Abstract

Abstract Urbanizing global populations spend over 90% of their time indoors where microbiome abundance and diversity are low. Chronic exposure to microbiomes with low abundance and diversity have demonstrated negative long-term impacts on human health. Sequencing-based analyses of environmental nucleic acids are critical to understanding the impact of the indoor microbiome on human health, however low DNA yields indoors, alongside sample collection and processing inconsistencies, currently challenge study replicability. This study presents a comparative assessment of a novel, passive, easily replicable sampling strategy using polydimethylsiloxane (PDMS) sheets alongside a representative swab-based collection protocol. Deployable, customizable PDMS films designed for whole-sample insertion into standardized extraction kits demonstrated 43% higher DNA yields per sample, and 76% higher yields per cm2 of sampler over swab-based protocols. These results indicate that this accessible, scalable method enables sufficient DNA collection to comprehensively evaluate indoor microbiome exposures and potential human health impacts using smaller, more space efficient samplers, representing an attractive alternative to swab-based collection. In addition, this process reduces the manual steps required for microbiome sampling which could address inter-study variability, transform the current microbiome sampling paradigm, and ultimately benefit the replicability and accessibility of microbiome exposure studies.

Abstract

Abstract Spaceflight induces an immune response in astronauts. To better characterize this effect, we generated single-cell, multi-ome, cell-free RNA (cfRNA), biochemical, and hematology data for the SpaceX Inspiration4 (I4) mission crew. We found that 18 cytokines/chemokines related to inflammation, aging, and muscle homeostasis changed after spaceflight. In I4 single-cell multi-omics data, we identified a “spaceflight signature” of gene expression characterized by enrichment in oxidative phosphorylation, UV response, immune function, and TCF21 pathways. We confirmed the presence of this signature in independent datasets, including the NASA Twins Study, the I4 skin spatial transcriptomics, and 817 NASA GeneLab mouse transcriptomes. Finally, we observed that (1) T cells showed an up-regulation of FOXP3, (2) MHC class I genes exhibited long-term suppression, and (3) infection-related immune pathways were associated with microbiome shifts. In summary, this study reveals conserved and distinct immune disruptions occurring and details a roadmap for potential countermeasures to preserve astronaut health.

Abstract

Abstract Lake Hillier is a hypersaline lake known for its distinctive bright pink color. The cause of this phenomenon in other hypersaline sites has been attributed to halophiles, Dunaliella, and Salinibacter, however, a systematic analysis of the microbial communities, their functional features, and the prevalence of pigment-producing-metabolisms has not been previously studied. Through metagenomic sequencing and culture-based approaches, our results evidence that Lake Hillier is composed of a diverse set of microorganisms including archaea, bacteria, algae, and viruses. Our data indicate that the microbiome in Lake Hillier is composed of multiple pigment-producer microbes, including Dunaliella, Salinibacter, Halobacillus, Psychroflexus, Halorubrum, many of which are cataloged as polyextremophiles. Additionally, we estimated the diversity of metabolic pathways in the lake and determined that many of these are related to pigment production. We reconstructed complete or partial genomes for 21 discrete bacteria (N = 14) and archaea (N = 7), only 2 of which could be taxonomically annotated to previously observed species. Our findings provide the first metagenomic study to decipher the source of the pink color of Australia’s Lake Hillier. The study of this pink hypersaline environment is evidence of a microbial consortium of pigment producers, a repertoire of polyextremophiles, a core microbiome and potentially novel species.

Purpose: In light of digital advancements, banks need to create customer experiences that strengthen loyalty and trust. For establishing a strong digital banking base, it is crucial for banks to make their processes efficient and fast. The purpose of the paper is to analyze the efficacy of banking functions on implementing Artificial Intelligence for enhancing customer engagement and improving customer satisfaction. It targets banks in metropolitan cities of India having tech-savvy customers, leading a fast-paced life who desire personalization and expect faultless and seamless services. Methodology: The study focusses on front, middle and back-office banking processes. The data for middle and back-office processes is collected through 10 interviews of senior officials and head of IT team in major banks. Literature review and theoretical research is carried out for various international and Indian banks with respect to the integration of AI to improve customer interactions and internal banking processes. For understanding the front-office user experience with AI-Banking, data has been gathered through a survey regarding usage of Chatbots on online banking platforms. A quantitative analysis using the Relative Importance Index reveals major use-cases ranked by customers. Spearman correlation is applied to find the relationship between the two most popular use-cases. Findings: The research paper reveals banking features integrated with AI. Chatbot use-cases on banking platforms are ranked based on customer experience. It is proved that there is a positive correlation (0.247) between the two most popular use-cases. The paper proposes IT Architecture and best practices for the digital banking sector. Practical/Theoretical implications: Based on the complete picture of AI integration with banking operations, evolving Indian banks could focus on the most popular use-cases to attract customers. A comparison with the features developed for various banks may provide a way for growth in the digital banking sector. The correlation between Chatbot use-cases may benefit the established Indian banks to further expand business. Originality/value: Implementation of AI in banking is identified for Indian Banks. It is proved that if a person uses Chatbot for assistance in customer service, they are likely to use Chatbot for recommendation regarding offers and discounts.

Abstract

Abstract Background Clean rooms of the Space Assembly Facility (SAF) at the Jet Propulsion Laboratory (JPL) at NASA are the final step of spacecraft cleaning and assembly before launching into space. Clean rooms have stringent methods of air-filtration and cleaning to minimize microbial contamination for exoplanetary research and minimize the risk of human pathogens, but they are not sterile. Clean rooms make a selective environment for microorganisms that tolerate such cleaning methods. Previous studies have attempted to characterize the microbial cargo through sequencing and culture-dependent protocols. However, there is not a standardized metagenomic workflow nor analysis pipeline for spaceflight hardware cleanroom samples to identify microbial contamination. Additionally, current identification methods fail to characterize and profile the risk of low-abundance microorganisms. Results A comprehensive metagenomic framework to characterize microorganisms relevant for planetary protection in multiple cleanroom classifications (from ISO-5 to ISO-8.5) and sample types (surface, filters, and debris collected via vacuum devices) was developed. Fifty-one metagenomic samples from SAF clean rooms were sequenced and analyzed to identify microbes that could potentially survive spaceflight based on their microbial features and whether the microbes expressed any metabolic activity or growth. Additionally, an auxiliary testing was performed to determine the repeatability of our techniques and validate our analyses. We find evidence that JPL clean rooms carry microbes with attributes that may be problematic in space missions for their documented ability to withstand extreme conditions, such as psychrophilia and ability to form biofilms, spore-forming capacity, radiation resistance, and desiccation resistance. Samples from ISO-5 standard had lower microbial diversity than those conforming to ISO-6 or higher filters but still carried a measurable microbial load. Conclusions Although the extensive cleaning processes limit the number of microbes capable of withstanding clean room condition, it is important to quantify thresholds and detect organisms that can inform ongoing Planetary Protection goals, provide a biological baseline for assembly facilities, and guide future mission planning. Abstract

The recent increase in publicly available metagenomic datasets with geospatial metadata has made it possible to determine location-specific, microbial fingerprints from around the world. Such fingerprints can be useful for comparing microbial niches for environmental research, as well as for applications within forensic science and public health. To determine the regional specificity for environmental metagenomes, we examined 4305 shotgun-sequenced samples from the MetaSUB Consortium dataset – the most extensive public collection of urban microbiomes, spanning 60 different cities, 30 countries, and 6 continents. We were able to identify city-specific microbial fingerprints using supervised machine learning (SML) on the taxonomic classifications, and we also compared the performance of ten SML classifiers. We then further evaluated the five algorithms with the highest accuracy, with the city and continental accuracy ranging from 85-89% to 90-94%, respectively. We then used these results to develop Cassandra, a random-forest-based classifier that identifies indicator species to aid in fingerprinting and can infer higher-order microbial interactions at each site. We further tested the Cassandra algorithm on the Tara Oceans dataset, the largest collection of marine-based microbial genomes, where it classified the oceanic sample locations with 83% accuracy. These results and code show the utility of SML methods and Cassandra to identify bioindicator species across both oceanic and urban environments, which can help guide ongoing efforts in biotracing, environmental monitoring, and microbial forensics (MF).

10. METABOLIC EFFECTS OF ANTIPSYCHOTIC TREATMENT IN SCHIZOPHRENIA

Abstract

BACKGROUND: It is well established that morbidity and mortality of Schizophrenia is signicantly increased due to the metabolic derangements caused by anti-psychotic medications which in turn are potent cardiovascular risk factors. Despite this widespread knowledge of metabolic risks of antipsychotic treatment, the metabolic parameters of the patients are rarely taken into consideration while prescribing antipsychotic medications in India. AIM:To determine the effects of antipsychotic treatment on the metabolic parameters in drug naïve patients of schizophrenia. METHOD: Eighty consecutive drug naive schizophrenia patients were assessed for 05 metabolic parameters which form the criteria of metabolic syndrome (NCEP- ATPIII). These parameters were again assessed after 06 months of treatment in the same set of patients. The data was then analyzed for changes in metabolic parameters in context of treatment with rst and second generation antipsychotics. RESULT: Our study found that there were signicant changes in the metabolic parameters after 06 months of treatment with anti-psychotic medications. CONCLUSION:Antipsychotic medications, both rst and second generation, cause signicant derangements in the metabolic parameters. Thus, metabolic parameters should always be taken into consideration before starting of antipsychotic treatment as well as during follow up.

11. PREVALENCE OF METABOLIC SYNDROME IN FIRST EPISODE DRUG NAÏVE SCHIZOPHRENIA PATIENTS

Abstract

BACKGROUND: It is well established that schizophrenia is associated with higher morbidity and mortality due to the nature of illness itself and cardiovascular risk factors. Among cardiovascular risk factors metabolic syndrome has been discussed extensively in psychiatric literature. However most of the studies focused on the recent pharmacological interventions are possible etiological factors of metabolic syndrome. In our study we are trying to nd out any association of metabolic syndrome with schizophrenia in drug naïve patients. METHOD: Eighty consecutive drug naïve rst episode patients with schizophrenia presenting at a tertiary care hospital in Western India were studied. They were assessed for prevalence of metabolic syndrome as per the criteria set by the NCEP (ATPIII) in Adults. RESULT: A high prevalence of metabolic syndrome in drug naïve rst episode patients with schizophrenia was found as compared to that in general population. CONCLUSION: There should be mandatory screening for the possibility of hyperlipidemia, high glucose levels and metabolic syndrome in drug naïve rst episode patients with schizophrenia before prescribing antipsychotic drugs

Abstract

Summary We present a global atlas of 4,728 metagenomic samples from mass-transit systems in 60 cities over 3 years, representing the first systematic, worldwide catalog of the urban microbial ecosystem. This atlas provides an annotated, geospatial profile of microbial strains, functional characteristics, antimicrobial resistance (AMR) markers, and genetic elements, including 10,928 viruses, 1,302 bacteria, 2 archaea, and 838,532 CRISPR arrays not found in reference databases. We identified 4,246 known species of urban microorganisms and a consistent set of 31 species found in 97% of samples that were distinct from human commensal organisms. Profiles of AMR genes varied widely in type and density across cities. Cities showed distinct microbial taxonomic signatures that were driven by climate and geographic differences. Abstract

In less than nine months, the Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) killed over a million people, including >25,000 in New York City (NYC) alone. The COVID-19 pandemic caused by SARS-CoV-2 highlights clinical needs to detect infection, track strain evolution, and identify biomarkers of disease course. To address these challenges, we designed a fast (30-minute) colorimetric test (LAMP) for SARS-CoV-2 infection from naso/oropharyngeal swabs and a large-scale shotgun metatranscriptomics platform (total-RNA-seq) for host, viral, and microbial profiling. We applied these methods to clinical specimens gathered from 669 patients in New York City during the first two months of the outbreak, yielding a broad molecular portrait of the emerging COVID-19 disease. We find significant enrichment of a NYC-distinctive clade of the virus (20C), as well as host responses in interferon, ACE, hematological, and olfaction pathways. In addition, we use 50,821 patient records to find that renin–angiotensin–aldosterone system inhibitors have a protective effect for severe COVID-19 outcomes, unlike similar drugs. Finally, spatial transcriptomic data from COVID-19 patient autopsy tissues reveal distinct ACE2 expression loci, with macrophage and neutrophil infiltration in the lungs. These findings can inform public health and may help develop and drive SARS-CoV-2 diagnostic, prevention, and treatment strategies.

Abstract

AimsThe recent pandemic of COVID-19 has not only shaken the healthcare but also economic structure around the world. In addition to these direct effects, it has also brought in some indirect difficulties owing to the information epidemic on social media. As India experienced a later outbreak of COVID-19 and a prolonged uninterrupted lockdown, we aimed to understand the nature of panic social media users in India are experiencing due to the flow of (mis)information. We further extend this investigation to other countries.MethodsWe performed a cross-sectional study by conducting survey on multiple social media platforms. We received 1075 responses (sex ratio 2:1) through opportunity sampling from social media users of 30 different countries (between April 11, 2020 and May 15, 2020). We performed both quantitative and qualitative analyses on the 935 respondents from India. Several hypotheses are statistically tested on them and are further examined on rest of the 140 social media users from 29 other countries. We also performed a separate Twitter hashtag analysis and sentiment analysis on the responses. We applied a citizen science approach to involve the respondents in the analysis pipeline after the survey.ResultsThis cross-sectional study on 1075 social media users from India and 29 other countries revealed a significant increase of social media usage and rise of panic over time in India. Middle-aged people and female exhibit a higher panic in India. The amount of panic was independent of the nature of association with COVID-19. The change of mental health was associated with panic level and productivity. Further qualitative analysis highlights the occurrences of information panic, economic panic, moral panic and spiritual panic, among other causes.ConclusionsSeveral panic behaviors are unique to social media users in India possibly because COVID-19 broke out relatively later in comparison with the other countries and the uninterrupted lockdown prolonged for a long time. The amount of social media usage might not be causal but has a significant role in generating panic among the people in India. A significantly higher level of panic among the middle-aged people can be attributed to their higher amount of responsibility. The popularity of different hashtags, including the names of drugs under trial for COVID-19, in limited countries highlight that the causes of panic are not the same everywhere. As some of the respondents took part as citizen scientists a robust perspective to the outcome is obtained.

Abstract

Although studies have shown that urban environments and mass-transit systems have distinct genetic profiles, there are no systematic worldwide studies of these dense, human microbial ecosystems. To address this gap in knowledge, we created a global metagenomic and antimicrobial resistance (AMR) atlas of urban mass transit systems from 60 cities, spanning 4,728 samples and 4,424 taxonomically-defined microorganisms collected for three years. This atlas provides an annotated, geospatial profile of microbial strains, functional characteristics, antimicrobial resistance markers, and novel genetic elements, including 10,928 novel predicted viral species, 1302 novel bacteria, and 2 novel archaea. Urban microbiomes often resemble human commensal microbiomes from the skin and airways, but also contain a consistent “core” of 31 species which are predominantly not human commensal species. Samples show distinct microbial signatures which may be used to accurately predict properties of their city of origin including population, proximity to the coast, and taxonomic profile. These data also show that AMR density across cities varies by several orders of magnitude, including many AMRs present on plasmids with cosmopolitan distributions. Together, these results constitute a high-resolution, global metagenomic atlas, which enables the discovery of new genetic components of the built human environment, highlights potential forensic applications, and provides an essential first draft of the global AMR burden of the world’s cities.

The present study was designed to directly compare two methods that elicit potentially different kinds of hindsight bias and to relate the magnitude of the bias to degree of handedness, a neurologically-related individual difference variable associated with differences in both memory performance and anchoring. 160 introductory psychology students were given 64 general knowledge questions, half under “memory instructions” and the other half under “hypothetical instructions”. Results indicated that individuals showed greater hindsight bias under hypothetical than memory instructions, replicating previous research. They also showed that consistent right-handers are relatively more affected under memory instructions, while inconsistent-handers are relatively more affected under hypothetical instructions. In sum, the current study demonstrates that two kinds of hindsight bias exist, and the kind that emerges depends on both the person and situational context. The findings also suggest that different mechanisms may be involved, these mechanisms may have neurological underpinnings, and a multicomponent model of hindsight bias may be best for future theorizing.

Abstract

The Microbe Directory (TMD) is a comprehensive database of annotations for microbial species collating features such as gram-stain, capsid-symmetry, resistance to antibiotics and more. This work presents a significant improvement to the original Microbe Directory (2018). This update adds 68,852 taxa, many new annotation features, an interface for the statistical analysis of microbiomes based on TMD features, and presents a portal for the broad community to add or correct entries. This update also adds curated lists of gene annotations which are useful for characterizing microbial genomes. Much of the new data in TMD is sourced from a set of databases and independent studies collating these data into a single quality controlled and curated source. This will allow researchers and clinicians to have easier access to microbial data and provide for the possibility of serendipitous discovery of otherwise unexpected trends.

Abstract

The goal of the current study was to explore information search and processing differences between individuals who are less and more numerate in an attempt to better understand the mechanisms that might differentiate the choices they make. We did so using a computerized process-tracing system known as MouseTrace, which presented monetary gambles in an alternative × attribute matrix with outcome (dollar amount) and probability information as attributes. This information was initially occluded but could be revealed by clicking on the cell that contained the desired information. Beginning with nine gambles offering the chance of gaining or losing a specified amount, participants (N = 110) narrowed down the options (to three and then one) using an inclusion or exclusion strategy. Consistent with previous research, inclusion was a more effortful strategy, and individuals who were higher in numeracy were more likely to select prospects with the highest expected value. Process measures revealed these individuals expended more effort (i.e., attended to and sought out more information and processed it in greater depth) and exhibited more compensatory processing than those who were lower in numeracy, but this sometimes depended on whether one was asked to include or exclude. These results serve as further evidence that individuals with higher levels of numeracy often engage in more elaborative processing of the decision task, which tends to lead to more optimal choices. Abstract

In the present study, the applicability of air atomized spray with the salt added water has been studied for ultra fast cooling (UFC) of a 6 mm thick AISI-304 hot steel plate. The investigation includes the effect of salt (NaCl and MgSO4) concentration and spray mass flux on the cooling rate. The initial temperature of the steel plate before the commencement of cooling is kept at 900 °C or above, which is usually observed as the “finish rolling temperature” in the hot strip mill of a steel plant. The heat transfer analysis shows that air atomized spray with the MgSO4 salt produces 1.5 times higher cooling rate than atomized spray with the pure water, whereas air atomized spray with NaCl produces only 1.2 times higher cooling rate. In transition boiling regime, the salt deposition occurs which causes enhancement in heat transfer rate by conduction. Moreover, surface tension is the governing parameter behind the vapour film instability and this length scale increases with increase in surface tension of coolant. Overall, the achieved cooling rates produced by both types of salt added air atomized spray are found to be in the UFC regime.

Abstract

Numeracy or one's ability to appropriately process and use numerical information has been shown to be an important individual difference factor in decision making. The current study utilized a risky decision-making task (called the “cups task”) in which choices are made to both earn and avoid losing hypothetical money. Critically, this design allowed investigators to examine numeracy-related differences in adaptive decision-making as measured by sensitivity to expected value (EV) differences over 54 paired-choice trials—some in which it was advantageous to take a risk and some in which it was not. Results showed that in an undergraduate sample of 114 individuals, the less numerate took more risks and were less sensitive to varying EV levels than the more numerate, especially when it was disadvantageous to take a risk and when the choice involved a potential loss (rather than a gain). These results are consistent with a dual processing account in which the more numerate are much better than the less numerate at extracting the precise affective “gist” of the numerical information, which is then used to determine the goodness or badness of a particular choice. Successful performance of a vigilance task in a particular modality can be affected by the allocation of attentional resources to other cognitive processes using same or different modalities. The present study investigated whether loss of hearing affects visual vigilance task performance and also if it’s a function of increased task complexity. A sample of 30 adolescents ( 15 with hearing loss and 15 with normal hearing) were selected by matching them in pairs according to their working memory functioning and other relevant organismic variables. A software program measured the performance on vigilance task with varying task complexity levels. Analysis of the obtained data indicates that the effect of hearing impairment on visual vigilance is moderated by task complexity and that is contingent to attentional resources. Here, deprivation in hearing ability negatively affects functioning of visual vigilance significantly only when the task complexity increases. But no significant differences are observed in hearing and hearing impaired adolescents when the task demanded less attentional resources.

Abstract

Studies on decision making have come to challenge the idea that having more choice is necessarily better. The Medicare prescription drug program (Part D) has been designed to maximize choice for the consumer but has simultaneously created a highly complex decision task with dozens of options. In this study, in a sample of 121 adults, we examined the impact that increasing choice options has on decision-making abilities in older versus younger adults. Consistent with our hypotheses, we found that participants performed better with less choice versus more choice, and that older adults performed worse than younger adults across conditions. We further examined the role that numeracy may play in making these decisions and the role of more traditional cognitive variables such as working memory, executive functioning, intelligence, and education. Finally, we examined how personality style may interact with cognitive variables and age in decision making. Regression analysis revealed that numeracy is related to performance across the lifespan. When controlling for additional measures of cognitive ability, we found that although age was no longer associated with performance, numeracy remained significant. In terms of decision style, personality characteristics were not related to performance. Our results add to the mounting evidence for the critical role of numeracy in decision making across decision domains and across the lifespan.

Abstract

Objective: Natural goitrogens present in plant foods or antithyroidal drugs both alter the morphology and normal functional status of the thyroid gland interfering the synthesis and secretion of thyroid hormones. In this study, the goitrogenic potential of bamboo shoots (BS) containing natural goitrogens has been compared against a potent synthetic antithyroidal drug thiourea.Methods: BS was fed to 6 rats by 1/3rdreplacement of 180grams (g) of their food i.e.60g of BS containing 35g of goitrogens of cyanogenic origin such that each rat ideally consumed 6 mg/100g of body weight per day and thiourea at a near equal dose of 6 mg/100g of body weight per day was force fed to another 6 rats for a period of 45 d. Changes in food consumption pattern, serum toxicity marker SGOT and SGPT, morphological changes like thyroid gland weight and histology and functional aspects like TPO activity, Na+-K+ATPase activity, urinary iodine excretion and serum T3,T4 and TSH levels induced by this natural and synthetic agent respectively were investigated. Results analyzed by ANOVA followed by post hoc tests.Results: Food consumption pattern was similar in all groups while serum toxicity markers SGOT, SGPT showed a non-significant increase in experimental groups. Increase in body and thyroid gland weight as compared to control was seen on treatment with both antithyroidal agents, a greater percentage of increase was seen in thiourea treated group. TPO activity, Na+-K+ATPase activity, serum T3 and T4levels were decreased significantly in both treated groups as compared to normal, with maximum inhibition in the thiourea treated group. Histological plates of BS treated thyroid revealed hypertrophy and thyroid follicular cell disruption with microcytic infiltration. Thiourea treated gland histological plate revealed complete abolition of normal histoarchitecture with marked changes in colloidal area and hyperplasia with folding and papillae on the epithelium.Conclusion: In overall, the data indicate that though the goitrogenic potentiality of BS cannot be considered having similar efficacy to that of thiourea but as a food entity by itself, it has potent antithyroidal activity even in presence of adequate iodine intake.

Abstract

Although disinfection is key to infection control, the colonization patterns and resistomes of hospital-environment microbes remain underexplored. We report the first extensive genomic characterization of microbiomes, pathogens and antibiotic resistance cassettes in a tertiary-care hospital, from repeated sampling (up to 1.5 years apart) of 179 sites associated with 45 beds. Deep shotgun metagenomics unveiled distinct ecological niches of microbes and antibiotic resistance genes characterized by biofilm-forming and human-microbiome-influenced environments with corresponding patterns of spatiotemporal divergence. Quasi-metagenomics with nanopore sequencing provided thousands of high-contiguity genomes, phage and plasmid sequences (>60% novel), enabling characterization of resistome and mobilome diversity and dynamic architectures in hospital environments. Phylogenetics identified multidrug-resistant strains as being widely distributed and stably colonizing across sites. Comparisons with clinical isolates indicated that such microbes can persist in hospitals for extended periods (>8 years), to opportunistically infect patients. These findings highlight the importance of characterizing antibiotic resistance reservoirs in hospitals and establish the feasibility of systematic surveys to target resources for preventing infections., Spatiotemporal characterization of microbial diversity and antibiotic resistance in a tertiary-care hospital reveals broad distribution and persistence of antibiotic-resistant organisms that could cause opportunistic infections in a healthcare setting.
