The radio-isotope rhenium-labeled N-[2-(triphenylmethyl)thioethyl]-3-aza-19-ethyloxycarbonyl-3-[2-(triphenylmethyl)thioethyl] octadecanoate) ligand (188Re-MN-16ET) is a novel therapeutic agent under preclinical evaluation for hepatoma. A reversed-phase high performance liquid chromatography coupled with a tandem mass spectrometric analysis method and diode array detector (DAD) involving a T type splitter was developed to characterize this pharmaceutical in rat liver tissue solution and determine its biotransformation rate. The separation was accomplished on a C18 column (chromolith silica, 4.6 mm × 100 mm) using an acetonitrile–ammonium acetate buffer gradient as the mobile phase. The detection was achieved by DAD set at 250 nm and tandem mass spectrometry using electrospray ionization in the positive ion mode. Re-MN-16ET displayed a retention time of 23.2 min and a transition ion pair corresponding to m/z 677 → 631 for multiple reaction monitoring. Its biotransformation reaction in rat liver homogenate proceeded for 90 min in a 37°C water bath. The characterization was conducted using aliquots that were extracted and concentrated from the reaction mixture for various incubation times. Re-MN-16ET exhibited a biotransformation half-life ( t1/2) of 8–9 min in liver tissue solution and was almost completely exhausted after 90 min. Two of its metabolites, consisting of the Re-labeled carboxylic acid derivative, predominately, and its corresponding demetallized disulfide ligand were found in the liver homogenate, providing a metabolism pathway for the radiopharmaceutical.

We report here the preparation and mechanistic explanation of a nitrogen rich platform, poly(melamine), and its application for efficient synthesis of carbon nanotube–metal nanoparticle hybrid. Poly(melamine) is electrochemically formed on the surface of a preanodized screen-printed carbon electrode (designated as SPCE*) under a suitable potential (1.5 V vs. Ag/AgCl) and pH (0.1 M HCl). Note that radical cation formation is the rate-determining step in melamine polymerization and an enough positive potential in highly acidic medium is essential for the formation and stabilization of radical cation. The XPS analysis indicates the generation of polymer by head-to-head coupling with NH NH bonding between melamine molecules. We demonstrate that this poly(melamine)-modified electrode is helpful in loading copper nanoparticles on multiwalled carbon nanotube for hybrid formation. Since the proposed method is easy for mass production, this report opens a new methodology to functionalize the carbon surface to prepare new materials in different fields.

BACKGROUND: Zeranol (Z) is a semi-synthetic mycotoxin that is used in some countries as a growth-promoting agent in livestock. In view of the known oestrogenic actions by Z and certain Z analogues, significant concerns exist with regard to the presence of Z residues in human foods and the potential for untoward effects, including carcinogenicity within the reproductive system. In order to confirm that foods are free from harmful Z residues, regulators need a quick and reliable analytical method that can be used for routine confirmation of Z-positive samples identified by enzyme-linked immunosorbent assay (ELISA) screening. In this study the authors have developed and validated a simple and rapid high-performance liquid chromatography method incorporating ultraviolet (UV) absorbance (wavelength 274 nm) and electrochemical (EC) dual-mode detection for simultaneous determination of Z-related mycotoxins produced from mouldy grain matrices, including rice, soybean and corn flakes. RESULTS: Recoveries for all analytes were around 80% and the limits of detection ranged from 10 to 25 ng mL−1 for UV and from 50 to 90 ng mL−1 for EC detection with good accuracy and reproducibility. Differential profiles and occurrence rates of Z, β-zearalenol, β-zearalanol and α-zearalenol in naturally moulded grain matrices were observed, indicating different metabolite patterns and possibly grain-specific effects of mycotoxin exposure for humans and animals. The strength of this dual detection method lies in its selectivity characterised by a carbon screen-printed electrode such that aflatoxin interference is precluded. CONCLUSION: The combined dual detection technique affords quick and reliable semi-confirmative and quantitative information on multiple types of Z analogues in mouldy grains without the necessity of using expensive mass spectrometry. Outbreaks of food-associated renal failure in pets occurred in Asia and the United States of America in 2004 and 2007. They were related to the combined intoxication of cyanuric acid and melamine. Our aims were to investigate cyanuric acid and melamine contamination of pet food and to examine subchronic toxicity in rats. Levels of 10%, 20%, 50%, and 50%–100% (w/w) of contaminated pet food were fed to rats for three months. Analytical results revealed that the tainted food contained significant levels of cyanuric acid and melamine in a ratio of 1:6.8. Rats fed the diet of 50%–100% for three months exhibited elevated serum blood urea nitrogen and creatinine, as well as dose-dependent melamine/cyanuric acid crystal-induced nephrotoxicity. The melamine/cyanuric acid crystals of various sizes were mixed with necrotic cell debris and inflammatory cells, accompanied by tubular dilation and interstitial fibrosis. The immunohistochemistry index of proliferative cellular nuclear antigen and osteopontin in the kidney of the 50%–100% group were elevated, indicating regeneration of renal cells and the formation of crystals. In conclusion, the combination ratio of cyanuric acid to melamine and the acidic urine content were two factors that, upon repeated exposure, determined the severity of the nephrotoxicity.

We report here a simple and easy electrochemical approach for sensitive detection of non-electroactive melamine using a disposable screen printed carbon electrode (SPCE) with uric acid as the recognition element. It is based on the competitive adsorptive behavior of melamine at the preanodized SPCE causing suppression in the oxidation current of uric acid. A linear range up to 126 ppb with a detection limit of 1.6 ppb (S/N=3) is achieved at the preanodized SPCE by differential pulse voltammetry. The electrochemical method is successfully applied to detect the melamine content in tainted milk powder and dog food.

A simple electrochemical approach, which does not require any expensive and complex instruments, is established for the selective and quantitative recognition of melamine in diary products and pet foods. During a preconcentration step (at 1.8 V versus Ag/AgCl), the formation of a polymer film from melamine on a preanodized screen-printed carbon electrode was identified by SEM and XPS. The as-formed polymer was found to be electroactive with a reversible redox peak, and hence square-wave voltammetry was applied to further increase the detection sensitivity to meet the detection limit for application in real sample analysis. Simply with a medium exchange procedure, melamine was selectively detected with a detection limit (S/N=3) of 0.8 μM (i.e., 98.3 ppb) by square-wave voltammetry. Lower than 1 ppm of melamine in real samples can be easily detected with good recoveries of 98.7-100.9% by the proposed approach. The recovery tests established for external calibration and standard addition techniques verified that the analysis can be done in a single-run measurement.
