The visual system of the fat-tailed dunnart (Sminthopsis crassicaudata), a small polyprotodont marsupial, has been examined both anatomically and behaviourally. The ganglion cell layer was examined in cresyl-violet stained wholemounts and found to contain a mean of 81,400 ganglion cells (SD ± 3,360); the identification of ganglion cells was supported by a correspondence to optic axon counts. Ganglion cells were distributed as a mid-temporally situated area centralis, embedded in a pronounced visual streak. Localised implants of horseradish peroxidase into retinal wholemounts revealed both A-type and B-type horizontal cells. Sections of the outer retina showed it to be rod-dominated, with a rod-to-cone ratio of 40:1 at the area centralis; cones were found to contain oil droplets but double cones were not a prominent feature. The retinal pigment epithelium consisted of squamous cells. Visual acuity, estimated from counts of peak ganglion cell density (8,300/mm2, SD ± 1,180) and measurements of posterior nodal distance (2.9 mm), was found to be 2.30 cycles per degree. The value was close to that of 2.36 cycles per degree estimated by behavioural tests using a Mitchell jumping stand; values were similar at low, intermediate and high light levels. Our findings are discussed in relation to the lifestyle of the dunnart.

This paper describes the ocular morphology of young adults of the southern hemisphere lamprey Geotria australis, the sole representative of the Geotriidae, and makes comparisons with those of holarctic lampreys (Petromyzontidae). As previously reported for the holarctic lamprey Ichthyomyzon unicuspis [Collin and Fritzsch, 1993], the lens of G. australis is non-spherical and possesses a cone-shaped posterior that may be capable of mediating variable focus. The avascular retina of G. australis is well differentiated, containing three retinal ganglion cell populations, three layers of horizontal cells and three photoreceptor types. In contrast to petromyzontids that contain only two photoreceptor types (short and long), G. australis possesses one rod-like (R1) and two cone-like (C1 and C2) photoreceptors. Although the rod-like receptor in G. australis may be homologous with the short receptors of holarctic lampreys, the two cone-like receptors have morphological characteristics that differ markedly from those of the long receptors of their holarctic counterparts. The features which distinguish the two cone-like receptors from those of the long receptor type in holarctic lampreys are the characteristics of the mitochondria and the presence of large amounts of two different types of stored secretory material in the endoplasmic reticulum of the myoid (refractile bodies). The endoplasmic reticulum of each receptor type has a different shape and staining profile and is polymorphic, each showing a continuum of distension. It is proposed that the presence of two cone-like photoreceptors with different characteristics would increase the spectral range of G. australis and thus be of value during the parasitic phase, when this lamprey lives in the surface marine waters. The irideal flap, present in G. australis but not petromyzontids, would assist in reducing intraocular flare during life in surface waters. The results of this study, which are discussed in the context of the proposed evolution of lampreys, emphasise that it is important to take into account the characteristics of the eyes of southern hemisphere lampreys when making generalizations about the eyes of lampreys as a whole.

The fine structure of the retinal pigment epithelium (RPE) of the cichlid Oreochromis niloticus was investigated in both light- and dark-adaptation. The eyes of four light-adapted and from four dark-adapted O. niloticus were fixed routinely for light and transmission electron microscopy. The RPE consisted of a single layer of columnar cells showing minimal basal infolding but plentiful apical processes that in light-adaptation interdigitated with the photoreceptor outer segments. The epithelial cells were joined by a series of basally-located tight junctions. These cells showed a large vesicular nucleus, plentiful smooth endoplasmic reticulum and polysomes, but only small amounts of rough endoplasmic reticulum. Phagosomes, lysosome-like bodies, lipid droplets, and myeloid bodies were observed. The choriocapillaris was a single layer of large-caliber capillaries, and Bruch's membrane (complexus basalis) was a trilaminate structure typical of teleosts. The RPE melanosomes moved basally (sclerally) in dark-adaptation and apically (vitreally) during light-adaptation. Other morphological features which changed at least to some degree during retinomotor responses were: the location of the RPE nucleus; the location and electron density of the mitochondria; and the location, number, and size of the myeloid bodies. A number of unique morphological changes take place within the RPE cells of this species during the circadian cycle in addition to the movement of melanosomes characterized in other vertebrates.

Cell generation and the early stages of maturation of the retinal pigment epithelium (RPE) and photoreceptors were examined in a marsupial, the quokka, Setonix brachyurus. Results are presented for animals aged up to postnatal day (P)250. RPE cell generation was studied by analysis of cell number from wholemounted retinae and by tritiated thymidine (3HThy) autoradiography in sectioned material. For 3HThy autoradiography, quokkas aged P1–P200 were injected with 3HThy and killed either 6–20 hours later (pulse-kill) or at P100 or P250 (pulse-leave). The extent of pigmentation of the RPE sheet was examined from sections of embryonic and early postnatal stages. Retinae from animals aged P5 to P160 were also examined at the electron microscope. By P100, RPE cell number is within the range found in adults. New RPE cells are generated in a peripheral band which moves outwards as cells leave the cell cycle in more central locations. RPE cells thus complete their last cell division in a centre-to-periphery wave centred about the optic nerve head. At any given retinal location, RPE cells complete their last cell division earlier than the overlying layers of the neural retina. Cells of the RPE rapidly develop a mature morphology. For example, melanin granules are observed at P5 and Verhoeff's membrane (the terminal bar complex) is evident by P25. By contrast, photoreceptor development in this species is protracted; cone inner segments are observed by P40, whilst the first rod inner segments are observed at P60. Despite being generated earlier, morphological maturation of the cones appears retarded and prolonged compared with that of the rods. The last stages of RPE cell maturation occur late in development, in synchrony with the generation of rods.

The pecten oculi of the American crow (Corvus brachyrhynchos) has been examined by both light and electron microscopy. In this species the pecten is very large and of the pleated type. It consists of 22-25 accordion folds that are joined apically by a bridge of tissue which holds the pecten in a fan-like shape widest at its base. Within each fold are numerous capillaries, larger supply and drainage vessels and many melanocytes. The capillaries are extremely specialized for transport functions and display extensive microfolds on both their luminal and abluminal borders. Except for the nuclear region which contains most of the organelles, the endothelial cell bodies are extremely thin. These capillaries are surrounded by thick fibrillar basal laminae which are conjectured to be structurally important. Pericytes are a common feature of these capillaries. The numerous pleomorphic melanocytes interspersed between the capillaries are also felt to be important in structural support of the pecten. The pecten is considered to be comparable to the falciform process of some teleosts, the conus papillaris of reptiles, the supraretinal vessels of amphibians and teleosts and the intraretinal vessels of mammals which are all felt to be alternative methods of bringing nutrients to the inner retina.

Summary The fine structure of the retinal epithelial (RPE) region has been investigated by light and electron microscopy in the American crow (Corvus brachyrhynchos). In this species the RPE consists of a single layer of cuboidal cells which display numerous deep basal (scleral) infoldings and plentiful apical (vitreai) microvillar processes which surround photoreceptor inner and outer segments. The RPE cells are joined laterally by a series of tight junctions (VERHOEFF'S membrane) located in the mid to basal region. Within the epithelial cells, smooth endoplasmic reticulum is very abundant while rough endoplasmic reticulum (RER) is scarce. Mitochondria of various shapes are abundant basally while polysomes are plentiful and widespread. In the light-adapted state RPE nuclei are large and vesicular and basally located while the melanosomes of these cells are predominantly located with the apical processes indicating photomechanical movements. Myeloid bodies are large and numerous and often have ribosomes on their outer surface. Bruch's membrane (complexus basalis) is typical of avian species in that it is pentalaminate and the lamina densa is displaced near the cho-riocapillaris. The endothelium of the choriocapillaris is thin facing Bruch's membrane but is only moderately fenestrated. Some of these fenestrations display a double-layered diaphragm while the majority show the more typical single-layered diaphragm noted in most species.

The morphology of the retinal photoreceptors of the great blue heron (Ardea herodias) has been investigated by light and electron microscopy. They consist of rods, single cones and double (unequal) cones present in a ratio of about 2:1:1 respectively. The rods are slender elongated cells with outer segments that reach to the retinal epithelial (RPE) cells and are surrounded by pigment-rich apical processes of the RPE cells in the light-adapted state. The rod inner segment displays an ellipsoid of mitochondria, an hyperboloid of glycogen, much rough ER, numerous polysomes, Golgi zones and autophagic vacuoles. The rod nucleus is located deep in the outer nuclear layer and the rod synaptic pedicle displays both invaginated and superficial synaptic sites. Single cones display a slightly tapered outer segment, a large electron lucent oil droplet and an ellipsoid of mitochondria in the apex of the inner segment. Double cones consist of a long thin chief member which shows an electron dense oil droplet and a shorter, stouter accessory cone with no oil droplet but a paraboloid of glycogen below the ellipsoid. As in the single cone, polysomes, RER and Golgi zones are present in the myoid region of both members of the double cone. All photoreceptor types have a connecting cilium joining inner and outer segments. Near the external limiting membrane, the chief and accessory cones show intercellular junctions. All cone photoreceptors are relatively small in diameter and hence tightly packed. While rods are felt to undergo retinomotor movements, cones are felt to move minimally or not at all. Both single and double cones display several invaginated (ribbon) synapses as well as numerous superficial (conventional) synaptic sites.

Summary As part of a comparative morphological study, the fine structure of the retinal epithelium (RPE), choriocapillaris and Bruch's membrane (complexus basalis) has been studied by electron microscopy in the red-tailed hawk (Buteo jamaicensis). In this species the RPE consists of a single layer of low cuboidal cells which display numerous basal (scleral) infoldings and extensive apical (vitreal) processes which interdigitate with photoreceptor outer segments. These epithelial cells are joined laterally by a series of basally located tight junctions. Internally SER is the most abundant cell organelle while only small amounts of RER are present. Polysomes are however abundant as are mitochondria. The RPE cell nucleus is large and vesicular. Melanosomes are mainly located in the apical processes of the RPE cells in light-adaptation. Myeloid bodies are large and numerous in light-adaptation and often show ribosomes on their outer border. Bruch's membrane (complexus basalis) shows the typical pentalami-nate structure noted in most vertebrates but with only a poorly defined central elastic layer. The endothelium of the choriocapillaris is very thin facing the RPE but is only moderately fenestrated. The choriocapillaris in this species is unusual however in that many of the fenestrae show a double-layered diaphragm.

We conducted laboratory experiments to determine relationships between water turbidity, piscivory, fork length, and development of the retina in juvenile walleyes Stizostedion vitreum. Walleyes 85 mm and longer ate a greater weight and number of fathead minnows Pimephales promelas in turbid water than in clear water. Feeding was inhibited at the highest turbidities (100 and 161 nephelometric turbidity units, NTU) in 1-h feeding trials but not at the highest turbidity (121 NTU) in 4-h trials. In contrast, walleyes in the shorter length-group (≤75 mm) fed at approximately the same rate in clear and turbid trials, but consumption was inhibited at the two highest turbidities (100 and 161 NTU) used in 1-h trials. The tapetum lucidum was present in the retina of walleyes used in all the experiments. Aggregation of photoreceptor cells in the retina into groups of 20 to 30 (to form macroreceptors) begins when walleyes are approximately 60 mm long. The morphology of the retinal epithelium (RPE) and closely associated choriocapillaris and Bruch's membrane (complexus basalis) has been investigated in the eye of the domestic cat (felis catus) by light and electron microscopy. The RPE consists of a single layer of cuboidal cells joined laterally by apically-located junctional complexes. Basally (sclerally) these cells display numerous infoldings while apically (vitreally) two distinct types of processes enclose photoreceptor outer segments. Internally the large vesicular nucleus is centrally located. Smooth endoplasmic reticulum, mitochondria, polysomes and lysosome-like bodies are abundant. Rough endoplasmic reticulum while present is not plentiful. Over the tapetum lucidum, melanosomes are absent from the RPE cells while in non-tapetal locations melanosomes are large and plentiful. The endothelium of the choriocapillaris is highly fenestrated and these capillaries are deeply indented into the epithelial layer over the tapetum. Bruch's membrane (complexus basalis) is non-tapetal regions is the typical pentalaminate structure noted for most mammals. Over the tapetum however it is reduced to a trilaminate structure and when associated with the indented capillary profiles is further reduced to a single thickened basal lamina.

The morphology of the retinal photoreceptors of the domestic cat (Felis catus) has been investigated by light and electron microscopy. In this species the photoreceptors can be readily differentiated and adequately described by the classical terminology of rods and cones, with rods being the more numerous. Rods are long slender cells with inner and outer segments of much the same diameter. Cones are shorter cells with a conical outer segment that tapers distally and an inner segment which is wider than the outer segment. Both rods and cones are highly polarized and compartmentalized cells and consist of an outer segment, a connecting cilium, an inner segment, a nuclear region and a synaptic process leading to an expanded synaptic ending. Morphological similarities and differences are noted between the rods and cones for the various regions of these cells.

The pecten oculi of the mallard duck (Anas platyrhynchos) has been examined by light and electron microscopy. In this species, the pecten is of the pleated type and 12–14 accordion folds are joined apically by a heavily pigmented bridge of tissue which holds the pecten in a fanlike shape, widest at the base. It is situated over the optic nerve head and extends out into the vitreous. The entire pecten is enclosed by a fine basal lamina and hyalocytes are often present on its outer surface. Within each fold are numerous capillaries, larger blood vessels, and melanocytes. The capillaries are surrounded by thick fibrillar basal laminae which often contain pericytes. These capillaries display extensive microfolds on both the luminal and abluminal borders. The endothelial cell bodies are extremely thin, with most organelles present in a paranuclear location. The melanocytes, which are most plentiful in the bridge region, form an incomplete sheath around the capillaries and other blood vessels. The morphology of the pecten in the mallard is indicative of a heavy involvement in the transport of materials.

Introduction: Recent studies in sheep have shown that repeated maternal injections of betamethasone are associated with adverse effects within the nervous system. Repeated fetal injections of betamethasone achieve serial improvements in preterm lung function in sheep and are a possible alternative to repeated maternal therapy. We have evaluated the effect of repeated fetal administration of betamethasone on nervous system maturation in an ovine model. Methods: Date-mated ewes In = 48) were randomized to receive ultrasound-guided fetal injections of beta-methasone or saline between days 104 to 124 of gestation and were delivered by cesarean section on day 125 or 145 (term = 150). Optic and sciatic nerves were prepared for light and electronmicroscopy. Eye diameters were measured and transverse sections of retinae were evaluated. Data were analyzed using a mixed model analysis of variance. Results: Repeated fetal administration of corticosteroid did not significantly affect optic nerve myelination but resulted in significant delays in sciatic axonal growth (p < 0.02) and retinal maturation (p < 0.04). The process of performing repeated fetal injections also significantly affected some retinal parameters. Conclusion: Repeated fetal administration of betamethasone alters some aspects of nervous system maturation in sheep. It is premature to plan trials of repeated fetal corticosteroid therapy in humans.

The choroidally located tapetum lucidum of the Port Jackson shark (Heterodontus phillipi) was examined by light and electron microscopy in light-adapted specimens. In this species the tapetum consists of a single layer of overlapping cells oriented at an angle of about 30 degrees to the incoming light and situated immediately external to the choriocapillaris. These tapetal cells alternate with and are separated from one another by melanocytes which extend beyond the tapetal cells to intervene between the tapetal cells and the incoming light. The tapetal cells and the melanocytes are flattened plate-like cells with their widest dimension facing the retina. Internally the tapetal cells display a peripherally located vesicular nucleus with most organelles in a paranuclear location. The bulk of a tapetal cell is packed with regularly spaced crystals reported to be guanine. The size and spacing of these reflective crystals is commensurate with the principles of constructive interference. In light adaptation, the melanosomes of the intervening melanocytes are widely dispersed and for the most part block the passage of light to the tapetal cells. Although dark-adapted specimens were not available, it seems reasonable to assume that in dark adaptation these melanosomes will retreat to unmask the tapetum and allow it to function as a known reflective layer.

The rod inner segment displays an ellipsoid of mitochondria, a large hyperboloid of glycogen, much RER, numerous polysomes, Golgi zones and autophagic vacuoles. Single cones show a slightly tapered outer segment, a large and usually heterogeneous oil droplet and an ellipsoid of mitochondria at the apex of the inner segment. Double cones consist of a longer, stouter chief member which displays a more homogeneous oil droplet and a prominent paraboloid of glycogen and a slightly shorter and thinner accessory member with no oil droplet or paraboloid. Both members of the double cone as well as the single cones show a prominent ellipsoid and plentiful polysomes, RER and Golgi zones in the inner segment. Along the length of the contiguous membranes of the two members of the double cone are presumed interreceptor junctions. All cone photoreceptors are relatively small in diameter and hence are tightly packed. Judging by their morphology in the light-adapted state neither rods nor cones are felt to undergo photomechanical movements in this species. Rods and cones (both types) display both invaginated (ribbon) synapses as well as numerous flat (conventional) synaptic sites.

Phagosomes are present, rough endoplasmic reticulum is scarce and myeloid bodies were not observed. Melanosomes are absent over the choroidally located tapetum lucidum, but are not abundant even in extratapetal areas. This paucity of melanosomes probably makes retinomotor movements unimportant. Bruch's membrane or complexus basalis is a pentalaminate structure. The endothelium of the choriocapillaris is thin but minimally fenestrated.

Both single and double cones display several invaginated synapses as well as numerous superficial synaptic sites.

The structure and arrangement of the retinal photoreceptors of the velvet cichlid fish (Astronotus ocellatus) have been studied by light and electron microscopy. Rods, single cones and double (twin) cones are present. In the light-adapted state, rods are very tall cells that reach deep into the retinal epithelial (RPE) layer. The long outer segment is composed of discs of uniform diameter displaying one or two incisures. The rod inner segment shows a distal ellipsoid of mitochondria, and then narrows dramatically in the myoid region. Rod nuclei are electron dense and located deep in the outer nuclear layer. Rod synaptic spherules are small and show two to three invaginated synaptic sites as well as superficial synapses. Single cones are similar to the individual members of a double cone and all display a short tapering outer segment, a large ellipsoid of mitochondria and a myoid rich in rough endoplasmic reticulum, polysomes, Golgi zones and autophagic vacuoles. Double cones have extensive subsurface cisternae along their entire contiguous surfaces. Cone nuclei are large and vesicular and located close to or through the external limiting membrane. The synaptic pedicles of cones are larger, more electron lucent, and display more invaginated (ribbon) synapses as well as conventional (superficial) synaptic sites than do the rod spherules. Rod photoreceptors certainly undergo retinomotor movements and it is probable that cones do as well. In the light-adapted state the cone photoreceptors are arranged in a repeating square mosaic pattern with one single cone surrounded by four double (twin) cones.

The morphology of the retinal pigment epithelium (RPE), choriocapillaris and Bruch's membrane (complexus basalis) have been studied by light and electron microscopy in the velvet cichlid (Astronotus ocellatus). The RPE is composed of a single layer of large columnar cells. The basal (scleral) border of these cells is minimally infolded, whereas the apical (vitreal) surface displays numerous pigment-laden processes which in light-adaptation surround both rod and cone outer segments. Laterally the RPE cells are joined by a series of basally located tight junctions. Wandering phagocytes are a constant feature within this epithelial membrane. The RPE cells display a large, vesicular nucleus, numerous mitochondria, much smooth endoplasmic reticulum, polysomes, myeloid bodies, phagosomes and melanosomes. Rough endoplasmic reticulum is relatively scarce within these cells. Although only light-adapted specimens were examined, it is thought that the melanosomes are capable of extensive retinomotor movement. The endothelium of the choriocapillaris facing Bruch's membrane is typically very thin but shows few fenestrations. Bruch's membrane is typical of other teleost species in that it is composed of only three layers.

The melanocytes which are most plentiful in the bridge region and peripherally in the pecten, form an incomplete sheath around the capillaries and other blood vessels. These melanocytes are also felt to be fulfilling a structural role within the pecten. The morphology of the pecten of the red-tailed hawk is indicative of a heavy involvement in the transport of materials to the avascular avian retina.

The fine structure of the choriocapillaris, Bruch's membrane and retinal epithelium was investigated by light and electron microscopy in both the tapetal and non-tapetal fundus of the domestic sheep. The choriocapillaries are heavily fenestrated on the side facing the retina. Overlying the tapetum, the choriocapillaris also displays fenestrae on the choroidal border. Bruch's membrane is pentalaminate throughout the retina and slightly thicker over the tapetal region. In all locations the retinal epithelium consists of a single layer of cells, which vary in height depending upon their location. The epithelial cells are joined laterally by apically located tight junctions and throughout the retina display numerous basal infoldings and apical processes which enclose rod outer segments. The epithelial cell nucleus is large and vesicular. All retinal epithelial cells are rich in smooth endoplasmic reticulum and phagosomes. Although not as abundant, mitochondria, Golgi zones, profiles of rough endoplasmic reticulum and polysomes are also noted in all locations. In non-tapetal areas, melanosomes are numerous whereas over the central tapetum fibrosum they are absent. While lysosomes are pressent throughout the epithelial layer, over the tapetal area they appear to be more numerous. The absence of melanosomes over a functional tapetum fibrosum is to be expected. The apparent increase in lysosomal numbers in this location may indicate an enhanced shedding of outer segment material over the tapetal region. Although some retinal epithelial cells are modified to accommodate a tapetum lucidum their morphology is basically similar throughout the retina and probably indicates that all regions of the retinal epithelium are capable of the normal functions of this indispensible retinal layer.

The structure of the retinal photoreceptors of the domestic pig has been studied by both light and electron microscopy. In this species the photoreceptors can be traditionally and adequately differentiated into rods and cones, with rods being the more numerous. Rods are long, slender cells, while cones are shorter and stouter. Both rods and cones consist of an outer segment, a connecting cilium, an inner segment, a nuclear region and a synaptic process leading to a synaptic ending. Differences are noted between rods and cones for the various regions of these cells. What are presumed to be the displaced cell bodies of both rods and horizontal cells are often noted within the outer plexiform layer of this species.

The retinal pigment epithelial (RPE) layer of a nocturnally active bird, the nighthawk (Chordeiles minor), has been investigated by electron microscopy. This layer is composed of a single layer of low cuboidal cells joined laterally by tight junctions. Basally (sclerally), the epithelial cells display deep infoldings while apically (vitreally) numerous processes enclose photoreceptor outer segments. Internally, the nucleus is large, vesicular and basally located. Smooth endoplasmic reticulum and mitochondria are abundant. Rough endoplasmic reticulum (RER) is scarce but numerous polysomes are present. Myeloid bodies are widespread and often appear to be continuous with the RER membranes. Phagosomes of outer segment material are present but not numerous in the light-adapted condition. Large spindle-shaped melanosomes as well as numerous rounded tapetal spheres are present in the apical region and apical processes of the epithelial cells. The composition of the reflective tapetal spheres is unknown. Bruch's membrane (complexus basalis) in most locations displays a pentalaminate appearance while in some regions it is trilaminate. The endothelium of the choriocapillaris facing Bruch's membrane is extremely thin but only sparsely fenestrated.

The retinal photoreceptors of the goldeye (Hiodon alosoides) are arranged in large bundles of 40–50 cells optically isolated from other bundles by the retinal epithelial cells. Within each bundle are found both rods and cones in roughly equal numbers. Rod photoreceptors show marked retinomotor responses to project beyond the photoreceptor bundle in light-adaptation and to lie entirely within the bundle in dark-adaptation. In all stages of the light cycle cone outer segments remain at the apex of the photoreceptor bundle. In light-adaptation, rod inner segments display an apical ellipsoid separated from a basal ellipsoid by the greatly elongated myoid. In dark-adaptation the rod inner segment is much the same diameter throughout its length. In both rods and cones, profiles of rough and smooth endoplasmic reticulum and Golgi zones are present in a supranuclear location. The nuclei of rods display little heterochromatin and are located vitreal to the external limiting membrane in light-adaptation, whereas in dark-adaptation more heterochromatin is noted and the nuclei lie scleral to the external limiting membrane. Cone nuclei display the same changes in chromatin pattern as rods but they show changes in nuclear location opposite that of the rods. Throughout its length, the rod photoreceptor cytoplasm is more electron dense than that of the cone. The synaptic spherule, of rods displays 2–3 invaginated synaptic, sites while the cone pedicle is larger and presents 8–10 invaginated synaptic sites. Both rods and cones also appear to have superficial synaptic sites. Membrane specializations are found along the length of the inner segments where rods and cones are contiguous. These may act as sites of intercellular communication and the whole photoreceptor bundle may therefore be considered, as a macroreceptor.

The fine structure of the retinal epithelium of the goldeye (Hiodon alosoides) has been investigated with electron microscopy in both the light- and dark-adapted condition. The retinal epithelial cells are extremely tall columnar cells. They form a single layer abutting on Bruch's membrane basally (sclerally) and interdigitating with bundles of photoreceptor cells apically (vitreally). The retinal epithelial cells and the photoreceptor bundles are arranged in a hexagonal fashion so that each epithelial cell normally contacts three photoreceptor bundles and three other epithelial cells. In the light-adapted state, melanosomes are sparsely but evenly distributed throughout the cell body except for the apices, which contain more melanosomes. In dark-adaptation, melanosomes become very concentrated in the cell apex between photoreceptor bundles, with some accumulation of melanosomes also noted basally. The predominant feature of these epithelial cells is the presence of numerous reflective bodies which act as a retinal tapetum lucidum. During light-adaptation these reflective bodies are scattered throughout the cell but in dark-adaptation they accumulate centrally. In the body of the epithelial cell these reflective particles are diffusely arranged, whereas they are regularly arranged where epithelial cells ensheath photoreceptor bundles. Circadian changes are also noted in the morphology of the myeloid bodies and mitochondria and in the location of the epithelial cell nucleus. Wandering phagocytes are present at the retinal epithelial-photoreceptor interface. Bruch's membrane is thick and trilaminate and the choriocapillaris endothelium is thin and sparsely fenestrated.

Summary The fine structure of the tapetum lucidum of the domestic cow was studied by electron microscopy. In this ungulate the reflective layer is a tapetum fibrosum situated in the choroid and mainly restricted to the superior fundus. The tapetum is composed of a large array of fine extracellular collagen fibrils separated into lamellae by fibrocytes. The collagen fibrils display the normal cross-striations of native collagen, are 0.2 μm in diameter and are arranged in a hexagonal pattern with a center-to-center spacing of about 0.2 μm. The diameter and spacing of these fibrils is consistent with the constructive interference of reflected light. A vascular lamella adjacent to the retinal epithelial layer contains the choriocapillaris and some irregularly arranged collagen fibrils. The tapetum is pierced by blood vessels which supply the choriocapillaris. Over the tapetum the retinal epithelium is non-pigmented while in nontapetal regions this layer is pigmented. The choriocapillaris is not indented into the retinal epithelium as is the case in species with a tapetum cellulosum.

The fine structure and arrangement of the photoreceptor cells of the archerfish (Toxotes jaculatrix) have been studied by electron microscopy. Rods, twin cones, and single cones are present. In the light-adapted state, the rods are very tall cells reaching almost to the base of the retinal epithelial cells. The outer segment is composed of membraneous discs of uniform diameter displaying a single incisure. The rod inner segment displays a distal small ellipsoid and an extremely thin myoid region. The nuclei of rods are electron-dense, and the synaptic spherule displays two or three invaginated sites. The single cone is similar to the individual members of a twin cone and displays a tapering outer segment and accompanying accessory outer segment. The wider cone inner segment contains a large, centrally loated ellipsoid and a peripheral region rich in endoplasmic reticulum, polysomes, and microfilaments. Twin cones display subsurface cisternae along their entire contiguous surfaces. The cone nuclei are large and vesicular and located vitread to the external limiting membrane. The synaptic pedicle of cones is lager and moe electron-lucent and contains more invaginated “ribbon” synaptic sites (ten to 12) than do rods. In addition, small “coated” invagination and larger synaptic vesicle-filled processes are also seen within cone pedicles. In the light-adapted state the cone photoreceptors are arranged in a repeating square mosaic pattern with one single cone surrounded by four twin cones.

The fine structure of the choriocapillaris, Bruch's membrane and retinal epithelium was investigated by light and electron microscopy in both the tapetal and non-tapetal fundus of the domestic sheep. The choriocapillaries are heavily fenestrated on the side facing the retina. Overlying the tapetum, the choriocapillaris also displays fenestrae on the choroidal border. Bruch's membrane is pentalaminate throughout the retina and slightly thicker over the tapetal region. In all locations the retinal epithelium consists of a single layer of cells, which vary in height depending upon their location. The epithelial cells are joined laterally by apically located tight junctions and throughout the retina display numerous basal infoldings and apical processes which enclose rod outer segments. The epithelial cell nucleus is large and vesicular. All retinal epithelial cells are rich in smooth endoplasmic reticulum and phagosomes. The normal morphology of the retinal epithelium and tapetum lucidum has been studied by transmission electron microscopy in the eye of a teleost; the giant danio (Danio malabaricus). The retinal epithelium forms a single layer of cells joined laterally by cell junctions. Centrally in the retina these cells are tall columnar, while more peripherally they become more cuboidal in shape. Apical processes of these cells enclose photoreceptor inner and outer segments. Few basal (scleral) infoldings are seen in the epithelial cells. Throughout the epithelial layer the cells display smooth endoplasmic reticulum, numerous mitochondria and phagosomes. Within the epithelial cells centrally located in the retina are found numerous lipid spheres which act as a tapetum lucidum. Only at the extreme periphery are these reflective spheres scarce or absent from the epithelial cells. Melanosomes, although found throughout the epithelial layer, are scarce centrally and more numerous peripherally. A population of wandering phagocytes appears to be a normal feature within the retinal epithelial layer of this species. Bruch's membrane shows a trilaminate composition rather than the usual pentalaminate structure seen in most vertebrates. The choriocapillaris endothelium facing Bruch's membrane is very thin but only minimally fenestrated.

The structure of the retinal pigment epithelium, choriocapillaris and Bruch's membrane has been studied by electron microscopy in the archerfish Toxotes jaculatrix. The pigment epithelium is composed of a single layer of large columnar cells. Each epithelial cell displays numerous small mitochondria, smooth endoplasmic reticulum, myeloid bodies, phagosomes and melanosomes. Rough endoplasmic reticulum and ribosomes while present are not abundant. The scleral or basal border of the epithelial cells is but minimally infolded whereas the vitreal or apical surface displays many elongated processes which surround the inner and outer segments of the photoreceptors. Laterally the epithelial cells are joined by a series of basally located cell junctions. Unattached phagocytic cells are a constant feature both between the retinal epithelial cells and within Bruch's membrane. The endothelium of the choriocapillaris bordering Bruch's membrane is typically very thin with a few fenestrations. Bruch's membrane lacks a central elastic layer and is composed of only three layers. Although only light-adapted specimens were examined, it is felt that the melanosomes are capable of retinomotor movements.

Morphogenesis of the retina of the Sprague-Dawley albino rat was studied by light microscopy from day 11 of gestation until 225 days after birth. A quantitative analysis during development of retinal volume, thickness of the entire neural retina and thickness of each of the retinal layers, both posteriorly and peripherally, was made. The results indicate that initially a single neuroblastic layer forms and continually thickens by mitosis at its outer border. The retinal layers then form in sequence, moving from the inner retinal border outward and always beginning posteriorly and then spreading peripherally. The transient layer of Chievitz does not appear. All adult layers are present by eight days after birth and each layer thins after reaching its maximal thickness. Total thickness of the retina excluding pigmented epithelium, is greatest on postnatal day 5, but retinal volume only reaches a peak on postnatal days 7 to 12. The nerve fiber, inner plexiform, outer plexiform and bacillary layers all continue to increase in thickness after the ganglion cell and inner and outer nuclear layers reach their maximal width and are beginning to become thinner.
