17. Intraspecific trait variation of carrion beetle species and communities across elevations.

- Abstract
Filtering processes across environmental gradients can structure patterns of trait variation within communities. The community‐weighted mean (CWM) is a metric that is commonly used to indicate the directionality of such filtering processes and the optimal adaptive strategy of taxa within community. Proximity to the CWM indicates higher fitness, and deviations from this optimal value result in changes in the relative abundances of coexisting species.We investigated patterns of intraspecific trait variation in four coexisting carrion beetle species (Coleoptera: Silphidae) across elevational gradients. The study was conducted in temperate forest ecosystems with distinct natural vegetation zones ranging from 950 m to 1700 m above sea level.Of the 12 traits measured, we found that intraspecific variation ranged from 34% (body length) to 100% (ratio of elytra length, head length and head width to body length) and accounted for a larger proportion of variation than interspecific variation in 7 traits. For most traits, trait range, which indicates the niche breadth of species at a given elevation, was positively correlated with relative abundance. The CWMs of traits associated with long‐distance dispersal decreased with elevation, whereas those associated with microhabitat use showed the opposite trend. Soil temperature influenced tibia length after controlling for the effects of species identity, but soil water content had no effect on trait variation. Patterns of variation in body width and thorax width of two beetle species supported the CWM‐optimality hypothesis; however, patterns of trait variation in body width and thorax length of another species showed an opposite trend.Our study identifies several traits that can be highly variable within carrion beetle species. Such high levels of intraspecific trait variation may enable populations to adapt across a wide range of elevations and vegetation types. [ABSTRACT FROM AUTHOR]

Long noncoding RNAs (lncRNAs) are commonly dysregulated in tumors, but only a handful are known to play pathophysiological roles in cancer. We inferred lncRNAs that dysregulate cancer pathways, oncogenes, and tumor suppressors (cancer genes) by modeling their effects on the activity of transcription factors, RNA-binding proteins, and microRNAs in 5,185 TCGA tumors and 1,019 ENCODE assays. Our predictions included hundreds of candidate onco- and tumor-suppressor lncRNAs (cancer lncRNAs) whose somatic alterations account for the dysregulation of dozens of cancer genes and pathways in each of 14 tumor contexts. To demonstrate proof of concept, we showed that perturbations targeting OIP5-AS1 (an inferred tumor suppressor) and TUG1 and WT1-AS (inferred onco-lncRNAs) dysregulated cancer genes and altered proliferation of breast and gynecologic cancer cells. Our analysis indicates that, although most lncRNAs are dysregulated in a tumor-specific manner, some, including OIP5-AS1, TUG1, NEAT1, MEG3, and TSIX, synergistically dysregulate cancer pathways in multiple tumor contexts.

This integrated, multiplatform PanCancer Atlas study co-mapped and identified distinguishing molecular features of squamous cell carcinomas (SCCs) from five sites associated with smoking and/or human papillomavirus (HPV). SCCs harbor 3q, 5p, and other recurrent chromosomal copy-number alterations (CNAs), DNA mutations, and/or aberrant methylation of genes and microRNAs, which are correlated with the expression of multi-gene programs linked to squamous cell stemness, epithelial-to-mesenchymal differentiation, growth, genomic integrity, oxidative damage, death, and inflammation. Low-CNA SCCs tended to be HPV(+) and display hypermethylation with repression of TET1 demethylase and FANCF, previously linked to predisposition to SCC, or harbor mutations affecting CASP8, RAS-MAPK pathways, chromatin modifiers, and immunoregulatory molecules. We uncovered hypomethylation of the alternative promoter that drives expression of the ΔNp63 oncogene and embedded miR944. Co-expression of immune checkpoint, T-regulatory, and Myeloid suppressor cells signatures may explain reduced efficacy of immune therapy. These findings support possibilities for molecular classification and therapeutic approaches.

We analyzed molecular data on 2,579 tumors from The Cancer Genome Atlas (TCGA) of four gynecological types plus breast. Our aims were to identify shared and unique molecular features, clinically significant subtypes, and potential therapeutic targets. We found 61 somatic copy-number alterations (SCNAs) and 46 significantly mutated genes (SMGs). Eleven SCNAs and 11 SMGs had not been identified in previous TCGA studies of the individual tumor types. We found functionally significant estrogen receptor-regulated long non-coding RNAs (lncRNAs) and gene/lncRNA interaction networks. Pathway analysis identified subtypes with high leukocyte infiltration, raising potential implications for immunotherapy. Using 16 key molecular features, we identified five prognostic subtypes and developed a decision tree that classified patients into the subtypes based on just six features that are assessable in clinical laboratories.

- Abstract
Springtails (Collembola) inhabit soils from the Arctic to the Antarctic and comprise an estimated ~32% of all terrestrial arthropods on Earth. Here, we present a global, spatially-explicit database on springtail communities that includes 249,912 occurrences from 44,999 samples and 2,990 sites. These data are mainly raw sample-level records at the species level collected predominantly from private archives of the authors that were quality-controlled and taxonomically-standardised. Despite covering all continents, most of the sample-level data come from the European continent (82.5% of all samples) and represent four habitats: woodlands (57.4%), grasslands (14.0%), agrosystems (13.7%) and scrublands (9.0%). We included sampling by soil layers, and across seasons and years, representing temporal and spatial within-site variation in springtail communities. We also provided data use and sharing guidelines and R code to facilitate the use of the database by other researchers. This data paper describes a static version of the database at the publication date, but the database will be further expanded to include underrepresented regions and linked with trait data.

In this paper, we propose a novel tactile servoing based pressure distribution control scheme of a manipulator using a convolutional neural network (CNN). The CNN significantly improves the performance of the tactile servoing scheme compared to the one based on the tactile Jacobian. LeNet-5, originally proposed for image classification problems, is applied to represent a nonlinear relationship between current and desired pressure distributions and the robot velocity command by using mean squared error as the loss function. In the proposed control scheme, the trained CNN directly generates the velocity command of the manipulator so that the pressure distribution converges to a given desired pressure distribution. Validation experiments are carried out to evaluate the performance of the proposed control scheme. Experimental results show that the proposed tactile servoing control scheme has better performance than the Jacobian-based tactile servoing control scheme.

Forest soils are a critical component of terrestrial ecosystems and host a large number of animal decomposer species. One diverse and abundant decomposer taxon is oribatid mites (Acari: Oribatida), whose species composition varies with forest type and tree species composition. We used functional traits that indicate different niche dimensions, to infer assembly processes of oribatid mite communities in monocultures and mixed forests of native and introduced tree species. We found that coexisting species differed more in the resource‐related niche dimension, i.e., reproductive mode and trophic guild, than in the morphological dimension, e.g., body length and width, sclerotization and concealability. These results suggest that both filtering and partitioning processes structure oribatid mite communities. In native European beech forests, but not in non‐native Douglas fir forests, oribatid mites were mainly structured by filtering processes acting via traits related both to environmental tolerance and to resources. Furthermore, oribatid mite trait diversity, but not phylogenetic diversity, differed significantly between monocultures and mixed forests, demonstrating that multidimensional diversity indices provide additional information on soil biodiversity. Overall, the study provides evidence that traits representing different niche dimensions need to be considered for understanding assembly processes in soil animal communities and thereby soil biodiversity. 43. Impact of Michelia champaca and Tectona grandis mono-species and their mixed plantation on chemical soil properties in a tropical semi-evergreen forest.

Understanding the effects of planted forests on soil properties is of great importance to meet increasing demands for wood production and contributing to forest ecosystem services and soil carbon sequestration. However, the influence of mono- and mixed tree plantations on soil chemical properties remains incompletely explained, particularly in tropical semi-evergreen forests. In this study, a monoplantation of Michelia champaca and Tectona grandis, as well as their mixed plantation, were studied on soil variables (pH, SOC, N, P, K) at different depths (0-10, 10-20, and 20-30 cm) in tropical semi-evergreen forest of the Khadimnagar National Park, Bangladesh. All examined pedovariables showed no significant differences among plantation types, notably in the surface soil layer (up to 10 cm). However, significant oscillations were found in the deepest soil layer (20- 30 cm), where M. champaca and mixed-species plantations obtained the lowest SOC (0.55-0.66%) and N (0.05-0.06%) concentrations. In the intermediate soil layer (10-20 cm) K concentrations were the lowest in M. champaca (19.6 mg/kg) and mixed-species plantation (17.6 mg/kg), while T. grandis recorded the highest K concentration (27.4 mg/kg). Additionally, the results indicated an increase in pH with soil depth (from 5.30 in the topsoil to 7.00 in the deepest layer), with T. grandis showing the lowest values across examined soil layers (5.76-5.95) and significantly differing from M. champaca, which exhibited the highest pH over the soil profile (6.29-6.51). Such results suggest the leaching of basic cations (Ca
2+ , Mg2+ , K+ ) over the soil profile due to pronounced monsoon rainfalls. - Abstract
Soil life supports the functioning and biodiversity of terrestrial ecosystems. Springtails (Collembola) are among the most abundant soil arthropods regulating soil fertility and flow of energy through above- and belowground food webs. However, the global distribution of springtail diversity and density, and how these relate to energy fluxes remains unknown. Here, using a global dataset representing 2470 sites, we estimate the total soil springtail biomass at 27.5 megatons carbon, which is threefold higher than wild terrestrial vertebrates, and record peak densities up to 2 million individuals per square meter in the tundra. Despite a 20-fold biomass difference between the tundra and the tropics, springtail energy use (community metabolism) remains similar across the latitudinal gradient, owing to the changes in temperature with latitude. Neither springtail density nor community metabolism is predicted by local species richness, which is high in the tropics, but comparably high in some temperate forests and even tundra. Changes in springtail activity may emerge from latitudinal gradients in temperature, predation and resource limitation in soil communities. Contrasting relationships of biomass, diversity and activity of springtail communities with temperature suggest that climate warming will alter fundamental soil biodiversity metrics in different directions, potentially restructuring terrestrial food webs and affecting soil functioning.
