Abstract: This White Paper presents an overview of the current status and future perspective of QCD research, based on the community inputs and scientific conclusions from the 2022 Hot and Cold QCD Town Meeting. We present the progress made in the last decade toward a deep understanding of both the fundamental structure of the sub-atomic matter of nucleon and nucleus in cold QCD, and the hot QCD matter in heavy ion collisions. We identify key questions of QCD research and plausible paths to obtaining answers to those questions in the near future, hence defining priorities of our research over the coming decades.

Abstract: Hot QCD physics studies the nuclear strong force under extreme temperature and densities. Experimentally these conditions are achieved via high-energy collisions of heavy ions at the Relativistic Heavy Ion Collider (RHIC) and the Large Hadron Collider (LHC). In the past decade, a unique and substantial suite of data was collected at RHIC and the LHC, probing hydrodynamics at the nucleon scale, the temperature dependence of the transport properties of quark-gluon plasma, the phase diagram of nuclear matter, the interaction of quarks and gluons at different scales and much more. This document, as part of the 2023 nuclear science long range planning process, was written to review the progress in hot QCD since the 2015 Long Range Plan for Nuclear Science, as well as highlight the realization of previous recommendations, and present opportunities for the next decade, building on the accomplishments and investments made in theoretical developments and the construction of new detectors. Furthermore, this document provides additional context to support the recommendations voted on at the Joint Hot and Cold QCD Town Hall Meeting, which are reported in a separate document., Comment: 190 pages, 69 figures

Abstract: This White Paper presents the community inputs and scientific conclusions from the Hot and Cold QCD Town Meeting that took place September 23-25, 2022 at MIT, as part of the Nuclear Science Advisory Committee (NSAC) 2023 Long Range Planning process. A total of 424 physicists registered for the meeting. The meeting highlighted progress in Quantum Chromodynamics (QCD) nuclear physics since the 2015 LRP (LRP15) and identified key questions and plausible paths to obtaining answers to those questions, defining priorities for our research over the coming decade. In defining the priority of outstanding physics opportunities for the future, both prospects for the short (~ 5 years) and longer term (5-10 years and beyond) are identified together with the facilities, personnel and other resources needed to maximize the discovery potential and maintain United States leadership in QCD physics worldwide. This White Paper is organized as follows: In the Executive Summary, we detail the Recommendations and Initiatives that were presented and discussed at the Town Meeting, and their supporting rationales. Section 2 highlights major progress and accomplishments of the past seven years. It is followed, in Section 3, by an overview of the physics opportunities for the immediate future, and in relation with the next QCD frontier: the EIC. Section 4 provides an overview of the physics motivations and goals associated with the EIC. Section 5 is devoted to the workforce development and support of diversity, equity and inclusion. This is followed by a dedicated section on computing in Section 6. Abstract: This report describes the physics case, the resulting detector requirements, and the evolving detector concepts for the experimental program at the Electron-Ion Collider (EIC). The EIC will be a powerful new high-luminosity facility in the United States with the capability to collide high-energy electron beams with high-energy proton and ion beams, providing access to those regions in the nucleon and nuclei where their structure is dominated by gluons. Moreover, polarized beams in the EIC will give unprecedented access to the spatial and spin structure of the proton, neutron, and light ions. The studies leading to this document were commissioned and organized by the EIC User Group with the objective of advancing the state and detail of the physics program and developing detector concepts that meet the emerging requirements in preparation for the realization of the EIC. The effort aims to provide the basis for further development of concepts for experimental equipment best suited for the science needs, including the importance of two complementary detectors and interaction regions. This report consists of three volumes. Volume I is an executive summary of our findings and developed concepts. In Volume II we describe studies of a wide range of physics measurements and the emerging requirements on detector acceptance and performance. Volume III discusses general-purpose detector concepts and the underlying technologies to meet the physics requirements. These considerations will form the basis for a world-class experimental program that aims to increase our understanding of the fundamental structure of all visible matter.

9. Direct and indirect constraints on CP-violating Higgs-quark and Higgs-gluon interactions

Abstract: We investigate direct and indirect constraints on the complete set of anomalous CP-violating Higgs couplings to quarks and gluons originating from dimension-6 operators, by studying their signatures at the LHC and in electric dipole moments (EDMs). We show that existing uncertainties in hadronic and nuclear matrix elements have a significant impact on the interpretation of EDM experiments, and we quantify the improvements needed to fully exploit the power of EDM searches. Currently, the best bounds on the anomalous CP-violating Higgs interactions come from a combination of EDM measurements and the data from LHC Run 1. We argue that Higgs production cross section and branching ratios measurements at the LHC Run 2 will not improve the constraints significantly. On the other hand, the bounds on the couplings scale roughly linearly with EDM limits, so that future theoretical and experimental EDM developments can have a major impact in pinning down interactions of the Higgs., Comment: Included analysis of Higgs decay processes. Abstract: Over the past decade, a large number of jet substructure observables have been proposed in the literature, and explored at the LHC experiments. Such observables attempt to utilize the internal structure of jets in order to distinguish those initiated by quarks, gluons, or by boosted heavy objects, such as top quarks and W bosons. This report, originating from and motivated by the BOOST2013 workshop, presents original particle-level studies that aim to improve our understanding of the relationships between jet substructure observables, their complementarity, and their dependence on the underlying jet properties, particularly the jet radius and jet transverse momentum. 12. Muon Flux Variations Measured by Low‐Cost Portable Cosmic Ray Detectors and Their Correlation With Space Weather Activity.

Abstract: We present a comparison of the measured cosmic ray (CR) muon fluxes from two identical portable low‐cost detectors at different geolocations and their sensitivity to space weather events in real time. The first detector is installed at Mount Wilson Observatory, CA, USA (geomagnetic cutoff rigidity Rc ∼ 4.88 GV), and the second detector is running on the downtown campus of Georgia State University in Atlanta, GA, USA (Rc ∼ 3.65 GV). The variation of the detected muon fluxes is compared to the changes in the interplanetary solar wind parameters at the L1 Lagrange point and geomagnetic indexes. In particular, we have investigated the muon flux behavior during three major interplanetary shock events and geomagnetic disturbances that occurred during July and August of 2022. To validate the interpretation of the measured muon signals, we compare the muon fluxes to the measurement from the Oulu neutron monitor (NM, Rc ∼ 0.8 GV). The results of this analysis show that the muon detector installed at Mount Wilson Observatory demonstrates a stronger correlation with a high‐latitude NM. Both detectors typically observe a muon flux decrease during the arrival of interplanetary shocks and geomagnetic storms. Interestingly, the decrease could be observed several hours before the onset of the first considered interplanetary shocks at L1 at 2022‐07‐23 02:28:00 UT driven by the high‐speed Coronal Mass Ejection and related geomagnetic storm at 2022‐07‐23 03:59:00 UT. This effort represents an initial step toward establishing a global network of portable low‐cost CR muon detectors for monitoring the sensitivity of muon flux changes to space and terrestrial weather parameters. Plain Language Summary: A pair of identical, low‐cost, and portable cosmic ray (CR) muon detectors is set up over 3,500 km apart for an exploratory study of monitoring space and terrestrial weather in real time at a global scale. One detector is installed on Mount Wilson, California and the other is in downtown Atlanta, Georgia. To validate the interpretation of the measured muon signals, the muon fluxes are compared to the well‐known neutron flux measurement from the Oulu neutron station in Finland. The results of this analysis show that the CR flux from both muon detectors typically decreases during geomagnetic storm events and that the muon detector installed on Mount Wilson is significantly correlated with the Oulu neutron monitor. Although this is yet an initial effort of building a global network of CR muon detectors for monitoring the space and earth weather in real time, the study provides evidence that muon network detection efficiency can be sufficient for a diagnostic of the major geoeffective space weather events. Abstract: Muon colliders provide a unique route to deliver high energy collisions that enable discovery searches and precision measurements to extend our understanding of the fundamental laws of physics. The muon collider design aims to deliver physics reach at the highest energies with costs, power consumption and on a time scale that may prove favorable relative to other proposed facilities. In this context, a new international collaboration has formed to further extend the design concepts and performance studies of such a machine. This effort is focused on delivering the elements of a $\sim$10 TeV center of mass (CM) energy design to explore the physics energy frontier. The path to such a machine may pass through lower energy options. Currently a 3 TeV CM stage is considered. Other energy stages could also be explored, e.g. an s-channel Higgs Factory operating at 125 GeV CM. Abstract: Among the post-LHC generation of particle accelerators, the muon collider represents a unique machine with capability to provide very high energy leptonic collisions and to open the path to a vast and mostly unexplored physics programme. However, on the experimental side, such great physics potential is accompanied by unprecedented technological challenges, due to the fact that muons are unstable particles. Their decay products interact with the machine elements and produce an intense flux of background particles that eventually reach the detector and may degrade its performance. Abstract: Over the past decade, a large number of jet substructure observables have been proposed in the literature, and explored at the LHC experiments. Such observables attempt to utilize the internal structure of jets in order to distinguish those initiated by quarks, gluons, or by boosted heavy objects, such as top quarks and W bosons. This report, originating from and motivated by the BOOST2013 workshop, presents original particle-level studies that aim to improve our understanding of the relationships between jet substructure observables, their complementarity, and their dependence on the underlying jet properties, particularly the jet radius and jet transverse momentum. This is explored in the context of quark/gluon discrimination, boosted W boson tagging and boosted top quark tagging.

34. Hyperproduction of alpha-hemolysin in a sigB mutant is associated with elevated SarA expression in Staphylococcus aureus.

Abstract: To evaluate the role of SigB in modulating the expression of virulence determinants in Staphylococcus aureus, we constructed a sigB mutant of RN6390, a prototypic S. aureus strain. The mutation in the sigB gene was confirmed by the absence of the SigB protein in the mutant on an immunoblot as well as the failure of the mutant to activate sigmaB-dependent promoters (e.g., the sarC promoter) of S. aureus. Phenotypic analysis indicated that both alpha-hemolysin level and fibrinogen-binding capacity were up-regulated in the mutant strain compared with the parental strain. The increase in fibrinogen-binding capacity correlated with enhanced expression of clumping factor and coagulase on immunoblots. The effect of the sigB mutation on the enhanced expression of the alpha-hemolysin gene (hla) was primarily transcriptional. Upon complementation with a plasmid containing the sigB gene, hla expression returned to near parental levels in the mutant. Detailed immunoblot analysis as well as a competitive enzyme-linked immunosorbent assay of the cell extract of the sigB mutant with anti-SarA monoclonal antibody 1D1 revealed that the expression of SarA was higher in the mutant than in the parental control. Despite an elevated SarA level, the transcription of RNAII and RNAIII of the agr locus remained unaltered in the sigB mutant. Because of a lack of perturbation in agr, we hypothesize that inactivation of sigB leads to increased expression of SarA which, in turn, modulates target genes via an agr-independent but SarA-dependent pathway.

Abstract: In this paper we report on the current status of studies on the expected performance for a detector designed to operate in a muon collider environment. Beam-induced backgrounds (BIB) represent the main challenge in the design of the detector and the event reconstruction algorithms. The current detector design aims to show that satisfactory performance can be achieved, while further optimizations are expected to significantly improve the overall performance. We present the characterization of the expected beam-induced background, describe the detector design and software used for detailed event simulations taking into account BIB effects. The expected performance of charged-particle reconstruction, jets, electrons, photons and muons is discussed, including an initial study on heavy-flavor jet tagging. A simple method to measure the delivered luminosity is also described. Overall, the proposed design and reconstruction algorithms can successfully reconstruct the high transverse-momentum objects needed to carry out a broad physics program., contribution to Snowmass 2021

Abstract: This White Paper presents the community inputs and scientific conclusions from the Hot and Cold QCD Town Meeting that took place September 23-25, 2022 at MIT, as part of the Nuclear Science Advisory Committee (NSAC) 2023 Long Range Planning process. A total of 424 physicists registered for the meeting. The meeting highlighted progress in Quantum Chromodynamics (QCD) nuclear physics since the 2015 LRP (LRP15) and identified key questions and plausible paths to obtaining answers to those questions, defining priorities for our research over the coming decade. In defining the priority of outstanding physics opportunities for the future, both prospects for the short (~ 5 years) and longer term (5-10 years and beyond) are identified together with the facilities, personnel and other resources needed to maximize the discovery potential and maintain United States leadership in QCD physics worldwide. This White Paper is organized as follows: In the Executive Summary, we detail the Recommendations and Initiatives that were presented and discussed at the Town Meeting, and their supporting rationales. Section 2 highlights major progress and accomplishments of the past seven years. It is followed, in Section 3, by an overview of the physics opportunities for the immediate future, and in relation with the next QCD frontier: the EIC. Section 4 provides an overview of the physics motivations and goals associated with the EIC. Section 5 is devoted to the workforce development and support of diversity, equity and inclusion. This is followed by a dedicated section on computing in Section 6. Abstract: Nitric oxide (NO)-sensitive soluble guanylyl cyclase (sGC), an enzyme that catalyzes the conversion of guanosine-5'-triphosphate (GTP) to cyclic guanosine-3',5'-monophophate (cGMP), transduces many of the physiological effects of the gasotransmitter NO. Upon binding of NO to the prosthetic heme group of sGC, a conformational change occurs, resulting in enzymatic activation and increased production of cGMP. cGMP modulates several downstream cellular and physiological responses, including but not limited to vasodilation. Impairment of this signaling system and altered NO-cGMP homeostasis have been implicated in cardiovascular, pulmonary, renal, gastrointestinal, central nervous system, and hepatic pathologies. sGC stimulators, small molecule drugs that synergistically increase sGC enzyme activity with NO, have shown great potential to treat a variety of diseases via modulation of NO-sGC-cGMP signaling. Here, we give an overview of novel, orally available sGC stimulators that Ironwood Pharmaceuticals is developing. We outline the non-clinical and clinical studies, highlighting pharmacological and pharmacokinetic (PK) profiles, including pharmacodynamic (PD) effects, and efficacy in a variety of disease models., (Copyright © 2018 Ironwood Pharmaceuticals Inc. 44. Analysis of genes encoding an alternative nitrogenase in the archaeon Methanosarcina barkeri 227.

Abstract: Methanosarcina barkeri 227 possesses two clusters of genes potentially encoding nitrogenases. We have previously demonstrated that one cluster, called nif2, is expressed under molybdenum (Mo)-sufficient conditions, and the deduced amino acid sequences for nitrogenase structural genes in that cluster most closely resemble those for the Mo nitrogenase of the gram-positive eubacterium Clostridium pasteurianum. The previously cloned nifH1 from M. barkeri shows phylogenetic relationships with genes encoding components of eubacterial Mo-independent eubacterial alternative nitrogenases and other methanogen nitrogenases. In this study, we cloned and sequenced nifD1 and part of nifK1 from M. barkeri 227. The deduced amino acid sequence encoded by nifD1 from M. barkeri showed great similarity with vnfD gene products from vanadium (V) nitrogenases, with an 80% identity at the amino acid level with the vnfD gene product from Anabaena variabilis. Moreover, there was a small open reading frame located between nifD1 and nifK1 with clear homology to vnfG, a hallmark of eubacterial alternative nitrogenases. Stimulation of diazotrophic growth of M. barkeri 227 by V in the absence of Mo was demonstrated. The unusual complement of nif genes in M. barkeri 227, with one cluster resembling that from a gram-positive eubacterium and the other resembling a eubacterial V nitrogenase gene cluster, suggests horizontal genetic transfer of those genes.

Abstract: Fibronectin-binding proteins (FnBPs) are thought to be important for the attachment of Staphylococcus aureus during infection. The regulation of the genes fnbA and fnbB by the global regulatory loci sar and agr was examined using site-specific regulatory mutants of S. aureus strain Newman. The results from binding assays using both aqueous and solid-phase fibronectin as well as ligand blotting with biotinylated fibronectin showed that the expression of FnBPA is enhanced in the agr mutant but inhibited in the sar mutant and the sar-agr double mutant. The same regulatory pattern was observed in Northern blot analysis using fnbA-specific probes. The introduction of sar on a multicopy plasmid increased the already enhanced fnbA transcription of the agr mutant. FnBPB was not detectable by ligand blotting and the fnbB promoter activity in promoter fusion assays was not affected by either sar or agr. The sequence encompassing ORF3 located upstream of sarA was found to be essential for the activation of fnbA transcription. We hypothesize that this sequence may modulate SarA expression and/or activity on the post-transcriptional level. Gel shift assays demonstrated that SarA binds to the fnbA promoter fragments, probably as a dimer. DNase I footprinting assays with SarA revealed a protected area of 102 bp upstream of fnbA.
