Abstract Smooth muscle cell (SMC) phenotypic modulation, primarily driven by PDGFRβ signaling, is implicated in occlusive cardiovascular diseases. However, the promotive and restrictive regulation mechanism of PDGFRβ and the role of protein tyrosine phosphatase non-receptor type 14 (PTPN14) in neointimal hyperplasia remain unclear. Our study observes a marked upregulation of PTPN14 in SMCs during neointimal hyperplasia. PTPN14 overexpression exacerbates neointimal hyperplasia in a phosphatase activity-dependent manner, while SMC-specific deficiency of PTPN14 mitigates this process in mice. RNA-seq indicates that PTPN14 deficiency inhibits PDGFRβ signaling-induced SMC phenotypic modulation. Moreover, PTPN14 interacts with intracellular region of PDGFRβ and mediates its dephosphorylation on Y692 site. Phosphorylation of PDGFRβY692 negatively regulates PDGFRβ signaling activation. The levels of both PTPN14 and phospho-PDGFRβY692 are correlated with the degree of stenosis in human coronary arteries. Our findings suggest that PTPN14 serves as a critical modulator of SMCs, promoting neointimal hyperplasia. PDGFRβY692, dephosphorylated by PTPN14, acts as a self-inhibitory site for controlling PDGFRβ activation.

Abstract The establishment of symbiotic interactions between leguminous plants and rhizobia requires complex cellular programming activated by Rhizobium Nod factors (NFs) as well as type III effector (T3E)-mediated symbiotic signaling. However, the mechanisms by which different signals jointly affect symbiosis are still unclear. Here we describe the mechanisms mediating the cross-talk between the broad host range rhizobia Sinorhizobium fredii HH103 T3E Nodulation Outer Protein L (NopL) effector and NF signaling in soybean. NopL physically interacts with the Glycine max Remorin 1a (GmREM1a) and the NFs receptor NFR5 (GmNFR5) and promotes GmNFR5 recruitment by GmREM1a. Furthermore, NopL and NF influence the expression of GmRINRK1, a receptor-like kinase (LRR-RLK) ortholog of the Lotus RINRK1, that mediates NF signaling. Taken together, our work indicates that S. fredii NopL can interact with the NF signaling cascade components to promote the symbiotic interaction in soybean.

Abstract Objective There is growing evidence supporting the utilization of the radial artery as a secondary arterial graft in coronary artery bypass grafting (CABG) surgery. However, debates continue over the recovery period of the radial artery following angiography. This study aims to evaluate the clinical outcomes and experiences related to the use of the radial artery post-angiography in total arterial coronary revascularization. Methods A retrospective analysis was performed on data from patients who underwent total arterial CABG surgery at the University of Hong Kong Shenzhen Hospital from July 1, 2020, to September 30, 2022. Preoperative assessments included ultrasound evaluations of radial artery blood flow, diameter, intimal integrity, and the Allen test. Additionally, pathological examinations of the distal radial artery and coronary artery CT angiography were conducted, along with postoperative follow-up to assess the safety and efficacy of using the radial artery in patients undergoing total arterial CABG. Results A total of 117 patients, compromising 102 males and 15 females with an average age of 60.0 ± 10.0 years, underwent total arterial CABG. The internal mammary artery was used in situ in 108 cases, while in 4 cases, it was grafted to the ascending aorta due to length limitations. Bilateral radial arteries were utilized in 88 patients, and bilateral internal mammary arteries in 4 patients. Anastomoses of the proximal radial arteries to the proximal ascending aorta included 42 cases using distal T-anastomosis and 4 using sequential grafts. The interval between bypass surgery and coronary angiography ranged from 7 to 14 days. Pathological examination revealed intact intima and continuous elastic membranes with no significant inflammatory infiltration or hyperplastic lumen stenosis in the radial arteries. There were no hospital deaths, 3 cases of perioperative cerebral infarction, 1 secondary thoracotomy for hemorrhage control, 21 instances of intra-aortic balloon pump (IABP) assistance, and 2 cases of poor wound healing that improved following debridement. CT angiography performed 2 weeks post-surgery showed no internal mammary artery occlusions, but 4 radial artery occlusions were noted. Conclusion Ultrasound may be used within 2 weeks post-angiography to assess the recovery of the radial artery in some patients. Radial arteries with intact intima may be considered in conjunction with the internal mammary artery for total arterial coronary CABG. However, long-term outcomes of these grafts require further validation through larger prospective studies.

Abstract Background The neonatal mammalian heart exhibits considerable regenerative potential following injury through cardiomyocyte proliferation, whereas mature cardiomyocytes withdraw from the cell cycle and lose regenerative capacities. Therefore, investigating the mechanisms underlying neonatal cardiomyocyte proliferation and regeneration is crucial for unlocking the regenerative potential of adult mammalian heart to repair damage and restore contractile function following myocardial injury. Methods The Tudor staphylococcal nuclease (Tudor-SN) transgenic (TG) or cardiomyocyte-specific knockout mice (Myh6-Tudor-SN −/− ) were generated to investigate the role of Tudor-SN in cardiomyocyte proliferation and heart regeneration following apical resection (AR) surgery. Primary cardiomyocytes isolated from neonatal mice were used to assess the influence of Tudor-SN on cardiomyocyte proliferation in vitro. Affinity purification and mass spectrometry were employed to elucidate the underlying mechanism. H9c2 cells and mouse myocardia with either overexpression or knockout of Tudor-SN were utilized to assess its impact on the phosphorylation of Yes-associated protein (YAP), both in vitro and in vivo. Results We previously identified Tudor-SN as a cell cycle regulator that is highly expressed in neonatal mice myocardia but downregulated in adults. Our present study demonstrates that sustained expression of Tudor-SN promotes and prolongs the proliferation of neonatal cardiomyocytes, improves cardiac function, and enhances the ability to repair the left ventricular apex resection in neonatal mice. Consistently, cardiomyocyte-specific knockout of Tudor-SN impairs cardiac function and retards recovery after injury. Tudor-SN associates with YAP, which plays important roles in heart development and regeneration, inhibiting phosphorylation at Ser 127 and Ser 397 residues by preventing the association between Large Tumor Suppressor 1 (LATS1) and YAP, correspondingly maintaining stability and promoting nuclear translocation of YAP to enhance the proliferation-related genes transcription. Conclusion Tudor-SN regulates the phosphorylation of YAP, consequently enhancing and prolonging neonatal cardiomyocyte proliferation under physiological conditions and promoting neonatal heart regeneration after injury. The immersed tunnel is considered an effective solution for traffic problems across rivers and seas. The sand filling layer, as an important part of immersed tunnel foundation treatments, directly affects segment attitude stability. Due to difficulties in quality control of concealed construction and the complex hydrodynamic environment, the sand filling layer is prone to compaction defects, further leading to changes in segment attitude. However, limited by structural concealment and state complexity, most studies consider the sand filling layer part of the foundation to study its impact on settlement while neglecting its influence on segment attitude. This research proposes an evaluation method for the sand filling layer state based on elastic wave testing and the elastic wave characteristic parameters selected come from analysis of the time domain, frequency domain and time–frequency domain. By classifying the elastic wave characteristic parameters through the K-means clustering method, the relationship between the state of the sand filling layer and the elastic wave characteristic parameters is established. The state of the sand filling layer is divided into dense, incompact, and void. A numerical model is established based on the Guangzhou BI-UT immersed tunnel with incompact and void sand filling layer states to simulate deformation and torsion. The results indicate that the settlement of the tunnel segment is low in the eastern region and high in the western region due to the presence of a less dense sand filling layer, with a maximum differential settlement of 0.04 m. The evaluation method plays a crucial role in guiding the construction of immersed tube tunnels.

ABSTRACT Two different sarbecoviruses, severe acute respiratory syndrome coronavirus (SARS-CoV) and SARS-CoV-2, have caused serious challenges to public health. Certain sarbecoviruses utilize angiotensin-converting enzyme 2 (ACE2) as their cellular receptor, whereas some do not, speculatively due to the two deletions in their receptor-binding domain (RBD). However, it remains unclear whether sarbecoviruses with one deletion in the RBD can still bind to ACE2. Here, we showed that two phylogenetically related sarbecoviruses with one deletion, BtKY72 and BM48-31, displayed a different ACE2-usage range. The cryo-electron microscopy structure of BtKY72 RBD bound to bat ACE2 identified a key residue important for the interaction between RBD and ACE2. In addition, we demonstrated that the mutations involving four types of core residues enabled the sarbecoviruses with deletion(s) to bind to human ACE2 (hACE2) and broadened the ACE2 usage of SARS-CoV-2. Our findings help predict the potential hACE2-binding ability to emerge sarbecoviruses and develop pan-sarbecovirus therapeutic agents.IMPORTANCEMany sarbecoviruses, including severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), possess the ability to bind to receptor angiotensin-converting enzyme 2 (ACE2) through their receptor-binding domain (RBD). However, certain sarbecoviruses with deletion(s) in the RBD lack this capability. In this study, we investigated two closely related short-deletion sarbecoviruses, BtKY72 and BM48-31, and revealed that BtKY72 exhibited a broader ACE2-binding spectrum compared to BM48-31. Structural analysis of the BtKY72 RBD-bat ACE2 complex identifies a critical residue at position 493 contributing to these differences. These findings offer crucial insights for accurately predicting the potential threat of newly emerging sarbecoviruses to human health.

BackgroundThe virulence of Yersinia pestis strains in the Rattus flavipectus plague focus is relatively low. The purpose of this study was to investigate the undetected, sporadic plague cases in plague foci and provide the basis for plague prevention and control.MethodsA 3-year-old plague-confirmed case was investigated in the R. flavipectus plague focus of Yunnan Province in 2020 due to the intensive screening for fever symptoms during the coronavirus disease 2019 (COVID-19) pandemic. Epidemiological investigation, laboratory testing, and clinical treatment were conducted for the case. The expanded survey was carried out around the case within a 7-km radius, including the resident population, domesticated dogs, and rats. PCR and indirect hemagglutination tests were performed on the collected samples.ResultsThe isolation rates of Y. pestis were 100.0% (7 out of 7) in dead rats and 4.00% (3 out of 75) in live rats in the survey area of the foci. A total of 5.00% (6 out of 120) of the domesticated dogs were F1 antibody positive. Nine local people were determined for plague infection recently (0.92%, 9 out of 978). The locations of human cases coincided with the Y. pestis epidemic area among the animals.ConclusionThis study discovered the existence of plague cases that had not been detected by routine surveillance in the R. flavipectus plague focus, and the actual epidemic of human infection may be underestimated.

In this paper, the Zn0.95Cu0.05O/p-Si heterojunction with lateral photovoltaic effect (LPE) is prepared that has the tuning effect and switching behavior of sunlight field. The dependence of the lateral photovoltage (LPV) on the laser position shows high sensitivity (20 mV mm−1) and strict linearity. Interestingly, the LPV eventually decreased to 0 mV as the sunlight intensity increased to 48 mW, under the constant laser intensity and position, indicating the application potential of a photo-switch. This light field effect results from the irradiation of sunlight, which changes the position of quasi-Fermi level, reduces the drive power for carriers by the space charge region in the heterojunction, and increases the concentration of carriers resulting in increased the scattering between carriers to decrease the lifetime of carriers produced by the laser. This work can expand the application of LPE in the field of multifunctional devices such as photo-switches, position sensors and self-powered devices.

This paper analyzes the current situation and existing problems of coal roadway driving technologies and equipment, introduces the new mode, technology, equipment of coal roadway rapid driving with integrated drilling and anchoring and its underground applications. The bolting technologies and equipment with integrated drilling and anchoring are developed which include the integrated drilling and anchoring bolt, pumpable resin, drilling boom and drill rig as well as bolts cartridge with integrated drilling and anchoring, pump unit and intelligence control system. The new technology simplifies the traditional six-step rock bolting process to one continuous step. Rock bolting can be realized automatically by pressing button once and the operation time for installing a single bolt can be reduced from 5 to 6 minutes to 3 minutes. The technologies and equipment of automatic spraying support are developed which include high-strength, rapid setting, and deformable surface supporting spraying materials with a tensile strength exceeding 9 MPa, elongation at break exceeding 100%, and a bonding strength exceeding 3 MPa. The spraying system with 6-degree-of-freedom manipulator is developed which can automatically and rapidly spray during roadway driving. The dynamic monitoring technology and system for roadway surrounding rock deformation are developed during driving which can achieve a surface deformation monitoring of roadway in environments with dust, multiple interference, and low illumination. The monitoring accuracy can meet the requirements of the surrounding rock stability evaluation. A new partitioned parallel collaborative rapid driving mode of excavation, spraying, bolting and transport is proposed and a complete set of roadway rapid driving equipment with integrated drilling and anchoring is developed. The displacement and stability of the surrounding rock during the driving process at the Caojiatan Coal Mine in Shaanxi Province are monitored. In terms of time, more than 80% of the surrounding rock displacement had already occurred in 2 to 3 hours after the excavation of the roadway. In terms of space, more than 90% of the surrounding rock displacement had already occurred at a distance equivalent to twice the width of the roadway from the excavation face. Therefore, the roadway surrounding rock displacement caused by rapid driving can be completed in a short time, requiring timely and rapid support. The nearly 6 000 m roadway driving project has been completed in the 10-metre-extra-high fully mechanized mining face of the Caojiatan Coal Mine, and over 60 000 bolts with integrated drilling and anchoring are installed. The time for installing a single bolt, the number of supporting workers, and the labor intensity of workers have been significantly reduced. The driving speed, efficiency, and automation level have been significantly improved. The displacement of the experimental roadway roof is small. The roadway surrounding rock stability is good as well as the support effect. The coal roadway rapid driving technology and equipment with integrated drilling and anchoring have achieved a success in underground field test. It has the ability to achieve a monthly driving footage of 1500 to 2000 m for ultra-large cross-section coal roadways.

Abstract DNA methylation is a pivotal epigenetic modification that affects gene expression. Tumor immune microenvironment (TIME) comprises diverse immune cells and stromal components, creating a complex landscape that can either promote or inhibit tumor progression. In the TIME, DNA methylation has been shown to play a critical role in influencing immune cell function and tumor immune evasion. DNA methylation regulates immune cell differentiation, immune responses, and TIME composition Targeting DNA methylation in TIME offers various potential avenues for enhancing immune cytotoxicity and reducing immunosuppression. Recent studies have demonstrated that modification of DNA methylation patterns can promote immune cell infiltration and function. However, challenges persist in understanding the precise mechanisms underlying DNA methylation in the TIME, developing selective epigenetic therapies, and effectively integrating these therapies with other antitumor strategies. In conclusion, DNA methylation of both tumor cells and immune cells interacts with the TIME, and thus affects clinical efficacy. The regulation of DNA methylation within the TIME holds significant promise for the advancement of tumor immunotherapy. Addressing these challenges is crucial for harnessing the full potential of epigenetic interventions to enhance antitumor immune responses and improve patient outcomes.

This paper reports a heterojunction laminate consisting of Ni doped BiFeO3 thin film and n-type single crystal silicon semiconductor, in which the repeatable modulation effect on the photoelectric properties were achieved by applying external magnetic fields. More obviously, the response of photocurrent and photovoltage to the magnetic field is perfectly linear. This functional effect of sensing is mainly related to the guidance of the magnetic field to the photogenerated carrier transfer process in the heterojunction. The linear relationships between magnetic field and photocurrent, magnetic field and photovoltage have been proved through the derivation of the formulas. The heterojunction perfectly combines the functions of the sensor and the controller to reflect both the intensity of the magnetic field and the intensity of the light in the environment. This work provides an alternative and facile way to realize the multi-degree of freedom control for electrical signal and practicability of multifunctional devices.

Abstract Researchers have been seeking for the most technically‐economical water electrolysis technology for entering the next‐stage of industrial amplification for large‐scale green hydrogen production. Various membrane‐based electrolyzers have been developed to improve electric‐efficiency, reduce the use of precious metals, enhance stability, and possibly realize direct seawater electrolysis. While electrode engineering is the key to approaching these goals by bridging the gap between catalysts design and electrolyzers development, nevertheless, as an emerging field, has not yet been systematically analyzed. Herein, this review is organized to comprehensively discuss the recent progresses of electrode engineering that have been made toward advanced membrane‐based electrolyzers. For the commercialized or near‐commercialized membrane electrolyzer technologies, the electrode material design principles are interpreted and the interface engineering that have been put forward to improve catalytic sites utilization and reduce precious metal loading is summarized. Given the pressing issues of electrolyzer cost reduction and efficiency improvement, the electrode structure engineering toward applying precious metal free electrocatalysts is highlighted and sufficient accessible sites within the thick catalyst layers with rational electrode architectures and effective ions/mass transport interfaces are enabled. In addition, this review also discusses the innovative ways as proposed to break the barriers of current membrane electrolyzers, including the adjustments of electrode reaction environment, and the feasible cell‐voltage‐breakdown strategies for durable direct seawater electrolysis. Hopefully, this review may provide insightful information of membrane‐based electrode engineering and inspire the future development of advanced membrane electrolyzer technologies for cost‐effective green hydrogen production.

Abstract This paper is aimed at the problems of multiple factors influencing the roof stability near a heading face and unreasonable values of the unsupported distance leading to a slow excavation speed. The behavior of the direct roof subsidence in the unsupported area is analyzed, and numerical simulation and the response surface method are used to study the key factors affecting the roof deformation and their interaction relations. The results indicate that roof strength is the most critical factor affecting deformation and failure, followed by soft rock thickness, unsupported distance, and support strength. The interaction analysis in Zhaozhuang Coal Mine's 33082 lane shows that it is difficult to control the roof subsidence when the unsupported distance is more than 2 m and the soft rock thickness is more than 4 m. After the unsupported distance is set to about 2 m and the support strength is increased to 0.25 MPa, the excavation speed approximately doubles, and the overall roadway is stable and controllable. By implementing field applications, the rationality of the research approach was confirmed.

Solid oxide fuel cells (SOFCs) offer a significant advantage over other fuel cells in terms of flexibility in the choice of fuel. Ammonia stands out as an excellent fuel choice for SOFCs due to its easy transportation and storage, carbon-free nature and mature synthesis technology. For direct-ammonia SOFCs (DA-SOFCs), the development of anode catalysts that have efficient catalytic activity for both NH3 decomposition and H2 oxidation reactions is of great significance. Herein, we develop a Mo-doped La0.6Sr0.4Fe0.8Ni0.2O3−δ (La0.6Sr0.4Fe0.7Ni0.2Mo0.1O3−δ, LSFNM) material, and explore its potential as a symmetrical electrode for DA-SOFCs. After reduction, the main cubic perovskite phase of LSFNM remained unchanged, but some FeNi3 alloy nanoparticles and a small amount of SrLaFeO4 oxide phase were generated. Such reduced LSFNM exhibits excellent catalytic activity for ammonia decomposition due to the presence of FeNi3 alloy nanoparticles, ensuring that it can be used as an anode for DA-SOFCs. In addition, LSFNM shows high oxygen reduction reactivity, indicating that it can also be a cathode for DA-SOFCs. Consequently, a direct-ammonia symmetrical SOFC (DA-SSOFC) with the LSFNM-infiltrated doped ceria (LSFNM-SDCi) electrode delivers a superior peak power density (PPD) of 487 mW cm−2 at 800 °C when NH3 fuel is utilised. More importantly, because Mo doping greatly enhances the reduction stability of the material, the DA-SSOFC with the LSFN-MSDCi electrode exhibits strong operational stability without significant degradation for over 400 h at 700 °C.

Abstract Background PRDM5 is considered a tumor suppressor in several types of solid tumors and is involved in multiple cellular processes. However, target genes regulated by PRDM5 in lung cancer and its potential mechanism are poorly defined. Methods Survival analysis was conducted using Kaplan‐Meier estimates based on the online databases. RNA‐sequencing and bioinformatics analysis were performed to identify the differentially expressed genes in PRDM5‐overexpressed A549 cells. Results We observed deregulated PRDM5 in several lung adenocarcinoma cell lines and its association with a poor prognosis. PRDM5 overexpression inhibited the proliferation of lung adenocarcinoma cells in vitro and suppressed tumor growth in a xenograft model. PRDM5 upregulated the promoter activity of SOCS1, which then inhibited the phosphorylation of JAK2 and STAT3. Conclusions Our study suggests that the low expression of PRDM5 promotes the proliferation of lung adenocarcinoma cells by downregulating SOCS1 and then upregulating the JAK2/STAT3 signaling pathway.

In plants, plasma membrane topologies are predominantly driven by the cell wall. In this study, the authors demonstrate that remorin proteins can take over these functions at specialized, unwalled plasma membranes such as infection droplets associated with symbiotic infection threads.

In order to study the energy distribution of the tandem hybrid tractor and achieve optimal fuel economy under the whole operating condition, an energy distribution strategy based on PMP (Pontryagin minimum principle) is proposed. The performance parameters of the relevant power components are obtained by constructing the test bench of the tandem hybrid tractor. At the same time, the mathematical model of energy distribution is established from the aspects of energy distribution objective function, state variable, and variable constraint. Based on the external characteristic test of the hub motor and the resistance analysis, the transport operation condition of the tractor is selected as the simulation target condition. The ADVISOR2002 and Simulink software are used to jointly simulate the three energy distribution strategies: the thermostat type, the power-following type, and the PMP type. The simulation results show that, compared with the other two, the fuel economy and battery power loss of the PMP-based energy distribution strategy are significantly improved. The fuel consumption per 100 km is decreased by 32.91% and 26.10%, respectively, which verifies the feasibility of the strategy. This study is of great significance for improving the production efficiency and reducing fuel consumption of hybrid tractors.

The reported BiFe _0.9 Co _0.1 O _3 film presents an interesting magnetic field tunning effect on the photoelectric properties and photoluminescence spectra. The change rate of the photocurrent up to 123.6% was achieved when applying a 400 Oe magnetic field to the film, which is attributed to the spin scattering of photoelectrons in the film. The experimental result of the magnetic field tuning photoluminescence spectra indicates that the magnetic field inhibits the radiative transition of the photogenerated carriers in the thin film and therefore reduces the intensity of photoluminescence. This research provides a basis for the research and application of devices in the field of multifunctional photoelectric materials.

Proton‐exchange‐membrane water electrolyzers (PEMWEs) are of particular interest for green hydrogen production, where the oxygen evolution reaction (OER) at the anode largely determines the overall efficiency. Up to now, only ultrafine IrO2 catalyst gives desirable performance, while its scarcity and high cost inhibit the widespread application. RuO2 catalyst is the most promising alternative, while its practical application is greatly hindered by poor durability. Herein, the greatly boosted performance of conventional sub‐micrometer RuO2 by compositing with Ag is reported, and both the morphology of Ag and the compositing way significantly affect the electrolysis performance. The PEMWE fabricated with a two‐layer RuO2/Ag nanowire (NWs) composite anode achieves 1.77 A cm−2 at 2.00 V, due to a prominent 44.6 times increase of the electronic conductivity, which greatly improves the catalyst utilization. In addition, mass transportation at high‐current‐density region is enhanced due to the highly porous feature of Ag NW layer. Long‐term stability under high current density of 1 A cm−2 for 100 h is proved with the composite anode, due to the suppressed degradation of RuO2 by silver compositing. This work may accelerate the widespread commercialization of PEMWEs by providing a new way for developing IrO2‐free anode.

Spin gapless semiconductor (SGS) presents abundant electric and magnetic properties and is highly sensitive to external factors, such as current, electric field, magnetic field, and stress. This paper reports on a PbPd0.9Co0.1O2/PMNPT(001) laminate thin film with the “SGS/ferroelectrics” structure, which exhibits significant current-induced resistance (I-ER) effect. More importantly, the colossal static electric-field-induced resistance (E-ER) effect in such a laminate film was observed for the first time. The introduction of lattice defects (Pb vacancies) induces a local electric field, electron spin ordering state, and ferroelectric polarization field effect to explain the excellent physical properties. The reported laminate thin film composite exhibits promising application potential in spintronic devices, composite sensor units, storage systems, and other low-energy semiconductor electronic device fields. This work proposes an alternative way to investigate the novel properties for spin gapless semiconductors and expand the research fields of multi-field modulation effect in magnetoelectric composites.

Abstract Until now, conventional nanogenerators could only produce electric pulses with relatively low-power densities. Herein, we invent a novel controllable growth technique for two-dimensional (2-D) cuprous oxide (p-Cu2O) single-crystal films, and on this basis, a new concept of 2-D single-crystal film flexoelectric nanogenerators (FENGs) are rationally designed and constructed for the first time, which has the characteristics of long-range order lattice, few grain boundaries and defects. More importantly, the accumulated built-in polarization potential in the bent 2-D p-Cu2O single-crystal film FENGs is in the same orientation as the output electricity, resulting in the first nanogenerator that can output continuous and stable electric signals with high voltage (Voc of 2.8 V), current (Jsc of 11.5 μA·cm−2) and power density (14.4 μW·cm−2), exhibiting great practical application potential for power generation and motion capture. This research breaks new ground and establishes a precedent for high-performance and continuous-output nanogenerators, as well as smart wearable sensors.

Background Following its resurgence in 1982, rodent plague has been linked to a wide range of circulation risks in Yunnan Province. The most serious public health concern associated with effective plague control is determining how various ecological variables influence the differential risk of transmission. Methods We investigated the population dynamics of the hosts and vectors using large-scale epidemiological surveillance data. In a seasonal eco-epidemiological model, we evaluated the impact of ecological conditions on the vectored flea index (VFI) to determine the rate of plague transmission. Results The findings revealed a changing species composition in natural foci over time. Additionally, shifting distributional ranges of species by elevation may be vital in modulating the VFI. The model estimates indicate that the dynamic VFI contributes to spatiotemporal variance in transmission. Conclusions The VFI could be a critical ecological indicator, allowing for real-time tracking and prompt intervention in the circulation of rodent plague. Understanding eco-epidemiological diversity can provide essential insights into effective responses to future plague resurgence. Author summary Plague have resurged among rodents in Yunnan Province since 1982, and intermittently occurs among humans. Yet the local risk of rodent plague dynamics has been highly diverse across ecological contexts. In natural foci, suggestions for this ecological diversity include high species richness and cyclic transmission between hosts and vectors. Here, we developed an eco-epidemiological framework to position the epidemiological dynamics of rodent plague in local ecological contexts. Our results suggest that diverse ecological conditions may have shaped the local risk of plague transmission in Yunnan Province. Typically, incremental of elevation has been associated with the tradeoff between the increased diversity and averted fraction of main host and vector species. More importantly, vectored flea index could be a critical ecological indicator for plague management.

In a sedimentary environment, the conventional one-dimensional (1D) inversion based on the horizontal layered model has difficulty restoring the resistivity distribution of the inclined strata when a coal seam has some dip angle or a small interval between layers. In such cases, the inversion resistivity exhibits horizontal discontinuities, which cannot accurately represent actual geological conditions. Therefore, in view of the good horizontal continuity of the underground electrical structure of sedimentary strata, we propose a high-resolution inversion method based on weighted horizontal and vertical constraints. As a quasi-two-dimensional (2D) inversion, this not only ensures the horizontal continuity of resistivity and recovers the inclined strata, but also improves the vertical resolution. Because the constrained factor has a significant influence on the inversion result, different constrained factors are applied in the horizontal and vertical directions to adjust the constraint strength on the model parameters of each layer and the continuity of the layer interface. In the numerical experiments, we design synthetic models with different tilt angles and layer spacings to test the inversion method and optimize the constrained factors used for coal seam detection. Finally, the transient electromagnetic (TEM) field data processing results in Inner Mongolia show that the resistivity distributions of sedimentary strata can be accurately restored by the new method, and the inversion results are consistent with known geological information.

Organic amine and nanosilica were combined to create a nano-demulsifier, which was employed in the oil–water separation process of a condensate emulsion. The nano-demulsifier has the structure of hyperbranched polymers and the skeleton structure of hyperbranched nanomaterials, and displays the demulsification impact of organic amine polymers as well as the synergistic effect of nanomaterials. This nano-demulsifier has the potential to drastically reduce the quantity of condensate demulsifiers utilized in the gathering station. The dehydration rate of the condensate lotion in the gas gathering station can reach more than 95% only at a concentration of 1.0 wt.%. Its application can significantly increase the separation efficiency of the condensate emulsion as well as the quality of condensate oil. It has a positive impact on cost reduction and efficiency in gas well production. The mechanism of action of the demulsifier was also studied, and the results show that the demulsifier is a phase reverse demulsifier.

Enhanced sulfurization has always been the focus of research on the flotation of copper oxide minerals. In this study, combined ammonium–amine salts were innovatively applied to improve the sulfurization of azurite. Flotation tests were carried out to evaluate the promoting effect of ammonium–amine co-activation on the sulfurization–xanthate flotation of azurite, and the microstructure evolution of sulfurized products was investigated to reveal the mechanism underlying this promoting effect. Compared with single ammonium (amine) salt activation, ammonium–amine co-activation improved the floatability of azurite to a greater extent, i.e., the flotation recovery increased by over 4 percentage points. ToF-SIMS, ICP-OES, FESEM-EDS, AFM, XRD, and UV-vis analyses indicated that ammonium–amine co-activation combined the advantages of inorganic ammonium for buffering pH and organic amine for copper ion complexation, thus promoting the growth of sulfurized crystal products (covellite) and enhancing the adhesion stability of sulfurized products on azurite. Therefore, increasing amounts of copper sulfide components were generated under the ammonium–amine–Na2S system, promoting the adsorption of additional xanthate on azurite. This study provides theoretical support for the application of combined ammonium–amine salts for the sulfurization flotation of copper oxide.

Abstract Subscapular tendon plays an important role in shoulder joint function. With the advance of magnetic resonance imaging technology and the popularization of arthroscopic shoulder surgery, subscapularis tears have been increasingly detected. However, reduction and fixation of subscapular tendon tears appears to be technically challenging. This study aims to describe an arthroscopic intra‐articular X‐shaped fixation technique: a procedure of subscapularis tendon repair performed with the aid of a suture passer using only a single anterior portal and a single suture anchor. By incorporating the advantages of a single anterior working portal for anchor placement and tear repair, this technique provides an easier way to use suture lasso and make knots in a limited working space, and the whole procedure is minimally invasive with a short learning curve. This technique has been applied in patients with subscapularis tears involving no intraoperative or postoperative complications. Our technology offers a valuable new treatment option for subscapularis tears.

Host-controlled intracellular accommodation of nitrogen-fixing bacteria is essential for the establishment of a functional Root Nodule Symbiosis (RNS). In many host plants, this occurs via transcellular tubular structures (infection threads - ITs) that extend across cell layers via polar tip-growth. Comparative phylogenomic studies have identified RPG (RHIZOBIUM-DIRECTED POLAR GROWTH) among the critical genetic determinants for bacterial infection. In Medicago truncatula, RPG is required for effective IT progression within root hairs but the cellular and molecular function of the encoded protein remains elusive. Here, we show that RPG resides in the protein complex formed by the core endosymbiotic components VAPYRIN (VPY) and LUMPY INFECTION (LIN) required for IT polar growth, co-localizes with both VPY and LIN in IT tip- and perinuclear-associated puncta of M. truncatula root hairs undergoing infection and is necessary for VPY recruitment into these structures. Fluorescence Lifetime Imaging Microscopy (FLIM) of phosphoinositide species during bacterial infection revealed that functional RPG is required to sustain strong membrane polarization at the advancing tip of the IT. In addition, loss of RPG functionality alters the cytoskeleton-mediated connectivity between the IT tip and the nucleus and affects the polar secretion of the cell wall modifying enzyme NODULE PECTATE LYASE (NPL). Our results integrate RPG into a core host machinery required to support symbiont accommodation, suggesting that its occurrence in plant host genomes is essential to co-opt a multimeric protein module committed to endosymbiosis to sustain IT-mediated bacterial infection.

IntroductionCurrently, the anti-oxidation of active ingredients in mulberry leaves (MLs) and their forage utilization is receiving increasing attention. Here, we propose that MLs supplementation improves oxidative resistance and immunity.MethodsWe conducted a trial including three groups of growing mutton sheep, each receiving fermented mulberry leaves (FMLs) feeding, dried mulberry leaves (DMLs) feeding or normal control feeding without MLs.ResultsTranscriptomic and metabolomic analyses revealed that promoting anti-oxidation and enhancing disease resistance of MLs is attributed to improved tryptophan metabolic pathways and reduced peroxidation of polyunsaturated fatty acids (PUFAs). Furthermore, immunity was markedly increased after FMLs treatment by regulating glycolysis and mannose-6-phosphate pathways. Additionally, there was better average daily gain in the MLs treatment groups.ConclusionThese findings provide new insights for understanding the beneficial effects of MLs in animal husbandry and provide a theoretical support for extensive application of MLs in improving nutrition and health care values.

This paper presents a PSBFEM approach that integrates the quadtree mesh generation technique based on digital images for solving seepage problems. The quantitative representation of the distribution of geometrical information and material parameters is achieved by utilizing the color intensity of each pixel, which can then be applied to seepage analysis. The presented method addresses the issue of hanging nodes by treating them as nodes of a polygonal element. We validate the proposed technique by solving three benchmark seepage problems. Results show that the image-based domain can be automatically discretized using a quadtree decomposition of the images. Furthermore, the computational efficiency and precision of the PSBFEM surpass that of the standard FEM. Therefore, the proposed technique allows for the convenient automatic discretization of the domain using pixel meshes to solve seepage problems in engineering applications.

Roadways with retained bottom coal are common in thick coal seam mining, and floor heaving is a prominent problem. In this study, based on the interaction between the floor and two sides of the roadway-surrounding rock, a Winkler elastic foundation beam model is established to analyze the floor heave problem. A 3DEC model was used to analyze the failure range, failure mode, and migration law of the floor-surrounding rock with different bottom coal thicknesses and coal body strengths. The results show that (1) an increase in the thickness of the bottom coal results in a decrease in the stiffness of the roadway side coal body (the foundation of the supporting rock layer) and an increase in the bending deformation range, the amount of floor rock beam deformation, and the extrusion force. This leads to an expansion in the range of the sides of the coal body that are squeezed by the floor rock layer, resulting in additional failure and deformation of the coal body sides. Therefore, the damage to the floor rock layer is extended and increased. (2) The expansion of the floor pressure-bearing arch and surrounding rock in the arch are the causes of floor heave in the deep coal roadway with retained bottom coal. (3) Because of an increase in the thickness of the bottom coal and a decrease in the coal body strength, the floor pressure-bearing arch expands to the deeper part; thus, the range of surrounding rock in the arch with deformation and failure increases, resulting in an increase in floor heave. The field practice indicates that the support strategy of the “high prestressed strong rock bolt (cable) supporting two sides and bottom corners in time” can effectively control the floor heave of a roadway with retained bottom coal.

Considering the Ligusticum wallichii-soil mixture is easy to block and the broken soil is difficult to separate when the Ligusticum wallichii is excavated and harvested，a vibration mechanism of double rocker threw broken soil is designed，and the kinematic characteristics of the mechanism are studied. The kinematic analysis and modeling of the vibrating earth breaking mechanism are carried out by analytical methods，the kinematics of the earth breaking vibration mechanism is obtained，the kinematic simulation of the earth breaking mechanism is carried out using Adams，the centroid displacement，velocity and acceleration of the mechanism are obtained，and the motion law and soil crushed ability of the vibrated soil crushed mechanism are verified. The results show that when the crank of the double rocker threw and crushed mechanism are at a turned angle of 22.4°，the mixture would be thrown away，and the horizontal backward movement distance is 67.1 mm，which is greater than the forward distance of the harvester 39.7 mm，realizes the backward conveyed of the mixture，the separation of the broken soil and avoids the occurrence of soil clogging. This research provides theoretical support for the design of the vibration crushing mechanism of the Ligusticum wallichii harvester.

A new variant of concern for SARS-CoV-2, Omicron (B.1.1.529), was designated by the World Health Organization on November 26, 2021. This study analyzed the viral genome sequencing data of 108 samples collected from patients infected with Omicron. First, we found that the enrichment efficiency of viral nucleic acids was reduced due to mutations in the region where the primers anneal to. Second, the Omicron variant possesses an excessive number of mutations compared to other variants circulating at the same time (median: 62 vs. 45), especially in the Spike gene. Mutations in the Spike gene confer alterations in 32 amino acid residues, more than those observed in other SARS-CoV-2 variants. Moreover, a large number of nonsynonymous mutations occur in the codons for the amino acid residues located on the surface of the Spike protein, which could potentially affect the replication, infectivity, and antigenicity of SARS-CoV-2. Third, there are 53 mutations between the Omicron variant and its closest sequences available in public databases. Many of these mutations were rarely observed in public databases and had a low mutation rate. In addition, the linkage disequilibrium between these mutations was low, with a limited number of mutations concurrently observed in the same genome, suggesting that the Omicron variant would be in a different evolutionary branch from the currently prevalent variants. To improve our ability to detect and track the source of new variants rapidly, it is imperative to further strengthen genomic surveillance and data sharing globally in a timely manner.

Transition metal oxide materials are promising oxygen catalysts that are alternatives to expensive and precious metal-containing catalysts. Integration of transition metal oxides with high activity for oxygen reduction reaction (ORR) and oxygen evolution reaction (OER) is an important pathway for good bifunctionality. In contrast to the conventional physical mixing and hybridization strategies, perovskite-type oxide provides an ideal structure for the integration of the transition metal element atoms on an atomic scale. Herein, B-site ordered double-perovskite-type La1.6Sr0.4MnCoO6 nanocrystallites with ultra-small cubic (20–50 nm) morphology and high specific surface areas (25 m2 g−1) were proposed. Rational designs were integrated to promote the ORR-OER catalysis, e.g., introducing oxygen vacancies via A-site cation substitution, further increasing surface oxygen vacancies via integration of a small amount of Pt/C and nanosizing of the material via a facile molten-salt method. The batteries with the La1.6Sr0.4MnCoO6 nanocrystallites and an aqueous alkaline electrolyte demonstrate decent discharge−charge voltage gaps of 0.75 and 1.10 V at 1 and 30 mA cm−2, respectively, and good cycling stability of 250 h (1500 cycles). A coin-type battery with a gel−polymer electrolyte also presents a good performance.

In this work, the growth of copper sulfide crystal onto azurite surfaces during sulfurization and its response to flotation are investigated. Filed emission scanning electron microscopy-energy dispersive X-ray spectroscopy (FESEM) and X-ray diffraction (XRD) studies confirmed that the sulfurization of azurite is not limited to the mineral surface, but rather penetrates into the bulk to form covellite crystal (syn-CuS), creating favorable conditions for the stable adsorption of xanthate and greatly promoting the azurite flotation. Additionally, as demonstrated by X-ray photoelectron spectroscopy (XPS) and time of flight secondary ion mass spectrometry (TOF-SIMS) analyses, a redox reaction occurred during this process, and Cu(Ⅱ) onto the mineral surface was reduced to Cu(I). Correspondingly, reduced S2- was oxidized to (S2)2-, (Sn)2-, and even to deeper oxidation state S0, (SxOy)n- and SO42-. Excess sodium sulfide strengthens copper sulfide to form onto the azurite surface, and provides enough raw material for crystal copper sulfide to grow, resulting in the formation of “flake-like” covellite with a better crystallinity. However, the floatability of azurite decreased dramatically under this condition, because the generated massive colloidal copper sulfide in flotation pulp deteriorates the flotation environment, resulting in a decreased effective adsorption of collector onto azurite surfaces.

The SARS-CoV-2 spike (S) protein mediates viral entry by binding of its receptor-binding domain (RBD) to the human angiotensin-converting enzyme 2 (ACE2) receptor and mutations of the S protein may have a great impact on virus transmissibility. Here, the authors characterize the interactions of six different SARS-CoV-2 RBD variants among them Alpha, Beta and Gamma and present crystal structures of these ACE2-RBD complexes.

Abstract Background The clinical pathologic stages (stage I, II, III-IV) of hepatocellular carcinoma (HCC) are closely linked to the clinical prognosis of patients. This study aims at investigating the gene expression and mutational profile in different clinical pathologic stages of HCC. Methods Based on the TCGA-LIHC cohort, we utilized a series of analytical approaches, such as statistical analysis, random forest, decision tree, principal component analysis (PCA), to identify the differential gene expression and mutational profiles. The expression patterns of several targeting genes were also verified by analyzing the Chinese HLivH060PG02 HCC cohort, several GEO datasets, HPA database, and diethylnitrosamine-induced HCC mouse model. Results We identified a series of targeting genes with copy number variation, which is statistically associated with gene expression. Non-synonymous mutations mainly existed in some genes (e.g.,TTN, TP53, CTNNB1). Nevertheless, no association between gene mutation frequency and pathologic stage distribution was detected. The random forest and decision tree modeling analysis data showed a group of genes related to different HCC pathologic stages, including GAS2L3 and SEMA3F. Additionally, our PCA data indicated several genes associated with different pathologic stages, including SNRPA and SNRPD2. Compared with adjacent normal tissues, we observed a highly expressed level of GAS2L3, SNRPA, and SNRPD2 (P = 0.002) genes in HCC tissues of our HLivH060PG02 cohort. We also detected the high expression pattern of GAS2L3, SEMA3F, SNRPA, and SNRPD2 in the datasets of GSE102079, GSE76427, GSE64041, GSE121248, GSE84005, and the qPCR assay using diethylnitrosamine-induced HCC mouse model. Moreover, SEMA3F and SNRPD2 protein were highly stained in the HCC tissues of the HPA database. The high expression level of these four genes was associated with the poor survival prognosis of HCC cases. Conclusions Our study provides evidence regarding the gene expression and mutational profile in different clinical pathologic stages of TCGA HCC cases. Identifying four targeting genes, including GAS2L3, SNRPA, SNRPD2, and SEMA3F, offers insight into the molecular mechanisms associated with different prognoses of HCC.

Soil microorganisms play vital roles in ecosystem functions, and soil microbial communities might be affected by heavy metal contamination caused by the anthropogenic activities associated with the coal-based industry. This study explored the effects of heavy metal contamination on soil bacterial and fungal communities surrounding different coal-based industrial fields (the coal mining industry, coal preparation industry, coal-based chemical industry, and coal-fired power industry) in Shanxi province, North China. Moreover, soil samples from farmland and parks away from all the industrial plants were collected as references. The results showed that the concentrations of most heavy metals were greater than the local background values, particularly for arsenic (As), lead (Pb), cadmium (Cd), and mercury (Hg). There were significant differences in soil cellulase and alkaline phosphatase activities among sampling fields. The composition, diversity, and abundance of soil microbial communities among all sampling fields were significantly different, particularly for the fungal community. Actinobacteria, Proteobacteria, Chloroflexi, and Acidobacteria were the predominant bacterial phyla, while Ascomycota, Mortierellomycota, and Basidiomycota dominated the studied fungal community in this coal-based industrially intensive region. A redundancy analysis, variance partitioning analysis, and Spearman correlation analysis revealed that the soil microbial community structure was significantly affected by Cd, total carbon, total nitrogen, and alkaline phosphatase activity. This study profiles the basic features of the soil physicochemical properties, the multiple heavy metal concentrations, and the microbial communities in a coal-based industrial region in North China.

The widespread application of protonic ceramic fuel cells is limited by the lack of oxygen electrodes with excellent activity and stability. Herein, the strategy of halogen doping in a Ba0.6Sr0.4Co0.7Fe0.2Nb0.1O3-δ (BSCFN) cathode is discussed in detail for improving cathode activity. Ba0.6Sr0.4Co0.7Fe0.2Nb0.1O3-x-δFx (x = 0, 0.05, 0.1) cathode materials are synthesised by a solid-phase method. The XRD results show that fluorine anion-doped BSCFN forms a single-phase perovskite structure. XPS and titration results reveal that fluorine ion doping increases active oxygen and surface adsorbed oxygen. It also confines chemical bonds between cations and anions, which enhances the cathode’s catalytic performance. Therefore, an anode-supported single cell with the configuration of Ni-BaZr0.1Ce0.7Y0.1Yb0.1O3-δ (BZCYYb)|BZCYYb|Ba0.6Sr0.4Co0.7Fe0.2Nb0.1O3-0.1-δF0.1 (BSCFN-F0.1) achieved a high peak power density of 630 mW cm−2 at 600 °C. Moreover, according to the symmetrical cell test, the BSCFN-F0.1 electrode demonstrated a superb stability for nearly 400 h at 600 °C. This work focuses on the influence of fluorine anion incorporation upon the performance of cathode materials. It also analyses and discusses the effects of different fluorine ion incorporation amounts to occupy different oxygen positions.

Abstract Background Emerging oncogenes were reportedly linked to the complicated subtypes and pathogenesis of clinical gliomas. Herein, we first comprehensively explored the potential correlation between growth‐arrest‐specific two family genes (GAS2, GAS2L1, GAS2L2, GAS2L3) and gliomas by bioinformatics analysis and cellular experiments. Methods Based on the available datasets of TCGA (The Cancer Genome Atlas), CGGA (Chinese Glioma Genome Atlas), and Oncomine databases, we performed a series of analyses, such as gene expression, survival prognosis, DNA methylation, immune infiltration, and partner enrichment. Mulberry leaves (ML) are a promising alternative fodder source due to their high protein content and the abundance of active components. A test of three inoculants in various combinations revealed that high-quality ML silage was produced at an inoculum ratio of 1:1:0 (50% Saccharomyces cerevisiae, 50% Lactobacillus plantarum, and 0% Bacillus subtilis). Using dry matter (DM) loss, pH, ammonia-N and amino acid contents, total antioxidant activity, and total flavonoids content to evaluate silage quality, this inoculant mixture was shown to produce high-quality silage within a range of inoculum size (5–15%), moisture contents (50–67%), ensiling temperatures (27–30°C), and ensiling duration (14–30 days). A third trial comparing silages produced after 30 days at 28°C and 50% moisture content revealed that silage E, prepared using an L. plantarum inoculant alone, displayed the lowest DM loss and pH, and low bacterial diversity, and it was dominated by Lactobacillus (88.6%), with low abundance of Enterobacter (6.17%). In contrast, silage B5, prepared with equal ratios of L. plantarum and S. cerevisiae, was dominated by Enterococcus (67.16%) and Lactobacillus (26.94%), with less marked yeast persistence, and reducing the DM content from 50 to 40% altered these relative abundances to 5.47 and 60.61, respectively. Control silages produced without an inoculant had the highest pH and ammonia-N content (indicative of poor quality), had the lowest antioxidant activity, had higher bacterial diversity, and were dominated by Carnobacterium (74.28%) and Enterococcus (17.3%). In summary, ensiling of ML conditions with proper inoculants yielded high-quality silage with a favorable microbial community composition.

Introduction: RET is well known as an important driver gene in NSCLC. Moreover, RET is a rare acquired resistance mechanism to EGFR-mutant NSCLC. Only 36 NSCLC cases of coexistence of EGFR and RET were reported previously worldwide. So far, there have been no reports on the following: (1) whether combination of EGFR tyrosine kinase inhibitor (TKI) and RET TKI works for meningeal metastasis; (2) the concentrations of EGFR TKI and RET TKI in the cerebrospinal fluid (CSF) and plasma; and (3) whether RET fusions and EGFR mutation happened in the same clone or not. Methods: We reported a patient with an EGFR-mutant NSCLC with acquired RET fusions and meningeal metastasis treated with pralsetinib and osimertinib; the specimen was analyzed by next-generation sequencing (Illumina NovaSeq 6000 platform). Symptom improvement and magnetic resonance imaging scan were used for effect evaluation. Furthermore, we determined the concentrations of pralsetinib and osimertinib in CSF and plasma by means of liquid chromatography tandem mass spectrometry. We also detected RET fusion and EGFR L858R mutation by methods of fluorescence in situ hybridization and immunohistochemistry with continuous sections to analyze whether RET fusions coexist with EGFR mutation in the same clone or not. Results: The allele frequency of the RET fusion was detected to be 12.88%. This patient achieved a partial response, indicating pralsetinib combined with osimertinib may be clinically beneficial for meningeal metastasis in patients harboring acquired coexistent RET fusions. The concentrations of pralsetinib in the CSF and plasma were 704.76 nM and 91.31 μM, whereas those of osimertinib in the CSF and plasma were 23.70 nM and 2148.94 nM, respectively. RET fusion was found in the same clone of EGFR L858R mutation. Conclusions: Our finding of this case indicated that RET fusion and EGFR mutation occur in the same population of cell clones, rather than in different cell clones. Combined pralsetinib may be an effective way to overcome the resistance, even for meningeal metastasis, owing to high CSF distribution of pralsetinib.

Abstract Background To evaluate the clinical outcomes of arthroscopic tight fibrous band release in the treatment of adult moderate-to-severe gluteal fibrosis using anterior and posterior portals during mid-term follow-up. Methods The data of 138 patients (58 males, 80 females) aged between 18 and 42 years (mean, 28.6 years), presenting with bilateral moderate-to-severe gluteal fibrosis (GF) from October 2013 to August 2019, was retrospectively analyzed. All patients underwent arthroscopic tight fibrous band release using anterior and posterior portals with radiofrequency energy. Under arthroscopic guidance through the posterior portal, we debrided the fatty tissue overlying the contracted band of the gluteal muscle and excised the contracted bands using a radiofrequency device introduced through the anterior portal. The pre- and post-operative gluteal muscle contracture disability (GD) scale and the patient satisfaction rate were compared to evaluate the curative effect of the operation. Results The average operation time was 18 min (range, 10–30 min) and the average blood loss was 4 ml (range, 2–10 ml) for unilateral arthroscopic release. Two cases of post-operative minimal hematomas, 2 cases of bruising and 2 cases of local subcutaneous edema were observed as early complications and were cured by conservative treatment. After surgery, all incisions healed in stage I, and no other complications such as wound infection, nerve and blood vessel injury were detected. One hundred eighteen patients were followed up for 6 to 72 months (mean, 36 months). No lateral instability of the hip was observed and all patients returned to normal gait. The degree of adduction of the hip joint in all these 118 patients was significantly improved relative to their pre-operative conditions. One hundred fifteen patients (97.5%) were able to crouch with knees close to each other after surgery. One hundred fourteen patients (96.6%) were able to cross the affected leg completely without any support. Most traditional artificial intelligence-based fault location methods are very dependent on fault signal selection and feature extraction, which is often based on prior knowledge. Further, these methods are usually very sensitive to line parameters and selected fault characteristics, so the generalization performance is poor and cannot be applied to different lines. In order to solve the above problems, this paper proposes a two-terminal fault location fusion model, which combines a convolutional neural network (CNN), an attention module (AM), and multi-head long short-term memory (multi-head-LSTM). First, the CNN is used to accomplish the self-extraction of fault data features. Second, the CBAM (convolutional block attention module) model is embedded into the convolutional neural network to selectively learn fault features autonomously. Furthermore, the LSTM is combined to learn the deep timing characteristics. Finally, a MLP output layer is used to determine the optimal weights to construct a fusion model based on the results of the two-terminal relative fault location model and then output the final location result. Simulation studies show that this method has a high location accuracy, does not require the design of complex feature extraction algorithms, and exhibits good generalization performance for lines with different parameters, which is of great importance for the development of AI-based methods of fault location.

In the preparation of a superamphiphobic surface, the most basic method is to reduce the surface free energy of the interface. The C—F bond has a very low surface free energy, which can significantly change the wettability of the solid–liquid interface and make it a hydrophobic or oleophobic, or even superamphiphobic surface. Based on the analysis of a large number of research articles, the preparation and application progress in fluoropolymer emulsion were summarized. After that, some corresponding thoughts were put forward combined with our professional characteristics. According to recent research, the status of the fluoropolymer emulsion preparation system was analyzed. In addition, all related aspects of fluoropolymer emulsion were systematically classified in varying degrees. Furthermore, the interaction between fluoropolymer structure and properties, especially the interaction with nanomaterials, was also explored. The aim of this review is to try to attract more scholars’ attention to fluorocarbon interfacial materials. It is expected that it will make a certain theoretical and practical significance in the preparation and application of fluoropolymer.

To ensure the high purity and biological activity of the adenovirus vector to be used for clinical applications, a stable and linearly scalable preparation method is highly imperative. During the adenovirus-harvesting process, the Triton X-100-based lysis method possesses the advantages of higher efficiency as well as easier linearization and amplification. Most Triton X-100 can be removed from the adenovirus sample by chromatographic purification. However, there is no report that a small amount of residual Triton X-100, present in adenovirus sample, can affect the particle integrity, infectivity, and structure of adenoviruses. Here, we found that although residual Triton X-100 affected the short-term stability, purity, infectivity, and structure of adenoviruses at 37°C, it did not hamper these properties of adenoviruses at 4°C. This study suggests that although the Triton X-100-based lysis method is a simple, efficient, and easy-to-scale process for lysing host cells to release the adenovirus, the storage conditions of adenovirus products must be taken into consideration.

Abstract Background In terms of biological behavior, gene regulation, or signaling pathways, there is a certain similarity between tumorigenesis and embryonic development of humans. Three germ layer structure exhibits the distinct ability to form specific tissues and organs. Methods The present study set out to investigate the genetic mutation characteristics of germ layer differentiation-related genes using the tumor cases of the cancer genome atlas (TCGA) database. Results These tumor samples were divided into three groups, including the ectoderm, mesoderm, and endoderm. Children cases less than 9 years old accounted for a larger proportion for the cases in the ectoderm and mesoderm groups; whereas the middle-aged and elderly individuals (from 50 to 89 years old) were more susceptible to tumors of endoderm. There was a better prognosis for the cases of mesoderm, especially the male with the race of White, compared with the other groups. A missense mutation was frequently detected for the cases of ectoderm and endoderm, while deletion mutation was common for that of mesoderm. We could not identify the ectoderm, mesoderm, or endoderm-specific mutated genes or variants with high mutation frequency. However, there was a relatively higher mutation incidence of endoderm markers (GATA6, FOXA2, GATA4, AFP) in the endoderm group, compared with the groups of ectoderm and mesoderm. Additionally, four members (SMO, GLI1, GLI2, GLI3) within the Hedgehog signaling pathway genes showed a relatively higher mutation rate in the endoderm group than the other two groups. Conclusions TCGA tumors of ectoderm, mesoderm, and endoderm groups exhibit the distinct subject distribution, survival status, and genomic alteration characteristics. The synergistic mutation effect of specific genes closely related to embryonic development may contribute to the tumorigenesis of tissues or organs derived from the specific germ layers. This study provides a novel reference for exploring the functional connection between embryogenesis and tumorigenesis.

EphA2 is the specific entry receptor for both human γ-herpesviruses Kaposi sarcoma associated herpesvirus (KSHV) and Epstein-Barr virus (EBV). Here, the authors present the crystal structures of the EphA2 ligand binding domain (LBD) bound to the viral glycoprotein gHgL from EBV and KSHV and further analyse EphA2 gHgL interactions with mutagenesis experiments in cell-based fusion assays and suggest that other animal γ-herpesviruses could also use EphA2 as an entry receptor.

Objective: Esophageal squamous cell carcinoma (ESCC) presents high morbidity and mortality. It was demonstrated that blood-derived vesicles can facilitate ESCC development and transmit regulating signals. However, the molecular mechanism of vesicle miRNA secreted by tumor cells affecting ESCC progression has not been explored.Methods: The mRNA-related signaling pathways and differentially expressed genes were screened out in TCGA dataset. The levels of miRNA-105-5p and SPARCL1 were determined by qRT-PCR. Protein level determination was processed using Western blot. The interaction between the two genes was verified with the dual-luciferase method. A transmission electron microscope was utilized to further identify extracellular vesicles (EVs), and co-culture assay was performed to validate the intake of EVs. In vitro experiments were conducted to evaluate cell function changes in ESCC. A mice tumor formation experiment was carried out to observe tumor growth in vivo.Results: MiRNA-105-5p expression was increased in ESCC, while SPARCL1 was less expressed. MiRNA-105-5p facilitated cell behaviors in ESCC through targeting SPARCL1 and regulating the focal adhesion kinase (FAK)/Akt signaling pathway. Blood-derived external vesicles containing miRNA-105-5p and EVs could be internalized by ESCC cells. Then, miRNA-105-5p could be transferred to ESCC cells to foster tumorigenesis as well as cell behaviors.Conclusion: EV-carried miRNA-105-5p entered ESCC cells and promoted tumor-relevant functions by mediating SPARCL1 and the FAK/Akt signaling pathway, which indicated that the treatment of ESCC via serum EVs might be a novel therapy and that miRNA-105-5p can be a molecular target for ESCC therapy.
