- Abstract
Maydis leaf blight (MLB) caused by Helminthosporium maydisY. Nisik. & C. Miyake is prevalent in almost all maize growing areas and is a major limiting factor in increasing yield. Knowing about the metabolic pathways that results in accumulation of enzymatic and non-enzymatic antioxidative biochemical compounds which constitutes a second line of defense, an in vitro experiment was conducted. Reactive oxygen species produced during stresses is one of the most instant defense reactions to arrest plant pathogen at the site of infection. Oxidative burst results in the production of ROS, mainly H2O2and superoxide at the invading site. Production of secondary metabolites and induction of ISR and SAR mediate biochemical defenses in plant. In vitro study to observe the effect of different elicitors applied exogenously against H. maydisby investigating the biochemical parameters viz., Total phenols, Proline, Hydrogen peroxide (H2O2) and, enzymatic anti-oxidative biochemicals viz., Superoxide dismutase (SOD), Polyphenol oxidase (PPO), Peroxidase (POD) and, Phenylalanine ammoniase (PAL) activity is conducted. The activity of different biochemical parameters was observed on the 0th day (before inoculation), 3rd day, 6th day and 9th day of inoculation with the help of spectrophotometer. Accumulation of H2O2concentration was recorded maximum (815.61 µmol g−1FW) at the 9th day after inoculation in 2,6-dichloroisonicotinic acid (INA)-treated plants. INA-treated plants showed highest proline accumulation (43.39 µmol g−1FW) and phenolics content (560.92 µg g−1FW) at the 6th and 3rd day after inoculation of the pathogen, respectively. In biochemical studies for elucidation of defence induction, biochemical markers reached maximum on the 3rd day after pathogen inoculation, and INA treatments were found most effective.

Passive stiffness of the heart is determined largely by extracellular matrix and titin, which functions as a molecular spring within sarcomeres. Titin stiffening is associated with the development of diastolic dysfunction (DD), while augmented titin compliance appears to impair systolic performance in dilated cardiomyopathy. We found that myofibril stiffness was elevated in mice lacking histone deacetylase 6 (HDAC6). Cultured adult murine ventricular myocytes treated with a selective HDAC6 inhibitor also exhibited increased myofibril stiffness. Conversely, HDAC6 overexpression in cardiomyocytes led to decreased myofibril stiffness, as did ex vivo treatment of mouse, rat, and human myofibrils with recombinant HDAC6. Modulation of myofibril stiffness by HDAC6 was dependent on 282 amino acids encompassing a portion of the PEVK element of titin. HDAC6 colocalized with Z-disks, and proteomics analysis suggested that HDAC6 functions as a sarcomeric protein deacetylase. Finally, increased myofibril stiffness in HDAC6-deficient mice was associated with exacerbated DD in response to hypertension or aging. These findings define a role for a deacetylase in the control of myofibril function and myocardial passive stiffness, suggest that reversible acetylation alters titin compliance, and reveal the potential of targeting HDAC6 to manipulate the elastic properties of the heart to treat cardiac diseases.

Introduction: Chronic Obstructive Pulmonary Disease (COPD) is a heterogeneous lung condition characterized by chronic respiratory symptoms (dyspnoea, cough, sputum production, exacerbations) due to abnormalities of the airways (bronchitis, bronchiolitis) and/or alveoli (emphysema) that cause persistent, often progressive, airflow obstruction. Aim: To study RCI as a marker for severity of COPD. Material and Methods: An observational study was conducted in Department of Pulmonary Medicine, K. N. Chest hospital, after approval from institution ethical committee, Dr. S. N. Medical College, Jodhpur. Result: In our study, 58 (58.0%) patients were Cigarettes, 36 (36.0%) patients were Ex-smoker and 6 (6.0%) patients were Non-Smoker. Conclusion: We conclude that, majority numbers of patients were Cigarettes in Stage 1 compared to Stage 2, Stage 3 and Stage 4 though it was not statistically significant. [ABSTRACT FROM AUTHOR]

Background: Asthma is a heterogeneous disease, usually represented by chronic airway inflammation leading to variable respiratory symptoms and variable expiratory airflow limitation. Vitamin D has been shown to have several effects on the innate and adaptive immune systems that might modulate the severity of asthma exacerbations. Vitamin D deficiency and insufficiency is common in asthma patient, which causes frequent asthma attacks, respiratory tract infections, reduced lung function, poor asthma control, poor steroid responsiveness and severe asthma exacerbations. Objective: To determine serum vitamin D levels among asthmatic patients and to correlate them with the severity of bronchial asthma. Methods: A total of 50 patients with stable bronchial asthma between 18 to 60 years ages groups were included in the study. This Observational Study was conducted from April 2022 to December 2022 in Department of Respiratory medicine, Kamla Nehru Chest Hospital, Jodhpur. After taking proper clinical history and system examination, spirometry was done in each patient to obtain forced expiratory volume (FEV1), FVC and FEV1/FVC ratio for determination of the severity of asthma. Asthma control was assessed by using of asthma control questionnaire. Serum vitamin D level and absolute eosinophils count were measured in all participants. Results: In our study out of 50 cases, male patients (n=24) and female (n=26) which Association of Allergic history in Group with Severity of asthma was statistically significant (p=0.0240) and mean Vitamin-D Levels (ng/ml) was significantly higher in Mild [29.2182± 6.0880] compared to Moderate [23.3583± 5.2467] and Severe [14.9067± 5.1568] (p<0.0001). The mean Vitamin D Levels was significantly higher in "well controlled" [27.9071 ± 6.3191] compared to "partly controlled" [22.8706 ± 6.8941] and "uncontrolled" [17.2158 ± 7.0243] (p=0.0002). Conclusion: Vitamin D deficiency and insufficiency are highly prevalent in asthma patients in which prolonged duration of illness, frequently asthma exacerbation and poor asthma control. Hence, vitamin D is a useful for prevention and treatment of bronchial asthma. [ABSTRACT FROM AUTHOR]

Introduction: Tuberculosis (TB) is a communicable disease that is a major cause of ill health and one of the leading causes of death worldwide. Monocytes are professional phagocytes that are highly skilled in defence against many pathogens, including MTB.4 They circulate in the blood and can differentiate into monocyte-derived macrophages and monocyte-derived dendritic cells that govern innate and adaptive immune responses and Lymphocytes (L) provide resistance to the spread of infection causing mycobacterial clearance. Aim & Objectives: To assess the monocyte lymphocyte ratio (M/L ratio) as a possible prognostic biomarker to see the outcome of Anti Tuberculous Therapy in terms of sputum status in active TB cases. Material & Methods: The present study was a Prospective observational study. This study was conducted from Feb 2021 and July 2022 at Department of respiratory medicine, Kamla Nehru Chest Hospital, Dr S N Medical College Jodhpur, a tertiary care Centre for respiratory diseases, Rajasthan, India. 130 patients were included in this study. Result: In our study, 7 (5.4%) patients had COPD, 23 (17.7%) patients had HTN and 18 (13.8%) patients had T2DM. The value of z is 9.8029. The value of p is < .00001. The result is significant at p <.05. In our study, 130 (100.0%) patients had Sputum Status at 0 month. In our study, 117 (90.0%) patients had Sputum conversion at 2nd month. The value of z is 12.8996. The value of p is < .00001. The result is significant at p <.05. The mean Monocytes at 0 Month of patients was [5.0769± 1.2519], mean Lymphocytes at 0 Month of patients was [17.7392± 4.3696], mean M/L ratio at 0 Month of patients was [.3057± .1206], mean Monocytes at 2nd Month of patients was [4.4862± 1.1302] and mean M/L ratio at 2nd Month of patients was [.2389± .0821]. Conclusion: Tuberculosis ss associated with Increased M/L Ratio, which Declines and Returns to Normal with Anti-Tuberculous Therapy. [ABSTRACT FROM AUTHOR]

Introduction: A pleural effusion is accumulation of excessive fluid in the pleural space due to an imbalance between the production and absorption of pleural fluid by pleurae (visceral as well as parietal). Pleural effusion is a common clinical manifestation and about 3000 per Million people in the world suffers from pleural disease. Aims: To assess usefulness of ratio of serum LDH to Pleural Fluid ADA in differentiating Malignant Pleural Effusion and Tubercular Pleural Effusion. Materials and Methods: The study was a Descriptive Observational Crosssectional study was conducted in Department of Respiratory Medicine, Kamala Nehru Chest Hospital, Dr S N Medical College Jodhpur (Rajasthan), and a tertiary care centre for Respiratory Disease in Rajasthan, India after getting clearance from the Ethical committee. Result: found that most patients with tubercular pleural effusion (n=20) had Pleural Fluid ADA levels >35 and majority of the patients had Biopsy proven Malignant pleural effusion [22 (44.0%)] and it was statistically significant (p< .00001), (z=4.1036). Conclusion: We found that, most number of patients had Tubercular Pleural Effusion. - Abstract
Ear biometrics has been found to be a good and reliable technique for human recognition. Initially ear biometrics could not gain popularity because there were doubts about its uniqueness. But, it started to gain momentum after a theory which came into existence and which said that it was very unlikely for any two years to be completely identical in all respects. The implemented methodology consists of steps such as pre-processing, feature extraction and matching based on the selected features. Our technique determines the extent to which these features support matching. The proposed work has been carried out on on a dataset containing 60 images for analyzing their features and matching of the source image with the dataset images. The results have been obtained on the basis of images correctly classified. The system accuracy telling us the extent to which matching could be performed on the basis of selected features.

- Abstract
Acute myeloid leukaemia (AML) is a heterogeneous disease characterized by transcriptional dysregulation that results in a block in differentiation and increased malignant self-renewal. Various epigenetic therapies aimed at reversing these hallmarks of AML have progressed into clinical trials, but most show only modest efficacy owing to an inability to effectively eradicate leukaemia stem cells (LSCs)1. Here, to specifically identify novel dependencies in LSCs, we screened a bespoke library of small hairpin RNAs that target chromatin regulators in a unique ex vivo mouse model of LSCs. We identify the MYST acetyltransferase HBO1 (also known as KAT7 or MYST2) and several known members of the HBO1 protein complex as critical regulators of LSC maintenance. Using CRISPR domain screening and quantitative mass spectrometry, we identified the histone acetyltransferase domain of HBO1 as being essential in the acetylation of histone H3 at K14. H3 acetylated at K14 (H3K14ac) facilitates the processivity of RNA polymerase II to maintain the high expression of key genes (including Hoxa9 and Hoxa10) that help to sustain the functional properties of LSCs. To leverage this dependency therapeutically, we developed a highly potent small-molecule inhibitor of HBO1 and demonstrate its mode of activity as a competitive analogue of acetyl-CoA. Inhibition of HBO1 phenocopied our genetic data and showed efficacy in a broad range of human cell lines and primary AML cells from patients. These biological, structural and chemical insights into a therapeutic target in AML will enable the clinical translation of these findings.

Interpenetrating polymer network hydrogel beads of pectin and sodium carboxymethyl xanthan were prepared by ionotropic gelation with Al(+3) ions and covalent cross-linking with glutaraldehyde for sustained delivery of diltiazem hydrochloride. Fourier transform infrared spectroscopy, X-ray diffraction, differential scanning colorimetry and scanning electron microscopy were used to characterise the hydrogel beads. The swelling of the hydrogel and the release of drug were relatively low in pH 1.2 buffer solutions. However, higher swelling and drug release were observed in pH 6.8 buffer solutions. The carboxyl functional groups of hydrogels undergo ionisation and the osmotic pressure inside the beads increases resulting in higher swelling and drug release in higher pH. The release of drug depends on concentration of polymer, amount and exposure time of cross-linker and drug content in the hydrogel matrices. The present study indicated that the hydrogel beads minimised the drug release in pH 1.2 buffer solutions and to prolong the drug release in pH 6.8 buffer solutions.

- Abstract
Long-term effect of nitrogen substitution (25 to 50%) through different organics, viz., FYM (farm yard ma-nure), GM (green manure) and WS (wheat straw) on crop yield, nutrition and physico-chemical properties of soil was studied under rice-wheat system. The data of long term experiment revealed that maximum grain yield of 46.83 qha-1 in wheat was obtained when 100% RDF applied through mineral fertilizers after 50% N of RDF being substituted with FYM in rice. Grain yield of wheat declined under control and sub-optimal fertilizer inputs (50% or 75% recommended fertilizer NPK), whereas positive yield increment was observed under treatments receiving organic supplements. The analysis of soil samples showed that soil pH reduced from initial value of 7.40 to 7.22, organic carbon build-up from 0.46% to 0.76%, available N from 194.00 to 225.95 kgha-1 available P2O5 from 23.60 to 49.54 kgha-1 and available K2O from 155.00 to 189.95 kgha-1 However, available Sulphur and DTPA-Zn increased from 7.74 to 14.41 kg ha-1 and 0.75 to 1.37 mg kg-1 respectively due to long-term (29 years) integrated nutrient management practices under rice-wheat system in alluvial soil. In conclusion, substitution of 50% and 25% N of RDF to rice through organics either FYM / Green manure / Wheat straw significantly increased the crop yield and nutrient uptake of wheat as well as build up the organic carbon, available N, P2O5, K2O, Sulphur, DTPA extractable Zn, Cu, Fe and Mn of post harvest soil after 29 years of the experiment.

- Abstract
The Hippo pathway has been identified as a key barrier for tumorigenesis, acting through downregulation of YAP/TAZ activity. Elevated YAP/TAZ activity has been documented in many human cancers. Ubiquitylation has been shown to play a key role in regulating YAP/TAZ activity through downregulation of a number of Hippo pathway components. Several ubiquitin ligase complexes have been implicated in this process, however, little is known about the deubiquitylating enzymes that counteract these activities to regulate YAP/TAZ. Here we identify the deubiquitylating enzyme USP9x as a regulator of YAP/TAZ activity. We demonstrate that USPx regulates ubiquitin-mediated turnover of the YAP inhibitor, Angiomotin. USP9x acts to deubiquitylate Angiomotin at lysine 496, resulting in stabilization of Angiomotin and lower YAP/TAZ activity. USP9x mRNA levels were reduced in several cancers. Clinically, USP9x mRNA levels were reduced in several cancers with low USPx expression correlating with poor prognosis in renal clear cell carcinoma. Our data indicate that USP9x may be a useful biomarker for renal clear cell carcinoma.

- Abstract
Colour in soils as well as other object is the visual perceptual property which is perceived by human eye. They are governed by spectrum of light corresponding to wavelength or reflected energy of the material. Developed model for soil acidity is based on visual interpretation, principal component and spectral enhancement techniques by using of the satellite image (IRS LISS III, 2014). In this context, red soil patch is much sensitive in red spectral band comparison to green and blue spectral bands and perceived as red tone by human eyes but same soil patch appears green in false colour composite (FCC) image of NIR (0.70-0.80?m), Red (0.60-0.70 ?m) and Green (0.50-0.60?m) bands. The maximum coverage of red soil patches having low pH < 6.5 (1:2.5) was recognized in 44.07 per cent of the total geographical area (3019.56 sq.km) under Banka district. Maximum red soil patches having their acidity were recognised in Katoria (18.56%), Chanan (15.15%), Bounsi (10.44%) and Banka (9.92%) blocks. Overall results indicated that variation of tone in different bands helps for the separation of red soil patches.

Activating mutations in the receptor tyrosine kinase FLT3 are frequently found in acute myelogenous leukemia patients and confer poor clinical prognosis. It is unclear how leukemic blasts escape cytokine control that regulates normal hematopoiesis. We have recently demonstrated that FLT3-internal tandem duplication (ITD), when localized to the biosynthetic compartment, aberrantly activates STAT5. Here, we show that one of the target genes induced by STAT5 is suppressor of cytokine signaling (SOCS)1 - a surprising finding for a known tumor suppressor. Although SOCS1 expression in murine bone marrow severely impaired cytokine-induced colony growth, it failed to inhibit FLT3-ITD-supported colony growth, indicating resistance of FLT3-ITD to SOCS1. In addition, SOCS1 coexpression did not affect FLT3-ITD-mediated signaling or proliferation. Importantly, SOCS1 coexpression inhibited interferon-αand interferon-γ signaling and protected FLT3-ITD hematopoietic cells from interferon-mediated growth inhibitory effects. In a murine bone marrow transplantation model, the coexpression of SOCS1 and FLT3-ITD significantly shortened the latency of a myeloproliferative disease compared with FLT3-ITD alone (P <.01). Mechanistically, SOCS proteins shield FLT3-ITD from external cytokine control, thereby promoting leukemogenesis. The data demonstrate that SOCS1 acts as a conditional oncogene, providing novel molecular insights into cytokine resistance in oncogenic transformation. Restoring cytokine control may provide a new way of therapeutic intervention.
