The purpose of this study is to define the geographic boundaries of commercial areas by creating a consistent definition, combining various commercial area types, including downtowns, retail centres, financial districts, and other employment subcentres. Our research involved the collection of office, retail and job density data from 69 metropolitan regions across USA and Canada. Using this data, we conducted an unsupervised image segmentation model and clustering methods to identify distinctive commercial geographic boundaries. As a result, we identified 23,751 commercial areas, providing a detailed perspective on the commercial landscape of metropolitan areas in the USA and Canada. In addition, the generated boundaries were successfully validated through comparison with previously established commerce-related boundaries. The output of this study has implications for urban and regional planning and economic development, delivering valuable insights into the overall commercial geography in the region. The commercial boundary and used codes are freely available on the School of Cities Github, and users can reuse, reproduce and modify the boundaries.

In 2007, the Japanese Ministry of Education revised the Education Law to pave the way for the introduction of a teaching license renewal system in 2009. The reform required all teachers to complete at least 30 hours of training in specially tailored university courses as a prerequisite for renewal of their teaching licenses every 10 years. In late 2021, however, the government abruptly announced it was ending the scheme in 2022 in a policy about face reminiscent of the abolition of "yutori kyoiku" ("education without pressure") in 2007. This qualitative study aims to examine the background, outline and issues of Japan's license renewal system in order to understand the reasons for its demise and illustrate the similarities with the "yutori kyoiku" experiment. Drawing on interview data and reflections from the author's experiences with the system over 12 years, it looks at some of Japan's unique training opportunities and, employing examples from other countries, examines what kind of 'training' is needed moving forward. It concludes by calling for a widening of the interpretation of 'training,' and indeed, the license itself, so as to increase diversity in what is a considerably restrictive employment sector.

J-domain proteins (JDPs) are the largest family of chaperones in most organisms, but much of how they function within the network of other chaperones and protein quality control machineries is still an enigma. Here, we report on the latest findings related to JDP functions presented at a dedicated JDP workshop in Gdansk, Poland. The report does not include all (details) of what was shared and discussed at the meeting, because some of these original data have not yet been accepted for publication elsewhere or represented still preliminary observations at the time.

Abstract Over the past five decades, a large number of wild animals have been individually identified by various observation systems and/or temporary tracking methods, providing unparalleled insights into their lives over both time and space. However, so far there is no comprehensive record of uniquely individually identified animals nor where their data and metadata are stored, for example photos, physiological and genetic samples, disease screens, information on social relationships. Databases currently do not offer unique identifiers for living, individual wild animals, similar to the permanent ID labelling for deceased museum specimens. To address this problem, we introduce two new concepts: (1) a globally unique animal ID (UAID) available to define uniquely and individually identified animals archived in any database, including metadata archived at the time of publication; and (2) the digital ‘home’ for UAIDs, the Movebank Life History Museum (MoMu), storing and linking metadata, media, communications and other files associated with animals individually identified in the wild. MoMu will ensure that metadata are available for future generations, allowing permanent linkages to information in other databases. MoMu allows researchers to collect and store photos, behavioural records, genome data and/or resightings of UAIDed animals, encompassing information not easily included in structured datasets supported by existing databases. Metadata is uploaded through the Animal Tracker app, the MoMu website, by email from registered users or through an Application Programming Interface (API) from any database. Initially, records can be stored in a temporary folder similar to a field drawer, as naturalists routinely do. Later, researchers and specialists can curate these materials for individual animals, manage the secure sharing of sensitive information and, where appropriate, publish individual life histories with DOIs. The storage of such synthesized lifetime stories of wild animals under a UAID (unique identifier or ‘animal passport’) will support basic science, conservation efforts and public participation.

Abstract Coral reef ecosystems are highly threatened and can be extremely sensitive to the effects of climate change. Multiple shark species rely on coral reefs as important habitat and, as such, play a number of significant ecological roles in these ecosystems. How environmental stress impacts routine, site-attached reef shark behavior, remains relatively unexplored. Here, we combine 8 years of acoustic tracking data (2013-2020) from grey reef sharks resident to the remote coral reefs of the Chagos Archipelago in the Central Indian Ocean, with a satellite-based index of coral reef environmental stress exposure. We show that on average across the region, increased stress on the reefs significantly reduces grey reef shark residency, promoting more diffuse space use and increasing time away from shallow forereefs. Importantly, this impact has a lagged effect for up to 16 months. This may have important physiological and conservation consequences for reef sharks, as well as broader implications for reef ecosystem functioning. As climate change is predicted to increase environmental stress on coral reef ecosystems, understanding how site-attached predators respond to stress will be crucial for forecasting the functional significance of altering predator behavior and the potential impacts on conservation for both reef sharks and coral reefs themselves.

This article considers the related concepts of 'rehearsal' [Harney, Stefano, and Fred Moten. 2013. "The Undercommons: Fugitive Planning & Black Study." Brooklyn: Minor Compositions] and 'catastrophe' [Stengers, Isabelle. 2015. "In Catastrophic Times: Resisting the Coming Barbarism." Translated by Andrew Goffey. London: Open Humanities Press] as activism, focussing upon eco-somatic drama processes engaged through "In Your Arms" (IYA). This participatory arts project for babies and their primary caregivers was co-created in Naarm (Melbourne), Australia, by a four-person artist ensemble, ten pre-crawling babies, and their mothers. Multi-modalities were used, including improvisatory movement scores, natural props, sound-making and listening practices. The article suggests new ways to consider drama activity at a micro-sensorial scale, offering practices that cultivate environmental attunement and care from the very first months of life.

Abstract Background Neuroblastoma is a common pediatric cancer, where preclinical studies suggest that a mesenchymal-like gene expression program contributes to chemotherapy resistance. However, clinical outcomes remain poor, implying we need a better understanding of the relationship between patient tumor heterogeneity and preclinical models. Results Here, we generate single-cell RNA-seq maps of neuroblastoma cell lines, patient-derived xenograft models (PDX), and a genetically engineered mouse model (GEMM). We develop an unsupervised machine learning approach (“automatic consensus nonnegative matrix factorization” (acNMF)) to compare the gene expression programs found in preclinical models to a large cohort of patient tumors. We confirm a weakly expressed, mesenchymal-like program in otherwise adrenergic cancer cells in some pre-treated high-risk patient tumors, but this appears distinct from the presumptive drug-resistance mesenchymal programs evident in cell lines. Surprisingly, however, this weak-mesenchymal-like program is maintained in PDX and could be chemotherapy-induced in our GEMM after only 24 h, suggesting an uncharacterized therapy-escape mechanism. Conclusions Collectively, our findings improve the understanding of how neuroblastoma patient tumor heterogeneity is reflected in preclinical models, provides a comprehensive integrated resource, and a generalizable set of computational methodologies for the joint analysis of clinical and pre-clinical single-cell RNA-seq datasets.

Abstract Animal-borne tags are effective instruments for collecting ocean data and can be used to fill spatial gaps in the observing network. We deployed the first conductivity, temperature, and depth (CTD) satellite tags on the dorsal fin of salmon sharks (Lamna ditropis) to demonstrate the potential of sharks to monitor essential ocean variables and oceanographic features in the Gulf of Alaska. Over 1360 km and 36 days in the summer of 2015, the salmon shark collected 56 geolocated, temperature-salinity profiles. The shark swam through a plume of anomalously salty water that originated from the “Blob” and encountered several mesoscale eddies, whose subsurface properties were altered by the marine heatwave. We demonstrate that salmon sharks have the potential to serve as submesoscale-resolving oceanographic platforms and substantially increase the spatial coverage of observations in the Gulf of Alaska.

This report highlights risk and prioritization factors for housing developments with expiring affordability protections, focused on preserving transit-accessible affordable housing. It presents a regional framework for identifying and preserving subsidized affordable housing in the Southern California Association of Governments (SCAG) region (Los Angeles, Imperial, Orange, Riverside, San Bernardino, and Ventura counties). First, we analyze data from the California Housing Partnership (CHPC) and the National Housing Preservation Database (NHPD) to understand risk factors for expiring housing units, and design a prioritization tool for entities in the region to use when prioritizing developments to focus preservation and anti-displacement efforts. Second, we highlight affordable housing preservation policy solutions and characteristics unique to the Southern California context. Third, we draw on the strategies and experiences of other jurisdictions in the United States with experience strategizing around affordable housing preservation efforts to present key lessons and takeaways.

In University life, the first women to achieve status or leadership positions inevitably are perceived as role models to their colleagues, peers and students. Role model theory suggests that although we generally understand the meaning of terms such as "role model" or "mentor", the position of role model has several dimensions and is distinct from mentor. This case study on Professor H.Y. Izan, the first woman professor of finance in Australia and New Zealand, adopts the role model theoretical lens to examine Izan as trailblazer in the context of the eighteen women professors appointed after her during the period 1990-2021.

ABSTRACT In long‐distance dispersal events, colonising species typically begin with a small number of founding individuals. A growing body of research suggests that establishment success of small founding populations can be determined by the context of the colonisation event and the new environment. Here, we illuminate the importance of these sources of context dependence. Using a spatially explicit, temporally dynamic, mechanistic, individual‐based simulator of a model amphibian species, the cane toad (Rhinella marina), we simulated colonisation scenarios to investigate how (1) the number of founding individuals, (2) the number of dispersal events, (3) landscape's spatial composition and configuration of habitats (‘spatially heterogeneous landscapes’) and (4) the timing of arrival with regards to dynamic environmental conditions (‘dynamic environmental conditions’) influence the establishment success of small founding populations. We analysed the dynamic effects of these predictors on establishment success using running‐window logistic regression models. We showed establishment success increases with the number of founding individuals, whereas the number of dispersal events had a weak effect. At ≥ 20 founding individuals, propagule size swamps the effects of other factors, to whereby establishment success is near‐certain (≥ 90%). But below this level, confidence in establishment success dramatically decreases as number of founding individuals decreases. At low numbers of founding individuals, the prominent predictors are landscape spatial heterogeneity and dynamic environmental conditions. For instance, compared to the annual mean, founding populations with ≤ 5 individuals have up to 18% higher establishment success when they arrive in ‘packed’ landscapes with relatively limited and clustered essential habitats and right before the breeding season. Accounting for landscape spatial heterogeneity and dynamic environmental conditions is integral in understanding and predicting population establishment and species colonisation. This additional complexity is necessary for advancing biogeographical theory and its application, such as in guiding species reintroduction efforts and invasive alien species management.

White sharks (Carcharodon carcharias) are among the most widespread, charismatic, and studied predators in the ocean. However, their conservation status is concerning in many ocean sectors, most notably the Mediterranean Sea, which hosts one of the least known and most endangered populations globally. Though they were historically abundant and widely distributed in the region, Mediterranean white sharks have declined to dangerously low abundance levels, impacted by centuries of coastal and, more recently, industrial fishing. The IUCN lists this species as Critically Endangered in the region, but information about its current abundance and ecology is scarce, hindering effective management and conservation. Here, we describe our initial effort to find and track the remaining Mediterranean white sharks and report what we have learned from these activities. In 2021-2023, we conducted three pilot expeditions in the Sicilian Channel, covering four major sites, collecting 159 eDNA samples, and carrying out 359 hrs of pelagic mid-water baited video surveys, 43 hours of deep-water benthic baited video surveys, and 111 hrs of fishing. Baited video surveys detected 42 species of bony fishes, elasmobranchs, marine mammals, and turtles. We detected white sharks at four sites from eDNA samples. Though we did not observe white sharks directly, these activities supported the identification of one of the last strongholds of this population in the region and started a multi-institutional white shark conservation program in the Mediterranean Sea, aiming to track the last white sharks in the region, estimate their abundance and extinction risk, characterize the species’ ecology and inform management and conservation.

Fusobacterium nucleatum as a Gram-negative anaerobe plays a key bridging role in oral biofilms. It is involved in periodontal and extraoral diseases, the most prominent being colorectal cancer. Five subspecies are recognised: animalis, fusiforme, nucleatum, polymorphum and vincentii. Subspecies interact with neutrophils constantly patrolling tissues to remove microbial intruders. Neutrophil antimicrobial activities include generation of reactive oxygen species (ROS), formation of neutrophil extracellular traps (NETs) and release of cytokines and neutrophil enzymes. Subspecies-specific differences in immunogenicity have previously been observed in a neutrophil-like cell line but were not investigated in human neutrophils. Additionally, neutrophil responses to planktonic and biofilm-grown F. nucleatum have not been studied to date. The aims of this study were to compare the immunogenicity of planktonic and biofilm-grown F. nucleatum and to investigate potential differences in human neutrophil responses when stimulated with individual F. nucleatum subspecies. Human neutrophils isolated from peripheral blood were stimulated with planktonic and biofilm-grown F. nucleatum subspecies. Generation of ROS and NET formation were quantified by luminescence and fluorescence assays, respectively. Secretion of cytokines (IL-1β, TNF-α, IL-6, IL-8), neutrophil elastase and matrix metalloproteinase-9 was quantified by enzyme-linked immunosorbent assay (ELISA). Neutrophil responses showed biofilm-grown bacteria induced a significantly higher total and intracellular ROS response, as well as shorter time to total ROS release. Biofilm-grown F. nucleatum led to significantly lower IL-1β release. We found significant differences among individual subspecies in terms of total, intracellular ROS and extracellular superoxide. Subspecies polymorphum stimulated the highest mean amount of NET release. Amounts of cytokines released differed significantly among subspecies, while no differences were found in lysosomal enzyme release. Immunogenicity of F. nucleatum in human neutrophils is highly subspecies-specific in vitro with regard to ROS release and cytokine production. Understanding subspecies-specific immunogenicity of F. nucleatum may facilitate the discovery of novel therapeutic targets in F. nucleatum-mediated diseases.

The concept that “the bladder is an unreliable witness” highlights the non-disease-specific nature of lower urinary tract symptoms (LUTS), which can arise regardless of anatomical differences between genders. This non-specificity has led to the broader classification of symptoms under LUTS, which include storage, voiding, and post-micturition symptoms. Overactive bladder (OAB) is characterised by urgency, often accompanied by frequency, nocturia, and, in some cases, incontinence. The symptoms of OAB do not always correlate with urodynamically proven idiopathic detrusor overactivity (IDO), indicating that afferent nerve activity plays a significant role. The complexity of bladder control involves a neural network spanning the brain, spinal cord, and peripheral ganglia. Current therapies, such as anticholinergics and beta-3 agonists, target both motor and sensory pathways, with emerging evidence suggesting that sensory modulation is crucial for effective treatment. Research using animal models has provided insights into the pharmacological mechanisms underlying bladder function, although these models have limitations in replicating human conditions. The importance of the sensory ‘web’ within the bladder’s mucosal and interstitial cell network is emphasised, as it significantly influences bladder compliance and sensory signalling. Recent advances, including the use of Onabotulinum toxin A and sacral neuromodulation, highlight the therapeutic potential of targeting sensory pathways in managing OAB symptoms.

Clean intermittent self-catheterisation (CISC) is a very common and largely well-tolerated intervention for people with neurological and urological voiding dysfunction; however, catheter-associated urinary tract infections (CAUTIs) are a troublesome and potentially major complication. The main risk factors predisposing to CAUTI associated with CISC are poor patient compliance, failure to achieve complete bladder emptying resulting in residual urine, and microtrauma during catheterisation, potentially weakening the bladder’s defences against UTI. An innovative development in the design of CISC catheters is Micro-hole Zone Technology (MHZT) used in the novel Luja™ CISC catheter, which aims to overcome some of the problems associated with conventional CISC catheters. Through a narrative review of the literature undertaken by a multi-disciplinary panel of experts, specialists and patient advocate, who reviewed all major factors contributing to CAUTI and the potential benefits of MHZT catheters over conventional two-eyelet CISC catheters (CECs). MHZT catheters potentially confer the following advantages over CECs in male and female patients: (1) more effective bladder drainage in one continuous flow; (2) reduced risk of blockages by preventing the occlusion of the catheter end-eyelets by bladder mucosa; (3) reduced risk of bladder mucosal microtrauma; and (4) reduced intra-catheter pressure if any flow stop occurs, hence minimising the unpleasant dragging sensation. These benefits are likely to improve the patient’s comfort, quality of life and compliance. However, the panel acknowledged the lack of robust clinical data as to whether MHZT catheters reduced the incidence of CAUTI. Hence, comparative studies between MHZT and CECs are needed before definitive conclusions can be drawn.

AbstractBackground Periodontitis is a chronic inflammatory non-communicable disease (NCD) characterised by the destruction of the tooth-supporting apparatus (periodontium), including alveolar bone, the presence of periodontal pockets, and bleeding on probing.Objectives To outline, for family doctors, the implications of the association between periodontal and systemic diseases; to explore the role of family doctors in managing periodontitis as an ubiquitous non-communicable disease (NCD).Methods The consensus reports of previous focused collaborative workshops between WONCA Europe and the European Federation of Periodontology (using previously undertaken systematic reviews), and a specifically commissioned systematic review formed the technical papers to underpin discussions. Working groups prepared proposals independently, and the proposals were subsequently discussed and approved at plenary meetings.Results Periodontitis is independently associated with cardiovascular diseases, diabetes, chronic obstructive pulmonary disease, obstructive sleep apnoea, and COVID-19 complications. Treatment of periodontitis has been associated with improvements in systemic health outcomes. The article also presents evidence gaps. Oral health care professionals (OHPs) and family doctors should collaborate in managing these conditions, including implementing strategies for early case detection of periodontitis in primary medical care centres and of systemic NCDs in oral/dental care settings. There is a need to raise awareness of periodontal diseases, their consequences, and the associated risk factors amongst family doctors.Conclusion Closer collaboration between OHPs and family doctors is important in the early case detection and management of NCDs like cardiovascular diseases, diabetes mellitus, and respiratory diseases. Strategies for early case detection/prevention of NCDs, including periodontitis, should be developed for family doctors, other health professionals (OHPs), and healthcare funders. Evidence-based information on the reported associations between periodontitis and other NCDs should be made available to family doctors, OHPs, healthcare funders, patients, and the general population.

The COVID-19 pandemic accelerated three trends that were already transforming economic development theory and practice. A backlash to economic restructuring and inequality, driven by globalization and technology, is now manifesting in reshoring and union movements. The resurgence of small and midsized cities, originally driven by increasing housing costs in coastal cities, has been reinforced by a rise in remote work. The uncertainty of today's complex economy is exacerbating long-term challenges of tracking economic change, making “shoot anything that flies” more important than ever. These trends highlight the need to focus economic development on building and supporting the workforce.

The aquatic landscapes of the Sacramento–San Joaquin Delta (hereafter, the Delta) and Suisun Bay represent both a significant past and future soil carbon stock. Historical alterations of hydrologic flows have led to depletion of soil carbon stocks via emissions of carbon dioxide (CO2), and loss of elevation as a result of subsidence. Optimizing ecosystem hydrology in the Delta and Suisun Bay could both reduce and reverse subsidence while also providing significant opportunities for climate mitigation and adaptation. Emissions of greenhouse gases (GHGs)—notably CO2, methane (CH4 ), and nitrous oxide (N2O)—contribute to global warming at different rates and intensities, requiring GHG accounting and modeling to assess the relative benefits of management options. Decades of data collection, model building, and map development suggest that past and current management actions have both caused—and can mitigate—losses of soil carbon. We review here the magnitude of potential GHG offsets, management options that may be achievable, and trade-offs of carbon storage under different land management. Using a land-use/land-cover framework to assess these management options, we describe the potential of three interventions (impoundment to reverse subsidence, agricultural management, and tidal reintroduction and/or maintained connectivity), both in acreage and radiative balance to clarify their relative influence on the region’s GHG balance today and in relation to its millennial history. From floodplains to farming to floating aquatic vegetation, we find specific scalable strategies to manage hydrology that can alter regional GHG balance. Preservation of soil carbon stocks and restoration of net atmospheric CO2 fluxes into soils are the primary route to net negative emissions in the Delta and Suisun Bay, with CH4 emission management occurring in a supporting role. Over a 40-year horizon of climate-mitigation markets, the resilience of different aquatic habitats introduces the most uncertainty, from expected and unexpected hydrologic changes associated with land, ocean, and operational water flows.

Abstract: The expectation of a mass movement out of cities due to the rise of remote work associated with the Covid-19 pandemic, is counter to longstanding theories of the benefits of agglomeration economies. It suggests centrifugal shifts of economic activity which could boost neighbourhood economies at the expense of the downtown core. Using mobile phone data from SafeGraph, we track migration and daily mobility patterns throughout the New York metropolitan area between July 2019 and June 2021. We find that diverse suburban centres and exurban areas have bounced back more quickly than the dense specialised commercial districts in and around Manhattan.

The social environment is a key factor that influences behavioural traits across a wide array of species. Yet, when investigating individual differences in behaviour, studies tend to measure animals in isolation from other conspecifics-even in social species. Surprisingly, whether behavioural traits measured in isolation are predictive of individual-level behaviour when in social groups is still poorly understood. Here, we repeatedly measured risk-taking behaviour (i.e. boldness; 741 total trials) in both the presence and absence of conspecifics in a social lizard, the delicate skink (Lampropholis delicata). Further, we manipulated food availability during group trials to test whether the effect of the social environment on risk-taking behaviour was mediated by competition over resources. Using 105 lizards collected from three independent populations, we found that individual risk-taking behaviour was repeatable when measured in either social isolation or within groups both with and without food resources available. However, lizards that were bolder during individual trials were not also bolder when in groups, regardless of resource availability. This was largely driven by individual differences in social behavioural plasticity, whereby individual skinks responded differently to the presence of conspecifics. Together, this resulted in a rank order change of individual behavioural types across the social conditions. Our results highlight the importance of the social environment in mediating animal personality traits across varying levels of resource availability. Further, these findings suggest that behavioural traits when measured in isolation, may not reflect individual variation in behaviour when measured in more ecologically realistic social groups.

Overlap between marine megafauna and maritime activities is a topic of global concern. Basking sharks (Cetorhinus maximus; CM) are listed as Globally Endangered under the IUCN, though reported sightings appear to be increasing in Ireland. While such trends in the region are welcome, increasing spatiotemporal overlap between CM and numerous water users poses an increased risk of boat strikes to the animals. To demonstrate the risk and impact of boat strikes on marine megafauna, we present camera-enabled animal-borne inertial measurement unit (IMU) data from a non-lethal boat strike on a CM within a proposed National Marine Park in Ireland. We tagged a ~7-m female CM in County Kerry, Ireland, which was struck by a boat ~6 h after tag deployment. Comparison of pre-strike data with 4 h of video and ~7.5 h of IMU data following the boat strike provides critical insight into the animal’s response. While the CM reacted momentarily with an increase in activity and swam to the seafloor, it quickly reduced its overall activity (i.e., overall dynamic body acceleration, tailbeat cycles, tailbeat amplitude, and vertical velocity) for the remainder of the deployment. Notably, the animal also ceased feeding for the duration of the video and headed towards deep offshore waters, which is in stark contrast to the pre-strike period where the animal was consistently observed feeding along the surface in shallow coastal water. This work provides insight into a CM’s response to acute injury and highlights the need for appropriate protections to mitigate risks for marine megafauna.
