Abstract

Parkinson’s disease (PD) is the second most common neurodegenerative disorder and a central role for α-synuclein (αSyn; SNCA) in disease aetiology has been proposed based on genetics and neuropathology. To better understand the pathological mechanisms of αSyn, we generated induced pluripotent stem cells (iPSCs) from healthy individuals and PD patients carrying the A53T SNCA mutation or a triplication of the SNCA locus and differentiated them into dopaminergic neurons (DAns). iPSC-derived DAn from PD patients carrying either mutation showed increased intracellular αSyn accumulation, and DAns from patients carrying the SNCA triplication displayed oligomeric αSyn pathology and elevated αSyn extracellular release. Transcriptomic analysis of purified DAns revealed perturbations in expression of genes linked to mitochondrial function, consistent with observed reduction in mitochondrial respiration, impairment in mitochondrial membrane potential, aberrant mitochondrial morphology and decreased levels of phosphorylated DRP1Ser616. Parkinson’s iPSC-derived DAns showed increased endoplasmic reticulum stress and impairments in cholesterol and lipid homeostasis. Together, these data show a correlation between αSyn cellular pathology and deficits in metabolic and cellular bioenergetics in the pathology of PD.

Abstract

Agroforestry, being grounded in traditional land use practices, has developed as an autonomous science to increase productivity and profitability for the farmers, while ensuring the land use sustainability. Agroforestry systems are widespread in many countries, supporting the coexistence of tree, crop and livestock components. Such complex ecological systems offer a wide range of economic, social and environmental benefits, occurring over a range of spatial and temporal scales. The integrated use of GIS, Remote Sensing and GPS technologies is particularly suited for assessing, mapping and quantifying the intrinsic spatial complexity of these systems. One of the major geospatial issues in Agroforestry is detecting, mapping and estimating the forest component of the systems: scattered trees or linear forest formations located either inside the field or along the field boundaries, also known as Trees Outside Forest (TOF) (FAO 1998, 2001). Data on TOF are scarce and the information available is fragmented at regional and national levels (Schnell et al., 2015). Beckschäfer et al. (2017) give an overview of inventory approaches suitable for the science-based assessment of TOF, specifically on agricultural lands. However, up to now there are no guidelines for TOF inventory in agroforestry systems. Aims Traditional tree-based agriculture systems involving different multipurpose trees such as chestnuts (Castanea spp.), oaks (Quercus spp.), and olive (Olea europa), (Eichhorn et al., 2006) are common in Italy and other Mediterranean countries. We investigated the integration of geospatial techniques for TOF inventory in traditional silvoarable systems located in Umbria region (central Italy), where oaks tree hedgerows (THRs) coexist with herbaceous crops. Methods We tested a procedure for the GIS inventory of THRs, through the semiautomatic photo interpretation of high-resolution multispectral Sentinel-2 satellite images and NDVI. Results were compared with GPS field measurements of THRs as control points to assess the ground truth. We also compared THRs picked up by remote sensing products with different spatial resolution (Google Digital Globe, Sentinel-2 and Landsat 8) using the same combination of spectral bands. Results and conclusion The THRs length detected, corresponding to the 14% of the total perimeter of the cultivated fields, fits accurately with the GPS field survey. The THRs' crowns cover the 3% of the total cultivated area, with an incidence of 67 m of linear tree rows for each hectare of cultivated land. We also observed that the THRs' spatial distribution improves the connection between forested patches in the study area, enhancing landscape connectivity. Further development is needed in order to include diverse landscape patterns: the high-resolution Sentinel-2 imagery appear especially suitable for the detection of most TOFs at landscape level.

Abstract

Agroforestry denotes land use systems combining trees with agricultural crops and/or livestock. The woody component, consisting of scattered or linear trees, can be located either inside the field or along the field boundaries as tree hedgerows. Such land use approach aims to optimize both ecological interactions and economical revenue and offers a number of ecosystem services, environmental benefits, occurring over a range of spatial and temporal scales. The resulting complexity of the landscape patterns can be detected combining Remote Sensing, GIS spatial analysis and field surveying in order to understand the interactions between woody and crop components, and for assessing, mapping and quantifying the socio-economic values of the agroforestry systems services. In this study, we aimed to map and estimate the extent of Tree Hedge Rows (THR) in an Italian agroforestry landscape and to assess the influence of THRs on the yield of crops at the plot-farm scale.

Abstract

Internet users can download software for their computers from app stores (e.g., Mac App Store and Windows Store) or from other sources, such as the developers' websites. Most Internet users in the US rely on the latter, according to our representative study, which makes them directly responsible for the content they download. To enable users to detect if the downloaded files have been corrupted, developers can publish a checksum together with the link to the program file; users can then manually verify that the checksum matches the one they obtain from the downloaded file. In this paper, we assess the prevalence of such behavior among the general Internet population in the US (N=2,000), and we develop easy-to-use tools for users and developers to automate both the process of checksum verification and generation. Specifically, we propose an extension to the recent W3C specification for sub-resource integrity in order to provide integrity protection for download links. Also, we develop an extension for the popular Chrome browser that computes and verifies checksums of downloaded files automatically, and an extension for the WordPress CMS that developers can use to easily attach checksums to their remote content. Our in situ experiments with 40 participants demonstrate the usability and effectiveness issues of checksums verification, and shows user desirability for our extension.

Abstract

To verify whether it is possible, in people with diabetes mellitus (DM) considered at very high cardiovascular (CV) risk, stratify this risk better and identify significant modifiable risk factor (including lifestyle habits) to help patients and clinicians improve CV prevention. People with DM and microvascular diseases or one or more CV risk factors (hypertension, hyperlipidemia, smoking, poor dietary habits, overweight, physical inactivity) included in the Risk and Prevention study were selected. We considered the combined endpoint of non-fatal acute myocardial infarction and stroke and CV death. A multivariate Cox proportional analysis was carried out to identify relevant predictors. We also used the RECPAM method to identify subgroups of patients at higher risk. In our study, the rate of major CV events was lower than expected (5 % in 5 years). Predictors of CV events were age, male, sex, heart failure, previous atherosclerotic disease, atrial fibrillation, insulin treatment, high HbA1c, heart rate and other CV diseases while being physically active was protective. RECPAM analysis indicated that history of atherosclerotic diseases and a low BMI defined worse prognosis (HR 4.51 95 % CI 3.04–6.69). Among subjects with no previous atherosclerotic disease, men with HbA1c more than 8 % were at higher CV risk (HR 2.77; 95 % CI 1.86–4.14) with respect to women. In this population, the rate of major CV events was lower than expected. This prediction model could help clinicians identify people with DM at higher CV risk and support them in achieving goals of physical activity and HbA1c.

Abstract

In central Italy, Prunus cerasus var. austera is cultivated as small stands or scattered trees in marginal areas for the production of jam and wine. Thanks to the healthy attributes of its products and its ability to grow in different environmental conditions, this variety has gained new interest in the development of marginal areas. We assessed the level of the genetic variability of P. cerasus var. austera germplasm from central Italy and identified a 'core collection' representative of the present genetic diversity. A total of 161 trees, morphologically identified as var. austera, and one tree, identified as var. caproniana were collected and genotyped by 14 SSRs. Two individuals provided by a commercial plant nursery, one of P. cerasus var. caproniana and one of P. cerasus var. austera, were used as control. Thirteen SSRs presented private alleles in austera. Seven individuals morphologically identified as austera revealed private alleles specific to caproniana. The PCoA and Bayesian clustering analysis showed a main genetic group including var. austera, while a second group included all the caproniana-like genotypes. A core collection of 31 trees (46% of austera genotypes) was selected. This study can be considered as a starting point for future investigations on this variety. [ABSTRACT FROM AUTHOR]

Abstract

Coordinated Research Project (CRP) on EBR-II Shutdown Heat Removal Tests (SHRT) was established by International Atomic Energy Agency (IAEA). The aim of the project is to support and to improve validation of simulation tools and projects for Sodium-cooled Fast Reactors (SFR). The Experimental Breeder Reactor II (EBR-II) plant was a uranium metalalloy- fuelled liquid-metal-cooled fast reactor designed and operated by Argonne National Laboratory (ANL) for the U.S. Department of Energy at the Argonne-West site. In the frame of this project, benchmark analysis of one of the EBR-II shutdown heat removal tests, protected loss-of-flow transient (SHRT-17), has been performed at the Gruppo di Ricerca Nucleare San Piero a Grado (GRNSPG) in Pisa, Italy. The aim of this paper is to present modelling of EBR-II reactor design using RELAP-3D, and to present results of the transient analysis of SHRT- 17. Complete nodalization of the reactor was made from the beginning. Model is divided in primary side that contains core, pumps, reactor pool and, for this kind of reactor specific, Z pipe, and intermediate side that contains Intermediate Heat Exchanger (IHX). After achievement of acceptable steady-state results, transient analysis was performed. Starting from full power and flow, both the primary loop and intermediate loop coolant pumps were simultaneously tripped and the reactor was scrammed to simulate a protected loss-of-flow accident. In addition, the primary system auxiliary coolant pump that normally had an emergency battery power supply was turned off. Despite early rise of the temperature in the reactor, the natural circulation characteristics managed to keep it at acceptable leveles and cooled the reactor down safely at decay heat power levels. Thermal-hydraulics characteristics and plant behaviour was focused on prediction of natural convection cooling by evaluating the reactor core flow and temperatures and their comparison with experimental data that were provided by ANL. The plan is to improve the model in future with replacing current models of core and pool with 3D models, and, eventually, coupling with neutronic codes for more accurate results.

Abstract

Rapid growth of human pressure on natural resources has led profound changes in the natural environment. Habitat destruction, fragmentation and isolation are widespread and resulting in human-dominated landscapes. Many species have suffered local extinctions due to fragmentation and loss of natural habitats, while their ability to expand and re-colonize is hampered by the low degree of connectivity between the landscape fragments. Biodiversity conservation depends, therefore, on the maintenance of functional ecological networks and on the specific capacity of adaptation in fragmented habitats. Initiatives aimed at sustainability and preservation of agro-forestry resources are needed to halt biodiversity loss and to restore habitat connectivity. Understanding the complex interactions between biological, physical and socio-cultural components that shaped the current landscape, has stimulated the development of interdisciplinary methods. They consider the landscape, and all the variables associated with it, as a fundamental element of analysis. Research in the landscape ecology field has improved understanding of both the causes and the consequences of spatial heterogeneity, and how these vary with scale, coming to an effective influence on management of both natural and human-shaped landscapes. An abundance of spatially-explicit datasets and GIS technology supply critical tools for exploring spatial variation in several ecological variables. GIS spatial analysis functions are now essential to investigate the spatial relationships among biological, physical and anthropic landscape components. In this study, we explored the possibility to combine spatial analysis techniques with analytical tools coming from different research fields, providing new insights in sustainability and conservation study of agro-forestry resources. Landscape genetics approach (Manel et al. 2003) that combine data and methods of landscape ecology and population genetics, was applied to understand the spatial genetic structure of forest species populations and the effects of the landscape composition (post-glacial expansion, environmental and physical barriers) on gene flow and genetic variability. We investigated the origin and the processes that have affected the current distribution of Castanea sativa in Europe, and Juglans regia in Asia, through the analysis of the spatial structure and genetic diversity of existing populations, in relation to landscape and environmental variables. We used the overlay technique, described by Holderegger et al. (2010), based on (1) identification of the main clusters of populations (genetic structure by Bayesian clustering), (2) detection of genetic discontinuities or barriers, and (3) interpolation of genetic variables/membership coefficient (Qi) among individuals or populations (kriging, IDW resulting in genetic isolines). The genetic clusters, genetic discontinuities, barriers, or isolines are overlaid on topographical or land cover/land use maps to search for spatial coincidences with landscape elements. Isoscapes, namely joining the terms isotopes and landscapes, are maps that represent spatial variability of isotope composition, as related to biological and biophysical processes. We explored the potential of combining stable isotope analysis techniques (IRMS) and spatial analysis to understand the spatial distribution of isotope ratio variation in rural and forest resources. Starting from a first attempt concerning the integration of isoscapes analysis in traceability studies of biological products, we present future research perspectives.

Abstract

Background Trials have shown a beneficial effect of n-3 polyunsaturated fatty acids in patients with a previous myocardial infarction or heart failure. We evaluated the potential benefit of such therapy in patients with multiple cardiovascular risk factors or atherosclerotic vascular disease who had not had a myocardial infarction. Methods In this double-blind, placebo-controlled clinical trial, we enrolled a cohort of patients who were followed by a network of 860 general practitioners in Italy. Eligible patients were men and women with multiple cardiovascular risk factors or atherosclerotic vascular disease but not myocardial infarction. Patients were randomly assigned to n-3 fatty acids (1 g daily) or placebo (olive oil). The initially specified primary end point was the cumulative rate of death, nonfatal myocardial infarction, and nonfatal stroke. At 1 year, after the event rate was found to be lower than anticipated, the primary end point was revised as time to death from cardiovascular causes or admission to the hospital for cardiovascular causes. Results Of the 12,513 patients enrolled, 6244 were randomly assigned to n-3 fatty acids and 6269 to placebo. With a median of 5 years of follow-up, the primary end point occurred in 1478 of 12,505 patients included in the analysis (11.8%), of whom 733 of 6239 (11.7%) had received n-3 fatty acids and 745 of 6266 (11.9%) had received placebo (adjusted hazard ratio with n-3 fatty acids, 0.97; 95% confidence interval, 0.88 to 1.08; P=0.58). The same null results were observed for all the secondary end points. Conclusions In a large general-practice cohort of patients with multiple cardiovascular risk factors, daily treatment with n-3 fatty acids did not reduce cardiovascular mortality and morbidity.
