanding of the interaction between virulence factors and immune response in S. aureus infective endocarditis would offer novel possibilities for the development of therapeutic strategies and specific diagnostic imaging markers.

Measuring photoionization time delays is an interesting and challenging topic. Here the authors demonstrate a method to measure the photoionization time delays using inner-shell ionization of CO molecule.

Within the frame of the EMPIrE test, four monolithic mini-plates were irradiated in the ATR reactor. In two of them, the monolithic U(Mo) foil had been PVD-coated with Zr before the plate manufacturing. Extensive microstructural characterizations were performed on a fresh archive mini-plate, using Optical Microscopy (OM), Scanning Electron Microscopy (SEM) combined with Energy Dispersive Spectroscopy (EDS), Electron Backscattered Diffraction (EBSD) and Focused Ion Beam (FIB)/Transmission Electron Microscopy (TEM) with nano EDS. A particular attention was paid to the examination of the U(Mo) foil, the PVD coating, the cladding/Zr and Zr/U(Mo) interfaces.The Zr coating has a thickness around 15 μm. It has a columnar microstructure and appears dense. The cohesion of the cladding/Zr and Zr/U(Mo) interfaces seems to be satisfactory. An almost continuous layer with a thickness of the order of 100–300 nm is present at the cladding/Zr interface and corresponds to an oxidized part of the Zr coating. At the Zr/U(Mo) interface, a thin discontinuous layer is observed. It could correspond to locally oxidized U(Mo).This work provides a basis for interpreting the results of characterizations on EMPIrE irradiated plates.

Abstract The production of peptides as active pharmaceutical ingredients (APIs) by recombinant technologies is of emerging interest. A reliable production platform, however, is still missing due the inherent characteristics of peptides such as proteolytic sensitivity, aggregation and cytotoxicity. We have developed a new technology named Numaswitch solving present limitations. Numaswitch was successfully employed for the production of diverse peptides and small proteins varying in length, physicochemical and functional characteristics, including Teriparatide, Linaclotide, human β-amyloid and Serum amyloid A3. Additionally, the potential of Numaswitch for a cost-efficient commercial production is demonstrated yielding > 2 g Teriparatide per liter fermentation broth in a quality meeting API standard.

Background: Immune cells show distinct motion patterns that change upon inflammatory stimuli. Monocytes patrol the vasculature to screen for pathogens, thereby exerting an early task of innate immunity. Here, we aimed to non-invasively analyse single patrolling monocyte behaviour upon inflammatory stimuli. Methods: We used time-lapse Magnetic Resonance Imaging (MRI) of the murine brain to dynamically track single patrolling monocytes within the circulation distant to the actual site of inflammation in different inflammatory conditions, ranging from a subcutaneous pellet model to severe peritonitis and bacteraemia. Abstract Interactions between water flow and patchy vegetation are governing the functioning of many ecosystems. Yet, numerical models that simulate those interactions explicitly at the submeter patch scale to predict geomorphological and ecological consequences at the landscape scale (order of km2) are still very computationally demanding. Here, we present a novel and efficient convolution technique to incorporate biogeomorphic feedbacks in numerical models across multiple spatial scales (from less than 1 m2 to several km2). This new methodology allows for spatially refining coarse‐resolution hydrodynamic simulations of flow velocities (order of m) around fine‐resolution patchy vegetation patterns (order of 10 cm). Although flow perturbations around each vegetation grid cell are not simulated with the same level of accuracy as with more expensive finer‐resolution models, we show that our approach enables spatial refinement of coarse‐resolution hydrodynamic models by resolving efficiently subgrid‐scale flow velocity patterns within and around vegetation patches (mean error, spatial variability, and spatial correlation improved by, respectively, 13%, 66%, and 49% on average in our test cases). We also provide evidence that our approach can substantially improve the representation of important biogeomorphic processes, such as subgrid‐scale effects on net sedimentation rate and habitable surface area for vegetation (respectively 66% and 39% better on average). Finally, we estimate that replacing a fine‐resolution model by a coarser‐resolution model associated with the convolution method could reduce the computational time of real‐life fluctuating flow simulations by several orders of magnitude. This marks an important step forward toward more computationally efficient multiscale biogeomorphic modeling.

Animal models of Staphylococcus aureus infective endocarditis (IE), especially in rodents, are commonly used to investigate the underlying pathogenesis, disease progression, potential diagnostic approaches, and therapeutic treatment. All these models are based on surgical interventions, and imply valve trauma by placing a polyurethane catheter at the aortic root. While the influence of endothelial damage and inflammation on the induction of IE has been studied intensively, the role of the catheter, as permanent source of bacteremia, and the interplay with bacterial virulence factors during the formation of IE is poorly understood. In our study, we aimed at identifying which set of preconditions is required for induction and formation of IE: (1) tissue injury, (2) permanent presence of bacteria, and (3) presence of the full bacterial repertoire of adhesion proteins. We investigated the manifestation of the disease in different modifications of the animal model, considering different degrees of endothelial damage and the presence or absence of the catheter. In four infection models the induction of IE was assessed by using two bacterial strains with different expression patterns of virulence factors – S. aureus 6850 and Newman. In vivo magnetic resonance imaging showed conspicuous morphological structures on the aortic valves, when an endothelial damage and a continuous bacterial source were present simultaneously. Cellular and inflammatory pathophysiology were characterized additionally by histology, real-time quantitative polymerase chain reaction analysis, and bacterial counts, revealing strain-specific pathogenesis and manifestation of IE, crucially influenced by bacterial adherence and toxicity. The severity of IE was dependent on the degree of endothelial irritation. However, even severe endothelial damage in the absence of a permanent bacterial source resulted in reduced valve infection. The spread of bacteria to other organs was also dependent on the pathogenic profile of the infectious agent.

Mangrove forests are valuable ecosystems, but their extent and diversity are increasingly threatened by sea-level rise and anthropogenic pressures. Here we develop a bio-morphodynamic model that captures the interaction between multiple mangrove species and hydro-sedimentary processes across a dynamic coastal profile. Numerical experiments are conducted to elucidate the response of mangrove assemblages under a range of sea-level rise and sediment supply conditions, both in the absence and presence of anthropogenic barriers impeding inland migration. We find that mangrove coverage can increase despite sea-level rise if sediment supply is sufficient and landward accommodation space is available. Tidal barriers are mainly detrimental to mangrove coverage and result in species loss. Importantly, we show that bio-morphodynamic feedbacks can cause spatio-temporal variations in sediment delivery across the forest, leading to upper-forest sediment starvation and reduced deposition despite extended inundation. As such, bio-morphodynamic feedbacks can decouple accretion rates from inundation time, altering mangrove habitat conditions and causing mangrove diversity loss even when total forest coverage remains constant or is increasing. A further examination of bio-morphodynamic feedback strength reveals that vegetation-induced flow resistance linked to mangrove root density is a major factor steering the inundation-accretion decoupling and as such species distribution. Our findings have important implications for ecosystem vulnerability assessments, which should account for the interactions between bio-morphodynamics and mangrove diversity when evaluating the impacts of sea-level rise on species assemblages.

In traditional dialectology, prominent geographical conditions are particularly made responsible for linguistic divergence between neighboring dialect areas (e. g. rivers, mountain ranges, etc.). Also modern national borders, territories of the Middle Ages or even older borders between tribal areas are made responsible as factors which consider linguistic areas as “mirror pictures of history” (Bach 1950: 31, translation: CS). Geographical as well as political borders seem to function as points of orientation that speakers use for constructing their cognitive surroundings and which consequently can result in linguistic divergence. In our contribution, we want to focus on the question of how the topographic nature of land-scapes influences the mental construction of linguistic areas by lay speakers. For pursuing this question, we will discuss results from German dialect areas that cover a broad range of different topographic landscape forms. The analyzed areas are located in the utter southwest of Germany (characterized by flat lands along the Rhine valley and low mountain ranges of the Black Forest) and in South Tyrol in the very north of Italy (characterized by mountains and deep valley cuttings). In our contribution, we will argue that speakers in their subjective dialect perception orient themselves toward national borders and topographically prominent points. With regard to topography primarily valleys are used as eponyms for subjective dialect areas.

The present paper focuses on conservative and innovative (transitional) dialect areas and the questions of 1) how such areas can be methodologically visualized and 2) how the outcomes can be interpreted. In the first part of this paper a geostatistical method of representing phonological features in space will be introduced: interpolation. This method is not entirely new to dialectology; it has been quite neglected, though, in comparison to other methods of mapping, such as the isogloss or dot symbol method that was mainly used in traditional dialect atlases. The interpolation method will be applied to a large corpus of spontaneous speech data from rural dialects spoken in southwest Germany. Methodological steps in data processing will be described, resulting in a data set that can be used as input for statistical analysis and the visual depiction of variation in space as interpolated grid plots. In the second part results will be discussed. The major outcome consists of an aggregate interpolation plot that includes variables from fifteen different etymological sound classes. These sound classes can be used for demonstrating the distribution of receding phonological variables in space. The interpolation shows two conservative areas where receding forms are still widespread. They lie within the centers of the two major dialect groups of southwest Germany: Alemannic and Swabian. The conservative areas are separated by a broad transitional zone characterized by intense variation between receding and innovative variants. It will be argued that this transitional zone is not due to the horizontal spread of the dialects into each other’s areas alone. Rather, variation is triggered by vertical standard influence that supports any dialect form to spread out horizontally as long as it is phonologically identical or similar to the standard form.

2D diagonal redox SDS-PAGE of proteins is used to detect intramolecular or intermolecular disulfide bridges using Chlamydomonas in this example (Stroeher and Dietz, 2008; Schwarz et al., 2012). Both dimensions consist of a conventional SDS-PAGE, except that the sample buffer for the first dimension lacks a reducing agent. Intermolecular disulfide bridges increase the apparent molecular weight of a protein in the first dimension, whereas intramolecular bridges decrease the apparent weight of the protein.

35S pulse labelling of proteins is used to attach a radioactive label to newly synthesized proteins, as sulfur is an element that is mainly present in proteins (Fleischmann and Rochaix 1999). Depending on your organism’s uptake mechanisms you need cysteine, methionine or sulfuric acid as a source of radioactive sulfur. This example uses Chlamydomonas cells and H235SO4 (Schwarz et al., 2012).

Metabolic control of gene expression coordinates the levels of specific gene products to meet cellular demand for their activities. This control can be exerted by metabolites acting as regulatory signals and/or a class of metabolic enzymes with dual functions as regulators of gene expression. However, little is known about how metabolic signals affect the balance between enzymatic and regulatory roles of these dual functional proteins. We previously described the RNA binding activity of a 63 kDa chloroplast protein from Chlamydomonas reinhardtii, which has been implicated in expression of the psbA mRNA, encoding the D1 protein of photosystem II. Here, we identify this factor as dihydrolipoamide acetyltransferase (DLA2), a subunit of the chloroplast pyruvate dehydrogenase complex (cpPDC), which is known to provide acetyl-CoA for fatty acid synthesis. Analyses of RNAi lines revealed that DLA2 is involved in the synthesis of both D1 and acetyl-CoA. Gel filtration analyses demonstrated an RNP complex containing DLA2 and the chloroplast psbA mRNA specifically in cells metabolizing acetate. An intrinsic RNA binding activity of DLA2 was confirmed by in vitro RNA binding assays. Results of fluorescence microscopy and subcellular fractionation experiments support a role of DLA2 in acetate-dependent localization of the psbA mRNA to a translation zone within the chloroplast. Reciprocally, the activity of the cpPDC was specifically affected by binding of psbA mRNA. Beyond that, in silico analysis and in vitro RNA binding studies using recombinant proteins support the possibility that RNA binding is an ancient feature of dihydrolipoamide acetyltransferases. Our results suggest a regulatory function of DLA2 in response to growth on reduced carbon energy sources. This raises the intriguing possibility that this regulation functions to coordinate the synthesis of lipids and proteins for the biogenesis of photosynthetic membranes.
