Abstract Background Perilla (Perilla frutescens L. Britt.) is an important oilseed and medicinal crop that frequently faces seasonal drought stress during seed germination, leading to a loss of dehydration tolerance (DT), which affects seed emergence and significantly reduces yield. DT has been successfully re-established for many species seeds. However, the physiological mechanisms and gene networks that regulate Perilla’s response to DT loss remain unclear. Results Phenotypic analysis determined that the window for DT in Perilla seeds occurs at radicle lengths of 0–4 mm. Integrating physiological and transcriptomic analyses revealed that the loss of DT promotes the production of reactive oxygen species (ROS) and regulates oxidase activity and gene expression. This implies that DT may influence seed germination by modulating ROS activity. Four radicle length (i.e., 0, 1, 3, and 4 mm) stages were analyzed, and 262 differentially expressed genes (DEGs) were identified that responded to DT. The majority of these genes were associated with epigenetics, cell function, and transport mechanisms. Analysis of expression data shows that desiccation inhibits the signaling network of genes encoding small secreted peptides (SSPs) and receptor-like protein kinases (RLKs). Finally, a relevant network diagram of DT response was proposed. Based on this information, we have revealed the metabolism regulation maps of the four main pathways involving these DEGs (i.e., metabolic pathways, cell cycle, plant hormone signal transduction, and motor proteins). Conclusions In conclusion, these findings deepen our understanding of gene network responses to DT during Perilla seed germination and provide potential target genes for the genetic improvement of drought resistance in this crop.

Abstract Controlling the nonlinear relationship between surface plasmon polariton (SPP) mode index and chemical potential of graphene can be used in the field of active transformation optics. Here, we propose an electrically tunable 2D Graded Photonic Crystal (GPC) lens based on graphene SPP platform. Our platform comprises a graphene monolayer integrated into a back-gated structure with nano-patterned gate insulators. When the chemical potential of the graphene surface is designed to operate in the nonlinear region, the designed GPC lens can be continuously transformed between a Maxwell’s fish-eye lens and a Luneburg lens by tuning the gate voltage. The range of the lens background chemical potential for allowing this transformation is systematically studied. To compensate for the significant errors inherent in the conventional effective medium theory (EMT) during the homogenization of photonic crystals (PCs), we propose a generalized effective medium theory (GEMT). The validity and accuracy of this approach are verified through comparisons with true values (based on rigorous eigenvalue solutions) and EMT values. Due to its advantages of on-site controls and easy fabrication characteristics, the proposed graphene GPC provides a new way for practical on-chip light manipulation.

ABSTRACT: Due to its beneficial effects on human health, Bifidobacterium is commonly added to milk powder. Accurate quantification of viable Bifidobacterium is essential for assessing the therapeutic efficacy of milk powder. In this study, we introduced a novel propidium monoazide (PMA)-antibiotic fluorescence in situ hybridization (AFISH)-flow cytometry (FC) method to rapidly and accurately quantify viable Bifidobacterium cells in milk powder. Briefly, Bifidobacterium cells were treated with chloramphenicol (CM) to increase their rRNA content, followed by staining with RNA-binding oligonucleotide probes, based on the AFISH technique. Then, the DNA-binding dye PMA was used to differentiate between viable and nonviable cells. The PMA-AFISH-FC method, including sample pretreatment, CM treatment, dual staining, and FC analysis, required approximately 2 h and was found to be better than the current methods. This is the first study to implement FC combined with PMA and an oligonucleotide probe for detecting Bifidobacterium.

Background/objective: Physical activity (PA) self-efficacy plays a crucial role in maintaining and enhancing PA behaviors in children. However, the effectiveness of eHealth interventions in boosting PA self-efficacy among children remains uncertain. Furthermore, which behavior change techniques (BCTs) used in eHealth interventions can positively influence children's PA self-efficacy needs further exploration for designing tailored eHealth interventions. Therefore, this systematic review and meta-analysis aimed to identify the effectiveness of eHealth interventions and BCTs in promoting children's PA self-efficacy. Methods: Following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines, a comprehensive search was conducted across six databases (PubMed, Web of Science, EBSCOhost, Ovid, SPORTDiscus, PsycINFO) up to January 8, 2024. Inclusion criteria included randomized controlled trials (RCT), quasi-experimental, and two-group experiments that examined the effect of eHealth interventions on PA self-efficacy among healthy children aged 0–18 years. The Physiotherapy Evidence Database (PEDro) scale was utilized to assess the risk of bias. Random effects meta-analysis was performed to determine the effectiveness of eHealth interventions and BCTs in selected studies. Results: Sixteen studies were screened, including 6020 participants with an average age of 11.58 years (SD = 2.87). The result showed small but significant intervention effects with high heterogeneity (I2 = 92.34 %) for postintervention PA self-efficacy (Hedges’ g = 0.315; 95 % CI = 0.069, 0.562, p = .012). Two BCTs were significantly associated with enhanced PA self-efficacy: instruction on performing the behavior (p = .003) and behavior demonstration (p = .036). Additionally, studies that adopted social support (unspecified) a nd prompt/cues were significantly less effective than studies that did not use these BCTs (p = .001). Conclusions: The findings showed that eHealth interventions positively affect children's PA self-efficacy. This review is the pioneer in focusing on BCTs in eHealth interventions for children. The insights gained provide valuable knowledge about tailored BCTs incorporated into eHealth interventions that promote children's PA self-efficacy. Abstract Gastric adenocarcinoma (STAD) is the most prevalent malignancy of the human digestive system and the fourth leading cause of cancer-related death. Calcium pools, especially Ca2+ entry (SOCE) for storage operations, play a crucial role in maintaining intracellular and extracellular calcium balance, influencing cell activity, and facilitating tumor progression. Nevertheless, the prognostic and immunological value of SOCE in STAD has not been systematically studied. The objective of this study was to develop a risk model for SOCE signature and to explore its correlation with clinical characteristics, prognosis, tumor microenvironment (TME), as well as response to immunotherapy, chemotherapy, and targeted drugs. We used the TCGA, GEO (GSE84437 and GSE159929), cBioPortal and TIMER databases to acquire mRNA expression data for STAD, along with patient clinical indicators, single-cell sequencing data, genomic variation information, and correlations of immune cell infiltration. An analysis of SOCE genes based on tumor vs. normal tissue comparisons, pan-cancer dimension, single-cell sequencing, DNA mutation, and copy number variation revealed that SOCE genes significantly impact the survival of STAD patients and are differentially involved in the immune response. SOCE co-expressed genes were identified by Pearson analysis, and subsequently protein-protein interaction (PPI) and gene function enrichment analysis indicated that coexpressed genes were associated with multicellular pathways. Based on TCGA and GSE84437 datasets, a multifactor Cox proportional hazard regression analysis was conducted to identify SOCE co-expressed genes associated with overall survival in STAD patients. Several mRNA prognostic genes, including PTPRJ, GPR146, LTBP3, FBLN1, EFEMP2, ADAMTS7 and LBH, were identified, which could be used as effective prognostic predictors for STAD patients. In both training and test datasets, receiver operating characteristic (ROC) curves were utilized to evaluate and illustrate the predictive capability of this characteristic in forecasting overall survival of STAD patients. The qPCR and human protein atlas (HPA) were employed to assess mRNA expression and protein levels. Furthermore, the ESTIMATE, TIMER, CIBERSORT, QUANTISEQ, MCPCOUNTER, xCell and EPIC algorithms were utilized to perform immune score and analyze immune cell infiltration. It effectively revealed the difference in prognosis and immune cell infiltration in TME between high-risk and low-risk groups based on the risk signature associated with SOCE. Notably, significant differences in tumor immune dysfunction and rejection (TIDE) scores between the two groups, suggesting that patients in the low-risk group may exhibit a more favorable response to ICIS treatment. The GDSC database and R packages for predictive analysis were utilized to analyze responses to chemotherapy and targeted drugs in high-risk and low-risk groups. In summary, the 7-gene signature associated with SOCE serves as a significant biomarker for evaluating the TME and predicting the prognosis of STAD patients. In addition, it may provide valuable insights for developing effective immunotherapy and chemotherapy regiments for patients with STAD.

Abstract Background The involvement of tetraspanins in cancer development has been widely implicated. In this study, the function and molecular mechanisms of tetraspanin 3 (TSPAN3) in non-small cell lung cancer (NSCLC) cells were explored. Methods Tissue samples from patients diagnosed with NSCLC were analyzed by immunohistochemistry, western blotting, and real-time polymerase chain reaction (PCR) to indicate the involvement of TSPAN3 in cancer progression. In the meantime, we also performed exhaustive mechanistic studies using A549 and H460 cells in vitro through a variety of methods including western blotting, real-time PCR, immunofluorescent staining, coimmunoprecipitation, cell proliferation assay, and nocodazole (NZ) washout assay. Proper statistical analysis was implemented wherever necessary in this study. Results TSPAN3 was found to be highly expressed in lung cancer cells and tissues. Moreover, high levels of TSPAN3 positively correlated with poor differentiation, lymph node involvement, advanced pathological tumor-node-metastasis stage, and poor prognosis in patients with NSCLC. TSPAN3 showed potential to promote the proliferation of NSCLC cells in vitro and in vivo. Specifically, TSPAN3 was found to interact with β1 integrin via the LEL domain, thereby facilitating the sorting of β1 integrin into Rab11a endosomes and promoting β1 integrin recycling and upregulation. Conclusions Our findings reveal TSPAN3 may represent a potentially valuable therapeutic target for NSCLC.

With accelerated urban sprawl, water scarcity and increased water stress have become global issues. Water of building industry and domestic water are two major contributors to total water consumption that cannot be ignored. This paper seeks to emphasize the effects of urban sprawl on water competition between building industry and residents. To achieve this objective, a framework for quantifying the water usage of the life cycle of the building was first constructed. Additionally, the degree of urban sprawl and water competition between domestic and building industry was computed. This study characterized the evolution path of urban sprawl and water competition using standard deviation ellipse. Spatial econometric models were used to determine the impact of urban sprawl on water competition. The results indicate that there is a high spatial correlation between urban sprawl and water competition. The center of gravity of China's urban sprawl is consistent with the north-to-south movement of water competition. As economic sprawl increases by 1%, water competition will rise by 0.440%. As the population expands by 1%, the water competition index shifts by 0.018%. Moreover, 0.017% more water was used for every 1% increase in land sprawl.

Lead-cooled fast reactors (LFRs) have a wide range of application scenarios, which require the thermal-hydraulic characteristics of LFRs to be reliable. In the present paper, the Lead-cooled fast reactor Thermal-Hydraulic Analysis Code LETHAC was developed, including the models of pipe, heat exchanger, and pool. To verify the correctness of LETHAC, two experimental facilities and three experimental cases were selected, including GFT and PLOFA tests for NACIE-UP and Test-1 for CIRCE. The calculated results show the same and consistent trend with the experimental data, but there are some discrepancies. It can be found that LETHAC is suitable and reliable in predicting the transient behavior of lead-cooled system.

BackgroundWe performed a network meta-analysis of phase III trials to compare the efficacy and safety of first-line regimens for patients with advanced esophageal squamous cell carcinoma (ESCC).MethodsA systematic review and Bayesian network meta-analysis were conducted by retrieving relevant literature from PubMed, Embase, the Cochrane Library, and the Web of Science. We included published sources of randomized clinical trials comparing immunotherapy combinations for treating advanced ESCC.ResultsWe analyzed seven studies involving eight immunotherapy combinations and 4688 patients. For patients without programmed death-ligand 1 (PD-L1) selection, it was found that the combination of toripalimab and chemotherapy provided better overall survival than chemotherapy alone (hazard ratio = 0.58, 95% confidence interval (CI) 0.43-0.78). Compared with chemotherapy alone, Sintilimab or camrelizumab plus chemotherapy seemed to achieve the best progression-free survival (hazard ratio = 0.56, 95% CI 0.46-0.68). Nivolumab plus chemotherapy appeared to provide the best objective response rate, with significant differences versus chemotherapy alone (odds ratio = 0.49, 95% CI 0.38-0.64). Nivolumab plus ipilimumab resulted in a relatively lower incidence of adverse events of grade ≥3 than other regimens.ConclusionsThe combination of immune checkpoint inhibitors (ICIs) with chemotherapy provided a high probability of more effective treatment in comparison with chemotherapy alone for patients with advanced ESCC. Toripalimab and sintilimab plus chemotherapy were ranked as providing the highest OS and PFS benefit in the first-line setting, respectively.

Abstract Damming profoundly affects downstream flow‐sediment regimes, altering channel bar dynamics and thereby affecting floods and riverine biodiversity. Here, we investigate the response of bars to upstream damming by examining patterns, mechanisms, and impacts in the Middle Yangtze River (∼1,000 km). Over a decade of post‐damming observational data reveal substantial increases in bar revegetation and Leaf Area Index. Shorter flood duration and stable bar size collectively drive bar greening. Consequently, denser vegetation has slowed flow velocity by 17% ± 2% and increased flow resistance by 21% ± 5%, offsetting the water‐level decrease from channel expansion due to scouring and even causing a slight rise in floodwater levels. Furthermore, damming has markedly altered river connectivity, thermal regimes, and solute dynamics, detrimentally affecting fish habitats and aquatic life. These findings, along with refined river stage simulation considering flow‐sediment‐vegetation interactions, facilitate sustainable reservoir operation and river management in big river systems.

Osteoarthritis (OA) commonly results in compromised mobility and disability, thereby imposing a significant burden on healthcare systems. Cartilage injury is a prevalent pathological manifestation in OA and constitutes a central focus for the development of treatment strategies. Despite the considerable number of studies aimed at delaying this degenerative process, their outcomes remain unvalidated in preclinical settings. Recently, therapeutic strategies focused on angiogenesis have attracted the growing interest from researchers. Thus, we conducted a comprehensive literature review to elucidate the current progress in research and pinpoint research gaps in this domain. Additionally, it provides theoretical guidance for future research endeavors and the development of treatment strategies.

Abstract Objectives The objective of this study was to evaluate the influence of marital status on overall survival (OS) and develop a nomogram for predicting 5-year OS in chondrosarcoma (CHS) patients. Methods We utilized the Surveillance, Epidemiology, and End Results (SEER) database to identify CHS patients diagnosed between 2010 and 2018. Survival rates were calculated using Kaplan–Meier analysis. Prognostic factors were identified through univariate and multivariate analyses. An independent cohort was used for external validation of the nomogram. Performance evaluation of the nomogram was conducted using Harrell's concordance index (C-index), calibration plot, and decision curve analysis (DCA). Objective Unilateral biportal endoscopic (UBE) surgery has recently been used as a minimally invasive procedure for the treatment of lumbar spinal stenosis and is associated with less perioperative blood loss. However, perioperative hidden blood loss (HBL) may be neglected during UBE. This study aimed to examine the volume of HBL and discuss the influential risk factors for HBL during unilateral biportal endoscopic surgery. Methods From January 2022 to August 2022, 51 patients underwent percutaneous unilateral biportal endoscopic surgery for lumbar spinal stenosis at the Department of Spinal Surgery of the Affiliated Traditional Chinese Medicine Hospital of Southwest Medical University and were enrolled in this study. The data included general indicators (age, sex and body mass index [BMI]), underlying disease (hypertension and diabetes), laboratory test results (prothrombin time [PT], activated partial thromboplastin time [APTT], fibrinogen [Fbg]), and preoperative and postoperative hematocrit and hemoglobin), related imaging parameters (severity of intervertebral disc [IVD] degeneration and soft tissue thickness of the interlaminar approach), number of operated vertebrae and operation time. Total blood loss (TBL) and HBL during surgical procedures were measured via the Gross formula. Influential factors were further analyzed by multivariate linear regression analysis and t‐tests. Results The mean HBL was 257.89 ± 190.66 mL for single‐operation patients and 296.58 ± 269.75 mL for two‐operation patients. Patients with lower PT (p = 0.044), deeper tissue thickness (p = 0.047), and diabetes mellitus were determined to have more HBL during UBE. The operation time might also be an important factor (p = 0.047). However, sex (p = 0.265), age (p = 0.771/0.624), BMI (p = 0.655/0.664), APTT (p = 0.545/0.751), degree of degenerated IVD (p = 0.932/0.477), and hypertension (p = 0.356/0.896) were not related to HBL. Conclusion This study determined the different influential factors of HBL during UBE. PT, tissue thickness, and diabetes mellitus are the independent risk factors that affect HBL incidence. Long PT may decrease the volume of HBL within a certain range. Tissue thickness and diabetes mellitus can lead to an increased volume of HBL.

Rheumatoid arthritis (RA) is a chronic inflammatory joint disease accompanied by energy depletion and accumulation of reactive oxygen species (ROS). Inorganic nanoparticles (NPs) offer great promise for the treatment of RA because they mostly have functions beyond being drug carriers. However, conventional nanomaterials become coated with a protein corona (PC) or lose their cargo prematurely in vivo, reducing their therapeutic efficacy. To avoid these problems, we loaded methotrexate (MTX) into hollow structured manganese dioxide nanoparticles (H-MnO2 NPs), then coated them with a ‘pseudo-corona’ of human serum albumin (HSA) at physiological concentrations to obtain HSA-MnO2@MTX NPs. Efficacy of MTX, MnO2@MTX, and HSA-MnO2@MTX NPs was compared in vitro and in vivo. Compared to MnO2@MTX, HSA-coated NPs were taken up better by lipopolysaccharide-activated RAW264.7 and were more effective at lowering levels of pro-inflammatory cytokines and preventing ROS accumulation. HSA-MnO2@MTX NPs were also more efficient at blocking the proliferation and migration of fibroblast-like synoviocytes from rats with collagen-induced arthritis. In this rat model, HSA-MnO2@MTX NPs showed better biodistribution than other treatments, specifically targeting the ankle joint. Furthermore, HSA-MnO2@MTX NPs reduced swelling in the paw, regulated pro-inflammatory cytokine production, and limited cartilage degradation and signs of inflammation. These results establish the therapeutic potential of HSA-MnO2@MTX NPs against RA.

BackgroundThe arteriosclerosis index, defined as the ratio of non-high density lipoprotein cholesterol to high density lipoprotein cholesterol (NHHR), has emerged as a novel biomarker for various diseases. The relationship between NHHR and lumbar bone mineral density (BMD) has not been previously examined.MethodsThis cross-sectional study analyzed data from the National Health and Nutrition Examination Survey (NHANES) 2011–2018. NHHR was calculated as (total cholesterol—high-density lipoprotein cholesterol)/high-density lipoprotein cholesterol. Lumbar BMD was calculated to Z scores. Weighted multivariate linear regression, subgroup analysis, interaction analysis, generalized additive model, and two-piecewise linear regression were used.ResultsA total of 8,602 participants were included. Hydrogen cyanide (HCN) is a toxic industrial chemical, necessitating low-level detection capabilities for safety and environmental monitoring. This study introduces a novel approach for detecting hydrogen cyanide (HCN) using a clamp-type custom quartz tuning fork (QTF) integrated with a dual-tube acoustic micro-resonator (AmR) for enhanced photoacoustic gas sensing. The design and optimization of the AmR geometry were guided by theoretical simulation and experimental validation, resulting in a robust on-beam QEPAS (Quartz-Enhanced Photoacoustic Spectroscopy) configuration. To boost the QEPAS sensitivity, an Erbium-Doped Fiber Amplifier (EDFA) was incorporated, amplifying the laser power by approximately 286 times. Additionally, a transformer-based U-shaped neural network, a machine learning filter, was employed to refine the photoacoustic signal and reduce background noise effectively. This combination yielded a significantly low detection limit for HCN at 0.89 parts per billion (ppb) with a rapid response time of 1 second, marking a substantial advancement in optical gas sensing technologies. Key modifications to the QTF and innovative use of AmR lengths were validated under various experimental conditions, affirming the system's capabilities for real-time, high-sensitivity environmental monitoring and industrial safety applications. This work not only demonstrates significant enhancements in QEPAS but also highlights the potential for further technological advancements in portable gas detection systems.

BackgroundDilated cardiomyopathy (DCM) is a heterogeneous myocardial disorder with diverse genetic or acquired origins. Notable advances have been achieved in discovering and understanding the genetics of DCM. This study aimed to depict the distribution of the main research forces, hotspots, and frontiers in the genetics of DCM, thus shaping future research directions.MethodsBased on the documents published in the Web of Science Core Collection database from 2013 to 2022, co-authorship of authors, institutions, and countries/regions, co-citation of references, and co-occurrence of keywords were conducted respectively to present the distribution of the leading research forces, research hotspots, and emerging trends in the genetics of DCM.Results4,141 documents were included, and the annual publications have steadily increased. Seidman, Christine E, Meder, Benjamin, Sinagra, Gianfranco were the most productive authors, German Centre for Cardiovascular Research was the most productive institution, and the USA, China, and Germany were the most prolific countries. The co-occurrence of keywords has generated 8 clusters, including DCM, lamin a/c, heart failure, sudden cardiac death, hypertrophic cardiomyopathy, cardiac hypertrophy, arrhythmogenic cardiomyopathy, and next-generation sequencing. Frequent keywords with average publication time after 2019 mainly included arrhythmogenic cardiomyopathy, whole-exome sequencing, RBM 20, phenotype, risk stratification, precision medicine, genotype, and machine learning.ConclusionThe research landscape of genetics in DCM is continuously evolving. Deciphering the genetic profiles by next-generation sequencing and illustrating pathogenic mechanisms of gene variants, establishing innovative treatments for heart failure and improved risk stratification for SCD, uncovering the genetic overlaps between DCM and other inherited cardiomyopathies, as well as identifying genotype-phenotype correlations are the main research hotspots and frontiers in this field.

BackgroundWith the increasing demand for joint replacement surgery in China, the government has successively issued the policies of national centralized procurement (NCP) and national volume-based procurement (NVBP) of artificial joints. The purpose of this study is to evaluate the impact of NCP and NVBP policies on hospitalization cost, rehospitalization and reoperation rate of total hip arthroplasty (THA).MethodsIn total, 347 patients who underwent THA from January 2019 to September 2022 were retrospectively analyzed. According to the implementation of NCP and NVBP, patients were divided into three groups: control group (n = 147), NCP group (n = 130), and NVBP group (n = 70). Patient-level data on the total hospitalization costs, rehospitalization rate, THA reoperation rate and inpatient component costs were collected before and after the implementation of the policies and Consumer Price Index was used to standardize the cost.ResultsAfter the implementation of NCP and NVBP, the total cost of hospitalization decreased by $817.41 and $3950.60 (p

Abstract Background Neuropathic pain (NP) is a kind of intractable pain. The pathogenesis of NP remains a complicated issue for pain management practitioners. SPARC/osteonectin, CWCV, and Kazal-like domains proteoglycan 2 (SPOCK2) are members of the SPOCK family that play a significant role in the development of the central nervous system. In this study, we investigated the role of SPOCK2 in the development of NP in a rat model of chronic constriction injury (CCI). Methods Sprague–Dawley rats were randomly grouped to establish CCI models. We examined the effects of SPOCK2 on pain hpersensitivity and spinal astrocyte activation after CCI-induced NP. Paw withdrawal threshold (PWT) and paw withdrawal latency (PWL) were used to reflects the pain behavioral degree. Molecular mechanisms involved in SPOCK2-mediated NP in vivo were examined by western blot analysis, immunofluorescence, immunohistochemistry, and co-immunoprecipitation. In addition, we examined the SPOCK2-mediated potential protein–protein interaction (PPI) in vitro coimmunoprecipitation (Co-IP) experiments. Results We founded the expression level of SPOCK2 in rat spinal cord was markedly increased after CCI-induced NP, while SPOCK2 downregulation could partially relieve pain caused by CCI. Our research showed that SPOCK2 expressed significantly increase in spinal astrocytes when CCI-induced NP. In addition, SPOCK2 could act as an upstream signaling molecule to regulate the activation of matrix metalloproteinase-2 (MMP-2), thus affecting astrocytic ERK1/2 activation and interleukin (IL)-1β production in the development of NP. Moreover, in vitro coimmunoprecipitation (Co-IP) experiments showed that SPOCK2 could interact with membrane-type 1 matrix metalloproteinase (MT1-MMP/MMP14) to regulate MMP-2 activation by the SPARC extracellular (SPARC_EC) domain. Conclusions Research shows that SPOCK2 can interact with MT1-MMP to regulate MMP-2 activation, thus affecting astrocytic ERK1/2 activation and IL-1β production to achieve positive promotion of NP.

Average and local convective heat transfer coefficients of nitrogen are measured experimentally in an electrically heated circular tube for a range of Reynolds number from 1.08 × 104 to 3.60 × 104, and wall-to-bulk temperature ratio from 1.01 to 1.77. The exit Mach number is up to 0.17, and the heat flux is up to 46 kW·m−2. The molybdenum test section has a 62 diameters heated section with an inside diameter of 5 mm and a 30 diameters entrance section to ensure the fully-developed flow. Uncertainty of Nusselt number is less than 1.6 % in this study. The results indicate that the average heat transfer correlations evaluated by both the bulk and the modified film Reynolds numbers agree well with the experimental data. The local heat transfer results based on bulk properties are compared with previous empirical correlations. New prediction correlations are recommended which are significantly affected by the property variation and heated length. The comparison between the proposed correlations and experimental points shows that 88 % of experimental data fall into an error of 10 %, and almost all data are within an error of 20 %.

Abstract Background The long-term nonunion of bone defects is always a difficult problem in orthopaedics treatment. Artificial bone implants made of polymeric materials are expected to solve this problem due to their suitable degradation rate and good biocompatibility. However, the lack of mechanical strength, low osteogenic induction ability and poor hydrophilicity of these synthetic polymeric materials limit their large-scale clinical application. Results In this study, we used bioactive glass (BG) (20%, W/W) and polycaprolactone (PCL, 80%, W/W) as raw materials to prepare a bone repair scaffold (PCL@BG20) using fused deposition modelling (FDM) three-dimensional (3D) printing technology. Subsequently, stromal cell-derived factor-1α (SDF-1α) chemokines were loaded into the PCL@BG20 scaffold pores with gelatine methacryloyl (GelMA) hydrogel. The experimental results showed that the prepared scaffold had a porous biomimetic structure mimicking that of cancellous bone, and the compressive strength (44.89 ± 3.45 MPa) of the scaffold was similar to that of cancellous bone. Transwell experiments showed that scaffolds loaded with SDF-1α could promote the recruitment of bone marrow stromal cells (BMSCs). In vivo data showed that treatment with scaffolds containing SDF-1α and BG (PCL@BG-GelMA/SDF-1α) had the best effect on bone defect repair compared to the other groups, with a large amount of new bone and mature collagen forming at the bone defect site. No significant organ toxicity or inflammatory reactions were observed in any of the experimental groups. Conclusions The results show that this kind of scaffold containing BG and SDF-1α serves the dual functions of recruiting stem cell migration in vivo and promoting bone repair in situ. We envision that this scaffold may become a new strategy for the clinical treatment of bone defects.

Abstract Background Osteoarthritis is a prevalent degenerative joint condition typically found in individuals who are aged 50 years or older. In this study, the focus is on PIWI-interacting RNA (piRNA), which belongs to a category of small non-coding RNAs. These piRNAs play a role in the regulation of gene expression and the preservation of genomic stability. The main objective of this research is to examine the expression of a specific piRNA called hsa_piR_019949 in individuals with osteoarthritis, to understand its impact on chondrocyte metabolism within this condition. Methods We analyzed piRNA expression in osteoarthritis cartilage using the GEO database. To understand the impact of inflammatory factors on piRNA expression in chondrocytes, we conducted RT-qPCR experiments. We also investigated the effect of piRNA hsa_piR_019949 on chondrocyte proliferation using CCK-8 and clone formation assays. Furthermore, we assessed the influence of piRNA hsa_piR_019949 on chondrocyte apoptosis by conducting flow cytometry analysis. Additionally, we examined the differences in cartilage matrix composition through safranine O staining and explored the downstream regulatory mechanisms of piRNA using transcriptome sequencing. Lentiviral transfection of NEAT1 and NLRP3 was performed to regulate the metabolism of chondrocytes. Results Using RNA sequencing technology, we compared the gene expression profiles of 5 patients with osteoarthritis to 3 normal controls. We found a gene called hsa_piR_019949 that showed differential expression between the two groups. Specifically, hsa_piR_019949 was downregulated in chondrocytes when stimulated by IL-1β, an inflammatory molecule. In further investigations, we discovered that overexpression of hsa_piR_019949 in vitro led to increased proliferation and synthesis of the extracellular matrix in chondrocytes, which are cells responsible for cartilage formation. Conversely, suppressing hsa_piR_019949 expression resulted in increased apoptosis (cell death) and degradation of the extracellular matrix in chondrocytes. Additionally, we found that the NOD-like receptor signaling pathway is linked to the low expression of hsa_piR_019949 in a specific chondrocyte cell line called C28/I2. Furthermore, we observed that hsa_piR_019949 can inhibit the expression of a long non-coding RNA called NEAT1 in chondrocytes. We hypothesize that NEAT1 may serve as a downstream target gene regulated by hsa_piR_019949, potentially influencing chondrocyte metabolism and function in the context of osteoarthritis. Conclusions PiRNA hsa_piR_019949 has shown potential in promoting the proliferation of chondrocytes and facilitating the synthesis of extracellular matrix in individuals with osteoarthritis. This is achieved by inhibiting the expression of a long non-coding RNA called NEAT1. The implication is that by using hsa_piR_019949 mimics, which are synthetic versions of the piRNA, as a therapeutic approach, it may be possible to effectively treat osteoarthritis.

Abstract Background To develop and validate a preoperative nomogram model combining the radiomics signature and clinical features for preoperative prediction of visceral pleural invasion (VPI) in lung nodules presenting as part‐solid density. Methods We retrospectively reviewed 156 patients with pathologically confirmed invasive lung adenocarcinomas after surgery from January 2016 to August 2019. The patients were split into training and validation sets by a ratio of 7:3. The radiomic features were extracted with the aid of FeAture Explorer Pro (FAE). A CT‐based radiomics model was constructed to predict the presence of VPI and internally validated. Multivariable regression analysis was conducted to construct a nomogram model, and the performance of the models were evaluated with the area under the receiver operating characteristic curve (AUC) and compared with each other. Results The enrolled patients were split into training (n = 109) and validation sets (n = 47). A total of 806 features were extracted and the selected 10 optimal features were used in the construction of the radiomics model among the 707 stable features. The AUC of the nomogram model was 0.888 (95% CI: 0.762–0.961), which was superior to the clinical model (0.787, 95% CI: 0.643–0.893; p = 0.049) and comparable to the radiomics model (0.879, 95% CI: 0.751–0.965; p > 0.05). The nomogram model achieved a sensitivity of 90.5% and a specificity of 76.9% in the validation dataset. Conclusions The nomogram model could be considered as a noninvasive method to predict VPI with either highly sensitive or highly specific diagnoses depending on clinical needs.

Shale hydration and swelling is the main obstacle to the development of shale gas utilizing water-based drilling fluids (WBDFs). In this work, the inhibition mechanism of alkylammonium inhibitor and alkylamine inhibitor adsorbed on sodium bentonite (Na+Bent) are investigated using infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), X-ray diffraction (XRD), zeta potential, particle size distribution tests, and thermogravimetry analysis (TGA). The results suggest that HTB and HMD can be inserted into the interlamination of Na+Bent and minimize the basal spacing compared to hydrated Na+Bent. HTB and HMD are inserted between the Na+Bent layers in a single-layer tiled manner and replace the sodium ions that are firmly fixed between the layers. Eventually, water molecules are removed from the interlayer Na+Bent. The interaction between the quaternary ammonium group and Na+Bent is more significant than between the primary amine group and Na+Bent. The inhibition performance suggests that HTB inhibits Na+Bent hydration and swelling more substantially than other inhibitors, indicating that the inhibition performance of the two quaternary ammonium groups is greater than that of the two primary amine groups. Therefore, HTB can be used as intercalation inhibition in WBDFs and has tremendous application value.

Abstract Breast cancer treatment has been a global puzzle, and apoptosis strategies based on mitochondrial Ca2+ overload have attracted extensive attention. However, various limitations of current Ca2+ nanogenerators make it difficult to maintain effective Ca2+ overload concentrations. Here, we constructed a multimodal Ca2+ nano-modulator that, for the first time, combined photothermal therapy (PTT) and mitochondrial Ca2+ overload strategies to inhibit tumor development. By crosslinking sodium alginate (SA) on the surface of calcium carbonate (CaCO3) nanoparticles encapsulating with Cur and ICG, we prepared a synergistic Ca2+ nano-regulator SA/Cur@CaCO3-ICG (SCCI). In vitro studies have shown that SCCI further enhanced photostability while preserving the optical properties of ICG. After uptake by tumor cells, SCCI can reduce mitochondrial membrane potential and down-regulate ATP production by producing large amounts of Ca2+ at low pH. Near-infrared light radiation (NIR) laser irradiation made the tumor cells heat up sharply, which not only accelerated the decomposition of CaCO3, but also produced large amounts of reactive oxygen species (ROS) followed by cell apoptosis. In vivo studies have revealed that the Ca2+ nano-regulators had excellent targeting, biocompatibility, and anti-tumor effects, which can significantly inhibit the proliferation of tumor cells and play a direct killing effect. These findings indicated that therapeutic strategies based on ionic interference and PTT had great therapeutic potential, providing new insights into antitumor therapy. Graphical Abstract

Abstract Background Both osteoporosis and cognitive impairment affect overall health in elderly individuals. This study aimed to investigate the association between cognitive impairment and the risk of osteoporosis. Methods PubMed, Web of Science, and the Cochrane Library were searched for studies on the association between osteoporosis and cognitive impairment from their inception until August 2023. The random-effects model was used to calculate the pooled risk ratio (RR) of osteoporosis in patients with cognitive impairment. Subgroup analysis was used to detect the sources of heterogeneity. Sensitivity analysis was used to test the robustness of the pooled results. Funnel plots, Egger’s test, and Begg’s test were used to test publication bias. Results Ten studies involving 9,872 patients were included in this meta-analysis. AbstractPrevious studies were mostly conducted based on two-dimensional feature space to monitor salinization, while studies on dense long-term salinization monitoring based on three-dimensional feature space have not been reported. Based on Landsat TM/ETM+/OLI images and three-dimensional feature space method, this study introduced six typical salinization surface parameters, including NDVI, salinity index, MSAVI, surface albedo, iron oxide index, wetness index to construct eight different feature space monitoring index. The optimal soil salinization monitoring index model was proposed base on field observed data and then the evolution process of salinization in Yellow River Delta (YRD) were analyzed and revealed during 1984–2022. The salinization monitoring index model of MSAVI-Albedo-IFe2O3 feature space had the highest accuracy with R2 = 0.93 and RMSE = 0.678g/kg. The spatial distribution of salinization in YRD showed an increasing trend from inland southwest to coastal northeast and the salinization intensity showed an increasing trend during 1984–2022 due to the implements of agricultural measures such as planting salt-tolerant crops, microbial remediation and fertility improvement. The rate of salinization deterioration in the northeast part was greater than others. Zones of salinization improvement were mainly located in cultivated land of the southwest parts.

Due to the complex intersection and control of multiple structural systems, the hydrogeological conditions of the Laiyuan Basin in China are complex. The depth of research on the relationship between geological structure and groundwater migration needs to be improved. The supply relationship of each aquifer is still uncertain. This paper systematically conducts research on the characteristics of hydrogen and oxygen isotopes, and combines magnetotelluric impedance tensor decomposition and two-dimensional fine inversion technology to carry out fine exploration of the strata and structures in the Laiyuan Basin, as well as comprehensive characteristics of groundwater migration and replenishment. The results indicate the following: (i) The hydrogen and oxygen values all fall near the local meteoric water line, indicating that precipitation is the main groundwater recharge source. (ii) The excess deuterium decreased gradually from karst mountain to basin, and karst water and pore water experienced different flow processes. (iii) The structure characteristics of three main runoff channels are described by MT fine processing and inversion techniques. Finally, it is concluded that limestone water moved from the recharge to the discharge area, mixed with the deep dolomite water along the fault under the control of fault F2, and eventually rose to the surface of the unconsolidated sediment blocked by fault F1 to emerge into an ascending spring.

In order to realize the optimal planning of grid–resource–storage for distribution networks (DNs) with high penetrated distributed photovoltaics (PVs), a cluster partition-based two-layer expansion planning for DNs is proposed. First, a comprehensive cluster partition index-based cluster partition method is proposed, which involves the indexes such as electrical distance, power balance of the cluster, and cluster size. Second, a cluster partition-based two-layer expansion planning model is proposed. In the upper layer, a line planning model for clusters is established to carry out the planning of cluster connection lines. In the lower layer, a robust source-storage planning model is established with the uncertainty of PVs and loads, and then, the optimal location and capacity of PVs and energy storages (ESs) can be obtained. In addition, the uncertainty regulation parameter is utilized to control the range of uncertainty sets, which can reduce the conservatism of the optimization. Finally, the proposed method is carried out in a real DN in China, which can effectively improve the economy of DN planning.

Many studies on horizontally loaded energy piles have tended to ignore the impact of axial frictional resistance in the analysis processes or lacked experimental validation. The effect of axial frictional resistance may be considered by conveniently calculating the frictional resistance and revealing the mechanism of axial frictional resistance. Our proposed displacement analysis approach takes into account pile-sand axial friction during heating. Validation was carried out through a centrifuge test case. Subsequent parametric analyses explored the influence of pile diameter and Young's modulus on the lateral response of the pile. Comparative results between the centrifuge test and proposed approach underscored its effectiveness in capturing the impact of friction resistance on energy piles during heating. The parametric analyses show that the increase in pile diameter from 0.5 m to 0.7 m and Young's modulus from 10 GPa to 20 GPa led to a decrease in normalized pile top displacement by 50.68% and 28.33%, a decrease in maximum soil pressure in front of the pile by 16.18% and 11.59%, and a decrease in maximum bending stress of the pile by 3.27% and an increase by 14.48%, respectively.

Abstract Background Osteosarcoma has the highest incidence among bone malignant tumors and mainly occurs in adolescents and the elderly, but the pathological mechanism is still unclear, which makes early diagnosis and treatment very difficult. Bone marrow mesenchymal stem cells (BMSCs) are considered to be one of the sources of osteosarcoma cells. Therefore, a full understanding of the gene expression differences between BMSCs and osteosarcoma cells is very important to explore the pathogenesis of osteosarcoma and facilitate the early diagnosis and treatment of osteosarcoma. Small noncoding RNAs (sncRNAs) are a class of RNAs that do not encode proteins but directly play biological functions at the RNA level. SncRNAs mainly include Piwi-interacting RNAs (piRNAs), small nucleolar RNAs (snoRNAs), small nuclear RNAs (snRNAs), repeat RNAs and microRNAs (miRNAs). Methods In this study, we compared the expression of sncRNAs in BMSCs and osteosarcoma cells by high-throughput sequencing and qPCR and looked for differentially expressed sncRNAs. CCK-8, clone formation and transwell assay were used to detect the effect of sncRNA in MG63 cells. Results We found that 66 piRNAs were significantly upregulated and 70 piRNAs were significantly downregulated in MG63 cells. As for snoRNAs, 71 snoRNAs were significantly upregulated and 117 snoRNAs were significantly downregulated in MG63 cells. As for snRNAs, 35 snRNAs were significantly upregulated and 17 snRNAs were significantly downregulated in MG63 cells. As for repeat RNAs, 6 repeat RNAs were significantly upregulated and 7 repeat RNAs were significantly downregulated in MG63 cells. As for miRNAs, 326 miRNAs were significantly upregulated and 281 miRNAs were significantly downregulated in MG63 cells. Overexpression of piRNA DQ596225, snoRNA ENST00000364830.2, snRNA ENST00000410533.1 and miRNA hsa-miR-369-5p inhibited the proliferation and migration of MG63 cells. Conclusions Our results provide a theoretical basis for the pathogenesis, early diagnosis and treatment of osteosarcoma.

Osteoarthritis (OA) causes disability and significant economic and social burden. Cartilage injury is one of the main pathological features of OA, and is often manifested by excessive chondrocyte death, inflammatory response, abnormal bone metabolism, imbalance of extracellular matrix (ECM) metabolism, and abnormal vascular or nerve growth. Regrettably, due to the avascular nature of cartilage, its capacity to repair is notably limited. Mesenchymal stem cells‐derived extracellular vesicles (MSCs‐EVs) play a pivotal role in intercellular communication, presenting promising potential not only as early diagnostic biomarkers in OA but also as efficacious therapeutic strategy. MSCs‐EVs were confirmed to play a therapeutic role in the pathological process of cartilage injury mentioned above. This paper comprehensively provides the functions and mechanisms of MSCs‐EVs in cartilage repair.

Abstract Transition metal-catalyzed carbonylative reaction with CO gas are among the central task in organic synthesis, enabling the construction of highly valuable carbonyl compound. Here, we show an earth-abundant nickel-catalyzed three-component tandem acylzincation/cyclization sequence of allene and alkylzinc reagent with 1 atm of CO under mild conditions. This protocol is featured by broad functional group tolerance with high reaction selectivity, providing a rapid and convenient synthetic method for the construction of diverse fully substituted benzotropone derivatives. Mechanistic studies reveal that the installation of a cyano group tethered to allene moiety enables the high regio- and stereoselectivity of this acylzincation of allene, allowing the selective formation of three consecutive C-C bonds in a highly efficient manner.

Abstract Small‐sample‐size problem is always a challenge for hyperspectral image (HSI) classification. Considering the co‐occurrence of land‐cover classes between similar scenes, transfer learning can be performed, and cross‐scene classification is deemed a feasible approach proposed in recent years. In cross‐scene classification, the source scene which possesses sufficient labelled samples is used for assisting the classification of the target scene that has a few labelled samples. In most situations, different HSI scenes are imaged by different sensors resulting in their various input feature dimensions (i.e. number of bands), hence heterogeneous transfer learning is desired. An end‐to‐end heterogeneous transfer learning algorithm namely domain‐invariant attention network (DIAN) is proposed to solve the cross‐scene classification problem. The DIAN mainly contains two modules. (1) A feature‐alignment CNN (FACNN) is applied to extract features from source and target scenes, respectively, aiming at projecting the heterogeneous features from two scenes into a shared low‐dimensional subspace. (2) A domain‐invariant attention block is developed to gain cross‐domain consistency with a specially designed class‐specific domain‐invariance loss, thus further eliminating the domain shift. The experiments on two different cross‐scene HSI datasets show that the proposed DIAN achieves satisfying classification results.

Small nucleolar RNAs (snoRNAs) play critical roles in various biological processes. The aberrant expression or depletion of snoRNAs is related to various diseases. In previous research, most of the snoRNAs were categorized as C/D box snoRNAs and H/ACA box snoRNAs, whose typical functions were thought of as regulation of 2′-O-ribose methylation and pseudouridylation of ribosome RNAs, respectively. However, in the past two decades, studies have revealed an increasing number of snoRNAs without specific targets or determined cell functions. These findings indicated that some potential roles of snoRNAs are still unknown. Numerous studies have indicated the correlation of snoRNAs with human diseases. SnoRNAs play various roles in abundant biological processes, and they have great potential in controlling human diseases. This new and rising field could benefit from investigations of the disease pathogenesis, biomarker identification, and the determination of novel therapeutic targets. This review summarized the reports on snoRNAs and the regulation of different diseases in recent years.

Abstract The development of atherosclerosis (AS) is closely linked to changes in the plaque microenvironment, which consists primarily of the cells that form plaque and the associated factors they secrete. The onset of inflammation, lipid deposition, and various pathological changes in cellular metabolism that accompany the plaque microenvironment will promote the development of AS. Numerous studies have shown that oxidative stress is an important condition that promotes AS. The accumulation of reactive oxygen species (ROS) is oxidative stress’s most important pathological change. In turn, the effects of ROS on the plaque microenvironment are complex and varied, and these effects are ultimately reflected in the promotion or inhibition of AS. This article reviews the effects of ROS on the microenvironment of atherosclerotic plaques and their impact on disease progression over the past five years and focuses on the progress of treatment strategies based on scavenging ROS of nanoparticles for AS. Finally, we also discuss the prospects and challenges of AS treatment.

Abstract Background Protocell refers to the basic unit of life and synthetic molecular assembly with cell structure and function. The protocells have great applications in the field of biomedical technology. Simulating the morphology and function of cells is the key to the preparation of protocells. However, some organic solvents used in the preparation process of protocells would damage the function of the bioactive substance. Perfluorocarbon, which has no toxic effect on bioactive substances, is an ideal solvent for protocell preparation. However, perfluorocarbon cannot be emulsified with water because of its inertia. Methods Spheroids can be formed in nature even without emulsification, since liquid can reshape the morphology of the solid phase through the scouring action, even if there is no stable interface between the two phases. Inspired by the formation of natural spheroids such as pebbles, we developed non-interfacial self-assembly (NISA) of microdroplets as a step toward synthetic protocells, in which the inert perfluorocarbon was utilized to reshape the hydrogel through the scouring action. Results The synthetic protocells were successfully obtained by using NISA-based protocell techniques, with the morphology very similar to native cells. Then we simulated the cell transcription process in the synthetic protocell and used the protocell as an mRNA carrier to transfect 293T cells. The results showed that protocells delivered mRNAs, and successfully expressed proteins in 293T cells. Further, we used the NISA method to fabricate an artificial cell by extracting and reassembling the membrane, proteins, and genomes of ovarian cancer cells. The results showed that the recombination of tumor cells was successfully achieved with similar morphology as tumor cells. In addition, the synthetic protocell prepared by the NISA method was used to reverse cancer chemoresistance by restoring cellular calcium homeostasis, which verified the application value of the synthetic protocell as a drug carrier. Conclusion This synthetic protocell fabricated by the NISA method simulates the occurrence and development process of primitive life, which has great potential application value in mRNA vaccine, cancer immunotherapy, and drug delivery. Transcription factors (TFs) are key players in the regulation of gene transcription in mammalian cells. Although high‐throughput screening can be used to identify differentially expressed genes between comparable groups, the precision of the corresponding datasets is far from optimal. Here, we establish Target Finder of Transcription Factor (TFoTF), a method for the prediction of TF‐targeted genes from genomic and cancer‐related transcriptomic data. TFoTF can identify potential TF‐targeted genes in large cancer datasets and efficiently estimate correlations between TFs and their targeted genes with a significant level of specificity, sensitivity, and precision. Overall, TFoTF is an easy‐to‐use tool that can be utilized to generate testable hypotheses in the context of cancer research projects.

Gas-cooled space reactor, which adopts He–Xe gas mixture as working fluid, is a better choice for megawatt power generation. In this paper, thermal-hydraulic characteristics of He–Xe gas mixture in 2 × 2 rod bundle wrapped with helical wires is numerically investigated. The velocity, pressure and temperature distribution of the coolant are obtained and analyzed. The results show that the existence of helical wires forms the vortexes and changes the velocity and temperature distribution. Hot spots are found at the contact corners between helical wires and fuel rods. The highest temperature of the hot spots reach 1600K, while the mainstream temperature is less than 400K. The helical wire structure increases the friction pressure drop by 20%–50%. The effect extent varies with the pitch and the number of helical wires. The helical wire structure leads to the reduction of Nusselt number. Comparing thermal-hydraulic performance ratios (THPR) of different structures, the THPR values are all less than 1. It means that gas-cooled space reactor adopting helical wires could not strengthen the core heat removal performance. This work provides the thermal-hydraulic design basis for He–Xe gas cooled space nuclear reactor.

Abstract Introduction The objective of this study was to investigate the ankle alignment alterations after the correction of knee varus deformity in MAKO robot-assisted total knee arthroplasty (MA-TKA). Methods A retrospective analysis was conducted for 108 patients with TKA from February 2021 to February 2022. Patients were divided into two groups based on MAKO robot involvement during the procedure: the MA-TKA group (n = 36) and the conventional manual total knee arthroplasty (CM-TKA) group (n = 72). The patients were divided into four subgroups according to the degree of surgical correction of the knee varus deformity. Seven radiological measurements were evaluated pre and post-surgery: mechanical tibiofemoral angle (mTFA), mechanical lateral distal femoral angle (mLDFA), medial proximal tibial angle (MPTA), lateral distal tibial angle (LDTA), tibial plafond inclination angle (TPIA), talar inclination angle (TIA), and tibiotalar tilt angle (TTTA). Multiple intersecting situational tasks in the field of aviation often cause air traffic controllers to face issues pertaining to interruption and task switching. To investigate the impact of task interruption on the situation awareness of air traffic controllers, two experiments were conducted. Experiment 1, which focused on 44 new graduates preparing to work in the field of air traffic control, revealed that task interruption reduced the participants' levels of situation awareness. Experiment 2, which focused on 80 new air traffic control graduates, employed a 2 (primary task modality: visual, auditory) × 2 (interruption modality: visual, auditory) between-subjects design and revealed that the negative effect of task interruption on situation awareness was moderated by task modality. Interruptions that occur in the same modality as the primary task were shown to result in greater decreases than were cross modal interruptions. Visual interruption led to a greater decrease in situation awareness than did auditory interruption, and interruption of the visual task also caused a greater decrease in situation awareness than did interruption of the auditory task. These findings might be valuable in attempts to enhance situation awareness among air traffic controllers by providing insights into the design of human‒computer interactions in the context of an air traffic control automation system.

Tumor-infiltrating immune cells (TICs) affect tumorigenesis and tumor development in head and neck squamous cell carcinoma (HNSCC). We constructed a novel predictive model for HNSCC based on immune-related genes (IRGs) from The Cancer Genome Atlas and the Immunology Database and Analysis Portal. After identifying the IRGs, a predictive model involving 13 IRGs with high stratification value of overall survival (OS) was constructed by multiple support vector machine recursive feature elimination and least absolute shrinkage and selection operator regression. We explored the relationship between the risk score (RS) and clinical characteristics. The nomogram showed high concordance and good agreement in OS. Four TICs affected the OS and were in agreement with the abundance analysis of the RS levels. Furthermore, the low-risk HNSCC group showed higher expression of PD-1, CTLA4, and TIGIT, while the high-risk group showed higher expression of EGFR. The high-risk HNSCC showed high sensitivity to eight drugs.

In order to reduce leakage flow, cavity tips are often used in high-pressure turbines, at the cost of higher thermal load compared to flat tips. To better control the leakage flow and heat transfer at the blade tip, a cavity-filling geometry is proposed in this paper, and the effects of the filling area are also investigated by numerical simulation. The results indicate that the new geometries maintain the advantages of cavity tips in terms of aerodynamic performance. With the cavity filling area increasing, the heat transfer of the new geometries shows a trend of initially increasing and then decreasing. When the front 15% of the blade tip chord length is filled, the average heat transfer of this configuration is reduced by 3.7% compared to the cavity blade tip, and the high heat transfer area is reduced by 23.3%, at the cost of more intensive hot spots. As the cavity-filling area increases from 5% to 15%, the size of the vortexes at the cavity reduces, resulting in a rise in the leakage. Similar behaviors of the tip heat transfer and leakage flow were also found when the turbine pressure ratios varied from 3 to 7, with the cavity-filling geometry.

AbstractPrevious studies on carbon storage simulation had ignored the difference of carbon intensity among various vegetation types inner the same land use. In this paper, The PLUS model was used to predict the land use change under multi-scenarios from 2030 to 2060, and the vegetation type data were supplemented by CA model to obtain the land cover-vegetation datasets from 2030-2060. Combined with the carbon density table of vegetation type, the future land use carbon storage during 2030-2060 under multi-scenarios in Beijing-Tianjin-Hebei region were analyzed. The main conclusions were as follows: (1) The spatial distribution of carbon storage in Beijing-Tianjin-Hebei region showed a pattern of ‘high in northeast-southwest and low in southeast-northwest’; (2) The carbon storage in Beijing-Tianjin-Hebei region during 1990-2020 showed a decreasing trend; (3) During 2030-2060, the carbon storage in Beijing-Tianjin-Hebei region showed a continuous decreasing trend in the absence of policy intervention, while that under the ecological protection and farmland protection scenarios showed an increasing trend; (4) Under different development scenarios, there were obvious significances of carbon storage in spatial distribution.

Background: Myocardial fibrosis (MF) is an advanced pathological manifestation of many cardiovascular diseases, which can induce heart failure and malignant arrhythmias. However, the current treatment of MF lacks specific drugs. Ginsenoside Re has anti-MF effect in rat, but its mechanism is still not clear. Therefore, we investigated the anti-MF effect of ginsenoside Re by constructing mouse acute myocardial infarction (AMI) model and AngⅡ induced cardiac fibroblasts (CFs) model. Methods: The anti-MF effect of miR-489 was investigated by transfection of miR-489 mimic and inhibitor in CFs. Effect of ginsenoside Re on MF and its related mechanisms were investigated by ultrasonographic, ELISA, histopathologic staining, transwell test, immunofluorescence, Western blot and qPCR in the mouse model of AMI and the AngⅡ-induced CFs model. Results: MiR-489 decreased the expression of α-SMA, collagenⅠ, collagen Ⅲ and myd88, and inhibited the phosphorylation of NF-κB p65 in normal CFs and CFs treated with AngⅡ. Ginsenoside Re could improve cardiac function, inhibit collagen deposition and CFs migration, promote the transcription of miR-489, and reduce the expression of myd88 and the phosphorylation of NF-κB p65. Conclusion: MiR-489 can effectively inhibit the pathological process of MF, and the mechanism is at least partly related to the regulation of myd88/NF-κB pathway. Ginsenoside Re can ameliorate AMI and AngⅡ induced MF, and the mechanism is at least partially related to the regulation of miR-489/myd88/NF-κB signaling pathway. Therefore, miR-489 may be a potential target of anti-MF and ginsenoside Re may be an effective drug for the treatment of MF.
