Abstract

Global warming is causing profound modifications of aquatic ecosystems and one major outcome appears to be a decline in adult size of many fish species. Over the last decade, sardine populations in the Gulf of Lions (NW Mediterranean Sea) have shown severe declines in body size and condition as well as disappearance of the oldest individuals, which could not be related to overfishing, predation pressure or epizootic diseases. In this study, we investigated whether this situation reflects a bottom-up phenomenon caused by reduced size and availability of prey that could lead to energetic constraints. We fed captive sardines with food items of two different sizes eliciting a change in feeding mode (filter-feeding on small items and directly capturing larger ones) at two different rations for several months, and then assessed their muscle bioenergetics to test for changes in cellular function. Feeding on smaller items was associated with a decline in body condition, even at high ration, and almost completely inhibited growth by comparison to sardines fed large items at high ration. Sardines fed on small items presented specific mitochondrial adjustments for energy sparing, indicating a major bioenergetic challenge. Moreover, mitochondria from sardines in poor condition had low basal oxidative activity but high efficiency of ATP production. Notably, when body condition was below a threshold value of 1.07, close to the mean observed in the wild, it was directly correlated with basal mitochondrial activity in muscle. The results show a link between whole-animal condition and cellular bioenergetics in the sardine, and reveal physiological consequences of a shift in feeding mode. They demonstrate that filter-feeding on small prey leads to poor growth, even under abundant food and an increase in the efficiency of ATP production. These findings may partially explain the declines in sardine size and condition observed in the wild.Le changement global entraîne de profondes modifications des écosystèmes aquatiques, l'une des principales étant le déclin de la taille des adultes chez de nombreuses espèces de poissons. Au cours de la dernière décennie, les populations de sardines du Golfe du Lion (Nord-Ouest de la Méditerranée) ont montré une importante diminution de leur taille et de leur condition corporelle ainsi qu'une disparition des individus les plus âgés, qui n'ont pas pu être liées à la surpêche, à la pression de prédation ou aux épizooties. Dans cette étude, nous avons cherché à savoir si cette situation reflète un phénomène ascendant causé par la réduction de la taille et de la disponibilité des proies qui pourrait entraîner des contraintes énergétiques chez la sardine. Nous avons ainsi nourri des sardines captives avec des granulés de deux tailles différentes provoquant un changement de mode d'alimentation (filtration des petits granulés et capture directe des plus gros) et à deux rations différentes pendant plusieurs mois, puis nous avons évalué leur bioénergétique musculaire pour tester les changements au niveau de leur fonction cellulaire. L’alimentation à base de petits granulés a été associée à un déclin de la condition corporelle, même à une ration élevée, et à une croissance quasiment inhibée par rapport aux sardines nourries avec des plus gros granulés à une ration élevée. Les sardines nourries avec des petits granulés ont également présenté des ajustements mitochondriaux spécifiques pour économiser de l’énergie, indiquant un défi bioénergétique majeur. De plus, les mitochondries des sardines en mauvaise condition présentaient une faible activité oxydative basale, mais une efficacité élevée de production d'ATP. Notamment, lorsque la condition corporelle était inférieure à une valeur seuil de 1,07, proche de la moyenne observée dans la nature, elle était directement corrélée à l'activité mitochondriale basale dans le muscle. Ces résultats montrent un lien entre la condition de l'animal entier et la bioénergétique cellulaire chez la sardine, et révèlent les conséquences physiologiques d'un changement de mode d'alimentation. Ils démontrent que le nourrissage via la filtration de petites proies entraîne une faible croissance, même en cas de nourriture abondante, et une augmentation de l'efficacité de la production d'ATP. Ces résultats peuvent expliquer en partie le déclin de la taille et de la condition des sardines observé dans la nature.

Abstract

Background Upon maturation in the bone marrow, polyploid megakaryocytes elongate very long and thin cytoplasmic branches called proplatelets. Proplatelets enter the sinusoids blood vessels in which platelets are ultimately released. Microtubule dynamics, bundling, sliding, and coiling, drive these dramatic morphological changes whose regulation remains poorly understood. Microtubule properties are defined by tubulin isotype composition and post-translational modification patterns. It remains unknown whether microtubule post-translational modifications occur in proplatelets and if so, whether they contribute to platelet formation. Results Here, we show that in proplatelets from mouse megakaryocytes, microtubules are both acetylated and polyglutamylated. To bypass the difficulties of working with differentiating megakaryocytes, we used a cell model that allowed us to test the functions of these modifications. First, we show that α2bβ3integrin signaling in D723H cells is sufficient to induce β1tubulin expression and recapitulate the specific microtubule behaviors observed during proplatelet elongation and platelet release. Using this model, we found that microtubule acetylation and polyglutamylation occur with different spatio-temporal patterns. We demonstrate that microtubule acetylation, polyglutamylation, and β1tubulin expression are mandatory for proplatelet-like elongation, swelling formation, and cytoplast severing. We discuss the functional importance of polyglutamylation of β1tubulin-containing microtubules for their efficient bundling and coiling during platelet formation. Conclusions We characterized and validated a powerful cell model to address microtubule behavior in mature megakaryocytes, which allowed us to demonstrate the functional importance of microtubule acetylation and polyglutamylation for platelet release. Furthermore, we bring evidence of a link between the expression of a specific tubulin isotype, the occurrence of microtubule post-translational modifications, and the acquisition of specific microtubule behaviors. Thus, our findings could widen the current view of the regulation of microtubule behavior in cells such as osteoclasts, spermatozoa, and neurons, which express distinct tubulin isotypes and display specific microtubule activities during differentiation. Abstract

Although individual electronic tagging using PIT (Passive Integrated Transponder) tags is well established, it is mainly used for fish > 60 mm in length. Since electronic tagging is an ideal identification method, we used RFID (Radio Frequency Identification) microtags (6 mm in length, 1 mm in diameter, 10 mg in mass) to characterize individual fish from the early stages of their development and throughout their lifecycles. We used sea bass, Dicentrarchus labrax (105 day-old fish weighing between 100 and 1100 mg), and studied the effects of intra-coelomic tagging in half the population using different endpoints including survival and tag reading, growth over 6 months and swimming responses. Dead fish were counted daily, biometric data were collected at 21 to 28 days intervals and fish were sexed at the end of the experiment. Behavioral swimming responses following a sudden dark challenge were evaluated after the first three biometric measurements (immediately after tagging, and then 21 and 42 days later). After 6 months, mean survival was 69%, the smallest surviving fish weighed 197 mg at tagging, and success in tag reading was 79% for the size class 300-400 mg after 63 days. No negative effects were observed on growth and most deaths occurred within 15 days after tagging with a first peak after 5 days but that did not particularly affect the smallest individuals. Differences in swimming responses were detected between tagged fish and untagged controls immediately after the surgical procedure with tagged fish swimming more than controls for 45 min. However, after 21 days, the tagged fish swam less than the untagged controls after being placed in the observation tank for 1 h and 45 min. At 42 days post-tagging, no further differences were observed. Overall, results suggested that the tagging method was suitable for fish as small as 400 mg and 36 mm in total length. This type of early tagging method is invaluable for the longitudinal monitoring of individual biological traits (e.g., growth) or for repeated assays with the same individual at distinct time points (e. g., behavior studies). We used it for a first evaluation of early growth differentiation between sexes and demonstrated a 31% mass gain difference in females that was visible as soon as the fish reached 105 days old. This confirms the potential of the method for the selection of early-expressed character traits which could lead to rearing cost savings for the aquaculture research field. (C) 2014 Elsevier B.V. Abstract

Replacing aquaculture feeds based on fisheries-derived resources with plant-based diets could be a relevant strategy to improve the sustainability of aquaculture. Recent studies on sea bass have shown that the total and early replacement of marine products by plant products would have a moderate effect on fish growth and body lipid content. Whether a plant-based diet impacts behavioural and physiological traits possibly linked to fish welfare, is not known, however. Here, we studied the effect of a totally plant-based diet introduced at an early stage of sea bass development on self-feeding behaviour, learning ability in a T-maze and stress biomarkers. We first compared learning processes in self-feeding conditions, between naive fish fed a plant-based diet (PBF) and fish fed a classic marine diet (MF). Then, we tested fish individually in a T-maze to compare the two feed groups for swimming activity, exploration and the ability to learn to discriminate between two two-dimensional objects associated with a reward. Blood physiological variables, including stress indicators (cortisol and glucose concentrations), were also determined. We did not find any indications of differences in self-feeding behaviour between PBF and MF in the first 30 days. A second experiment showed similar swimming activities in both fish categories. The "no-choice" percentage was high in both fish categories (similar to 60%), but all the fish moved preferentially toward the reward. Their first turns indicated an ability to discriminate between two two-dimensional objects to complete a simple task. However, the high percentage of "no-choice" responses in both fish categories could have rendered the results non significant. The T-maze test procedure induced the production of high concentrations of cortisol, indicating acute stress in fish of both groups during testing. Plasma cortisol concentration was higher in MF than PBF, suggesting that the plant-based diet may affect the short-term release of cortisol. This study provides the first insight into the impact of a plant-based diet on sea bass behavioural traits, and confirms the effect of this diet on cortisol release in response to stress. Overall, in this first experiment, we did not find any major impact of a plant-based diet on sea bass behavioural traits, which is an interesting point for the development potential of such a sustainable aquaculture strategy.

Chantier qualité GA; International audience; We estimated the heritability of growth-related traits (weight and length at ages one summer, first spring and two summers) in a synthetic mirror carp strain (HSM) in the Czech Republic. The four generation pedigree was obtained from parentage assignment of three factorial mating designs with microsatellite markers, and included 195 fish without phenotypes (48 G0, 147 G1) and 1321 fish with phenotypes (674 G2, 647 G3). Animal model heritability estimates over generations were in the range 0.21–0.33 for length and in the range 0.31–0.44 for weight. The genetic correlation between length and weight was high (0.97). The correlations between growth measurements in the first and in the second summer of age were moderate to low (0.34–0.67). Divergent selection for length at two summers of age was performed on G2 fish, and response to selection was evaluated by comparing the offspring of the selected sires in G3, in a communal test where genetic groups were identified by microsatellite parentage assignment. The response to upwards selection was moderate and indicated a realized heritability value of 0.24 to 0.34. No response to downwards selection was observed. We conclude that although selection for growth seems to be feasible in common carp, it would be a long-term process before results are visible.

Abstract

Little is known about the genetic basis of residual feed intake (RFI) variation in fish, since this trait is highly sensitive to environmental influences, and feed intake of individuals is difficult to measure accurately. The purpose of this work was (i) to assess the genetic variability of RFI estimated by an X-ray technique and (ii) to develop predictive criteria for RFI. Two predictive criteria were tested: loss of body weight during feed deprivation and compensatory growth during re-feeding. Ten heterozygous rainbow trout clones were used. Individual intake and body weight were measured three times at three-week intervals. Then, individual body weight was recorded after two cycles of a three-week feed deprivation followed by a three-week re-feeding. The ratio of the genetic variance to the phenotypic variance was found high to moderate for growth, feed intake, and RFI (VG/VP = 0.63 +/- 0.11, 0.29 +/- 0.11, 0.29 +/- 0.09, respectively). The index that integrates performances achieved during deprivation and re-feeding periods explained 59% of RFI variations. These results provide a basis for further studies on the origin of RFI differences and show that indirect criteria are good candidates for future selective breeding programs.

Abstract

253 full-sib families from 33 males and 23 females of European seabass were produced in a partly factorial mating design. All fish were reared in the same tank during 14 months, then 7000 of them were dispatched in four farms to different locations (France, Israel, Italy, Portugal) representing a wide variety of environmental conditions. Around 400 g mean weight, 1177 to 1667 fish in each site were weighed and length was measured. Condition factor (K) was calculated. Pedigrees were redrawn a posteriori using microsatellites markers: parental origin could be retraced for 99.2% of fish. Due to a high incidence of deformities, the useful sample size was reduced to 491-670 fish per site. Maternal effects were small. Using a simple animal model, heritability of weight ranged from 0.38 +/- 0.14 to 0.44 +/- 0.14 in the different sites. Length was highly correlated to weight, with similar heritabilities. GxE interaction, estimated through genetic correlations of weight across the different environments ranged from 0.70 +/- 0.10 to 0.99 +/- 0.05. National audience; In order to improve Our understanding of factors determining egg quality in fish, seabass egg batches obtained from 34 females held under standard conditions, were assessed for biological performances and a fraction from each batch was subjected to proteomic analysis. Comparison of proteomic patterns from eggs of different quality revealed differential protein spots among which one was identified as Nucleoside Diphosphate Kinase, a highly conserved protein involved in cell proliferation. This study shows that molecular markers may help understand factors determining egg quality in fish.

Abstract

National audience; Proportions of offspring from five common carp contributing to a sperm pool composed of equalized sperm concentrations (N-progeny) or equal sperm volumes (V-progeny) were each compared to a uniform distribution. Four microsatellite markers (MFW1, MFW6, MFW7, MFW28) were used to determine the paternity of the progeny. The homogeneity of offspring numbers from each male for the two types of progeny, were tested using an exact test for the likelihood-ratio chi-square. Numbers of offspring in the progeny groups were highly variable (0.4–50% in V-progeny, 2.4–41.2% in N-progeny) and highly significantly different as shown by Pearson chi-square statistics (χ2 = 189, 4 df, P < 0.0001 in V-progeny and χ2 = 139, 4 df, P < 0.0001 in N-progeny). Significant heterogeneity between treatments (P < 0.05) together with reduction of χ2 value from 189 to 139 showed that equalization of sperm concentration reduced heterogeneity in numbers of offspring. Number of sperm per male, sperm motility (71–98%), sperm velocity (97–155 μm s− 1) and control hatching rates (81–91%) all affected the observed number of offspring sired by each of the males, but the high variability in proportion of progeny among the 5 males remained unexplained.

Abstract

International audience; In aquaculture, deformities occur in all types of rearing systems, and are always a problem as they reduce the marketing value of fish. Deformities may have numerous origins, among which genetics is usually considered marginal (see review Alfonso, Montero, Rabaina, Astorga, Izquierdo & Gines 2000), although in some cases it appears to be significant (Alfonso et al. 2000; Gjerde, Pante & Baeverfjord 2005). In common carp, deformities also occur under practical conditions, and despite the general knowledge, genetics is often implicated as being a plausible cause by the farmers. One reason for this may be that commercial lines of common carp are often propagated with limited numbers of broodfish, and are often suspected to be quite inbred (e.g. Kohlmann, Gross, Murakaeva & Kersten 2003). In addition, Kirpichnikov (1981) has shown that several fin abnormalities could be attributed to genetics in this species. During the course of a selective breeding experiment on common carp under the semi-intensive pond conditions of Central Europe, we observed the appearance of a significant proportion of deformed fish, and so we decided to investigate the possibility of whether these could be of the genetic origin.

Abstract

We characterised, for the first-time, the sound production of black-chinned tilapia Sarotherodon melanotheron and show differences with that of Nile tilapia Oreochromis niloticus in a hybridization pairing context. Although both species were able to produce drum sounds, they showed different acoustic features. Drum sounds were produced in aggressive (chasing or lateral attack) and non-aggressive (courtship) contexts by O. niloticus but only in aggressive situations (fleeing or avoidance) by S. melanotheron. The second type of sounds produced by O. niloticus were grunts, produced in both aggressive (chasing and after biting) and non-aggressive contexts (nest building). The second type of sound produced by S. melanotheron was a rolling sound, produced only during courtship. Each species was able to produce common sounds (drum) and species-specific sounds (grunts and rolling). This implies that species can communicate without being able to understand each other because the sounds emitted may probably have different significance. Drumming corresponded only to aggressivity in S. melanotheron, whereas this was not true for O. niloticus. 11-ketotestosterone (11-kt) levels were significantly higher in male O. niloticus than male S. melanotheron, but there was no significant correlation between 11-kt or estradiol concentrations and the number of sounds produced in aggressive or non-aggressive behavioural contexts in either species. During interspecies interactions, O. niloticus drum sounds are likely considered to be aggressive by S. melanotheron and could potentially constitute a reproductive barrier between the two species., (Copyright © 2020 Elsevier GmbH. Abstract

Mature megakaryocytes extend long processes called proplatelets from which platelets are released in the blood stream. The Rho GTPases Cdc42 and Rac as well as their downstream target, p21-activated kinase 2 (PAK2), have been demonstrated to be important for platelet formation. Here we address the role, during platelet formation, of PAK1, another target of the Rho GTPases. PAK1 decorates the bundled microtubules (MTs) of megakaryocyte proplatelets. Using a validated cell model which recapitulates proplatelet formation, elongation and platelet release, we show that lack of PAK1 activity increases the number of proplatelets but restrains their elongation. Moreover, in the absence of PAK1 activity, cells have hyperacetylated MTs and lose their MT network integrity. Using inhibitors of the tubulin deacetylase HDAC6, we demonstrate that abnormally high levels of MT acetylation are not sufficient to increase the number of proplatelets but cause loss of MT integrity. Taken together with our previous demonstration that MT acetylation is required for proplatelet formation, our data reveal that MT acetylation levels need to be tightly regulated during proplatelet formation. We identify PAK1 as a direct regulator of the MT acetylation levels during this process as we found that PAK1 phosphorylates the MT acetyltransferase MEC-17 and inhibits its activity.

Background: Most fish breeding programs aim at improving growth rate and include feed conversion ratio (FCR) neither in the breeding goal nor in the selection index, although decreasing FCR is known to increase farm profit and decrease environmental impacts. This is because FCR is difficult to measure in fish that live in groups and FCR is assumed to have a favourable (negative) genetic correlation with growth, although the magnitude of this correlation is unknown. We investigated the effect of the genetic correlation between growth and FCR on the economic and environmental responses of a two-trait breeding goal (growth and FCR), compared to a single-trait breeding goal (growth only). Next, we evaluated the weights to assign to growth and FCR in a two-trait breeding goal to maximize sustainability of fish production., Methods: We used pseudo-best linear unbiased prediction (BLUP) index calculations to simulate a breeding program for sea bass. For the single-trait breeding goal, the trait in the breeding goal and in the index was thermal growth coefficient (TGC) and for the two-trait breeding goal, the traits in the breeding goal were TGC and FCR and the traits in the index were TGC and percentage of fat in the dorsal muscle (an indirect measure of FCR). We simulated responses to selection for genetic and phenotypic correlations between TGC and FCR ranging from 0 to - 0.8. Then, in the two-trait breeding goal, we calculated the economic return and the change in eutrophication when using economic values (EV) or environmental values (ENV)., Results: When the genetic correlation between TGC and FCR was lower than - 0.45, we found major differences in economic returns and in eutrophication between single and two-trait breeding programs. At a correlation of - 0.25, the two-trait breeding goal based on EV increased economic return by 25% compared to the single-trait breeding goal, while using ENV decreased eutrophication by 1.34% per ton of fish produced after one generation of selection., Conclusions: The genetic correlation between TGC and FCR affects the magnitude of economic losses due to omitting FCR in the breeding program. In addition, the genetic correlation affects the importance of choosing EV or ENV to reduce eutrophication and increase profit.

Background: Improving feed efficiency in fish is crucial at the economic, social and environmental levels with respect to developing a more sustainable aquaculture. The important contribution of genetic improvement to achieve this goal has been hampered by the lack of accurate basic information on the genetic parameters of feed efficiency in fish. We used video assessment of feed intake on individual fish reared in groups to estimate the genetic parameters of six growth traits, feed intake, feed conversion ratio (FCR) and residual feed intake in 40 pedigreed families of the GIFT strain of Nile tilapia, Oreochromis niloticus. Feed intake and growth were measured on juvenile fish (22.4 g mean body weight) during 13 consecutive meals, representing 7 days of measurements. We used these data to estimate the FCR response to different selection criteria to assess the potential of genetics as a means of increasing FCR in tilapia., Results: Our results demonstrate genetic control for FCR in tilapia, with a heritability estimate of 0.32 ± 0.11. Response to selection estimates showed FCR could be efficiently improved by selective breeding. Due to low genetic correlations, selection for growth traits would not improve FCR. However, weight loss at fasting has a high genetic correlation with FCR (0.80 ± 0.25) and a moderate heritability (0.23), and could be an easy to measure and efficient criterion to improve FCR by selective breeding in tilapia., Conclusion: At this age, FCR is genetically determined in Nile tilapia. A selective breeding program could be possible and could help enabling the development of a more sustainable aquaculture production.

Background: Upon maturation in the bone marrow, polyploid megakaryocytes elongate very long and thin cytoplasmic branches called proplatelets. Proplatelets enter the sinusoids blood vessels in which platelets are ultimately released. Microtubule dynamics, bundling, sliding, and coiling, drive these dramatic morphological changes whose regulation remains poorly understood. Microtubule properties are defined by tubulin isotype composition and post-translational modification patterns. It remains unknown whether microtubule post-translational modifications occur in proplatelets and if so, whether they contribute to platelet formation., Results: Here, we show that in proplatelets from mouse megakaryocytes, microtubules are both acetylated and polyglutamylated. To bypass the difficulties of working with differentiating megakaryocytes, we used a cell model that allowed us to test the functions of these modifications. First, we show that α2bβ3integrin signaling in D723H cells is sufficient to induce β1tubulin expression and recapitulate the specific microtubule behaviors observed during proplatelet elongation and platelet release. Using this model, we found that microtubule acetylation and polyglutamylation occur with different spatio-temporal patterns. We demonstrate that microtubule acetylation, polyglutamylation, and β1tubulin expression are mandatory for proplatelet-like elongation, swelling formation, and cytoplast severing. We discuss the functional importance of polyglutamylation of β1tubulin-containing microtubules for their efficient bundling and coiling during platelet formation., Conclusions: We characterized and validated a powerful cell model to address microtubule behavior in mature megakaryocytes, which allowed us to demonstrate the functional importance of microtubule acetylation and polyglutamylation for platelet release. Furthermore, we bring evidence of a link between the expression of a specific tubulin isotype, the occurrence of microtubule post-translational modifications, and the acquisition of specific microtubule behaviors. Thus, our findings could widen the current view of the regulation of microtubule behavior in cells such as osteoclasts, spermatozoa, and neurons, which express distinct tubulin isotypes and display specific microtubule activities during differentiation.

Toxicity of polyethylene microplastics (PE-MP) of size ranges similar to their natural food to zooplanktonic organisms representative of the main taxa present in marine plankton, including rotifers, copepods, bivalves, echinoderms and fish, was evaluated. Early life stages (ELS) were prioritized as testing models in order to maximize sensitivity. Treatments included particles spiked with benzophenone-3 (BP-3), a hydrophobic organic chemical used in cosmetics with direct input in coastal areas. Despite documented ingestion of both virgin and BP-3 spiked microplastics no acute toxicity was found at loads orders of magnitude above environmentally relevant concentrations on any of the invertebrate models. In fish tests some effects, including premature or reduced hatching, were observed after 12 d exposure at 10 mg L -1 of BP-3 spiked PE-MP. The results obtained do not support environmentally relevant risk of microplastics on marine zooplankton. Similar approaches testing more hydrophobic chemicals with higher acute toxicity are needed before these conclusions could be extended to other organic pollutants common in marine ecosystems. This study aims to investigate the effects of ammonia nitrogen loading rates and salinity levels on nutrients removal rates and substrate enzyme activities of constructed wetland (CW) microcosms planted with Salicornia bigelovii treating mariculture wastewater. Activities of urease (UA), dehydrogenase (DA), protease (PrA) and phosphatase (PA) were considered. Using principal component analysis (PCA), nutrient removal index (NRI) and enzyme activity index (EAI) were developed to evaluate the effects. The results revealed that increasing ammonia nitrogen loading rates had positive effects on nitrogen removal rates (i.e. NH 4 -N and DIN) and enhanced substrate enzyme activities. Compared with low salinity (i.e. 15 and 22), high salinity levels (i.e. 29 and 36) enhanced nutrients removal rates, DA and UA, but weaken PA and PrA. Abstract

Fish learning and cognition are usually approached by testing single individuals in various devices such as mazes that have serious drawbacks, especially in gregarious species, including the stress induced by the test procedure. This might impair the results and lead to misinterpretation about the learning abilities of the targeted species. In order to provide an alternative to the individual-based tests, we investigated for the first time the operant conditioning of four similar groups (50 individuals per tank) of sea bass. We used two computerized self-feeder devices per tank, each coupled with individual electronic identification and that were alternately activated during varying positive appetitive reinforcement period of time (7 to 1 day). Learning abilities were examined at both group and individual levels. At the group level, the operant conditioning was demonstrated as the triggering activity significantly decreased when the device was turned off and increased when it was turned on, whatever the reinforcement period duration. The individual level analysis revealed a more complex situation with fish showing different learning performances that can be best explained through the producer-scrounger game theory.

Abstract

In sea cage fish farming, production quotas aim to constrain the impact of fish farming on the surrounding ecosystem. It is unknown how these quotas affect economic profitability and environmental impact of genetic improvement. We combined bioeconomic modelling with life cycle assessment (LCA) to calculate the economic (EV) and environmental (ENV) values of thermal growth coefficient (TGC) and feed conversion ratio (FCR) of sea bass reared in sea cages, given four types of quota commonly used in Europe: annual production (Qprod), annual feed distributed (Qannual&#95;feed), standing stock (Qstock), and daily feed distributed (Qdaily&#95;feed). ENV were calculated for LCA impact categories climate change, eutrophication and acidification. ENV were expressed per ton of fish produced per year (ENV(fish)) and per farm per year (ENV(farm)). Results show that irrespective of quota used, EV of FCR as well as ENV(fish) and ENV(farm) were always positive, meaning that improving FCR increased profit and decreased environmental impacts. However, the EV and the ENV(fish) of TGC were positive only when quota was Qstock or Qdaily&#95;feed. Moreover, the ENV(farm) of TGC was negative in Qstock and Qdaily&#95;feed quotas, meaning that improving TGC increased the environmental impact of the farm. We conclude that Qstock quota and Qdaily&#95;feed quota are economically favorable to a genetic improvement of TGC, a major trait for farmers. However, improving TGC increases the environmental impact of the farm. Improving FCR represents a good opportunity to balance out this increase but more information on its genetic background is needed to develop breeding programs improving FCR.

Abstract

Variable and low egg quality is a major limiting factor for the development of efficient aquaculture production. This stems from limited knowledge on the mechanisms underlying egg quality in cultured fish. Molecular analyses, such as transcriptomic studies, are valuable tools to identify the most important processes modulating egg quality. However, very few studies have been devoted to this aspect so far. Within this study, the microarray-based transcriptomic analysis of eggs (of different quality) of sea bass (Dicentrarchus labrax) was performed. An Agilent oligo microarray experiment was performed on labelled mRNA extracted from 16 batches of eggs (each batch obtained from a different female) of sea bass, in which over 24,000 published probe arrays were used. We identified 39 differentially expressed genes exhibiting a differential expression between the groups of low (fertilization rate < 60 %) and high (fertilization rate > 60 %) quality. The mRNA levels of eight genes were further analyzed by quantitative PCR. Seven genes were confirmed by qPCR to be differentially expressed in eggs of low and high quality. This study confirmed the importance of some of the genes already reported to be potential molecular quality indicators (mainly rnf213 and irf7), but we also found new genes (mainly usp5, mem-prot, plec, cenpf), which had not yet been reported to be quality-dependent in fish. These results suggest the importance of genes involved in several important processes, such as protein ubiquitination, translation, DNA repair, and cell structure and architecture; these probably being the mechanisms that contribute to egg developmental competence in sea bass.

Abstract

To understand the genetic basis of coping style in European seabass, fish from a full factorial mating (10 females x 50 males) were reared in common garden and individually tagged. Individuals coping style was characterized through behavior tests at four different ages, categorizing fish into proactive or reactive: a hypoxia avoidance test (at 255 days post hatching, dph) and 3 risk-taking tests (at 276, 286 and 304 dph). We observed significant heritability of the coping style, higher for the average of risk-taking scores (h2 = 0.45 ± 0.14) than for the hypoxia avoidance test (h2 = 0.19 ± 0.10). The genetic correlations between the three risk-taking scores were very high (rA = 0.96-0.99) showing that although their repeatability was moderately high (rP = 0.64-0.72), successive risk-taking tests evaluated the same genetic variation. A mild genetic correlation between the results of the hypoxia avoidance test and the average of risk-taking scores (0.45 ± 0.27) suggested that hypoxia avoidance and risk-taking tests do not address exactly the same behavioral and physiological responses. Genetic correlations between weight and risk taking traits showed negative values whatever the test used in our population i.e. reactive individual weights were larger. The results of this quantitative genetic analysis suggest a potential for the development of selection programs based on coping styles that could increase seabass welfare without altering growth performances. Abstract

The shy-bold continuum is both a fundamental aspect of human behavior and a relatively stable behavioral trait for many other species. Here we assessed whether shy individuals prefer familiar congeners, taking the European sea bass, a recently domesticated fish showing similar behavioral responses to wild fish, as a model to better understand the inter-individual variability in social behavior previously observed in this species. In the wild, the link between familiarity i.e., the preference of fish for familiar congeners and boldness could be part of the mechanism underlying shoaling formation in fish. Thirty fish were individually tested in a device designed to assess the preference for a familiar vs. an unfamiliar congener on the basis of visual cues only. An open field test (OFT) with shelter was performed on the same fish 32 days later to assess the boldness of each individual. Variables of interest included the proportion of time spent in the shelter, border and center zone of the arena and variables of activity. Variables measured in OFT were collapsed into first principal component scores using Principal Components Analysis (PCA) which allowed characterizing a shy-bold continuum. Time spent near the familiar congener was negatively correlated with boldness i.e., shy individuals spent most of the time near the familiar congener. We discuss the relevance of these findings to the understanding of the behavior of European sea bass and suggest that the link between familiarity and shyness is a general aspect of both animal and human behavior., (Copyright © 2016 Elsevier B.V. Abstract

Background: European sea bass (Dicentrarchus labrax) is one of the most important farmed species in Mediterranean aquaculture. The observed sexual growth and maturity dimorphism in favour of females adds value towards deciphering the sex determination system of this species. Current knowledge indicates the existence of a polygenic sex determining determination system that interacts with temperature. This was explored by restriction-site associated DNA (RAD) marker analysis in a test panel of 175 offspring that originated from a factorial cross between two dams and four sires from a single full-sib family., Results: The first high-density single nucleotide polymorphism (SNP) based linkage map for sea bass was constructed, consisting of 6706 SNPs on 24 linkage groups. Indications for putative sex-determining QTL (quantitative trait loci) that were significant at the genome-wide threshold were detected on linkage groups 6, 11 and 18 to 21, although a genome-wide association study (GWAS) did not identify individual significant SNPs at a genome-wide threshold. A preliminary genomic prediction approach that tested the efficiency of SNP-based selection for female sea bass showed a slight advantage compared to traditional pedigree-based selection. However, when the same models were tested on the same animals for selection for greater length, a clear advantage of the SNP-based selection was observed., Conclusions: Overall, the results of this study provide additional support to the polygenic sex determination hypothesis in sea bass. In addition, identification of sex-ratio QTL may provide new opportunities for sex-ratio control in sea bass.

Abstract

In fish farming, economic values (EV) of breeding goal traits are lacking, even though they are key parameters when defining selection objectives. The aim of this study was to develop a bioeconomic model to estimate EV of 2 traits representing production performances in fish farming: the thermal growth coefficient (TGC) and the feed conversion ratio (FCR). This approach was applied to a farm producing African catfish (Clarias gariepinus) in a recirculating aquaculture system (RAS). In the RAS, 2 factors could limit production level: the nitrogen treatment capacity of the biofilter or the fish density in rearing tanks at harvest. Profit calculation includes revenue from fish sales, cost of juveniles, cost of feed, cost of waste water treatment, and fixed costs. In the reference scenario, profit was modeled to zero. EV were calculated as the difference in profit per kilogram of fish between the current population mean for both traits (µt) and the next generation of selective breeding (µt+Δt) for either TGC or FCR. EV of TGC and FCR were calculated for three generations of hypothetical selection on either TGC or FCR (respectively 6.8% and 7.6% improvement per generation). The results show that changes in TGC and FCR can affect both the number of fish that can be stocked (number of batches per year and number of fish per batch) and the factor limiting production. The EV of TGC and FCR vary and depend on the limiting factors. When dissolved NH3-N is the limiting factor for both µt and µt+Δt, increasing TGC decreases the number of fish that can be stocked but increases the number of batches that can be grown. As a result, profit remains constant and EVTGC is zero. Increasing FCR, however, increases the number of fish stocked and the ratio of fish produced per kilogram of feed consumed ("economic efficiency"). The EVFCR is 0.14 €/kg of fish, and profit per kilogram of fish increases by about 10%. When density is the limiting factor for both µt and µt+Δt, the number of fish stocked per batch is fixed; therefore, extra profit is obtained by increasing either TGC, which increases the annual number of batches, or by decreasing FCR, which decreases annual feed consumption. EVTGC is 0.03 €/kg of fish and EVFCR is 0.05-0.06 €/kg of fish. These results emphasize the importance of calculating economic values in the right context to develop efficient future breeding programs in aquaculture.

Although food deprivation is a major ecological pressure in fishes, there is wide individual variation in tolerance of fasting, whose mechanistic bases are poorly understood. Two thousand individually tagged juvenile European sea bass were submitted to two 'fasting/feeding' cycles each comprising 3 weeks of food deprivation followed by 3 weeks of ad libitum feeding at 25°C. Rates of mass loss during the two fasting periods were averaged for each individual to calculate a population mean. Extreme fasting tolerant (FT) and sensitive (FS) phenotypes were identified that were at least one and a half standard deviations, on opposing sides, from this mean. Respirometry was used to investigate two main hypotheses: (1) tolerance of food deprivation reflects lower mass-corrected routine metabolic rate (RMR) in FT phenotypes when fasting, and (2) tolerance reflects differences in substrate utilisation; FT phenotypes use relatively less proteins as metabolic fuels during fasting, measured as their ammonia quotient (AQ), the simultaneous ratio of ammonia excretion to RMR. There was no difference in mean RMR between FT and FS over 7 days fasting, being 6.70±0.24 mmol h(-1) fish(-1) (mean ± s.e.m., N=18) versus 6.76±0.22 mmol h(-1) fish(-1) (N=17), respectively, when corrected to a body mass of 130 g. For any given RMR, however, the FT lost mass at a significantly lower rate than FS, overall 7-day average being 0.72±0.05 versus 0.90±0.05 g day(-1) fish(-1), respectively (P<0.01, t-test). At 20 h after receiving a ration equivalent to 2% body mass as food pellets, ammonia excretion and simultaneous RMR were elevated and similar in FT and FS, with AQs of 0.105±0.009 and 0.089±0.007, respectively. At the end of the period of fasting, ammonia excretion and RMR had fallen in both phenotypes, but AQ was significantly lower in FT than FS, being 0.038±0.004 versus 0.061±0.005, respectively (P<0.001, t-test). There was a direct linear relationship between individual fasted AQ and rate of mass loss, with FT and FS individuals distributed at opposing lower and upper extremities, respectively. Thus the difference between the phenotypes in their tolerance of food deprivation did not depend upon their routine energy use when fasting. Abstract

The European sea bass, Dicentrarchus labrax L., tolerates a range of salinities from freshwater to hyper-saline. To study differences in protein expression, fish were reared in both freshwater and seawater. After 3-month acclimation, gill and intestine epithelia were collected and the soluble protein extracted. In all, 362 spots were differentially expressed in the gills and intestines of fishes reared in seawater compared to those from freshwater. Fifty differential protein spots were excised from a colloidal Coomassie-stained gel. Nine separate protein spots were identified unambiguously by mass spectrometry and database searching. Among the six proteins over-expressed in gill cells in seawater, five were cytoskeleton proteins and one was the aromatase cytochrome P450. In gill cells under freshwater conditions, the two over-expressed proteins identified were the prolactin receptor and the major histocompatibility complex class II beta-antigen. In intestinal cells under freshwater conditions, the Iroquois homeobox protein Ziro5 was upregulated over ninefold. The expression of these proteins, their possible direct or indirect roles in the adaptation of D. labrax to salinity, and their correspondences with a previous study are discussed.

Abstract

A preliminary study on tetraploid gynogenetic induction in the European sea bass was performed by pressure-blocking the second polar body release and the first cleavage in eggs fertilized with ultraviolet-irradiated sperm. Fertilization of eggs with genetically inactivated sperm produced only haploid development that terminated around hatching. Pressure treatments (8.500 psi for 2 min) applied at 6 and 65 min after fertilization (a.f.) produced variable levels (7-95%) of tetraploid larvae at hatching. A small proportion of mosaics (3.8n/4.2n) was also recorded.
