4. Association between rs56066773 and rs56232250 polymorphisms of FOXP3 gene in target site of microRNA with rheumatoid arthritis in patients referred to Emam Ali clinic of Shahrekord

Abstract

Background and aims: Rheumatoid arthritis is an autoimmune disease in which the numder or function of regulatory T cells (Treg) is impaired. FOXP3 is one of the major factors of Treg function. Among the factors involved on FOXP3 expression are microRNAs (MIR) which bind to 3´UTR. A nucleotide substitution in the sequence of the target site of microRNA can affect the regulation of microRNA. Polymorphisms of rs56066773 and rs56232250 in the 3´UTR of gene FOXP3 can relate with rheumatoid arthritis through the target gene. This study was aimed to investigate the relationship between two polymorphisms with rheumatoid arthritis. Methods: In this case-control study, 98 RA patients were recruited from Emam Ali rheumatology clinic and 124 healthy individuals (without the negative history of autoimmune diseases) served as control. Rs56066773 and rs56232250 polymorphisms in the 3´UTR of FOXP3 gene were investigated with Polymerase Chain Reaction (PCR)- Restricted Fragment Length Polymorphism (RFLP). The data were analyzed using SPSS. Results: In the study population, the frequency of the A/G in rs56066773 in patients and control was 3.1 and 1.6%, respectively. The frequency of the same genotype in rs56232250 was 1 and 1.6% in patients and controls respectively. It was not found any significant relationship between two polymorphism and disease. Conclusion: Although not found a significant relationship between polymorphisms and arthritis, previous studies has been established the relationship between polymorphisms in the microRNA target site with a number of diseases. Therefore, it is necessary to examine other FOXP3 gene 3'UTR polymorphisms.

Abstract

An unsymmetrical tridentate Schiff base 4-((E)-(2-amino-5-nitrophenylimino)methyl)-5-(hydroxymethyl)- 2-methylpyridin-3-ol is newly synthesized and characterized experimentally. Its geometrical parameters, the assignment of IR bands and NMR chemical shifts are also computed by the density functional theory (DFT) method. In addition, the atoms in molecules (AIM) analysisis employed to investigate its geometry. Only one of the diamine-NH2 groups undergoes the condensation reaction. In the structure of the synthesized Schiff base, the remaining aminogroup lies in the para position with respect to the nitro group (isomer 1). In both gas and solution phases, isomer 1 is more stable than isomer 2 with the meta orientation of the amino and nitro groups. The NMR chemical shifts and the AIM analysis show that isomer 1 is a more favorite structure for the synthesized Schiff base. It has no planar structure. The phenolic proton is engaged in the intramolecular hydrogen bond with the azomethine nitrogen atom. The experimental results are in good agreement with the theoretical ones, confirming the validity of the optimized geometry. [ABSTRACT FROM AUTHOR]

16. On-line measurement of heat of combustion

Abstract

An experimental method for an on-line measurement of heat of combustion of a gaseous hydrocarbon fuel mixture of unknown composition is developed. It involves combustion of a test gas with a known quantity of air to achieve a predetermined oxygen concentration level in the combustion products. This is accomplished by a feedback controller which maintains the gas volumetric flow rate at a level consistent with the desired oxygen concentration in the products. The heat of combustion is determined from a known correlation with the gas volumetric flow rate. An on-line microcomputer accesses the gas volumetric flow data, and displays the heat of combustion values at desired time intervals.

17. Feasibility of a nuclear gauge for fuel quantity measurement aboard aircraft

Abstract

Capacitance fuel gauges have served as the basis for fuel quantity indicating systems in aircraft for several decades. However, there have been persistent reports by the airlines that these gauges often give faulty indications due to microbial growth and other contaminants in the fuel tanks. This report describes the results of a feasibility study of using gamma ray attenuation as the basis for measuring fuel quantity in the tanks. Studies with a weak Am-241 59.5-keV radiation source indicate that it is possible to continuously monitor the fuel quantity in the tanks to an accuracy of better than 1 percent. These measurements also indicate that there are easily measurable differences in the physical properties and resultant attenuation characteristics of JP-4, JP-5, and Jet A fuels. The experimental results, along with a suggested source-detector geometrical configuration are described.

18. Measurement of viscosity of gaseous mixtures at atmospheric pressure

Abstract

Coefficients of viscosity of various types of gas mixtures, including simulated natural-gas samples, have been measured at atmospheric pressure and room temperature using a modified capillary tube method. Pressure drops across the straight capillary tube section of a thermal mass flowmeter were measured for small, well-defined, volume flow rates for the test gases and for standard air. In this configuration, the flowmeter provides the volumetric flow rates as well as a well-characterized capillary section for differential pressure measurements across it. The coefficients of viscosity of the test gases were calculated using the reported value of 185.6 micro P for the viscosity of air. The coefficients of viscosity for the test mixtures were also calculated using Wilke's approximation of the Chapman-Enskog (C-E) theory. The experimental and calculated values for binary mixtures are in agreement within the reported accuracy of Wilke's approximation of the C-E theory. However, the agreement for multicomponent mixtures is less satisfactory, possible because of the limitations of Wilkes's approximation of the classical dilute-gas state model.

19. Investigation of paramagnetic response of metallic epoxies

Abstract

The paramagnetic properties of epoxies which were impregnated with metal ions were examined as the primary task in this research. A major conclusion was that the quality control of the epoxies was insufficient to permit reliable evaluation. Subsequently, a new set of specimens is being prepared. As an additional task, a new method is investigated for estimating heats of combustion for saturated hydrocarbons. The results of that investigation have shown that the empirical approach is a promising method for on-line measurements.

Abstract

Using density functional theory, the mechanism of adsorption of 2-chlorophenol from water in the presence of γ-Fe2O3 nanoparticles was investigated. Fe2O3 nanoparticles were modelled using Fe6(OH)18(H2O)6 ring clusters. 2-chlorophenol can coordinate to the γ-Fe2O3 nanoparticles via its own OH or Cl groups. The process produces two intermediates which will be converted into final products through two pathways (Cl pathway and OH pathway). The activation energy and activation Gibbs free energy of the two pathways have been calculated and compared with each other. It was found that the OH pathway is under thermodynamic control and the C1 pathway is under kinetic control. All of the calculations were performed using a hybrid density functional method (B3LYP) in the solution phase (PCM model). [ABSTRACT FROM AUTHOR]

Abstract

Background and Objective: Nicotine is an addictive substance and Ritalin is a medicine which has been prescribed for treatment of hyperactivity / attention deficit disorder (ADHD). This study was done to evaluate the effect of Ritalin and nicotine and combination of Ritalin and nicotine on daily sperm production and epididymal sperm reserve in mice. Methods: In this experimental study, 120 adult male BALB/c mice were randomly allocated into one control group and 11 experimental (treatment) groups. Animals, in the first, second and third treated groups were received nicotine at the doses of 100, 200 and 400 microgr/kg/bw, respectively. Mice in fourth and fifth treated groups were received Ritalin at doses of 2 and 10 mg/kg/bw, respectively. Animals in sixth and seventh treatment groups were received nicotine in 400 microgr/kg/bw and Ritalin in 2 and 10 mg/kg/bw and in eighth and ninth groups, nicotine at dose of 200 microgr/kg/bw and Ritalin at doses of 2 and 10 mg/kg/bw, respectively. Animals in tenth and eleventh treated groups were received nicotine as dose of 100 microgr/kg/bw and Ritalin at doses of 2 and 10 mg/kg/bw, respectively. Ritalin and nicotine were administrated orally for 40 days. At the end of study, daily sperm production and epididymal sperm reserve were measured. Results: The daily sperm production was significantly reduced in the groups with high consumption doses of nicotine and different doses of Ritalin and the majority of groups which used the combination of Ritalin and nicotine (P<0.05). The epididymal sperm reserve was significantly increased in experimental groups of 8, 9, 10 and 11 which were received the combination of Ritalin and nicotine in different doses (P<0.05). Conclusion: The combination of Ritalin and nicotine reduces daily sperm production and it increases epididymal sperm reserve in adult BALB/c mice. [ABSTRACT FROM AUTHOR]

24. An Experimental and Analytical Investigation of Friction Factors for Fully Developed Flow in Internally Finned Triangular Ducts

Abstract

Friction factors for fully developed flow in triangular ducts with fins of various height and width are investigated for Reynolds numbers ranging from 150 to 90,000. Two triangular ducts having apex angles of 60 and 38.8 deg are studied. Results are presented in the form of standard plots of friction factor as a function of Reynolds number. Friction factor values for the smooth triangular duct cases are in good agreement with the existing results. For the finned-duct cases, the fully developed axial velocity profiles in laminar flow are determined by solving the x-momentum equation iteratively by the Gauss–Seidel finite-difference technique. The theoretically determined friction factors for these cases are in good agreement with the experimental values of friction factors based on pressure drop measurements.

Abstract

An unsymmetrical tridentate Schiff base 4-((E)-(2-amino-5-nitrophenylimino)methyl)-5-(hydroxymethyl)- 2-methylpyridin-3-ol is newly synthesized and characterized experimentally. Its geometrical parameters, the assignment of IR bands and NMR chemical shifts are also computed by the density functional theory (DFT) method. In addition, the atoms in molecules (AIM) analysisis employed to investigate its geometry. Only one of the diamine-NH2 groups undergoes the condensation reaction. In the structure of the synthesized Schiff base, the remaining aminogroup lies in the para position with respect to the nitro group (isomer 1). In both gas and solution phases, isomer 1 is more stable than isomer 2 with the meta orientation of the amino and nitro groups. The NMR chemical shifts and the AIM analysis show that isomer 1 is a more favorite structure for the synthesized Schiff base. It has no planar structure. The phenolic proton is engaged in the intramolecular hydrogen bond with the azomethine nitrogen atom. Abstract

Purpose: To present the effectiveness and safety of radiofrequency ablation (RFA) in parathyroid adenoma (PTA)., Materials and Methods: In this retrospective study, 60 patients with a single PTA were evaluated for changes in biochemical and ultrasonographic features up to 6 months after RFA of the lesion. Adenomas were ablated with an alternative technique so called "Nik jet dissection" which incorporates full hydrodissection and polar artery coagulation. Complications as well as the variations in biochemical data and nodule volumes were analyzed between baseline measurements and at each follow-up interval data (first day, 1, 3, and 6 months after ablation) were analyzed., Results: A significant reduction in serum intact parathyroid hormone and calcium levels was observed 6 months after ablation, with a mean difference of - 83.4 ± 104.1 pg/mL, p < 0.001, and - 0.29 ± 0.22 mmol/L, p < 0.001, respectively. Serum phosphorus levels increased significantly with a mean difference of 0.09 ± 0.19 mmol/L, p = 0.040 at the end of the follow up. We observed a significant volume reduction rate of parathyroid adenomas with 89 ± 20.8 percent, p < 0.001. Also, 51% of adenomas disappeared at the end of the follow up. In this study, two cases of hematoma and one case of transient hoarseness (grade 1 of the CIRSE classification) were encountered., Conclusion: Our study showed that RFA with the alternative technique, called "Nik jet dissection" is a safe and effective modality in management of PTA. Abstract

Preeclampsia, the more severe manifestation of gestational hypertensive disorders, is a major cause of maternal and perinatal morbidity and mortality worldwide. Genetic polymorphisms in long non-coding RNAs (lncRNAs) are considered as potential genetic preeclampsia. This study aimed to explore the association between SENCR rs555172 SNP and PE risk in healthy pregnant women compared to women with preeclampsia. A total of 140 healthy pregnant women and 130 preeclampsia cases were included in the study. The rs555172 genotype was determined using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), and the expression of the SENCR gene was analyzed in 40 placenta tissue samples from both groups. Various statistical approaches were employed to assess the genotypic and allelic frequencies. The results showed no significant difference in the frequency of the rs555172 polymorphism between healthy pregnant women and those with preeclampsia in terms of the dominant (p=0.82), recessive (p=0.39), and over-dominant (p=0.42) models. Additionally, the analysis of SENCR relative expression revealed no significant difference between the two groups (p=0.48). In conclusion, the LncRNA SENCR rs555172(G/A) seems not associated with an increased risk of Preeclampsia in pregnant women.

31. Cycle Threshold Values Predict COVID-19 Severity and Mortality but Are not Correlated with Laboratory Markers.

Abstract

Many studies have evaluated the possible utility of cycle threshold (Ct) values as a predictor of Coronavirus disease 2019 (COVID-19) severity and patient outcome. Given the inconsistent results, we aimed to evaluate the association between severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) Ct values and disease severity, inflammatory markers, and outcomes in Iranian patients with COVID-19. A retrospective study of 528 patients with COVID-19 hospitalized from September 2020 to October 2021 was conducted. Demographic, clinical, and laboratory data of patients were retrieved from electronic medical records. Ct values were analyzed as a continuous variable after subcategorizing into 3 groups: low (Ct values<20), medium (Ct values 20 to 30), and high (Ct values>30). Of the 528 patients (45.1% female) aged 13 to 97 years, 109 patients had low Ct values, 312 patients had medium, and 107 patients had high Ct values. Patients with low Ct values were more likely to present with critical COVID-19, require invasive mechanical ventilation and develop complications such as acute respiratory distress syndrome and pneumonia. Furthermore, patients with low or medium Ct values were more likely to die compared to patients with high Ct values. Multivariate analysis showed that patients with low or medium Ct values were more likely to have severe COVID-19 compared with patients with high Ct values. The multivariate analysis also showed a higher risk of mortality in patients with low Ct values compared to patients with high Ct values, although this was not statistically significant. Our findings revealed that Ct values were an independent predictor of COVID-19 severity. The risk of mortality was higher in patients with low Ct values. However, further investigation is needed to address the correlation between Ct values and inflammatory factors.

Abstract

Precise diagnosis of thyroid nodules is challenging due to non-diagnostic/inconclusive results and uncertainties about the malignancy of follicular neoplasms (FNs), even in frozen-section pathology. Therefore, surgical management, especially in Bethesda III and IV categories, may be complicated, and sometimes a second surgery may be required. The Thyroid Nodule Impedance Measurement System (TN-IMS) consists of a metallic patch attached to submental skin and a G20 I.V. cannula inserted into the targeted nodules. Two impedance-based parameters named Z1kHz and impedance phase slope (IPS) in 100 kHz to 500 kHz of the thyroid nodules are recorded and compared with their histopathological results as the gold standard. TN-IMS was intra-surgically applied to 103 human thyroid nodules and normal thyroid tissues. A remarkable consistency between defined co-ranges of Z1kHz/IPS and the histopathological status of specimens was achieved (p < 0.001). Based on these measurements, it was concluded that intraoperative bioelectrical impedance scanning of thyroid nodules would be a helpful complementary approach to detecting high-risk excision-required thyroid nodules.

Abstract

Background and Aims: This study aimed to assess the level of the expression of CCL-18 on nasal inferior turbinate mucosa in patients with mild (M) and moderate-to-severe (M/S) persistent allergic rhinitis (PAR)., Methods: The participants in this case-controlled study were divided into three groups: patients with M/S PAR, patients with M PAR, and the healthy control group. Biopsies of nasal inferior turbinate mucosa were obtained from the participants. Expression of CCL-18 mRNA was evaluated by real-time PCR. The serum levels of CCL-18 were determined by ELISA. Total serum IgE levels and specific serum IgE levels were measured. The clinical manifestations were assessed using the total nasal syndrome score (TNSS)., Results: Gene expression and the serum level of CCL-18 in patients with M/S PAR increased significantly compared to the control group and patients with M PAR. The serum level of CCL-18 was found to correlate with TNSS in patients with M/S PAR. There was a statistical correlation between the serum level of CCL-18 and the total serum IgE in the treatment groups., Conclusion: The results of the study indicate that there could be a relationship between the expression of CCL-18 in nasal turbinate mucosa and the severity of allergic rhinitis.

Abstract

The new 3H-imidazo[4,5-a]acridine-11-carbonitriles were prepared from the reaction of 1-alkyl-5-nitro-1H-benzoimidazoles with 2-(4-methoxyphenyl)acetonitrile and benzyl cyanide by nucleophilic substitution of hydrogen in high yields. Physical spectral and analytical data have confirmed the structures of the synthesized dyes. The optical and solvatochromic properties of these compounds were investigated and the results showed that they show very interesting photophysical properties. Density functional theory (DFT) calculations of fluorescent dyes were performed to provide the optimized geometries and relevant frontier orbitals. Furthermore, intra- and intermolecular interactions in fluorescent imidazo[4,5-a]acridines were evaluated by AIM (Atoms in Molecules) analysis.

Abstract

Nowadays, there is a trend toward early diagnosis and treatment of rheumatoid arthritis (RA) especially in patients with early signs of bone erosion which can be detected by magnetic resonance imaging (MRI). The aim of following study is to compare the sensitivity and specificity of ultrasonography (US) and conventional radiography (CR) compared to MRI for early detection of bone erosion in RA patients. In 12 patients with RA diagnosis, 120 first to fifth metacarpophalangeal joints and 96 second to fifth proximal interphalangeal joints were examined. Non-contrast MRI, US and CR were performed for bone erosion evaluation. For further analysis, the patients were divided in two equal groups according to disease activity score (DAS28). The overall sensitivity and specificity of US compared to MRI in detecting bone erosion were 0.63 and 0.98, respectively with a considerable agreement (kappa = 0.68, p < 0.001). Sensitivity and specificity of CR compared to MRI in detecting bone erosion were 0.13 and 1.00, respectively (kappa = 0.20, p < 0.001). In patients with more active disease, the sensitivity and specificity were 0.67 and 0.99 (kappa = 0.74, p < 0.001) compared to 0.59 and 0.97 (kappa = 0.61, p < 0.001) for the rest of patients according to DAS28. Conclusively, these findings reveal an acceptable agreement between US and MRI for detection of bone erosion in patients with early RA but not CR. US might be considered as a valuable tool for early detection of bone erosion especially when MRI is not available or affordable. Besides, it seems the US could be more reliable when the disease is more active.
