1. The E302 instability-versus-efficiency experiment at FACET-II

Abstract

We discuss plans for the E302 instability-efficiency experiment, starting in 2024 at the recently upgraded FACET-II facility at SLAC National Accelerator Laboratory. The beam-breakup instability will be the main area of study for the E302 experiment. With the imaging spectrometer at FACET-II, we introduce a novel technique for observing and quantifying the amplitude growth of the trailing bunch due to the transverse instability. Using the transverse position on the spectrometer screen and the transfer matrix of the magnetic lattice used for the spectrometer, we aim to extract a $x'$-$E$ charge distribution that can be used to quantify the amplitude of the beam. By varying the trailing bunch's charge and, hence, the beam loading of the accelerating field, we aim to adjust the wake-to-beam power transfer efficiency in the E302 experiment. We plan to quantify the amplitude for different configurations of the beam charge and, hence, investigate the relationship between the beam-breakup instability and efficiency. We use a combination of particle-in-cell (PIC) codes to simulate a beam-driven plasma wakefield accelerator from start-to-end with a FACET-II-like spectrometer and demonstrate the methodology that will be used for the instability studies at the E302 experiment., Comment: 9 pages, 9 figures, conference proceeding for the 6th European Advanced Accelerator Concepts workshop (EAAC2023), 17-23 September 2023

5. A hexa-herbal Chinese formula for treatment of atopic dermatitis : phytochemical analysis and selected anti-inflammatory activities

Abstract

Diverse pharmacological activities and the solid clinical performance of Chinese herbal medicines have attracted worldwide attention in terms of its modernization. Here, a hexaherbal Chinese formula (HHCF) for treating atopic dermatitis (AD) topically has been studied from chemical and biological perspectives. The HHCF comprises the rootstock of Scutellaria baicalensis Georgi (SCU), Rheum tanguticum Maxim. Ex Balf. (RHE), Sophora flavescens Aiton (SOP); the root bark of Dictamnus dasycarpus Turcz. (DIC); the bark of Phellodendron chinense C.K. Schneid. (PHE) and the fruit of Kochia scoparia (L.) Schrad. (KOC). In this thesis the chemical composition of the HHCF has been profiled and characterized using liquid-chromatography (LC) coupled with triple quadruple mass spectrometry (MS). 68 chemical compounds including alkaloids, anthraquinones, coumarins, flavonoids, naphthalene derivatives, phenylbutanone glucosides, phenolic acids, pterocarpans, stilbenes and tannins were putatively identified in the LC-MS profile of the HHCF based on mass measurement and characteristic fragment ions. The source(s) of these chemical compounds has been analyzed using the developed EXCEL template and PHE, RHE and SOP contributed the largest number of chemical compounds identified in the HHCF, while KOC seems to contribute very little. To evaluate the anti-inflammatory effects of the HHCF for treating AD/skin inflammation, the thymus and activation-regulated chemokine (CCL17), Prostaglandin E2 (PGE2), IL-8, IL-6 and hyaluronidase inhibitory effects were studies in vitro. Results showed that the HHCF inhibited hyaluronidase activity, TNF-α- plus IFN--induced CCL17 production in spontaneously immortalized human epidermal keratinocytes (HaCaT) and IL-1β-induced PGE2 in human fibroblasts with no effects on IL-8 and IL- 6. Among the chemical compounds characterized in LC-MS profile of the HHCF, multivariate regression models indicated the major contributor to the CCL17 inhibition were berberine, catechin dimers, gallic acid, galloylglucose, 4-(4'-hydroxylphenyl)-2- butanone 4'-O-β-D-glucoside, pyrogallol and resveratrol 4'-O-β-D-(6''-O-galloyl) glucoside. The effects of the HHCF against other pathogenesis-related targets need to be further study to ascertain its therapeutic efficacy against AD.

Abstract

The performance of a double sided silicon strip detector (DSSSD), used for position and energy detection of heavy ions, is reported. The analysis shows that the incomplete charge collection (ICC) and charge sharing (CS) effects of the DSSSD give rise to a loss of energy resolution, however the position information is recorded without ambiguity. Representations of ICC/CS events in the energy spectra are shown and their origins are confirmed by correlation analysis of the spectra from both junction side and ohmic side of the DSSSD.

Abstract

In antineutrophil cytoplasmic autoantibody (ANCA)-associated vasculitides (AAV), the two major target antigens of ANCA are proteinase 3 (PR3) and myeloperoxidase (MPO). Evidence is accumulating that there are distinct differences between patients with PR3-AAV and those with MPO-AAV. In the present study, the clinicopathological features and prognosis of patients with PR3-AAV and MPO-AAV from a single center in China were retrospectively analyzed. A total of 212 Chinese patients with AAV were recruited in the present study; 189/212 (89.15%) patients were classified as having MPO-AAV and 23/212 (10.85%) patients as having PR3-AAV. Compared with those in the PR3-AAV group, patients in the MPO-AAV group were older and less frequently had ear, nose and throat or ophthalmic involvement. MPO-AAV patients had higher levels of serum creatinine and proteinuria at baseline. No significant difference was observed with regard to the pathological changes of the glomeruli and tubulointerstitium between the two groups. The probability of developing end-stage renal disease was significantly higher in patients with MPO-AAV compared with that in patients with PR3-AAV. There was no significant difference in the one-year patient survival rate between the two groups. However, differences in certain clinical characteristics and outcomes were observed between MPO-AAV and PR3-AAV patients. A large national investigation of AAV is required to confirm the concept that PR3-AAV and MPO-AAV are distinct disease entities.Copyright © 2021 Spandidos Publications. All rights reserved.

18. Application of point-of-care ultrasound in patients receiving enteral nutrition.

Abstract

OBJECTIVE: Enteral nutrition (EN) is the first-choice nutritional support, as it is more in line with normal physiological processes. During EN, the major goals to achieve include accurate confirmation of the feeding tube position, monitoring the gastric residual volume, assessing gastrointestinal motility, and monitoring the nutritional status of patients. With rapid development in technology, point-of-care ultrasound (POCUS) has become a more convenient and effective technical tool for monitoring critically ill patients receiving EN. In this review, we have summarized and discussed the value of POCUS in the implementation, monitoring, and evaluation of EN therapy to provide a reference for nutritional support of critically ill patients in critical care settings. MATERIALS AND METHODS: This is a narrative review. A literature search for Scopus-indexed articles was performed randomly using PubMed and MEDLINE databases as the primary sources. No specific term was used for the search. RESULTS: POCUS can be used for positioning of nasogastric and nasointestinal tubes, evaluation of gastric residuals and gastrointestinal motility as well as monitoring of nutritional status. CONCLUSIONS: POCUS is a real-time, highly repeatable, radiation-free, and non-invasive visual inspection technique, with high application value in assessing the nutritional status of patients receiving EN and guiding the development of further nutritional treatment plans. 20. Tin stable isotopes in magmatic-affected coal deposits : insights in the geochemical behavior of tin

Abstract

Tin (Sn) is a redox-sensitive element and geochemically incompatible that accumulates in evolved magmas and associated hydrothermal fluids. Here, a Sn stable isotope approach is used to characterize sources and geochemical behaviors of Sn in magmatic intruded coal deposits by combining with petrographic and elemental analysis. Sn isotope ratios of twelve coal samples with various magmatic-affected degrees show a delta Sn-120/118 variation from -0.06 to 0.22%. (2 sigma of 0.07 parts per thousand, relative to NIST 3161a). Basing on Sn isotope data and geochemical characteristics of the samples, we suggest binary source mixing and redox state related isotope shift as two mechanisms accounting for the isotope variations. The first mechanism is an admixture of magmatichydrothermal fluids, characterized by high delta(120/118), into coal deposits with low delta(120/118). A binary mixing model is used to estimate Sn contributions of the two end-members. The second process results in a positive isotope shift caused by an oxidation of sample states when they are affected by magmatic intrusions. In fact, magmatic intrusions could trigger the oxidation of Sn, resulting in heavy Sn isotope enrichment in the magmatic affected samples. The change in coal redox-state is supported by a concomitant change of redox proxies, such as V/(V + Ni), Mn/Cr and V/Cr. The enrichment of heavy Sn isotopes in samples impacted by magmatic hydrothermal fluids compared to normally-deposited coals could be explained by the conversion of dominant Sn species from Sn (II)-S to Sn (IV)-O under the influence of oxidative magma, which favors the enrichment of heavier Sn isotopes in Sn (IV)-O due to its shorter bond length. This study shows that Sn isotope ratios are useful tools for investigating Sn geochemical behavior in different Earth reservoirs and during redox sensitive processes.

42. An Improvement on Design of Sheet Extrusion Dies

Abstract

On the basis of the method of representative values and the flow pattern, that is, the position of the isobars parallels to the die orifice (PIPDO), the design of dies for the sheet and annular parisons is improved in the present paper. Since this pattern includes the effects of the sloped manifold and the die design for the circular manifold are independent of flow rate and the polymer viscosity, the distribution system of a die gives better uniformity of flow. In the previous study [1] a defect is found, that is, the calculated position of the path of the island area lags behind actual position. This defect is eliminated, and the modified equation is obtained. Another flow pattern, the pressure loss on all flow path is equal (PLFPE), is introduced. The equivalency of the above two flow patterns is proved theoretically. Although the flow uniformity isn’t perfect in the simplified analytical design of dies proposed in the previous paper, this simplified design is still of advantages. Therefore, its design criterion is put forward in this paper.

Abstract

Background IgE‐expressing (IgE+) plasma cells (PCs) provide a continuous source of allergen‐specific IgE that is central to allergic responses. The extreme sparsity of IgE+ cells in vivo has confined their study almost entirely to mouse models. Objective To characterize the development pathway of human IgE+ PCs and to determine the ontogeny of human IgE+ PCs. Methods To generate human IgE+ cells, we cultured tonsil B cells with IL‐4 and anti‐CD40. Using FACS and RT‐PCR, we examined the phenotype of generated IgE+ cells, the capacity of tonsil B‐cell subsets to generate IgE+ PCs and the class switching pathways involved. Results We have identified three phenotypic stages of IgE+ PC development pathway, namely (i) IgE+germinal centre (GC)‐like B cells, (ii) IgE+ PC‐like ‘plasmablasts’ and (iii) IgE+ PCs. The same phenotypic stages were also observed for IgG1+ cells. Total tonsil B cells give rise to IgE+ PCs by direct and sequential switching, whereas the isolated GC B‐cell fraction, the main source of IgE+ PCs, generates IgE+ PCs by sequential switching. PC differentiation of IgE+ cells is accompanied by the down‐regulation of surface expression of the short form of membrane IgE (mIgES), which is homologous to mouse mIgE, and the up‐regulation of the long form of mIgE (mIgEL), which is associated with an enhanced B‐cell survival and expressed in humans, but not in mice. Conclusion Generation of IgE+ PCs from tonsil GC B cells occurs mainly via sequential switching from IgG. The mIgEL/mIgES ratio may be implicated in survival of IgE+ B cells during PC differentiation and allergic disease.

47. Circ-ABCB10 acts as an oncogene in glioma cells via regulation of the miR-620/FABP5 axis.

Abstract

OBJECTIVE: This study aims to investigate the biological function of circular RNA ABCB10 (circ-ABCB10) in regulating the progression of glioma and to study the possible underlying mechanisms. PATIENTS AND METHODS: The expression levels of circ-ABCB10, miR-620 and FABP5 mRNA in glioma tissues, normal surrounding tissues and glioma cell lines were measured by Real-time PCR (RT-PCR). Circ-ABCB10 was silenced by siRNA in glioma cell lines (U87, T98G). The proliferation, migration and invasion of glioma cells were measured by MTT, wound healing and transwell assays, respectively. The relationship between circ-ABCB10, miR-620 and FABP5 was tested by Dual-Luciferase assay. The expression of proteins was measured by Western blot. The cell cycle distribution and apoptosis were measured by flow cytometry. RESULTS: The expression levels of circ-ABCB10 and FABP5 in glioma tissues and cells were significantly higher than those in their normal counterparts. Moreover, the expression of miR-620 was lower in glioma tissues. Silencing of circ-ABCB10 in glioma cells significantly inhibited the proliferation, migration and invasion of glioma cells. Moreover, downregulation of circ-ABCB10 induced cell cycle arrest and apoptosis in glioma cells. Furthermore, inhibition of miR-620 showed the opposite effects to silencing circ-ABCB10 on glioma cells. Dual-Luciferase reporter assays demonstrated that circ-ABCB10 could bind to miR-620 and that FABP5 was a direct target of miR-620. Western blot results showed that circ-ABCB10 could stabilize the expression of FABP5, while miR-620 decreased the expression of FABP5. Furthermore, overexpression of FABP5 abrogated the silencing effects of circ-ABCB10 in glioma cells. CONCLUSIONS: These data suggest that circ-ABCB10 affects glioma progression by regulating the miR-620/FABP5 axis, and circ-ABCB10 might be used as a potential target for the treatment of glioma.
