Questions have been raised apropos the emerging problem of microbial resistance, which may pose a great hazard to the human health. Among biosafe compounds are essential oils which captured consumer draw due to their multifunctional properties compared to chemical medication drugs. Here, we examined the chemical profile and the mechanism(s) of action of the Thymus vulgaris essential oil (TVEO) against a Gram-negative bacterium Salmonella enterica Typhimurium ATTCC 10028 (S. enterica Typhimurium ATTCC 10028) and two Gram-positive bacteria Staphyloccocus aureus ATCC 6538 (S. aureus ATCC 6538) and Listeria monocytogenes ATCC 19117 (L. monocytogenes ATCC 19117). Findings showed that TVEO was principally composed of thymol, o-cymene, and γ-terpinene with 47.44, 16.55, and 7.80%, respectively. Molecular docking simulations stipulated that thymol and β-sesquiphellandrene (a minor compound at 1.37%) could target multiple bacterial pathways including topoisomerase II and DNA and RNA polymerases of the three tested bacteria. This result pointed plausible impairments of the pathogenic bacteria cell replication and transcription processes. Through computational approach, the VEGA quantitative structure–activity relationship (QSAR) model, we revealed that among twenty-six TVEO compounds, sixteen had no toxic effects and could be safe for human consumption as compared to the Food and Drug Administration (FDA) approved drugs (ciprofloxacin and rifamycin SV). Assessed by the SwissADME server, the pharmacokinetic profile of all identified TVEO compounds define their absorption, distribution, metabolism, and excretion (ADME) properties and were assessed. In order to predict their biological activity spectrum based on their chemical structure, all TVEO compounds were subjected to PASS (Prediction of Activity Spectra for Substances) online tool. Results indicated that the tested compounds could have multiple biological activities and various enzymatic targets. Findings of our study support that identified compounds of TVEO can be a safe and effective alternative to synthetic drugs and can easily combats hazardous multidrug-resistant bacteria.

Among them, beetroot leaves (BL) are known for their therapeutic properties. Growing worldwide, the genus Ephedra (family Ephedraceae) had a medicinal, ecological, and economic value. The extraordinary morphological diversity suggests that Ephedra was survivor of an ancient group, and its antiquity is also supported by fossil data. It has recently been suggested that Ephedra appeared 8–32 million years ago, and a few megafossils document its presence in the Early Cretaceous. Recently, the high analytical power provided by the new mass spectrometry (MS) instruments is making the characterization of Ephedra metabolites more feasible, such as ephedrine series. In this regard, the chemical compounds isolated from crude extracts, fractions, and few isolated compounds of Ephedra species were characterized by MS-based techniques (LC-MS, LC-ESI-MS, HPLC-PDA-ESI/MS, LC-DAD-ESI/MSn, LC/Orbitrap MS, etc.). Moreover, we carry out an exhaustive review of the scientific literature on biomedicine and pharmacotherapy (anticancer, antiproliferative, anti-inflammatory, antidiabetic, antihyperlipidemic, antiarthritic, and anti-influenza activities; proapoptotic and cytotoxic potential; and so on). Equally, antimicrobial and antioxidant activities were discussed. This review is focused on all these topics, along with current studies published in the last 5 years (2015–2019) providing in-depth information for readers.

The chemical composition and the biological activities of aromatic plants may be influenced by seasonal changes. Therefore, the essential oil of Inula graveolens (IGEO), collected at Chebba salt marsh, was studied in terms of yields, compositions and biological activities, throughout four different seasons, namely spring, summer, autumn and winter (April, July, October and January, respectively). GC/MS analysis identified 30 compounds. Mostly quantitative rather than qualitative, variation was observed in the oil composition of each sample. It had been revealed a predominant presence of bornyl acetate and borneol, as well as significant differences of several compound amounts in function of the seasonal change. Biological activities were also related to the harvest season; hence, IGEO from spring gave the best antioxidant activity results and IGEO from autumn seemed to be the most potent against pathogenic microorganisms, while oils from summer and winter were the strongest inhibitors of acetylcholinesterase and tyrosinase.A matrix linking IGEO major compounds to biological was composed to identify relationships between concentrations of the volatile molecules and the biological activities of the samples. Furthermore, the seasonal variation of these main volatile constituents was also investigated through principal compound analysis (PCA). The obtained results revealed that each biological activity depends on the seasonal fluctuation of the amounts of certain chemical compounds. A new series of ruthenium(II) N-heterocyclic carbene complexes has been synthesized via transmetalation. These complexes are easily handled in very good yields via synthesis of silver–NHC complexes and subsequent reaction with [RuCl2(p-cymene)]2 in dichloromethane (DCM) under argon atmosphere. The new ruthenium(II)–NHC complexes were applied to transfer hydrogenation of ketone derivatives using 2-propanol as a hydrogen source and KOH as a co-catalyst. The results show that the corresponding alcohols could be obtained in good yield with high catalyst activity (up to 76%) under mild conditions. All new compounds were characterized by spectroscopic and analytical methods. The antibacterial activities of the corresponding ruthenium(II)–NHC complexes 4a–i were tested against positive and negative bacteria using the agar dilution procedure and their IC50 values have been determined. A cytotoxicity study of the ruthenium(II)–NHC complexes 4 was performed against the two human cancer cell lines MDA-MB-231 and MCF-7. In addition, the scavenging activity of the synthesized complexes for the DPPH radical was also determined.

New benzimidazolium salts (2) and their copper N-heterocyclic carbene complexes (3–4) were designed, synthesized and structurally characterized by NMR (1H and 13C), IR, HRMS and elemental analysis. All obtained complexes especially compounds 3d and 3e, presented significant inhibitory activity against the tested food-borne pathogens and clinical microorganisms. The compound 3b presents against S. aureus a MIC value of 0.0195 mg/ml quite similar to that of ampicillin (0.0195 mg/ml) used as standard. Compounds 3a and 3b, from a concentration of 0.5 mg/ml, present an identical scavenging activity to that of the two used controls butylated hydroxytoluene (BHT) and gallic acid (GA). Concerning acetylcholinesterase inhibition activity (AChEI), compound 3e presents an interesting AChEI activity with 32.80% of inhibition.

In this study, a series of unsymmetrical N,N-disubstituted benzimidazolium salts were synthesized as N-heterocyclic carbene (NHC) (2a-2j) and their AgI complexes. Their structures were characterized by elemental analyses, 1H and 13C nuclear magnetic resonance (NMR) and infrared (IR) spectroscopy techniques. Further, both the salts and their silver(I)-NHC complexes have been evaluated for their potential antibacterial properties against a panel of bacterial strains namely, Micrococcus luteus, Listeria monocytogenes, Salmonella Typhimurium, Staphylococcus aureus and Pseudomonas aeruginosa. The results show that silver complexes are effective against SalmonellaTyphimurium, Listeria monocytogenes, and Micrococcus luteus with moderate to high activity, and their minimum inhibitory concentrations ranging from 0.0034 to 1.26 mg mL−1. In addition, the benzimidazoles salts 2a-2j and silver-NHC complexes 3a-3j were screened for their antitumor activity. Complex 3e exhibited the highest antitumor effect with half-maximal inhibitory concentration (IC50) value of 4.2 µg mL-1 against MCF-7.

Extraction of Tunisian pomegranate peels was employed with different solvents such as ethyl acetate, acetonitrile, and water. Total phenolic and flavonoids contents, antioxidant activity, and antibacterial capacity against five foodborne pathogenic bacteria were evaluated. The highest values of polyphenols (351 mg gallic acid equivalent/g), flavonoids (104 mg quercetin/g), and DPPH and ABTS inhibition were recorded in the ethyl acetate extract followed by the aqueous extract. The latter present the maximum antibacterial potential against S. enterica, P. aeruginosa, and E. coli. The potential use of the lyophilized aqueous extract (AE), used for safety reason and being rich in phenolic, as biopreservative in minced beef meat was described. AE was incorporated at 0.1, 0.5, and 1% and compared with 0.1% butylated hydroxytoluene (BHT). During 21 days at 4°C, AE at 1% could appreciably retard the microflora proliferation (p<0.05), the accumulation of MetMb and the carbonyl group (p<0.05), slowing down the loss of sulphydryl proteins (p<0.05), and led to a decrease (p<0.05) in primary (peroxide value and conjugated dienes) and secondary lipid oxidation (TBARS) in treated meat. By the 14th day, AE-treated minced meat obtained higher sensory scores than untreated and BHT samples. Based on these results, lipid and protein oxidation changes and sensorial attributes were useful in discriminating meat samples by overall acceptability prediction. Generally, AE at 1% presented the potent preservative effect that could be utilized as an application on meat-substituting synthetic antioxidant.

In previous work we have isolated and identified a new strain calledEnterococcus faeciumFL31. The active compound secreted by this strain, “BacFL31”, has been purified and characterized. In the present study, safety aspect, assessed by microbiological and molecular tests, demonstrated thatEnterococcus faeciumFL31 was susceptible to relevant antibiotics, free of hemolytic, gelatinase, DNase, and lipase activities. In addition, it did not harbor virulence and antibiotic resistance genes. Combined SYTOX Green dye and UV-absorbing experiments, along with released extracellular potassium and transmembrane electrical potential measurements, showed that pure BacFL31 at a concentration of 1×MIC (50 μg/mL) could damage cytoplasmic membrane of the pathogenListeria monocytogenesATCC19117. The same concentration causes the leakage of its intracellular constituents and leads to the destruction of this pathogenic microorganism. In summary, this work reflected characteristics ofEnterococcus faeciumFL31 strain and its bacteriocin in terms of functional and safety perspectives.

A series of Cu-N-heterocyclic carbene (Cu-NHC) complexes, 3a-c, were synthesized and characterized by spectroscopic methods such as NMR, HRMS and elemental analysis. The performance of these (Cu-NHC) complexes, 3a-c showed excellent activity in 1,3-dipolar cycloaddition reactions between alkynes and azides to obtain 1,4-disubstituted triazoles 6a-d in 80%–91% isolated yields. All obtained complexes especially compounds 3a and 3c, presented significant inhibitory activity against the tested food-borne pathogens and clinical microorganisms. The compound 3a presents against S. aureus a MIC value of 4.8 μg/ml quite similar to that of ampicillin (4 μg/ml) used as standard. Compounds 3a and 3b, from a concentration of 0.5 mg/ml, present an identical scavenging activity to that of the two used controls butylated hydroxytoluene (BHT) and gallic acid (GA). Concerning acetylcholinesterase inhibition activity (AChEI), compound 6a presents an interesting AChEI activity with 68.3% of inhibition.
