The family of WD40-repeat (WDR) proteins is one of the largest in eukaryotes, but little is known about their function in brain development. Among 26 WDR genes assessed, we found 7 displaying a major impact in neuronal morphology when inactivated in mice. Remarkably, all seven genes showed corpus callosum defects, including thicker (Atg16l1, Coro1c, Dmxl2, and Herc1), thinner (Kif21b and Wdr89), or absent corpus callosum (Wdr47), revealing a common role for WDR genes in brain connectivity. We focused on the poorly studied WDR47 protein sharing structural homology with LIS1, which causes lissencephaly. In a dosage-dependent manner, mice lacking Wdr47 showed lethality, extensive fiber defects, microcephaly, thinner cortices, and sensory motor gating abnormalities. We showed that WDR47 shares functional characteristics with LIS1 and participates in key microtubule-mediated processes, including neural stem cell proliferation, radial migration, and growth cone dynamics. In absence of WDR47, the exhaustion of late cortical progenitors and the consequent decrease of neurogenesis together with the impaired survival of late-born neurons are likely yielding to the worsening of the microcephaly phenotype postnatally. Interestingly, the WDR47-specific C-terminal to LisH (CTLH) domain was associated with functions in autophagy described in mammals. Silencing WDR47 in hypothalamic GT1-7 neuronal cells and yeast models independently recapitulated these findings, showing conserved mechanisms. Finally, our data identified superior cervical ganglion-10 (SCG10) as an interacting partner of WDR47. Taken together, these results provide a starting point for studying the implications of WDR proteins in neuronal regulation of microtubules and autophagy.

Homeotherm animals adapt to external temperature fluctuations to maintain a constant body temperature. We hypothesised that the gut microbiota is involved in this adaptation, and identified change in the gut flora population in conditions of cold (6°C) and warm (34°C) temperatures. We showed that cold adapted gut microbiota could promote white adipose tissue browning associated with a better cold tolerance, improved insulin sensitivity and fat loss. The extinction of the bacteria A. muciniphila mediated by cold was sufficient to increase intestinal, villi and microvilli lengths that maximise the absorptive capacity of the intestine, thus helping compensating for the increased energy expenditure. In contrast, warm adapted gut microbiota induced whitening of adipose tissue and reduced body temperature. Additionally, warm exposure or warm adapted microbiota transplantation could improve the structural parameters and strength of the bone. Accordingly, both warm exposure and warm microbiota transplantation were effective in the prevention of osteoporosis.

This thesis is divided into two parts. A first part is dedicated to the study of diffusion phenomena in non trivial geometries. First, a work on the effects of irregularities of the geometry of a surface on a Galilean Brownian motion evolving on this surface is presented. This work has potential applications in biology, in modeling lateral diffusion phenomena on interfaces for instance. Studies concerning relativistic stochastic processes are then displayed. On the one hand, simple models of diffusion in an expanding universe are proposed and fluctuation-dissipation relations satisfied by these models are presented. On the other hand, a unified approach of the various relativistic stochastic processes existing in the literature, namely the ROUP, the Franchi-Le~Jan process and the Dunkel-Hänggi process, is introduced. A second part deals with two applications of the recently developed mean feld theory for General Relativity. The theory is applied to a Schwarzschild black hole and to an extreme Reisner-Nordström black hole. The characteristics of the mean space-time obtained in each of these applications are presented. The results are linked to observations of astrophysical black holes and to cosmological observations. The effects of the averaging on the thermodynamical properties of these black holes are also investigated., Cette thèse se compose de deux parties. Une première partie est dédiée à l'étude de phénomènes de diffusions dans des géométries non triviales. Je présente tout d'abord un travail sur les effets des irrégularités de la géométrie d'une surface sur un mouvement brownien galiléen évoluant sur cette surface. Ce problème est susceptible d'applications en biologie, notamment dans la modélisation des phénomènes de diffusions latérales sur des interfaces. J'expose ensuite des travaux réalisés sur des processus stochastiques relativistes. J'introduis d'une part des modèles simples de diffusion dans un univers en expansionet présente des relations de fluctuation-dissipation vérifiées par ces modèles. J'expose d'autre part une approche unifiée des différents processus stochastiques relativistes existant dans la littérature, à savoir, le ROUP, le processus de Franchi-Le~Jan et le processus de Dunkel-Hänggi. Dans une seconde partie, je présente deux applications de la récente théorie champ moyen de la relativité générale. La théorie a été appliquée à un trou noir de Schwarzschild et à un trou noir de Reisner-Nordström extrême. Les caractéristiques de l'espace-temps moyen obtenu dans chacun de ces deux cas sont présentées. Les résultats sont mis en relation avec des observations de trous noirs astrophysiques et avec des observations cosmologiques. Les effets de la moyennisation sur les propriétés thermodynamiques de ces trous noirs sont également étudiés.

In 2000 Ifremer launched and coordinated the development of a monitoring strategy for the REBENT network (REseau BENThique) in order to assess the aftermath of the “Erika” oil spill which took place in December 1999. Its aim is to provide consistent baseline knowledge on coastal benthic habitats, as well as to develop a monitoring tool that detects changes at various spatial and temporal scales. Sector-based seabed habitat mapping in shallow water (< 50 m deep), is currently being conducted throughout Brittany's coastal waters using a combination of geoacoustic marine systems together with ground-truthing using biological grab sampling and seabed observations. Due to the extended area, the overlap of soft and rocky bottoms, and contrasting hydrological conditions, the region of Tregor-Goelo is characterized by a mosaic of seabed habitats which host highly diversified fauna and flora. Six major soft-bottom macrofaunal communities were identified from 19 sampling stations. Detailed mapping of their distribution was achieved by means of morphosedimentary analyses based on a combination of remote sensing geoacoustic systems (multibeam echosounder and sidescan sonar), grab samplings and video observations. If at a regional scale, the description of benthic populations in the western Channel was made by Cabioch in 1968, the coastal area of Tregor-Goelo did not have so far been mapped for benthic habitats., Le REBENT (REseau BENThique), initié en 2000 à la suite de la catastrophe de l’Erika, a pour objectif de suivre les habitats benthiques côtiers et de détecter les changements à différentes échelles de temps et d’espace. L’approche sectorielle, opérée dans les petits fonds (< 50 m) des côtes bretonnes, combine des moyens de prospection acoustique, afin de délimiter les principaux types de substrats, avec des prélèvements et des observations biologiques pour caractériser les peuplements macrobenthiques. Du fait de l’étendue de la zone, de l’imbrication prononcée des fonds meubles et rocheux et de conditions hydrologiques contrastées, le secteur Trégor-Goëlo offre une mosaïque d’habitats particulièrement riches et diversifiés, tant en ce qui concerne la flore que la faune. Six unités majeures de peuplements ont été identifiées en substrats meubles, à partir des 19 stations biologiques échantillonnées. La cartographie détaillée de leur distribution a été rendue possible grâce aux travaux morphosédimentaires préalables s’appuyant sur la mise en oeuvre d’outils acoustiques (sondeur multifaisceaux et sonar à balayage latéral) et de validations terrain sous la forme de prélèvements sédimentaires et d’observations vidéo. Si à l’échelle régionale, la carte des peuplements benthiques de la Manche occidentale avait été réalisée par Cabioch en 1968, la zone côtière du Trégor-Goëlo n’avait, jusqu’à présent, fait l’objet d’aucune cartographie de ses peuplements benthiques.

The study area in the bay of Quiberon is characterised by a large array of sedimentological facies, mainly composed of sands and muds which, according to particle size, host different benthic communities : • a fine to medium slightly muddy sand Abra alba-Aponuphis bilineata community, • a sandy mud Abra alba-Ampharete finmarchica community, • a sandy mud to muddy sand Maldane glebifex community. The entrance into the bay is made through the Teignouse strait, which is subject to complex hydrodynamics that create a patchwork of strongly displaced fine and gravelly sand sediments. Here several population units, covering a small surface area, were identified. Included in the main characteristic species were Callista Chione, Travisia forbesii or Branchiostoma lanceolatum., Le REBENT (REseau BENThique), initié en 2000 à la suite de la catastrophe de l’Erika, a pour objectif de suivre les habitats benthiques côtiers et de détecter les changements à différentes échelles de temps et d’espace. L’approche sectorielle, opérée dans les petits fonds (< 50 m) des côtes bretonnes, combine des moyens de prospection acoustique, afin de délimiter les principaux types de substrats, avec des prélèvements et des observations biologiques pour caractériser les peuplements macrobenthiques. Le secteur étudié en baie de Quiberon se réfère pour l’essentiel à un vaste ensemble sédimentaire, composé de sables et de vases, qui, selon le degré d’envasement, abrite notamment : • un peuplement de sable fin à moyen, légèrement envasé, à Abra alba-Aponuphis bilineata, • un peuplement de vase sableuse à Abra alba-Ampharete finmarchica, • un peuplement de vase sableuse-sable vaseux à Maldane glebifex. L’entrée de la baie, communiquant avec le large par le passage de la Teignouse, est soumise à un hydrodynamisme complexe qui conditionne un important mosaïquage de sédiments fins et sablo-graveleux fortement remaniés. Plusieurs unités de peuplements, de faible superficie, y ont été identifiées avec, parmi les principales espèces caractéristiques, Callista Chione, Travisia forbesii ou encore Branchiostoma lanceolatum.
