Hookworm infections cause a neglected tropical disease (NTD) affecting ~740 million people worldwide, principally those living in disadvantaged communities. Infections can cause high morbidity due to their impact on nutrient uptake and their need to feed on host blood, resulting in a loss of iron and protein, which can lead to severe anaemia and impaired cognitive development in children. Currently, only one drug, albendazole is efficient to treat hookworm infection and the scientific community fears the rise of resistant strains. As part of on-going efforts to control hookworm infections and its associated morbidities, new drugs are urgently needed. We focused on targeting the blood-feeding pathway, which is essential to the parasite survival and reproduction, using the laboratory hookworm model Nippostrongylus brasiliensis (a nematode of rodents with a similar life cycle to hookworms). We established an in vitro-drug screening assay based on a fluorescent-based measurement of parasite viability during blood-feeding to identify novel therapeutic targets. A first screen of a library of 2654 natural compounds identified four that caused decreased worm viability in a blood-feeding-dependent manner. This new screening assay has significant potential to accelerate the discovery of new drugs against hookworms.

RIVM rapport:Het ministerie van Infrastructuur en Waterstaat (IenW) heeft het RIVM gevraagd een instrument te ontwikkelen waarmee het mogelijk is te peilen hoe veilig omwonenden van chemische industrie zich voelen. In een pilot is de eerste opzet van het instrument ontwikkeld en getest in twee steden? één met veel chemische industrie in de omgeving (Zaandam), en één met enkele chemiebedrijven in de stad (Deventer). Het belangrijkste doel van dit onderzoek is het ontwikkelen van een nieuw instrument. Het ontwikkelde instrument bestaat uit een vragenlijst die inzicht geeft in hoe veilig omwonenden activiteiten met gevaarlijke stoffen ervaren. De antwoorden maken ook duidelijk welke factoren samenhangen met hun veiligheidsbeleving. De vragenlijst blijkt geschikt te zijn om veiligheidsbeleving te meten en kan periodiek worden ingezet om trends te kunnen waarnemen. Ook doet het RIVM concrete suggesties om de vragenlijst te verbeteren, zoals het aanpassen van de volgorde van de vragenlijst. Daarnaast moet een effectieve werkwijze worden gekozen om voldoende representatieve deelnemers te werven. Naast de ontwikkelde methode bevat dit rapport ook een enkele inhoudelijke opbrengsten van de pilot. Omdat de deelnemers aan de pilot de inwoners van de twee steden onvoldoende weerspiegelden, zijn de inhoudelijke opbrengsten van de pilot indicatief. Uit dit onderzoek blijkt dat deelnemers uit de stad met enkele chemiebedrijven de veiligheid positiever beleven dan deelnemers uit de stad die nabij veel chemische industrie ligt. De factor die het meest samenhangt met de veiligheidsbeleving is de mate waarin mensen positief zijn over de omgeving waarin zij wonen. Hoe positiever mensen hun woonomgeving ervaren hoe positiever de veiligheidsbeleving en andersom. Ook de mate waarin mensen erop vertrouwen dat overheden en bedrijven een ongeval kunnen voorkómen, hangt samen met de veiligheidsbeleving. Hoe groter dit vertrouwen, hoe groter de veiligheidsbeleving en andersom. The substantive result of the pilot should be considered indicative, as the participants of the pilot did not form a sufficiently representative sample of the residents of the two cities. It might be obvious, however the answers indicate that residents of a city with a few chemical companies feel safer than residents of a city located near a great deal of chemical industry. The factor that is most closely related to the feeling of safety is the degree to which people are positive about their living environment. The more positive the experience of the living environment is, the more positive the feeling of safety is and vice versa. Beter inzicht in de oorzaken van het ontstaan van beroepsziekten biedt kansen om ze eerder en beter te herkennen. Op basis van die informatie kunnen preventieve maatregelen worden opgesteld. Een onderzoeksmodel van het RIVM dat achterliggende oorzaken van ernstige arbeidsongevallen in kaart brengt (Storybuilder), blijkt ook voor beroepsziekten te kunnen worden gebruikt. Aanbevolen wordt de kennis uit deze verkenning te toetsen bij professionals uit de praktijk, om zicht te krijgen op de waarde ervan voor praktijk en beleid. Een beroepsziekte is een ziekte die mensen hebben opgelopen hoofdzakelijk als gevolg van het werk of de werkomstandigheden. Een voorbeeld is het ontstaan van mesothelioom door te werken met asbest. Het RIVM heeft voor deze verkenning twintig dossiers bekeken: tien dossiers van mensen met de schildersziekte (OPS/CTE) en tien dossiers van mensen met onherstelbare rugklachten. Het blijkt mogelijk om met Storybuilder bestaande casussen in beeld te krijgen. De eerste Storybuilder-modellen van de twee beroepsziekten zijn in dit rapport beschreven, maar deze werkwijze is nog niet gevalideerd of met praktijkprofessionals besproken. RIVM examined 20 dossiers for this exploratory study: 10 case files of people with OPS (organic psychosyndrome) / CTE (chronic toxic encephalopathy) and 10 case files of people with irreversible back complaints. It turned out to be possible to use Storybuilder to get a picture of existing case studies. The first Storybuilder models of the two occupational diseases are described in the report, but this working method has not yet been validated or discussed with professionals from the field. The researchers also observed that the dossiers studied do not contain all the information needed to give a complete factual description of how the occupational conditions arise. It is recommended that an inventory should be made of additional information sources for the information that is as yet missing. If this succeeds, it will be possible to carry out studies in greater depth with the Storybuilder models for occupational diseases and if possible extend them to other types of occupational diseases.
