A survey conducted in the 1920s, prior to the chestnut blight, indicated that chestnuts and oaks were codominant canopy species in White Oak Canyon, Shenandoah National Park, Virginia. A second survey in 1977 suggested that chestnuts were being replaced by tree species present before the blight, particularly oaks. In 2021, we resurveyed the 10 sites included in our 1977 survey and also recorded canopy and understory trees that grew above remnant chestnut sprouts. The canopy changed more substantially during the second interval (since 1977). Birch and maples were now more abundant. Hemlock declined, and oaks were less common in the canopy. In general, the trees considered as early to mid-successional have replaced oaks and hemlock. Chestnut sprouts have become much less common since 1977, presumably as repeated cycles of diebacks have weakened rootstocks. Those sites where chestnut sprouts have persisted until 2021 differed from neighboring sites without them. Chestnut sprouts were rare in sites with birch and hemlock; chestnut has persisted in locations with red oaks in the canopy and with few other understory competitors. This survey has been conducted over a longer time interval than previous studies that asked similar questions and our results suggest that changes to the forest composition following the loss of the American chestnut may be greater than previously recognized although the relative contribution of losing this codominant species is unclear. This article examines the use of white space in W.B. Yeats's poem "Among School Children" and its significance in conveying meaning. The author argues that the white space between stanzas invites readers to infer moments of contemplation and allusion to Yeats's personal life. The poem reflects Yeats's feelings of inadequacy and alienation as a schoolchild, as well as his reflections on his love for Maud Gonne. The white space serves as a transition between different thoughts and emotions in the poem. Additionally, the text discusses the philosophical themes in the poem, where Yeats questions the ability of ancient philosophers to prevent aging and death, and instead finds hope in memory and art. Yeats believed in the timeless nature of beauty and the enduring spirit of the soul. Dryocosmus kuriphilus, commonly known as the chestnut gall wasp, belongs to the family Cynipidae and is native to China. It is a highly invasive insect species causing serious damage to chestnut trees and has rapidly spread to various continents, including Europe, North America, and Oceania. The D. kuriphilus has become one of the important pests of chestnut plants in the world and is listed as a quarantine object by the European and Mediterranean Plant Protection Organization (EPPO). In this study, we used PacBio long reads, Illumina short reads, and Hi-C sequencing data to construct a chromosome-level assembly of the D. kuriphilus genome. The assembled genome includes 14,729 contigs with a total length of 2.28 Gb and a contig N50 of 0.8 Mb. With Hi-C technology, 2.17 Gb (95.02%) of contigs were anchored and oriented into the 10 pseudochromosomes with the scaffold N50 of 198.8 Mb and the scaffold N90 of 158.8 Mb. In total, 24,086 protein-coding genes were predicted in the assembled D. kuriphilus genome as the reference gene set. A total of 1.82 Gb repeats (occupying 79.7% of the genome), including 1.42 Gb of transposable elements and 0.40 Gb of tandem repeats, were identified in D. kuriphilus genome. In the evaluation of completeness, the BUSCO analysis determined a level of 98.1% completeness for the assembled genome sequences based on the Insecta database (OrthoDB version 10). The high-quality genome assembly of D. kuriphilus will not only provide a valuable reference for the study of its evolutionary history and genetic structure but also facilitate the research of host-pest interactions and invasiveness. Moreover, this genome assembly will promote in the development of effective management strategies to mitigate the economic and ecological impacts of this invasive pest on chestnut trees and ecosystems. Vegetable tannins are environmentally friendly tanning agents. However, they generally impart a dark colour to the tanned leather and highly contribute to the organic load in wastewaters. In this study, we employed a purification protocol separately on chestnut tannin (CT) and sulfited quebracho tannin (QT) to obtain the purified fractions (PCT and PQT). These samples were characterised by GPC, 1H NMR, 13C NMR, FT-IR, and HPLC–DAD techniques and applied for tanning tests. Through the purification process, non-tannin components and smaller molecules such as gallic acid, glucopyranose, and catechin were effectively removed from CT and QT, which consequently led to the reduced moisture content, pH value, and lighter colour of purified fractions. The crust leathers processed with PCT and PQT showed desirable light shades. Moreover, the organic loads in PCT and PQT tanning wastewater were reduced by 13.5% and 19.1%, respectively, when compared to those in traditional CT and QT tanning wastewater. Additionally, the physical and mechanical characteristics of crust leathers processed with PCT and PQT were comparable to those processed with CT and QT. Thus, purification of vegetable tannins may serve as a feasible strategy for producing light-colored vegetable-tanned leather while minimizing organic pollutant discharge during the vegetable tanning process. Colibacillosis, caused by avian pathogenic Escherichia coli (APEC), is one of the most prevalent and economically damaging bacterial diseases affecting poultry globally. Managing colibacillosis is difficult and frequently ineffective because APEC strains have developed widespread resistance to antibiotics, and the strict regulations and public concerns towards using antimicrobial agents in poultry further complicate the situation. This study aimed to evaluate the antibacterial properties of hydrolysable (sweet chestnut extract) and condensed (quebracho extract) tannins on Escherichia coli (E. coli) isolates from poultry, exploring their potential as antibiotic alternatives in managing colibacillosis. E. coli was isolated from the internal organs of deceased poultry across 18 farms, including layers, broilers, and broiler breeders. Each isolate was assessed for the presence of APEC strain predictors (virulence genes iutA, hlyF, iss, iroN, and ompT), antimicrobial resistance to 14 antibiotics using the disc diffusion method, and the presence of resistance genes for specific antibiotics (ampicillin, gentamicin, tetracycline, and quinolones). Out of 43 isolates, 27 (62.8%) were classified as APEC, 30 (69.8%) showed resistance to three or more antibiotic classes, and 32 (74.4%) carried at least one AMR gene. The minimum inhibitory concentrations (MICs) for the hydrolysable tannins from sweet chestnut extract (Castanea sativa Mill.) (SwCh) ranged from 0.5 to 3 mg/mL, while for the condensed tannins from quebracho extract (Schinopsis lorentzii) (Que), the MICs ranged from 1.5 to 4.5 mg/mL. The results indicate that both hydrolysable and condensed tannins possess significant in vitro antimicrobial activity against APEC, offering a potentially valuable alternative for controlling colibacillosis in the poultry industry. The chestnut tree (Castanea sativa Mill.) is a widespread plant in Europe, rich in high-value compounds, which can be divided mainly into monomeric polyphenols and tannins. These compounds exhibit various biological activities, such as antioxidant, as well as anticarcinogenic and antimicrobial properties. Chestnut wood (CW) extracts were prepared using different extraction techniques, process conditions, solvents, and their mixtures. This work aimed to test various extraction techniques and determine the optimal solvent for isolating enriched fractions of vescalagin, castalagin, vescalin, and castalin from CW residues. Supercritical CO2 extraction with a more polar cosolvent was applied at different pressures, which influenced solvent density. According to the results, the proportions of the components strongly depended on the solvent system used for the extraction. In addition, HPLC-DAD was used for semiqualitative purposes to detect vescalagin, castalagin, vescalin, and castalin. The developed valorization protocol allows efficient fractionation and recovery of the polyphenolic components of CW through a sustainable approach that also evaluates pre-industrial scaling-up. Castanea sativa is a high-value tree species whose stands have faced significant threats over the past century. The occurrence of one such threat, Chestnut Red Stain—a heartwood discoloration caused by the fungus Fistulina hepatica—has recently increased. This disease devalues the timber by up to 70% due to the rejection of stained wood by the industry. This study aimed to evaluate the effects of three management strategies on the occurrence of F. hepatica in chestnut coppice stands. Additionally, the co-occurrence of F. hepatica and Cryphonectria parasitica, both highly prevalent in the study area, was assessed. In 2013, three different silvicultural treatments were applied to five plots. Seven years later, the stand characteristics were measured, and the incidence of F. hepatica was evaluated using molecular tools. Data modeling revealed that the quadratic mean diameter before the interventions was related to the incidence of F. hepatica. Our results suggest that the thinning at earlier stand stages may be more critical than the type or intensity of the thinning in reducing the incidence of the fungus in the mid-term. Chestnut (Castanea sativa Mill.) forests in the Mediterranean region are facing increasing abandonment due to a combination of factors, ranging from climate change to socioeconomic issues. The recovery of chestnut ecosystems and their preservation and valorization are key to ensuring the supply of the wide spectrum of ecosystem services they provide and to preventing detrimental environmental shifts. The study's objective was to provide evidence on the effects of different management options on the ecophysiology of chestnut forests, with diverse pruning intensities (low, medium, and high intensity versus no pruning) tested in an abandoned chestnut stand in central Italy with the aim of recovering and rehabilitating it for fruit production. Innovative Internet of Things (IoT) 'Tree Talker' devices were installed on single trees to continuously monitor and measure ecophysiological (i.e., water transport, net primary productivity, foliage development) and microclimatic parameters. Results show a reduction in water use in trees subjected to medium- and high-intensity pruning treatments, along with a decrease in the carbon sequestration function. However, interestingly, the results highlight that trees regain their usual sap flow and carbon sink activity at the end of the first post-pruning growing season and fully realign during the following year, as also confirmed by the NDVI values. As such, this paper demonstrates the efficacy of recovering and managing abandoned chestnut forests, and the initial setback in carbon sequestration resulting from pruning is rapidly remedied with the advantage of reviving trees for fruit production. Additionally, the reduced water demand induced by pruning could represent a promising adaptation strategy to climate change, bolstering the resilience of chestnut trees to prolonged and intensified drought periods, which are projected to increase under future climate scenarios, particularly in the Mediterranean region. In this study, the concentrations of cadmium (Cd), iron (Fe), manganese (Mn), nickel (Ni), lead (Pb) and zinc (Zn) in plants sampled from Mt. Madra were investigated. Furthermore, the distribution characteristics and source identification of potentially toxic metals were investigated with the application of Positive Matrix Factorization (PMF) modelling. Samples of 26 different plant species were taken from Mt. Madra at elevations ranging from 177 to 1347 m using the multi-point sampling approach. The metal quantities measured by ICP-OES are the following sequences (mean ± SD) (mg/kg): Fe (974.96 ± 29.6) > Mn (111.81 ± 2.6) > Zn (27.28 ± 0.2) > Ni (2.17 ± 0.03) > Pb (0.77 ± 0.01) > Cd (0.12 ± 0.01). According to the plant samples in which the highest values were determined, the metals are as follows: Cd (Lathyrus laxiflorus, 0.401 mg/kg), Fe (Ajuga orientalis, 7621.207 mg/kg), Mn (Castanea sativa, 724.927 mg/kg), Ni (Prunella laciniata, 6.947 mg/kg), Pb (Crataegus stevenii, 3.955 mg/kg) and Zn (Prunella laciniata, 50.802 mg/kg). The results of the PMF model showed that Cd had an atmospheric transport factor originated and transported from industrial activites, Ni had a substrate factor, Fe, Mn, Pb and Zn were influenced by different anthropogenic factors. Chestnuts become sweetened with better tastes for fried products after cold storage, but the possible mechanism is not clear. The dynamics of sugar components and related physiological responses, as well as the possible molecular mechanism in chestnuts during cold storage, were investigated. Sucrose accumulation and starch degradation contributed to taste improvement. Sucrose content reached the peak after two months of cold storage, along with the accumulation of reducing sugars of maltose, fructose and glucose to a much lesser extent. Meanwhile, alpha-amylase and beta-amylase maintained high levels, and the activities of acid invertase and sucrose synthase increased. Transcriptome data demonstrated that differentially expressed genes (DEGs) were significantly enriched in the process of starch and sucrose metabolism pathway, revealing the conversion promotion of starch to sucrose. Furthermore, DEGs involved in multiple phytohormone biosynthesis and signal transduction, as well as the transcription regulators, indicated that sucrose accumulation might be interconnected with the dormancy release of chestnuts, with over 90% germinated after two months of cold storage. Altogether, the results indicated that cold storage improved the taste of chestnuts mainly due to sucrose accumulation induced by DEGs of starch and sucrose metabolism pathway in this period, and the sweetening process was interconnected with dormancy release. The production of high-quality aged marc spirits includes a minimum period of six months of ageing in oak barrels. Lastly, producers are interested in alternative botanical origin wood. The present study is aimed at investigating the influence of Greek oak (Quercus trojana) and Greek chestnut (Castanea sativa) compared with French (Quercus petraia) and American oak (Quercus alba) on the chemical composition and sensorial characteristics of aged tsipouro, produced from marc from the Black Muscat variety. Gas chromatography–olfactometry–mass spectrometry (GC–O–MS) was used to identify volatile compounds of the aged tsipouro. Also, colour and polyphenol measurements were made, and an organoleptic evaluation was performed by 16 trained tasters. The findings revealed rather similar results between the tsipouro made from different wood species, especially between Greek and French oaks, and relative differentiation for that of chestnut. All the aged distillates exhibited a pleasant and rich aromatic potential, dominated by floral and fruity terpene varietal aromas as well as wood-related volatiles. Chestnut, with a high phenolic potential, gives pleasant organoleptic effects over time and can be an alternative wood for ageing spirits. Escin, extracted from horse chestnut (Aesculus hippocastanum) has anti-edema and anti-inflammatory effects. It is used to treat several clinical conditions, including venous insufficiency, pain, inflammation, and edema. Considering escin's pharmacodynamic, the inhibition of the bradykinin pathway represents a particular effect, decreasing the local edema and conferring an advantage in comparison to other compounds. Chestnut internal rot is the main disease of chestnut during postharvest storage, which causes rot and mildew of chestnut kernel and brings huge economic losses to production. In order to clarify the pathogenic bacteria and its biological characteristics, the strain ZHZF21 was obtained by tissue isolation method. The strain was identified by morphological characteristics and ITS, TUB2 and CAL multi-gene sequence analysis. The pathogenicity of strain ZHZF21 was determined by mycelial inoculation method, and its biological characteristics were studied. The results showed that the colony of strain ZHZF21 was dark green ring concentric circles, with sexual and asexual reproduction stages. This strain was clustered with Colletotrichum fructicola numbered NDSTY31 in GenBank based on multi-gene sequences. Combined with colony morphological characteristics and molecular tree, strain ZHZF21 was preliminarily identified as Colletotrichum fructicola. The brown lesions after inoculation of strain ZHZF21 were consistent with the natural symptoms. The strain ZHZF21 could grow at 5~35 °C, the optimum temperature was 25 °C, the lethal temperature was 45~50 °C, and the optimum pH was 6. Under the condition of full darkness, strain ZHZF21 grew fastest, the utilization rates of arabinose and yeast were the highest, the utilization rate of lactose was the lowest, and urea could hardly be used. Background: Castanea sativa Mill. is important for ecosystems and societies. Its rich historical and cultural significance, remarkable ecological contributions, and diverse applications have inspired scientific research. This comprehensive review and conceptual mapping of chestnut research consolidates existing knowledge, identifies emerging trends, and highlights untapped potential to inform future investigations. First, the Scopus database was searched to retrieve all data-driven articles in English related to C. sativa published in English from 1951 to 2023. Second, the R language, Scopus Analytics, and VOSviewer were used to analyze the year of publication, authors, countries, affiliations, keywords, and citations. Finally, network analysis was performed to evaluate the hotspots and developmental trends of C. sativa. A total of 1889 research articles were recovered. Results: The articles showed an exponential progression, with a regression coefficient of 0.9435 (R2) and an annual growth rate of 6.28%. Italy (19.87%), Spain (15.56%), Portugal (13.39%), Turkey (6.92%), and Switzerland (5.17%) were the most prolific countries. C. sativa (798), sweet chestnut (54), cryphonectria parasitica (44), honey (38), phenolic compounds (35), chestnut blight (33) and antioxidant activity (31) are the keywords that occur the most frequently. The main research groups in the thematic map are "forest management," "Cryphonectria parasitica," "wood," "Spain" and "ethnobotany." Research on C. sativa has all the basic, motor, niche, and emerging or declining themes. Forest management, drought, Gnomoniopsis smithogilvyi, C. sativa shells, amino acids, honey, phenolic compounds, hydrolyzable tannins, antioxidant capacity, antioxidants, and extractives are trending topics. Conclusions: This bibliometric analysis highlights the importance of C. sativa research, revealing its ecological contributions, cultural significance, and diverse applications. Fresh-cut Chinese water chestnuts (CWCs) are susceptible to yellowing and browning during storage due to mechanical damage and the loss of protective outer skin, adversely affecting their marketability and shelf life. Methyl jasmonate (MeJA) is currently extensively used for food preservation, but it has not been used in Chinese water chestnuts. This study investigated the effect of MeJA treatment on the quality of fresh-cut CWCs. Fresh-cut CWCs immersed in 20 μM MeJA solution for 10 min and stored at 10°C for 5 d effectively delayed the yellowing process, reduced the respiration rate, and minimized the weight and soluble solids loss during storage. In addition, MeJA treatment induced the activities of superoxide dismutase (SOD) and catalase (CAT), which improved the antioxidant capacity of fresh-cut CWCs and inhibited the generation of reactive oxygen species (ROS). Meanwhile, MeJA treatment inhibited the activities of phenylalanine aminotransferase (PAL), polyphenol oxidase (PPO) and peroxidase (POD). The results of quantitative real-time PCR (qRT-PCR) showed that MeJA down-regulated the expression of CwCHS1, CwCHS2, CwCHS3 and CwCHI2 in freshly cut CWCs and inhibited the accumulation of flavonoids, thus delaying the surface discoloration of freshly cut CWCs. Phytophthora cinnamomi Rands devastates forest species worldwide, causing significant ecological and economic impacts. The European chestnut (Castanea sativa) is susceptible to this hemibiotrophic oomycete, whereas the Asian chestnuts (Castanea crenata and Castanea mollissima) are resistant and have been successfully used as resistance donors in breeding programs. The molecular mechanisms underlying the different disease outcomes among chestnut species are a key foundation for developing science-based control strategies. However, these are still poorly understood. Dual RNA sequencing was performed in C. sativa and C. crenata roots inoculated with P. cinnamomi. The studied time points represent the pathogen's hemibiotrophic lifestyle previously described at the cellular level. Phytophthora cinnamomi expressed several genes related to pathogenicity in both chestnut species, such as cell wall-degrading enzymes, host nutrient uptake transporters, and effectors. However, the expression of effectors related to the modulation of host programmed cell death (elicitins and NLPs) and sporulation-related genes was higher in the susceptible chestnut. After pathogen inoculation, 1,556 and 488 genes were differentially expressed by C. crenata and C. sativa, respectively. The most significant transcriptional changes occur at 2 h after inoculation (hai) in C. sativa and 48 hai in C. crenata. Nevertheless, C. crenata induced more defense-related genes, indicating that the resistant response to P. cinnamomi is controlled by multiple loci, including several pattern recognition receptors, genes involved in the phenylpropanoid, salicylic acid and ethylene/jasmonic acid pathways, and antifungal genes. Importantly, these results validate previously observed cellular responses for C. crenata. Collectively, this study provides a comprehensive time-resolved description of the chestnut-P. cinnamomi dynamic, revealing new insights into susceptible and resistant host responses and important pathogen strategies involved in disease development. Castanea sativa Mill. (C. sativa) processing and pruning generate several by-products, including leaves, burs, and shells (inner and outer teguments), which are considered an important source of high-value phytochemicals. Ellagitannins from C. sativa leaf extracts have been described to impair H. pylori viability and inflammation in gastric cells. Furthermore, chestnut shells showed an important anti-inflammatory effect in gastric epithelial cells. Dietary polyphenols, including tannins, have been reported to interfere with targets of inflammation, including the nuclear factor κB (NF-κB). A promising role as a further therapeutical target for gut disorders has been recently proposed for the regulatory subunit of hypoxia-inducible factor (HIF-1α), as a potential stabilizer of intestinal barrier integrity. Therefore, the main objective of this work is the chemical characterization of several chestnut by-products (bud, spiny bur, wood, pericarp and episperm), together with the exploitation of their anti-inflammatory properties in intestinal cells, scavenging capacity, and stability following gastrointestinal digestion. The chemical characterization confirmed the presence of bioactive polyphenols in the extracts, including ellagitannins. In CaCo-2 cells stimulated by an IL-1β-IFN-γ cocktail, nearly all chestnut by-products (50 µg/mL) inhibited the release of proinflammatory mediators (CXCL-10, IL-8, MCP-1, ICAM), along with the NF-κB-driven transcription, and induced the HRE-driven transcription. The stability of the most promising extracts, identified through PCA and cluster analysis, was addressed by in vitro gastrointestinal digestion. Despite the significant reduction in total polyphenol index of chestnut bud and wood after gastric and intestinal digestion, the activity of these extracts on both scavenging and anti-inflammatory parameters remained promising. These data contribute to exploit the potential of chestnut by-products as sources of dietary polyphenols with anti-inflammatory properties at the intestinal level. Moreover, this study could represent an important step to encourage the recycling and valorization of chestnut by-products, promoting the circular economy and reducing the environmental impact related to the management of agriculture waste. Kynurenic acid (KYNA) is an antioxidant degradation product of tryptophan that has been shown to have a variety of cytoprotective, neuroprotective and neuronal signalling properties. However, mammalian transporters and receptors display micromolar binding constants; these are consistent with its typically micromolar tissue concentrations but far above its serum/plasma concentration (normally tens of nanomolar), suggesting large gaps in our knowledge of its transport and mechanisms of action, in that the main influx transporters characterized to date are equilibrative, not concentrative. In addition, it is a substrate of a known anion efflux pump (ABCC4), whose in vivo activity is largely unknown. Exogeneous addition of L-tryptophan or L-kynurenine leads to the production of KYNA but also to that of many other co-metabolites (including some such as 3-hydroxy-L-kynurenine and quinolinic acid that may be toxic). With the exception of chestnut honey, KYNA exists at relatively low levels in natural foodstuffs. However, its bioavailability is reasonable, and as the terminal element of an irreversible reaction of most tryptophan degradation pathways, it might be added exogenously without disturbing upstream metabolism significantly. Many examples, which we review, show that it has valuable bioactivity. The current study investigated the impact of germination duration on the functional components (vitamin C, γ-aminobutyric acid (GABA), polyphenols, flavonoids) and antioxidant activity of germs and cotyledons of the germinated Chinese chestnut (Castanea mollissima). We utilized seeds of the "Zaofeng" Chinese chestnut to germinate, and sowed the seeds in wet sand at 22 °C and 85% relative humidity. The germination rate, length, diameter, and fresh weight of the sprouts were investigated at 0, 2, 4, 6, 8, and 10 days after sowing, and the kinetic changes of amylose, amylopectin, sugar components, soluble protein, vitamin C, GABA, total phenols, flavonoids, and the DPPH and ABTS free radical scavenging activity in the germs and cotyledons were monitored, respectively. The findings revealed that the germination rate and germ biomass increased continuously during germination. The germination rate reached 90% on the 8th day after sowing. Germination reduced amylose in cotyledons from 42.3% to 34.2%, amylopectin from 42.9% to 25.8%, total sugar from 12.6% to 11.4%, and vitamin C from 1.45 mg/g to 0.77 mg/g. Meanwhile, soluble protein in the embryos rose from 0.31% to 0.60%, vitamin C from 21.1 to 29.4 mg/g, GABA from 0.49 to 1.68 mg/g, total flavonoids from 53.6 to 129.7 mg/g, and ABTS antioxidant activity from 1.52 to 3.27 μmol TE/g. The average contents of D-fructose, inositol, vitamin C, GABA, polyphenols, and flavonoids and the DPPH and ABTS antioxidant activity in germs were as high as 22.5, 6, 35, 7.5, 10, 20, and 10 and 20-fold those of cotyledons, respectively. Especially, the average content of glucose in germ was as high as 80-fold that of cotyledon. D-xylulose, D-galacturonic acid, and D-ribose were only found in germs, but not in cotyledons. Considering the germ biomass and functional components content, germs of Chinese chestnuts germinated at 22 °C for 8 days are considered the most suitable raw material for functional food products. In conclusion, controlled germination not only enhances the physicochemical and functional properties of Chinese chestnut germs but also reduces the caloric content and improves the nutritional composition of the cotyledons appropriately. Moreover, the comprehensive evaluation of compositional changes and functionality in the embryo and cotyledon of Chinese chestnuts will provide a solid foundation for subsequent functional food processing utilizing germinated Chinese chestnuts. Eleocharis dulcis (Burm. f.) Trin. ex Hensch., commonly known as Chinese water chestnut, is a traditional aquatic vegetable in China, and now is widely cultivated throughout the world because of its high nutritional value and unique tastes. Here, we report the assembly of a 493.24 Mb telomere-to-telomere (T2T) genome of E. dulcis accomplished by integrating ONT ultra-long reads, PacBio long reads and Hi-C data. The reference genome was anchored onto 111 gap-free chromosomes, containing 48.31% repeat elements and 33,493 predicted protein-coding genes. Whole genome duplication (WGD) and inter-genomic synteny analyses indicated that chromosome breakage and genome duplication in E. dulcis possibly occurred multiple times during genome evolution after its divergence from a common ancestor with Rhynchospora breviuscula at ca. 35.6 Mya. A comparative time-course transcriptome analysis of corm development revealed different patterns of gene expression between cultivated and wild accessions with the highest number of differentially expressed genes (DEGs, 15,870) at the middle swelling stage and some of the DEGs were significantly enriched for starch metabolic process. This study was designed to investigate some physicochemical properties (color, electrical conductivity, ash, water-insoluble and mineral matter contents) of chestnut honey produced in the Ayancık, Türkeli and Erfelek regions of Sinop province, where chestnut forests are dense. The experimental data showed that most of the honey samples had different amber color tones. The electrical conductivity values and the amounts of water-insoluble matter for all chestnut honeys were determined above 0.8 mS/cm and below 0.1 g per 100 g, respectively. In addition, honey samples contained mainly potassium and calcium and other minerals such as magnesium, sodium, aluminum and iron. It has been demonstrated that these chestnut honeys are registered with Geographical Indication and meet the criteria required in the Turkish Food Codex Honey Communiqué of the Ministry of Agriculture and Forestry. In this study, antagonistic endophytic fungi were isolated from postharvest chestnut fruits; endophytic antagonistic fungi and their combination of inhibitory effects on the fungal pathogen Neofusicoccum parvum were evaluated. A total of 612 endophytic fungi were isolated from 300 healthy chestnut kernels, and 6 strains out of them including NS-3, NS-11, NS-38, NS-43, NS-56, and NS-58 were confirmed as antagonistic endophytic fungi against Neofusicoccum parvum; these were separately identified as Penicillium chermesinum, Penicillium italicum, Penicillium decaturense, Penicillium oxalicum, Talarmyces siamensis, and Penicillium guanacastense. Some mixed antagonistic endophytic fungi, such as NS-3-38, NS-11-38, NS-43-56, and NS-56-58-38, exhibited a much stronger antifungal activity against N. parvum than that applied individually. Among them, the mixture of NS-3-38 showed the highest antifungal activity, and the inhibition rate was up to 86.67%. The fermentation broth of NS-3, NS-38, and their combinations exhibited an obvious antifungal activity against N. parvum, and the ethyl acetate phase extract of NS-3-38 had the strongest antifungal activity, for which the inhibitory rate was up to 90.19%. The NS-3-38 fermentation broth combined with a chitosan coating significantly reduced N. parvum incidence in chestnuts from 100% to 19%. Furthermore, the fruit decay and weight loss of chestnuts during storage were significantly decreased by the NS-3-38 fermentation broth mixture along with a chitosan coating. Therefore, a mixture of P. chermesinum and P. decaturense could be used as a potential complex biocontrol agent to control postharvest fruit decay in chestnuts. [ABSTRACT FROM AUTHOR]

Chestnut weevil (Cuculio sp.) is one of the most nefarious insect pests, adversely affecting chestnut cultivation. Despite the economic importance of chestnut, genetic differences of susceptibility to weevil infestation among cultivars has not been clarified. We estimated the environmental variance for the infestation rate of chestnut weevil (IRCW) using seven cultivars/selections with two single-tree replicates for 11 years. Analysis of variance using root-squared-transformed values of the IRCW revealed significant effects of genotype, year, the genotype × year interactions, and tree within genotype. Year variance consisted of 66% of the total variance, while genetic variance and genotype × year interaction account for a certain percentage (11.9% and 12.9%, respectively). Linear regression analysis of the IRCW and nut harvesting date (NHD) revealed their close association. Because NHD is a high heritability trait, the genetic effect of IRCW would be mostly explained by the difference in NHD among cultivars. Interestingly, 'Riheiguri,' a Japanese–Chinese hybrid cultivar, has a lower IRCW than others even after adjustment for the effect of NHD. A 2 years evaluation of IRCW with five single-tree replicates confirmed that the IRCW of 'Riheiguri' was lower than 'Tsukuba,' a Japanese cultivar with almost the same NHD. Our results demonstrated that the year variance was the largest variance component of the IRCW, whereas the genetic difference was closely associated with the difference in NHD, but was also related to factor(s) other than NHD. Chestnut production is considered one of the most important economic resources of rural mountainous areas in Greece. Lately, producers report a steep rise in the incidence of brown rot disease caused by the fungus Gnomoniopsis smithogilvyi (Gnomoniaceae, Diaporthales), which results in severe chestnut rot. The pathogen is considered an emerging pathogen in many countries worldwide (Italy, France, Switzerland, Australia, New Zealand). This study aimed at (a) exploring the incidence of the brown rot disease in Vria (Regional Unit of Pieria, Region of Central Makedonia, Greece), (b) isolating and identifying the causal agent of the disease, (c) exploring the fungus presence at different phenological stages of the chestnut trees, and (d) implementing species-specific Bar- High Resolution Melting Analysis (HRM) for the early detection of G. smithogilvyi in chestnuts. G. smithogilvyi occurrence in chestnut tissues was more severe in June (59 %), nearly disappeared in July (19 %) and August (7 %) and increased again during harvesting time in September (57 %). This result could be attributed to a sum of different factors, including climate conditions. Moreover, it was demonstrated that G. smithogilvyi can be identified using a Bar-HRM analysis of chestnut tissues (buds, flowers and nuts). Results of this study clearly demonstrate that Bar-HRM can be used for the accurate, rapid and reliable identification of G. smithogilvyi universally on infected samples from different localities. Wood extractives have proven strong anti-oxidative properties which may be used to mitigate surface deterioration caused by photolytic effects and free radical formations. An interesting challenge regarding wood extractives is understanding how they behave in terms of treating natural wood surfaces to reduce anti-oxidative processes that arise from exposure to the main environmental factors. In this study, the possible efficacy of chestnut (Castanea sativa Mill.) extractives derived from native (CH) and thermally modified wood (TMT CH), as a means of protecting against UV exposure in poplar (Populus spp.) and spruce (Picea abies Karst.) wood, was evaluated. Chestnut wood was first thermally modified at 180 °C for 3 h, and the extractives were obtained by the accelerated solvent extraction technique (ASE) and subsequently used to treat wood surfaces. Samples were immersed in extractive solutions and exposed to artificial UV-weathering exposure, then contact angle and colour stability were monitored during the process. An FTIR analysis of the photo-degradation process of poplar and spruce surfaces was also executed. Extractives of TMT chestnut changed the total colour variation in both poplar and spruce wood. A much darker colour compared to the extractives of native wood was observed and an increase in ∆E* from 9.75 to 30.76 and 6.24 to 22.97 in poplar and spruce was calculated. The stability of the colour depended both on the surface wood and the type of extractive. The initial contact angle remained almost unchanged in the poplar wood surface and only slightly increased in spruce regardless of whether they were treated with extractives from native or TMT chestnut wood. A strong reduction in contact angle after the accelerated UV exposure test was observed, especially in spruce treated with CH extractives. FTIR analysis confirmed the lower levels of chemical degradation of surfaces observed by colorimetry, where TMT CH extractives formed more stable chemical bonds than native extractives. The comparative analysis in this study clarified the complex relationships between the effects of high-temperature modification of wood and the potential protective role of TMT extractives on some wood surfaces. The aim of this study was to investigate the differences between Castanea sativa Mill. and Castanea crenata Siebold & Zucc. × Castanea sativa Mill. in rooting ability in relation to endogenous levels of auxin, auxin cofactors and inhibitors that influence rooting success. Leafy cuttings of the two commercial cultivars 'Marsol' and 'Maraval' (Castanea crenata × Castanea sativa) and the native accession 'Kozjak' (Castanea sativa) were analyzed. Endogenous indole-3-acetic acid (IAA) concentration was assessed at the beginning of propagation (day 0); in addition, strigolactones, flavonoids, rooting ability and quality were assessed 120 days after. The concentration of endogenous IAA in 'Maraval' (324.34 ± 28.66 ng g−1) and 'Marsol' (251.60 ± 35.44 ng g−1) was significantly higher than in 'Kozjak' (112.87 ± 35.44 ng g−1). The best rooting result was observed with the genotypes 'Maraval' (100.00 ± 0.00%) and 'Marsol' (90.48 ± 6.15%). A significantly lower strigol concentration was observed in the roots of 'Maraval' (75.54 ± 17.93 ng g−1) compared with other genotypes. The total flavonoid concentration in 'Maraval' was significantly higher (2794.99 ± 187.13 μg g−1) than in 'Kozjak' (1057.38 ± 61.05 μg g−1). Our results indicate that the concentration of endogenous IAA has a significant influence on rooting success. The results further indicate that in the case of flavonoids and strigolactones, not only the individual compounds but also their ratio is important for rooting success. Correlation coefficients calculated between analyzed compounds and rooting success point toward specific functions of flavonoids and strigolactones in the rooting of Castanea that need to be functionally analyzed. The demand for wooden raw materials is constantly on the rise. Meeting this interest requires developing new timber supply chains, tackling illegal trade, using timber more efficiently, and enhancing the service life of wood-based products. From this point of view, sweet chestnut wood can be successfully used, alone or mixed with poplar, to manufacture Oriented Strand Board (OSB). This article assesses the fungal resistance of OSB/3 (load-bearing boards for use in humid conditions) made with different fractions of poplar (P) and sweet chestnut (SC) wood, in order to take advantage of the protective action of the tannins contained in the latter. Four types of OSB/3 were manufactured on an industrial scale: 100% P; 80% P and 20% SC; 50% P and 50% SC; 100% SC. The durability of the experimental boards was determined in accordance with the EN 113-3 standard (2023). No significant differences in mass loss were found for OSB/3 with different sweet chestnut wood fractions. Instead, the mass loss of these panels was significantly lower (i.e. fungal resistance was higher) than that of the poplar OSB/3 and poplar plywood tested for comparison. Based on the results, the use of sweet chestnut wood has interesting potential for the manufacturing of OSB/3 made with adequate gluing quality, suitable for applications in environments where fungal decay may occur. Characterization of the Cryphonectria parasitica population was initially done by a phenotypical assessment of 40 in vitro grown isolates obtained from 52 healing cankers collected from eight important chestnut-growing provinces of Turkey. The results of Bavendamm test, often correlated indirectly to hypovirulence, suggested 31 possibly hypovirulent and 9 virulent isolates. PCR tests amplified two regions of ORFs A and B of Cryphonectria hypovirus 1 (CHV-1) from 36 of 40 isolates. The PCR test confirmation was more sensitive than the Bavendamm test. Partial ORFA sequencing revealed 36 CHV-1 haplotypes belonging to Italian subtype (I), with all hypovirulent isolates being of EU-1 vc type. The CHV-1 from 10 native EU-1 isolates were first transferred to six European vc type testers, EU-2, EU-3, EU-5, EU-7, EU-26, and EU-44, having heteroallelism at one vic locus. The presence of the vic locus difference generally reduces virus transmission. The easiest and highest frequency virus transfer was obtained by vic4 and vic6 allelic differences, while the differences vic2 and vic7 made the transfer more challenging. Finally, in this study we successfully transferred CHV-1 to an EU-1 isolate obtained from the Bursa province to an EU-12 European tester isolate. [ABSTRACT FROM AUTHOR]

Street art, with a particular emphasis on murals, plays a crucial role in shaping the cultural DNA of contemporary cities. A prime example of this is the mural "Four Seasons with Kora" in Warsaw, which is dedicated to the renowned Polish artist Kora (Olga Jackowska). This large-scale mural, which combines the artist's portrait with a chestnut tree motif, visually changes with the season, influencing the artist's social perception. This study analyzed murals' functions in social, cultural, and ecological contexts, highlighting their role in informal education and as a platform for social dialogue and integration. Using research methods such as visual analysis and examining comments and reactions on social media, this work aimed to understand how a mural integrates with its surroundings and is perceived throughout different seasons. The results indicated that the mural has become an important element of public space, not only for beautifying the city but also for stimulating social and cultural reflection. Graphene was mixed with varnishes at different ratios and applied by spraying method on different cross-sections of various wood materials, and their wear and adhesion performances were determined. Graphene (0.25%, 0.50%, 1%); varnishes (water-based and polyurethane varnish) and wood materials (beech (Fagus orientalis Lipsky), chestnut (Castanea sativa Miller), yellow pine (Pinus sylvestris L.), and spruce (Picea orientalis (L.) Link.)) were used. Adhesion and abrasion tests were performed. A total of 480 test specimens were prepared, 5 specimens for each wood type, cross-sectional direction, graphene ratio, and varnish type for adhesion and abrasion tests. The adhesion of the samples was determined by ASTM D 4541-09E1 pull-off test and abrasion resistance was determined in accordance with ASTM 4060-10. The data obtained were statistically analyzed and the significance values within and between groups were determined. As a result, abrasion resistance and adhesion increased in graphene 2 (0.50%) in both varnish types.

Abstract Vegetable tannins are environmentally friendly tanning agents. However, they generally impart a dark colour to the tanned leather and highly contribute to the organic load in wastewaters. In this study, we employed a purification protocol separately on chestnut tannin (CT) and sulfited quebracho tannin (QT) to obtain the purified fractions (PCT and PQT). These samples were characterised by GPC, 1H NMR, 13C NMR, FT-IR, and HPLC–DAD techniques and applied for tanning tests. Through the purification process, non-tannin components and smaller molecules such as gallic acid, glucopyranose, and catechin were effectively removed from CT and QT, which consequently led to the reduced moisture content, pH value, and lighter colour of purified fractions. The crust leathers processed with PCT and PQT showed desirable light shades. Moreover, the organic loads in PCT and PQT tanning wastewater were reduced by 13.5% and 19.1%, respectively, when compared to those in traditional CT and QT tanning wastewater. Additionally, the physical and mechanical characteristics of crust leathers processed with PCT and PQT were comparable to those processed with CT and QT. Thus, purification of vegetable tannins may serve as a feasible strategy for producing light-colored vegetable-tanned leather while minimizing organic pollutant discharge during the vegetable tanning process. Abstract Flavonoids are crucial secondary metabolites that possess the ability to mitigate UV damage and withstand both biotic and abiotic stresses. Therefore, it is of immense significance to investigate the flavonoid content as a pivotal indicator for a comprehensive assessment of chestnut's drought tolerance. This study aimed to determine the flavonoid content and drought tolerance-related physiological and biochemical indices of six chestnut varieties (clones) grafted trees—Qianxi 42 (QX42), Qinglong 45 (QL45), Yanshanzaofeng (YSZF), Yanzi (YZ), Yanqiu (YQ), and Yanlong (YL)-under natural drought stress. The results were used to comprehensively analyze the drought tolerance ability of these varieties. The study revealed that the ranking of drought tolerance indices in terms of their ability to reflect drought tolerance was as follows: superoxide (oxide) dismutase (SOD) activity, ascorbate peroxidase (APX) activity, flavone content, catalase (CAT) activity, proline (PRO) content, soluble sugar content, peroxidase (POD) activity, betaine content, flavonol content, hydrogen peroxide (H2O2) content, soluble protein content, superoxide ion (OFR) content, superoxide (ion OFR) production rate, malondialdehyde (MDA) content, chlorophyll content. Through principal component analysis, the contents of flavonoids and flavonols can be used as indicators for comprehensive evaluation of drought tolerance of chestnut. The comprehensive evaluation order of drought tolerance of grafted trees of 6 chestnut varieties (Clones) was: QL45 > QX42 > YQ > YZ > YSZF > YL.

Wood constitutes a unique and valuable material that has been used from ancient times until nowadays in a wide variety of applications, in which the surface quality of wood often constitutes a critical factor. In this study, the influence of different wood areas and therefore, of different anatomical characteristic areas of chestnut wood (Castanea sativa Mill.) on the surface quality, was thoroughly studied, in terms of surface roughness. Five different chestnut tree trunks were harvested, from which five different disks were obtained corresponding to five different trunk heights. Surface roughness was measured on these disks on the transverse, radial, and tangential planes, on the areas of sapwood and heartwood, measuring the roughness in each point both vertically and in parallel to the wood grain. The results revealed that the examined roughness indexes (Ra, Rz, Rq) follow a parallel path to one another. In the case of all surfaces (transverse, radial, tangential) of the disks examined, when the measurement was implemented perpendicularly to the wood grain, a significantly higher roughness was recorded, compared to the wood grain measurements being implemented in parallel with the wood grain. Significant differences between heartwood and sapwood roughness were not demonstrated, although sapwood often appeared to exhibit a higher surface roughness than heartwood sites. Among the roughness values of the three different surfaces, the highest roughness in the vertical-to-wood-grain measurements was recorded by tangential surfaces, with slightly lower values on the transverse surfaces and the lowest roughness on radial surfaces. Meanwhile, for the measurements in parallel with the wood grain, the transverse surfaces presented significantly higher roughness values compared to the tangential and radial surfaces. Significant roughness differences were not detected among the surfaces at different trunk heights. Although, significant differences in roughness were recorded among different trees, it was observed that all the studied trees align with the identified and described within-tree trends. In this paper, chestnut and tea polyphenols were used as the main raw materials to develop tea polyphenol chestnut juice, aiming to explore the stability of tea polyphenols on the stability of cloudy chestnut juice and the digestive characteristics of chestnut starch.By adding ethyl gallate (EG), epigallocatechin gallate (EGCG), epicatechin gallate (ECG), epigallocatechin (EGC), epicatechin (EC), and mixed tea polyphenols to chestnut juice, it was found that the concentration of the five monomeric tea polyphenols with the least impact on the stability was 0.8 mg/mL.Tea polyphenols EC had the least impact on the stability of chestnut juice.Adding mixed tea polyphenols was most likely to cause instability in chestnut juice.The rheology analysis of chestnut juice showed that tea polyphenols could aggregate the starch particles in the juice through intermolecular force, leading to the reduction of the viscosity of chestnut juice and easy sedimentation.The addition of tea polyphenols would inhibit the digestion of chestnut juice, among which the addition of EG and EGCG chestnut juice had a better inhibition effect.According to the results of light transmittance, sedimentation accumulation, and in vitro digestion tests, it could be found that the addition of tea polyphenols would have an impact on the stability of chestnut juice and the digestive properties of chestnut starch.It provides a new research direction for the development of a tea polyphenol chestnut juice compound drink. BACKGROUND RESULTS CONCLUSION Chestnut fruit quality is affected by fungal contamination. The study of the patterns of contamination in the postharvest is crucial to individuate the critical phases and propose solutions. To understand how fungal colonization varies on fruits, the composition of mycobiota was investigated in postharvest handling and in between tissues (shell and kernel).Fungal sequences were clustered into 308 operational taxonomic units (OTUs). Biodiversity was higher in shell than kernel tissues. Results evidenced the risk of new contamination in specific phases such as the ‘cold bath’ and storage. Genera known as mycotoxin producers were detected in all phases. Specifically, 47 OTUs belonging to Penicillium, eight to Fusarium and two to Aspergillus genera were identified. While Fusarium spp. was sensitive to ‘warm bath’ phase, Penicillium spp. was largely insensitive and accumulated in storage conditions. Surprisingly, Aspergillus spp. was poorly represented. Aflatoxin, ochratoxin A, fumonisins and T‐2/HT‐2 detection was performed for shell and kernel, and process phases. Higher contamination was observed on shell than in kernel samples. While aflatoxins were within the European Union (EU) limits for dry fruits, Ochratoxin exceeded the EU limits. The present study represents the first report of fumonisins and T‐2/HT‐2 detection in chestnuts.Fungal contamination taxa is high in chestnut fruits following postharvest handling and storage. A parametrization of process phases such as the ‘warm bath’ is functional to reduce the risk for some taxa. Summary: Chestnut rice wine, also called banli mijiu (BLMJ), uses Chinese chestnut (Castanea mollissima Blume) in the fermentation of traditional glutinous rice wine. This study characterised the microbial communities and volatile compounds in BLMJ and explored their correlations. Headspace solid‐phase microextraction‐gas chromatography–mass spectrometry (HS‐SPME‐GC–MS) revealed that the major volatile compounds during BLMJ fermentation include eighteen important volatile compounds (OAV > 1). High‐throughput sequencing (HTS) demonstrated that the abundance and diversity of bacteria were significantly greater than those of fungi during the fermentation process. Among them, Kropensteria and Saccharomyces were the most abundant bacteria and fungi at the genus level during the fermentation of BLMJ. Spearman's correlation analysis revealed that eighteen important volatile compounds were significantly positively correlated with seven microbial genera (|r| > 0.9, P < 0.01), including six bacterial genera (Rhodococcus, Lactococcus, Pseudomonas, Janthinobacterium, Unclassified_f_Bacillaceae and Burkholderia‐Caballeronia‐Paraburkholderia) and one fungal genus (Saccharomyces). Therefore, these bacterias and fungi may be core functional microorganisms during BLMJ fermentation. Summary: In the current study, polyphenols were extracted from chestnut (Castanea sativa) shells (CSS) by two eco‐friendly techniques, namely microwave‐assisted extraction (MAE) and ultrasound‐assisted extraction (UAE). The bioaccessibility, phenolic composition and bioactivity of CSS extracts were explored upon in vitro gastrointestinal digestion considering a future nutraceutical use. The phenolic and flavonoid concentrations, bioaccessibility and antioxidant/antiradical properties of both extracts decreased during digestion. The bioaccessibility was 25% and 26%, respectively, for UAE and MAE extracts. The phenolic profile demonstrated that gallic acid, caftaric acid and catechin were the most bioaccessible compounds for both extraction techniques. The CSS extracts unveiled notable radicals scavenging efficiency, particularly against HOCl (18.32–75.44% inhibition) and ONOO− (16.78–86.11% inhibition), as well as hypoglycaemic (6.95–17.99% α‐amylase inhibition) and neuroprotective (16.44–54.66% AChE inhibition) properties upon digestion. Considering the undigested extracts, the UAE extract revealed better results than the MAE extract. Oppositely, among digested fractions the MAE digests disclosed the most promising outcomes. The remarkable contribution of phenolic compounds to the bioactivity of CSS extracts after in vitro digestion was proved by heatmap correlations, indicating that their pro‐healthy properties were modified after gastrointestinal digestion, being ascribed to the distinct phenolic composition. This study presents the preliminary outcomes of a methodology for the physical and mechanical characterization of various chestnut elements in different states of preservation. Strategizing conservation and retrofit interventions for timber is necessary, and to do this, it is necessary to establish an estimation of physical (transmissivity, thermal conductivity, humidity level, etc.) and mechanical properties (density, compressive or bending strength, etc.). This essential information is typically associated with timber defects, but there are lack of correlations. The primary objective is to establish correlations between thermal and density properties with the aim of preserving original assets. The investigation delves into the relationship between timber density and thermal properties through experimental non-destructive testing (NDT). Two NDTs were employed with the aim of correlating: penetrometric testing and active thermography investigations. The parametric study on the excitation period yielded valuable insights into the temporal dynamics of heat transfer within the timber, underscoring the significance of selecting appropriate excitation periods to capture precise thermal properties. Tabular data on relative humidity for salified, dried, and new samples provided a quantitative backdrop to these observations, unveiling the nuanced effects of humidity on the timber's thermal response. The results of this study are positioned to inform future conservation efforts by laying the groundwork for a comprehensive understanding of timber's mechanical properties. Particularly, the challenge lies in accurately estimating density, where surface tests are often less reliable than in-depth ones. Therefore, it is crucial to seek validation through other NDT tests, such as thermographic analysis and visual inspection, and hygrometric tests recognizing their importance in enhancing the reliability of density assessments. Simple Summary: Due to the growing world population, the sustainability of food and feed sources with high nutritional value has become a crucial issue. In this scenario, insects can constitute a low-impact protein source with high nutritional value. The growth and nutrient composition of insects are potentially influenced by rearing conditions, particularly by the selected growth substrate. In this study, we evaluated the chemical and functional characteristics of Tenebrio molitor larvae reared on different growth substrates: a traditional wheat bran substrate and an innovative substrate consisting of wheat bran supplemented with chestnut shell, a by-product of the chestnut agro-industrial chain. The results showed that the innovative growth substrates positively influenced the insects' survival suggesting a beneficial effect on larval health. The enrichment of the growth substrate with chestnut shell modified the protein and amino acid profile of insect meals, possibly indicating a shift in their metabolism. In addition, insect meals obtained from larvae reared on chestnut-shell-enriched substrate exhibited higher antibacterial and antioxidant activity, suggesting a potential beneficial effect when included in animal feed. Our results showed positive outcomes related to the design of innovative strategies for insect rearing, enriching larvae meal with beneficial health properties in line with sustainability and One Health principles. Tenebrio molitor larvae represent a sustainable protein source for food and feed. The aim of this study was to evaluate the supplementation of chestnut shell, a by-product of the agro-industrial chain, in growth substrates for T. molitor larvae rearing. Seven-week-old larvae were reared on three different growth substrates: the control group (CTRL) was fed wheat bran, treatment group one was fed wheat bran supplemented with 12.5% w/w chestnut shell (TRT1), and treatment group two was fed wheat bran supplemented with 25% w/w chestnut shell (TRT2). Larval weight, substrate consumption, and mortality were recorded weekly. After 14 days, insect meals were produced for bromatological and colorimetric analysis, and bacterial inhibition activity assay using a microdilution method. The amino acid profile of insects was determined using quantitative nuclear magnetic resonance spectroscopy. Our results showed a lower feed conversion ratio and higher larval survival rate % in TRT2 compared to CTRL (p < 0.05). Proteins and lipids of TRT2 were higher than other groups (p < 0.05). Important differences were observed in the amino acid profile of TRT1 and TRT2 compared to CTRL (p < 0.05). TRT1 and TRT2 showed higher E. coli inhibitory activity than CTRL (p < 0.05). In conclusion, chestnut shell supplementation improved the survival and functional characteristics of larvae and likely impacted the insects' metabolism. [ABSTRACT FROM AUTHOR]

This study delved into the larval development and the morphological and anatomical transformations that occur in the galls of chestnut trees (Castanea mollissima BL.) and are induced by the chestnut gall wasp Dryocosmus kuriphilus Yasumatsu (GWDK) across various stages: initial, growth, differentiation, maturity, and lignification. Chestnut galls in the five development stages were collected. Gall structural characteristics were observed with an anatomical stereomicroscope, and anatomical changes in galls were analyzed with staining and scanning electron microscope techniques. The chestnut gall wasp laid its eggs on young leaves and buds. Chestnut gall wasp parasitism caused plant tissues to form a gall chamber, with parenchyma, protective, and epidermal layers. The development of the gall structure caused by the infestation of the GWDK gall led to the weakening of the reactive oxygen species (ROS) elimination ability of the host. The accumulation of ROS led to cell wall peroxidation, resulting in structural damage and diminished host resistance, and the parenchyma layer exhibited significant nutrient supply and thickening. The thickness of the protective and epidermal layers varied notably across different growth stages. The oviposition of the chestnut gall wasp induced modifications in the original plant tissues, with gall formation being most favorable in young tissues, correlating with the maturity level of the host plant tissues. Variances in the internal structures of the galls primarily stemmed from nutrient supplementation, while those in the external structure were attributed to defensive characteristics. This research contributes a foundational understanding of gall development induced by the chestnut gall wasp in Chinese chestnut, offering valuable insights into the intricate interplay between insect infestation and plant physiology. Flavonoids are crucial secondary metabolites that possess the ability to mitigate UV damage and withstand both biotic and abiotic stresses. Therefore, it is of immense significance to investigate the flavonoid content as a pivotal indicator for a comprehensive assessment of chestnut's drought tolerance. This study aimed to determine the flavonoid content and drought tolerance-related physiological and biochemical indices of six chestnut varieties (clones) grafted trees—Qianxi 42 (QX42), Qinglong 45 (QL45), Yanshanzaofeng (YSZF), Yanzi (YZ), Yanqiu (YQ), and Yanlong (YL)-under natural drought stress. The results were used to comprehensively analyze the drought tolerance ability of these varieties. The study revealed that the ranking of drought tolerance indices in terms of their ability to reflect drought tolerance was as follows: superoxide (oxide) dismutase (SOD) activity, ascorbate peroxidase (APX) activity, flavone content, catalase (CAT) activity, proline (PRO) content, soluble sugar content, peroxidase (POD) activity, betaine content, flavonol content, hydrogen peroxide (H2O2) content, soluble protein content, superoxide ion (OFR) content, superoxide (ion OFR) production rate, malondialdehyde (MDA) content, chlorophyll content. Through principal component analysis, the contents of flavonoids and flavonols can be used as indicators for comprehensive evaluation of drought tolerance of chestnut. Objectives: The sweet chestnut Castanea sativa Mill. is the only native Castanea species in Europe, and it is a tree of high economic value that provides appreciated fruits and valuable wood. In this study, we assembled a high-quality nuclear genome of the ancient Italian chestnut variety 'Marrone di Chiusa Pesio' using a combination of Oxford Nanopore Technologies long reads, whole-genome and Omni-C Illumina short reads. Data description: The genome was assembled into 238 scaffolds with an N50 size of 21.8 Mb and an N80 size of 7.1 Mb for a total assembled sequence of 750 Mb. The BUSCO assessment revealed that 98.6% of the genome matched the embryophyte dataset, highlighting good completeness of the genetic space. After chromosome-level scaffolding, 12 chromosomes with a total length of 715.8 and 713.0 Mb were constructed for haplotype 1 and haplotype 2, respectively. The repetitive elements represented 37.3% and 37.4% of the total assembled genome in haplotype 1 and haplotype 2, respectively. A total of 57,653 and 58,146 genes were predicted in the two haplotypes, and approximately 73% of the genes were functionally annotated using the EggNOG-mapper.
