Schisandra chinensis is a functional fruit with tonic effect and was widely used by traditional Chinese medicine for treatment and health care. The quality of Schisandra chinensis fruit may vary by different storage condition. In this study, the influence of ambient temperature, humidity, packaging materials and period of storage on the quality of Schisandra chinensis fruits were investigated. The contents of main active components lignans and organic acids were simultaneously determined by ultra-performance liquid chromatography coupled to quadrupole electrostatic field orbitrap high resolution mass spectrometry (UPLC Orbitrap HRMS). The antioxidant activity was determined using DPPH radical scavenging capacity, ABTS+ inhibition rate and FRAP value. The correlation of multicomponent and antioxidant activity was analyzed by grey relevance analysis. Taking the changes of multicomponent and antioxidant activity as investigation index, Schisandra chinensis fruits under different storage conditions was comprehensively evaluated. Schisandrol A, malic acid, sorbic acid, schizandrin A, schizandrin B, and schisandrol B were the main effective components of antioxidant activity. Ambient temperature at 5 °C and humidity at 40 % were more suitable for Schisandra chinensis fruits and kraft paper bag was better packaging material. Do not exceed 1 year was the effective storage period. For the safety evaluation, no aflatoxin was detected within the storage period of 2 years, demonstrated the storage was satisfactory. This study provided a reference for the high-quality storage and standardized operating procedures for storage of Schisandra chinensis fruits.

Ginsenoside Rd is a tetracyclic triterpenoid derivative, widely existing in Panax ginseng, Panax notoginseng and other traditional Chinese medicines. Many studies have proved that ginsenoside Rd have a variety of significant biological activities on certain types of cancer. However, the mechanism of ginsenoside Rd remains unclear in lung cancer. Continuous cropping obstacles seriously constrained the sustainable development of the ginseng industry. The allelopathic autotoxicity of ginsenosides is the key “trigger” of continuous cropping obstacles in ginseng. During harvest, the ginseng plants could be broken and remain in the soil. The decomposition of ginseng residue in soil is one of the important release ways of ginsenosides. Therefore, the allelopathic mechanism of ginsenosides through the decomposed release pathway needs an in-depth study. To investigate this allelopathic regulation mechanism, the integrated analysis of transcriptomics and metabolomics was applied. The prototype ginsenosides in ginseng were detected converse to rare ginsenosides during decomposition. The rare ginsenosides caused more serious damage to ginseng hairy root cells and inhibited the growth of ginseng hairy roots more significantly. By high-throughput RNA sequencing gene transcriptomics study, the significantly differential expressed genes (DEGs) were obtained under prototype and rare ginsenoside interventions. These DEGs were mainly enriched in the biosynthesis of secondary metabolites and metabolic pathways, phytohormone signal transduction, and protein processing in endoplasmic reticulum pathways. Based on the functional enrichment of DEGs, the targeted metabolomics analysis based on UPLC-MS/MS determination was applied to screen endogenous differential metabolized phytohormones (DMPs). The influence of prototype and rare ginsenosides on the accumulation of endogenous phytohormones was studied. These were mainly involved in the biosynthesis of diterpenoid, zeatin, and secondary metabolites, phytohormone signal transduction, and metabolic pathways. After integrating the transcriptomics and metabolomics analysis, ginsenosides could regulate the genes in phytohormone signaling pathways to influence the accumulation of JA, ABA, and SA. The conclusion was that the prototype ginsenosides were converted into rare ginsenosides by ginseng decomposition and released into the soil, which aggravated its allelopathic autotoxicity. The allelopathic mechanism was to intervene in the response regulation of genes related to the metabolic accumulation of endogenous phytohormones in ginseng. This result provides a reference for the in-depth study of continuous cropping obstacles of ginseng.

One of the malignant tumors with a high occurrence rate worldwide is gastric carcinoma, which is an epithelial malignant tumor emerging from the stomach. Natural product polysaccharides are a kind of natural macromolecular polymers, which have the functions of regulating immunity, anti-oxidation, anti-fatigue, hypoglycemia, etc. Natural polysaccharides have remarkable effectiveness in preventing the onset, according to studies, and development of gastric cancer at both cellular and animal levels. This paper summarizes the inhibitory mechanisms and therapeutic significance of plant polysaccharides, fungi polysaccharides, and algal polysaccharides in natural product polysaccharides on the occurrence and development of gastric cancer in recent years, providing a theoretical basis for the research, development, and medicinal value of polysaccharides.

ABSTRACTReductive soil disinfestation (RSD) is an effective method to inhibit soilborne pathogens. However, it remains unclear how RSD combined with different types of organic materials affects the soil ecosystems of perennial plants. Pot experiments were conducted to investigate the effects of RSD incorporated with perilla (PF), alfalfa (MS), ethanol, and acetic acid on soil properties, enzyme activities, microbial communities and functions, and seedling growth. Results showed that RSD-related treatments improved soil properties and enzyme activities, changed microbial community composition and structure, enhanced microbial interactions and functions, and facilitated seedling growth. Compared with CK, RSD-related treatments increased soil pH, available nitrogen, and available potassium contents, sucrase and catalase activities, and decreased soil electric conductivity values. Meanwhile, RSD-related treatment also significantly reduced the relative abundance of Fusarium while increasing the relative abundance of Arthrobacter, Terrabacter, and Gemmatimonas. The reduction was more evident in PF and MS treatment, suggesting the potential for RSD combined with solid agricultural wastes to suppress pathogens. Furthermore, the microbial network of RSD-related treatment was more complex and interconnected, and the functions related to carbon, nitrogen, sulfur, and hydrogen cycling were significantly increased, while the functions of bacterial and fungal plant pathogens were decreased. Importantly, RSD-related treatments also significantly promoted seed germination and seedling growth. In summary, RSD combined with solid agricultural wastes is better than liquid easily degradable compounds by regulating the composition and function of microbial communities to improve soil quality and promote plant growth.IMPORTANCEReductive soil disinfestation (RSD) is an effective agricultural practice. We found that RSD combined with solid agricultural wastes is better than that of liquid easily degradable compounds, may improve soil quality and microbial community structure, inhibit the proliferation of pathogenic bacteria, and contribute to the growth of replanted crops. Thus, RSD combined with solid agricultural wastes is more effective than liquid easily degradable compounds.

Abstract Background Neuroblastoma is one of the common extracranial tumors in children (infants to 2 years), accounting for 8 ~ 10% of all malignant tumors. Few special drugs have been used for clinical treatment currently. Results In this work, herbal extract ginsenosides were used to synthesize fluorescent ginsenosides carbon nanodots via a one-step hydrothermal method. At a low cocultured concentration (50 µg·mL− 1) of ginsenosides carbon nanodots, the inhibition rate and apoptosis rate of SH-SY5Y cells reached ~ 45.00% and ~ 59.66%. The in vivo experiments showed tumor volume and weight of mice in ginsenosides carbon nanodots group were ~ 49.81% and ~ 34.14% to mice in model group. Since ginsenosides were used as sole reactant, ginsenosides carbon nanodots showed low toxicity and good animal response. Conclusion Low-cost ginsenosides carbon nanodots as a new type of nanomedicine with good curative effect and little toxicity show application prospects for clinical treatment of neuroblastoma. It is proposed a new design for nanomedicine based on bioactive carbon nanodots, which used natural bioactive molecules as sole source.

Berberine (BBR), as a natural isoquinoline alkaloid, has demonstrated various pharmacological activities, and is widely applied in the treatment of diseases. The quantitative analysis of BBR is important for pharmacological studies and clinical applications. In this work, utilizing the specific interaction between BBR and triplex DNA, a sensitive and selective fluorescent detecting method was established with DNA-templated silver nanoclusters (DNA-AgNCs). After binding with the triplex structure in the template of DNA-AgNCs, BBR quenched the fluorescence of DNA-AgNCs and formed BBR-triplex complex with yellow–green fluorescence. The ratiometric fluorescence signal showed a linear relationship with BBR concentration in a range from 10 nM to 1000 nM, with a detection limit of 10 nM. Our method exhibited excellent sensitivity and selectivity, and was further applied in BBR detection in real samples.

Objective: To better study and make use of the medicinal value of Hericium erinaceus and ginseng. Methods: In this paper, Hericium erinaceus was used as the fermentation strain and ginseng was used as the drug substrate (PGP) for two-way solid fermentation to obtain Hericium erinaceus ginseng two-way solid fermentation bacteria (HEP). The contents of polysaccharides and proteins of fermentation bacteria and different alcohol precipitation components of ginseng drug substrate group in different fermentation periods were determined by phenol sulfuric acid method and Coomassie brilliant blue method. And DPPH·, ·OH and ABTS+· methods were used to determine the antioxidant capacity of different alcohol precipitation components. Results: The total sugar content of 40% alcohol precipitation component was the most. At the 9th day of fermentation, the total sugar content of 40% alcohol precipitation component HEP-40 in fermentation substrate group reached 0.98 mg/g, and the total sugar content of 40% alcohol precipitation component PGP-40 in ginseng matrix group reached 1.03 mg/g. The content of reducing sugar and protein in 90% alcohol precipitation component was the most. At the 30th day of fermentation, the content of reducing sugar in 90% alcohol precipitation component HEP-90 of fermentation substrate group reached 0.18 mg/g, and the content of reducing sugar in 90% alcohol precipitation component PGP-90 of ginseng matrix group reached 0.24 mg/g; At the 40th day of fermentation, the protein content of HEP-90 and PGP-90 reached 0.54 and 0.46 mg/g respectively. The scavenging of DPPH radicals by all alcoholic fractions of the fermentation plasm up to the 24th day of fermentation was good, with DPPH scavenging rates of 37.66%, 60.06% and 73.58% for the polysaccharides HEP-40, HEP-70 and HEP-90 in the fermentation plasm, respectively; At the 40th day of fermentation, the clearance rates of HEP-40, HEP-70 and HEP-90 on OH radical were 33.71%, 54.32% and 94.90%, respectively; When the fermentation reached the 40th day, the clearance ability of the HEP group reached the peak, and the clearance rates of HEP-40, HEP-70 and HEP-90 on ABTS radical were 44.83%, 87.90% and 98.90%, respectively. Conclusion: After two-way solid fermentation of Hericium erinaceus and ginseng, the antioxidant capacity of the pharmacodynamic active components in the fermentation products (HEP) was higher than that of the pharmacodynamic active components in ginseng without fermentation treatment (PGP).

The allelopathic autotoxicity of ginsenosides is an important cause of continuous cropping obstacles in ginseng planting. There is no report on the potential molecular mechanism of the correlation between polarity of ginsenoside components and their allelopathic autotoxicity. This study applied a combination of metabolomics and transcriptomics analysis techniques, combined with apparent morphology, physiological indexes, and cell vitality detection of the ginseng hairy roots, through which the molecular mechanism of correlation between polarity and allelopathic autotoxicity of ginsenosides were comprehensively studied. The hairy roots of ginseng presented more severe cell apoptosis under the stress of low-polarity ginsenoside components (ZG70). ZG70 exerted allelopathic autotoxicity by regulating the key enzyme genes of cis-zeatin (cZ) synthesis pathway, indole-3-acetic acid (IAA) synthesis pathway, and jasmonates (JAs) signaling transduction pathway. The common pathway for high-polarity ginsenoside components (ZG50) and ZG70 to induce the development of allelopathic autotoxicity was through the expression of key enzymes in the gibberellin (GA) signal transduction pathway, thereby inhibiting the growth of ginseng hairy roots. cZ, indole-3-acetamid (IAM), gibberellin A1 (GA1), and jasmonoyl-L-isoleucine (JA-ILE) were the key response factors in this process. It could be concluded that the polarity of ginsenoside components were negatively correlated with their allelopathic autotoxicity.

Folate-mediated one-carbon metabolism (FOCM) is closely associated with postharvest preservation. This study investigated the effects of exogenous nitric oxide (NO) on FOCM, storage quality, energy metabolism, and mitochondrial membrane integrity in cold-storage peach fruit. In this experiment, peaches were soaked with 1.5 mmol L−1S-nitrosoglutathione (GSNO) as NO donor, and the negative treatment (NT) solution containing 5 μmol L−1 carboxy-PTIO (c-PTIO, NO scavenger), 200 μmol L−1 NG-Nitro-L-arginine methyl ester (L-NAME, NO synthase-like enzyme inhibitor), and 200 μmol L−1 sodium tungstate dihydrate (nitrate reductase inhibitor) and stored at 0°C. The results showed that NO decreased the activity of S-adenosylmethionine synthase and S-adenosylhomocysteine hydrolase and increased the activity of methionine sulfoxide reductase A, as well as the content of N5-methyl-THF, the ratio of tetrahydrofolate (THF), homocysteine, methionine, S-adenosylmethionine (SAM), and SAM to S-adenosylhomocysteine compared with the control, indicating that NO effectively increased FOCM flux by affecting the activity of FOCM enzymes. Meanwhile, NO increased the activities of H+-ATPase, Ca2+-ATPase, cytochrome c oxidase, succinate dehydrogenase, and the contents of adenosine triphosphate and adenosine diphosphate, and maintained high energy charge in peaches during storage. NO retarded the increase in mitochondrial permeability transition, reactive oxygen species content, and the decrease in mitochondrial membrane fluidity, membrane potential, and swelling. NT treatment exhibited the opposite results. In conclusion, these results suggested that NO could induce the accumulation of folate and FOCM flux and maintain mitochondrial energy levels, which might be responsible for maintaining the quality of peaches during cold storage.

Root exudates contain plant metabolites secreted by the roots into the soil, such as ginsenosides secreted by the ginseng root. However, little is known about ginseng root exudate and its impact on the chemical and microbial properties of soil. In this study, the effect of increasing concentrations of ginsenosides on the chemical and microbial properties of soil was tested. Chemical analysis and high-throughput sequencing techniques were used to evaluate the soil chemical properties and microbial characteristics following exogenous application of 0.1 mg·L−1, 1 mg·L−1, and 10 mg·L−1 ginsenosides. Ginsenosides application significantly altered soil enzyme activities; SOM-dominated physicochemical properties were significantly reduced which altered the composition and structure of the soil microbial community. In particular, treatment with 10 mg∙L−1 ginsenosides significantly increased the relative abundance of pathogenic fungi such as Fusarium, Gibberella and Neocosmospora. These findings indicate that ginsenosides in root exudates are important factors that may lead to increased deterioration of soil during ginseng cultivation and provided new research direction for the subsequent study on the mechanism of interaction between ginsenosides and soil microbial communities.

Cinobufacini injection (CI), an aqueous extract of Cutis Bufonis, is clinically used for cancer therapy in China, but its molecular mechanism for the treatment of osteosarcoma (OS) remains unclear. We constructed U2OS ectopic subcutaneous tumor model to verify the anti-OS effect of CI in vivo. Meanwhile, cell proliferation of U2OS and MG63 cells was monitored in vitro using the CCK-8 assay, colony formation and morphological changes. Cell cycle arrest and apoptosis were detected by flow cytometry and western blot, which showed that CI significantly inhibited proliferation, induced cell cycle arrest and apoptosis in human OS cells. The further RNA-seq results identified that the Hippo signaling pathway was involved in the anti-OS effect of CI. YAP/TAZ are two major components of the Hippo pathway in breast cancer and are positively regulated by prolyl isomerase PIN1, we assessed their role in OS using both clinicopathological sections and western blots. CI also inhibited PIN1 enzyme activity in a dose-dependent manner, which resulted in impaired PIN1, YAP, and TAZ expression in vitro and in vivo. Additionally, 15 potential compounds of CI were found to occupy the PIN1 kinase domain and inhibit its activity. In summary, CI plays an anti-OS role by down-regulating the PIN1-YAP/TAZ pathway.

IntroductionSaposhnikovia divaricata is a traditional Chinese medicine in China, which is widely used in clinic. The root of S. divaricata is often used as medicine, but little research has been done on its other tissues.MethodsIn this study, the contents of root and leaf of S. divaricata were determined by HPLC, the differentially expressed genes were screened by transcriptome sequencing at molecular level, and then verified by network pharmacology.ResultsThe results showed that the content of 4’-O-β-D-glucosyl-5-O-methylvisamminol in the leaves was significantly higher than that in the roots, which was about 3 times higher than that in the roots. In addition, 10 differentially expressed key enzyme genes were screened in plant hormone signal transduction, phenylpropanoid and flavonoid biosynthetic pathways. C4H and CYP98A were up-regulated in root, while F3H was down-regulated in root. They can be used as important candidate genes for the mechanism of quality difference of S. divaricata. Finally, network pharmacological validation showed that 5-O-methylvesamitol plays an important role in the treatment of ulcerative colitis.DiscussionThese findings not only provide insight into flavonoid biosynthesis in S. divaricata associated molecular regulation, but also provide a theoretical basis for the development and utilization of S. divaricata.

Panax ginseng is a famous valuable folk herb, the price of which particularly depends on its cultivation age. Since the classical chemical analysis usually needs to destroy the plants which is not suitable for the old wild ginseng due to the huge cost, in the current study, we employed the headspace solid-phase microextraction (HS-SPME) GC-MS methodology which has the advantage of time-saving, nondestructive, and green to analyze the growth age of ginseng (roots) via determining the volatile components of ginseng stem and leaves (GSLs) combined with metabolic profiles way. 100 mg of finely ground GSL samples (3–7 years) were extracted by a 65 μm PDMS-DVB fiber in the headspace and then analyzed within 20 mins. The content assay of total saponins, crude polysaccharides, and total protein was also included. As a result of GC-MS-based profiling, the orthogonal partial least-squares discriminant analysis’ (OPLS-DA) score plots showed excellent classification of four comparison groups (the 3- and 4-, 3-and 5-, 3- and 6-, and 3- and 7-year-old GSL) with 100% discrimination rate, respectively. 263 common differential variables were gained referring to the VIP and P values. 32 volatile metabolites were identified and showed good age discrimination capacities with an area under the curve (AUC) value of more than 0.8 by the receiver operating characteristic (ROC) analysis. Remarkably, there was one volatile component of sesquiterpenes, 1-cyclohexene-1-carboxaldehyde, 2, 6, 6-trimethyl-, which showed an excellent prediction with the AUC value of 1.0 among all the four compared groups, which could be the potential biomarker to distinguish all the four compared groups. The total contents of different growth ages of components of ginseng stems and leaves were distinct. The contents of total protein (4.80 ± 0.27%) and crude polysaccharide (24.15 ± 0.89%) in the 4th year of GSLs were the highest among the five cultivation years. The content of total saponins (4.66 ± 0.38%) in the 5th year of GSLs was the highest. These results could provide a novel reference for the GSLs’ harvest and application periods. Moreover, the current study could also supply a promising way to set up a nondestructive, in situ, and green approach to discriminate the growth age of Panax ginseng rapidly without digging the roots out instead of detecting the VOCs of GSLs.

P. ginseng C.A. Meyer is a valuable Chinese herbal medicine that belongs to the Araliaceae family. Major obstacles to the continuous cropping of ginseng have severely restricted the sustainable development of the ginseng industry. The allelopathic effects of triterpenoid saponins play an important role in disorders related to continuous cropping; however, the mechanisms underlying the allelopathic autotoxicity of triterpenoid ginsenosides remain unknown. In this study, we performed mRNA and miRNA sequencing analyses to identify candidate genes and miRNAs that respond differentially to ginsenoside Ro stress in ginseng and their targets. The growth of the ginseng hairy roots was significantly inhibited under Ro stress (0.5 mg/L, Ro-0.5). The inhibition of root growth and injury to root-tip cells promoted the accumulation of the endogenous hormones indole-3-acetic acid and salicylic acid and inhibited the accumulation of abscisic acid and jasmonate acid. The accumulation of ginsenosides, except Rg3, was significantly inhibited under Ro-0.5 stress. An mRNA analysis of the Ro-0.5 and control groups showed that differentially expressed genes were mostly concentrated in the hormone signal transduction pathway. ARF7 and EFM were upregulated, whereas XTH23 and ZOX1 were downregulated. These genes represent important potential candidates for hormone-responsive continuous cropping diseases. In total, 74 differentially expressed miRNAs were identified based on the miRNA sequencing analysis, of which 22 were upregulated and 52 were downregulated. The target genes of ptc-miR156k_L + 1, mtr-miR156b-5p, gma-miR156a_R + 1, and mtr-miR156e all belonged to TRINITY_DN14567_c0_g4, which is a gene in the plant hormone signal transduction pathway. These four miRNAs were all negatively correlated with mRNA, indicating their likely involvement in the response of ginseng to continuous cropping disorders and the regulation of ginsenoside synthesis. Our findings provide useful insights for removing the barriers to continuous ginseng cropping and have important implications in the genetic engineering of plant stress responses.

Background: Floccularia luteovirens (Alb. & Schwein.) Pouzar, is an extremely rare edible and medicinal mushroom in China. The crude polysaccharides of F. luteovirens (FLPs) has significant antioxidant and anti-inflammation activities and exerts excellent protective functions in diabetic nephropathy (DN) complications, but the material basis of the pharmacological effects of FLPs and the molecular mechanism of its pharmacological action are still unclear. Methods: First, we performed systemic composition analysis on extracted and isolated FLPs. Next, the spontaneous db/db mouse DN model was used to investigate the mitigation and protection functions of FLPs in DN and the underlying mechanism through the mammalian target of the rapamycin (mTOR)/GSK-3β/NRF-2 pathway. Results: FLPs contained 65.0% total sugars, 7.2% reducing sugars, 7.93% proteins, 0.36% total flavonoids, 17 amino acids, 13 fatty acids, and 8 minerals. After intragastric administration of FLPs with concentrations of 100, 200 and 400 mg/kg for 8 weeks, FLPs inhibited excessive weight gain, relieved the symptoms of obesity, and significantly improved glucose metabolism and lipid metabolism in the db/db mice. In addition, FLPs were also involved in regulating the indicators of various oxidases and inflammatory factors in the serum and kidney of db/db mice. Conclusions: FLPs effectively improved and relieved kidney tissue injury caused by high glucose, targeted and regulated phospho-GSK-3β, and suppressed inflammatory factor accumulation. Furthermore, FLPs activated the nuclear factor erythroid 2-related factor 2/heme oxygenase 1 (NRF2/HO-1) pathway and enhanced the activity of catalase (CAT) to further play a role in relieving and treating T2DM and nephropathy complications.

Background: Ganoderma lucidum spore powder (GLSP) has abundant pharmacological activities. However, the difference in the hepatoprotective function of sporoderm-broken and sporoderm-unbroken Ganoderma spore powder has not been studied. This study is the first to investigate the effects of both sporoderm-damaged and sporoderm-intact GLSP on the improvement of acute alcoholic liver injury in mice and gut microbiota of mice. Methods: Serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels and interleukin 1β (IL-1β), interleukin 18 (IL-18), and tumor necrosis factor-α (TNF-α) levels in liver tissues from mice in each group were detected by enzyme-linked immunosorbent assay (ELISA) kits, and histological analysis of liver tissue sections was performed to evaluate the liver-protecting effects of both sporoderm-broken and sporoderm-unbroken GLSP. Additionally, 16S rDNA sequencing of feces from the bowels of mice was performed to compare the regulatory effects of both sporoderm-broken and sporoderm-unbroken GLSP on the gut microbiota of mice. Results: Compared with those in the 50% ethanol model group (MG), sporoderm-broken GLSP significantly reduced serum AST and ALT levels (p < 0.0001) and the release of the inflammatory factors, including IL-1β, IL-18, and TNF-α (p < 0.0001), and effectively improved the pathological state of liver cells; sporoderm-unbroken GLSP significantly reduced the ALT content (p = 0.0002) and the release of the inflammatory factors, including IL-1β (p < 0.0001), IL-18 (p = 0.0018), and TNF-α (p = 0.0005), and reduced the serum AST content, but the reduction was not significant; compared with the gut microbiota of the MG, sporoderm-broken GLSP reduced the levels of Verrucomicrobia and Escherichia_Shigella, increased the relative abundance of beneficial bacteria such as Bacteroidetes, and decreased the abundance levels of harmful bacteria, such as Proteobacteria and Candidatus_Saccharibacteria; sporoderm-unbroken GLSP could reduce the abundance levels of harmful bacteria, such as Verrucomicrobia and Candidatus_Saccharibacteria; and GLSP treatment alleviates the downregulation of the levels of translation, ribosome structure and biogenesis, and lipid transport and metabolism in liver-injured mice; Conclusions: GLSP can alleviate the imbalance of gut microbiota and improve liver injury, and the effect of sporoderm-broken GLSP is better.

A valid and encyclopedic evaluation method for assessing the quality of Saposhnikovia divaricata has been set up based on the analysis of a high-performance liquid chromatography (HPLC) fingerprint combined with the cluster analysis (CA), principal component analysis (PCA), partial least square discriminant analysis (PLS-DA), and similarity analysis. 15 peaks of the common model were obtained and used for the similarity analysis, CA analysis, PCA analysis, and PLS-DA analysis. The fingerprint of S. divaricata was established, and 15 common peaks were calibrated. The four common peaks were identified as prim-o-glucosylcimifugin, 4-O-β-D-glucosyl-5-O-methylvisamminol, cimifugin, and sec-o-glucosylhamaudol by comparison with the reference substance. The similarity of the fingerprints of the 33 batches of S. divaricata is above 0.9. Cluster analysis divides the 33 batches of S. divaricata into 2 categories. Principal component analysis (PCA) roughly divides them into 4 categories. Partial least squares method-discriminant analysis (PLS-DA) screened to obtain 2 differential markers, the different components were designated by the reference substance as 4-O-β-D-glucosyl-5-O-methylvisamminol and cimifugin. The fingerprint established by this study combined with chemometrics analysis is reasonable, effective, accurate, and simple, which makes the information more comprehensive and can provide a scientific basis and reference for quality control and quality evaluation of S. divaricata.

Reductive soil disinfestation (RSD) and soil fumigant chloropicrin (SFC) are two common agricultural strategies for the elimination of soil-borne pathogens. However, the differences in soil environmental factors, soil bacterial microbiome, and root performance between SFC and RSD are poorly understood. In this study, three soil treatments, untreated control (CK), SFC with 0.5 t⋅ha–1 chloropicrin, and RSD with 15 t⋅ha–1 animal feces, were compared. We evaluated their effects on soil environmental factors, bacterial community structure, and root activity using chemical analysis and high-throughput sequencing. RSD treatment improved soil composition structure, bacterial diversity, and root performance to a greater extent. Carbon source utilization preference and bacterial community structure were strikingly altered by SFC and RSD practices. Bacterial richness, diversity, and evenness were notably lowered in the SFC- and RSD-treated soil compared with the CK-treated soil. However, RSD-treated soil harbored distinct unique and core microbiomes that were composed of more abundant and diverse potentially disease-suppressive and organic-decomposable agents. Also, soil bacterial diversity and composition were closely related to soil physicochemical properties and enzyme activity, of which pH, available Na (ANa), available Mg (AMg), available Mn (AMn), total Na (TNa), total Ca (TCa), total Cu (TCu), total Sr (TSr), urease (S-UE), acid phosphatase (S-ACP), and sucrase (S-SC) were the main drivers. Moreover, RSD treatment also significantly increased ginseng root activity. Collectively, these results suggest that RSD practices could considerably restore soil nutrient structure and bacterial diversity and improve root performance, which can be applied as a potential agricultural practice for the development of disease-suppressive soil.

Saposhnikovia divaricata is derived from the dried roots of Saposhnikovia divaricata (Turcz.) Schischk and used as a Chinese herbal medicine for treating respiratory, immune, and nervous system diseases. The continuously increasing market demand for traditional Chinese medicine requires the commercial cultivation of Saposhnikovia divaricata using standardized methods and high yielding genotypes, such as double-headed root plants, for achieving consistent quality and a reliable supply. In this study, we aimed to identify the quantitative differences in chromone, a precursor of flavonoid biosynthesis, between plants with single- and double-headed roots using high-performance liquid chromatography and further explore the two phenotypes at the transcriptomic and metabolomic levels. Our results showed that the chromone content was significantly higher in plants with double-headed roots than in those with single-headed roots. Transcriptomic analysis revealed six significantly differentially expressed genes between the two phenotypes, including five key genes in the flavonoid biosynthesis pathway (4-coumarate-CoA ligase, chalcone synthase 1, vinorine synthase, chalcone-flavonone isomerase 1, and flavanone 3 beta-hydroxylase) and one key gene in the abscisic acid biosynthetic pathway (zeaxanthin epoxidase). Moreover, metabolomic analysis showed that the 126 differentially expressed metabolites were mainly enriched in the biosynthesis of secondary metabolites and phytohormones. Overall, our results suggest that plants with double-headed roots have higher medicinal value than those with single-headed roots, probably due to differences in various biosynthetic pathways. These data might help select the genotypes with superior yield and therapeutic properties.

Hydrogen sulfide (H2S), nitric oxide (NO), and reactive oxygen species (ROS) play essential signaling roles in cells by oxidative post-translational modification within suitable ranges of concentration. All of them contribute to the balance of redox and are involved in the DNA damage and repair pathways. However, the damage and repair pathways of mitochondrial DNA (mtDNA) are complicated, and the interactions among NO, H2S, ROS, and mtDNA damage are also intricate. This article summarized the current knowledge about the metabolism of H2S, NO, and ROS and their roles in maintaining redox balance and regulating the repair pathway of mtDNA damage in plants. The three reactive species may likely influence each other in their generation, elimination, and signaling actions, indicating a crosstalk relationship between them. In addition, NO and H2S are reported to be involved in epigenetic variations by participating in various cell metabolisms, including (nuclear and mitochondrial) DNA damage and repair. Nevertheless, the research on the details of NO and H2S in regulating DNA damage repair of plants is in its infancy, especially in mtDNA.

Two polysaccharides were obtained from steamed ginseng via ultrafiltration, and their physical–chemical properties, solution properties and antifatigue activities were studied. WSGP-S3 and WSGP-G3 were acid heteropolysaccharides with the molecular weights of 2.03 × 104 and 4.86 × 104, respectively. They were composed of different molar ratios of the monosaccharides Rha, GlcA, GalA, Glc, Gal, and Ara. The results of size-exclusion chromatography–multiangle laser light scattering analysis, Conge red staining and Circular dichroism spectroscopy revealed that WSGP-S3 exhibited a random conformation of branched clusters in solution. By contrast, WSGP-G3 exhibited an ordered conformation, including helix-like conformations in aqueous solution. Antifatigue activity tests proved that WSGP-S3 markedly prolonged the exhaustive swimming time of fatigued mice; increased liver and muscle glycogen levels and superoxide dismutase, catalase, glutathione peroxidase activities and decreased blood lactic acid, nitrogen and malondialdehyde levels compared with the control treatment. Moreover, it enhanced spleen cell proliferation in fatigued mice. By contrast, WSGP-G3 had no significant effect on fatigued mice. The results showed that WSGP-S3 might have a major contribution to the antifatigue effects of steamed ginseng polysaccharides and could be a potential anti-fatigue polysaccharide.

Panax ginseng is a valuable traditional herbal medicine material with numerous applications. Ginsenosides are the key bioactive compounds in ginseng. Cold stress can activate stress tolerance mechanisms that regulate biomass and biosynthesis in ginseng tissue. In this study, the effects of short- and long-term cold stress (5°C) on the physiological characteristics, tissue-specific ginsenoside distributions, and ginsenoside synthesis gene expressions of 3-year-old P. ginseng during the flowering period were investigated. Short-term cold stress significantly reduced ginseng biomass (root fresh weight and dry weight), and increased malondialdehyde, proline, soluble sugar, and soluble protein concentrations. Superoxide dismutase, peroxidase, and catalase activities also increased significantly under cold stress. With prolongation of the cold stress period, all antioxidant enzyme activity decreased. The protopanaxatriol-type ginsenoside concentrations in the taproots (phloem and xylem) and fibrous roots, as well as the protopanaxadiol-type ginsenoside concentrations in the leaves, increased significantly under short-term cold stress. The key genes (SE, DS-II, CYP716A52v2, and CYP716A53v2) involved in the ginsenoside biosynthesis pathway were significantly positively correlated with the ginsenoside accumulation trends. Thus, short-term cold stress can stimulate membrane lipid peroxidation, in turn stimulating the antioxidant enzyme system to alleviate oxidative damage and increasing the expression of key enzyme genes involved in ginsenoside biosynthesis. During agricultural production, protopanaxadiol/protopanaxatriol ratios could be manipulated by low-temperature storage or treatments.

To explore a green and efficient extraction technology for the extraction of active ingredients of Asarum, the deep eutectic solvent combined with ultrasonic was applied to compare the extraction efficiency of 10 kinds of deep eutectic solvents, taking the extraction rate of methyl eugenol and asarinin as indices. Single-factor experiments were adopted to investigate the influence of molar ratio, liquid-to-solid ratio, eddy time, ultrasonic time, and temperature of the deep eutectic solvent on the extraction rate of methyl eugenol and asarinin. Based on single-factor experiments, the surface response methodology was used to optimize the extraction process conditions. The results showed that the optimum extracting process conditions of methyl eugenol and asarinin in Asarum consisted of a ratio of choline chloride to glycerol of 1 : 3, a DES volume of 2 mL, an ultrasonic temperature of 60°C, an ultrasonic time of 30 min, and a vortex oscillation of 7 min. Under the optimum extracting process conditions, the contents of methyl eugenol and asarinin were 1.9428 mg/g and 0.9989 mg/g, respectively, and the comprehensive index was 2.3280 (RSD of 1.91%). The results were close to the predicted values of the response surface model, demonstrating the applicability of the model. The extraction rate of methyl eugenol and asarinin in Asarum by this method was higher than that of water extraction and alcohol extraction, which fully indicated the high efficiency of ultrasonic-assisted green deep eutectic solvent extraction technology. The results provide data support for further development and utilization of Asarum.

This work aimed at improving the water solubility of Ginsenoside (G)-Re by forming an inclusion complex. The solubility parameters of G-Re in alpha (α), beta (β), and gamma (γ) cyclodextrin (CD) were investigated. The phase solubility profiles were all classified as AL-type that indicated the 1:1 stoichiometric relationship with the stability constants Ks which were 22 M−1 (α-CD), 612 M−1 (β-CD), and 14,410 M−1 (γ-CD), respectively. Molecular docking studies confirmed the results of phase solubility with the binding energy of −4.7 (α-CD), −5.10 (β-CD), and −6.70 (γ-CD) kcal/mol, respectively. The inclusion complex (IC) of G-Re was prepared with γ-CD via the water-stirring method followed by freeze-drying. The successful preparation of IC was confirmed by powder X-ray diffraction (XRD), Fourier transform-infrared spectroscopy (FT-IR), differential scanning calorimetry (DSC), and scanning electron microscopy (SEM). In-vivo absorption studies were carried out by LC-MS/MS. Dissolution rate of G-Re was increased 9.27 times after inclusion, and the peak blood concentration was 2.7-fold higher than that of pure G-Re powder. The relative bioavailability calculated from the ratio of Area under the curve AUC0–∞ of the inclusion to pure G-Re powder was 171%. This study offers the first report that describes G-Re’s inclusion into γ-CD, and explored the inclusion complex’s mechanism at the molecular level. The results indicated that the solubility could be significantly improved as well as the bioavailability, implying γ-CD was a very suitable inclusion host for complex preparation of G-Re.

BACKGROUND: Dickkopf-1 (DKK1) is an antagonist of Wnt/β-catenin signaling implicated in tumorigenesis. However, the biological role of DKK1 and β-catenin involved in chondrosarcoma has not been sufficiently investigated. This study was designed to investigate the expression profiles of DKK1 and β-catenin, and to clarify their clinical values in chondrosarcoma. METHODS: The mRNA and protein levels of DKK1 and β-catenin in fresh chondrosarcoma and the corresponding non-tumor tissues were analyzed by Real-time PCR and Western blot, respectively. The protein expression patterns of DKK1 and β-catenin were investigated by immunohistochemistry. The associations among DKK1 level, β-catenin accumulation, clinicopathological factors and the overall survival were separately evaluated. RESULTS: Both DKK1 and β-catenin levels were remarkably elevated in chondrosarcoma compared with the corresponding non-tumor tissues. High DKK1 level and positive β-catenin accumulation in chondrosarcoma specimens were 58.7% and 53.9%, respectively. Elevated DKK1 level significantly correlated with positive β-catenin accumulation, and they were remarkably associated with histological grade and Musculoskeletal Tumor Society stage. Furthermore, DKK1 level and β-catenin accumulation had significant impacts on the prognosis of chondrosarcoma patients. Multivariate analysis revealed that DKK1 level was an independent prognostic factor for overall survival. CONCLUSIONS: Elevated DKK1 levels associated with β-catenin accumulation play a crucial role in chondrosarcoma. DKK1 can serve as a novel predictor of poor prognosis in patients with chondrosarcoma.
