Abstract

Stellar-mass and supermassive black holes abound in the Universe, whereas intermediate-mass black holes (IMBHs) of ~10^2-10^5 solar masses in between are largely missing observationally, with few cases found only. Here we report the real-time discovery of a long-duration X-ray transient, EP240222a, accompanied by an optical flare with prominent H and He emission lines revealed by prompt follow-up observations. Its observed properties evidence an IMBH located unambiguously in the halo of a nearby galaxy and flaring by tidally disrupting a star -- the only confirmed off-nucleus IMBH-tidal disruption event so far. This work demonstrates the potential of sensitive time-domain X-ray surveys, complemented by timely multi-wavelength follow-ups, in probing IMBHs, their environments, demographics, origins and connections to stellar-mass and supermassive black holes., Comment: 64 pages, 15 figures, submitted

Abstract

Massive stars end their life as core-collapse supernovae, amongst which some extremes are Type Ic broad-lined supernovae associated with long-duration gamma-ray bursts (LGRBs) having powerful relativistic jets. Their less-extreme brethren make unsuccessful jets that are choked inside the stars, appearing as X-ray flashes or low-luminosity GRBs. On the other hand, there exists a population of extragalactic fast X-ray transients (EFXTs) with timescales ranging from seconds to thousands of seconds, whose origins remain obscure. Known sources that contribute to the observed EFXT population include the softer analogs of LGRBs, shock breakouts of supernovae, or unsuccessful jets. Here, we report the discovery of the bright X-ray transient EP240414a detected by the Einstein Probe (EP), which is associated with the Type Ic supernova SN 2024gsa at a redshift of 0.401. The X-ray emission evolution is characterised by a very soft energy spectrum peaking at < 1.3 keV, which makes it distinct from known LGRBs, X-ray flashes, or low-luminosity GRBs. Follow-up observations at optical and radio bands revealed the existence of a weak relativistic jet that interacts with an extended shell surrounding the progenitor star. Located on the outskirts of a massive galaxy, this event reveals a new population of explosions of Wolf-Rayet stars characterised by a less powerful engine that drives a successful but weak jet, possibly owing to a progenitor star with a smaller core angular momentum than in traditional LGRB progenitors., Comment: 43 pages, 9 figures, 4 tables, submitted. Abstract

Long gamma-ray bursts (GRBs) are believed to originate from core collapse of massive stars. High-redshift GRBs can probe the star formation and reionization history of the early universe, but their detection remains rare. Here we report the detection of a GRB triggered in the 0.5--4 keV band by the Wide-field X-ray Telescope (WXT) on board the Einstein Probe (EP) mission, designated as EP240315a, whose bright peak was also detected by the Swift Burst Alert Telescope and Konus-Wind through off-line analyses. At a redshift of $z=4.859$, EP240315a showed a much longer and more complicated light curve in the soft X-ray band than in gamma-rays. Benefiting from a large field-of-view ($\sim$3600 deg$^2$) and a high sensitivity, EP-WXT captured the earlier engine activation and extended late engine activity through a continuous detection. With a peak X-ray flux at the faint end of previously known high-$z$ GRBs, the detection of EP240315a demonstrates the great potential for EP to study the early universe via GRBs., Comment: 41 pages, 8 figures, 7 tables

Abstract

In the single-degenerate scenario of Type Ia supernovae (SNe Ia), the interaction between high-speed ejected material and the donor star in a binary system is expected to lead to mass being stripped from the donor. A series of multi-dimensional hydrodynamical simulations of ejecta-donor interaction have been performed in previous studies most of which adopt either a simplified analytical model or the W7 model to represent a normal SN Ia explosion. Whether different explosion mechanisms can significantly affect the results of ejecta-donor interaction is still unclear. In this work, we simulate hydrodynamical ejecta interactions with a main-sequence (MS) donor star in two dimensions for two near-Chandrasekhar-mass explosion models of SNe Ia, the W7 and N100 models. We find that about 0.30 and 0.37 Msun of hydrogen-rich material are stripped from a 2.5 Msun donor star in a 2 day orbit by the SN Ia explosion in simulations with the W7 deflagration and N100 delayed-detonation explosion model, respectively. The donor star receives a kick of about 74 and 86 km/s, respectively, in each case. The modal velocity, about 500 km/s, of stripped hydrogen-rich material in the N100 model is faster than the W7 model, with modal velocity of about 350km/s, by a factor 1.4. Based on our results, we conclude that the choice of near-Chandrasekhar-mass explosion model for normal SNe Ia seems to not significantly alter the ejecta-donor interaction for a given main-sequence donor model, at least in 2D., Comment: 11 pages, 8 figures, 1 table, accepted for publication in MNRAS

18. A Framework for Self-Regulated Digital Learning (SRDL)

Abstract

This article develops a framework for self-regulated digital learning, which supports for self-regulated learning (SRL) in e-learning systems. The framework emphasizes 8 features: learning plan, records/e-portfolio and sharing, evaluation, human feedback, machine feedback, visualization of goals/procedures/concepts, scaffolding, and agents. Each feature facilitates or supports one or more SRL skills, including planning, monitoring and evaluating learning, applying appropriate cognitive strategies, and setting standards of products or performance. The implementation in domain-general and -specific systems as illustrated by web-based inquiry and problem-solving are discussed. Examples and learning effects are elicited from the literature to demonstrate various designs. Approaches for designing SRL systems, educational implications, and new directions for future research incorporating SRL into digital learning are presented.

26. Assessment of tensile residual stress mitigation in Alloy 22 welds due to laser peening

Abstract

This paper examines the effects of laser peening on Alloy 22 (UNS N06022), which is the proposed material for use as the outer layer on the spent-fuel nuclear waste canisters to be stored at Yucca Mountain. Stress corrosion cracking (SCC) is a primary concern in the design of these canisters because tensile residual stresses will be left behind by the closure weld. Alloy 22 is a nickel-based material that is particularly resistant to corrosion; however there is a chance that stress corrosion cracking could develop given the right environmental conditions. Laser peening is an emerging surface treatment technology that has been identified as an effective tool for mitigating tensile redisual stresses in the storage canisters. The results of laser-peening experiments on Alloy 22 base material and a sample 33 mm thick double-V groove butt-weld made with gas tungsten arc welding (GTAW) are presented. Residual stress profiles were measured in Alloy 22 base material using the slitting method (also known as the crack-compliance method), and a full 2D map of longitudinal residual stress was measured in the sample welds using the contour method. Laser peening was found to produce compressive residual stress to a depth of 3.8 mm in 20 mm thick base material coupons. The depth of compressive residual stress was found to have a, significant dependence on the number of peening layers and a slight dependence on the level of irradiance. Abstract

Background & Aims: Bile salt export pump (BSEP) deficiency frequently necessitates liver transplantation in childhood. In contrast to two predicted protein truncating mutations (PPTMs), homozygous p.D482G or p.E297G mutations are associated with relatively mild phenotypes, responsive to surgical interruption of the enterohepatic circulation (siEHC). The phenotype of patients with a compound heterozygous genotype of one p.D482G or p.E297G mutation and one PPTM has remained unclear. We aimed to assess their genotype-phenotype relationship. Methods: From the NAPPED database, we selected patients with homozygous p.D482G or p.E297G mutations (BSEP1/1; n = 31), with one p.D482G or p.E297G, and one PPTM (BSEP1/3; n = 30), and with two PPTMs (BSEP3/3; n = 77). We compared clinical presentation, native liver survival (NLS), and the effect of siEHC on NLS. Results: The groups had a similar median age at presentation (0.7-1.3 years). Overall NLS at age 10 years was 21% in BSEP1/3 vs. 75% in BSEP1/1 and 23% in BSEP3/3 (p <0.001). Without siEHC, NLS in the BSEP1/3 group was similar to that in BSEP3/3, but considerably lower than in BSEP1/1 (at age 10 years: 38%, 30%, and 71%, respectively; p = 0.003). After siEHC, BSEP1/3 and BSEP3/3 were associated with similarly low NLS, while NLS was much higher in BSEP1/1 (10 years after siEHC, 27%, 14%, and 92%, respectively; p <0.001). Conclusions: Individuals with BSEP deficiency with one p.E297G or p.D482G mutation and one PPTM have a similarly severe disease course and low responsiveness to siEHC as those with two PPTMs. This identifies a considerable subgroup of patients who are unlikely to benefit from interruption of the enterohepatic circulation by either surgical or ileal bile acid transporter inhibitor treatment. Abstract

Background: Radiotherapy (RT) is a common treatment for prostate cancer, yet the risk of second primary colorectal cancer (SPCRC) in patients with prostate cancer undergoing RT has not been adequately studied. Methods: This study employed a population-based cohort design using the US Surveillance, Epidemiology, and End Results (SEER) database to identify individuals diagnosed between January 1975 and December 2015. The cumulative incidence of SPCRC was estimated using Fine–Gray competing risk regression. Poisson regression analysis was used to estimate the risk associated with RT. Survival outcomes of patients with SPCRC were evaluated using the Kaplan–Meier method. Results: A total of 287,607 patients diagnosed with prostate cancer were identified. The cumulative incidences were higher in patients who did not receive RT (2.00%) compared to those who underwent RT (2.47%) after 25 years. After adjustment for multiple variables, RT was associated with an increased risk of developing combined SPCRC (adjusted HR 1.590). Additionally, the overall survival was significantly lower in patients who developed colorectal cancer after receiving RT as compared to those who did not receive RT. Conclusion: These findings underscore the need for diligent long-term monitoring and effective management strategies to detect SPCRC in patients treated with RT for prostate cancer. 33. Age-related genes affecting the immune cell infiltration in ulcerative colitis revealed by weighted correlation network analysis and machine learning.

Abstract

OBJECTIVE: The crosstalk between age and immunity in the context of ulcerative colitis (UC) remains incompletely understood. Our objective is to elucidate the specific age-associated genetic factors that modulate immune cell infiltration in UC, with the aim of identifying innovative therapeutic targets for the treatment of this disease. MATERIALS AND METHODS: Potential batch effects between samples were removed by R package "inSilicoMerging". Unsupervised clustering analysis via the "ConsensusClusterPlus" R package was utilized to perform consensus molecular subtyping of immune subtypes in UC. The construction of a heat map was accomplished through the utilization of the R package "pheatmap", while functional enrichment analysis was executed by means of the Metascape database. The identification of the age-related gene module was achieved by performing weighted gene co-expression network analysis (WGCNA) analysis using the R package "WGCNA". The support vector machine (SVM), least absolute shrinkage and selector operation (LASSO), and random forest algorithms were performed via the "e1071", "glmnet" and "randomForest" packages in R, respectively. The diagnostic performance of the parameter was assessed using the receiver operating characteristic (ROC) curve. Correlation analysis was performed by Spearman correlation. The "XSum" package in R was employed to identify potential small-molecule drugs for UC utilizing the Connectivity Map (CMap) database. Molecular docking was performed with Autodock Vina molecular docking software. RESULTS: A significantly greater frequency of UC patients aged below 40 years was observed in the group with extensive disease extent as compared to those with non-extensive disease extent (70% vs. 47%; Chi-square test, p = 0.02). The application of unsupervised clustering analysis allowed for the stratification of UC patients into two distinct immune subtypes, namely cluster C1 and cluster C2. The distribution of immune subtypes was significantly different between different age categories (Chi-square test, p = 0.00219). The UC samples that were grouped under cluster C1 were distinguished by a higher abundance of macrophages and an elevated number of neutrophils relative to those in cluster C2. Based on both WGCNA and Limma analysis, 146 age-related genes were identified, which exhibited a predominant enrichment in the biological process of cellular senescence. Two age-related genes (MIDN, and PLD6) affecting the immune cell infiltration in UC were identified based on machine learning algorithms (SVM, LASSO, and random forest). The diagnostic performance of MIDN (AUC = 0.93) and PLD6 (AUC = 0.90) in discerning UC patients belonging to cluster C1 was found to be satisfactory, as demonstrated by ROC curve analysis. MIDN demonstrated a positive correlation (r = 0.50, p < 0.0001) with Neutrophil, while PLD6 exhibited a negative correlation (r = -0.52, p < 0.0001) with Neutrophil levels. The "XSum" algorithm revealed that Entinostat has therapeutic potential for UC. The docking glide score between Entinostat and MIDN, and PLD6 protein was -8.9 kcal/mol and -6.8 kcal/mol, respectively. CONCLUSIONS: We have identified two age-related genes, MIDN and PLD6, that are involved in immune cell infiltration in patients with ulcerative colitis. Furthermore, a small molecule drug (Entinostat) with potential therapeutic effects for UC was screened out. Abstract

OBJECTIVE: Due to the impact of excessive glucose on osteogenic differentiation, diabetic osteopathy frequently results in skeletal fragility, osteoporosis, and bone pain. Zoledronic acid, a bisphosphonate (BP) that effectively inhibits osteoclastic bone resorption is given yearly to improve bone mineral density (BMD) in patients with osteoporosis. However, the detailed molecular mechanisms remained unclear. This study investigates the possible pathways by which zoledronic acid regulates osteogenesis when blood glucose levels are high. MATERIALS AND METHODS: MC3T3-E1 cells were treated with one mM zoledronic acid or not in a standard or high glucose culture medium. A quantitative polymerase chain reaction (qPCR) assay was utilized to assess the expression of the target candidate genes, including RUNX2, MALAT1, miR-133, miR-20a, and miR-204. RESULTS: In a high-glucose condition, zoledronic acid treatment significantly lowered MALAT1 (p < 0.0001) and miR-20a (p < 0.0001) expression. Conversely, in a high-glucose condition, RUNX2, miR-133, and miR-204 expressions were found to be significantly increased in the zoledronic acid treatment group as compared to no treatment (all p < 0.0001). CONCLUSIONS: In conclusion, under a high-glucose environment, zoledronic acid can modulate the expression of the RUNX2 transcription factor through epigenetic regulation. [ABSTRACT FROM AUTHOR]

Abstract

While there is an international definition of social work as a profession, little is known about whether there is also a shared pedagogy in social work cross-nationally. To our knowledge, this paper is the first empirical study which aims to fill this gap by applying the concept of signature pedagogy in social work education to explore the commonality of social work pedagogy across countries. The study uses a multi-site case study (six universities in five European countries) through applying a ‘critical teacher-researcher’ approach in generating the data, followed by a two-phased thematic analysis. The study evidenced a shared principle of social work pedagogy which nurtures social work student to think and perform like a social worker and develop the professional self through developing relationships and dialogue, professional practice, group work, self-reflection and critical thinking. It is argued from, this exploratory study, that even between countries which have different welfare ideology as well as social work history and education systems, there is some common ground in social work pedagogy where one can learn from another through the use of ‘teacher as researcher’ methodologically., Det finns en internationell definition av socialt arbete som profession men vi vet mindre om huruvida det finns en gemensam pedagogik i hur socialt arbete lärs ut mellan länder. Utifrån vår kännedom är detta det första paper som genom en empirisk studie utforskar vad vi har gemensamt i hur vi lär ut socialt arbete. Studien använder en multi-site case study (sex universitet i fem europeiska länder) genom att använda ”critical teacher researcher” som metod för att samla in data som sedan följs av en tematisk analys i två steg. Studien visade ett gemensamt förhållningssätt i den pedagogik som lär ut socialt arbete där vi alla fokuserar på att ge studenterna en förmåga att tänka och verka som socialarbetare och utveckla det professionella ”jaget” genom att utveckla relationer och dialog, professionell praktik, grupparbete, självreflektion och kritiskt tänkande. Det finns, utifrån den här utforskande studien, grund för att påstå att även länder som inte har en gemensam välfärdsideologi, historik eller utbildningssystem, ändå har några gemensamma principer i pedagogiken i socialt arbete där vi kan lära från varandra genom att använda ”teacher as reasercher” metoden.
