Seasonal outbreaks of respiratory viral infections remain a global concern, with increasing morbidity and mortality rates recorded annually. Timely and false responses contribute to the widespread of respiratory pathogenic diseases owing to similar symptoms at an early stage and subclinical infection. The prevention of emerging novel viruses and variants is also a big challenge. Reliable point-of-care diagnostic assays for early infection diagnosis play a critical role in the response to threats of epidemics or pandemics. We developed a facile method for specifically identifying different viruses based on surface-enhanced Raman spectroscopy (SERS) with pathogen-mediated composite materials on Au nanodimple electrodes and machine learning (ML) analyses. Virus particles were trapped in three-dimensional plasmonic concave spaces of the electrode via electrokinetic preconcentration, and Au films were simultaneously electrodeposited, leading to the acquisition of intense and in-situ SERS signals from the Au–virus composites for ultrasensitive SERS detection. The method was useful for rapid detection analysis (< 15 min), and the ML analysis for specific identification of eight virus species, including human influenza A viruses (i.e., H1N1 and H3N2 strains), human rhinovirus, and human coronavirus, was conducted. The highly accurate classification was achieved using the principal component analysis-support vector machine (98.9%) and convolutional neural network (93.5%) models. This ML-associated SERS technique demonstrated high feasibility for direct multiplex detection of different virus species for on-site applications.

Nanoscale plasmonic hotspots play a critical role in the enhancement of molecular Raman signals, enabling the sensitive and reliable trace analysis of biomedical molecules via surface-enhanced Raman spectroscopy (SERS). However, effective and label-free SERS diagnoses in practical fields remain challenging because of clinical samples' random adsorption and size mismatch with the nanoscale hotspots. Herein, we suggest a novel SERS strategy for interior hotspots templated with protein@Au core-shell nanostructures prepared via electrochemical one-pot Au deposition. The cytochrome c and lysates of SARS-CoV-2 (SLs) embedded in the interior hotspots were successfully functionalized to confine the electric fields and generate their optical fingerprint signals, respectively. Effective hotspot engineering with facile and cost-effective fabrication procedures is critical for the practical application of surface-enhanced Raman spectroscopy (SERS). We propose a SERS substrate composed of a metal film over polyimide nanopillars (MFPNs) with three-dimensional (3D) volumetric hotspots for this purpose. The 3D MFPNs were fabricated through a two-step process of maskless plasma etching and hydrogel encapsulation. The probe molecules dispersed in solution were highly concentrated in the 3D hydrogel networks, which provided a further enhancement of the SERS signals. SERS performance parameters such as the SERS enhancement factor, limit-of-detection, and signal reproducibility were investigated with Cyanine5 (Cy5) acid Raman dye solutions and were compared with those of hydrogel-free MFPNs with two-dimensional hotspots. The hydrogel-coated MFPNs enabled the reliable detection of Cy5 acid, even when the Cy5 concentration was as low as 100 pM. We believe that the 3D volumetric hotspots created by introducing a hydrogel layer onto plasmonic nanostructures demonstrate excellent potential for the sensitive and reproducible detection of toxic and hazardous molecules.

Nanostructured materials offer the potential to drive future developments and applications of electrochemical devices, but are underutilized because their nanoscale cavities can impose mass transfer limitations that constrain electrochemical signal generation. Here, we report a new signal-generating mechanism that employs a molecular redox capacitor to enable nanostructured electrodes to amplify electrochemical signals even without an enhanced reactant mass transfer. The surface-tethered molecular redox capacitor engages diffusible reactants and products in redox-cycling reactions with the electrode. Such redox-cycling reactions are facilitated by the nanostructure that increases the probabilities of both reactant-electrode and product-redox-capacitor encounters (i.e., the nanoconfinement effect), resulting in substantial signal amplification. Using redox-capacitor-tethered Au nanopillar electrodes, we demonstrate improved sensitivity for measuring pyocyanin (bacterial metabolite). This study paves a new way of using nanostructured materials in electrochemical applications by engineering the reaction pathway within the nanoscale cavities of the materials.

We introduce a facile and low-cost method for fabricating gold nanostructures on cellulose filter paper (CFP) to prepare a paper-based surface-enhanced Raman scattering (SERS) sensor for label-free molecular detection. Polymerized dopamine (PD) was used as an adhesive layer on the CFP and simultaneously functioned as a reducing agent for gold nanoparticle (AuNP) nucleation. The size of the AuNPs was dependent on the pH of the gold precursor solution, and nanoparticles with an average size of 102 nm were formed on the PD-coated CFP at a pH 3, exhibiting high SERS activity. Finite-difference time-domain (FDTD) simulations of the electromagnetic field enhancement of AuNPs with different sizes and interparticle distances were performed to identify the origin of the SERS effect. The developed paper-based SERS substrate showed uniform and excellent molecular sensitivity with a limit of detection (LOD) of 10−7 M for methylene blue, as measured by a portable Raman spectrometer. Furthermore, as a field application test, surfaces of apples were pretreated with diquat (DQ) and paraquat (PQ) pesticides, which were then detected down to a concentration of 1 ppm after simple attachment of the sensor on the apple peels and performing a SERS measurement. The developed paper-based SERS sensor is expected to be applicable as a label-free sensor for a variety of chemical and biological molecules.

Engineering of interior hotspots provides a paradigm shift from traditional surface-enhanced Raman spectroscopy (SERS), in which the detection sensitivity depends on the positioning of adsorbed molecules. In the present work, we developed an Ag–Au bimetallic nanocomposite (SGBMNC) SERS platform with interior hotspots through facile chemical syntheses. Ag nanoparticles replaced by Au via the galvanic replacement reaction (GRR) provided hotspot regions inside the SGBMNC that remarkably enhanced the plasmonic activity compared to the conventional SERS platforms without the internal hotspots. The diffusion of analytes into the proposed interior hotspots during the GRR process enabled sensitive detections within 10 s. The SERS behaviors of the SGBMNC platform were investigated using methylene blue (MB) as a Raman probe dye. A quantitative study revealed excellent detection performance, with a limit of detection (LOD) of 42 pM for MB dye and a highly linear correlation between peak intensity and concentration (R2 ≥ 0.91). The SGBMNC platform also enabled the detection of toxic benzyl butyl phthalate with a sufficient LOD of 0.09 ppb (i.e., 280 pM). Therefore, we believe that the proposed methodology can be used for SERS assays of hazardous materials in practical fields.

The sensitivity and limit-of-detection (LOD) of the traditional surface-enhanced Raman spectroscopy (SERS) platform suffer from the requirement of precise positioning of small analytes, including DNAs and bacteria, into narrow hotspots. We report on a quasi-three-dimensional (3D) plasmonic nanowell array with high structural uniformity for molecular detection. The quasi-3D plasmonic nanowell array was composed of periodic hexagonal Au nanowells whose surface is densely covered with gold nanoparticles (Au NPs), separated by an ultrathin dielectric interlayer. The uniform array of the Au nanowells was fabricated by nanoimprint lithography and deposition of Au thin film. A self-assembled monolayer (SAM) of perfluorodecanethiol (PFDT) was coated on the Au surface, on which Au was further deposited. Interestingly, the PFDT-coated Au nanowells were fully covered with Au NPs with an ultra-high density of 375 &mu, m&minus, 2 rather than a smooth film due to the anti-wetting property of the low-energy surface. The plasmonic nanogaps formed among the high-density Au NPs led to a strong near-field enhancement via coupled localized surface plasmon resonance and produced a uniform surface-enhanced Raman spectroscopy (SERS) response with a small relative standard deviation of 5.3%. Importantly, the highly uniform nanostructure, featured by the nanoimprint lithography and 3D growth of densely-packed Au NPs, minimizes the spatial variation of Raman intensity, potentially providing quantitative analysis. Moreover, analyte molecules were highly concentrated and selectively deposited in nanowells when a water droplet containing the analyte was evaporated on the plasmonic substrate. The analyte formed a relatively thick overcoat in the nanowells near the triple line due to the coffee-ring effects. Combining 3D plasmonic nanowell substrates with molecular enrichments, highly sensitive detection of lactic acid was demonstrated. Given its combination of high sensitivity and signal uniformity, the quasi-3D plasmonic nanowell substrate is expected to provide a superior molecular detection platform for biosensing applications.

Plasmonic silver nanoparticles (AgNPs) arranged on polyimide (PI) nanopillars (AgNP/PI) were developed as a stable photothermal surface-enhanced Raman spectroscopy (SERS) platform. The AgNP/PI substrates were prepared via maskless plasma etching and metal deposition using an in situ vacuum sputtering system. The AgNP/PI with high periodicity and high areal density led to high sensitivity, with a SERS enhancement factor (EF) of 2.2 × 108, and excellent reproducibility (relative standard deviation of 7.6%), as evaluated using the high-precision Raman mapping technique. To support the high SERS EF and sensitivity, the near-field enhancements of the dense AgNP array were calculated using a finite-difference time domain method. Notably, compared with AgNPs on polyethylene terephthalate (PET) nanopillar substrates, the AgNPs formed on the thermally stable PI nanopillar substrates exhibited excellent photothermal stability under illumination with a high-powered laser. The PI and PET surface changes induced by photoinduced local heating were investigated in terms of the glass-transition temperature, coefficient of thermal expansion, and thermal conductivity. The new plasmonic SERS platform has strong potential for reliable molecular detection in practical sensing applications with high-powered laser illuminations.

We utilized a fast Raman spectral mapping technique for fast detection of bacterial pathogens. Three-dimensional (3D) plasmonic nanopillar arrays were fabricated using the nanolithography-free process consisting of maskless Ar plasma treatment of a polyethylene terephthalate substrate and subsequent metal deposition. Bacterial pathogens were immobilized on the positively charged poly(l-lysine)-coated 3D plasmonic substrate through electrostatic interactions. Then, the bacterial surfaces were selectively labeled with antibody-conjugated surface-enhanced Raman scattering (SERS) nanotags, and Raman mapping images were collected and statistically analyzed for quantitative analysis of bacteria. Salmonella typhimurium was selected as a model pathogen bacterium to confirm the efficacy of our SERS imaging technique. Minimum number of Raman mapping points with statistical reliability was determined to reduce assay time. It was possible to get a statistically reliable standard calibration curve for 529 pixels (laser spot with 60 μm interval), which required a total mapping time of 45 min to get a standard calibration curve for five different concentrations of bacteria in the 0 to 10

Developing a sensor that identifies and quantifies trace amounts of analyte molecules is crucially important for widespread applications, especially in the areas of chemical and biological detection. By non-invasively identifying the vibrational signatures of the target molecules, surface-enhanced Raman scattering (SERS) has been widely employed as a tool for molecular detection. Here, we report on the reproducible fabrication of wafer-scale dense SERS arrays and single-nanogap level near-field imaging of these dense arrays under ambient conditions. Plasmonic nanogaps densely populated the spaces among globular Ag nanoparticles with an areal density of 120 particles per μm2 upon application of a nanolithography-free simple process consisting of the Ar plasma treatment of a polyethylene terephthalate substrate and subsequent Ag sputter deposition. The compact nanogaps produced a high SERS enhancement factor of 3.3 × 107 and homogeneous (coefficient of variation of 8.1%) SERS response. The local near fields at these nanogaps were visualized using photo-induced force microscopy that simultaneously enabled near-field excitation and near-field force detection under ambient conditions. A high spatial resolution of 3.1 nm was achieved. Taken together, the generation of a large-area SERS array with dense plasmonic nanogaps and the subsequent single-nanogap level characterization of the local near field have profound implications in the nanoplasmonic imaging and sensing applications.

The ability to monitor metabolites related to cell communication is important for understanding bacterial physiology, identifying bacterial species in a cellular community, and providing insights into the stage of infection. Here, we developed a nanostructured electrochemical sensor comprising a 3D-redox-capacitor-coated electrode, which was prepared by grafting a redox capacitor molecule, catechol/o-quinone, at high density to a chitosan layer. It provided high sensitivity in detecting redox-active bacterial metabolites through a redox cycling reaction in the nanoconfined structures of the electrode. We used it to monitor the secretion of signalling-related metabolites at different growth phases of Pseudomonas aeruginosa. We also used the electrode to detect signalling-related metabolites in diverse simulated body fluids mixed with P. aeruginosa culture. It was able to provide information on bacterial communication and help in sensitive diagnostic monitoring of bacterial infection.

A bioinspired nanoflower structure was fabricated on microneedle (MN) to utilize as a surface-enhanced Raman scattering (SERS) sensor for intradermal sensing applications. On the surface of poly (lactic-co-glycolic acid) (PLGA) MN, polydopamine (PD) was coated as an interlayer, and the hydroxyapatite (HA) which is a flower-like-structured bone mineral was crystalized via a bio-mineralization in simulated body fluid (SBF). Au was deposited on the HA-coated microneedle (HA-MN) to generate SERS effect. Scanning electron microscopy (SEM) images showed the nanoflower structures of Au-coated HA-MN (Au-nanoflower on microneedle; NFMN) and dense Au nanoislands (AuILs) formed on each lath-like petal surface. After SERS characterization of the NFMN-SERS sensor, electromagnetic field distribution was investigated by finite-difference time-domain (FDTD) simulation to identify origin of the SERS effect. On the NFMN-SERS sensor, methylene blue (MB) was detected down to a concentration of 50 nM with uniform tip-to-tip signal intensity. Furthermore, the feasibility of the NFMN-SERS sensor for intradermal sensing was investigated using a skin phantom model. The MB molecule contained in the dermis region was detected down to 1 nM after simple insertion and withdrawal. The developed NFMN-SERS sensor is expected to be applied for various intradermal sensing, especially for chemical biomarkers in interstitial fluids of skin.

Enhancing light–matter interactions is essential to improving nanophotonic and optoelectronic device performance. In the present work, we developed a new design for 3D plasmonic nanostructures with enhanced near-field interactions among the plasmonic nanomaterials. The 3D plasmonic nanostructures consisted of multilayered bottom Ag/polydimethylsiloxane (PDMS) nanostructures, an alumina middle layer, and top Ag nanoparticles (NPs). High areal density PDMS nanoprotrusions were self-organized by a simple maskless plasma etching process. The conformal deposition of alumina using atomic layer deposition and Ag deposition produced 3D plasmonic nanostructures. These structures induced multiple near-field interactions between the ultrahigh-areal-density (1400 μm−2) top Ag NPs and the underlying Ag nanostructures, and among the top Ag NPs themselves. The high density of hot spots across the 3D space yielded highly efficient and widely tunable plasmonic responses across the entire visible range. The SERS signal enhancement measured at the 3D plasmonic nanostructures was 3.9 times the signal measured at the 2D multilayered structures and 48.0 times the signal measured at a Ag NP layer deposited onto a Si substrate. Finally, the 3D plasmonic nanostructures exhibited excellent uniformity with a variation of 6.8%, based on a microscale Raman mapping analysis. The excellent Raman signal uniformity can be attributed to the ultrahigh areal density of the Ag NPs and the uniform thickness of the alumina spacing layer.

Highly roughened Au-decorated 3D ZnO nano-structures were prepared using a combination of prism holographic lithography and atomic layer deposition techniques. Prism holographic lithography is a simple and rapid method for fabricating ordered 3D nanostructures using the optical interference effects of multiple beams derived from a specially designed prism. Highly ordered reproducible surface-enhanced Raman scattering (SERS) substrates are needed for the reliable calibration of target analyte concentrations. A high density of Au nanoparticles separated by nanoscale gaps was generated on the Au-coated ZnO inverse structures. The nanogaps may function as strong hot spots for highly sensitive SERS-based chemical/biological sensors. The optimized SERS intensity from the prepared Au-coated 3D ZnO inverse structures was 20 times the intensity obtained from an Au-coated flat glass control substrate. The surfaces could be reused after the photocatalytic degradation and removal of adsorbates in the presence of ZnO. The Au-coated 3D ZnO structures described here offer an alternative to traditional single-use SERS substrates.

9 pags. 5 figs., Plasmonic biosensors have demonstrated superior performance in detecting various biomolecules with high sensitivity through simple assays. Scaled-up, reproducible chip production with a high density of hotspots in a large area has been technically challenging, limiting the commercialization and clinical translation of these biosensors. A new fabrication method for 3D plasmonic nanostructures with a high density, large volume of hotspots and therefore inherently improved detection capabilities is developed. Specifically, Au nanoparticle-spiked Au nanopillar arrays are prepared by utilizing enhanced surface diffusion of adsorbed Au atoms on a slippery Au nanopillar arrays through a simple vacuum process. This process enables the direct formation of a high density of spherical Au nanoparticles on the 1 nm-thick dielectric coated Au nanopillar arrays without high-temperature annealing, which results in multiple plasmonic coupling, and thereby large effective volume of hotspots in 3D spaces. The plasmonic nanostructures show signal enhancements over 8.3 × 10-fold for surface-enhanced Raman spectroscopy and over 2.7 × 10-fold for plasmon-enhanced fluorescence. The 3D plasmonic chip is used to detect avian influenza-associated antibodies at 100 times higher sensitivity compared with unstructured Au substrates for plasmon-enhanced fluorescence detection. A surface-enhanced Raman scattering (SERS) sensor comprising silver nanowires (AgNWs) and genetically engineered M13 bacteriophages expressing a tryptophan-histidine-tryptophan (WHW) peptide sequence (BPWHW) was fabricated by simple mixing of BPWHW and AgNW solutions, followed by vacuum filtration onto a glass-fiber filter paper (GFFP) membrane. The AgNWs stacked on the GFFP formed a high density of SERS-active hot spots at the points of nanowire intersections, and the surface-coated BPWHW functioned as a bioreceptor for selective pesticide detection. The BPWHW-functionalized AgNW (BPWHW/AgNW) sensor was characterized by scanning electron microscopy, confocal scanning fluorescence microscopy, atomic force microscopy, and Fourier transform infrared spectroscopy. The Raman signal enhancement and the selective pesticide SERS detection properties of the BPWHW/AgNW sensor were investigated in the presence of control substrates such as wild-type M13 bacteriophage-decorated AgNWs (BPWT/AgNW) and undecorated AgNWs (AgNW). The BPWHW/AgNW sensor exhibited a significantly higher capture capability for pesticides, especially paraquat (PQ), than the control SERS substrates, and it also showed a relatively higher selectivity for PQ than for other bipyridylium pesticides such as diquat and difenzoquat. Furthermore, as a field application test, PQ was detected on the surface of PQ-pretreated apple peels, and the results demonstrated the feasibility of using a paper-based SERS substrate for on-site residual pesticide detection. The developed M13 bacteriophage-functionalized AgNW SERS sensor might be applicable for the detection of various pesticides and chemicals through modification of the M13 bacteriophage surface peptide sequence.

3D hybrid plasmonic nanomaterials are composed of 3D-stacked Ag nanowires and nanoparticles separated by a nanoscale-thick alumina interlayer. The 3D hybrid plasmonic nanostructures exhibit strong plasmonic coupling between the ultrahigh populations of plasmonic nanomaterials, overcoming the physical limitation of inefficient plasmonic coupling of the Ag nanowire stacks.

Highly uniform 3D ZnO hollow shell structures were prepared by combining prism holographic lithography (PHL) and atomic layer deposition (ALD). As a dense ZnO film was obtained by using the ALD process, no volume shrinkage occurred during the subsequent calcination to remove the sacrificial polymer template. No volume shrinkage during heat treatment is crucial for achieving excellent optical properties and mechanical stability of inverse photonic crystals (PCs).

8 pags., 6 figs., Positioning probe molecules at electromagnetic hot spots with nanometer precision is required to achieve highly sensitive and reproducible surface-enhanced Raman spectroscopy (SERS) analysis. In this article, molecular positioning at plasmonic nanogaps is reported using a high aspect ratio (HAR) plasmonic nanopillar array with a controlled surface energy. A large-area HAR plasmonic nanopillar array is generated using a nanolithography-free simple process involving Ar plasma treatment applied to a smooth polymer surface and the subsequent evaporation of metal onto the polymer nanopillars. The surface energy can be precisely controlled through the selective removal of an adsorbed self-assembled monolayer of low surface-energy molecules prepared on the plasmonic nanopillars. This process can be used to tune the surface energy and provide a superhydrophobic surface with a water contact angle of 165.8° on the one hand or a hydrophilic surface with a water contact angle of 40.0° on the other. We report new 3D hybrid plasmonic nanostructures exhibiting highly sensitive SERS-based sensing performance, utilizing efficient plasmonic light absorption and analyte-enrichment effect. The hybrid plasmonic nanostructures composed of 3D-stacked Ag NWs and NPs separated by a thin hydrophobic dielectric interlayer. A hydrophobic polydimethylsiloxane (PDMS) interlayer provides dielectric nanogap between Ag NWs and NPs, and analyte-enrichment effect due to the inhibition of drop spreading. The 3D hybrid PDMS-interlayered Ag nanostructures showed hydrophobicity with initial contact angle of 137.6°. Utilizing the analyte-enrichment strategy, the PDMS-interlayered Ag nanostructures exhibited an enhanced sensitivity of methylene blue molecules by a factor of 10 (limit of detection, LOD of 1.5 nM), compared to the alumina-separated 3D hybrid Ag nanostructures.

Advances in flexible optoelectronic devices have led to an increasing need for developing highly efficient, low-cost, flexible transparent conducting electrodes. Copper-based electrodes have been unattainable due to the relatively low optical transmission and poor oxidation resistance of copper. Here, we report the synthesis of a completely continuous, smooth copper ultra-thin film via limited copper oxidation with a trace amount of oxygen. The weakly oxidized copper thin film sandwiched between zinc oxide films exhibits good optoelectrical performance (an average transmittance of 83% over the visible spectral range of 400–800 nm and a sheet resistance of 9 Ω sq−1) and strong oxidation resistance. These values surpass those previously reported for copper-based electrodes; further, the record power conversion efficiency of 7.5% makes it clear that the use of an oxidized copper-based transparent electrode on a polymer substrate can provide an effective solution for the fabrication of flexible organic solar cells., Light enters and exits optoelectronic devices through transparent conductive electrodes, which are one of their most expensive components. Here, the authors develop stable transparent conductive electrodes based on copper and oxide layers that lead to efficient flexible organic solar cells.
