Abstract

Continued efforts are made on the development of earth-abundant metal catalysts for dehydrogenation/hydrolysis of amine boranes. In this study, complex [K-18-crown-6-ether][(NO)2Fe(μ-MePyr)(μ-CO)Fe(NO)2] (3-K-crown, MePyr = 3-methylpyrazolate) was explored as a pre-catalyst for the dehydrogenation of dimethylamine borane (DMAB). Upon evolution of H2(g)from DMAB triggered by 3-K-crown, parallel conversion of 3-K-crowninto [(NO)2Fe(N,N′-MePyrBH2NMe2)]−(5) and an iron-hydride intermediate [(NO)2(CO)Fe(μ-H)Fe(CO)(NO)2]−(A) was evidenced by X-ray diffraction/nuclear magnetic resonance/infrared/nuclear resonance vibrational spectroscopy experiments and supported by density functional theory calculations. Subsequent transformation of Ainto complex [(NO)2Fe(μ-CO)2Fe(NO)2]−(6) is synchronized with the deactivated generation of H2(g). Through reaction of complex [Na-18-crown-6-ether][(NO)2Fe(η2-BH4)] (4-Na-crown) with CO(g)as an alternative synthetic route, isolated intermediate [Na-18-crown-6-ether][(NO)2(CO)Fe(μ-H)Fe(CO)(NO)2] (A-Na-crown) featuring catalytic reactivity toward dehydrogenation of DMAB supports a substrate-gated transformation of a pre-catalyst [(NO)2Fe(μ-MePyr)(μ-CO)Fe(NO)2]−(3) into the iron-hydride species Aas an intermediate during the generation of H2(g).

Abstract

We previously developed pluripotent rabbit embryonic stem cells (rbES) using a culture system supplemented with basic fibroblast growth factor (bFGF) and leukemia inhibitory factor (LIF), noggin and Y-27632 (referred to as iFLY). In present work, we explored multiple approaches to enhance the chance of deriving domed pluripotent rbES cells by inhibition of MEK, GSK, and PKC signaling pathways. Domed stated rbES were derived in defined medium supplemented with 15% KOSR, 10(3) IU/mL mouse LIF, 10 ng/mL bFGF and three inhibitors to the MEK (PD0325901, 1 µM), GSK3 (CHIR99021, 3 µM) and PKC (Gö6983, 5 µM) (3i). Domed rbES were passaged every 3-4 days till passage 3-4 for the designated experiments. We showed that bFGF and LIF are indispensable for the derivation and maintenance of rbES; whereas the 3i medium containing inhibitors to the MEK (PD0325901), GSK3 (CHIR99021) and PKC (Gö6983) were necessary for deriving domed rbES. Domed rbES possessed naïve ES markers as Rex1 and ERAS in addition to Oct4, Klf4, Sox 2 and c-myc by RT-PCR. Domed rbES showed positive staining for Rex1, Fgf4, Klf4, Nanog and Oct4 by immunofluorescence chemistry. Further deleting either one factor in 3i medium as CHIR99021, PD0325901, Gö6983 or bFGF resulted in disappearing of domed rbES colonies. The optimal concentrations of 3i contained 0.75 µM PD0325901, 2.25 µM CHIR99021, and 4.5 µM Gö6983. Our work, in combination of different inhibitors for deriving rabbit ES, supports that the network of signal pathways plays an important role in ES self-renew, propagation and maintenance, and sheds light on deriving authentic properties of rbES in an important yet understudied model animal species.

Abstract

Liver cancer is one of the most common malignant tumors globally and is the second leading cause of cancer deaths in Taiwan. The activity of α‐fucosidase (AFU) in the human body has been confirmed by many clinical studies and experiments to be applicable in the early diagnosis of primary liver cancer. Herein, an electrochemical method for determining AFU activity was developed based on measuring p‐nitrophenol (pNP), which is a product of the reaction between AFU and p‐nitrophenyl‐α‐L‐fucopyranoside. The electrochemical detection of pNP in serum samples exhibited a wide linear range (2.5–250 μM), which corresponds to approximately 2.5–250 U/L AFU. The recovery of AFU measured upon enzyme addition to standard serum was 97–110%. Moreover, no potential interferants had a significant effect on pNP quantification. In contrast, an optical absorption method was subject to interference by the serum color. Further, the linear range for pNP was only 2.5–50 μM and the recovery of AFU was 165–219%. The performance of this absorption method could lead to a misjudgment of the disease course. Abstract

Chloramphenicol (CAP) is a veterinary antibiotic that has been banned due to its severe side effects in humans. Through the application of manure, veterinary antibiotics can enter the soil, where they can be taken up by crops and vegetables and pose a potential health hazard to humans. Thus, it is highly desirable to develop a rapid and sensitive tool for on‐site detection of CAP to ensure food safety and to control the abuse of antibiotics. To this end, nitrogen‐doped graphene quantum dots (N‐GQDs) were successfully prepared via microwave‐assisted synthesis using citric acid and urea as carbon and nitrogen sources, respectively. Analytical results suggested that the interaction between N‐GQDs and CAP could occurs via π‐π stacking, which quenched N‐GQD fluorescence. CAP spiked into chicken feed could be rapidly extracted with ethanol and quantified based on N‐GQD fluorescence quenching without further separation. This method showed good recovery (97–102.6%), a low detection limit (1.8 ppm), and was not affected by interference from florfenicol, and thiamphenicol, legal substitute antibiotics. Abstract

This research resolves an inconsistency problem that arises from assessing circularity of workpiece measured by coordinate measuring machines (CMMs). Although the notion of circularity is employed to constrain two-dimensional circular features, in practice the measured points are obtained in a three-dimensional space and are in general not in the same plane, let alone being in a 'perpendicular' cross-section. All of the algorithms currently used for assessing circularity deal with data in a single plane that is perpendicular to the axis of a cylindrical feature from which the circular feature is extracted. This discrepancy causes the assessed circularity significantly departing from the actual circularity and resulting in rejection of in-tolerance parts. This research designs a compensation procedure for deriving two-dimensional data from three-dimensional biased measured points. The circularity is assessed based on the compensated two-dimensional data points. The assessment results with the compensation are compared with those without compensation. A program containing a variety of implementations of form fitting algorithms is developed and used to illustrate the improvement on the accuracy of assessment. [DOI: 10.1115/1.1312336]
