Abstract Due to their high reliability and cost‐efficiency, submarine pipelines are widely used in offshore oil and gas resource engineering. Due to the interaction of waves, currents, seabed, and pipeline structures, the soil around submarine pipelines is prone to local scour, severely affecting their operational safety. With the Yellow River Delta as the research area and based on the renormalized group (RNG) k‐ε turbulence model and Stokes fifth‐order wave theory, this study solves the Navier–Stokes (N–S) equation using the finite difference method. The volume of fluid (VOF) method is used to describe the fluid‐free surface, and a three‐dimensional numerical model of currents and waves–submarine pipeline–silty sandy seabed is established. The rationality of the numerical model is verified using a self‐built waveflow flume. On this basis, in this study, the local scour development and characteristics of submarine pipelines in the Yellow River Delta silty sandy seabed in the prototype environment are explored and the influence of the presence of pipelines on hydrodynamic features such as surrounding flow field, shear stress, and turbulence intensity is analyzed. The results indicate that (1) local scour around submarine pipelines can be divided into three stages: rapid scour, slow scour, and stable scour. The maximum scour depth occurs directly below the pipeline, and the shape of the scour pits is asymmetric. (2) As the water depth decreases and the pipeline suspension height increases, the scour becomes more intense. (3) When currents go through a pipeline, a clear stagnation point is formed in front of the pipeline, and the flow velocity is positively correlated with the depth of scour. This study can provide a valuable reference for the protection of submarine pipelines in this area.

Abstract Impaired fatty acid oxidation (FAO) is a prominent feature of metabolic remodeling observed in pathological myocardial hypertrophy. Hepatocyte nuclear factor 4alpha (HNF4α) is closely associated with FAO in both cellular processes and disease conditions. Pellino 1 (Peli1), an E3 ligase containing a RING-like domain, plays a crucial role in catalyzing polyubiquitination of various substrates. In this study, we aimed to investigate the involvement of HNF4α and its ubiquitination, facilitated by Peli1, in FAO during pressure overload-induced cardiac hypertrophy. Peli1 systemic knockout mice (Peli1KO) display improved myocardial hypertrophy and cardiac function following transverse aortic constriction (TAC). RNA-seq analysis revealed that changes in gene expression related to lipid metabolism caused by TAC were reversed in Peli1KO mice. Importantly, both HNF4α and its downstream genes involved in FAO showed a significant increase in Peli1KO mice. We further used the antagonist BI6015 to inhibit HNF4α and delivered rAAV9-HNF4α to elevate myocardial HNF4α level, and confirmed that HNF4α inhibits the development of cardiac hypertrophy after TAC and is essential for the enhancement of FAO mediated by Peli1 knockout. In vitro experiments using BODIPY incorporation and FAO stress assay demonstrated that HNF4α enhances FAO in cardiomyocytes stimulated with angiotension II (Ang II), while Peli1 suppresses the effect of HNF4α. Mechanistically, immunoprecipitation and mass spectrometry analyses confirmed that Peli1 binds to HNF4α via its RING-like domain and promotes HNF4α ubiquitination at residues K307 and K309. These findings shed light on the underlying mechanisms contributing to impaired FAO and offer valuable insights into a promising therapeutic strategy for addressing pathological cardiac hypertrophy.

Abstract Since 2019, SARS-CoV-2 has evolved rapidly and gained resistance to multiple therapeutics targeting the virus. Development of host-directed antivirals offers broad-spectrum intervention against different variants of concern. Host proteases, TMPRSS2 and CTSL/CTSB cleave the SARS-CoV-2 spike to play a crucial role in the two alternative pathways of viral entry and are characterized as promising pharmacological targets. Here, we identify compounds that show potent inhibition of these proteases and determine their complex structures with their respective targets. Furthermore, we show that applying inhibitors simultaneously that block both entry pathways has a synergistic antiviral effect. Notably, we devise a bispecific compound, 212-148, exhibiting the dual-inhibition ability of both TMPRSS2 and CTSL/CTSB, and demonstrate antiviral activity against various SARS-CoV-2 variants with different viral entry profiles. Our findings offer an alternative approach for the discovery of SARS-CoV-2 antivirals, as well as application for broad-spectrum treatment of viral pathogenic infections with similar entry pathways.

The root of Angelica sinensis is utilized in Traditional Chinese medicine to enhance blood replenishment and facilitate blood circulation. The early bolting and flowering (EBF) of A. sinensis, however, compromises the quality of the roots and restricts the yield of medicinal substances. The study was conducted to compare the transcriptomic and metabolomic profiles between EBF plants and normal plants of two cultivars of A. sinensis, followed by validation of the transcriptome results using qRT-PCR. There were 3677 DEGs in EBF plants compared to normal plants of cultivar 2 (Mingui No.2), and cultivar 4 (Mingui No.4) was 3354. The main differential metabolites in the EBF and normal plants were phenolic acids, flavonoids, lignans, and coumarins. The analysis of 5 EBF-related pathways revealed 28 genes exhibiting differential expression and 5 metabolites showing differential accumulation. The expression of the Lhcb5, Lhcb2, Lhcb6, Lhcb1, Lhca4, ATPG1, EGLC, CELB, AMY, glgA, CYCD3, SnRK2, PYL, AHK2, AUX1, BSK, FabI/K, ACACA and FabV decreased and the expression of the PsbR, PsbA, LHY, FT, CO, malQ, HK, GPI and DELLA increased in EBF plants. In addition, the Abscisic acid, d-Glucose-6P, α-d-Glucose-1P, NADP+, and ADP were more significantly enriched in EBF plants. The findings offer novel perspectives on the EBF mechanisms in A. sinensis and other medicinal plants of the Apiaceae family.

Abstract Scutellaria baicalensis has been one of the most commonly used traditional Chinese medicinal plants in China for more than 2000 years. The three new varieties cultivated could not be distinguished by morphology before flowering. It will hinder the promotion of later varieties. Chloroplast DNA has been widely used in species identification. Moreover, previous studies have shown that complete chloroplast genome sequences have been suggested as super barcodes for identifying plants. Therefore, we sequenced and annotated the complete chloroplast genomes of three cultivated varieties. The chloroplast genomes of SBW, SBR, and SBP were 151,702 bp, 151,799 bp, and 151,876 bp, which contained 85 protein-coding genes, 36 tRNA genes, and 8 rRNA genes. The analysis of the repeat sequences, codon usage, and comparison of chloroplast genomes shared a high degree of conservation. However, the sliding window results show significant differences among the three cultivated varieties in matK-rps16 and petA-psbJ. And we found that the matK-rps16 sequence can be used as a barcode for the identification of three varieties. In addition, the complete chloroplast genome contains more variations and can be used as a super-barcode to identify these three cultivated varieties. Based on the protein-coding genes, the phylogenetic tree demonstrated that SBP was more closely related to SBW, in the three cultivated varieties. Interestingly, we found that S. baicalensis and S. rehderiana are closely related, which provides new ideas for the development of S. baicalensis. The divergence time analysis showed that the three cultivated varieties diverged at about 0.10 Mya. Overall, this study showed that the complete chloroplast genome could be used as a super-barcode to identify three cultivated varieties of S. baicalensis and provide biological information, and it also contributes to bioprospecting.

Abstract Regulation of intron retention (IR), a form of alternative splicing, is a newly recognized checkpoint in gene expression. Since there are numerous abnormalities in gene expression in the prototypic autoimmune disease systemic lupus erythematosus (SLE), we sought to determine whether IR was intact in patients with this disease. We, therefore, studied global gene expression and IR patterns of lymphocytes in SLE patients. We analyzed RNA-seq data from peripheral blood T cell samples from 14 patients suffering from systemic lupus erythematosus (SLE) and 4 healthy controls and a second, independent data set of RNA-seq data from B cells from16 SLE patients and 4 healthy controls. We identified intron retention levels from 26,372 well annotated genes as well as differential gene expression and tested for differences between cases and controls using unbiased hierarchical clustering and principal component analysis. We followed with gene-disease enrichment analysis and gene-ontology enrichment analysis. Finally, we then tested for significant differences in intron retention between cases and controls both globally and with respect to specific genes. Overall decreased IR was found in T cells from one cohort and B cells from another cohort of patients with SLE and was associated with increased expression of numerous genes, including those encoding spliceosome components. Different introns within the same gene displayed both up- and down-regulated retention profiles indicating a complex regulatory mechanism. These results indicate that decreased IR in immune cells is characteristic of patients with active SLE and may contribute to the abnormal expression of specific genes in this autoimmune disease.

Biomass is a direct reflection of community productivity, and the allocation of aboveground and belowground biomass is a survival strategy formed by the long-term adaptation of plants to environmental changes. However, under global changes, the patterns of aboveground–belowground biomass allocations and their controlling factors in different types of grasslands are still unclear. Based on the biomass data of 182 grasslands, including 17 alpine meadows (AMs) and 21 desert steppes (DSs), this study investigates the spatial distribution of the belowground biomass allocation proportion (BGBP) in different types of grasslands and their main controlling factors. The research results show that the BGBP of AMs is significantly higher than that of DSs (p < 0.05). The BGBP of AMs significantly decreases with increasing mean annual temperature (MAT) and mean annual precipitation (MAP) (p < 0.05), while it significantly increases with increasing soil nitrogen content (N), soil phosphorus content (P), and soil pH (p < 0.05). The BGBP of DSs significantly decreases with increasing MAP (p < 0.05), while it significantly increases with increasing soil phosphorus content (P) and soil pH (p < 0.05). The random forest model indicates that soil pH is the most important factor affecting the BGBP of both AMs and DSs. Climate-related factors were identified as key drivers shaping the spatial distribution patterns of BGBP by exerting an influence on soil nutrient availability. Climate and soil factors exert influences not only on grassland biomass allocation directly, but also indirectly by impacting the availability of soil nutrients.

BackgroundProlonged fasting, characterized by restricting caloric intake for 24 h or more, has garnered attention as a nutritional approach to improve lifespan and support healthy aging. Previous research from our group showed that a single bout of 36-h water-only fasting in humans resulted in a distinct metabolomic signature in plasma and increased levels of bioactive metabolites, which improved macrophage function and lifespan in C. elegans.ObjectiveThis secondary outcome analysis aimed to investigate changes in the plasma lipidome associated with prolonged fasting and explore any potential links with markers of cardiometabolic health and aging.MethodWe conducted a controlled pilot study with 20 male and female participants (mean age, 27.5 ± 4.4 years; mean BMI, 24.3 ± 3.1 kg/m2) in four metabolic states: (1) overnight fasted (baseline), (2) 2-h postprandial fed state (fed), (3) 36-h fasted state (fasted), and (4) 2-h postprandial refed state 12 h after the 36-h fast (refed). In order to study the chemical constituents in the leaves of Champereia manillana var. longistaminea, the chemical composition was analyzed by the silica gel column chromatography (CC), thin-layer chromatography (TLC), dextran gel column chromatography (Sephadex LH-20), reversed-phase silica gel (RPC18) column chromatography and high performance liquid chromatography (HPLC). The ethyl acetate extraction part of the ethanol extraction was separated and six monomer compounds were obtained. The structures of these compounds were identified using nuclear magnetic resonance spectroscopy (NMR), high resolution mass spectrometry (HR-ESI-MS) and the data of the literature. Six compounds were elucidated as taraxerol (1), 1H-indole-3-carboxylic acid (2), (24R)-cycloartane-3β, 24, 25-triol (3), (24R, S)-3β-24,31-epoxy-24-methylcycloartane(4), 1-O-linolenoyl-3-O-β-D-galactospyranosyl-snglycerol (5), and hyloglyceride (6). All compounds 1-6 were isolated from this plant for the first time.

The worsening global environmental crisis highlights the urgency of integrating Environmental Education (EE) throughout the curriculum including in English as a Foreign Language (EFL) curricula. However, challenges exist, such as the development of appropriate curriculum documents and educators' understanding of approaches to EE. This study looked at Indonesian EFL textbooks for elementary schools, focusing mainly on the images and how they can portray environmental themes. These images and some brief texts that accompany the images were examined using tools from ecolinguistics literature, including Stibbe's (2021) nine stories and the United Nations' EE objectives (UNESCO-UNEP, 1975). This study intends to fill a gap in the EE literature by analyzing images in elementary school textbooks in the specific context of Indonesia, generalizing the idea of EE to other contexts. Findings revealed that while some EE efforts were evident, the majority of the material leaned towards an anthropocentric (human centered) perspective, indicating a need for greater incorporation of insights from ecolinguistics to enhance the content. Moreover, in terms of EE objectives, the textbooks provided ecological knowledge but lacked emphasis on developing skills and encouraging student participation in solving environmental problems. Examples demonstrating the ability to evaluate and address environmental issues were also absent. Suggestions are made for future EE content in EFL materials.

Magnesium alloys have great potential in biomedical applications due to their unique combination of satisfactory mechanical property and decent biodegradability. However, their poor corrosion resistance limits their applications in biomedical fields. In this work, we employ a chemical conversion deposition method to prepare a Zr-based conversion film on the surface of AZ31 magnesium alloy to serve as a passivation layer. The mechanism for the film formation was studied and it showed the deposition process consists of four steps: substrate dissolution, nucleation, film growth, and film equilibrium. The film is mainly composed of Zr(OH) _4 /ZrO and Mg(OH) _2 /MgO with small amount of MgF _2 and ZrF _4 . The protective performance of the Zr-based film was investigated by electrochemical and immersion tests in simulated body fluid (SBF). Electrochemical results showed a significant decrease in the corrosion current density ( I _corr ), a positive shift of corrosion potential ( E _corr ), a bigger capacitive loop diameter and higher impedance values for the Zr-coated substrate as compared with an uncoated one. Immersion results indicated the corrosion rate of the Zr-coated sample was ∼20% lower than that of an uncoated one. All above results corroborate the great potential of Zr-based coating in enabling AZ31 alloy for biomedical applications.

Optimizing the flood limit water level (FLWL) of reservoirs in sediment-laden rivers under changing water and sediment conditions is an important research topic that could improve comprehensive utilization benefits. Because reservoir operation has multiple objectives in sediment-laden rivers, this study established a water–sediment mathematical model, a comprehensive benefit evaluation model, and an evaluation index system. Taking the Xiaolangdi Reservoir of the Yellow River as an example, the operation mode of the FLWL under changing water and sediment conditions was studied. Under the scenarios of incoming sediment amounts of 300–800 million tons, when using the operation mode of gradually raising the FLWL, the sediment retention period was 4–13 years longer; the lower average annual siltation of the downstream channel and minimum bank-full discharge of the downstream channel after 50 years was larger by 150–260 m3/s than the operation mode of raising the FLWL at one time. However, with enhanced benefits of sediment blocking and siltation reduction, other benefits such as water resources supply, hydropower generation, and ecological improvement are reduced. The average annual number of days that do not meet the downstream water resources supply requirements, irrigation, and ecological improvement was increased by 0.64–2.16 days, and 91–197 million kW·h reduced average annual hydropower generation. The critical amount of incoming sediment was 350 million for conversion between the two FLWL operation modes, and it will increase to 450 million tons if the incoming runoff of the Yellow River increases by 20%. After constructing the Guxian Reservoir in the middle of the Yellow River, the critical amount of incoming sediment will increase to 600 million tons. This study is of great significance for improving the utilization efficiency of water resources and promoting the socio-economic development of river basins.

Geological disasters refer to adverse geological phenomena that occur under the influence of natural or human factors and cause damage to human life and property. Establishing prevention and control zones based on geological disaster risk assessment results in land planning and management is crucial for ensuring safe regional development. In recent years, there has been an increase in extreme rainfall events, so it is necessary to conduct effective geological hazard and risk assessments for different extreme rainfall conditions. Based on the first national geological disaster risk survey results, this paper uses the analytic hierarchy process (AHP) combined with the information method (IM) to construct four extreme rainfall conditions, namely, 10-year, 20-year, 50-year, and 100-year return periods. The susceptibility, hazard, vulnerability, and risk of geological disasters in the Laoshan District in eastern China are evaluated, and prevention and control zones are established based on the evaluation results. The results show that: (1) There are 121 collapse geological disasters in Laoshan District, generally at a low susceptibility level. (2) A positive correlation exists between extreme rainfall and hazards/risks. With the rainfall condition changing from a 10-year return period to a 100-year return period, the proportion of high-hazard zones increased from 20% to 41%, and high-risk zones increased from 31% to 51%, respectively. The Receiver operating characteristic (ROC) proved that the assessment accuracy was acceptable. (3) Key, sub-key, and general prevention zones have been established, and corresponding prevention and control suggestions have been proposed, providing a reference for geological disaster prevention and early warning in other regions.

To find the gene hypervariable regions of three varieties of Hansenia forbesii H. Boissieu and determine their phylogenetic relationship, the chloroplast (cp) genome of these three varieties were firstly sequencing by the Illumina hiseq platform. In this study, we assembled the complete cp genome sequences of Hansenia forbesii LQ (156,954 bp), H. forbesii QX (157,181 bp), H. forbesii WQ (156,975 bp). They all contained 84 protein-coding genes, 37 tRNAs, and 8 rRNAs. The hypervariable regions between three cp genomes were atpF-atpH, petD, and rps15-ycf1. Phylogenetic analysis showed that H. forbesii LQ and H. forbesii WQ were closely related, followed by H. forbesii QX. This study showed that the three varieties of H. forbesii could be identified by the complete cp genome and specific DNA barcode (trnC-GCA-petN) and provided a new idea for germplasm identification of similar cultivated varieties.

Despite its powerful influence on student learning, assessment feedback has received relatively less attention in translation education. The mainstream assessment practices in translation education have relied mainly on a static approach to translation competence. The consequences of a static approach include a partial representation of translation competence development and a deficit view of students and their learning. Alternatively, this paper argues for an ecological approach to contextualize assessment feedback in translation education. The ecological approach emphasizes the spatial and temporal context for translation assessment. While detailed contextual information is essential to the ecological assessment approach, assessing translation performance across tasks and time is a considerable challenge. In response to such a challenge, this conceptual paper proposes a corpus-assisted approach to translation assessment. It discusses how a longitudinal student translation corpus can be developed to assist ecological assessment feedback on translation performance. A project in progress based on a translation education program is reported as a case in point for illustrative purposes. The paper has suggested ways forward for future assessment feedback practice and research in translation education.

Abstract High-density lipoprotein (HDL) particles have multiple beneficial and cardioprotective roles, yet our understanding of their full structural and functional repertoire is limited due to challenges in separating HDL particles from contaminating plasma proteins and other lipid-carrying particles that overlap HDL in size and/or density. Here we describe a method for isolating HDL particles using a combination of sequential flotation density ultracentrifugation and fast protein liquid chromatography with a size exclusion column. Purity was visualized by polyacrylamide gel electrophoresis and verified by proteomics, while size and structural integrity were confirmed by transmission electron microscopy. This HDL isolation method can be used to isolate a high yield of purified HDL from a low starting plasma volume for functional analyses. This method also enables investigators to select their specific HDL fraction of interest: from the least inclusive but highest purity HDL fraction eluting in the middle of the HDL peak, to pooling all of the fractions to capture the breadth of HDL particles in the original plasma sample. We show that certain proteins such as lecithin cholesterol acyltransferase (LCAT), phospholipid transfer protein (PLTP), and clusterin (CLUS) are enriched in large HDL particles whereas proteins such as alpha-2HS-glycoprotein (A2HSG), alpha-1 antitrypsin (A1AT), and vitamin D binding protein (VDBP) are enriched or found exclusively in small HDL particles.

The interaction between soil and structure is a research hotspot in ocean engineering, and the shear performance of interfaces is an essential factor affecting the bearing capacity of offshore structures. Taking the Yellow River Underwater Delta as the research area, the Softening/Hardening damage model of the silt–steel interface and the determination method of model parameters are proposed based on the statistical damage theory. Through the interface monotonic shear test under the conditions of different normal stress, roughness and water content, the shear mechanical properties and volumetric deformation laws on the silt–steel interface are analyzed, and the damage model parameters are obtained. Finally, a FRIC subroutine for the damage model was developed based on ABAQUS. The research results indicate the following: (1) The interface between silt and steel exhibits two characteristics, softening/hardening and shear shrinkage/expansion, under different conditions. Roughness significantly impacts interfacial cohesion, while water content mainly affects the internal friction angle. (2) The softening model based on the classic rock damage model can better simulate the stress–strain relationship of the silt–steel interface under high normal stress and low water content. In contrast, the hardening model based on the classic hyperbola model can better simulate the stress–strain relationship under low normal stress and high water content. The calculated results of the softening/hardening model agree with the experimental results, and the model has 7 parameters. (3) The developed FRIC subroutine can effectively simulate the nonlinear mechanical behavior of the interface between silt and steel. The research results provide a reference for exploring the stability analysis of offshore structures considering interface weakening effects.

Maximizing the carbon sequestration of forested land is important for achieving carbon neutrality. Although some studies have discussed forest carbon sequestration efficiency (FCSE) from the perspective of total factor production, it is being increasingly recognized that forestland use regulates sequestration and emissions. When viewing forestland use as input and carbon emissions as output, there is a lack of empirical evidence on FCSE and its influencing factors. Here, a superefficiency slacks-based measurement model was applied to estimate FCSE for 66 counties in Zhejiang Province, China. The influencing factors and spatial spillover effects of FCSE were also analyzed using a spatial autocorrelation model. The findings showed that over the sample observation period, county FCSE ranged from 0.199 to 1.258, with considerable gaps. The global Moran’s I index showed that county-level FCSE was markedly spatially autocorrelated. Spatially, forestland use, cutting, pests, and diseases had negative spatial spillover effects on FCSE, whereas average annual temperature and precipitation displayed positive spillover effects. These findings suggest that the overall coordination of forest resource supervision and management among counties should be strengthened. The implementation of forestry management models aimed at consolidating or increasing forest carbon sequestration should be emphasized to improve forest quality, thereby promoting FCSE enhancement.

Classification-based forest management (CFM) is generally regarded as an important political means of achieving sustainable forest development. However, in the upsurge of publicly managed forest devolution, the impact of CFM policies on forestland restoration remains uncertain and needs to be explored. This study contributes to the scant literature on this topic in China, where CFM has long been implemented based on the ecological welfare forestland (EWF) certification program. We use provincial data from China to examine the relationship between EWF-certified areas and forest restoration. Based on inter-provincial panel data from the third to the ninth consecutive forest resource inventories in China (1984–2018), we use a dynamic spatial autoregressive model to analyze the impact of forest classification management on forest restoration. The results show that, contrary to appearances, increasing EWF-certified areas promotes forest restoration. However, after controlling for other possible influencing factors, increasing EWF-certified areas plays a minimal role in promoting forest restoration and regrowth by inhibiting investment in forest management and even has a negative impact on forest restoration in the southern collective forest area.

As a drug delivery system (DDS), traditional mesoporous silica nanoparticles (MSNs) suffer from bioaccumulation in vivo and premature drug release in systemic circulation due to low degradation rate and lack of protective gatekeeper. Herein, we developed a safe and intelligent DDS with characteristics of pH-responsive biodegradation and controlled drug release based on mesoporous silica composite nanoparticles (MSCNs) capped with ZnO quantum dots (ZnO QDs). Acidic degradable MSCNs were successfully synthesized by doping Ca2+ and PO43− into the MSNs’ framework. The in vitro doxorubicin hydrochloride (DOX) release was inhibited at neutral pH 7.4 but triggered significantly at pH 5.0 due to the dissociation of ZnO caps. The internalization behavior and cytotoxicity of 4T1 cells indicated MSCNs-ZnO could efficiently deliver DOX into the cells with significant antitumor activity. Such a DDS with pH-responsive biodegradation and controlled drug release has promising potential for cancer therapy.

Pine forests (Pinus tabulaeformis) have been in danger of defoliation by a caterpillar in the west Liaoning province of China for more than thirty years. This paper aims to assess and predict the degree of damage to pine forests by using remote sensing and ancillary data. Through regression analysis of the pine foliage remaining ratios of field plots with several vegetation indexes of Landsat data, a feasible inversion model was obtained to detect the degree of damage using the Normalized Difference Infrared Index of 5th band (NDII5). After comparing the inversion result of the degree of damage to the pine in 29 years and the historical damage record, quantized results of damage assessment in a long time-series were accurately obtained. Based on the correlation analysis between meteorological variables and the degree of damage from 1984 to 2015, the average degree of damage was predicted in temporal scale. By adding topographic and other variables, a linear prediction model in spatiotemporal scale was constructed. The spatiotemporal model was based on 5015 public pine points for 24 years and reached 0.6169 in the correlation coefficient. This paper provided a feasible and quantitative method in the spatiotemporal prediction of forest pest occurrence by remote sensing.
