The medium used for bottle cultivation of mushrooms is made according to a specific workflow; this includes mixing the raw materials, adjusting the moisture content, placing the mixture into bottles, and sterilizing and cooling the medium. Medium composition should be kept constant throughout the year. placed in the bottle, choice of materials, moisture content, weight of the medium, bulk density, nitrogen content, three-phase medium, such as suitable conditions. To achieve constant medium composition a comprehensive model has been established in accordance with the technology developed in the work process with respect to medium compositions piecemeal. By comprehensively applying the various methods to establish medium composition, our country can expect to grow at over four seasons and overcome the many difficulties in mushroom mycelial growth periods, cultivation periods, quantity, and quality.

Two phosphate solubilizing bacteria, Pantoea rodasii PSB-11and Enterobacter aerogenes PSB-12, were isolated from button mushroom compost and employed to assess their synergistic effect in liquid medium and on growth of green gram plants by single and co-inoculation of the strains. Co-inoculation of two strains was found to release the highest content of soluble phosphorus (521 μg ml -1 ) into the medium, followed by single inoculation of Pantoea strain (485 μg ml -1 ) and Enterobacter strain (470 μg ml -1 ). However, there was no significant difference between single inoculation of bacterial strain and co-inoculation of two bacterial strains in terms of phosphorous release. The highest pH reduction, organic acid production and glucose consumption was observed in the E. aerogenes PSB-12 single inoculated culture medium rather than those of co-inoculation. According to the plant growth promotion bioassay, co-inoculated mung bean seedlings recorded 10.6% and 10.7% higher shoot and root growth respectively compared to the control. Therefore, in concluding, coinoculation of the strains P. rodasii and E. aerogenes displayed better performance in stimulating plant growth than inoculation of each strain alone. However, being short assessment period of the present study, we recommend in engaging further works under field conditions in order to test the suitability of the strains to be used as bio-inoculants.

This study analyzed the physicochemical characteristics of Astragalus membranaceus (AM) fermented with seven different mushroom mycelia. Physicochemical characteristics, such as contents of moisture, pH, total reducing sugars, free sugar, and isoflavonoid, were investigated. The moisture content was increased in most of the samples. The pH values of AM fermented with Phellinus linteus and Flammulina velutipes were increased, while the pH of other samples were similar to that of non-fermented AM. The reducing sugar content was in the range of 211.69~391.74 mg/100 g. The extraction yield using water was higher than that when extracted with 80% ethanol. The free sugar content was increased through fermentation with mushroom mycelia. However, the glucose contents of the 80% ethanol and water extracts were decreased. Finally, the calycosin and formononetin contents in 80% ethanol and water extracts of AM fermented with Phellinus linteus were 2,549.24 mg/g, and 827.66 mg/g for calycosin, and 1,366.69 mg/g and 221.28 mg/g for formononetin, respectively. These results suggest that fermentation with mushroom mycelia could be used to increase the bioactivity of AM. The mycelium-fermented AM might be a valuable source of functional material and edible resource for industry.

In order to isolate compost-promoting bacteria with high activity of cellulase and xylanase, spent mushroom substrates with sawdust were collected from mushroom cultivation farm, Jinju, Gyeongnam in Korea. Among of the isolates, one strain, designated CA105 was selected by agar diffusion method. The strain CA105 was identified as members of the Bacillus subtilis by biochemical characteristics using VITEK 2 system. Comparative 16S rRNA gene sequence analysis showed that isolate CA105 formed a distinct phylogenetic tree within the genus Bacillus and was most closely related to Bacillus subtilis with 16S rRNA gene sequence similarity of 98.9%. On the basis of its physiological properties, biochemical characteristics and phylogenetic distinctiveness, isolate CA105 was classified within the genus Bacillus subtilis, for which the name Bacillus subtilis CA105 is proposed. The cellulase and xylanase activity of B. subtilis CA105 was slightly increased according to bacterial population from exponential phase to stationary phase in growth curve for Bacillus sp. CA105.

Button mushroom compost (BMC) was prepared by fermenting the mixture of waste button mushroom bed collected from Boryeong area in South Korea (4): sawdust (8) : pig and fowl manure (1) for 40 days at 30°C. The BMC compromised diverse microorganisms including aerobic bacteria 8.1 × 106 cfu g-1, Gram negative bacteria 1.7 × 10 7 cfu g -1 , genus Bacillus 6.4 × 10 6 cfu g -1 , genus Pseudomonas 1.5 × 10 4 cfu g -1 , actinomycetes 1.0 × 10 4 cfu g -1 , and fungi 3.5 × 10 3 cfu g -1 . BMC was used as a microbial inoculant for estimating the mobilization of heavy metals in soil or plant. When metal solubilization potential of BMC was assessed in a batch experiment, the inoculation of BMC was shown to increase the concentrations of water soluble Co, Pb, Cd, and Zn by 29, 26, 27, and 43% respectively, than those of non-inoculated soils. BMC-assisted growth promotion and metal uptake in sunflower (Helianthus annuus) was also evaluated in a pot experiment. In comparison with non-inoculated seedlings, the inoculation led to increase the growth of H. annuus by 17, 15, 18, and 21% respectively in Co, Pb, Cd, and Zn contaminated soils. Moreover, enhanced accumulation of Co, Pb, Cd, and Zn in the shoot and root systems was observed in inoculated plants, where metal translocation from root to the above-ground tissues was also found to be enhanced by the BMC. The apparent results suggested that the BMC could effectively be employed in enhancing phytoextraction from the soils contaminated with heavy metals such as Co, Pb, Cd, and Zn.

This study was carried out to investigated optimum additive ratio of barley flour for substitution of rice bran at cultivation of winter mushrooms. Mycerial growth was faster according to increase of barley flour ratio than those of controls, but only some slow at the addition of 10% barley flour. Yields of fruiting body show the highest to 165.4 g/850 ml of medium which are the addition 30% of barley flour and not significant difference of yields up to the addition of 70% barley flour. Diameter of pileus was the highest at the addition of 30% barley flour. Hardness of pileus and stipes were the highest at the addition of 10% barley flour. The L value of stipes were the hight at the addition of barley flour, but the L value of pileus were decreased at the addition of barley flour, but there was no significant difference in the a-value and the b-value.

This report is the result of devising a method for utilizing the device of the load cell to maintain a constant water content of the medium every day to prepare a cultural substrates with the mixer for growing mushrooms bottle cultivation. A load cell was device under the medium mixer. It is developed when the device reaches the weight calculated as amount of substrate bottled and number of the bottle, it is automatically terminated by water injection. In addition, measuring the water content of each medium and the total weight of the medium reaches the target moisture content were calculated by using the program Cheong et al. (2015). Enter the total weight of the medium on the display unit of the load cell, when starting the water supply to reach the weight-based mixing media, the water supply is stopped. This method can improve the convenience by reducing the user's trouble in repeated work medium prepared by automating water supply. The suitable moisture content of the mixed medium for some kind of mushroom can be improved by the composition accuracy. And mycelial culture period, primordial period, mushroom growing period is maintained even of the medium can be produced stably. Therefore, it is possible to achieve a stable management of the mushroom farm according to mushroom quality and quantity stable throughout the year.

A lipase producing bacterium was isolated from button mushroom bed, which showing high clear zone on agar media containing Tributyrin as the substrate. The strain was identified as Burkholderia cepacia by analysis of 16S rDNA gene sequence. Crude lipase (CL) was partially purified from 70% ammonium sulfate precipitation using the culture filtrate of B. cepacia. Immobilized lipases were prepared by cross-linking method with CL from B. cepacia and Novozyme lipase (NL) onto silanized Silica-gel as support. Residual activitiy of the immobilized CL (ICL) and immobilized NL (INL) was maintained upto 61% and 72%, respectively. Biodiesel (Fatty acid methyl ester, FAME) was recovered by transesterification and methanolysis of Canola oil using NaOH, CL and ICL as the catalysts to compare the composition of fatty acids and the yield of FAME. Total FAME content was NaOH 781 mg L-1, CL 681 mg L-1 and ICL 596 mg L-1, in which the highest levels of FAME was observed to 50% oleic acid (C18:1) and 22% stearic acid (C18:0). In addition, the unsaturated FAME (C18:1, C18:2) decreased, while saturated FAME (C16:0, C18:0) increased according to increasing the reaction times with both CL and ICL, supporting CL possess both transesterification and interesterification activity. When reusability of ICL and INL was estimated by using the continuous reaction of 4 cycles, the activity of ICL and INL was respectively maintained 66% and 79% until the fourth reaction.

The potential ability of Button mushroom compost (BMC) to solubilize heavy metals was estimated with metal contaminated soils collected from abandoned mines of Boryeong area in South Korea. The bacterial strains in BMC were isolated for investigating the mobilization of metals in soil or plant by the strains and identified according to 16S rRNA gene sequence analysis. When metal solubilization potential of BMC was assessed in a batch experiment, the BMC was found to be capable of solubilizing metals in the presence of metals (Co, Pb and Zn) and the results showed that inoculation of BMC could increase the concentrations of water soluble Co, Pb and Cd by 35, 25 and 45% respectively, than those of non-inoculated soils. BMC-assisted growth promotion and metal uptake in sunflower (Helianthus annuus) was also evaluated in a pot experiment. In comparison with non-inoculated seedlings, the inoculation led to increase the growth of H. annuus by 27, 25 and 28% respectively in Co, Pb and Zn contaminated soils. Moreover, enhanced accumulation of Co, Pb and Zn in the shoot and root systems was observed in inoculated plants, where metal translocation from root to the above-ground tissues was also found to be enhanced by the BMC. The apparent results suggested that the BMC could effectively be employed in enhancing phytoextraction of Co, Pb and Zn from contaminated soils.

Mycerial growth was some fast at addition of R. verniciflua sawdust, and not significant difference by increase of R. verniciflua substrate. Yields of fruiting body show the highest to 150.2 g/850 ml of medium which are addition 20% of R. verniciflua sawdust, and diameter of pileus and thick of stipes were the highest at addition 10% of R. verniciflua sawdust. The L value of stipes were the highest at addition 10% of R. verniciflua sawdust and the L value of pileus were decreased by increase of R. verniciflua substrate, but there was no significant difference in the a-value and the b-value.

BACKGROUND: Excessive metals in the soil have become one of the most significant environmental problems. Phytoremediation has received considerable attention as a method for restoring the contaminated soils. The microbes having remarkable metal tolerance and plant growth-promoting abilities could also play a significant role in remediation of metal-contaminated soils, because bioaugmentation with such microbes could promote phytoextraction of metals. Therefore, the present study was focused on evaluating the phytoextraction of heavy metals (Co, Pb and Zn) in Helianthus annuus (sunflower) induced by bioaugmentation of a phosphate solubilizing bacterium. METHODS AND RESULTS: A phosphate solubilizing bacterium was isolated from metal-contaminated soils based on the greater halo size (>3 mm) with solid NBRIP agar medium containing 10 g glucose, 5 g Ca3(PO4)2, 5 g MgCl2⋅6H2O, 0.25 g MgSO4.7H2O, 0.2 g KCl, 0.1 g (NH4)2SO4 in 1 L distilled water. Isolated bacterial strain was assessed for their resistance to heavy metals; CoCl2.6H2O, 2PbCO3.Pb(OH)2, and ZnCl2 at various concentrations ranging from 100-400 µg/mL (Co, Pb and Zn) using the agar dilution method. A pot experiment was conducted with aqueous solutions of different heavy metals (Co, Pb and Zn) to assess the effect of bacterial strain on growth and metal uptake by Helianthus annuus (sunflower). The impact of bacterial inoculation on the mobility of metals in soil was investigated under laboratory conditions with 50 mL scaled polypropylene centrifuge tubes. The metal contents in the filtrate of plant extracts were determined using an atomic absorption spectrophotometer (Perkinelmer, Aanalyst 800, USA). CONCLUSION: Inoculation with Enterobacter ludwigii PSB 28 resulted in increased shoot and root biomass and enhanced accumulation of Co, Pb and Zn in Helianthus annuus plants. The strain was found to be capable of promoting metal translocation from the roots to the shoots of H. annuus. Therefore, Enterobacter ludwigii PSB 28 could be identified as an effective promoter of phytoextraction of Co, Pb and Zn from metal-contaminated soils.

A gram-negative bacterium was isolated from spent substrate of Agaricus bisporus and showed marked antagonistic activity against Pseudomonas tolaasii. The bacterium was identified as Pseudomonas azotoformans by based on the cultural, biochemical and physiological characteristics, and 16S rRNA gene sequence. The isolated bacterium was saprophytic but not parasitic nor pathogenic to cultivation mushroom. The isolated bacterium for P. tolaasii cell was not sufficient for inhibition in vitro. Control efficacy of Pseudomonas azotoformans HC5 to brown blotch of P. tolaasii was 73, 78, and 71% on A. bisporus, Flammulina velutipes, and Pleurotus ostreatus, respectively. In the future, the suppressive bacterium may be useful for development of a biocontrol system.

A bacterial strain with the potential ability to solubilize heavy metals was isolated from heavy metal contaminated soils collected from abandoned mines of Boryeong area in South Korea. The bacterial strain with the highest degree of metal resistance was shown to have close proximity with Shewanella xiamenensis FJ589031, according to 16S rRNA sequence analysis, and selected for investigating the mobilization of metals in soil or plant by the strain. The strain was found to be capable of solubilizing metals both in the absence and in the presence of metals (Co, Pb and Cd). Metal mobilization potential of the strain was assessed in a batch experiment and the results showed that inoculation could increase the concentrations of water soluble Co, Pb and Cd by 48, 34 and 20% respectively, compared with those of non-inoculated soils. Bacterial-assisted growth promotion and metal uptake in sunflower (Helianthus annuus) was evaluated in a pot experiment. In comparison with non-inoculated seedlings, the inoculation led to increase the growth of H. annuus by 24, 18 and 16% respectively in Co, Pb and Cd contaminated soils. Moreover, enhanced accumulation of Co, Pb and Cd in the shoot and root systems was observed in inoculated plants, where metal translocation from root to the above-ground tissues was also found to be enhanced by the strain. Plant growth promotion and metal mobilizing potential of the strain suggest that the strain could effectively be employed in enhancing phytoextraction of Co, Pb and Cd from contaminated soils.

본 시험은 팽이버섯 병재배용 배지의 혼합수준에 따른 전탄소(T-C), 전질소(T-N) 함량과 균배양기간, 버섯발생기간, 자실체 수량 등 재배적 특성을 조사하였다. 배지재료의 혼합비율을 달리하였을 때 팽이버섯의 경우 [콘코브75+미강20+건비지5] 배지에서 자실체 수량이 190.5 g/850 ml로 가장 높았으나(20.4% 증가), 균 배양기간이 28일로 대조구보다 8일 지연되었다(대조구 [미송톱밥75+미강25]: 수량 158.2g, 배양기간 20일). [미송톱밥75+미강20+건비지5] 배지는 팽이버섯의 자실체 수량이 172.7 g(9.1% 증가), 균 배양기간이 21일로 대조구보다 1일 지연되었다. 【The experiments were conducted to provide information on the chemical concentrations and cultural characteristics in the periods of hyphal incubation, primordial formation, and fruiting bodies yield of winter mushroom, Flammulina velutipes at the mixture ratio of the raw materials. Substrates were analyzed for pH, total carbon (T-C), total nitrogen (T-N), and C/N ratio. In case of Flammulina velutipes, yield of fruiting bodies were 190.5 g/850 ml in the substrates, [Corn-cob + Rice bran + Soybean curd residue (75:20:5)], which was increased 20.4% more than the control, [Douglas fir sawdust + Rice bran (75:25)]. But the periods of hyphal incubation took 28 days, which was 7 days longer than the control. Also, in the substrates [Douglas fir sawdust + Rice bran + Soybean curd residue (75:20:5)], the yields was 172.7 g/850 ml and the periods of hyphal incubation was 21 days.】

균상느타리버섯에서 발생하는 곰팡이 병해 중에서 가장 많은 문제를 발생하는 것은 Trichoderma disease 또는 green mould 등으로 불리우는 푸른곰팡이병이다. 이 병은 Trichoderma, Penicillium, Aspergillus 등의 속(genus)에 포함되는 종들에 의해 발병되는 병을 총칭하는 것으로 발생빈도와 피해가 가장 큰 것은 Trichoderma 속의 병원균에 의해 발생하였다. 느타리버섯 균상에 분리된 병원균은 T. longibrachiatum, T. koningii, Trichoderma virens, T atroviride, T. hazianum, T. pseudokoningii 6종이었으며, 발생빈도가 높은 것은 T. virens, T. hazianum, T. longibrachiatum 3종이었고, 각각의 균은 배양적 특성 및 병원성도 각기 다른 특성을 보이고 있었다. T. longibrachiatum 균사생장이 매우 빠르고, 포자형성이 빠르며, 특히 후막포자형성이 잘되는 특성을 있으며, 후기에는 진한 녹색으로 변색된다. Trichoderma koningii는 균사생장은 빠르고, 공중균사가 많은 편이며, 포자는 공중균사에 밀집되어 형성된다. 특히 Trichoderma virens 같은 경우에는 포자형성에 의한 색깔변화가 느리며, 감염후기에 발견되어 그 피해가 심한 편이었다. Trichoderma atroviride. 균사생장은 보통이며, 포자형성은 공중균사에 드물게 형성되며, 후기에는 진녹색을 띤다. 이균의 특징은 coconut 냄새와 유사한 향기가 나는 것이다. T. hazianum은 초기에는 균사가 백색으로 매우 빠르고, 공중균사에 포자가 형성하며, 후기에는 진한 녹색으로 변색된다. 공중 균사에 형성된 포자는 육안적으로 pustules or tufts의 형태를 나타내었다. Trichoderma pseudokoningii 균사생장은 빠르며, 포자형성은 접종부위를 중심으로 형성되며, 후기에는 환무니를 형성하며, 특히 배지 색이 변색되고 후막포자 잘 형성되는 특징이 있었다. 【Green mold disease caused by Trichoderma species has recently caused considerable damage to oyster mushroom industries in Korea. This disease Trichoderma, Penicillium, Aspergillus, such as in (genus) to be included in a disease caused by a species that collectively the largest incidence and damage is caused by the pathogen Trichoderma genus. T. longibrachiatum, Trichoderma koningii, Trichoderma virens, T. hazianum, T. atroviride, and T. pseudokoningii were detected on oyster mushroom beds and, of them, T. virens, T. hazianum, T. longibrachiatum was the most frequently detected. The knowledge concerning physiological and ecological properties of Trichoderma spp. was essential for their effective control. T. longibrachiatum hyphal growth is very fast, spore formation, and, particularly well-chlamydospore formation characteristics, and reviews are dark green discoloration. T. koningii, fast mycelial growth, aerial hyphae and spores in aerial hyphae formation is concentrated. T. virens, especially if the color change caused by spore-forming, slow, late in infection, the more severe the damage is discovered. T. hazianum fast mycelial growth, white aerial hyphae and late turns dark green. After spore formation hyphae glob of white pustules or tufts on the top of the formation. T. atroviride. aerial hyphae usually the mycelial growth and spore formation in the unlikely event of the formation and smells similar to the smell of coconut is that. Fast T. pseudokoningii mycelial growth, spore formation is formed around the inoculation site, discoloration of the medium color and well formed chlamydospores.】

This study was carried out to provide standardization for mushroom production of high quality at farmhouse. The standardization does much to improve merchantable quality, distribution efficiency and fair dealings by shipping of the standard agricultural products. Therefore, modification of these standards is required to fit farmhouse situations. The standardization for production of Pleurotus eryngii of high quality follows like this. A grades were above 90 g at average weight, more 50 mm at stipe thick, 1.6~1.7 at stipe length/stipe thick rate and 1.1~1.2 at pileus diameter/stipe thick ratio. B grades were more 45 g at average weight, above 40 mm at stipe thick, 1.8~1.9 at stipe length/stipe thick ratio and 1.2~1.4 at pileus diameter/stipe thick ratio.

Several bacteria are known as the causal agents of diseases of the cultivated button mushroom(Agaricus bisporus) and oyster mushroom(Pleurotus ostreatus). Pseudomonas tolaasii is the causal agent of brown blotch disease of commercial mushrooms. Pseudomonas sp. HC1 is a potent biological control agent to control brown blotch disease caused by Pseudomonas tolaasii. This can markedly reduce the level of extracellular toxins (i.e., tolaasins) produced by Pseudomonas tolaasii, the most destructive pathogen of cultivated mushrooms. To define the optimum conditions for the mass production of the Pseudomonas sp. HC1, we have investigated optimum culture conditions and effects of various nutrient source on the bacterial growth. The optimum initial pH and temperature were determined as pH 5.0 and , respectively. The optimal culture medium for the growth of tolaasin inhibitor bacterium was determined as follows: 0.9% dextrin, 1.5% yest extract, 0.5% , 4mM , and 3.0% cysteine.

This study was carried out to investigated optimum mixing ratio of Korean natural Juglans mandshurica using as functional plants. Total nitrogen and carbon source of J. mandshurica was 0.21% and 46.0%, respectively and C/N ratio was 219. Total nitrogen source and pH of substrate mixed with J. mandshurica was 2.3~2.9 and 5.0, respectively. The contents of CaO, MgO and at J. mandshurica media were higher at harvest media than inoculation media. Mycelial growth was the fastest at J. mandshurica 20%, and slower by increase of J. mandshurica substrate. Yields of fruiting body show the highest to 163 g/850 mL of medium which are addition 10% of J. mandshurica sawdust, and diameter and thick of pileus were the highest, too. The L value of pileus and stipes were increased by increase of J. mandshurica substrate, but there was no significant difference in the a-value and the b-value. The contents of and of fruiting body were increased at J. mandshurica substrate, but there was no significant difference in contents of CaO, MgO and . The contents of Cu of fruiting body was decreased by increase of J. mandshurica substrate, but Fe, Mn and Zn were increased.

This study was carried out to improve standardization of agricultural products and investigate quality changes during storage at different treatments. The standardization does much to improve merchantable quality, distribution efficiency and fair dealings by shipping of the standard agricultural products. Therefore, modification of these standards is required to fit farmhouse situations. Average pileus diameter and thickness of A grades was 38.9 mm and 4.5 mm at shelf culture and 22.2 mm and 3.9 mm at bottle culture. Average stipe length of A grades was 49.6 mm at shelf culture and 66.7 mm at bottle culture, and stipes thick was the highest in A grades of shelf culture. The diameter of pileus of Pleurotus ostreatus stored for 14 days at and showed no significant difference. but the thickness of pileus decreased rapidly after 7 days. L-value and hardness of stipes and pileus were higher at shelf culture than bottle culture.

 The major species of fungus gnats which caused the severe damage in shiitake farm were identified as a Bradysia difformis, B. alpicola, and Camtomyia cortocalis on oak log beds cultivation. The B. difformis occurred early in the middle of March while B. alpicola and C. cortocalis appeared since the beginning of May. The occurrence rate for adults of B. difformis showed highly at the end of July (11.9~1,774.2 in dong-myeon and 0.4~2,583.3 in pungse-myeon) in 2012 and mid-June (10.7~4,650 in dong-myeon and 36.8~4740 in pungse-myeon) in 2013. The counting numbers on the traps for B. alpicola reached highest peak in the middle of June (2.1~63.2 in dong-myeon and 1.0~21.7 in pungse-myeon) and the end of May (0.8~163.7 in dong-myeon and 0.5~280.5 in pungse-myeon) in 2012 and 2013, respectively. The number of C. cortocalis showed high record in the middle of May in 2012 (0.6~4.7) and in the middle of June (2.1~17.3) in 2013 in dong-myeon whereas showed the peaks in the middle of May (0.6~4.7) in 2012 and in the late of May (1.3~17.6) in 2013 in pungse-myeon. The fruiting bodies of shiitake mushroom by fungus gnats were severely damaged from mid-June to late-July and the damage rate were 0.625.5% (2012) and 0.7~30.5% (2013) in dong-myeon and 1.5%~21.6% (2012) and 1.9~36.8%(2012) in pungse-myeon. To investigate the control effect for fungus gnats by Hypoaspis aculeifer, H. aculeifer (30 mixutre of nymph and adult per m) were treated to oak log beds shiitake cultivation for six times (May 2 and 28, June 25, July 10 and 25 and August 28). The occurrence rate of adults and damage rate of fruiting bodies of 3 major species reduced 79.3% (adult numbers) and 74.8% (fruiting bodies) in dong-myeon and 64.1% (adult numbers) and 65.5% (fruiting bodies) in pungse-myeon, respectively, compared to non-treatment. Accordingly, H. aculeifer effectively controlled the fungus gnats on shiitake mushroom and can be used as good control agent.

The project spanned from 2009 to 2012, including 19 kinds of oyster varieties, 34 were advertised in the national 387 places. As a result, the proliferation of demonstration farms spread around the 30% (2009) share of domestic varieties 45% (2012) raised to the level. Early varieties of these domestic supply system more efficient, the central and local governments in order to promote dissemination of the technology of that business was expanded to promote.

This study was carried out to improve standardization of agricultural products and investigate quality changes during preservation at various treatments. The standardization does much to improve merchantable quality, distribution efficiency and fair dealings by shipping of the standard agricultural products. Mushrooms notified as the standard are five; Pleurotus ostreatus, Pleurotus eryngii, Flammulina velutipes, Agaricus bisporus and Ganoderma lucidum. But many farmers are suffering from strict standards. Therefore, modification of these standards is required to fit farmhouse situations. Fruit-body weight of A, B and C grades of the marketed Agaricus bisporus was 19.3g, 12.2g and 10.4, respectively. Average pileus diameter of A grades was 43.3mm but B and C grades did not show significant difference. Average stipe length of A, B and C grades was 9.8mm, 13.1mm and 11.6mm, and stipes thick was the highest in A grades. The weight loss rate of stipe cutting of Agaricus bisporus was 12.6% in harvested mushrooms and the rate of stipe cutting length was 60.8%. The diameter of Agaricus bisporus stored for 5 days was 3.5~4.5cm but decreased rapidly in 15 days. The weight loss rate of the high temperature samples lowered rapidly than that of the low temperature samples. Conversely, the L value of the low temperature samples was higher than that of the high temperature samples.

This study was carried out to investigated optimum mixing ratio of Korean natural Acer tegmentosum for production of functional oyster mushroom. Total nitrogen and carbon sources of Acer tegmentosum were 0.2% and 44.4%, respectively and C/N ratio was 234. Total nitrogen source and pH of substrate mixed with Acer tegmentosum were 2.7~2.9 and 4.8~5.0, respectively. The contents of , and MgO at Acer tegmentosum media were higher than that of the control. Mycerial growth was the fastest at Acer tegmentosum 10%, and slower by increase of Acer tegmentosum substrate. Yields of fruiting body was the highest to 159 g/850 mL at 10% of Acer tegmentosum and dimeter and thick of pileus were the highest, too. The L value of pileus and stipes were decreased by increase of Acer tegmentosum substrate, but there was no significant difference in the a-value and the b-value. The contents of and of fruiting body were increased at Acer tegmentosum substrate, but there was no significant difference in contents of CaO, MgO and .

C. Among the nine agro-chemicals, oxolinic acid WP,streptomycin + copper hydroxide WP and streptomycin WP were found to be effective for the inhibition ofthe pathogen in vitro. The results of scanning electron microscopic investigation showed that onion bulbs wasmacerated by infection of B. gladioli pv. alliicola CH1. B. gladioli pv. alliicola CH1 was able to producepolygalacturonase but did not produce pectin lyase and carboxymethylcellulase. In analysis of thepolygalacturonase activity of the isolated pectin-degradation enzymes from B. gladioli pv. alliicola CH1 totalprotein, three activity bands 45 kDa, 35 kDa, and 29 kDa were detected by the direct (or in-gel) activitystaining on SDS-PAGE. From black spots on winter mushroom (Flammulina velutipes), fluorescent bacteria were repeatedly isolated during surveys at places of production in the years 2009–2010 in Korea. From these lesions three bacterial strains (designated CHM13, CHM16, CHM17) were isolated which, following inoculation of mushroom stipes and caps, yielded characteristic black spots and sunken lesions, which developed into a severe black rot. Results of Gram stain and biochemical tests preliminarily identified these isolates as Pseudomonas tolaasii. This was confirmed by pathogenicity to winter mushroom, physiological and biochemical properties, analysis of the 16S rRNA and rpoB gene sequences, fatty acids profile, specific and sensitive PCR assays and, lipopeptide detection. This is the first report of the isolation of Pseudomonas tolaasii from cultivated winter mushroom in Korea.

To provide a basic information on the chemical concentrations of different raw materials used in mushroom cultivation, the raw materials were collected from 13 Flammulina velutipes, 14 Pleurotus eryngii, and 8 P. ostreatus farms and analyzed to calculate moisture contents, pH, total carbon(T-C), total nitrate(T-N), total phosphate(T-P) and 13 different cations. In our results, the C : N ratios of cotton seed meal, beet pulp, and corn-cob were 6~17, 12~29 and 56~127, respectively. Depending on the companies which process these materials, the range of C:N ratio of soybean curd residue was approximately either 8~9 or 14~17 with wider range of C : N ratio of the raw materials imported from other countries without the detection of heavy metals. In this study, the formula was provided to calculate the composition of mixed media for mushroom cultivation based on the ingredient chart of different raw materials.

This study was carried out to investigate interaction between mushroom mycelium and microor- ganisms in cotton waste media for the shelf cultivation of oyster mushroom. Two oyster mushroom farms was selected for this experiment. One was good mushroom farm (farmhouse I) and the other failed mushroom farm (farmhouse II). In farmhouse I, the inhibition microorganisms were higher toward the end of growth stage than the early stage, but the result of farmhouse II was opposite. Effects of the mycelium growth on plate culture showed same results on mushrooms as the earlier one. And the mycelium growth was influenced by secretory materials of microorganisms. Among of the isolates, Only few microorganism had inhibitory effects on either P. tolaasii or T. harzianum causing the disease of oyster mushrooms. But more microorganisms had inhibition effects on P. agarici.

Morphological characteristics of the fungal fruiting bodies produced on the insect sampleswere examined. A total of 542 entomopathogenic fungal isolates were obtained from the insect samples and iden-tified based on their morphological and cultural characteristics. Three species of Beauveria, eight species ofCordyceps and four species of Isaria were mainly isolated from the insect samples. The diversity of microflora according to growth stage of Pleurotus ostreatus and the correlation between microbe and medium fermentation were investigated. In farmhouse I, the aerobic bacteria and fungi as longer of growing period were increased. And, thermophilic bacteria and fluorescent Pseudomonas sp. showed high density at the early stage of spawn inoculation. The thermophilic actinomycetes were distributed evenly during all the growing period, but mesophilic actinomycetes were not observed. In farmhouse II, thermophilic actinomycetes were not observed in fermented medium and density of fungi were suddenly increased at 60 days after spawn inoculation. And also, mushrooms can hardly be harvested due to Penicillium spp. After medium fermentation, density of aerobic bacteria, thermophilic bacteria, and fluorescent Pseudomonas sp. was higher at farmhouse I than those of farmhouse II. In farmhouse I, Bacillus sp. and Pseudomonas sp. dominated at early stage of mushroom growth but as time goes by, density of Bacillus sp. was higher than the others. And also, the kind of microbe showed a few at early stage of mushroom growth but increased as time goes by. In farmhouse II, Bacillus sp. was dominated at early stage of mushroom growth. And the growth of Bacillus sp. and Pseudomonas sp. showed intersect aspect each other in the farmhouse I but Bacillus sp. dominated during all growth periods in the farmhouse II.

Phylogenetic relationships were investigated in the mushroom genus Agrocybe based on sequence data from the internal transcribed spaces(ITS) of nuclear ribosomal DNA(rDNA). Thirty strains including Agrocybe aegerita, A. grocybe cylindracea, A. grocybe praecox, A. grocybe pediades and Unknown Agrocybe species were subjected for the analysis. The Agrocybe spp. were separated into eight distinct groups. Phylogenetic analysis divided these species into eight groups as follows: A. chaxingu within group I, A. salicacola within group II, A. cylindracea and A. aegerita within group III, Agrocybe sp. within group IV and V, and other Agrocybe spp. were grouped within VI, VII, and VIII.

The internal stipe necrosis of cultivated mushrooms (Agaricus bisporus) is caused by the bacterium Ewingella americana, a species of the Enterobacteriaceae. Recently, Ewingella americana was isolated from cultivated white button mushrooms in Korea evidencing symptoms of internal stipe browning. Its symptoms are visible only at harvest, and appear as a variable browning reaction in the center of the stipes. From these lesions, we isolated one bacterial strain (designated CH4). Inoculation of the bacterial isolate into mushroom sporocarps yielded the characteristic browning symptoms that were distinguishable from those of the bacterial soft rot that is well known to mushroom growers. The results of Gram stain, flagellal staining, and biochemical tests identified these isolates as E. americana. This was verified by pathogenicity, physiological and biochemical characteristics, and the results of an analysis of the 16S rRNA gene sequences and the fatty acids profile. This is the first report of the isolation of E. americana from cultivated white button mushrooms in Korea.

This study was carried out to obtain the basic data for the mycelial growth conditions of cauliflower mushroom, Sparassis crispa. Twenty-one isolates were collected from domestic and abroad. The optimal temperature and pH for the mycelial growth of these isolates were and pH 6.0, respectively. The mycelial growth was the best in the HBA medium, but very poor in the Lilly medium. However no mycelial growth in the CzapekDox medium. The utilization of carbon source was the best with fructose, and that of nitrogen source was the best with glutamine when compared to other tested sources. The selected isolate ASI150010 produced the highest mycelial weight in liquid culture containing soybean mill () and potato () extract. And uncleaned rice, wheat and barley were found to be good substrates for the mycelial growth S. crispa.

In some cases, the problem with the mycelium of Tricholoma giganteum is delayed mycelial growth or non-regeneration. Therefore, this study was conducted to understand the mycelial storage condition of T. giganteum and to investigate the regeneration ratio of mycelial growth. T. giganteum obtained from KACC in RDA was evaluated for its simple preservation at and subcultured on different media. Mycelium of T. giganteum was cultured on the PBA (potato bamboo extract medium) for seven days at . Using simple preservation method, the mycelium of T. giganteum (MKACC 50852) and Pleurotus ostreatus (Chunchu No. 2) were stored on six different media in two kinds of storage vessels (tube and vial) for a period of 1-12 months at , and storage temperatures. At storage condition, mycelial regeneration was failed in all agar media, but was successful in the sawdust medium for 3 months. At storage, mycelial activity was maintained up to six months. On the other hand, P. ostreatus produced well-regenerated mycelia both at and storage up to 12 months. In conclusion, it is estimated that the mycelia storage condition of T. giganteum must be done at and subcultured within six months.
