Objective To investigate the clonal rearrangement characteristics and clinical application value of IGH gene in B-cell non-Hodgkin’s lymphoma (B-NHL). Methods Demographic and clinical data as well as IGH sequencing results of 55 patients with B-NHL who underwent next-generation sequencing (NGS) testing were collected, and IGH gene clonal rearrangement was detected. The characteristics of IGH gene clonal rearrangement, IGHV gene usage, and the clinical application value of NGS for IGH clonal rearrangement were analyzed. Results Among 55 patients with B-NHL and IGH clonal rearrangement, single dominant clones were mainly detected (85.45%, 47/55); a few patients had two (12.73%, 7/55) and three dominant clones (1.82%, 1/55). In terms of preference for IGHV gene usage, IGHV3 gene had the highest frequency of access in B-NHL, followed by IGHV4. Among the IGHV subtypes, IGHV3-23 had the highest frequency in chronic lymphocytic leukemia/small lymphocytic lymphoma, and IGHV4-34 had the highest frequency in primary central nervous system diffuse large B-cell lymphoma and not otherwise specified diffuse large B-cell lymphoma. Conclusion A preference for IGHV gene usage in clonal rearrangement of IGH genes is noted in B-NHL patients with different pathological types. Using NGS to detect IGH clonal rearrangement can identify subclones and clonal correlations, and assist in disease diagnosis.

[Objective] In order to understand the spatiotemporal changes in the wetland of the Genhe Basin, the impacts of climate change and land use change on wetland landscape pattern was clarified. [Methods] The driving factors of changes in wetland landscape pattern in the Genhe Basin between 1990 and 2019 were investigated by analyzing the changes in wetland extent and combining them with climate change during the same period. [Results] The results showed that over the past 30 years, the area of wetlands in the Genhe Basin had decreased by 31.89%, the number of patches had increased by 3.74%, and the landscape fragmentation index had increased by 4.48. The overall degradation of wetland areas in the watershed was significant, while the overall number of wetland patches in the watershed had increased. From 1990 to 2004, wetland areas in the watershed showed a significant degradation trend (p0.05), and the average annual temperature showed a non-significant increasing trend (p>0.05). There was a significant negative correlation between landscape fragmentation index and average annual temperature, which might be due to the melting of shallow permafrost in the watershed due to rising temperatures, leading to a decrease in wetland fragmentation during this period. [Conclusion] The research results are of great significance for maintaining biodiversity and stability of wetland ecosystems in the Genhe Basin, promoting coordinated and interactive wetland-dryland relationships, and ensuring sustainable development of wetland resources.

Intractable hypokalemia and hypocalcemia are rare complications in leukemia patients. This article reports a patient with chronic myelomonocytic leukemia (CMML)with refractory hypokalemia, hypocalcemia, hypochloridemia, hypomagnesemia-a 48-year-old man complained of fatigue, nausea, and face numbness for over one month. The number of monocytosis in peripheral blood and bone marrow increased remarkably. At the same time, the patient developed hypokalemia, hypochloridemia, hypocalcemia, hypomagnesemia, metabolic alkalosis, massive proteinuria, and increase in plasma aldosterone and renin. After two coursea of treatment using Azacitidine therapy, complete remission of bone marrow was achieved and the electrolyte disturbance was almost corrected, except hypomagnesemia. We summarize the clinical characteristics of the patient so as to raise the clinical awareness of such cases.

Background: The aim of this study was to investigate the frequency dependence of electroacupuncture (EA) in alleviating chronic visceral pain in patients with irritable bowel syndrome (IBS) and the differences in the gut microbiota and metabolites as potential mechanisms to explain frequency dependence. Methods: A visceral hyperalgesia model was established by colorectal instillation of 2,4,6‐trinitrobenzene sulfonic acid in rats, and EA treatment at 2/10 Hz, 2/50 Hz and 2/100 Hz was applied at ST25. Visceral sensation was quantified by the abdominal withdrawal reflex score and the area under the curve of the rectus abdominis electromyogram in response to colorectal distension. Ultrastructural morphological damage of colonic tissue of the rats was examined by transmission electron microscopy. 16S rRNA gene sequencing and 1H‐nuclear magnetic resonance spectroscopy were applied to study the differences in the gut microbiota and to perform metabonomic profiling of the colonic tissue. Key Results: EA at ST25 at different frequencies attenuated chronic visceral pain, ultrastructural morphological damage to colonic tissue and disruption of the gut microbiota in IBS rats. The frequency of 2/100 Hz has more regulatory pathways than 2/10 Hz and 2/50 Hz. In addition, IBS rats exhibited colonic metabolic disorders, and pantothenate was significantly upregulated after EA treatment at different frequencies. Very low‐density lipoprotein and 2‐hydroxybutyrate were significantly increased in the 2/10 Hz group, while low density lipoprotein, very low‐density lipoprotein, 2‐hydroxybutyrate, methylmalonate and alpha‐hydroxyisobutyric acid were significantly increased in the 2/100 Hz group. Conclusions and Inferences: EA at ST25 at different frequencies attenuated chronic visceral pain through different gut microbiota and metabolic pathways. [ABSTRACT FROM AUTHOR]

The rice stem borer (RSB) Chilo suppressalis is a devastating rice pest with resistance to a number of insecticides. Recently, the new meta-diamide insecticide cyproflanilide has been considered an effective insecticide to control RSB. However, its resistance risk has not been reported. In the present study, we aimed to assess the resistance risk and evaluate the fitness cost after the RSB was exposed to cyproflanilide. After five generations of selection, the resistance level of RSB increased by 1.5-fold. When h2 was 0.125, a 10-fold resistance increase in the LD50 values was expected in fourteen and thirty-one generations at the selection intensity of 90% and 50%, respectively. The selected population (RSB-SEL) had significant differences in the developmental duration of eggs, 1st, 2nd, 3rd, and 6th instar larvae, and female pupae compared to the unselected population (RSB-UNSEL). Besides, the adult longevity was shortened, and the average pupal weight of males, the emergence rate, the sex ratio, the oviposition, the mean fecundity, and the full life cycle rate were decreased in RSB-SEL. The intrinsic rate of increase (r), the net reproductive rate (R0), and the finite rate of increase (λ) of RSB-SEL were significantly lower than those of RSB-UNSEL, while the mean generation time (T) of RSB-SEL was significantly longer than that of RSB-UNSEL. Based on the results of the prediction of the generations required for a 10-fold resistance increase in the LD50, a potential risk of resistance development exists in RSB after continuous and excessive use of cyproflanilide. Background: Depression remains one of the most prevalent psychiatric disorders, with many patients not responding adequately to available treatments. Electroacupuncture (EA), a nonpharmacologic therapy, holds great promise for alleviating depressive symptoms. In this study, RNA sequencing (RNA‐Seq) was used to identify genome‐wide alterations in the hippocampus of rats after chronic unpredictable mild stress (CUMS) and EA treatments to further elucidate the mechanism by which EA ameliorates depression to provide a basis for the clinical application of EA in stress‐related diseases. Methods: The sucrose preference test (SPT), open field test (OFT), and forced swimming test (FST) were used to investigate the ability of EA at Baihui (GV20) and Taichong acupoints (LR3) to improve depression‐like behavior in rats subjected to CUMS. Subsequently, RNA‐Seq analysis revealed transcriptomic profiles of the hippocampus of rats subjected to CUMS in which EA ameliorated depressive behavior. Finally, the expression profiles of major differentially expressed genes were tested by real‐time quantitative polymerase chain reaction (qRT‐PCR) to determine the accuracy of the RNA‐Seq results. Results: Rats subjected to CUMS exhibited depressive‐like behaviors, such as decreased sucrose consumption in the SPT (p <.001), decreased time in the central area of the OFT (p <.001), and increased immobility in the FST (p <.01). Importantly, rats subjected to CUMS and treated with EA showed increased sucrose consumption (p <.001), increased time spent in the central area of the OFT (p <.001) and decreased immobility in the FST (p <.01). Sixty‐three genes that were differentially expressed following CUMS were altered by EA; most of these were associated with immune pathways. Compared with those in the control group, the expression levels of Colla2 (p <.001), Col3a1 (p <.001), Psmb9 (p <.01), and Tap1 (p <.01) in the hippocampus of rats subjected to CUMS were lower. The changes in the expression of these genes were reversed by EA treatment. Conclusion: EA at GV20 and LR3 attenuated CUMS‐induced depression‐like behaviors by regulating the expression of specific genes such as Colla2, Col3a1, Psmb9, and Tap1. [ABSTRACT FROM AUTHOR]

Under a warming climate, occurrences of wildfires have been increasingly more frequent in boreal and arctic forests during the last few decades. Wildfires can cause radical changes in the forest ecosystems and permafrost environment, such as irreversible degradation of permafrost, successions of boreal forests, rapid and massive losses of soil carbon stock, and increased periglacial geohazards. Since 2016, we have gradually and more systematically established a network for studying soil nutrients and monitoring the hydrothermal state of the active layer and near-surface permafrost in the northern Da Xing'anling (Hinggan) Mountains in Northeast China. The dataset of soil moisture content (0–9.4 m in depth), soil organic carbon (0–3.6 m), total nitrogen (0–3.6 m), and total phosphorus and potassium (0–3.6 m) have been obtained by field sampling and ensuing laboratory tests. Long-term datasets (2017–2022) of ground temperatures (0–20 m) and active layer thickness have been observed by thermistor cables permanently installed in boreholes. The present data can be used to simulate changes in permafrost features under a changing climate and wildfire disturbances and to explore the changing interactive mechanisms of the fire-permafrost-carbon system in the hemiboreal forest. Furthermore, can provide baseline data for studies and action plans to support the carbon neutralization initiative and assessment of ecological safety and management of the permafrost environment. Downward solar radiation (DSR) and air temperature (Ta) have significant influences on the thermal state of frozen ground. These parameters are also important forcing terms for physically based land surface models (LSMs). However, the quantitative influences of inaccuracies in DSR and Ta products on simulated frozen ground temperatures remain unclear. In this study, three DSR products (CMFD‐SR, Tang‐SR, and GLDAS‐SR) and two Ta products (CMFD‐Ta and GLDAS‐Ta) were used to force an LSM model in an alpine watershed in Northwest China, to investigate the sensitivity of simulated ground temperatures to different DSR and Ta products. Compared to a control model (CTRL) forced by in situ observed DSR, ground temperatures simulated by the experimental model forced by GLDAS‐SR are obviously decreased because GLDAS‐SR is much lower than in situ observations. Instead, simulation results in models forced by CMFD‐SR and Tang‐SR are much closer to those of CTRL. Ta products led to significant errors in simulated ground temperatures. In conclusion, both CMFD‐SR and Tang‐SR could be used as good alternatives to in situ observed DSR for forcing a model, with acceptable errors in simulation results. However, more care need to be paid for models forced by Ta products instead of Ta observations, and conclusions should be carefully drawn. [ABSTRACT FROM AUTHOR]
