Recently, a Pediococcus pentosaceus SL4 (PP) strain that secretes the probiotic-derived anti-cancer protein P8 (PP-P8) has been developed as an anti-colorectal cancer (CRC) biologic by Cell Biotech. We initially identified a Lactobacillus rhamnosus (LR)-derived anti-cancer protein, P8, that suppresses CRC growth. We also showed that P8 penetrates specifically into CRC cells (DLD-1 cells) through endocytosis. We then confirmed the efficacy of PP-P8, showing that oral administration of this agent significantly decreased tumor mass (∼ 42%) relative to controls in a mouse CRC xenograft model. In terms of molecular mechanism, PP-P8 induces cell-cycle arrest in G2 phase through down-regulation of Cyclin B1 and Cdk1. In this study, we performed in vivo toxicology profiling to obtain evidence that PP-P8 is safe, with the goal of receiving approval for an investigational new drug application (IND).Methods: Based on gene therapy guidelines of the Ministry of Food and Drug Safety (MFDS) of Korea, the potential undesirable effects of PP-P8 had to be investigated in intact small rodent or marmoset models prior to first-in-human (FIH) administration. The estimated doses of PP-P8 for FIH are 1.0× 1010 – 1.0× 1011 CFU/person (60 kg). Therefore, to perform toxicological investigations in non-clinical animal models, we orally administered PP-P8 at doses of 3.375 × 1011, 6.75 × 1011, and 13.5× 1011 CFU/kg/day; thus the maximum dose was 800– 8000-fold higher than the estimated dose for FIH.Results: In our animal models, we observed no adverse effects of PP-P8 on clinicopathologic findings, relative organ weight, or tissue pathology. Abstract

Aging, often considered a result of random cellular damage, can be accurately estimated using DNA methylation profiles, the foundation of pan-tissue epigenetic clocks. Here, we demonstrate the development of universal pan-mammalian clocks, using 11,754 methylation arrays from our Mammalian Methylation Consortium, which encompass 59 tissue types across 185 mammalian species. These predictive models estimate mammalian tissue age with high accuracy (r > 0.96). Age deviations correlate with human mortality risk, mouse somatotropic axis mutations and caloric restriction. We identified specific cytosines with methylation levels that change with age across numerous species. These sites, highly enriched in polycomb repressive complex 2-binding locations, are near genes implicated in mammalian development, cancer, obesity and longevity. Our findings offer new evidence suggesting that aging is evolutionarily conserved and intertwined with developmental processes across all mammals.

Abstract

Motile cells navigate through tissue by relying on tactile cues from gradients provided by extracellular matrix such as ligand density or stiffness. Mesenchymal stem cells (MSCs) and fibroblasts encounter adhesive or ‘haptotactic’ gradients at the interface between healthy and fibrotic tissue as they migrate towards an injury site. Mimicking this phenomenon, we developed tunable RGD and collagen gradients in polyacrylamide hydrogels of physiologically relevant stiffness using density gradient multilayer polymerization to better understand how such ligand gradients regulate migratory behaviors. Independent of ligand composition and fiber deformation, haptotaxis was observed in mouse 3T3 fibroblasts. Human MSCs however, haptotaxed only when cell-substrate adhesion was indirectly reduced via addition of free soluble matrix ligand mimetic peptides. Under basal conditions, MSCs were more contractile than fibroblasts. However, the presence of soluble adhesive peptides reduced MSC-induced substrate deformations; increased contractility may contribute to limited migration, but modulating cytoskeletal assembly was ineffective at promoting MSC haptotaxis. When introduced to gradients of increased absolute ligand concentrations, 3T3s displayed increased contractility and no longer haptotaxed. These data suggest that haptotactic behaviors are limited by adhesion and that although both cell types may home to tissue to aid in repair, fibroblasts may be more responsive to ligand gradients than MSCs.

Abstract

Heat transport in polymer nanocomposites reinforced with graphite nanoplatelets (GNPs) is studied using high-precision thermal conductivity measurements. The resistance to heat conduction across interfaces between GNPs and the polymer matrix has a strong effect on energy transport in the nanocomposites. The thermal conductivity is observed to increase when GNPs are pretreated with nitric acid to improve interfacial bonding. The improvement in the thermal conductivity, however, is much smaller than the corresponding improvement in mechanical properties. The thermal interface resistance extracted from the present thermal conductivity data is comparable to that obtained from the previously reported data on carbon nanotube suspensions. Abstract

A user may use a software bundle provided by vendors to control Consumer Electronics (CEs) in a home network environment. Software was developed as a bundle composed of various components and registered to an OSGi service registry, thus, a user can select a suitable bundle and implement it in a service gateway. There's no need to consider hardware when using a single bundle, because this was handled by hardware, service gateway specifications. But using several bundles to implement a business process for simultaneous control of various CEs is a different matter. If hardware resource sharing was not considered in development, then Quality of Service (QoS) of the business process cannot be guaranteed. Thus, we need an evaluation method for a user-defined business process, to guarantee QoS in business process modeling. This paper suggests a representative evaluation algorithm by considering metadata about bundle distribution as evaluation items, such as method response time, component response time, memory space and CPU capacity. Also, this paper suggests a business process simulator. It enables constructions of simulations for the efficiency of evaluation items and methods. A user uses a simulator to select a suitable bundle for hardware and easily makes and verifies a suitable business process for their goals. Abstract

When a televiewer watches Digital TV (DTV), one may encounter some program previews. If the televiewer wants to transcribe the main program, one has to search it and pre-engage for a transcription by using Electronic Program Guide (EPG) after memorizing the date, time, title, channel and so on. This happens because Program and System Information Protocol (PSIP), contains program information using EPG in DTV, is not served program information in the case of previews. Therefore, we propose a method and algorithm to provide program information for teletranscription without any change of the PSIP standard and implement them in DTV. This paper will introduce a convenient method to transcribe TV programs by taking advantage of Event Information Table (EIT), among the standards of PSIP, which contains information like title and on-air time. Abstract

The aim of this paper is to propose a Metadata-Net as ontology to make a relation with similar metadata by analyzing structure and semantics of multimedia content metadata. Also, it is to suggest a Multimedia Contents Management System based on a Metadata-Net ([M.sup.2]SM) as a methodology to provide integrated services for managing and searching various multimedia contents created and stored in Home Network-accessible Consumer Electronics (CEs). The proposed system helps users, who use a mobile device outside home, search and view highly relevant multimedia contents. When user wants to search some multimedia contents by metadata, [M.sup.2]SM provides query extension and modification and restriction using Metadata-Net in order to reduce increasing searching time caused by traversing unnecessary CEs in Home Network. The proposed system was implemented on a home server that acts as a gateway for establishing connections between Home Network and Internet. It supports various standards about Home Network and controls CEs to manage and search multimedia contents. In this way, [M.sup.2]SM reduces the search results delivered to mobile devices from Home Network, so power efficiency of mobile devices increase because CPU and network usage are declined. Abstract

Graphite nanoplatelets (GNPs) are an attractive class of reinforcement materials as they offer both high modulus and high strength. GNPs have a thickness of less than 100 nm although their planar dimensions can be as large as a few micrometers. As with any other nanoscale particles, their effectiveness as a reinforcement phase depends on the surface functionalization, which controls dispersion and interfacial bonding. A simple surface oxidation using nitric acid is shown to yield as good an improvement in composite properties as a few other functional groups chemically bonded to the surface do. The properties discussed include modulus, strength, fracture toughness, adhesive strength, thermal and electrical conductivities, and dielectric constants. Methods of further exfoliating graphite nanoplatelets are discussed together with the associated benefits on composite properties. Potential applications of graphite nanoplatelet composites are presented to conclude the report.
