Abstract

Introduction: Leukemic stem cells (LSCs) are the transcriptionally low/silent cells which are resistant to the tyrosine kinase inhibitor. These have been found to play a pivotal role in disease relapse in chronic myeloid leukemia (CML) cases. The present study evaluated the correlation of absolute CML‐LSC count in the peripheral blood (PB) at diagnosis and achievement of major molecular response (MMR) at 12 months in patients of CML‐CP. Methods: This was a prospective, observational, non‐interventional single center study including newly diagnosed adult (>18 yrs) CML‐CP patients. Absolute CD26 + CML‐LSC quantification was done by multiparametric flow cytometry. Patients were treated with Imatinib treatment and subsequently monitored at 3‐month intervals for BCR::ABL transcript levels. MMR was defined as a BCR::ABL1 transcript level of less than 0.1% on international scale. Results: A total of 89 patients were enrolled in the study out of which 40.5% achieved MMR at 12 months. There was a significant difference in the median absolute CML‐LSC count of the patients who achieved MMR at 12 months as compared to those who did not (58.5 vs 368.1 cells/μL; p value <0.001). Using a ROC analysis, a count of <165.69 CML LSC/μL was identified to have a sensitivity of 83.8% and specificity of 72.4%, in predicting the MMR at 12 months. Conclusion: Absolute CML‐LSC count at diagnosis in the PB predicts the MMR achievement at 12 months. An absolute count of less than 165 cells/μL is highly predictive of achieving MMR at 12 months. Abstract

The CryoGrid community model is a flexible toolbox for simulating the ground thermal regime and the ice-water balance for permafrost and glaciers, extending a well-established suite of permafrost models (CryoGrid 1, 2, and 3). The CryoGrid community model can accommodate a wide variety of application scenarios, which is achieved by fully modular structures through object-oriented programming. Different model components, characterized by their process representations and parameterizations, are realized as classes (i.e., objects) in CryoGrid. Standardized communication protocols between these classes ensure that they can be stacked vertically. For example, the CryoGrid community model features several classes with different complexity for the seasonal snow cover, which can be flexibly combined with a range of classes representing subsurface materials, each with their own set of process representations (e.g., soil with and without water balance, glacier ice). We present the CryoGrid architecture as well as the model physics and defining equations for the different model classes, focusing on one-dimensional model configurations which can also interact with external heat and water reservoirs. We illustrate the wide variety of simulation capabilities for a site on Svalbard, with point-scale permafrost simulations using, e.g., different soil freezing characteristics, drainage regimes, and snow representations, as well as simulations for glacier mass balance and a shallow water body. The CryoGrid community model is not intended as a static model framework but aims to provide developers with a flexible platform for efficient model development. In this study, we document both basic and advanced model functionalities to provide a baseline for the future development of novel cryosphere models.

A novel methodology is proposed here for drivers commanding the vehicle through his gestures and haptic sensation that are connected to Master control system (MCS) which acts as an master interface between the driver and the different machinery sensors inside the vehicle, all such machinery is controlled by the respective actuators, that are commanded by the MCS correlated by the gesture, haptic sensation of driver. Inputs from vehicle i.e. surrounding situation and traffic positions are collected by HD video cameras placed at different positions on the vehicle hardware also the system is thoroughly by CDAS for better efficiency.This system has direct applications in situations where human life is very precious for example in serious conflicts ; also it counters the situation as faced by humanity during Covid19.

Abstract

Peatlands form part of the boreal landscapes exhibiting diverse types and microforms that have different characteristics of topography, hydrology, vegetation, and soil. Our understanding is still limited concerning how boreal peatlands, especially their inherent heterogeneities, affect the regional biosphere-atmosphere exchange of carbon and related climate effects, and what level of detail is needed to characterize them in land cover maps. By combining remote sensing information, field measurements, and biogeochemical modeling, we estimated that the peatlands within the study area (26% of the area) contributed 22% of the landscape-scale carbon dioxide uptake and 93% of methane emissions. We also showed that, among different land cover types, peatlands played the dominant role in the variability of radiative climate effect due to carbon exchanges. Possible aggregation and misclassification of peatland types could induce significant biases in the regional balances and their radiative effect, but the distinction of non-inundated dry and inundated wet peatland types could reduce these biases effectively.

Abstract

Peatlands, with high spatial variability in ecotypes and microforms, constitute a significant part of the boreal landscape and play an important role in the global carbon (C) cycle. However, the effects of this peatland heterogeneity within the boreal landscape are rarely quantified. Here, we use field-based measurements, high-resolution land cover classification, and biogeochemical and atmospheric models to estimate the atmosphere-ecosystem C fluxes and corresponding radiative effect (RE) for a boreal landscape (Kaamanen) in northern Finland. Our result shows that the Kaamanen catchment currently functioned as a sink of carbon dioxide (CO2) and a source of methane (CH4). Peatlands (26% of the area) contributed 22% of the total CO2 uptake and 89% of CH4 emissions; forests (61%) accounted for 78% of CO2 uptake and offset 6% of CH4 emissions; water bodies (13%) offset 7% of CO2 uptake and contributed 11% of CH4 emissions. The heterogeneity of peatlands accounted for 11%, 88%, and 75% of the area-weighted variability (deviation from the area-weighted mean among different land cover types (LCTs) within the catchment) in CO2 flux, CH4 flux, and the combined RE of CO2 and CH4 exchanges over the 25-yr time horizon, respectively. Aggregating peatland LCTs or misclassifying them as non-peatland LCTs can significantly (p < 0.05) bias the regional CH4 exchange and RE estimates, while differentiating between drier non-inundated and wetter inundated peatlands can effectively reduce the bias. Current land cover products lack such details in peatland heterogeneity, which would be needed to better constrain boreal C budgets and global C-climate feedbacks.
