Objective: To study the effect and mechanism of luteoloside on the differentiation and lipid metabolism of 3T3-L1 preadipocytes. Methods: The cytotoxicity of different concentrations of luteoloside on 3T3-L1 preadipocyte was detected using methyl thiazolyl tetrazolium (MTT) assay. Cell differentiation was induced by the cocktail method, and lipid droplets were observed by oil red O staining. The contents of triglyceride and total cholesterol in differentiated cells, as well as the secretion of related inflammatory factors such as tumor necrosis factor-α (TNF-α), interleukin (IL)-6, IL-10, leptin (LEP) and Adiponectin (ADPN) were measured. The relative expression levels of peroxisome proliferators activated receptor (PPAR) γ, CCAAT-enhancer-binding proteins (C/EBP)-α, sterol regulatory element-binding protein 1 (SREBP1), PPARα, uncoupling protein 1 (UCP-1), and carnitine palmitoyltransferase 1 (CPT-1) were analyzed by Western blotting. Results: The survival rate of 3T3-L1 preadipocytes decreased with the increase in luteoloside concentration, being above 80% at luteoloside concentrations of 5–60 μg/mL. Treatment with luteoloside significantly inhibited the differentiation of 3T3-L1 preadipocytes and reduced lipid accumulation. Luteoloside down-regulated the contents of TC and TG, the secretion of TNF-α, IL-6 and LEP, but up-regulated the secretion of IL-10 and ADPN. Luteoloside down-regulated the protein expression levels of PPARγ, C/EBP-α and SREBP1, but up-regulated those of PPARα, UCP-1 and CPT-1. Conclusion: Luteoloside can suppression the differentiation and lipid metabolism of 3T3-L1 preadipocytes. The mechanism may be that luteoloside down-regulates the protein expression of PPARγ, C/EBP-α, and SREBP1 to inhibit fat synthesis. Furthermore, it activates PPARα to upregulate downstream proteins and promote fat consumption. Luteoloside may also regulate the secretion of cytokines and promote cellular lipolysis, thus improving lipid metabolism imbalance.

Objective To focus on the efficacy at variable frequency stimulation (VFS) after deep brain stimulation (DBS) for the treatment of axial symptoms in Parkinson's disease (PD). Methods Clinical data of 12 patients with PD who underwent bilateral DBS followed by VFS more than one year from January 2018 to June 2022 at Tianjin Huanhu Hospital, Tianjin University were retrospectively analyzed. Axial symptoms were assessed using Unified Parkinson's Disease Rating Scale (UPDRS)-gait posture, Timed Up and Go Test (TUGT) time and the occurrence of freezing of gait (FOG), The Freezing of Gait - Questionnaire (FOG - Q), and Berg Balance Scale (BBS). The above indicators were recorded one week before DBS surgery and in the drug "off" period (baseline), at the end of high frequency stimulation (the end of HFS), at one month follow-up of VFS (short-term VFS), and at one year or more follow-up of VFS (long-term VFS). Results Compared with baseline, BBS scores increased at the end of HFS (t = - 2.845, P = 0.008); UPDRS-gait posture (t = 12.703, P = 0.000; t = 11.737, P = 0.000), TUGT time (t = 8.923, P = 0.000; t = 8.025, P = 0.000) and the occurrence of FOG (t = 4.448, P = 0.000; t = 4.181, P = 0.000), and FOGQ scores (t = 9.186, P = 0.000; t = 8.186, P = 0.000) decreased, while the BBS scores increased (t = - 7.686, P = 0.000; t = - 6.106, P = 0.000) in both short-term VFS and long-term VFS. Compared with the end of HFS, UPDRS-gait posture (t = 11.583, P = 0.000; t = 10.618, P = 0.000), TUGT time (t = 6.922, P = 0.000; t = 6.022, P = 0.000) and the occurrence of FOG (t = 3.612, P = 0.001; t = 3.344, P = 0.002), and FOG-Q scores (t = 7.915, P = 0.000; t = 6.915, P = 0.000) decreased, while the BBS scores increased (t = - 4.842, P = 0.000; t = - 3.262, P = 0.003) in short-term VFS and long-term VFS. Conclusions For PD patients with HFS but poor axial symptoms efficacy after DBS, the use of VFS significantly improves multifaceted axial symptoms with reliable long-term efficacy.

In order to develop and utilize Lentinus edodes protein, the effect of glutamine transaminase (TGase) cross-linking on the physicochemical and processing characteristics of L. edodes protein was studied. The results showed that the addition of 0%–20% TGase gradually reduced the contents of total thiol, free thiol and β-sheet of L. edodes protein, while the fluorescence intensity, particle size, β-turn content and surface hydrophobicity showed a trend of initially increasing and then decreasing. As the shear rate increased, the shear stress of the protein increased and the apparent viscosity decreased, indicating that TGase cross-linking promoted the generation of macromolecular polymers. In addition, the cross-linked protein exhibited a dense texture similar to that of the original protein, indicating the formation of a stable three-dimensional network structure. As the addition level of TGase increased from 4% to 12%, the protein network structure became increasingly dense. Moreover, an appropriate amount of TGase significantly improved the solubility, emulsifying properties, thermal stability, and oil-holding capacity of L. edodes protein. These results indicate that TGase cross-linking improves the physicochemical and processing properties of L. edodes protein to varying degrees.

Background Mild cognitive impairment (MCI) is an age-related chronic noncommunicable disease facing a certain caregiving burden. At present, most studies focus on the caregiving burden and social support of caregivers of patients with moderate-to-severe cognitive impairment, but there are few studies on the social support and related factors of caregivers of MCI patients. Objective To explore the social support and related factors of caregivers of MCI patients, and provide evidence for feasible social support for caregivers of MCI patients. Methods A total of 102 family caregivers of patients initially diagnosed as MCI in Weifang Community Health Service Center, Pudong New Area, Shanghai from April 2020 to August 2021 were recruited. A cognitive impairment management group was established to investigate basic information, social support rate score (SSRS), positive aspects of caregiver (PAC) score and Zarit Caregiver Burden Questionnaire (ZBI) score of the caregivers of patients with MCI. The related factors of social support for the participants were analyzed. Results The average score of social support of caregivers of MCI patients was (31.26±6.87), including the average score of objective support as (7.02±2.10), subjective support as (17.93±4.62) and support utilization as (6.31±1.96). Objective: Glucocorticoid-induced osteoporosis (GIOP) represents a major complication arising from the long-term use of glucocorticoids, which are widely prescribed for various inflammatory and autoimmune conditions. Despite its prevalence, the current therapeutic options for GIOP are limited in terms of efficacy, safety profiles, and patient compliance. The Modified Danggui Buxue Decoction (DGBXD), a traditional Chinese herbal formulation, has shown promise in preliminary studies for its potential osteoprotective effects. The present study aimed to explore the mechanistic underpinnings of DGBXD's action on GIOP using network pharmacology and molecular docking approaches, bridging traditional medicine with modern pharmacological insights. Method: Network pharmacology is applied to screen drug-active compounds and potential core target proteins for disease treatment and to explore the drugs’ therapeutic mechanisms. Result: Altogether, 78 DGBXD active compounds and 223 DGBXD-related, 146 component-disease common, and 2168 GIOP-associated target genes were obtained. The PPI network had 43 nodes and 462 edges, and a total of 10 core target genes, including TP53, JUN and MAPK3, were identified. The results of the GO enrichment analysis implied that DGBXD might participate in biological activities, including responses to oxidative stress and nutrient levels. The outcomes of the KEGG pathway enrichment analysis showed that DGBXD may treat GIOP through TNF, IL-17, and phosphatidylinositol 3-kinase (PI3K)-Akt signaling pathways. Based on to the molecular docking results, biologically active compounds (beta-carotene, formononetin, luteolin, and isorhamnetin) exhibited good binding to AKT1 and ESR1. Conclusion: DGBXD may aid in GIOP treatment by modulating multiple therapeutic targets and signaling pathways.

Abstract Background Patients with hematological malignancies received multiple hypodermic injections of recombinant human granulocyte colony-stimulating factor. Procedural pain is one of the most common iatrogenic causes of pain in patients with hematological malignancies. It is also identified as the most commonly occurring problem in clinical care in the Department of Hematology and Oncology at Shenzhen University General Hospital. However, providing immediate relief from pain induced by hypodermic injection of recombinant human granulocyte colony-stimulating factor remains a major challenge. This trial aims to evaluate the safety and analgesic efficacy of a fixed nitrous oxide/oxygen mixture for patients with hematological malignancies and experiencing procedural pain caused by hypodermic injection of recombinant human granulocyte colony-stimulating factor in the department. Methods The nitrous oxide/oxygen study is a single-center, randomized, double-blind, placebo-controlled trial involving patients with hematological malignancies who require hypodermic injections of recombinant human granulocyte colony-stimulating factor for treatment. This trial was conducted in the Hematology and Oncology Department of Shenzhen University General Hospital. A total of 54 eligible patients were randomly allocated to either the fixed nitrous oxide/oxygen mixture group (n = 36) or the oxygen group (n = 18). Neither the investigators nor the patients known about the randomization list and the nature of the gas mixture in each cylinder. Outcomes were monitored at the baseline (T0), immediately after hypodermic injection of recombinant human granulocyte colony-stimulating factor (T1), and 5 min after hypodermic injection of recombinant human granulocyte colony-stimulating factor (T2) for each group. The primary outcome measure was the score in the numerical rating scale corresponding to the highest level of pain experienced during hypodermic injection of recombinant human granulocyte colony-stimulating factor. Secondary outcomes included the fear of pain, anxiety score, four physiological parameters, adverse effects, total time of gas administration, satisfaction from both patients and nurses, and the acceptance of the patients. Discussion This study focused on the safety and analgesic efficacy during hypodermic injection of recombinant human granulocyte colony-stimulating factor procedure. Data on the feasibility and safety of nitrous oxide/oxygen therapy was provided if proven beneficial to patients with hematological malignancies during hypodermic injection of recombinant human granulocyte colony-stimulating factor and widely administered to patients with procedural pain in the department. Studying the adsorption behavior of Yangjiang granite to Sr2+ is very meaningful for the safety assessment of the low- and medium-level nuclear waste repository in Yangjiang. In this study, experimental samples were obtained from the granite rock formations near the low- and medium-level nuclear waste repository in Yangjiang. The static adsorption experiment was performed study the adsorption behavior of Yangjiang surrounding rocks granite on Sr2+. And the effect of contact time, initial concentration, reaction temperature, pH, as well as cation and anion on the adsorption behavior of granite was studied in detail. The physical and chemical compositions of Yangjiang granite were investigated by X-ray diffraction spectra(XRD), X-ray fluorescence spectrometer(XRF), scanning electron microscopy-energy dispersive spectroscopy(SEM-EDS), Fourier transform infrared spectra(FTIR) and Zeta potential analyzer. The characterization studies show the granite surface becomes rough after adsorption. The results of static adsorption experiments indicate that adsorption equilibrium time is about 5 days. The Sr2+ adsorption decreases with the increase of Sr2+ concentration which conforms to the Langmuir isotherm adsorption model and belongs to single-layer adsorption. Under neutral and higher temperature conditions, the adsorption effect is better. The fitting thermodynamic equation suggests that adsorption is a spontaneous endothermic reaction. The ion in groundwater such as Ca2+, Mg2+, Fe3+, NO3- all can inhibit the adsorption of Sr2+ by Yangjiang granite, while CO32- can interfere with adsorption by forming polymers with Sr2+. The results of this study can provide a certain reference for the safety evaluation of the low- and medium-level nuclear waste repository in Yangjiang.
