Abstract

Phononic crystals and metamaterials have a unique band structure that allows for the existence of topologically protected surface states. The topologically protected edge states can guide elastic waves without significant scattering or loss of energy. One of the most promising applications of topological insulators in wave guiding is in the field of acoustics, where they can be used to design highly efficient and robust acoustic wave guides. However, the high efficiency, precision, reconfigurability, and robustness of elastic waves remains challenging. The topological insulators provide a feasible method to design high-efficiency, robust, and low-backscattering waveguides. In this work, a novel design of hexagonal metamaterial plates composed of a base plate and piezoelectric patches is proposed. The hexagonal metamaterial plate can generate robust topologically protected edge waves via active control. The paths of the topologically protected edge waves can be tuned by adjusting the control parameters. The robustness and efficiency of the proposed hexagonal metamaterial plate are testified to by the numerical examples. These findings provide systematic theoretical guidelines for designing reconfigurable wave guides, elastic wave splitters, and novel elastic wave devices and hold great promise for the development of high-performance and versatile wave guide technologies with potential applications in a wide range of fields. Abstract

The global COVID-19 pandemic emerged at the end of December 2019. Acute respiratory distress syndrome (ARDS) and acute lung injury (ALI) are common lethal outcomes of bacterial lipopolysaccharide (LPS), avian influenza virus, and SARS-CoV-2. Toll-like receptor 4 (TLR4) is a key target in the pathological pathway of ARDS and ALI. Previous studies have reported that herbal small RNAs (sRNAs) are a functional medical component. BZL-sRNA-20 (Accession number: B59471456; Family ID: F2201.Q001979.B11) is a potent inhibitor of Toll-like receptor 4 (TLR4) and pro-inflammatory cytokines. Furthermore, BZL-sRNA-20 reduces intracellular levels of cytokines induced by lipoteichoic acid (LTA) and polyinosinic-polycytidylic acid (poly (I:C)). We found that BZL-sRNA-20 rescued the viability of cells infected with avian influenza H5N1, SARS-CoV-2, and several of its variants of concern (VOCs). Acute lung injury induced by LPS and SARS-CoV-2 in mice was significantly ameliorated by the oral medical decoctosome mimic (bencaosome; sphinganine (d22:0)+BZL-sRNA-20). Our findings suggest that BZL-sRNA-20 could be a pan-anti-ARDS ALI drug.

Abstract

Background: Jieyuanshen decoction (JYAS-D) - a traditional Chinese medicine was invented by Professor Nie based on classic formulas, chaihu jia longgu muli decoction has been proved as having favorable curative effects on depression in clinical practices. The aim of this study was to investigate the antidepressant effects and its molecular mechanism of JYAS-D.Materials and Methods: The model of depression was established by Chronic Unpredictable Stress. Different doses (8.2 g/kg, 16.3 g/kg, 32.7 g/kg) of JYAS-D was orally administered; Fluoxetine was orally administered with 10mg/kg. All treatments lasted for 28 days. Sucrose preference and open-field tests were adopted to observe the behavior of rats. OPA (ortho-phthalaldehyde) derivatization method was used to detect the contents of amino acid neurotransmitter. RIA (Radiation immunity analysis) method was used to measure the serum concentrations of CORT (Corticosterone), ACTH (Adrenocorticotropic hormone) and CRH (Corticotropin-releasing hormone). ELISA (Enzyme linked immunosorbent assay) method was adopted to examine the contents of Glucocorticoid receptor (GR) and Mineralocorticoid receptor (MR) in hippocampus.Results: Compared with the model group, sucrose preference was increased in all treatment groups. The concentration of serum CORT was reduced in the middle dose of JYAS-D and control groups; the concentration of serum ACTH was reduced in the low and high-dose of JYAS-D; the concentration of serum CRH was reduced in the middle and high-dose of JYAS-D. The content of hippocampus GR was increased in the middle and high-dose of JYAS-D; the content of hippocampus Glu (Glutamic acid) was reduced among the low, middle and high-dose of JYAS-D and fluoxetine group, the ratio of Glu/γ-GABA (γ-aminobutyric acid was reduced in the low and high-dose of JYAS-D.Conclusion: JYAS-D had a significant antidepressant-like effect on rat model through regulating serum concentration of CORT, ACTH and CRH, increasing the content of hippocampus GR and regulating the equilibrium of amino acids neurotransmitter.Keywords: Hypothalamic-pituitary-adrenal (HPA) axis; Glucocorticoid/ Mineralocorticoid receptor (G/MR); Amino acid neurotransmitter; Chronic Unpredictable Stress (CUS); Jieyuanshen decoction(JYAS-D)

42. [The Relationship between TSG101 Protein and Notch3 Receptor in Lung Cancer.].

Abstract

Background: TSG101 protein is one of the key factors in Endosomal sorting pathway. The abnormality of this pathway could decrease degradation of Notch receptor which leads to disorder of cell differentiation and development in drosophila. In mammal, the Notch3 receptors correlate with lung tissue development and lung carcinogenesis. The purpose of this study is to explore the relationship between TSG101 protein and Notch3 receptor in human lung cancer., Methods: Immunohistochemical method (S-P method) and Western blot were applied to detect the expressions of TSG101 protein and Notch3 receptor in lung cancer tissues and cell lines, in comparison with the corresponding normal tissues and cell lines. Besides, the expression of Notch3 receptor was observed when TSG101 protein was blocked by its specific antibody., Results: The TSG101 protein expressions in tumor tissues were significantly lower than those in the neighboring noncancerous tissue, with higher expressions of Notch3 receptor in tumor tissues, which were correlated with the cellular differentiation and lymph node metastases. The expression of Notch3 receptor increased after blockage of TSG101 protein by its specific antibody., Conclusions: Down-regulation of TSG101 protein was correlated with the up- regulation of Notch 3 receptor in lung cancer.

43. [Expression and its significance of TSG101 in lung cancer tissue and lung cancer cell lines.].

Abstract

Background: It has been proven that TSG101 is a candidate tumor suppressor gene whose deletion in NIH3T3 cells leads to spontaneous lung metastases in nude mice. Aberrant transcripts of TSG101 have been identified in primary breast carcinomas without evidence of intragenic deletions. To investigate the possible role of TSG101 and aberrant transcripts of TSG101 in lung cancer, we performed transcript analysis and protein analysis in lung cancer cell lines and lung cancer tissue., Methods: Immunohistochemical method (S-P method) was used to detect the expression of TSG101 in 79 human squamous carcinoma and adenocarcinoma cases with the neighboring noncancerous tissue. RT-PCR was adopted to detect a common shortened TSG101 transcript because of aberrant alternative splicing and the wild-type transcript in lung cancer lines. Western Blot method was adopted to detect the expression of TSG101 protein in lung cell cells., Results: The expression of TSG101 protein in tumor tissues was significantly lower than that in the neighboring noncancerous tissue. Reverse transcriptase RT-PCR analysis detected a common shortened TSG101 transcript because of aberrant alternative splicing, which was co-expressed with the wild-type transcript in seven lung cancer lines. Although shortened transcript was detected in all of lung cell cells being involved in our experiment, there is more aberrant transcipts in small cell lung cancer (SCLC) lines .Western blot analysis have detected the same differences in protein level., Conclusions: TSG101 is a candidate tumor suppressor in non small cell lung cancer. However the common TSG101 aberrant transcript may be associated with types of lung cancer, and can be used as valuable biomarkers to evaluate lung cancer poor prognosis.
