5. RandNet: deep learning with compressed measurements of images

Abstract

Principal component analysis, dictionary learning, and auto-encoders are all unsupervised methods for learning representations from a large amount of training data. In all these methods, the higher the dimensions of the input data, the longer it takes to learn. We introduce a class of neural networks, termed RandNet, for learning representations using compressed random measurements of data of interest, such as images. RandNet extends the convolutional recurrent sparse auto-encoder architecture to dense networks and, more importantly, to the case when the input data are compressed random measurements of the original data. Compressing the input data makes it possible to fit a larger number of batches in memory during training. Moreover, in the case of sparse measurements,training is more efficient computationally. We demonstrate that, in unsupervised settings, RandNet performs dictionary learning using compressed data. In supervised settings, we show that RandNet can classify MNIST images with minimal loss in accuracy, despite being trained with random projections of the images that result in a 50% reduction in size. Abstract

Transcribing and replicating a double-stranded genome require protein modules to unwind, transcribe/replicate nucleic acid substrates, and release products. Here we present in situ cryo-electron microscopy structures of rotavirus dsRNA-dependent RNA polymerase (RdRp) in two states pertaining to transcription. In addition to the previously discovered universal "hand-shaped" polymerase core domain shared by DNA polymerases and telomerases, our results show the function of N- and C-terminal domains of RdRp: the former opens the genome duplex to isolate the template strand; the latter splits the emerging template-transcript hybrid, guides genome reannealing to form a transcription bubble, and opens a capsid shell protein (CSP) to release the transcript. These two "helicase" domains also extensively interact with CSP, which has a switchable N-terminal helix that, like cellular transcriptional factors, either inhibits or promotes RdRp activity. The in situ structures of RdRp, CSP, and RNA in action inform mechanisms of not only transcription, but also replication.

Abstract

The large H II complex N11 in the Large Magellanic Cloud contains OB associations at several different stages in their life histories. We have obtained ROSAT PSPC and HRI X-ray observations, Curtis Schmidt CCD images, echelle spectra in H-alpha and [N II] lines, and IUE interstellar absorption line observations of this region. The central bubble of N11 has an X-ray luminosity a factor of only 3-7 brighter than predicted for an energy-conserving superbubble, making this the first detection of X-ray emission from a superbubble without a strong X-ray excess. The region N11B contains an extremely young OB association analogous to the central association of the Carina nebula, apparently still embedded in its natal molecular cloud. We find that N11B emits diffuse X-ray emission, probably powered by stellar winds. Finally, we compare the tight cluster HD32228 in N11 to R136 in 30 Dor. Abstract

BackgroundRecent developments in radiotherapeutic technology have resulted in a new approach to treating patients with localized lung cancer. We report preliminary clinical outcomes using stereotactic radiosurgery with real-time tumor motion tracking to treat small peripheral lung tumors.MethodsEligible patients were treated over a 24-month period and followed for a minimum of 6 months. Fiducials (3-5) were placed in or near tumors under CT-guidance. Non-isocentric treatment plans with 5-mm margins were generated. Patients received 45-60 Gy in 3 equal fractions delivered in less than 2 weeks. CT imaging and routine pulmonary function tests were completed at 3, 6, 12, 18, 24 and 30 months.ResultsTwenty-four consecutive patients were treated, 15 with stage I lung cancer and 9 with single lung metastases. Pneumothorax was a complication of fiducial placement in 7 patients, requiring tube thoracostomy in 4. All patients completed radiation treatment with minimal discomfort, few acute side effects and no procedure-related mortalities. Following treatment transient chest wall discomfort, typically lasting several weeks, developed in 7 of 11 patients with lesions within 5 mm of the pleura. Grade III pneumonitis was seen in 2 patients, one with prior conventional thoracic irradiation and the other treated with concurrent Gefitinib. A small statistically significant decline in the mean % predicted DLCO was observed at 6 and 12 months. All tumors responded to treatment at 3 months and local failure was seen in only 2 single metastases. There have been no regional lymph node recurrences. At a median follow-up of 12 months, the crude survival rate is 83%, with 3 deaths due to co-morbidities and 1 secondary to metastatic disease.ConclusionRadical stereotactic radiosurgery with real-time tumor motion tracking is a promising well-tolerated treatment option for small peripheral lung tumors.

Abstract

Simple Summary: Cancer is the main cause of human death. Nanomedicine provides a new strategy for the treatment of diverse cancers. Among them, superparamagnetic artificial cell PLGA-Fe3O4 micro/nanocapsules demand great attention due to their excellent properties, such as targeted delivery, magnetic response, and biocompatibility. The aim of this review is to summarize the application of superparamagnetic artificial cells (PLGA-Fe3O4) micro/nanocapsules in cancer drug targeted delivery and their preparation methods, providing insights into the future development of superparamagnetic artificial cells (PLGA nano/microcapsules) for magnetic targeting in cancer treatment. Artificial cells have been extensively used in many fields, such as nanomedicine, biotherapy, blood substitutes, drug delivery, enzyme/gene therapy, cancer therapy, and the COVID-19 vaccine. The unique properties of superparamagnetic Fe3O4 nanoparticles have contributed to increased interest in using superparamagnetic artificial cells (PLGA-Fe3O4 micro/nanocapsules) for targeted therapy. In this review, the preparation methods of Fe3O4 NPs and superparamagnetic artificial cell PLGA-drug-Fe3O4 micro/nanocapsules are discussed. This review also focuses on the recent progress of superparamagnetic PLGA-drug-Fe3O4 micro/nanocapsules as targeted therapeutics. We shall concentrate on the use of superparamagnetic artificial cells in the form of PLGA-drug-Fe3O4 nanocapsules for magnetic hyperthermia/photothermal therapy and cancer therapies, including lung breast cancer and glioblastoma.
