Abstract

The aggressive nature and poor prognosis of lung cancer led us to explore the mechanisms driving disease progression. Utilizing our invasive cell-based model, we identified methylthioadenosine phosphorylase (MTAP) and confirmed its suppressive effects on tumorigenesis and metastasis. Patients with low MTAP expression display worse overall and progression-free survival. Mechanistically, accumulation of methylthioadenosine substrate in MTAP-deficient cells reduce the level of protein arginine methyltransferase 5 (PRMT5)-mediated symmetric dimethylarginine (sDMA) modification on proteins. We identify vimentin as a dimethyl-protein whose dimethylation levels drop in response to MTAP deficiency. The sDMA modification on vimentin reduces its protein abundance but trivially affects its filamentous structure. In MTAP-deficient cells, lower sDMA modification prevents ubiquitination-mediated vimentin degradation, thereby stabilizing vimentin and contributing to cell invasion. MTAP and PRMT5 negatively correlate with vimentin in lung cancer samples. Taken together, we propose a mechanism for metastasis involving vimentin post-translational regulation.

33. Structuring Cooperative Learning in Teaching English Pronunciation

Abstract

Classrooms incorporating Cooperative Learning (CL) structures facilitate a supportive learning environment for English Language Learners (ELLs). Accurate pronunciation by ELLs is important for communication, and also benefits academic achievement. The known benefits of CL for ELLs make it a desirable learning environment to teach pronunciation skills. Thus, sufficient consideration must be given to its incorporation in curricula and delivery format. However, in implementing CL structures in ELL classrooms, attention must be paid to incubation time for CL elements, social and cultural factors. This quasi-experiment, with a pre-post test two-treatment, formed by expanding across program design investigated the impact of a CL environment in the pronunciation skills of ELLs. Results showed that the difference between the improvement of the CL (experimental) group and the control group was not significant enough to claim that CL was a major differential factor. Factors affecting the implementation of CL in ELL classrooms are discussed.

Abstract

There are no antivirals or vaccines available to treat Enterovirus 71 (EV71) infections. Although the type I interferon response, elicited upon virus infection, is critical to establishing host antiviral innate immunity, EV71 fails to induce this response efficiently. Here we provide new insights into potential anti-EV71 therapy by showing that neutralization of EV71-induced miR-146a prevents death in mice by restarting the production of type I interferon. EV71 infection upregulates miR-146a, which targets IRAK1 and TRAF6 involved in TLR signalling and type I interferon production. We further identify AP1 as being responsible for the EV71-induced expression of miR-146a. Surprisingly, knocking out miR-146a or neutralizing virus-induced miR-146a by specific antagomiR restores expressions of IRAK1 and TRAF6, augments IFNβ production, inhibits viral propagation and improves survival in the mouse model. Our results suggest that enterovirus-induced miR-146a facilitates viral pathogenesis by suppressing IFN production and provide a clue to developing preventive and therapeutic strategies for enterovirus infections.

Abstract

Background: Examining patients with first-episode psychosis (FEP) provides opportunities to better understand the mechanism underlying these illnesses. By incorporating quantitative measures in FEP patients, we aimed to (1) determine the baseline distribution of clinical features; (2) examine the impairment magnitude of the quantitative measures by comparing with external controls and then the counterparts of schizophrenia patients of different familial loadings; and (3) evaluate whether these quantitative measures were associated with the baseline clinical features. Methods: Patients with FEP were recruited from one medical center, two regional psychiatric centers, and two private clinics in northern Taiwan with clinical features rated using the Positive and Negative Syndrome Scale (PANSS) and Personal and Social Performance (PSP) scale. Quantitative measurements included the Continuous Performance Test (CPT), Wisconsin Card Sorting Test (WCST), niacin response abnormality (NRA), and minor physical anomalies and craniofacial features (MPAs). To evaluate the relative performance of the quantitative measures in our FEP patients, four external comparison groups from previous studies were used, including three independent healthy controls for the CPT, WCST, and NRA, respectively, and one group of treatment-resistant schizophrenia patients for the MPAs. Additionally, patients from simplex families and patients from multiplex families were used to assess the magnitude of FEP patients’ impairment on the CPT, WCST, and NRA. Results: Among the 80 patients with FEP recruited in this study (58% female, mean age = 25.6 years, mean duration of untreated psychosis = 132 days), the clinical severity was mild to moderate (mean PANSS score = 67.3; mean PSP score = 61.8). Patients exhibited both neurocognitive and niacin response impairments (mean Z-scores: −1.24 for NRA, − 1.06 for undegraded d', − 0.70 for degraded d', − 0.32 for categories achieved, and 0.44 for perseverative errors) but did not show MPAs indicative of treatment resistance. Among these quantitative measures, three of the four neurocognitive indices were correlated with the baseline clinical features, whereas NRA did not show such correlation. Conclusions: This FEP study of Taiwanese patients revealed the presence of neurocognitive performance and niacin response and their different relationships with clinical features, rendering this sample useful for future follow-up and incorporation of multiomics investigation. Abstract

Background: Mercaptopurine-induced neutropenia can interrupt chemotherapy and expose patients to infection during childhood acute lymphoblastic leukemia (ALL) treatment. Previously, six candidate gene variants associated with mercaptopurine intolerance were reported. Herein, we investigated the association between the mean tolerable dose of mercaptopurine and these genetic variants in Taiwanese patients. Methods: In total, 294 children with ALL were treated at the National Taiwan University Hospital from April 1997 to December 2017. Germline variants were analyzed for NUDT15, SUCLA2, TPMT, ITPA, PACSIN2, and MRP4. Mean daily tolerable doses of mercaptopurine in the continuation phase of treatment were correlated with these genetic variants. Results: Mercaptopurine intolerance was significantly associated with polymorphisms in NUDT15(Pvalue < 0.0001). Patients with SUCLA2variants received lower mercaptopurine doses (Pvalue?=?0.0119). The mean mercaptopurine doses did not differ among patients with TPMT, ITPA, MRP4, and PACSIN2polymorphisms (Pvalue?=?0.9461, 0.5818, and 0.7951, respectively). After multivariable linear regression analysis, only NUDT15variants retained their clinically significant correlation with mercaptopurine intolerance (Pvalue < 0.0001). Conclusion: In this cohort, the major genetic determinant of mercaptopurine intolerance was NUDT15in Taiwanese patients. Impact:

The NUDT15variant is a stronger predictor of mercaptopurine intolerance than TPMTin a Taiwanese cohort. This finding is similar with studies performed on Asian populations rather than Caucasians.

Pre-emptive genotyping of the patients’ NUDT15before administering mercaptopurine may be more helpful than genotyping TPMTin Asians.

47. Design and Optimization of a Wearable Under-actuated Mechanism for Spinal Posture Measurement

Abstract

This paper proposes a novel wearable device to monitor and record the posture and alignment of spine. The proposed device adopts an underactuated mechanism design which allows it to adapt to the multiple-degrees-of-freedom spinal posture with minimum weight and complexity. To ensure the validity of measurement and comfort of wearing, the mechanism parameters are determined firstly by considering a special posture then are fine-tuned using an optimization algorithm so that uniform contact forces for several selected spinal postures can be achieved. Experiments demonstrate that the device can automatically maintain contact with the wearer's back and offer real-time spinal posture and alignment data for medical diagnosis and treatment.
