The multifaceted chemo-immune resistance is the principal barrier to achieving cure in cancer patients. Identifying a target that is critically involved in chemo-immune-resistance represents an attractive strategy to improve cancer treatment. iRhom1 plays a role in cancer cell proliferation and its expression is negatively correlated with immune cell infiltration. Here we show that iRhom1 decreases chemotherapy sensitivity by regulating the MAPK14-HSP27 axis. In addition, iRhom1 inhibits the cytotoxic T-cell response by reducing the stability of ERAP1 protein and the ERAP1-mediated antigen processing and presentation. To facilitate the therapeutic translation of these findings, we develop a biodegradable nanocarrier that is effective in codelivery of iRhom pre-siRNA (pre-siiRhom) and chemotherapeutic drugs. This nanocarrier is effective in tumor targeting and penetration through both enhanced permeability and retention effect and CD44-mediated transcytosis in tumor endothelial cells as well as tumor cells. Inhibition of iRhom1 further facilitates tumor targeting and uptake through inhibition of CD44 cleavage. Co-delivery of pre-siiRhom and a chemotherapy agent leads to enhanced antitumor efficacy and activated tumor immune microenvironment in multiple cancer models in female mice. Targeting iRhom1 together with chemotherapy could represent a strategy to overcome chemo-immune resistance in cancer treatment.

BackgroundGenome-wide association studies (GWAS) provide a powerful means to identify associations between genetic variants and phenotypes. However, GWAS techniques for detecting epistasis, the interactions between genetic variants associated with phenotypes, are still limited. We believe that developing an efficient and effective GWAS method to detect epistasis will be a key for discovering sophisticated pathogenesis, which is especially important for complex diseases such as Alzheimer's disease (AD).ResultsIn this regard, this study presents GenEpi, a computational package to uncover epistasis associated with phenotypes by the proposed machine learning approach. GenEpi identifies both within-gene and cross-gene epistasis through a two-stage modeling workflow. In both stages, GenEpi adopts two-element combinatorial encoding when producing features and constructs the prediction models by L1-regularized regression with stability selection. The simulated data showed that GenEpi outperforms other widely-used methods on detecting the ground-truth epistasis. As real data is concerned, this study uses AD as an example to reveal the capability of GenEpi in finding disease-related variants and variant interactions that show both biological meanings and predictive power.ConclusionsThe results on simulation data and AD demonstrated that GenEpi has the ability to detect the epistasis associated with phenotypes effectively and efficiently. The released package can be generalized to largely facilitate the studies of many complex diseases in the near future.

The orphan nuclear receptors estrogen-related receptors (ERRs) bind to the estrogen-related receptor response element (ERRE) to regulate transcriptional programs in cellular metabolism and cancer cell growth. In this study, we evaluated the potential for a pyrrole-imidazole polyamide to block ERRα binding to ERREs to inhibit gene expression. We demonstrated that the ERRE-targeted polyamide 1 blocked the binding of ERRα to the consensus ERRE and reduced the transcriptional activity of ERRα in cell culture. We further showed that inhibiting ERRα transcriptional activity with polyamide 1 led to reduced mitochondrial oxygen consumption, a primary biological effect regulated by ERRα. Finally, our data demonstrated that polyamide 1 is an inhibitor for cancer cell growth.

Further technological development of perovskite solar cells (PSCs) will require improvements in power conversion efficiency and stability, while maintaining low material costs and simple fabrication. In this Research Article, we describe top-illuminated ITO-free, stable PSCs featuring microcavity structures, wherein metal layers on both sides on the active layers exerted light interference effects in the active layer, potentially increasing the light path length inside the active layer. The optical constants (refractive index and extinction coefficient) of each layer in the PSC devices were measured, while the optical field intensity distribution was simulated using the transfer matrix method. The photocurrent densities of perovskite layers of various thicknesses were also simulated; these results mimic our experimental values exceptionally well. To modify the cavity electrode surface, we deposited a few nanometers of ultrathin MoO3 (2, 4, and 6 nm) in between the Ag and poly(3,4-ethylenedioxythiophene):polystyrenesulfonate (PEDOT:PSS) layers provide hydrophobicity to the Ag surface and elevate the work function of Ag to match that of the hole transport layer. We achieved a power conversion efficiency (PCE) of 13.54% without hysteresis in the device containing a 4 nm-thick layer of MoO3. In addition, we fabricated these devices on various cavity electrodes (Al, Ag, Au, Cu); those prepared using Cu and Au anodes displayed improved device stability of up to 72 days. Furthermore, we prepared flexible PSCs having a PCE of 12.81% after incorporating the microcavity structures onto poly(ethylene terephthalate) as the substrate. These flexible solar cells displayed excellent stability against bending deformation, maintaining greater than 94% stability after 1000 bending cycles and greater than 85% after 2500 bending cycles performed with a bending radius of 5 mm.

In recent years, the display information of head-up displays for vehicles has gradually developed from single-depth to multi-depth. To reduce the workload of driving and the number of eye adjustments, researchers use the visual perception of human eyes to realize the image information integrated with the real world. In this study, HoloLens2 is used to demonstrate head-up displays of different depths. An electroencephalogram, an electro-ophthalmogram, and a NASA-TLX questionnaire were used to evaluate the fatigue of drivers during long-term driving. The development of creativity plays a decisive role in the future development of human life as it stimulates divergent thinking and grants the critical ability to innovate and solve problems. Therefore, the question of how to encourage students' creativity has also attracted the attention of related research in various fields. Our study used the Scratch visual programming tool to allow students to create their own digital games. We used different technological devices as external stimuli during the research process to stimulate students' creative ideas. We provided four control devices: a keyboard and mouse, PicoBoard, a 65-inch touch screen, and a Wii remote control for 92 fifth-grade students in four S-shaped classes. After a 12-week experimental process, students designed their original Scratch game. The results of the study found that differences in device properties correspond to different items of creativity: devices highly relevant to students' life experiences improve the flexibility and elaboration of creativity. Innovative controls promote creative originality. The freedom of device control can increase the fluency of creativity. Therefore, providing control devices with different properties will allow teachers to establish learning environments that foster creativity. Identifying accurate biomarkers of cognitive decline is essential for advancing early diagnosis and prevention therapies in Alzheimer's disease. The Alzheimer's disease DREAM Challenge was designed as a computational crowdsourced project to benchmark the current state-of-the-art in predicting cognitive outcomes in Alzheimer's disease based on high dimensional, publicly available genetic and structural imaging data. This meta-analysis failed to identify a meaningful predictor developed from either data modality, suggesting that alternate approaches should be considered for prediction of cognitive performance.
