Neuronal activities of the human brain responsible for cognitive features have been theorized through several animal models that exhibited various complementary spatial learning modes by generating a flexible repertoire of behavioral strategies. However, for such studies associated with a neurodegenerative disease, which can be further manipulated to provide therapeutic strategies, the animal models employed in their developmental stages have been preferred over the adult ones. This pilot work was incepted to underscore the spatial memory capabilities that strengthened the intricate mechanism of memory acquisition potential in one of the low-order evolutionarily conserved species, such as zebrafish larvae. Initially, a reliable and more easily reproducible microfluidic platform integrating simple and intricate paths was designed to learn and test the spatial information in zebrafish larvae of 4-6 d.p.f. under non-invasive acoustic stimuli. Further, to acquire spatial information as the representation of spatial memory formation in zebrafish larvae, the acoustic startle responses were evaluated by quantifying various dynamic behaviors under distinct operating parameters. After significant conditioning sessions, the spatial memory was tested by employing variable 'freezing'. By the end of the 30 min-long test session, 6 d.p.f. larvae were found to exhibit the highest value of freezing of approximately 43% and 20% in the short and long paths, respectively. Even though a substantial rate of memory loss was observed, it can be envisaged to serve several behavioral strategies that process the dynamic cognitive memory among distinct spatiotemporal environments. Further, the proposed behavioral paradigm had the advantage of being more adaptable and reliably replicable by other researchers. As a consequence, different hypotheses can be readily tested to generate more reproducible findings towards distinct neurobehavioral characteristics. Therefore, the proposed paradigm for the consolidation of spatial memory based on the non-invasive spatial avoidance strategies could provide an enduring framework of reference for behavioral studies using zebrafish larvae.

Dementia is one of neurodegenerative disease without preventive medicine currently. Dextromethorphan (DXM) has been reported to reduce neuronal damage and neurodegeneration in animal and human models. The effect of DXM on the dementia has not been fully examined. We examined the medical records over 40 years old in Taiwan’s National Health Insurance Research Database between 2000 and 2015 to establish matched cohorts. We used a Cox regression hazard model to identify risk factors of dementia during 16 years of follow-up, and the results indicate that a significantly lower percentage of subjects with DXM use ( P < .001) developed dementia compared with those without DXM use (11.38%, 4541/39 895 vs 18.66%, 29 785/159 580). After adjustment for age and other variables [adjusted hazard ratio: .567 (95% confidence interval: .413-.678, P < .001)], this study also demonstrated that DXM use appeared to reduce the risk of developing dementia. DXM use may potentially provide a protective effect against dementia.

This study proposed a new experimental approach for the vascular and phenotype evaluation of the non-anesthetized zebrafish with representative imaging orientations for heart, pectoral fin beating, and vasculature views by means of the designed microfluidic device through inducing the optomotor response and hydrodynamic pressure control. In order to provide the visual cues for better positioning of zebrafish, computer-animated moving grids were generated by an in-house control interface which was powered by the larval optomotor response, in conjunction with the pressure suction control. The presented platform provided a comprehensive evaluation of internal circulation and the linked external behaviors of zebrafish in response to the cardiovascular parameter changes. The insights from these imaging sections was extended to identify the linkage between the cardiac parameters and behavioral endpoints. In addition, selected chemicals such as ethanol and caffeine were employed for the treatment of zebrafish. The obtained findings can be applicable for future investigation in behavioral drug screening serving as the forefront in psychopharmacological and cognition research.

BACKGROUND: Ovarian cancer (OCa) is the deadliest of all gynecologic cancers in the United States. Currently approved therapies have improved OCa survival for clinically localized disease, however, the majority (~90%) of patients with high-grade serous OCa (HGSOC) experience relapse with incurable metastases. There is a dire need for new therapeutic approaches. We hypothesized that the high basal endoplasmic reticulum stress (ERS) in OCa represents a critical and targetable vulnerability and may overcome the tumor heterogeneity. The objective of this project is to exploit increased ERS in ovarian cancer cells by engaging the novel target LIPA using the unique compound ERX-41. METHODS: The utility of ERX-41 as a new therapy was evaluated using MTT and CellTiter-Glo Cell Viability Assays. We used multiple established and patient derived OCa cell lines. The effect of ERX-41 on the Cell viability of patient-derived organoids (PDO) was measured using CellTiter-Glo 3D Assay. Long term effects of ERX-41 on cell survival were measured using colony formation assays. Apoptosis was measured using Annexin V and Caspase-Glo® 3/7 Assays. Cell cycle analysis was analyzed by Flow Cytometry. Mechanistic studies were done using LIPA knockout (KO) cells, RT-qPCR, and western blotting. Status of LIPA in OCa was determined using TNMplot database. In vivo efficacy of ERX-41 was tested using both cell line derived (CDX) and patient derived (PDXs) xenografts. RESULTS: TNM plot results showed that LIPA is highly expressed in OCa tumors compared to normal tissues and LIPA expression correlated with clinical grade. Kaplan-Meier plotter analyses of TCGA data revealed that LIPA expression is negatively correlated with overall survival in OCa patients. MTT and CellTitre-Glo assay results showed that ERX-41 significantly reduced the cell viability of both established and primary OCa cells, and PDO’s with an IC50 of ~500nM. ERX-41 treatment also significantly reduced the cell survival, increased S-phase arrest, and promoted apoptosis of OCa cells. A time course study revealed a robust and consistent induction of ERS markers (CHOP and sXBP1) in OCa cells by ERX-41 within 4h. Western blotting analyses also confirmed increased expression of ERS markers including CHOP, elF2α, PERK, and ATF4 upon ERX-41 treatment confirming that ERX-41 induces ERS. In xenograft studies, ERX-41 treatment resulted in ~66% reduction of tumor volume measured by Xenogen-IVIS. Further, in studies using PDX tumors, treatment with ERX-41 resulted in a significant reduction (~60%) of tumor volume and tumor weight. CONCLUSION: Collectively, our results suggest that ERX-41 is a novel therapeutic agent that targets the LIPA with a unique mechanism of action and implicate ERX-41 binding to LIPA induces ER stress, and apoptosis of OCa cells. Further molecular characterization of how ERX-41 binding to LIPA induces ER stress in OCa cells is ongoing. Citation Format: Alexia B. Background: Ovarian cancer (OCa) is the deadliest of all gynecologic cancers in the United States. Here, we tested the hypothesis that the high basal level of endoplasmic reticulum stress (ERS) in OCa represents a critical vulnerability and drugs that further aggravate this already engaged system in OCa may exhaust its protective features and contribute to apoptosis induction. The objective of this proposal is to identify a hit compound that enhances ERS in OCa and to conduct mechanistic studies. Methods: We synthesized a small library of >200 chemically distinct oligobenzamide analogs with maintenance of the chemical backbone but altered R groups of ERX-11. We performed the primary screening of this library to evaluate the induction of mRNA levels of two canonical ERS/UPR (unfolded protein response) genes- sXBP1 and CHOP. Biological activity of ERX-208 was validated using multiple OCa cells. Mechanistic studies were conducted using CRISPR/Cas9 KO, Western blotting, reporter gene assays, IHC and RNA-seq analysis. PK (pharmacokinetics) and toxicity studies were done using C57BL/6 mice. Cell line-derived xenografts (CDXs), patient-derived xenografts (PDXs), patient-derived explants (PDEs), and patient-derived organoids (PDO) were used for preclinical evaluation. Results: From a screen of a curated ERX-11 derived oligobenzamide library, we identified a hit compound, ERX-208 that potently (IC50~100nM) induces ERS/UPR and apoptosis in multiple OCa cells in vitro. CRISPR KO screen identified the lysosomal acid lipase A (LIPA) protein as the critical target of ERX-208. LIPA KO abrogates response to ERX-208, while reconstitution of LIPA restores ERX-208 response. The time course studies showed a robust and consistent induction (>15-fold CHOP, and >10-fold sXBP1) by ERX-208 treatment within 24h. We confirmed induction of classic UPR components peIF2α, CHOP and LC3B using Western blotting in multiple OCa cells. Functionally, ERX-208 causes growth inhibition of OCa cells, as noted by MTT cell viability assays using 15 OCa cells with an IC50 of ~50-100nM. The activity of ERX-208 is distinct among oligobenzamides as ERX-11 has limited/no activity against OCa cells. RNA-seq analysis confirmed that ERX-208 induces significant ERS, UPR, and apoptosis. Further, ERX-208 reduced the growth of OCa PDO’s in vitro, PDEs ex vivo and CDXs and PDXs in vivo. ERX-208 treatment did not show any signs of toxicity and body weight of mice was not affected. IHC analyses showed increased activation of ERS/UPR markers such as GRP78, p-PERK and decreased proliferation measured by Ki67. Conclusions: Collectively, our results demonstrated the utility of ERX-208 and will establish a novel therapeutic paradigm in OCa that overcomes tumor heterogeneity by targeting LIPA and enhancing ERS leading to apoptosis. Introduction: Estrogen receptor gene (ESR1) drives approximately 70-80% of breast cancer cases, which are primarily treated with endocrine therapies. Mutations in the ESR1 exclusively occur in the ligand binding domain (LBD) and are significantly enriched following endocrine therapy. These mutations retain the estrogen receptor (ER) expression thus conferring ligand-independent growth, which is a paramount contributor to endocrine therapy resistant disease. Worse progression-free survival is consistently associated with ESR1 mutation status; therefore, there is an unmet need for therapies against these mutations. To this end, we recently developed TX-245, a bis-benzamide molecule that hampers the function of the two most common ESR1 mutations - Y537S and D538G. Methods: TX-245 was evaluated in vitro using MTT cell proliferation assay and colony formation assay carried out in multiple ER+ breast cancer cells harboring wild type (WT) ESR1 or mutant (MT) ESR1. Approaches such as tumor spheroid cultures were also implemented to study the effect of TX-245 compared to selected SERDs/SERMs via CellTiter-Glo 3D viability assay. Mechanistic insights were obtained performing TR-FRET assay, fluorescence polarization assay, RNA-seq, ChIP-seq, western blot, and qRT-PCR. The therapeutic efficacy in vivo was evaluated using cell xenografts, patient derived xenografts (PDX), patient derived explants (PDE), and metastatic mouse models of MT-ER breast cancer. Results: TX-245 demonstrated greater anti-proliferative dose-dependent activity in models harboring WT and MT ESR1, including those resistant to fulvestrant and tamoxifen, when compared to other SERDs/SERMs. RNAseq data suggests that TX-245 dramatically alters the transcription of ERα-regulated genes, with repression of canonical estradiol-upregulated and induction of estradiol-repressed genes. TX-245 was able to antagonize ER-chromatin interaction and ER-transcriptional signaling in breast cancer cells driven by ESR1 mutation. TX-245 treatment downregulated ER protein expression and decreased expression of ER targeted genes (i.e., GREB and PGR) in multiple ER+ cell lines. TX-245 treatment as a single agent or in combination with palbociclib resulted in tumor regression in xenografts expressing WT, Y537S or D538G ESR1 mutation. PDX and PDE models consistently exhibited substantial tumor growth inhibition as well as decreased proliferation (Ki67 staining). Metastases studies using intracardiac and intratibial injection demonstrated that TX-245 inhibited the progression of established metastatic niches in mice injected with Y537S or D538G ER+ breast cancer cells. Conclusion: Our results indicate that TX-245 inhibits cell proliferation in cell lines that express WT or mutant ESR1, induce ER degradation, suppress ER-mediated signaling and reduce tumor burden in mouse models. These studies highlight the utility of TX-245 in targeting mutations in the ER-LBD. Artificial cilia-based microfluidics is a promising alternative in lab-on-a-chip applications which provides an efficient way to manipulate fluid flow in a microfluidic environment with high precision. Additionally, it can induce favorable local flows toward practical biomedical applications. The endowment of artificial cilia with their anatomy and capabilities such as mixing, pumping, transporting, and sensing lead to advance next-generation applications including precision medicine, digital nanofluidics, and lab-on-chip systems. This review summarizes the importance and significance of the artificial cilia, delineates the recent progress in artificial cilia-based microfluidics toward microfluidic application, and provides future perspectives. The presented knowledge and insights are envisaged to pave the way for innovative advances for the research communities in miniaturization.

3D bioprinting is one of the rapidly evolving fields of tissue engineering where microengineering meets cells biology within an unprecedented precision to construct tissue structures of various forms with complexity. However, enabling simultaneous printing of heterogeneous biomaterial along with scaffold components through the currently available printers is still considered as a major challenge due to the lack of instrumentation. Flow control is one of the major issues associated with the process. In this aspect, a microfluidic nozzle head equipped with two shape-memory alloy (SMA) actuators was proposed and integrated with a commercially available 3D printer to assist the biomaterial printing in a more systematic manner. The SMA actuator restrains the amount of flows for fabricating the desired scaffold components. Experimental results illustrated that the use of SMA actuator ensued a rapid and precise flow control of biomaterial and can further facilitate to maintain the width of any printed structures. As a proof of concept for the profound biomedical applications with the present manufacturing configuration, a 3D printed hydrogel platform was fabricated with demonstrated characters for later cell seeding after the printing further opens a new chapter in terms of biomaterial printing.

OBJECTIVEDespite intensive medical treatment, patients with glioblastoma (grade IV glioma [GBM]) have a low 5-year survival rate of 5.5%. In this study, the authors tried to improve currently used therapies by identification of a therapeutic target, IGFBP3, for glioma treatment.METHODSIGFBP3 RNA expression in 135 patients newly diagnosed with glioma was correlated with clinicopathological factors. Immunohistochemical analysis was performed to determine IGFBP3 protein expression in glioma specimens. The effect of IGFBP3 depletion on cell proliferation was examined using IGFBP3 knockdown glioma cells. Intracranial infusion of IGFBP3 siRNAs was performed to evaluate the effect of IGFBP3 depletion in mouse intracranial xenograft models.RESULTSWe demonstrated higher IGFBP3 expression in GBM than in tumor margin and grade II glioma. IGFBP3 expression was not only positively correlated with tumor grades but also associated with tumor histology and IDH1/2 mutation status. Additionally, higher IGFBP3 expression predicted shorter overall survival in glioma and GBM proneural subgroup patients. In vitro cell culture studies suggested IGFBP3 knockdown suppressed cell proliferation and induced cell cycle G2/M arrest as well as apoptosis in glioma cells. Also, accumulation of DNA double-strand breaks and γH2AX was observed in IGFBP3 knockdown cells. IGFBP3 knockdown delayed in vivo tumor growth in mouse subcutaneous xenograft models. Furthermore, convection-enhanced delivery of IGFBP3 siRNA to mouse brain suppressed intracranial tumor growth and prolonged survival of tumor-bearing mice.CONCLUSIONSOur findings suggest IGFBP3 predicts poor outcome of glioma patients and is a potential therapeutic target for which depletion of its expression suppresses tumor growth through inducing apoptosis and accumulation of DNA damage in glioma cells.

Low-lying triplet metal-to-ligand charge transfer (3MLCT) transitions of osmium complexes induced by spin–orbit coupling (SOC) are promising for extending the photocurrent response in dye-sensitized solar cells. In this study, we present a new osmium complex (CYC-33O), incorporating a 2-thiohexyl-3,4-ethylenedioxythiophene functionalized bipyridyl ancillary ligand to red-shift the absorption and enhance the absorbance of singlet and triplet metal-to-ligand charge transfer (1MLCT and 3MLCT) transitions. Time-dependent density functional theory (TDDFT) calculations show that the reinforced 1MLCT and 3MLCT transitions of CYC-33O are mainly from osmium to the 4,4′,4′′-tricarboxy-2,2′:6′,2′′-terpyridine anchoring ligand, increasing the heterogeneous electron transfer between CYC-33O and TiO2. The device sensitized with CYC-33O exhibits panchromatic conversion beyond 1000 nm, yielding a photocurrent density of 19.38 mA cm−2, which is much higher than those of the cells based on the ruthenium analogue (CYC-33R) and model osmium complex (Os-3) sensitizers.

This research proposes a novel 4H-SiC power device structure—different concentration floating superjunction MOSFET (DC-FSJ MOSFET). Through simulation via Synopsys Technology Computer Aided Design (TCAD) software, compared with the structural and static characteristics of the traditional vertical MOSFET, DC-FSJ MOSFET has a higher breakdown voltage (BV) and lower forward specific on-resistance (Ron,sp). The DC-FSJ MOSFET is formed by multiple epitaxial technology to create a floating P-type structure in the epitaxial layer. Then, a current spreading layer (CSL) is added to reduce the Ron,sp. The floating P-type structure depth, epitaxial layer concentration and thickness are optimized in this research. This structure can not only achieve a breakdown voltage over 3300 V, but also reduce Ron,sp. Under the same conditions, the Baliga Figure of Merit (BFOM) of DC-FSJ MOSFET increases by 27% compared with the traditional vertical MOSFET. Ron,sp is 25% less than that of the traditional vertical MOSFET.

The diarylgermylenes Ge(ArMe6)2 and Ge(AriPr4)2 (ArMe6 = C6H3-2,6-(C6H3-2,4,6-Me3)2), AriPr4 = C6H3-2,6-(C6H3-2,6-iPr2)2) are shown to react reversibly with ethylene. It is demonstrated that the more sterically crowded Ge(AriPr4)2 also reversibly binds propylene. The germirane products Ar2GeCH2CHR (Ar, R = ArMe6, H (1a), AriPr4, H (1b), and AriPr4, Me (1c)) were characterized via 1H and 13C NMR spectroscopy and by X-ray crystallography in the case of 1b. Thermolysis of Ge(AriPr4)2 under ethylene resulted in Ge–C bond homolysis and the formation of the digermene [(AriPr4)Ge(Et)]2 (1d). The thermodynamic parameters of the reactions were determined by variable-temperature 1H NMR spectroscopy.

The thermodynamic parameters of the reactions were determined by variable-temperature 1H NMR spectroscopy, and the experimental Gibbs free energies indicated their near-thermoneutrality and relatively weak alkyne coordination.

Photocatalysts as a semiconductor material are widely used in the field of water and environment cleaning applications as it equips with favorite features such as chemical and physical stability, easy availability, inexpensive, and non-toxic in nature. Additionally, photocatalysts can convert the light energy of the irradiation into the chemical energy of the electron-hole pairs. Most commonly used TiO2 can function as the efficient photocatalysts in the presence of light. However it is a material which majorly requires UV light for its activation, and it is not practically being useful. In this aspect, the proposed study demonstrated that with a combination of SnFe2O4 nanoparticles with magnetic artificial cilia, a highly efficient catalytic activity can be achieved under the visible light due to the rapid and uniform mixing within the microfluidic device with least energy budget. To identify the optimal advanced oxidation process using the selected photocatalyst running with the microfluidics, a micro-particle image velocimetry analysis was carried out through three modes of artificial cilia rotation. The study also determined the evolution curves of the degradation rate with respect to time for all of three modes of cilia rotation, and a superior performance was achieved with a maximum degradation rate of 81.7% in 60 min using the presented design concept.

The molecular characteristics of glioma patients, such as 1p/19q codeletion, MGMT promoter region methylation, and IDH1/IDH2 mutations, carry important information for tumor classification, biological behavior, and the prediction of patient survival. We sought to address the clinical characteristics of brain tumor patients in the Taiwanese population. Our aim was to compare the H&E-based classification and the 2016 WHO classification in glioma patients. 192 patients were recruited in this study including glioblastomas (GBMs) and lower-grade gliomas (LGGs). We performed assays to determine the statuses of 1p/19q codeletion, MGMT promoter methylation, and IDH mutations in tumors and assessed their associations with survival time. We also compared the difference between H&E and WHO 2016 classification. Using the H&E classification, the overall survival time (OST) was associated with IDH status in astrocytoma and oligoastrocytoma patients but not in oligodendroglioma and GBM patients. However, OST showed no significant correlation with MGMT promoter methylation in all the groups. Furthermore, no significant association was observed between 1p/19q codeletion and OST in GBM patients. Using the WHO 2016 classification, our results indicated that astrocytoma and oligodendroglioma patients with mutant IDH showed a significantly prolonged OST than patients with wild-type IDH. However, only oligodendroglioma patients with MGMT hypermethylation demonstrated statistically significant difference. Prognostic factors, including the age at diagnosis, IDH mutations, MGMT promoter methylation, and 1p/19q codeletion, could independently predict patient survival. Based on the WHO 2016 classification of glioma, the new histological groups provide a better and objective method to categorize glioma patients and predict patient survival.

Advances in microfluidics for zebrafish processing are critical to facilitate the bioassay in new drug development. Still, a manipulation platform for zebrafish relies mainly on the “static” agarose material and is coupled with the conventional fish facility to ensure the time-history investigation with the homogenous zebrafish population for drug screening. A fully automated IoT (Internet of Things) microfluidics system is necessary to be developed to enable smart and high-speed zebrafish testing without the need for fish facility support. In this work, a smart microfluidic device was presented to enable (i) highly efficient temperature control with the precision of ± 0.1 °C, (ii) remote zebrafish transport through light patterning, and (iii) perfusion and dynamic culturing of zebrafish all together assembled for on-chip high-quality imaging. A new microscale manipulation is envisaged to initiate a new chapter of running bioassays.

To generate immunity against human papillomavirus (HPV), the use of a recombinant DNA vaccine to carry an appropriate target gene is a promising and cost-effective approach. Granulocyte-macrophage colony-stimulating factor (GM-CSF) is a potent immunomodulatory cytokine that enhances the efficacy of vaccines by promoting the development and prolongation of humoral and cellular immunity. In this study, we linked codon-optimized GM-CSF (cGM-CSF) to the HPV16 E7 sequence as fused protein and evaluated the immunogenic potential of this DNA vaccine.We have demonstrated that cGM-CSF enhanced immunity against tumor challenges by generating and promoting the proliferation of HPV16 E7-specific CD8+ T cells, which secrete IFN-γ in the murine model. In this study, we aimed to evaluate the immunogenic potential of DNA vaccine that constructed by linking codon-optimized GM-CSF to HPV16 E7 sequence in the animal model. We study the half-life of RNA decay and cellular location of HPV16 E7 by Q-PCR and Western blot. We also assess immune response in the animal model by flow cytometry and ELISA.The cGM-CSF-E7 sequence increased and extended the expression of E7 mRNA, in comparison with the E7 sequence alone. Mice vaccinated with the cGM-CSF-E7 DNA vaccine exhibited a slower rate of tumor growth than those vaccinated with the unconjugated E7 DNA vaccine. We also found that the CD4 and CD8+ T cells from these mice showed strong secretion of IFN-γ.Through in vivo antibody depletion experiments, we demonstrated that both CD4+ and CD8+ T cells play an important role in the suppression of tumor growth.

Food coloring is often used as a coloring agent in foods, medicines and cosmetics, and it was reported to have certain carcinogenic and mutagenic effects in living organisms. Investigation of physiological parameters using zebrafish is a promising methodology to understand disease biology and drug toxicity for various drug discovery on humans. Zebrafish (Danio rerio) is a well-acknowledged model organism with combining assets such as body transparency, small size, low cost of cultivation, and high genetic homology with humans and is used as a specimen tool for the in-vivo throughput screening approach. In addition, recent advances in microfluidics show a promising alternative for zebrafish manipulation in terms of drug administration and extensive imaging capability. This pilot work highlighted the design and development of a microfluidic detection platform for zebrafish larvae through investigating the effects of food coloring on cardiovascular functionality and pectoral fin swing ability. The zebrafish embryos were exposed to the Cochineal Red and Brilliant Blue FCF pigment solution in a concentration of (0.02‰, 0.2‰) cultured in the laboratory from the embryo stage to hatching and development until 9 days post fertilization (d.p.f.). In addition, zebrafish swimming behaviors in terms of pectoral fin beating towards the toxicity screening were further studied by visualizing the induced flow field. It was evidenced that Cochineal Red pigment at a concentration of 0.2‰ not only significantly affected the zebrafish pectoral fin swing behavior, but also significantly increased the heart rate of juvenile fish. The higher concentration of Brilliant Blue FCF pigment (0.2%) increased heart rate during early embryonic stages of zebrafish. However, zebrafish exposed to food coloring did not show any significant changes in cardiac output. The applications of this proposed platform can be further extended towards observing the neurobiological/hydrodynamic behaviors of zebrafish larvae for practical applications in drug tests.

Genetic manipulation of zebrafish results in thousands of mutant strains and to efficiently preserve them for future use, zebrafish sperms have been cryopreserved in various cryopreservation centers. However, cryopreservation protocols are known to alter genetic entities. Therefore, there is an urgent need for an efficient method that can select morphologically superior and progressively motile zebrafish sperms after their activation for in vitro fertilization success. However, unlike those of other mammalian species, fish sperms do not take any physical or chemical cues to travel towards the egg. Their inertness towards any external cues makes the control of their orientation in a microfluidic environment difficult. In this aspect, a new microfluidic concept was demonstrated where PDMS baffles were inserted in the sidewalls to form microscale confinement creating a flow stagnation zone towards sperm retention. Two distinct microfluidic device designs were selected to evidence the improvement in sperm retention through the unique hydrodynamic feature provided by the microchannel design. Under similar flow conditions, 44% improvement was noticed for the device with a modified baffle design in terms of sperm retrieving efficiency. It was further noticed that with a flow tuning of 0.7 μL min-1, 80% of the total sperms swimming into the retention zones was retained within a specific time window. The present work further explains the significance of the hydrodynamic dependency of zebrafish sperm kinematics that paves the way for highly efficient spermatozoan manipulation.

In the current scenario of endovascular intervention, surgeons have to manually navigate the catheter within the complex vasculature of the human body under the guidance of X-ray. This manual intervention upsurges the possibilities of vessel damage due to frequent contact between the catheter and vasculature wall. In this context, a shape memory alloy-based miniaturized actuator was proposed in this study with a specific aim to reduce vessel wall related damage by improving the bending motions of the guidewire tip in a semi-automatic fashion. The miniaturized actuator was integrated with a FDA-approved guidewire and tested within a patient-specific vascular network model to realize its feasibility in the real surgical environment. The results illustrate that the miniaturized actuator gives a bending angle over 23° and lateral displacement over 900 µm to the guide wire tip by which the guidewire can be navigated with precision and possible vessel damage during the catheter intervention can certainly be minimized. In addition to it, the dynamic responses of the presented actuator were further investigated through numerical simulation in conjunction with the analytic analysis.

The zebrafish sperm activation profoundly depends upon the homogeneous mixing of the sperm cells with its diluent in a quick succession as it alters the cell’s extracellular medium and initiates their motility. Manual stirring, the traditional method for zebrafish sperm activation is tedious, time-consuming, and has a poor outcome. In this aspect, an artificial cilia embedded serpentine microfluidic is designed through which hydrodynamic factors of the microfluidic environment can be precisely regulated to harness uniform mixing, hence ensuring a superior sperm activation. To quantify the sperm motility, computer assisted sperm analysis software (CASA) was used whereas to quantify the generated flow field, micro particle image velocimetry (μPIV) was used. With this proposed microfluidic, 74.4% of the zebrafish sperm were activated which is 20% higher than its currently existing manual measurements. The μPIV analysis demonstrates that the curvature of the microchannel induces an orbital rotation to the flow field along the length of the microchannel together with the artificial cilia actuation which instigates a local rotation to the flow field of the artificial cilia location. The collective rotation in the whole flow field induce vorticity that promotes the change in temporal dynamics of the sperm cells towards their activation.

Flow propulsion and manipulation in a microscale flow regime are essential for the rapid processing of biomedical analytical assays that are performed on lab-on-a-chip platform. However, inherited from typically conical movement of artificial cilia in a cyclic manner, the generated backflow and flow oscillations during artificial cilia actuation are inevitably significant and post a significant barrier to the practical use of artificial cilia for accurate flow control. To address this problem, in this study we have hypothesized that by minimizing the traversing path of the artificial cilia during the recovery stroke could minimize the generated back flow and will result in an increment in the net flow propulsion. In this aspect, we have initiated the concept of the triangular beating pattern and compared its performance with the typical circular beating pattern. Upon comparison, it was observed that in the case of triangular beating pattern, the generated peak net flow velocity is almost double than the case of circular beating pattern. In particular, the underlying hydrodynamics induced during the actuation of the aforementioned two distinct beating patterns of artificial cilia were visualized and quantified with delineated comparison. This comparison was conducted based on flow dynamic characteristics measured through a micro-particle image velocimetry method. These results are important given that previous researchers do not explicitly recognize the role of the triangular beating pattern which possesses a significantly hydrodynamic advantage that can reduce the amount of back flow and surrounding flow fluctuations. The proposed concept provides a novel perspective on the microscale flow manipulation with promising applications in micropropulsion.
