Abstract

Being capable of characterizing DNA local bending is essential to understand thoroughly many biological processes because they involve a local bending of the double helix axis, either intrinsic to the sequence or induced by the binding of proteins. Developing a method to measure DNA bend angles that does not perturb the conformation of the DNA itself or the DNA-protein complex is a challenging task. Here, we propose a joint theory-experiment high throughput approach to rigorously measure such bend angles using the Tethered Particle Motion (TPM) technique. By carefully modeling the TPM geometry, we propose a simple formula based on a kinked Worm-Like Chain model to extract the bend angle from TPM measurements. Using constructs made of 575 base-pair DNAs with in-phase assemblies of 1 to 7 6A-tracts, we find that the sequence CA6CGG induces a bend angle of 19 [4] {\deg}. Our method is successfully compared to more theoretically complex or experimentally invasive ones such as cyclization, NMR, FRET or AFM. We further apply our procedure to TPM measurements from the literature and demonstrate that the angles of bends induced by proteins, such as Integration Host Factor (IHF) can be reliably evaluated as well., Comment: in Nucleic Acids Research published March 12, 2015

11. Evolution of mechanical properties of Ti6242S alloy after oxidation in air at 560°C: influence of solid salts deposits

Abstract

Titanium alloys are widely used in aerospace applications due to their good ratio of weight versus mechanical properties. When exposed to air at 560°C, Ti6242S titanium alloy presents very good oxidation resistance: a very thin oxide layer forms at its surface and oxygen dissolution inside the metallic material is rather limited. However, in real functioning conditions of the plane, near seas or oceans, the atmosphere contains NaCl, that can crystallise at the surface of hot parts. An active corrosion mechanism is established in these conditions, with catastrophic effect on the material behaviour at high temperature: very thick and brittle oxide scales and very important damaging of the metal outer part. Another issue is the formation of Na2SO4 specie by reaction of NaCl with kerosene combustion gases (SO2/SO3), leading to mixed NaCl/Na2SO4 deposits. The effect of exposure conditions on the mechanical properties of titanium alloy Ti6242S was evaluated through tensile tests performed on the raw alloy and after oxidation in air at 560°C of the specimens: without any deposit, with NaCl solid deposit, with NaCl/Na2SO4 solid deposit. The evolution of mechanical properties was interpreted in connexion with the microstructural modifications that occur during the high temperature ageing.

Abstract

International audience; In the past two decades, fibre metal laminates (FMLs) have been widely used to manufacture aircraft and aerospace components. FMLs are hybrid composite structures based on thin sheets of metal alloys reinforced by organic matrix composites (Sinmazçelik et al., 2011). When compared to mono-material solutions, FMLs provide added functionalities such as high specific bending strength, fatigue resistance, acoustical absorption, vibration transmissibility and damping characteristics. They overcome most of the limitations of metal and polymeric reinforced composites. The first generation of FMLs was based on thermoset resins. To shorten the process cycle times, lower the production cost and offer interesting end-of life options, a new generation of FMLs based on thermoplastic matrices was developed in a second time. Increasing environmental concerns currently encourage researchers to develop new ranges of recyclable composites based on plant fibres (Gomes Vieira et al., 2017). This last generation of sustainable and lightweight structural components could be integrated in a wider variety of applications including urban infrastructures and mobile street furniture. This work proposes an assessment of the durability of FMLs made of flax/polypropylene composite designed in view of their use for street furniture. The evolution of the static and dynamic (impact) properties of hybrid materials will be presented as a function of hydrothermal ageing time.

Abstract

Abstract Background Human T-cell leukemia virus type 1 and type 2 are related human retroviruses. HTLV-1 is the etiological agent of the Adult T-cell Leukemia/Lymphoma and of the Tropical Spastic Paraparesis/HTLV-1 Associated Myelopathy, whereas, HTLV-2 infection has not been formally associated with any T-cell malignancy. HTLV-1 and 2 genomes encode, respectively, the Tax1 and Tax2 proteins whose role is to transactivate the viral promoter. HTLV-1 and HTLV-2 Tax sequences display 28% divergence at the amino acid level. Tax1 is a shuttling protein that possesses both a non canonical nuclear import (NLS) and a nuclear export (NES) signal. We have recently demonstrated that Tax1 and Tax2 display different subcellular localization and that residues 90–100 are critical for this process. We investigate in the present report, whether Tax2 also possesses a functional NES. Results We first used a NES prediction method to determine whether the Tax2 protein might contain a NES and the results do suggest the presence of a NES sequence in Tax2. Using Green Fluorescent Protein-NES (GFP-NES) fusion proteins, we demonstrate that the Tax2 sequence encompasses a functional NES (NES2). As shown by microscope imaging, NES2 is able to mediate translocation of GFP from the nucleus, without the context of a full length Tax protein. Furthermore, point mutations or leptomycin B treatment abrogate NES2 function. However, within the context of full length Tax2, similar point mutations in the NES2 leucine rich stretch do not modify Tax2 localization. Finally, we also show that Tax1 NES function is dependent upon the positioning of the nuclear export signal "vis-à-vis" GFP. Conclusion HTLV-2 Tax NES is functional but dispensable for the protein localization in vitro.

35. Discovery of a new human T-cell lymphotropic virus (HTLV-3) in Central Africa

Abstract

Abstract Human T-cell Leukemia virus type 1 (HTLV-1) and type 2 (HTLV-2) are pathogenic retroviruses that infect humans and cause severe hematological and neurological diseases. Both viruses have simian counterparts (STLV-1 and STLV-2). STLV-3 belongs to a third group of lymphotropic viruses which infect numerous African monkeys species. Among 240 Cameroonian plasma tested for the presence of HTLV-1 and/or HTLV-2 antibodies, 48 scored positive by immunofluorescence. Among those, 27 had indeterminate western-blot pattern. PCR amplification of pol and tax regions, using HTLV-1, -2 and STLV-3 highly conserved primers, demonstrated the presence of a new human retrovirus in one DNA sample. tax (180 bp) and pol (318 bp) phylogenetic analyses demonstrated the strong relationships between the novel human strain (Pyl43) and STLV-3 isolates from Cameroon. The virus, that we tentatively named HTLV-3, originated from a 62 years old Bakola Pygmy living in a remote settlement in the rain forest of Southern Cameroon. The plasma was reactive on MT2 cells but was negative on C19 cells. The HTLV 2.4 western-blot exhibited a strong reactivity to p19 and a faint one to MTA-1. On the INNO-LIA strip, it reacted faintly with the generic p19 (I/II), but strongly to the generic gp46 (I/II) and to the specific HTLV-2 gp46. The molecular relationships between Pyl43 and STLV-3 are thus not paralleled by the serological results, as most of the STLV-3 infected monkeys have an "HTLV-2 like" WB pattern. In the context of the multiple interspecies transmissions which occurred in the past, and led to the present-day distribution of the PTLV-1, it is thus very tempting to speculate that this newly discovered human retrovirus HTLV-3 might be widespread, at least in the African continent.

TalentCampus was created in 2012 by a cluster of universities and higher education institutions, with a national agency for PhD support (ABG Intelli’agence). It was awarded funding through a national research call (IDEFI – french government investment for the future scheme). The aim of TalentCampus is to increase excellence in higher education by exploring the development of soft skills. However, the training sessions are not only for higher education students. Indeed, the idea is for participants to discover themselves through exchanges with people from many different backgrounds. Social background, age and disciplinary diversity are key factors in the success of TalentCampus sessions, which bring together high school pupils, students (from undergraduates to doctoral students), juniors and seniors professionals and job seekers. TalentCampus training sessions are based on active pedagogy, innovative content learning and collective immersion. It also creates training programs for companies or universities who have identified specific needs. Set up as 3 to 5-day workshops during the school holidays, TalentCampus develops skills to complement academic ability, including self-confidence, communication, leadership, emotional intelligence, and stress management. The courses are supervised by specialists from many different fields: for example, sport teachers or actors.Each training session is evaluated in order to know if the trainees have achieved the intended educational objectives. We will see in this communication what learners think about active pedagogy and mixed group during the training sessions. The sessions started in 2012 in Bourgogne Franche-Comté region. National and international developments are in progress as the target of TalentCampus is to disseminate and to let diffuse its best practices at a national and international levels. Session development in education system, lifelong training, as well as scientific research development aims to reach this target.; Desde el 2012, el proyecto francés TalentCampus ha creado cursos permitiendole a personas, desarrollar sus propias habilidades. De hecho, la idea es que personas se descubran a través de intercambios con diferentes personas de otros niveles. También sabemos que las personas se sienten más motivadas y quieren poner en marcha proyectos personales y profesionales. Abstract

Recently, unusual Raman signals were observed in different works conducted on thin zirconia layers grown on zirconium alloys simulating in‐reactor materials after high fluence ion irradiation or for samples cut from fuel rods irradiated in nuclear plants. As such, these spectra clearly do not correspond to any known spectrum of a pure standard zirconia phase. Therefore, the analysis conducted in this paper aims to provide a better understanding of these peculiar Raman features. For that purpose, specific ion‐irradiated samples were analysed. In situ Raman spectroscopy was first used to follow the irradiation process. Then, samples were characterized using different excitation wavelengths. Finally, the effects of oxygen isotopic substitution were examined in details. Results are discussed in terms of disorder and size‐related effects. Un atelier, proposé lors des sessions TalentCampus, a été animé lors des journées du séminaire Novatris. L'objectif de cet article est d'en présenter les objectifs et les résultats, ainsi que les premiers indices d'impact sur les compétences sociales des apprenants.

CIN3 Software; International audience; Many papers deal with the effect of water vapor on the high-temperature corrosion behavior of metallic materials. Experimental procedures are an essential consideration in the study of oxidation in steam, O2-H2O or air-H2O atmospheres, H2-H2O atmospheres, etc. The influence of various laboratory-controlled experimental parameters on the steam oxidation response of materials has been reviewed, and best practice recommendations are proposed in many papers. There are two main ways of enriching a gas in water vapor. The first consists in bubbling a gas (H2, air, N2, Ar, etc.) through a water bath held at a fixed temperature (thermostat). For example, synthetic air bubbled through a water bath held at 46°C gives a 10% H2O enrichment of the gas. The SiMap laboratory in Grenoble uses two successive water baths to obtain the desired water vapor content. The first bath is used to pre-enrich the gas by bubbling at a temperature below the target temperature. The second bath is heated to the temperature required for water vapor enrichment. The LICB team in Dijon and the LVVEEM team at Le Puy en Velay use a slightly different setup that involves a cooling tower: the gas is made to flow through a distilled water bath held at 70-80°C, and the H2O fraction is controlled and adjusted by passing the gas through a cooling tower heated to the correct temperature. Hayashi et al. use the same process to perform water vapor experiments. The experimental setup presented in Figure 1 possesses three water sources, which makes it very convenient for conducting long-term experiments as it is possible to switch from the first to the second or third bath, as many times as needed. Successive experiments in H216O, H218O and D2O can also be carried out by placing each isotope in a separate bath.
