Also, it gained importance last year because of the increasing industrial demand of biodiesel. The objective of this study was to study the effect of the pH (10, 11 and 12) of extraction of proteins from castor bean cake on some physical properties of films based on these proteins modified by tannin. Increasing pH improved the protein extraction yield without affecting their amino acid profile. The films prepared using these proteins by casting process showed a structural matrix sufficiently cohesive, intense total color difference (?E* = 83–85) and relatively high opacity (8–15%). The values of tensile strength, elongation at break, Young Modulus, water vapor permeability and hydrophobicity of films were affected by the protein extraction pH. The protein extraction in pH = 12 resulted in more rigid, more resistant, with enhanced extensibility, a lower rate of water diffusion and less hydrophilic films. The films showed homogeneous surfaces and dense internal microstructure. FT-IR spectra of all films showed characteristic bands and peaks typical of proteins with no differences in positions and intensity due to pH variations. It is possible to produce films from proteins extracted from castor bean cake by casting process. Grosso Tese (doutorado) - Universidade Estadual de Campinas, Faculdade de Engenharia de Alimentos Resumo: Uma alternativa interessante na produção de filmes comestíveis com propriedades mecânicas e de permeabilidade melhoradas é a mimetização de estruturas biológicas como a parede celular e a cutícula dos tecidos vegetais por conterem na sua composição polissacarídeos, proteínas e lipídios. O objetivo do presente trabalho foi produzir e caracterizar filmes compostos de glucomanana, pectina, metilcelulose, gelatina e lipídios. O trabalho foi desenvolvido em três etapas: (1) produção de filmes a partir dos polissacarídeos: glucomanana, pectina e metilcelulose, (2) produção de filmes à base de polissacarídeos e gelatina em diferentes proporções e pHs e (3) produção de filmes à base de misturas de polissacarídeos/gelatina e lipídios: cera de carnaúba, ácidos graxos esteárico e palmítico, estearoil-2-lactil-lactato de sódio (SSL) e éster de sacarose (ES). Nesta última etapa, os lipídios foram incorporados na forma de emulsão (contendo todos os lipídios) e na forma de dispersão (contendo SSL ou ES). Os filmes foram caracterizados quanto às suas propriedades mecânicas, permeabilidade ao vapor de água (PVA), morfologia, solubilidade, absorção de água, opacidade e estrutura (infravermelho e raios X). Na primeira etapa foram obtidos filmes com propriedades mecânicas melhoradas a partir da mistura metilceluloseglucomanana- pectina na proporção 1:4:1, respectivamente. Estes filmes mostraram superfícies homogêneas sem separação de fases e um valor de PVA intermediário em relação às outras formulações. Também foram evidenciadas interações entre os polissacarídeos nesta mistura, através dos espectros de infravermelho e raios X. Na segunda etapa foram obtidos filmes com PVA reduzida a partir da mistura de polissacarídeos e gelatina nas proporções 90/10 e 10/90, respectivamente, em pH 5. Para estes filmes foram observados altos valores de absorção de água a 95% de umidade relativa em relação às outras formulações. A seção transversal dos filmes com PVA reduzida mostrou uma estrutura pouco compacta e em camadas, e as propriedades mecânicas não foram melhoradas significativamente. Na terceira etapa também foram obtidos filmes com PVA reduzida a partir da mistura polissacarídeo/gelatina com adição de lipídios, onde as dispersões não prejudicaram as propriedades mecânicas. Os filmes polissacarídeo/gelatina contendo as emulsões mostraram glóbulos de tamanhos variados dispersos na superfície, enquanto que, os produzidos com as dispersões apresentaram superfícies lisas, com microfuros e granulosidade. A opacidade aumentou significativamente em função do uso das emulsões, enquanto que, as dispersões provocaram mudanças pouco significativas. Filmes comestíveis com bom desempenho, ou seja, com altos valores de tensão na ruptura e elongação foram obtidos através da mistura dos polissacarídeos e; filmes com baixos valores de PVA foram obtidos a partir da combinação dos polissacarídeos com a gelatina, ou da combinação polissacarídeo/gelatina com os lipídios Abstract: One interesting alternative in edible films production with improved mechanical properties and water vapor permeability is the mimic of biological structures like cell wall and cuticle of plant tissue by containing in their composition polysaccharides, proteins and lipids. The objective of this work was to produce and to characterize composite films of glucomannan, pectin, methylcellulose, gelatin and lipids. This work was developed in three stages: (1) films production from polysaccharides: glucomannan, pectin and methylcellulose, (2) films production from polysaccharides and gelatin in different ratios and pHs and (3) films production from mixture of gelatin/polysaccharides and lipids: carnauba wax, stearic and palmitic fatty acid, sodium stearoyl-2-lactylate (SSL) and sucrose ester (ES). In this last stage, lipids were added like emulsions (containing all lipids) and like dispersion (containing SSL or ES). The films were characterized by their mechanical properties, water vapor permeability (WVP), morphology, solubility, water absorption, opacity and structure (infrared spectroscopy and x ray). In the first stage, films with improved mechanical properties were obtained from methylcellulose-glucomannan-pectin mixture in the ratio of 1/4/1, respectively. These films showed homogeneous surfaces without phases separation and WVP intermediate values with regard to others formulations. Also, interactions were verifyed between the polysaccharides in this mixture through infrared spectroscopy and x ray. In the second stage, films with reduced WVP were obtained from mixture of gelatin/polysaccharides in the ratios of 90/10 and 10/90, respectively, in pH 5. For these films were observed high water absorption values at 95% relative humidity with regard to other formulations. The transversal section microscopy of films with reduced WVP showed a structure few compact and in layers, and the mechanical properties were improved significantly. In the third stage, films with reduced WVP were obtained from mixture of gelatin/polysaccharides and lipids, where the dispersions had not impaired the mechanical properties. The films polysaccharides/gelatin containing emulsions showed globules of different size dispersed in the surface, whereas, the ones containing the dispersions showed homogeneous surfaces with micro holes and granularity. The opacity increased significantly by use of emulsions, whereas, the dispersions caused few changes. Orientador: Miriam Dupas Hubinger Dissertação (mestrado) - Universidade Estadual de Campinas, Faculdade de Engenharia de Alimentos Resumo: No mundo atual é crescente a demanda dos consumidores por produtos frescos, devido a sua qualidade nutricional, além de oferecer benefícios para a saúde como, por exemplo, na prevenção de algumas doenças. Frente a esta demanda, o presente trabalho estudou o uso de coberturas comestíveis de fécula de mandioca na manutenção da qualidade dos frutos de morango (Fragaria ananassa Duch.). Os frutos de morango receberam tratamentos combinando o processamento mínimo com a utilização de cobertura comestível de fécula de mandioca, cuja finalidade foi prolongar a vida útil destes em temperaturas de refrigeração. Os frutos após o processamento foram mergulhados por 10 minutos em suspensão de 3% (p/v) de fécula de mandioca e submetidos à secagem naturalmente à temperatura ambiente e armazenados em temperaturas de 10 ?C e 15 ?C. Na primeira etapa, o objetivo foi avaliar as características da cobertura aplicada nos morangos (propriedades mecânicas: forças de ruptura e penetração, cor dos morangos, taxa de respiração e resistência ao vapor de agua) depois de 24 horas do tratamento. A aplicação da cobertura nos morangos não alterou significativamente as propriedades mecânicas (forças de ruptura e penetração), cor dos morangos, taxa de respiração e resistência ao vapor de água em comparação aos morangos sem coberturas. Na segunda etapa foi realizado o estudo da vida útil dos morangos com e sem cobertura, armazenados a temperaturas de 10 ?C e 15 ?C. Na perda de peso verificou-se diferença significativa ao longo do tempo de armazenamento diferença significativa, e no tempo fixo não se observou diferenças significativas entre os tratamentos, a taxa de respiração mostrou um incremento significativo em todos os tratamentos ao longo no tempo de armazenamento e para tempo fixo não presentaram diferenças significativo entre os tratamentos exceto no dia 1 se observou que o tratamento 15 ?C com cobertura diminui significativamente com respeito a 15 ?C sem cobertura. O teor de antocianinas apresentou uma diminuição significativa ao longo do tempo de armazenamento para os tratamentos 10 ?C sem cobertura e 15 C sem e com cobertura em ambas temperaturas são não significativo nos dia 5 e 8. As amostras 10 ?C com e sem cobertura e 15 ?C com e sem cobertura foram avaliadas sensorialmente os morangos com cobertura apresentaram notas acima do limite de aceitação (4,5), o fator que determinou o tempo de vida útil foi a contaminação microbiológica observada nas amostras. As amostras armazenadas a 10 ?C com e sem cobertura apresentaram 8 dias de vida útil e as amostras armazenadas 15 ?C com e sem cobertura apresentaram 5 dias de vida útil Abstract: Nowadays there is a growing consumer demand for fresh products, due to their nutritional quality and the health benefits they provide, for example, in the prevention of some diseases. Faced with this demand, the present work analyzed the use of edible cassava starch coating for preserving the quality of strawberry fruits (Fragaria ananassa Duch.). Strawberries underwent treatment combining minimal processing with the use of edible cassava starch coating, aiming to extend the lifetime of these fruits at refrigerated temperatures. To this aim, after the processing stage, the fruits were immersed for 10 minutes in a 3% (w/v) cassava starch suspension; then they were naturally dried at room temperature and stored at temperatures of 10?C and 15?C. During the first stage, the objective was to evaluate the characteristics of the starch-based coating applied to strawberries ¿ that is, mechanical properties (compression and penetration strengths) strawberry color, respiration rate and water vapor resistance ¿ after a 24-hour treatment. The application of the coating on the strawberries did not significantly modify the mechanical properties (compression and penetration strengths), strawberry color, respiration rate and resistance to water vapor, when compared to uncoated strawberries. During the second stage we studied the lifetime of coated and uncoated strawberries stored at temperatures of 10?C and 15?C. In the weight loss treatments, we were able to verify significant differences during the storage time, and in the fixed time no significant differences were observed, the respiration rate showed significant increase in all treatments during storage time, and in the fixed time, no significant differences among treatments were seen, except for day 1, when it was observed that the coated fruits under treatment at 15 ?C showed a significant decrease with regard to the uncoated fruits treated at 15?C. The anthocyanin content showed significant decrease during storage time for uncoated fruits at 10?C and for coated and uncoated fruits at 15?C; at both temperatures there were no significant results on days 5th and 8th. Sensory analysis was performed on samples at coated and uncoated stored at 10?C and 15?C . Coated strawberries showed numbers above the acceptance limit (4.5). The factor that determined the lifetime was the microbiological contamination observed in the samples. Orientador: Miriam Dupas Hubinger Tese (doutorado) - Universidade Estadual de Campinas, Faculdade de Engenharia de Alimentos Resumo: A alcachofra (Cynara scolymus L.) é rica em compostos com propriedades funcionais, como frutooligossacarídeos. Os resíduos da fabricação de conservas de alcachofra, que representam 70% do total da flor de alcachofra, possuem os mesmos compostos bioativos que o coração utilizado na fabricação de conservas, porém são descartados. Assim, o objetivo deste trabalho foi produzir extratos a partir de resíduos de alcachofra, clarificá-los e concentrá-los por processos de separação por membranas, obtendo, ao final do processo, um extrato rico em frutooligossacarídeos. O desenvolvimento experimental foi iniciado pelo estudo da cinética de extração aquosa por três diferentes métodos: convencional (agitação com alta temperatura), ultrassom com sonicação direta e com sonicação indireta. A técnica que apresentou maior rendimento de extração de açúcares prebióticos, ultrassom com sonicação direta, foi selecionada e as variáveis de processo (tempo, temperatura e potência) foram estudadas. Encontrados os níveis ótimos para extração (10 min, 60 °C e 360 W), os açúcares prebióticos do extrato foram identificados e quantificados. Em seguida, esse extrato foi submetido à clarificação por microfiltração, utilizando unidade de filtração laboratorial, duas membranas de diferentes tamanhos de poros e três pressões de trabalho (1, 2 e 3 bar). Nesta etapa foi avaliado: o perfil do fluxo de permeado ao longo do processo, a incrustação, a redução do teor de sólidos e a rejeição dos compostos de interesse. A membrana MP005 (com material de polietersulfona e tamanho de poro de 0,05 ?m) foi selecionada para clarificação, pois apresentou a menor queda de fluxo de permeado, menor nível de incrustação e alta recuperação dos açúcares prebióticos no permeado. Desta maneira, o extrato clarificado foi submetido à nanofiltração, utilizando também unidade de filtração laboratorial, três membranas de diferentes massas molares de corte (MWCO) e três pressões de trabalho (10, 15 e 20 bar), avaliando-se os mesmos parâmetros descritos anteriormente para microfiltração. A membrana NF270 (com material de poliamida e MWCO de 150-300 g/mol) alcançou o maior fator de concentração de açúcares prebióticos, possuindo alto fluxo, sendo assim selecionada. Ao final, a microfiltração foi avaliada em sistema piloto de filtração tangencial e a nanofiltração em unidade piloto com módulo espiral, o qual alcançou redução de volume de 20 vezes, e alta concentração de açúcares prebióticos Abstract: Artichoke (Cynara scolymus L.) is rich in compounds with functional properties, such as fructooligosaccharides. The waste from artichoke canning industry, which are about 70% of the total artichoke flower, contain the same bioactive compounds that edible portion of the plant, however they are discarded. Therefore, the objective of this work was producing extracts from artichoke waste, then clarify and concentrate them using membrane separation processes, in order to obtain, in the end of process, an extract rich in fructooligosaccharides. The experimental development was initiated by kinetic study of aqueous extraction by three different methods: conventional (shaking with high temperature), ultrasound with direct and indirect sonication. The technique that showed higher extraction yield of prebiotic sugar, ultrasound with direct sonication, was selected and the process variables (time, temperature and power) were studied. The optimum level for extraction were found (360 W, 60 °C e 10 min) and the prebiotic sugars of optimized extract was identified and quantified. Thereafter, this extract was subjected to clarification by microfiltration, using a laboratory filtration unit, testing two membranes with different pore sizes and three pressures (1, 2 and 3 bar). In this step, permeate flux, fouling of membrane, reduction of solid content and rejection of compounds of interest were evaluated. MP005 membrane (with polyethersulfone material and pore size of 0.05 ?m) was selected for clarification; since it showed the lowest permeate flux decay and fouling, and high recovery of prebiotic sugars in permeate side. Then, the clarified extract was subjected to nanofiltration, using also a laboratory filtration unit, three membranes with different molecular weight cut-off and three pressures (10, 15 and 20 bar), evaluating the same parameters previously described for microfiltration. NF270 membrane (with polyamide material and mass molecular weight of 150-300 g/mol) achieved the highest concentration factor of prebiotic sugars, showing high flux, therefore this membrane was selected for posterior step. Finally, the microfiltration was evaluated in a tangential filtration unit and the nanofiltration was tested in spiral wound module membrane, which achieved a volume reduction of 20 times and, consequently, high prebiotic sugar concentration Doutorado Engenharia de Alimentos Doutora em Engenharia de Alimentos
