However, the relationship between CXCR4 and ALK5 and the possible mechanisms promoted by ALK5 in the development of pain have not been evaluated.Materials and Methods: Tumor cell implantation (TCI) technology was used to generate a model of cancer-induced bone pain (CIBP) in rats; intrathecal (i.t.) injections of small interfering (si) RNAs targeting CXCR4 and the ALK5-specific inhibitor, RepSox, were performed. Behavioral outcomes, Western blotting, and immunofluorescence techniques were used to evaluate the expression of the aforementioned specific target proteins in the CIBP model.Results: The results revealed that i.t. administration of siRNAs targeting CXCR4 resulted in significant reductions in both mechanical and thermal hyperalgesia in rats with CIBP and likewise significantly reduced the expression of ALK5 in the spinal cord. Similarly, i.t. administration of RepSox also resulted in significant reductions in mechanical and thermal hyperalgesia in rats with CIBP together with diminished levels of spinal p-Smad3.Conclusion: Taken together, our results suggest that CXCR4 expression in the spinal cord may be a critical mediator of CIBP via its capacity to activate ALK5 and downstream signaling pathways.Keywords: chemokines, neoplasms, sensation, fluorescent antibody technique, cancer pain

Nevertheless, the function and underlying mechanisms of ceRNAs in esophageal cancer (EC) are still largely unknown. Methods: In this study, profiles of DEmRNAs, DElncRNAs, and DEmiRNAs between normal and EC tumor tissue samples were obtained from the Cancer Genome Atlas database using the DESeq package in R by setting the adjusted P2 as the cutoff. The ceRNA network (ceRNet) was initially constructed to reveal the interaction of these ceRNAs during carcinogenesis based on the bioinformatics of miRcode, miRDB, miRTarBase, and TargetScan. Then, independent microarray data of GSE6188, GSE89102, and GSE92396 and correlation analysis were used to validate molecular biomarkers in the initial ceRNet. Finally, a least absolute shrinkage and selection operator logistic regression model was built using an oncogenic ceRNet to diagnose EC more accurately. Results: We successfully constructed an oncogenic ceRNet of EC, crosstalk of hsa-miR372-centered CADM2-ADAMTS9-AS2 and hsa-miR145-centered SERPINE1-PVT1. In addition, the risk-score model −0.0053*log2(CADM2)+0.0168*log2(SERPINE1)-0.0073*log2(ADAMTS9-AS2)+0.0905*log2(PVT1)+0.0047*log2(hsa-miR372)–0.0193*log2(hsa-miR145), (log2[gene count]) could improve diagnosis of EC with an AUC of 0.988. Conclusion: We identified two novel pairs of ceRNAs in EC and its role of diagnosis. The pairs of hsa-miR372-centered CADM2-ADAMTS9-AS2 and hsa-miR145-centered SERPINE1-PVT1 were likely potential carcinogenic mechanisms of EC, and their joint detection could improve diagnostic accuracy. To identify and characterize individuals holding misperceptions around their weight status, it may be informative for clinicians to assess self-reported body mass index (BMI) classification (ie, underweight, normal, overweight, obese) in addition to clinical weight measurement. Methods: Self-reported weight classification data from the 2007 Current Visit Information – Child and Adolescent Survey collected at Mayo Clinic in Rochester, MN, were compared with measured clinical height and weight for 2,993 adolescents. Results: While, overall, 74.2% of adolescents accurately reported their weight status, females, younger adolescents, and proxy (vs self) reporters were more accurate. Controlling for demographic and behavioral characteristics, the higher an individual's BMI percentile, the less likely there was agreement between self-report and measured BMI percentile. Those with high BMI who misperceive their weight status were less likely than accurate perceivers to attempt weight loss. Conclusion: Adolescents’ and proxies’ misperception of weight status increases with BMI percentile. Obtaining an adolescent's self-perceived weight status in addition to measured height and weight offers clinicians valuable baseline information to discuss motivation for weight loss. Abstract

Referring to the dependence of income and occupational opportunities on education as well as to the lower compatibility between career and parenthood for women, educational differences of family foundation rates are being interpreted as an expression of different opportunity costs by many researchers (opportunity cost hypothesis). This hypothesis is discussed and analysed in this article for the German case drawing on the German Family Survey (Familiensurvey). The analysis considers two aspects of fertility motivation: on the one hand, it refers to educational level and the perceived value of children for (potential) parents; on the other, it examines the behavioural relevance of these fertility motivations. The empirical results from national representative longitudinal data question the opportunity cost hypothesis. Neither the perception of the incompatibility between career and motherhood nor its effect on the desire to become a parent are stronger for more highly educated women than for women with a lower formal educational level. Moreover, the article provides empirical evidence that the well-known educational differentiation of family formation rates is associated with child-related utility expectations, such as stimulation and affect.

Abstract

Nuclear factor-κB (NF-κB), a transcription factor that is activated by various stimuli, is associated with the pathogenesis of several cancers. One functional polymorphism, -94 insertion/deletion ATTG (rs28362491), in the human NFKB1 gene (one member of the NF-κB gene family) is associated with increased risk of various cancers. However, only one study has reported that rs28362491 is significantly associated with ovarian cancer. The aim of this study was to analyze the association between single nucleotide polymorphisms (SNPs) and haplotypes in the NFKB1 gene and the risk of ovarian cancer in a Chinese population. We examined the potential association between ovarian cancer and 15 SNPs (rs28362491, rs3774932, rs1598856, rs230531, rs230530, rs230528, rs230521, rs230498, rs230539, rs1005819, rs3774956, rs4648055, rs4648068, rs3774964, rs3774968) of the NFKB1 gene using the MassARRAY system. Participants included 411 patients with ovarian cancer and 438 healthy controls. The results showed that the allelic or genotypic frequencies of three polymorphisms, including rs28362491 (promoter region), rs230521 (intron 4), and rs4648068 (intron 12), in the patients with ovarian cancer, were significantly different from those in the healthy controls. Strong linkage disequilibrium was observed in four blocks (D' > 0.9). Significantly more A-C (block 2: rs230528-rs230521) haplotypes (P = 0.0003 after Bonferroni's corrections) and G-A-A (block 4: rs4648068-rs3774964-rs3774968) haplotypes (P = 0.021) were found in the patients with ovarian cancer. These findings point to a role of the NFKB1 polymorphism in patients with ovarian cancer among a Chinese Han population, and may be informative for future genetic or biological studies on ovarian cancer.

Abstract

We have measured the dijet angular distribution in $\sqrt{s}$=1.8 TeV $p\bar{p}$ collisions using the D0 detector. Order $\alpha^{3}_{s}$ QCD predictions are in good agreement with the data. At 95% confidence the data exclude models of quark compositeness in which the contact interaction scale is below 2 TeV., Comment: 11 pages, Latex, 3 postscript figures

28. Fuzzy tolerance multilevel approach for structural topology optimization

Abstract

This paper presents a novel methodology, fuzzy tolerance multilevel programming approach, for applying fuzzy set theory and sequence multilevel method to multi-objective topology optimization problems of continuum structures undergoing multiple loading cases. Ridge-type nonlinear membership functions in fuzzy set theory are applied to embody fuzzy and uncertain characteristics essentially involved by the objective and constraint functions. Sequence multilevel method is used to characterize the different priorities of loading cases at different levels making contribution to the final optimum solution, which is practically beneficial to reduce the subjective influence transferred by using weighted approaches. The solid isotropic material with penalization (SIMP) is adopted as the density-stiffness interpolation scheme to relax the original optimization problem and indicate the dependence of material properties with element pseudo-densities. Sequential linear programming (SLP) is used as the optimizer to solve the multi-objective optimization problem formulated using fuzzy tolerance multilevel programming scheme. Numerical instabilities, such as checkerboards and mesh dependencies are summarized and a duplicate sensitivity filtering method, in favor of contributing to the mesh-dependent optimum designs, is subsequently proposed to regularize the singularity of the optimization problem. The validation of the methodologies presented in this work has been demonstrated by detailed examples of numerical applications. © 2005 Elsevier Ltd. All rights reserved.

29. Multiple stiffness topology optimizations of continuum structures

Abstract

This paper presents a new multi-objective topology optimization algorithm for continuum structures under multiple loading cases. An expert evaluation method of weights based on grey system theory is proposed to calculate the objective weights when the compromise programming approach is employed as a multi-objective optimization scheme converting the multi-objective problem to a single objective problem. A modified updating scheme with a self-adaptive move limit for design variables is also suggested, SIMP is regarded as density-stiffness interpolation scheme and the optimality criteria method is used as the optimizer. Numerical instabilities, such as checkerboards and mesh dependencies, are also discussed. The validities of these methods in this paper are demonstrated by some numerical applications.

Abstract

A measurement of the cross section for production of single, isolated photons is reported for transverse energies in the range of 10-125 GeV for two regions of pseudorapidity, |η| < 0.9 and 1.6 < |η| < 2.5. The data represent 12.9 pb-1 of integrated luminosity accumulated in p̄p collisions at √s = 1.8 TeV and recorded with the DO detector at the Fermilab Tevatron Collider. The background, predominantly from jets which fragment to neutral mesons, is estimated using the longitudinal shower shape in the calorimeter. In both pseudorapidity regions the cross section is found to agree with the next-to-leading order QCD prediction for 30 ≲ ET y ≲ 80 GeV.

Abstract

We present results from a search for anomalous WW and WZ production in p p̄ collisions at √s = 1.8 TeV. We used p p̄ →jjX events observed during the 1992-1993 run of the Fermilab Tevatron collider. corresponding to an integrated luminosity of 13.7 ± 0.7 pb -1. A fit to the transverse momentum spectrum of the W boson yields direct limits on the CP-conserving anomalous WWγ and WWZ coupling parameters of -0.9 < Δκ < 1.1 (with λ = 0) and -0.6 < λ < 0.7 (with Δκ = 0) at the 95% confidence level, for a form factor scale Λ = 1.5 TeV, assuming that the WWγ and WWZ coupling parameters are equal.

Abstract

The global topologies of inclusive three- and four-jet events produced in p̄p interactions are described. The three- and four-jet events are selected from data recorded by the DO detector at the Fermilab Tevatron Collider operating at a center-of-mass energy of √s = 1800 GeV. The measured, normalized distributions of various topological variables are compared with parton-level predictions of tree-level QCD calculations. The parton-level QCD calculations are found to be in good agreement with the data. The studies also show that the topological distributions of the different subprocesses involving different numbers of quarks are very similar and reproduce the measured distributions well. The parton-shower Monte Carlo generators provide a less satisfactory description of the topologies of the three- and four-jet events. © 1996 The American Physical Society.

Abstract

We report on a search for right-handed W bosons (WR) with mass greater than 200 GeV/c2. We used data collected with the DO detector at the Fermilab Tevatron p p̄ collider at √s = 1.8 TeV to search for WR decays into an electron and a massive right-handed neutrino WR +- → e+-NR. Using the inclusive electron data, we set mass limits independent of the NR decay: mWR > 650 GeV/c2 and mWR > 720 GeV/c2 at the 95% confidence level, valid for mNR > 1/2mWR and mNR ≪ mWR. respectively. The latter also represents a new lower limit on the mass of a heavy left-handed W boson (W') decaying into ev. In addition, limits on mWRvalid for larger values of the NR mass are obtained assuming that NR decays to an electron and two jets.

Abstract

We present a search for pair produced top squarks, the supersymmetric partners of the top quark, using the D0 detector at the Fermilab Tevatron pp̄ collider. We consider a scenario in which the lighter of the two top squarks t̃ l decays with 100% branching fraction to a charm quark and the lightest neutralino X̃ 1 0 yielding a signal of two acollinear jets with missing transverse energy. We observe 3 events while we expect 3.5 ± 1.2 events from the known standard model processes. We exclude at the 95% confidence level a significant region of the mt̃1-mX̃10 parameter space. The highest mt̃1 value we exclude is 93 GeV/c 2 with a corresponding mmX̃10 value of 8 GeV/c 2.

Abstract

A study of the particle multiplicity between jets with large rapidity separation has been performed using the D0 detector at the Fermilab Tevatron pp̄ Collider operating at √s = 1.8 TeV. A significant excess of low-multiplicity events is observed above the expectation for color-exchange processes. The measured fractional excess is 1.07 ± 0.10(stat) -0.13 +0.25(syst)%, which is consistent with a strongly interacting color-singlet (colorless) exchange process and cannot be explained by electroweak exchange alone. A lower limit of 0.80% (95% C.L.) is obtained on the fraction of dijet events with color-singlet exchange, independent of the rapidity gap survival probability.

38. Sexual abuse and lifetime diagnosis of somatic disorders: a systematic review and meta-analysis.

Abstract

Context: Many patients presenting for general medical care have a history of sexual abuse. The literature suggests an association between a history of sexual abuse and somatic sequelae.Objective: To systematically assess the association between sexual abuse and a lifetime diagnosis of somatic disorders. Data Sources and Extraction A systematic literature search of electronic databases from January 1980 to December 2008. Pairs of reviewers extracted descriptive, quality, and outcome data from included studies. Odds ratios (ORs) and 95% confidence intervals (CIs) were pooled across studies by using the random-effects model. The I(2) statistic was used to assess heterogeneity.Study Selection: Eligible studies were longitudinal (case-control and cohort) and reported somatic outcomes in persons with and without history of sexual abuse.Results: The search identified 23 eligible studies describing 4640 subjects. There was a significant association between a history of sexual abuse and lifetime diagnosis of functional gastrointestinal disorders (OR, 2.43; 95% CI, 1.36-4.31; I(2) = 82%; 5 studies), nonspecific chronic pain (OR, 2.20; 95% CI, 1.54-3.15; 1 study), psychogenic seizures (OR, 2.96; 95% CI, 1.12-4.69, I(2) = 0%; 3 studies), and chronic pelvic pain (OR, 2.73; 95% CI, 1.73-4.30, I(2) = 40%; 10 studies). There was no statistically significant association between sexual abuse and a lifetime diagnosis of fibromyalgia (OR, 1.61; 95% CI, 0.85-3.07, I(2) = 0%; 4 studies), obesity (OR, 1.47; 95% CI, 0.88-2.46; I(2) = 71%; 2 studies), or headache (OR, 1.49; 95% CI, 0.96-2.31; 1 study). We found no studies that assessed syncope. When analysis was restricted to studies in which sexual abuse was defined as rape, significant associations were observed between rape and a lifetime diagnosis of fibromyalgia (OR, 3.35; 95% CI, 1.51-7.46), chronic pelvic pain (OR, 3.27; 95% CI, 1.02-10.53), and functional gastrointestinal disorders (OR, 4.01; 95% CI, 1.88-8.57).Conclusion: Evidence suggests a history of sexual abuse is associated with lifetime diagnosis of multiple somatic disorders. Abstract

Background: Differential counting (DC) of different cell types in bone marrow (BM) aspiration smears is crucial for diagnosing hematological diseases. However, a clinically applicable method for automatic DC has yet to be developed., Objective: This study developed and validated an artificial intelligence (AI)-based algorithm for identifying and classifying nucleated cells in BM smears., Methods: In the development phase, a mask region-based convolutional neural network (Mask R-CNN)-based AI model was trained to detect and classify individual BM cells. We used a large data set of expert-annotated images representing a variety of disease categories. The BM slides were stained with Liu's stain or Wright-Giemsa stain. Consensus meetings were held to ensure experts from different institutes applied consistent criteria in classifying cells. Subsequently, the performance of the AI algorithm in identifying cell images and determining cell ratios was evaluated using a multinational clinical dataset., Results: The AI model was trained on 542 slides (85.1 % stained with Liu's stain and 14.9 % with Wright-Giemsa stain) containing 597,222 annotated cells. It achieved an accuracy of 0.94 for the testing dataset containing 26,170 cells. The performance of the AI model was further validated using another multinational real-world dataset (data obtained from three centers in Taiwan and one in the United States) comprising 200,639 cells. The AI model achieved an accuracy of 0.881 in classifying individual cells and demonstrated high precision in classifying blasts (0.927), bands and polymorphonuclear neutrophils (0.955), plasma cells (0.930), and lymphocytes (0.789). When the differential counting percentage of each cell type was assessed, a strong correlation (ρ > 0.8) between the AI and manual methods was observed for most cell categories., Conclusions: In this study, an AI algorithm was developed and clinically validated using large, multinational datasets. Abstract

In this paper, we introduce an R package EATME, which is known as Exponentially weighted moving average (EWMA) control chart with Adjustments To Measurement Error. The main purpose of this package is to correct for measurement error effects in continuous or binary random variables and develop the corrected control charts based on the EWMA statistic. In addition, the corrected control charts can detect out-of-control process accurately. The package contains a function to generate synthetic data and includes functions to determine the reasonable coefficient of control limit as well as estimate average run length. Moreover, for the visualization, we also provide the control charts to show the monitoring of in-control and out-of-control process. The article "MiR-15a-3p suppresses the growth and metastasis of ovarian cancer cell by targeting Twist1" by B. Fan, L.-P. Chen, Y.-H. Yuan, H.-N. Xiao, X.-S. Lv, Z.-Y. Xia, published in Eur Rev Med Pharmacol Sci 2019; 23 (5): 1934-1946-DOI: 10.26355/eurrev&#95;201903&#95;17232-PMID: 30915736 has been retracted by the Editor in Chief. The authors contacted the journal, claiming that, after carefully examining the published article, they found that two of the figures in the article were duplicates. They also stated that they did not have access to the original figures. Therefore, they requested the manuscript to be retracted. After the authors' email, the journal started an investigation and found that the article was also questioned on PubPeer (link: https://pubpeer.com/publications/3FA0DDA41DFC73C5E8E8E1C505DBAA). The journal's investigation uncovered a duplication between Figures 3D and Figure 6D. Consequently, the Editor in Chief mistrusts the results presented and has decided to retract the article. Abstract

In causal inference, the estimation of the average treatment effect is often of interest. For example, in cancer research, an interesting question is to assess the effects of the chemotherapy treatment on cancer, with the information of gene expressions taken into account. Two crucial challenges in this analysis involve addressing measurement error in gene expressions and handling noninformative gene expressions. While analytical methods have been developed to address those challenges, no user-friendly computational software packages seem to be available to implement those methods. To close this gap, we develop an R package, called AteMeVs, to estimate the average treatment effect using the inverse-probability-weighting estimation method to handle data with both measurement error and spurious variables. This developed package accommodates the method proposed by Yi and Chen (2023) as a special case, and further extends its application to a broader scope. Abstract

Objectives: To study the effects and mechanisms of tetramethylpyrazine (TMP) on tumor necrosis factor-α (TNF-α)-induced inflammatory injury in human coronary artery endothelial cells (HCAEC)., Methods: HCAEC were randomly divided into four groups: the control group (no treatment), the model group (treated with TNF-α, 50 ng/mL for 24 hours), the TMP group (pre-treated with TMP, 80 μg/mL for 12 hours followed by TNF-α treatment for 24 hours), and the SIRT1 inhibitor group (pre-treated with TMP and the specific SIRT1 inhibitor EX527 for 12 hours followed by TNF-α treatment for 24 hours). Cell viability was assessed using the CCK-8 method, lactate dehydrogenase (LDH) activity was measured using an LDH assay kit, reactive oxygen species (ROS) levels were observed using DCFH-DA staining, expression of pyroptosis-related proteins was detected by Western blot, and SIRT1 expression was analyzed using immunofluorescence staining., Results: Compared to the control group, the model group showed decreased cell viability, increased LDH activity, ROS level and expression of pyroptosis-related proteins, and decreased SIRT1 expression ( P <0.05). Compared to the model group, the TMP group exhibited increased cell viability, decreased LDH activity, ROS level and expression of pyroptosis-related proteins, and increased SIRT1 expression ( P <0.05). In comparison to the TMP group, the SIRT1 inhibitor group showed decreased cell viability, increased LDH activity, ROS level and expression of pyroptosis-related proteins, and decreased SIRT1 expression ( P <0.05)., Conclusions: TMP may attenuate TNF-α-induced inflammatory injury in HCAEC, which is associated with the inhibition of pyroptosis and activation of the SIRT1 signaling pathway.

Abstract

Background and Objectives: High-flow nasal cannula (HFNC) is used in up to 50% of children admitted with bronchiolitis. Consensus recommendations for feeding these children have not been established, and there is variability in practice. We sought to identify factors influencing feeding decisions for general care patients admitted with bronchiolitis on HFNC from a national sample of interdisciplinary care team members., Methods: In this qualitative study, we conducted semi-structured virtual interviews with care team members involved in making feeding decisions at 10 US hospitals from June 2022 to March 2023. Eligible participants included 1 nurse, respiratory therapist, speech language pathologist, and physician from each site. Interviews were audio-recorded, transcribed, and conducted until reaching sufficiency. Three researchers analyzed transcript data using content analysis guided by the Systems Engineering Initiative for Patient Safety 2.0 model., Results: We interviewed 29 participants, including 19 nurses, respiratory therapists, and speech language pathologists and 10 physicians. Participants identified 11 factors influencing feeding decisions related to hospital work systems and processes outlined in the Systems Engineering Initiative for Patient Safety model, including people (child and parent characteristics, care team experience), tools and technology (guideline, protocol, ordersets), organization (institutional culture, education), environment (time of day, care location), task (interventions to optimize feeding), and process (clinical assessment, feeding trial, communication)., Conclusions: Our findings suggest that feeding decisions are driven by factors related to the child, care team experience, institutional tools, and culture. Abstract

Full-endoscopic lumbar interbody fusion (FELIF) is a critical yet challenging procedure. However, extensive analyses of discectomy and cartilage endplate preparation techniques are limited. This can be attributed to the lack of universal protocols owing to diverse surgical practices and equipment preferences. Therefore, this narrative review presents a comprehensive overview of discectomy and cartilage endplate preparation techniques in FELIF. A literature search of the PubMed, Embase, and Google Scholar databases in December 2023 retrieved 490 studies, of which 53 met the predefined inclusion criteria, and 1373 patients were included in the analyses. Spinal endoscopic disc and cartilage endplate removal can be categorized into 2 main types: removal under direct endoscopic visualization and removal under radiographic guidance with the protection of a working sheath following the endoscope's removal. Removal under direct visualization ensures the safety and precision of the procedure. Radiographic guidance can enhance the efficiency of the removal process. Specially designed instruments can be utilized through the narrow working channels of spinal endoscopes for the scraping surgery. Moreover, many traditional spinal endoscopic instruments, through specific techniques and manipulations, can also aid in cartilage removal. The approaches and techniques vary significantly among physicians, but overall, these instruments and techniques aim to achieve a safe and efficient disc-scraping outcome. Thus, this review may offer a comprehensive guidance to surgeons in selecting the most efficient practices for FELIF. 50. Botulinum toxin type A-targeted SPP1 contributes to neuropathic pain by the activation of microglia pyroptosis.

Abstract

Background: Neuropathic pain (NP) is the primary symptom of various neurological conditions. Patients with NP often experience mood disorders, particularly depression and anxiety, that can severely affect their normal lives. Microglial cells are associated with NP. Excessive inflammatory responses, especially the secretion of large amounts of pro-inflammatory cytokines, ultimately lead to neuroinflammation. Microglial pyroptosis is a newly discovered form of inflammatory cell death associated with immune responses and inflammation-related diseases of the central nervous system., Aim: To investigate the effects of botulinum toxin type A (BTX-A) on microglial pyroptosis in terms of NP and associated mechanisms., Methods: Two models, an in vitro lipopolysaccharide (LPS)-stimulated microglial cell model and a selective nerve injury model using BTX-A and SPP1 knockdown treatments, were used. Key proteins in the pyroptosis signaling pathway, NLRP3-GSDMD, were assessed using western blotting, real-time quantitative polymerase chain reaction, and immunofluorescence. Inflammatory factors [interleukin (IL)-6, IL-1β, and tumor necrosis factor (TNF)-α] were assessed using enzyme-linked immunosorbent assay. We also evaluated microglial cell proliferation and apoptosis. Furthermore, we measured pain sensation by assessing the delayed hind paw withdrawal latency using thermal stimulation., Results: The expression levels of ACS and GSDMD-N and the mRNA expression of TNF-α , IL-6 , and IL-1β were enhanced in LPS-treated microglia. Furthermore, SPP1 expression was also induced in LPS-treated microglia. Notably, BTX-A inhibited SPP1 mRNA and protein expression in the LPS-treated microglia. Additionally, depletion of SPP1 or BTX-A inhibited cell viability and induced apoptosis in LPS-treated microglia, whereas co-treatment with BTX-A enhanced the effect of SPP1 short hairpin (sh)RNA in LPS-treated microglia. Finally, SPP1 depletion or BTX-A treatment reduced the levels of GSDMD-N, NLPRP3, and ASC and suppressed the production of inflammatory factors., Conclusion: Notably, BTX-A therapy and SPP1 shRNA enhance microglial proliferation and apoptosis and inhibit microglial death.
