Abstract Background Fixation instability serves as an initial trigger for femoral head necrosis in patients with femoral neck fractures undergoing internal fixation. The configuration of screw trajectories is critical in determining the stability of the fixation. Our previously published study indicates that abduction of screw trajectories may enhance postoperative angular stability. However, the effectiveness of a wider screw distribution on the sagittal plane in optimizing fixation stability and reducing the risk of femoral head necrosis remains uncertain. Methods This retrospective study reviewed imaging data from patients with femoral neck fractures who were treated with inverse triangle cannulated screw fixation. Postoperative lateral radiographs were used to evaluate the screw configuration strategy. The distance between the two cranial screws and the femoral neck width at its narrowest point were measured. The ratio of these two measurements was calculated to represent the screw spread grade. Demographic and imaging parameters were compared between patients who developed femoral head necrosis and those who did not. Regression analysis was conducted to identify potential risk factors associated with femoral head necrosis. Additionally, numerical models were employed to simulate the biomechanical impact of variations in screw spread grade. Results The clinical analysis revealed that a lower screw distance-to-femoral width ratio was associated with a higher incidence of femoral head necrosis. This ratio was also identified as an independent risk factor for femoral head necrosis. Biomechanical simulations corroborated these findings, demonstrating that models with a lower screw-to-femoral width ratio exhibited reduced fixation stability and increased stress distribution. Conclusions Wider distribution of screws along the sagittal plane may significantly mitigate the risk of femoral head necrosis by enhancing fixation stability. The optimization of screw trajectories, particularly through broader screw spacing facilitated by a navigation system, emerges as a promising strategy for improving the local biomechanical environment and reducing the likelihood of femoral head necrosis.

Abstract Background Uncertainty in speech perception and emotional disturbances are intertwined with psychiatric symptoms. How prosody embedded in target speech affects speech-in-noise recognition (SR) and is related to psychiatric symptoms in patients with schizophrenia (SCHs) remains unclear. This study aimed to examine the neural substrates of prosodic SR deficits and their associations with psychiatric symptom dimensions in patients with schizophrenia. Methods Fifty-four SCHs and 59 healthy control participants (HCs) completed the SR task (participants were required to identify the contents of the target pseudo-sentences expressed in neutral, happy, sad, angry, fearful, and disgusted prosody by actors), positive and negative syndrome scale (PANSS) assessment, and magnetic resonance imaging scanning. We examined the deficits of the six prosodic SRs in schizophrenia patients and their associations with brain gray matter volume (GMV) and psychiatric symptoms. Results Negative emotional (sad, angry, fearful, and disgusted) prosodies of the target sentences worsened SR across groups. Both participant groups had equal SR between the neutral and happy prosody conditions. Compared to the anger and disgusted conditions, SCHs and HCs had better SR under sad and fearful conditions. Better prosodic SR was associated with shorter duration and lower local shimmer of target sentences. A partial least squares (PLS) component of GMV (covering 47 brain regions with group differences) was associated with six prosodic SRs. The happy SR was associated with the PANSS total, negative, and general scores, adjusting for covariates. Conclusions Negative emotional prosodies of the target sentences dampened the recognition of the target sentences. The prosodic SR abnormalities in SCHs were associated with not only brain GMV reductions in the regions involved in the processing of sensorimotor, speech, and emotion but also with negative and general psychiatric symptoms. These findings suggest the possibility of improving negative symptoms by improving a happy SR in schizophrenia patients based on neuroplasticity.

Abstract Background This study aims to conduct a finite element analysis (FEA) to assess the bio-mechanical properties of C2 sagittal-parallel pedicle screw (PPS) in fixation for atlantoaxial instability, thereby providing a theoretical foundation for its clinical application. Methods A total of 5 intact C1-2 finite element models were established. Based on this, instability models were developed and two different fixation methods were applied for each model: C1 lateral mass screw (LMS) combined with C2 sagittal-parallel pedicle screw (C1LMS + C2PPS), and C1 lateral mass screw combined with C2 traditional pedicle screw (C1LMS + C2PS). Under a physiological load of 40 N, a pure moment of 1.5 Nm was used to simulate movements of the cervical spine in flexion, extension, lateral bending, and axial rotation. The von Mises stress of implants and the segment range of motion (ROM) were analyzed and compared statistically. Results The intact model was validated and showed good consistency with other studies in terms of range of motion (ROM). In flexion and extension, the C1LMS + C2PPS resulted in lower segment ROM (12.4% and 6.3% decrease) and stress concentration (15.9% decrease in flexion) compared to C1LMS + C2PS. However, in lateral bending and axial rotation, the C1LMS + C2PPS exhibited higher segment ROM (42.9% and 5.9% increase) and stress concentration (8.7% and 21.4% increase) compared to C1LMS + C2PS. Conclusion Both methods were safe and stable for the fixation of atlantoaxial instability. Compared to C1LMS + C2PS, the use of C1LMS + C2PPS may provide better stability and a lower risk of implant failure in flexion and extension. Clinically, it is feasible to utilize the C2 sagittal-parallel pedicle screw in fixing atlantoaxial instability.

Abstract Background This study aimed to identify the risk factors of acute ischemic stroke (AIS) occurring during hospitalization in patients following off-pump coronary artery bypass grafting (OPCABG) and utilize Bayesian network (BN) methods to establish predictive models for this disease. Methods Data were collected from the electronic health records of adult patients who underwent OPCABG at Beijing Anzhen Hospital from January 2018 to December 2022. Patients were allocated to the training and test sets in an 8:2 ratio according to the principle of randomness. Subsequently, a BN model was established using the training dataset and validated against the testing dataset. The BN model was developed using a tabu search algorithm. Finally, receiver operating characteristic (ROC) and calibration curves were plotted to assess the extent of disparity in predictive performance between the BN and logistic models. Results A total of 10,184 patients (mean (SD) age, 62.45 (8.7) years; 2524 (24.7%) females) were enrolled, including 151 (1.5%) with AIS and 10,033 (98.5%) without AIS. Female sex, history of ischemic stroke, severe carotid artery stenosis, high glycated albumin (GA) levels, high D-dimer levels, high erythrocyte distribution width (RDW), and high blood urea nitrogen (BUN) levels were strongly associated with AIS. Type 2 diabetes mellitus (T2DM) was indirectly linked to AIS through GA and BUN. The BN models exhibited superior performance to logistic regression in both the training and testing sets, achieving accuracies of 72.64% and 71.48%, area under the curve (AUC) of 0.899 (95% confidence interval (CI), 0.876–0.921) and 0.852 (95% CI, 0.769–0.935), sensitivities of 91.87% and 89.29%, and specificities of 72.35% and 71.24% (using the optimal cut-off), respectively. Conclusion Female gender, IS history, carotid stenosis (> 70%), RDW-CV, GA, D-dimer, BUN, and T2DM are potential predictors of IS in our Chinese cohort. The BN model demonstrated greater efficiency than the logistic regression model. Hence, employing BN models could be conducive to the early diagnosis and prevention of AIS after OPCABG.

Abstract Background The risk of liver fibrosis in HBeAg-positive chronic hepatitis B (CHB) patients with indeterminate phase is not well characterized. We aimed to compare the presence of liver fibrosis in HBeAg-positive CHB patients between indeterminate phase and immune-tolerant phase. Methods This multi-center, retrospective cohort study included 719 treatment-naïve HBeAg-positive CHB patients with normal alanine aminotransferase (ALT). Patients with HBV DNA > 106 IU/mL were categorized into immune-tolerant phase, whereas those with HBV DNA ≤ 106 IU/mL were classified into indeterminate phase. Significant liver fibrosis and cirrhosis were determined by APRI, FIB-4, transient elastography, or liver biopsy. Results The median age of patients was 33.0 years and 59.8% of patients were male. 81.5% and 18.5% of patients were in the immune-tolerant phase and indeterminate phase, respectively. Abstract To investigate physiological function of α-synuclein is important for understanding its pathophysiological mechanism in synucleinopathies including Parkinson’s disease. Employing knockout mice, we found that Snac/α-synuclein deletion induced aberrant projection of olfactory sensory neurons and hyposmia. We identified 9 axon guidance associated differentially expressed proteins using iTRAQ based Liquid Chromatograph Mass Spectrometer. NCK2 is most significantly down-regulated protein among them. We further found that either α-synuclein deletion or NCK2 deficiency induced Eph A4 inactivation. Re-expressing Snac/α-synuclein in its knockout neurons reversed the down-regulation of NCK2, as well as the inactivation of EphA4. Overexpression of Snac/α-synuclein in α-synuclein deleted mice reversed the down-regulation of NCK2 and pEphA4, and improved the olfactory impairment of mice. Correlation analysis showed that there is a significant correlation between the protein level of α-synuclein, NCK2, and pEphA4, respectively. Nonetheless, immunoprecipitation analysis showed that NCK2 was associated with both EphA4 and Rho A, suggesting that NCK2 as a scaffolding protein to modulate Eph A4/Rho A pathway. Moreover, Rho A activity was significantly lower in α-synuclein deficient mice. Thus, α-synuclein regulates olfactory neurons projection through NCK2 dependent EphA4/Rho A pathway. Malfunction of α-synuclein because of deletion may cause aberrant olfactory neurons projection. This extended our knowledge of α-synuclein functions, which may explain why olfaction is usually impaired in some synucleinopathies.

Abstract In view of the cost and reliability issues caused by mechanical position sensors, position sensorless control (PSC) of permanent magnet synchronous motors (PMSMs) has received widespread attention. A series of rotor position estimation (RPE) schemes have been developed, in which the phase‐locked loop (PLL) plays an important role. However, the conventional phase‐locked loops face two thorny issues, one is the degradation of RPE performance under motor acceleration and deceleration conditions, and the other is high noise sensitivity. To this end, an adaptive gain PLL (AG‐PLL) is proposed for PSC of PMSM drives. The gains of the loop filter in the proposed AG‐PLL are adaptively adjusted with the estimation error in order to find a compromise between the estimation performance under motor acceleration and deceleration conditions and the noise sensitivity. Moreover, the proposed scheme avoids increasing the order of the PLL, so its dynamic performance is also guaranteed. The proposed scheme is tested based on the hardware‐in‐the‐loop platform.

Abstract Background Fatty acid binding protein 1 (FABP1), a low molecular weight intracellular protein, has been proposed as a potential useful serum biomarker for liver injury. However, limited investigations have been conducted in chronic hepatitis B virus (HBV)-related liver disease. Objective To investigate the diagnostic potential of FABP1 in disease progression among patients with chronic HBV-related liver disease. Methods A prospective study was conducted on 293 patients with chronic HBV-related liver diseases, including chronic asymptomatic carrier (ASC), chronic hepatitis B (CHB). FABP1 was measured in serum samples collected at admission and some selected liver biopsies. Results Immunohistochemical analysis revealed abundant cytoplasmic expression of FABP1 in hepatocytes. A significant negative correlation was observed between FABP1 expression and inflammation grades in liver tissue (Spearman's r = -0.355, P = 0.017). However, no statistically significant correlation was found with fibrosis (P > 0.05). Abstract Background It is very important to provide the correct nursing care for patients with intracerebral hemorrhage (ICH), but the level of critical care needs in patients with intracerebral hemorrhage is not clear. The purpose of this study is to establish a risk model based on the epidemiological and clinical characteristics of ICH patients, to help identify the critical care needs of ICH patients. Methods The clinical data of ICH patients from January 2018 to September 2023 were analyzed retrospectively. The full cohort was used to derive the clinical prediction model and the model was internally validated with bootstrapping. Discrimination and calibration were assessed using the area under curve (AUC) and the Hosmer-Lemeshow tests, respectively. Results 611 patients with ICH were included for model development. 61.21% (374/611) ICH patients had received critical care interventions. The influencing factors included in the model were Glasgow Coma Scale (GCS) score, intraventricular hemorrhage, past blood pressure control, systolic blood pressure on admission and bleeding volume. The model’s goodness-of-fit was evaluated, which yielded a high area under the curve (AUC) value of 0.943, indicating a good fit. For the purpose of model validation, a cohort of 260 patients with ICH was utilized. The model demonstrated a Youden’s index of 0.750, with a sensitivity of 90.56% and a specificity of 78.22%. Conclusion GCS, systolic blood pressure, intraventricular hemorrhage, bleeding volume and past blood pressure control are the main factors affecting the critical care needs of patients with ICH. This study has deduced a clinical predictive model with good discrimination and calibration to provide scoring criteria for clinical health care providers to accurately evaluate and identify the critical care needs of ICH patients, to improve the rational integration and allocation of medical resources.

Abstract Alzheimer’s disease (AD) is a common neurodegenerative disease characterized by progressive cognitive and physical impairment. Neuroinflammation is related to AD, and the misfolding and aggregation of amyloid protein in the brain creates an inflammatory microenvironment. Microglia are the predominant contributors to neuroinflammation, and abnormal activation of microglia induces the release of a large amount of inflammatory factors, promotes neuronal apoptosis, and leads to cognitive impairment. In this study, we used microglial membranes containing caffeic acid-coupled carbon quantum dots to prepare a novel biomimetic nanocapsule (CDs-CA-MGs) for the treatment of AD. The application of CDs-CA-MGs via nasal administration can bypass the blood‒brain barrier (BBB) and directly target the site of inflammation. After treatment with CDs-CA-MGs, AD mice showed reduced inflammation in the brain, decreased neuronal apoptosis, and significantly improved learning and memory abilities. In addition, CDs-CA-MGs affect inflammation-related JAK-STAT and Toll-like receptor signaling pathways in AD mice. CDs-CA-MGs significantly downregulated interleukins (IL-1β and IL-6) and tumor necrosis factor (TNF-α). This finding suggested that CDs-CA-MGs may improve cognitive impairment by modulating inflammatory responses. In conclusion, the use of CDs-CA-MGs provides a possible therapeutic strategy for the treatment of AD. Graphical abstract

Taking the No.4 coal seam in the return air roadway in the upper formation No.1 coal mining area of a mine as the background, based on the energy dissipation theory, the energy dissipation mechanical model of arch roadway was established based on the energy balance equation and the energy analysis theory of surrounding rock loose circle, and the failure range of surrounding rock loose circle was positively correlated with the energy dissipation. Based on the Moore-Coulomb strain softening constitutive model, the energy dissipation fish program based on FLAC3D software was developed and numerical simulation was carried out, and the average energy dissipation range of the roadway in the model was determined to be 5.9 m. Combined with the field drilling imaging test, the failure range of the loose ring of the surrounding rock of the roadway was determined to be 5.9 m and 6.04 m respectively. The above content proves that the energy dissipation theory can be well applied to the analysis of the loose zone of surrounding rock.

The genus Phyllothelys Wood-Mason, 1876 (Mantodea, Hymenopodidae) includes 22 species, with China being a major diversity hotspot, hosting 15 species in four groups.A new species of this genus from Malipo, Yunnan, China, is described herein, namely Phyllothelys qingjinum sp. nov. A comprehensive taxonomic description, including geometric morphometric analysis, genetic distance and molecular phylogenetic analysis, were employed to illustrate its distinctiveness. This discovery fills a significant distribution gap for the P. werneri species group in southern Yunnan and emphasises the need for further exploration in this region.

Abstract The intricate causes of Alzheimer's disease (AD) hinder effective, lasting treatment. Although the dietary modulation of the brain–gut axis was explored for AD therapy, the exact mechanism remains unclear. This study suggested that 140 days of the whey protein hydrolysate (WPH) intake could attenuate the AD pathologic symptoms in APP/PS1 transgenic mice via a bidirectional action of the gut microbe–SCFA (short‐chain fatty acid)–brain axis. Abstract Radiotherapy (RT) is a crucial treatment for colorectal cancer (CRC) patients, but it often fails to induce systemic antitumor immunity. CD73, an immunomodulatory factor, is upregulated after RT and associated with poor prognosis in CRC patients. This study aims to elucidate the mechanisms driving RT-induced CD73 upregulation in CRC and investigate how combining RT with CD73 blockade stimulates immune responses and induces abscopal effects. Findings revealed that RT-induced CD73 upregulation is mediated by the ataxia telangiectasia and Rad3-related (ATR) pathway and correlated with RT tolerance, as demonstrated through flow cytometry, immunofluorescence, and Western Blotting. Using flow cytometry and multicolor immunofluorescence, experiments demonstrated that in CRC subcutaneous tumor models, combination therapy reduces the infiltration of myeloid-derived suppressor cells (MDSCs), tumor-associated macrophages (TAMs), and regulatory T cells (Tregs) while increasing dendritic cells (DCs) and CD8 + T cells, resulting in superior antitumor responses. Additionally, results from flow cytometry, Western Blot, and RNA sequencing demonstrated that combination therapy enhances the antigen-presenting ability of DCs and activates tumor antigen-specific CD8 + T cells, improving their function and delaying their depletion. The activation of the cGAS-STING and IFN-I pathways is crucial for this effect. In summary, the integration of RT with CD73 blockade effectively reverses the immunosuppressive TME and invigorates CD8 + T cell-driven, specific antitumor immune responses. These insights shed fresh light on the mechanisms governing the synergistic modulation of immunity by RT and CD73 blockade in CRC, offering promising avenues for the advancement of therapeutic strategies against CRC.

Abstract Allosteric drugs offer a new avenue for modern drug design. However, the identification of cryptic allosteric sites presents a formidable challenge. Following the allostery nature of residue-driven conformation transition, we propose a state-of-the-art computational pipeline by developing a residue-intuitive hybrid machine learning (RHML) model coupled with molecular dynamics (MD) simulation, through which we can efficiently identify the allosteric site and allosteric modulator as well as reveal their regulation mechanism. For the clinical target β2-adrenoceptor (β2AR), we discover an additional allosteric site located around residues D792.50, F2826.44, N3187.45 and S3197.46 and one putative allosteric modulator ZINC5042. Using Molecular Mechanics/Generalized Born Surface Area (MM/GBSA) and protein structure network (PSN), the allosteric potency and regulation mechanism are probed to further improve identification accuracy. Benefiting from sufficient computational evidence, the experimental assays then validate our predicted allosteric site, negative allosteric potency and regulation pathway, showcasing the effectiveness of the identification pipeline in practice. We expect that it will be applicable to other target proteins.

Abstract Background Combining antitumor proprietary Chinese medicine (pCm) with radiotherapy and chemotherapy can effectively improve tumor cure rates and enhance patients’ quality of life. Gastric cancer (GC) severely endangers public health. Despite satisfactory therapeutic effects achieved by using antitumor pCm to treat GC, its underlying mechanism remains unclear. Objective To integrate existing research data, construct a database of antitumor pCm, and study the intervention mechanisms in GC by focusing on their monomer components. Methods We constructed an antitumor pCm database based on China’s medical insurance catalog, and employed network pharmacology, molecular docking methods, cell experiments, transcriptomics, and bioinformatics to investigate the intervention mechanisms of effective pCm components for GC. Results The study built an antitumor pCm database including 55 pCms, 171 Chinese herbal medicines, 1955 chemical components, 2104 targets, and 32 disease information. Network pharmacology and molecular docking technology identified norcantharidin as an effective component of antitumor pCm. In vitro experiments showed that norcantharidin effectively inhibited GC cell proliferation, migration, and invasion; blocked the G2/M cell cycle phase; and induced GC cell apoptosis. Transcriptomic results revealed that norcantharidin affected biological processes, such as cell adhesion, migration, and inflammatory responses by influencing PI3K-AKT, NF-κB, JAK-STAT, TNF-α signaling pathways, and EMT-related pathways. Core molecules of norcantharidin involved in GC intervention include SERPINE1, SHOX2, SOX4, PRDM1, TGFR3, TOX, PAX9, IL2RB, LAG3, and IL15RA. Additionally, the key target SERPINE1 was identified using bioinformatics methods. Conclusion Norcantharidin, as an effective component of anti-tumor pCm, exerts its therapeutic effects on GC by influencing biological processes such as cell adhesion, migration, and inflammation. This study provides a foundation and research strategy for the post-marketing re-evaluation of antitumor pCms.

Abstract Conventional dielectric solid materials, both natural and artificial, lack electromagnetic self-duality and thus require additional coatings to achieve impedance matching with free space. Here, we present a class of dielectric metamaterials that are effectively self-dual and vacuum-like, thereby exhibiting full-polarization omnidirectional impedance matching as an unusual Brewster effect extended across all incident angles and polarizations. With both birefringence and reflection eliminated regardless of wavefront and polarization, such anisotropic metamaterials could establish the electromagnetic equivalence with “stretched free space” in transformation optics, as substantiated through full-wave simulations and microwave experiments. Our findings open a practical pathway for realizing unprecedented polarization-independence and omnidirectional impedance-matching characteristics in pure dielectric solids.

Aiming at the problem of large deformation and instability control of narrow coal pillar in the vertical stratification section of extra-thick coal seam, the stability and control technology of the surrounding rock of narrow coal pillar in extra-thick coal seam are studied by combining theoretical analysis, numerical simulation and field test. Based on the calculation of the triaxial stress in the narrow coal pillar, the yield criterion of the plane strain of the narrow coal pillar is obtained based on the Mohr-Coulomb failure strength, the failure characteristics and the evolution law of the damage degree of the narrow coal pillar are qualitatively analyzed, and the instability mechanism and size effect of the narrow coal pillar are further analyzed. Finally, the key technologies of surrounding rock control of the narrow coal pillar are given. It is verified by numerical simulation and field test. The research results show that ① The damage degree of the middle part of the narrow coal pillar is greater than that of the two sides, and the damage degree of the middle part from the top to the bottom develops from serious to slight. With the increase of width-height ratio, the damage degree and the proportion of serious damage area in the narrow coal pillar gradually decrease. When the width-height ratio is greater than 1∶1, a large range of slight damage area begins to appear in the middle and lower part of the narrow coal pillar. When the width-height ratio is greater than 5∶3, the proportion of slight damage area exceeds 50%. When C≥3 MPa, or φ≥20°, the damage degree of the two sides of the narrow coal pillar becomes slight. ② The side of the mining roadway in the working face is a low-stress bearing area of the narrow coal pillar. Under the action of high stress in the upper part, the coal body in the large-scale low-stress bearing area moves to the roadway, resulting in a continuous large deformation of the two sides, which in turn affects the stability of the roof. The height of coal pillar is the main controlling factor for the stability of the narrow coal pillar in the middle and bottom layered sections. The increase of width-height ratio of the narrow coal pillar has little influence on the bearing strength of high strength bearing area, while the bearing strength of low strength bearing area increases obviously. Reasonable width-height ratio of the narrow coal pillar can balance the proportion of high and low strength bearing area in coal pillar. ③ The stability of surrounding rock should be considered in the design of layered narrow coal pillars when both sides of the working face are mined out, so as to ensure that the range of high-strength bearing area inside the coal pillar is greater than half of the width and height of the coal pillar. Through the joint strengthening support measures, multiple joint control areas from shallow to deep are formed in the coal pillar to jointly maintain the self-stability of the coal pillar.

Abstract Background During wound healing, fibroblast to myofibroblast transition is required for wound contraction and remodeling. While hypoxia is an important biophysical factor in wound microenvironment, the exact regulatory mechanism underlying hypoxia and fibroblast-to-myofibroblast transition remains unclear. We previously found that tetraspanin CD9 plays an important role in oxygen sensing and wound healing. Herein, we investigated the effects of physiological hypoxia on fibroblast-to-myofibroblast transition and the biological function and mechanism of CD9 in it. Methods Human skin fibroblasts (HSF) and mouse dermis wounds model were established under physiological hypoxia (2% O2). The cell viability and contractility of HSF under hypoxia were evaluated by CCK8 and collagen gel retraction, respectively. The expression and distribution of fibroblast-to-myofibroblast transition markers and CD9 in HSF were detected by Western blotting and immunofluorescence. CD9 slicing and overexpressing HSFs were constructed to determine the role of CD9 by small interfering RNA and recombinant adenovirus vector. The association of TβR2 and TβR1 was measured by immunoprecipitation to explore the regulatory mechanism. Additionally, further validation was conducted on mouse dermis wounds model through histological analysis. Results Enhanced fibroblast-to-myofibroblast transition and upregulated CD9 expression was observed under hypoxia in vitro and in vivo. Besides, reversal of fibroblast-to-myofibroblast transition under hypoxia was observed when silencing CD9, suggesting that CD9 played a key role in this hypoxia-induced transition. Moreover, hypoxia increased fibroblast-to-myofibroblast transition by activating TGF-β1/Smad2/3 signaling, especially increased interaction of TβR2 and TβR1. Ultimately, CD9 was determined to directly affect TβR1–TβR2 association in hypoxic fibroblast. Conclusion Collectively, these findings suggest that CD9 promotes TβR2–TβR1 association, thus driving the transition of human dermal fibroblasts to myofibroblast under hypoxia.

Abstract Background Stroke patients often experience limb dysfunction, which can significantly impact their quality of life and daily living abilities. This study aimed to explore the effectiveness of nursing programs that incorporate multidisciplinary continuing care with the participation of nursing staff for patients with stroke and limb dysfunction. Methods This was a randomized controlled trial (RCT) conducted from August 2021 to August 2023. Ninety stroke patients were randomly assigned to a control group (n = 45) and an observation group (n = 45). The control group received routine discharge care, while the observation group received multidisciplinary continuing care with the participation of nursing staff. Outcomes measured included Fugl-Meyer Assessment (FMA) scores for upper and lower limb function, quality of life, daily living ability (Barthel Index, MBI), and adverse reactions. Results The FMA scores for upper and lower limbs were significantly higher in the observation group compared to the control group. The observation group also had significantly higher scores in all quality of life dimensions and MBI scores compared to the control group. There were 10 adverse reactions reported in the observation group and 22 in the control group. Conclusions Implementing multidisciplinary continuing care with the participation of nursing staff for stroke patients with limb dysfunction has a positive effect on improving limb function, quality of life, and daily living abilities, while also being relatively safe.

Abstract Background Hemorrhagic fever with renal syndrome (HFRS), a life-threatening zoonosis caused by hantavirus, poses significant mortality risks and lacks specific treatments. This study aimed to delineate the transcriptomic alterations during the recovery phases of HFRS. Methods RNA sequencing was employed to analyze the transcriptomic alterations in peripheral blood mononuclear cells from HFRS patients across the oliguric phase (OP), diuretic phase (DP), and convalescent phase (CP). Twelve differentially expressed genes (DEGs) were validated using quantitative real-time PCR in larger sample sets. Results Our analysis revealed pronounced transcriptomic differences between DP and OP, with 38 DEGs showing consistent expression changes across all three phases. Notably, immune checkpoint genes like CD83 and NR4A1 demonstrated a monotonic increase, in contrast to a monotonic decrease observed in antiviral and immunomodulatory genes, including IFI27 and RNASE2. Furthermore, this research elucidates a sustained attenuation of immune responses across three phases, alongside an upregulation of pathways related to tissue repair and regeneration. Conclusion Our research reveals the transcriptomic shifts during the recovery phases of HFRS, illuminating key genes and pathways that may serve as biomarkers for disease progression and recovery.

High resolution isotope ratio mass spectrometry (HR-IRMS) is often used to measure the abundance of deuterium to verify the efficiency of isotope isolation of a cryogenic distillation column system. In order to achieve precise quantification of hydrogen isotope abundance by HR-IRMS, a fine tuning procedure was performed by using molecular hydrogen to optimize the source parameters, steering parameters and analyzer parameters. The mass resolution can reach 10 000 after tuning, which is high enough to separate all potential hydrogen and helium isotopes except for 3He+ and T+. The test results of peak center stability, peak top flatness, resolution and sensitivity stability showed that the performance of the mass spectrometer can meet the demand for hydrogen isotope analysis. The inlet pressure should be in the range of 4-33 Pa to obtain stable sensitivity, and the accelerating voltage of the ion source should not be less than 8 V because it is negatively correlated with the production rate of trimmers (H3+, D3+, H2D+ and HD2+ etc.). As the accelerating voltage decreasing, the production rate of trimers increases. Another key factor affecting the production rate of trimers is the inlet pressure, the square of which is proportional to the production rate of trimers. Therefore, optimizing the ion source parameters to obtain higher sensitivity is a better way than increasing the sample inlet pressure. On this basis, the equilibrium hydrogen isotope standards which are essential to obtain the sensitivity of each component of hydrogen isotopes were prepared by a self-made standard gas preparation device, and the sensitivity values of each component were corrected by pure gases and the self-prepared standard with verified compositions. The molecular leak rate for the inlet hole was determined by monitoring the pressure for H2 and D2 as a function of time as the gas leaked out, and the leak rate of the other components could be calculated using the leak rate of H2 or D2 because the leak rate is inversely proportional to the square root of the molar mass. Standard mixtures were analyzed to test precision and trueness of this method. The results showed that the measurement precision is better than 1% for samples with natural deuterium abundance and better than 0.5% for samples with medium to high abundance of deuterium. The measured values are in good agreement with the standard values. Compared to the low resolution mass spectrometry, this method is simple and reliable, does not need to resolve the mass overlap peak, and the accuracy of the measurement does not depend on the degree of hydrogen isotope balance in the sample.

Abstract The family of coinage-metal-based cyclic trinuclear complexes exhibits abundant photophysical properties, promising for diverse applications. However, their utility in biochemistry is often hindered by large particle size and strong hydrophobicity. Meanwhile, the investigation into multi-photon excited luminescence within this family remained undocumented, limiting their potential in bio-imaging. Herein, we unveil the multi-photon excited luminescent properties of pyrazolate-based trinuclear gold(I) clusters, facilitated by excimeric gold(I)···gold(I) interactions, revealing a nonlinear optical phenomenon within this family. Furthermore, to address issues of poor biocompatibility, we employ electrospinning coupled with hydroxypropyl-beta-cyclodextrin as the matrix to fabricate a flexible, durable, transparent, and red emissive film with a photoluminescence quantum yield as high as 88.3%. This strategy not only produces the film with sufficient hydrophilicity and stability, but also achieves the downsizing of trinuclear gold(I) clusters from microscale to nanoscale. Following the instantaneous dissolution of the film in the media, the released trinuclear gold(I) nanoparticles have illuminated cells and bacteria through a real-time, non-toxic, multi-photon bio-imaging approach. This achievement offers a fresh approach for utilizing coinage-metal-based cyclic trinuclear complexes in biochemical fields.

Abstract Objective To investigate the clinical significance of using 3D printing guides in modified unilateral puncture percutaneous vertebroplasty (PVP) for the treatment of osteoporotic vertebral compression fractures (OVCF), and to explore a new method for preventing paravertebral vein leakage during PVP in conjunction with a previous study of the optimal puncture-side bone cement/vertebral volume ratio(PSBCV/VV%). Methods This retrospective study analyzed 99 patients who underwent unilateral puncture PVP between January 2023 and December 2023. Patients were divided into a guide plate group (46 patients) and a conventional group (53 patients). The guide plate group underwent modified unilateral puncture PVP with the guidance of 3D printing guides, while the conventional group underwent unilateral puncture PVP using the conventional pedicle approach. The distribution of bone cement, surgical outcomes, and the occurrence of cement leakage into paravertebral veins were observed in both groups. Results The guide plate group had significantly shorter operating time and required fewer fluoroscopies compared to the conventional group. Abstract Objectives The purpose of this study is to compare the efficacy of mind–body practices (MBPs) and multiple psychological methods, and identify the optimal method for relieving work-related stress among healthcare workers (HCWs) by network meta-analysis (NMA). Methods We applied six electronic databases, namely PubMed, Web of Science, Embase, PsycINFO, Cochrane, and Chinese National Knowledge Infrastructure to identify relevant RCTs from inception to September 16, 2023, and implemented a search strategy based on the PICOS principles. Data selection, extraction, and analysis of bias were carried out independently and in duplicate by separate researchers. State 16.0 was used to conduct NMA for comparing the effectiveness of various therapies. Results We identified 23 studies including MBPs and three different psychological therapies, namely mindfulness-related therapy (MRT), psychoeducational therapy (PT), and comprehensive therapy (CT), which were divided into eleven specific techniques, namely yoga, meditation techniques (MT), Qigong, muscle relaxation(MR), biofeedback therapy (BT), mindfulness-based interventions (MBIs), modified mindfulness-based stress reduction (MBSR-M), mindfulness-based interventions combined with others (MBIs-C), mindfulness-based awareness(MBA), PT and CT. Our NMA results of MBPs and three psychological therapies showed MBPs (SMD = -0.90, CrI:-1.26, -0.05, SUCRA = 99%) were effective for occupational stress in HCWs, followed by MRT(SMD = -0.48, CrI:-0.87, -0.08, SUCRA = 66.5%). NMA results of eleven specific techniques showed yoga (SMD = 1.36, CrI:0.91, 1.81, SUCRA = 97.5%) was the most effective technique in relieving the stress of HCWs, followed by MR (SMD = 1.36, CrI:0.91, 1.81, SUCRA = 87.3%). Conclusions Our study suggested MBPs may be the most effective intervention to improve the occupational stress of HCWs. Furthermore, yoga is likely to be the most optimal of MBPs. Hospital managers should attach importance to yoga in addressing occupational stress among medical workers.

Abstract Electrostatic capacitors play a crucial role as energy storage devices in modern electrical systems. Energy density, the figure of merit for electrostatic capacitors, is primarily determined by the choice of dielectric material. Most industry-grade polymer dielectrics are flexible polyolefins or rigid aromatics, possessing high energy density or high thermal stability, but not both. Here, we employ artificial intelligence (AI), established polymer chemistry, and molecular engineering to discover a suite of dielectrics in the polynorbornene and polyimide families. Many of the discovered dielectrics exhibit high thermal stability and high energy density over a broad temperature range. One such dielectric displays an energy density of 8.3 J cc−1 at 200 °C, a value 11 × that of any commercially available polymer dielectric at this temperature. We also evaluate pathways to further enhance the polynorbornene and polyimide families, enabling these capacitors to perform well in demanding applications (e.g., aerospace) while being environmentally sustainable. These findings expand the potential applications of electrostatic capacitors within the 85–200 °C temperature range, at which there is presently no good commercial solution. More broadly, this research demonstrates the impact of AI on chemical structure generation and property prediction, highlighting the potential for materials design advancement beyond electrostatic capacitors.

Abstract Background Nosocomial infections with heavy disease burden are becoming a major threat to the health care system around the world. Through long-term, systematic, continuous data collection and analysis, Nosocomial infection surveillance (NIS) systems are constructed in each hospital; while these data are only used as real-time surveillance but fail to realize the prediction and early warning function. Study is to screen effective predictors from the routine NIS data, through integrating the multiple risk factors and Machine learning (ML) methods, and eventually realize the trend prediction and risk threshold of Incidence of Nosocomial infection (INI). Methods We selected two representative hospitals in southern and northern China, and collected NIS data from 2014 to 2021. Thirty-nine factors including hospital operation volume, nosocomial infection, antibacterial drug use and outdoor temperature data, etc. Five ML methods were used to fit the INI prediction model respectively, and to evaluate and compare their performance. Results Compared with other models, Random Forest showed the best performance (5-fold AUC = 0.983) in both hospitals, followed by Support Vector Machine. Nitrogen is an indispensable element in the growth and development stages of various crops, as well as an important component of soil nutrients. It plays an important role in farmland ecosystems. Studying the spatial distribution of soil total nitrogen (STN) is essential for improving the efficiency of soil fertility management and for a better understanding of the soil nitrogen cycle. In the study, the STN content data of surface soil (0–20 cm) were obtained from 19,119 sampling sites within an area of 7.47 × 104 km2 based on a multi-purpose regional geochemical survey established in Zhejiang Province, China, from 2002 to 2018. The spatial distribution and main driving factors of STN were determined using the random forest model. The results showed that the average STN content in the topsoil of the Zhejiang Province was 1.48 g⋅kg−1. Among the different land-use types, grassland soil had the highest STN content of 1.55 g⋅kg−1, and alfisol had the highest STN content of 2.59 g⋅kg−1. The spatial distribution of STN content was high in the northern part and low in the southern part of the study area. The results of the random forest analysis showed that nitrogen deposition, pH, and mean annual precipitation (MAP) were the most important factors influencing the spatial variability of STN. The STN content exhibited a nonlinear decrease with an increase in pH and MAP and a nonlinear increase with an increase in nitrogen deposition. This study provides decision-makers with synthesized information on the spatial variability of STN and its driving factors in Zhejiang Province, which is important for accurately grasping the spatial distribution characteristics of STN to implement precision agriculture management measures.

Biochar has the potential to become a more promising carbon source with a broad range of applications in soil agroecosystems. The depletion of soil minerals is a big problem due to soil erosion and leaching of nutrients. Biochar is a soil conditioner used in crop production to improve soil profile, increase fertilizer use efficiency, plant growth and development, carbon sequestration, and reduce greenhouse gas emissions. It is persistent in the environment and retains water, nutrients, and contaminants. It can also be applied in environmental rehabilitation for remediating contaminated soil. Applied biochar significantly enhanced the nutrients in the rhizospheric soil and reduced the bioavailability and uptake of heavy metals. The enhancement of soil nutrient values was the major functional mechanism for enhancing plant protection and production. Overall, the results of this mini-review are significant because they provide the strategies and technological direction for using biochar in sustainable agricultural systems.

Predicting hepatitis B surface antigen (HBsAg) clearance is important for chronic hepatitis B (CHB) patients receiving pegylated interferon-alfa (Peg-IFN) therapy. We aimed to determine the predictive value of serum hepatitis B core antibody (anti-HBc) for HBsAg clearance. A total of 189 HBeAg-negative CHB patients who received Peg-IFN based therapy were retrospectively included and classified into two groups: nucleos(t)ide analogues (NAs) add-on Peg-IFN group (add-on group, n = 94) and Peg-IFN combined with NAs or Peg-IFN monotherapy group (combination or monotherapy group, n = 95). After 48 weeks of treatment, 27.5% (52/189) and 15.9% (30/189) of patients achieved HBsAg clearance and seroconversion, respectively. Patients in the combination or monotherapy group tended to achieve relatively higher HBsAg clearance (31.6% vs. 23.4%, p = 0.208) and seroconversion (21.1% vs. 10.6%, p = 0.050) rates than those in the add-on group. An understanding of the relationship between CO2 emissions and land use can provide a deeper understanding of how urbanization affects climate change, provide insights into sustainable urban development, and reveal CO2 emission dynamics in the context of urbanization. However, the accuracy of these predictions is limited due to the poor understanding of the influence of differences in urban hierarchy on land use transition rules. This study proposes an integrated CO2 emission prediction method that utilizes a newly developed future land use simulation (partitioned-FLUS) model and a Long Short-Term Memory (LSTM) model. Notably, the partitioned-FLUS model considers regional heterogeneities based on urban hierarchies. Based on data from the Yangtze River Delta, zonal partitioning was found to improve Kappa coefficients by at least 13% compared to conventional FLUS models. The results indicated that a significant urbanization belt was likely to form both along and south of the Yangtze River over the next 30 years; resulting in areas of concentrated CO2 emissions. The relationship between land use and CO2 emissions varied greatly between cities of different urban hierarchies. This study suggests that construction land and farmland in Level 1 and 2 cities should be reallocated to Level 3 cities, thereby reducing carbon emissions.

Norovirus (NV) infection causes acute gastroenteritis in children and adults. Upon infection with NV, specific CD8+ T cells, which play an important role in anti-infective immunity, are activated in the host. Owing to the NV’s wide genotypic variability, it is challenging to develop vaccines with cross-protective abilities against infection. To aid effective vaccine development, we examined specific CD8+ T-cell responses towards viral-structural protein (VP) epitopes, which enable binding to host susceptibility receptors. We isolated peripheral blood mononuclear cells from 196 participants to screen and identify predominant core peptides towards NV main and small envelope proteins using ex vivo and in vitro intracellular cytokine staining assays. Human leukocyte antigen (HLA) restriction characteristics were detected using next-generation sequencing. Three conservative immunodominant VP-derived CD8+ T-cell epitopes, VP294–102 (TDAARGAIN), VP2153–161 (RGPSNKSSN), and VP1141–148 (FPHIIVDV), were identified and restrictively presented by HLA-Cw * 0102, HLA-Cw * 0702, and HLA-A *1101 alleles, separately. Our findings provide useful insights into the development of future vaccines and treatments for NV infection.

AbstractA series of novel benzimidazole derivatives were designed and synthesised based on the structures of reported oral available ALK inhibitor and HDAC inhibitor, pracinostat. In enzymatic assays, compound 3b, containing a 2-acyliminobenzimidazole moiety and hydroxamic acid side chain, could inhibit both ALK and HDAC6 (IC50 = 16 nM and 1.03 µM, respectively). Compound 3b also inhibited various ALK mutants known to be involved in crizotinib resistance, including mutant L1196M (IC50, 4.9 nM). Moreover, 3b inhibited the proliferation of several cancer cell lines, including ALK-addicted H2228 cells. To evaluate its potential for treating cancers in vivo, 3b was used in a human A549 xenograft model with BALB/c nude mice. At 20 mg/kg, 3b inhibited tumour growth by 85% yet had a negligible effect on mean body weight. These results suggest a attracting route for the further research and optimisation of dual ALK/HDAC inhibitors.

Stochastic techno-economic analysis (TEA) is pivotal in assessing the financial viability and risks inherent in biofuel production processes. In this method, the Monte Carlo approach entails the random sampling of input variables and multiple runs of the TEA model to create probability distributions of economic metrics. However, traditional Monte Carlo TEA, reliant on iterative calls to process simulation, is resource-intensive and time-consuming, hindering widespread adoption. To address these challenges, we present an accessible framework that harnesses machine learning methods to estimate techno-economic uncertainty in biofuel production pathways. Our approach streamlines the conventional simulation process by automating dataset generation and machine learning model training. These trained models enable rapid predictions of minimum fuel selling prices at any scale, accommodating randomized input variables based on their defined distributions. We illustrate the efficacy of our framework through examples from sustainable aviation fuel production pathways. Our research entails identifying the primary factors influencing uncertainties in minimum selling prices, exploring the synergistic effects of pathway inputs, and assessing how price variability is impacted by financial, technical, and supply chain factors. These examples underscore the framework's effectiveness in addressing breakeven price uncertainties in biofuel production across diverse input scenarios.

ABSTRACT Aim Although neuroticism has a negative impact on burnout among healthcare workers, there is little research exploring the specific interactions between different components of burnout and neuroticism. This study aims to investigate this relationship among nurses in Emergency Departments (ED) through network analysis method (NAM). Design A cross‐sectional investigative survey was conducted. Methods A total of 408 Chinese nurses were recruited from ED of five hospitals in 2023 through Questionnaire Star. Components of burnout were measured by the 15‐item Maslach Burnout Inventory‐General Survey (MBI‐GS), while components of neuroticism were measured by a subscale of the Eysenck Personality Questionnaire‐Revised, Short Scale for Chinese (EPQ‐RSC) questionnaire. NAM was applied to build relationship between components of burnout and neuroticism and identify the bridge expected influence (BEI). Results Within burnout‐neuroticism network, the relations between the components of burnout and neuroticism were mainly positive. Nt5 (‘Anxious’)‐EE1 (‘Exhausted’; weight = 0.281) and Nt5 (‘Anxious’)‐EE5 (‘Breakdown’; weight = 0.280) had stronger connections. Among the neuroticism, Nt5 (‘Anxious’) showed the strongest positive BEI, while Nt12 (‘Guilty’) showed the strongest negative BEI. Feeling anxiety and less guilt conscience in neuroticism were the critical factors to impact burnout of nurses in ED. Awareness of these relationships can help to develop strategies to identify and curtail burnout. Patient or Public Contribution A total of 408 emergency nurses participated in this survey.

Although severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) variants raise concerns about decreased vaccine efficacy, vaccines continue to confer robust protection in humans, implying that immunity beyond neutralization contributes to vaccine efficacy. In addition to neutralization, antibodies can mediate various Fc-dependent effector functions, including antibody-dependent cellular phagocytosis (ADCP), antibody-dependent neutrophil phagocytosis (ADNP) and antibody-dependent cellular cytotoxicity (ADCC). However, the specific role of each Fc-mediated effector function in contributing to COVID-19 disease attenuation in human remains unclear. To fully define the potential immune correlates of Fc-mediated effector functions, we comprehensively analyzed the above Fc-mediated effector functions in two study cohorts. In the CoronaVac vaccinee cohort, individuals without breakthrough infection exhibited higher levels of ADCP and ADNP activities with a greater degree of cross-reactivity compared to those who had breakthrough infection. A predictive model was established incorporating ADNP activity and IgG titer, achieving an area under the curve (AUC）of 0.837. In the COVID-19 patient cohort, BA.5-specific ADCP and ADNP responses were significantly reduced in COVID-19 patients with fatal outcomes compared to milder outcomes. The prognostic model incorporating WT, BA.5, and XBB.1.5 spike-specific ADNP demonstrated effective predictive ability, achieving an AUC of 0.890. Meanwhile, transcriptomic analysis of peripheral blood mononuclear cells (PBMCs) from COVID-19 patients in the acute phases of infection highlighted remarkably upregulation of neutrophil activity and phagocytic function, further reinforcing the essential role of ADNP. Collectively, our findings underscored Fc-mediated effector activities, especially neutrophil phagocytosis, as significant antibody biomarkers for the risk of SARS-CoV-2 breakthrough infection and COVID-19 prognosis.

This research investigates the influence of performance metrics on match outcomes and constructs a predictive model using data from the Qatar World Cup. Employing magnitude-based decision and an array of machine learning algorithms, such as Decision Trees, Logistic Regression, Support Vector Machines, AdaBoost, Random Forests, and Artificial Neural Network, we examined data from 59 matches, excluding extra time. Fourteen performance indicators were integrated into the model, with two types of match outcomes—winning and non-winning—serving as the output variables. The ANN model exhibited the highest predictive performance, achieving an accuracy of 75.42%, an AUC of 76.96%, a precision of 72.73%, a recall of 65.31%, a specificity of 77.03%, and an F1 score of 68.82%. SHAP analysis revealed that “On Target”, “Shooting Opportunity”, and “Ball Progressions” were the most influential features. These findings underscore the critical role of shooting accuracy and the creation of scoring opportunities in determining match outcomes. Consequently, this study developed an accurate model for predicting match outcomes and meticulously analyzed the match performance. Coaches should prioritize the sensitive indicators identified in this study during training and structure training sessions accordingly.

IntroductionThe hepatitis C virus (HCV) poses a major global health challenge, with its non-structural proteins being essential for viral replication and pathogenesis. Mutations in these proteins significantly contribute to drug resistance, necessitating innovative therapeutic strategies. This study aims to identify epitope-based therapeutic targets in the non-structural proteins of HCV genotype 1, employing in-depth in silico tools to counteract emerging drug resistance.MethodsWe retrieved approximately 250 sequences of each non-structural protein from the NCBI database, capturing a broad spectrum of variability and sequence alignments, variability analysis and physicochemical property analysis were conducted. We utilized the TEPITOOL server by IEDB to predict cytotoxic T lymphocyte (CTL) epitopes. Following this, we assessed the efficiency of TAP transport and proteasomal cleavage using IEDB’s combined predictor tool. The epitopes were selected based on conservancy analysis, immunogenicity, allergenicity, and presence in non-glycosylated regions, ensuring high predictive scores and suitability as vaccine candidates. Epitopes were docked with the HLA-A*02:01 allele and Toll-like receptor-3 using the ClusPro server. The immune response potential of the epitopes was evaluated through in-silico immune stimulation.ResultsThe study identified 27 potential CTL epitopes from the non-structural proteins, including NS3, NS4a, NS4b, NS5a, and NS5b. Out of these, three lead epitopes demonstrated high conservation (>90%), strong binding affinities to HLA-A*02:01 and TLR-3, and robust immune response potential. These epitopes also showed favorable characteristics such as being non-allergenic and non-glycosylated.ConclusionThis comprehensive in-silico analysis provides a promising foundation for developing an epitope-based vaccine targeting HCV non-structural proteins, offering a novel approach to overcoming drug resistance in HCV treatment.

Background and aims“Astringent seed” is a type of abortive seed frequently observed in Chinese fir (Cunninghamia lanceolata). It is widely recognized but poorly understood for its underlying causes. This study investigates the potential of astringent seeds to alleviate the toxic effects of active aluminum ions.MethodsThis study involved treating seeds and seedlings with two distinct concentrations of astringent seeds water extracts under the aluminum ion stress. Then the germination of seeds and growth of seedlings were evaluated and compared.ResultsUnder aluminum stress, both seed germination and seedling growth were notably inhibited. Treatment with a low-concentration of the extract significantly alleviated this inhibition. Root elongation in the seedlings increased by 36.95% compared to the control group, and the aluminum ion accumulation at the root tips was reduced by 38.89% relative to the aluminum-stressed group. This treatment also normalized the levels of malondialdehyde (MDA) in the roots and leaves, enhanced the activities of antioxidative enzymes such as superoxide dismutase (SOD) and catalase (CAT), and restored the levels of endogenous hormones including gibberellin (GA3), indole-3-acetic acid (IAA), methyl jasmonate (Ja-ME), and abscisic acid (ABA). Furthermore, the low-concentration of the extract positively impacted the disorganized chloroplast structures. In contrast, a high-concentration of the extract failed to revert most of these stress indicators.ConclusionLow concentrations of astringent seed water extract effectively alleviate the inhibitory effects of aluminum ions on seed and seedling. This implies that in natural environments, the proximity of healthy seeds to astringent seeds could potentially enhance their growth.

Summary: Capturing the genetic diversity of different wild populations is crucial for unraveling the mechanisms of adaptation and establishing links between genome evolution and local adaptation. The Asian corn borer (ACB) moth has undergone natural selection during its adaptative evolution. However, structural variants (SVs), which play significant roles in these adaptation processes, have not been previously identified. Here, we constructed a multi-assembly graph pan-genome to highlight the importance of SVs in local adaptation. Our analysis revealed that the graph pan-genome contained 176.60 Mb (∼37.33%) of unique sequences. Subsequently, we performed an analysis of expression quantitative trait loci (QTLs) to explore the impact of SVs on gene expression regulation. Notably, through QTL mapping analysis, we identified the FTZ-F1 gene as a potential candidate gene associated with the traits of larval development rate. In sum, we explored the impact of SVs on the local adaptation of pests, therefore facilitating accelerated pest management strategies.

Summary: Many insect species, including the economically important pest Helicoverpa armigera, avoid unfavorable conditions by suspending development. This form of phenotypic plasticity—facultative diapause—is a complex trait, though its evolution and intricate genetic architecture remain poorly understood. To investigate how such a polygenic trait could be locally adapted, we explore its genetic architecture. We map a large-effect diapause-associated locus to the Z chromosome by crossing high- and low-latitude populations. By generating multiple chromosome-scale assemblies, we identify an ∼5.93-Mb chromosomal inversion that constitutes the locus. Within this inversion, 33 genes harbor divergent non-synonymous mutations, notably including three circadian rhythm genes: Period, Clock, and Cycle. CRISPR-Cas9 knockout experiments confirm that each gene is independently essential for pupal diapause. Thus, a diapause supergene arose within H. armigera via a Z chromosome inversion, enabling local climatic adaptation in this economically important crop pest.

Summary: Current solid- and liquid-state electrode materials with extreme physical states show inherent limitation in achieving the ultra-stable batteries. Herein, we present a colloidal electrode design with an intermediate physical state to integrate the advantages of both solid- and liquid-state materials. The colloidal electrode was designed based on the inherent water competition effect of (SO4)2− from the aqueous electrolyte and inherently water-soluble polyethylene glycol (PEG)/ZnI2 from the cathode. The constructed aqueous Zn||PEG/ZnI2 colloid battery demonstrated ultra-stable cycling performance with Coulombic efficiencies approaching 100% and a capacity retention of 86.7% over 10,700 cycles, without requiring anodic modification. In addition, the battery also exhibits compatibility with multiple operating conditions including fluctuating charging, limited self-discharging rate, different charging statuses, and fast charging. Moreover, the battery also shows practical potential by integrating with a photovoltaic solar panel charging. This design provides a broad platform for building the next-generation aqueous batteries with ultra-long lifetime.

IntroductionThe oxygen reduction reaction (ORR) is a crucial determinant of the energy transformation capacity of fuel cells. This study investigates the performance of N and B dual-doped carbon in ORR.MethodsSix models using density functional theory (DFT) are developed to compare the performance of different doping strategies. A highly efficient dual-doped carbon ORR catalyst (S-850-1) is synthesized from Saccharina japonica, containing 4.54 at% N and 1.05 at% B atom.ResultsElectrochemical analysis reveals that S-850-1 significantly outperforms the nitrogen mono-doped carbon S-850, exhibiting a higher half-wave potential of 0.861 V and a greater limited current density of −5.60 mA cm⁻2, compared to S-850’s 0.838 V and −5.24 mA cm⁻2. Furthermore, S-850-1 surpasses the performance of 20% Pt/C, demonstrating enhanced durability and exceptional resistance to CO and methanol. The 1.40 V open circuit voltage produced by S-850-1 when integrated into a Zn-air battery can power an LED light.DiscussionBoth theoretical and practical evaluations validate the excellent ORR performance of nitrogen and boron dual-doped carbon, as evidenced by the agreement between the electrochemical results and DFT calculations. This work not only extends the range of ORR catalysts derived from biomass but also provides guidance on creating and producing affordable, effective catalysts that utilize natural resources.
