Zn.sup.2+ and Cu.sup.2+ related to transition metal ions present in foods play essential roles in the health of human and animal metabolism. However, the excess of these ions contaminates foods and causes many kinds of diseases. In recent years, designing chemosensors for detection of the two targets has been widespread but is still a great challenge for practical application. In this study, by reducing expenses, simplifying analytical procedures, and enabling multifunctions, a functional chemosensor was proposed by using tetraphenylethylene derivative (H.sub.4tcbpe) which could selectively and sensitively response to Zn.sup.2+ and Cu.sup.2+. H.sub.4tcbpe can be used to detect Zn.sup.2+ with fluorescence enhancement based on aggregation induced emission effect, while fluorescence of H.sub.4tcbpe can be quenched by Cu.sup.2+ due to the disassembly of H.sub.4tcbpe aggregates in a solvent/nonsolvent binary mixture of EtOH and H.sub.2O. Fluorophotometry, UV-Vis, and SEM were used to characterize behavior of the fluorescence sensor. In this work, the detection in liquid solutions was conducted, and a fluorescent test pen for the assay of Zn.sup.2+ and Cu.sup.2+ on a piece of filter paper was fabricated. The proposed fluorescent test pen contained H.sub.4tcbpe as a 'fluorescent probe ink' and made it possible to write words on the area of a filter paper spiked with solutions with different concentrations of Zn.sup.2+ and Cu.sup.2+. The detection limits for Zn.sup.2+ and Cu.sup.2+ are 43 nM and 0.5 [mu]M, respectively. This fluorescent sensor was applied to sense ions in black fungus samples. Effective treatment against glioma remains challenging nowadays because the protective blood-brain barrier (BBB) impedes drug penetration into brain and the limited efficacy of conventional chemotherapy. While strong positively charged nanoparticles have good permeability through the BBB, they often come with the caveat of cationic toxicity to healthy tissues and organs during blood circulation. Here we show a neutrally charged nanoprobe with a surface decorated with γ-glutamyl moieties that can be cleaved by γ-glutamyl transpeptidase, an enzyme overexpressed on brain capillaries. Upon the cleavage, positively charged primary amines are generated, facilitating the effective crossing of the nanoprobe through BBB via the adsorption-mediated transcytosis pathway, while avoiding the caveat of cationic toxicity. In addition, when reaching the acidic tumor microenvironment, the nanoprobe co-encapsulating sonosensitizer and immune agonist swells, which results in an accelerated drug release under ultrasound irradiation to induce a combined immune response, ultimately leading to a robust anticancer effect. Overall, we report an effective drug delivery nanoplatform across the BBB for an enhanced therapy of glioma. Chemotherapy of glioma is limited by delivering drugs across blood-brain barrier (BBB) to tumor sites in brain. Immunotherapy has made great progress in various solid tumors. However, the "cold" tumor immune microenvironment of microsatellite stable subtype colorectal cancer (MSS‐CRC) hinders the effectiveness of immunotherapy. Therefore, reshaping the immunosuppressive microenvironment and initiating efficient antitumor immune responses are critical for immunotherapy of MSS‐CRC. According to the analysis of clinical samples, it is found that the levels of fat mass and obesity‐associated protein (FTO) and M2‐like tumor‐associated macrophages (TAMs) infiltration are significantly elevated in CRC tissue, which has driven one to construct a targeted cationic liposome to simultaneously enhance the RNA methylation and inhibit the CD47 immune checkpoint expression of tumor cells in the hope of promoting the M1‐like TAMs polarization and phagocytosis. By upregulating the m6A modification of tumor cells, the lactate secretion is decreased to promote the TAMs repolarized into M1‐like. Meanwhile, CD47 siRNA codelivered by the cationic liposomes downregulates the expression of immune checkpoint CD47 on the cancer cell surface, which enhances the phagocytic ability of the M1‐like TAMs. The combination treatment scheme is expected to provide a new option for treating MSS‐CRC, which may also be extended for treating other immunologically "cold" tumors.
