Abstract

This article optimizes the inference performance of the Qwen-1.8B model by performing Int8 quantization, vectorizing some operators in llama.cpp, and modifying the compilation script to improve the compiler optimization level. On the Yitian 710 experimental platform, the prefill performance is increased by 1.6 times, the decoding performance is increased by 24 times, the memory usage is reduced to 1/5 of the original, and the accuracy loss is almost negligible.

14. Luminescence-activated Pt(IV) prodrug for in situtriggerable cancer therapy

Abstract

Anticancer platinum prodrugs that can be controllably activated are highly desired for personalized precision medicine and patient compliance in cancer therapy. However, the clinical application of platinum(IV) prodrugs (Pt(IV)) is restricted by tissue penetration of external irradiation. Here, we report a novel Pt(IV) activation strategy based on endogenous luminescence of tumor microenvironment responsiveness, which completely circumvents the limitation of external irradiation. The designed Pt(IV)-Lu, a mixture of trans, trans, trans-[Pt(N3)2(OH)2(py)2] and luminol (Lu), has controllable activation property: it remains inert in reductant environment and normal tissues, but under tumor microenvironment, Lu will be oxidized to produce blue luminescence, which rapidly reduce Pt(IV) to Pt(II) without the need of any external activator. Pt(IV)-Lu shows excellent responsive antitumor ability both in vitroand in vivo. Compared to cisplatin, the median lethal dose in BALB/c mice increased by an order of magnitude. Our results suggest that Pt(IV)-Lu exhibits highly controllable activation property, superior antitumor activity, and good biosafety, which may provide a novel strategy for the design of platinum prodrugs.

15. Association between gut microbiota and menstrual disorders: a two-sample Mendelian randomization study.

Abstract

Background: Evidence from observational studies and clinical trials suggests that the gut microbiota is associated with gynecological diseases. However, the causal relationship between gut microbiota and menstrual disorders remains to be determined., Methods: We obtained summary data of gut microbiota from the global consortium MiBio-Gen's genome-wide association study (GWAS) dataset and data on menstrual disorders from the IEU Open GWAS project. MR-Egger, weighted median, inverse variance weighted, simple mode, and weighted mode were used to examine the causal association between gut microbiota and menstrual disorders. Thorough sensitivity studies were performed to confirm the data's horizontal pleiotropy, heterogeneity, and robustness., Results: Through MR analysis of 119 kinds of gut microbiota and 4 kinds of clinical phenotypes, it was discovered that 23 different kinds of gut microbiota were loosely connected to menstrual disorders. After FDR correction, the results showed that only Escherichia/Shigella ( p  = 0.00032, P FDR  = 0.0382, OR = 1.004, 95%CI = 1.002-1.006) is related to menstrual disorders., Conclusion: According to our MR Analysis, there are indications of a causal relationship between menstrual disorders and gut microbiota.
