42. Solvent structure effects in the macroscopic theory of van der Waals forces.

Abstract

The theory of van der Waals forces is generalized in order to include the effects of solvent structure by explicitly dealing with the spatial correlations between the solvent molecules. This is achieved within the framework of nonlocal dielectric response theory, with the specular reflection approximation to apply the nonlocal response of the structured solvent in the bulk to give the response of such solvent intercalated between the interacting macroscopic surfaces. A closed form expression for the zero-order contribution to the van der Waals free energy is derived for two semi-infinite dielectric regions (characterized by a standard, local dielectric response and fixed dielectric constant), interacting across a layer of a structured solvent described by a nonlocal dielectric response function. The nonlocal character of the latter gives rise to several quantitatively and qualitatively new features of the dispersion interaction; the most important ones are similar in consequences to the effects of spatial variation of the Hamaker coefficient, as seen from the particular case of solvents with an exponentially decaying spatial intersolvent correlations. 43. Nanostructures in n-Octanol Equilibrated with Additives and/or Water

Abstract

Fluid fatty alcohols are believed to be nanostructured but broadly amorphous (i.e., noncrystalline) fluids and solvents, including the most popular fatty tissue mimetic, hydrated n-octanol (i.e., hydro-octanol). To check this premise, we studied dry octanol and hydro-octanol as a model of relatively short fluid n-alkanols with small-angle X-ray scattering (SAXS). We also combined this alkanol with the matching alkane (i.e., octane) and with a common anti-inflammatory pain killer (ketoprofen). This revealed that (hydro-)octanol and arguably any other short fatty alcohol form a mesophase. Its basic structural motif are regularly packed polar nanoclusters, reflected in the inner peak in the SAXS diffractogram of (hydro-)octanol and other fluid n-alkanols. The nanoclusters arguably resemble tiny, (inverse) hydrated bilayer fragments, located on a thermally smeared para-crystalline lattice. Additives to hydro-octanol can change the nanoclusters only moderately, if at all. For example, octane and the drug ketoprofen added to hydro-octanol enlarge the nanoclusters only little because of the mixture’s packing frustration. To associate with and to bring more water into hydro-octanol, an additive must hence transform the nanoclusters: it expands them into irregularly distributed aqueous lacunae that form a proto-microemulsion, reflected in the previously unknown Guinier’s SAXS signal. A “weak” (i.e., a weakly polar or nonpolar) additive can moreover create only size-limited lacunae. Coexistence of nanoclusters and lacunae as well as size variability of the latter in hydro-octanol subvert the concept of octanol–water partition coefficient, which relies on the studied compartment homogeneity. In turn, it opens new possibilities for interfacial catalysis. Reinterpreting “octanol–water partition coefficient” data in terms of octanol–water association or binding constant(s) could furthermore diminish the variability of molecular lipophilicity description and pave the ground toward a more precise theoretical quantification and prediction of molecular properties.

Abstract

Die Struktur und der Entstehungsprozess multilamellarer Komplexe aus kationischen Lipidvesikeln und DNS wurde mit Hilfe der Cryo-Transmissionselektronenmikroskopie (Cryo-TEM) und der energiedispersiven Röntgenreflektometrie untersucht. Mehrere Schritte der Komplexbildung wurden identifiziert. Hierzu gehören die Adsorption und Fusion von Vesikeln und ein Vorgang, bei dem ein adsorbiertes Vesikel aufbricht und seine Lipidschicht auf der Substratoberfläche abrollt. Als Schlussfolgerung aus den Cryo-TEM-Untersuchungen wird ein aus diesen Schritten zusammengefügter Mechanismus vorgeschlagen, demzufolge die Komplexbildung zwei miteinander verknüpften Pfaden folgt. Die Entstehung des multilamellaren Komplexes ist nach diesem Mechanismus ein sequentieller Prozess, bei dem ausgehend von einem einzelnen Substratvesikel eine Schicht nach der anderen absorbiert. Die röntgenreflektometrischen Messungen bestätigen und ergänzen die Schlussfolgerungen aus den Cryo-TEM-Untersuchungen. Die Multilamellen entstehen in drei Schritten: ein Vesikel kommt durch Kollision in Kontakt mit dem Substrat; das Vesikel adsorbiert und verformt sich dabei; es bricht auf und rollt seine Lipiddoppelschicht über das Substrat. Weitere Schichten adsorbieren auf gleiche Weise. Dabei übernimmt die DNS die Rolle eines molekularen Klebstoffs. Die DNS-Adsorption kann mit einem Diffusionsmodell, die Lamellenbildung mit einem irreversiblen Multischicht-Adsorptionsmodell beschrieben werden. Schließlich wurde auch die Rolle der Ionen im Medium betrachtet. Further layers form the same way. DNA acts as a molecular glue. The DNA adsorption can be explained using a diffusion model. To describe the formation of the lamellae, a irreversible multilayer adsorption model was applied. Finally the effect of bulk ions on the complex formation was studied. Surprisingly, the concentration of EDTA in the solution greatly affected complex formation.
