In this work, we examine the use of environment-sensitive fluorescent dyes in fluorescence lifetime imaging microscopy (FLIM) biosensors. We screened merocyanine dyes to find an optimal combination of environment-induced lifetime changes, photostability, and brightness at wavelengths suitable for live-cell imaging. FLIM was used to monitor a biosensor reporting conformational changes of endogenous Cdc42 in living cells. The ability to quantify activity using phasor analysis of a single fluorophore (e.g., rather than ratio imaging) eliminated potential artifacts. We leveraged these properties to determine specific concentrations of activated Cdc42 across the cell.

Cutaneous basal cell carcinoma in situ is a recently proposed subtype of this skin cancer. It is characterized by either restriction of the tumor cells within the epidermis or the presence of tumor cells contiguous with the overlying epidermis that extend into the underlying dermis, or both. Importantly, cancer invasion-demonstrated by non-contiguous aggregates of basaloid tumor cells in the dermis-is not a feature of in situ basal cell carcinoma of the skin. A 63-year-old woman with cutaneous basal cell carcinoma in situ-superficial type that presented as an erythematous scaly plaque on her abdomen and a 61-year-old man with a cutaneous basal cell carcinoma in situ-fibroepithelioma type that presented as a flesh-colored smooth exophytic nodule on his back are reported. The characteristics of in situ basal cell carcinoma of the skin in these individuals are summarized. In conclusion, similar to other cutaneous malignant neoplasms-such as squamous cell carcinoma, malignant melanoma, and Merkel cell carcinoma-basal cell carcinoma of the skin can also present as an in situ cancer.

Background: The heterogeneity of high-grade serous ovarian carcinoma (HGSOC) has hindered the clinical treatment, and our current study aims to characterize the change in tumor microenvironment (TME) with the progression of HGSOC via single cell RNA sequencing (scRNA-seq). Methods: The single-cell landscape in HGSOC was downloaded from the dataset GSE184880, which included 7 HGSOC and 5 normal samples and then applied for the filtering and annotation of cell clusters. The differentially expressed marker genes in these clusters were analyzed via "FindAllMarker" function in Seurat package and the functional enrichment analyses were implemented using clusterProflier package. Finally, the CellChat package was applied for the cell–cell communication analysis. Cellular experimental were determined Real-time Reverse Transcription Polymerase Chain Reaction (RT-qPCR). Results: 45,448 single cells were categorized into 10 cell clusters. The proportion of NK/T cells (49.5%), epithelial cells (15.3%) and myeloid cells (14%) was higher in the HGSOC samples. The heterogeneity and different enriched pathways of epithelial cells have been revealed with the progression of HGSOC from early to late stage, concurrent with the reduced activity of cytotoxic NK/T cells and the decreased capabilities of recruiting immune cells and presenting antigens in macrophages. Besides, the cell–cell communication analysis has revealed a strong communication of CXCL and CCL signal between M1 macrophages and cytotoxic NK/T cells in early stage of HGSOC. Moreover, RT-qPCR indicated that CCL4/5 and CCR1/5 levels were upregulated in tumor cell SK-OV-3. Conclusion: The investigation using scRNA-seq has depicted the change in cytotoxic NK/T cells, epithelial cells and myeloid cells of the TME of HGSOC, which may provide another insight into the specific mechanisms underlying the progression of HGSOC. This paper is an attempt to generalize the nitrogen isotope information of the molecules that make up the cell from our point of view. Nitrogen in the cell exists as 20 proteinaceous amino acids, nucleobases, hemes, chlorophylls, and others, and their composition is similar among organisms. Based on a physiologically simple autonomous system that maintains a balance between inputs and outputs, it is theoretically predicted that 15N is distributed to each cellular compound with a certain regularity, and thus a specific relationship in nitrogen isotopic ratios among compounds. Previous studies essentially confirm this. The nitrogen isotopic ratio of compound i constituting the cell can be generalized as δ15Ni = δ15Nplant + Δi (TP − 1) + γi, where TP represents the trophic position, and Δi and γi are the trophic discrimination factor and the intracellular 15N distribution of compound i, respectively. Knowing reliable values of Δi and γi will help us to better understand nitrogen dynamics in the biosphere and advance our understanding of the picture of the Earth's environment through biogeochemistry. Central metabolism is organised through high‐flux, Nicotinamide Adenine Dinucleotide (NAD+/NADH) and NADP+/NADPH systems operating at near equilibrium. As oxygen is indispensable for aerobic organisms, these systems are also linked to the levels of reactive oxygen species, such as H2O2, and through H2O2 to the regulation of macromolecular structures and activities, via kinetically controlled sulphur switches in the redox proteome. Dynamic changes in H2O2 production, scavenging and transport, associated with development, growth and responses to the environment are, therefore, linked to the redox state of the cell and regulate cellular function. These basic principles form the 'redox code' of cells and were first defined by D. P. Jones and H. Sies in 2015. Here, we apply these principles to plants in which recent studies have shown that they can also explain cell‐to‐cell and even plant‐to‐plant signalling processes. The redox code is, therefore, an integral part of biological systems and can be used to explain multiple processes in plants at the subcellular, cellular, tissue, whole organism and perhaps even community and ecosystem levels. As the environmental conditions on our planet are worsening due to global warming, climate change and increased pollution levels, new studies are needed applying the redox code of plants to these changes. The aqueous phase mass transfer coefficient (Kaq) for ethanol transfer to the organic phase during fermentation has been successfully correlated with various system parameters in the presence of suspended yeast flocs under agitated conditions. In particular, a semi-empirical correlation of Kaq with the agitation speed of the stirrer, the void space in the broth, and the surface area of dead cells has been developed. In general, it has been found that Kaq increases with the agitation speed of the stirrer and the ratio of the void volume to the dead cell surface area. Kaq increases marginally though with higher incubation time at a given agitation speed of stirrer. The correlation is valid within a period of 12–30 h of incubation time. The agitation speed range is 200–300 rpm, and the ratio of void volume to dead cell surface area ratio ranged from 3.86 × 10−4 to 1.69 × 10−3 m. The fermentation time required was only 25–30 h compared to conventional fermentation systems, where it takes a minimum of 40–48 h. The organic phase mass transfer coefficient (Korg) is 300–500 times higher than Kaq. This is due to the drag of the yeast floc surfaces to the ethanol transferring eddies from the bulk to the interface. [ABSTRACT FROM AUTHOR]

Exploring the chemical content of individual cells not only reveals underlying cell‐to‐cell chemical heterogeneity but is also a key component in understanding how cells combine to form emergent properties of cellular networks and tissues. Recent technological advances in many analytical techniques including mass spectrometry (MS) have improved instrumental limits of detection and laser/ion probe dimensions, allowing the analysis of micron and submicron sized areas. In the case of MS, these improvements combined with MS's broad analyte detection capabilities have enabled the rise of single‐cell and single‐organelle chemical characterization. As the chemical coverage and throughput of single‐cell measurements increase, more advanced statistical and data analysis methods have aided in data visualization and interpretation. This review focuses on secondary ion MS and matrix‐assisted laser desorption/ionization MS approaches for single‐cell and single‐organelle characterization, which is followed by advances in mass spectral data visualization and analysis. Cladophora is commonly found in marine and freshwater around the globe and provides productivity for littoral zone microorganisms and invertebrates. The eutrophication of the water body has led to the abnormal proliferation of Cladophora in some water, often in river coast channel outbreaks. Even under the nutritional deficiency systems, such as the central trunk canal of the South-to-North Water Diversion in China, Cladophora's blooms affect water quality and seriously jeopardize human health. Thus, the structural characteristics of Cladophora cells and spores, the factors affecting the growth of Cladophora cells, and the mechanisms of damage and destruction of Cladophora cells and spores were investigated. Cladophora cells are cylindrical with very thick cell walls. The inner layer of the spore wall is a thin membrane which contains the nucleus of the spore. The growth and spreading of Cladophora cells are affected by various environmental factors such as light, temperature, water depth, water level, nutrient salts, pH, etc. Some physical treatment measures, such as ultrasounds, would destroy the cell walls and membranes of Cladophora by its high-intensity mechanical action. Chemicals and aquatic plant measures can destroy Cladophora cells' photosynthesis system, antioxidant enzyme systems, proteins, and ultrastructure. Based on the mechanisms for these cell damage and destruction, a combination of measures that are likely to inhibit the growth of Cladophora cells effectively was suggested. Furthermore, the damaged cells of Cladophora could provide some environmental benefits. Optical tweezers are a means to manipulate objects with light. Optical tweezers can obtain a nanometer space, piconewton force, and a millisecond resolution; thus, they are excellently suited for studying biological processes from the single-cell to the single-molecule level. Optical tweezers can screen and manipulate tumor cells, study single molecule, and monitor cancer treatments. Thus, optical tweezers will promote the progress of cancer research and treatments.

The article presents a case study of an 82-year-old man who experienced alternating blurred vision in both eyes, initially attributed to corneal deposits but later diagnosed with non-arteritic anterior ischemic optic neuropathy (NA-AION). It states that the case underscores the variability of giant cell arteritis (GCA) presentations and the critical role of ultrasound in confirming diagnoses without typical symptoms.

This research investigated the effect of cadmium on the tissue and cell of kidney of the turtle Mauremys reevesii. Twenty turtles were injected with cadmium at 0, 7.5, 15, 30 mg/kg separately and five turtles were taken in each group at two weeks after exposure. Kidneys were immediately excised and macroscopic pathological changes were observed, then the kidneys were fixed in 4% paraformaldehyde for histopathological examination and fixed in 2.5% (v/v) glutaraldehyde for examination of ultra-structure. The tissues of kidney presented varying degrees of histopathological lesions in cadmium treated turtles by a dose-dependent manner under the light microscope. Under transmission electron microscope, renal tubules cells presented varying degrees of dose-dependent lesions. The results indicated that cadmium can cause cell damages to the kidney, in particular to the mitochondria. Mitochondria can be used as one biomarker in the monitoring of cadmium pollution and its quantitative risk assessments. [ABSTRACT FROM AUTHOR]

The problem of getting rid of expensive and metal-intensive current transformers has been declared by CIGRE as strategically important for the electric power industry. However, almost all traditional current protections receive information from measuring current transformers. In this work, a resource-saving reed switch overcurrent protection without current transformers is suggested, which can be used as an alternative to traditional current protections for 6–10 kV electrical installations connected to a switchgear cell. The protection is designed following the novel method we have developed based on inductance coils. Inductance coils measure the electromotive force under different operation modes of an electrical installation and at different points inside the switchgear cell it is connected to; the EMF values are recalculated in the values of magnetic induction, and reed switches are mounted instead of inductance coils at the points where the magnetic induction is maximal. Moreover, these values are sufficient to detect phase-to-phase short circuits in the electrical installation. The dependence of the induction value on the position of an inductance coil inside the cell is derived with the use of the simplest formula of the Biot–Savart law. The results can be used at large and small industrial enterprises, electric power stations, and substations of plants; they can be interesting for the scientific community because they help to solve the topical problem of the electric power industry. Ernst Brücke's 1861 essay Die Elementarorganismen has often been cited as a watershed in the history of physiology as well as in the history of cell theory. In its time it was widely read as a reform of animal cell theory, shifting the concept of the cell away from Schleiden and Schwann's original cell schema of a membranous vesicle with a nucleus, and towards the protoplasm theory that had developed in botany, centered on the cell's living contents. It was also notorious for its arguments against the necessity of both the nucleus and the cell membrane. Brücke's essay was not only an intervention into cell theory: historians can read it as a continuation of debates on the nature of the organism and theories of organization, and as an epistemological meditation on the microscope. Cells evolved some 4 billion years ago, and since then the integrity of the structural and functional continuity of cellular life has been maintained via highly conserved and ancient processes of cell reproduction and division. The plasma membrane as well as all the cytoplasmic structures are reproduced and inherited uninterruptedly by each of the two daughter cells resulting from every cell division. Although our understanding of the evolutionary emergence of the very first cells is obscured by the extremely long timeline since that revolutionary event, the generally accepted position is that the de novo formation of cells is not possible; all present cells are products of other prior cells. This essential biological principle was first discovered by Robert Remak and then effectively coined as Omnis Cellula e Cellula (every cell of the cell) by Rudolf Virchow: all currently living cells have direct structural and functional connections to the very first cells. Based on our previous theoretical analysis, all cells are endowed with individual sentient cognition that guides their individual agency, behaviour and evolution. There is a vital consequence of this new sentient and cognitive view of cells: when cells assemble as functional tissue ecologies and organs within multicellular organisms, including plants, animals and humans, these cellular aggregates display derivative versions of aggregate tissue‐ and organ‐specific sentience and consciousness. This innovative view of the evolution and physiology of all currently living organisms supports a singular principle: all organismal physiology is based on cellular physiology that extends from unicellular roots. Reactive oxygen species (ROS) involving superoxide anion, hydrogen peroxide and hydroxyl radical play important role in human health. ROS are known to be the markers of oxidative stress associated with different pathologies including neurodegenerative and cardiovascular diseases, as well as cancer. Accordingly, ROS level detection in biological systems is an essential problem for biomedical and analytical research. Electrochemical methods seem to have promising prospects in ROS determination due to their high sensitivity, rapidity, and simple equipment. This review demonstrates application of modern electrochemical sensors for ROS detection in biological objects (e.g., cell lines and body fluids) over a decade between 2011 and 2021. Particular attention is paid to sensors materials and various types of modifiers for ROS selective detection. The ultrasound, as one of the non-invasive intervention or regulation measures in mechanotherapy, has attracted much attention. The methods, effects and action mechanisms of 4 types of ultrasounds for tissues/ cells, namely, the high-frequency high-intensity ultrasound, the low-frequency high-intensity ultrasound, the low-frequency low-intensity ultrasound and the high-frequency low-intensity ultrasound were reviewed, and the emerging ultrasound therapeutic instrumentations were introduced. Then the future development trends of ultrasonic mechanotherapeutics were discussed. This review helps to promote the technical progress and clinical application of ultrasound mechanotherapy in tumor treatment, neurologic diseases (such as Alzheimer's disease and Parkinson's disease) treatment and so on. For many biomedical applications, laser-assisted methods are essential to enhance the three-dimensional (3D) resolution of a light microscope. In this report, we review possibilities to improve the 3D imaging potential by axial tomography. This method allows us to rotate the object in a microscope into the best perspective required for imaging. Furthermore, images recorded under variable angles can be combined to one image with isotropic resolution. With the expansion of ICP-MS application into the field of bioanalysis, there is an urgent need for novel element tags today. Here, we report the design of a dual-element Ir-Eu tag, opening the door to simultaneous fluorescent imaging and ICP-MS quantification. The ratio of 153Eu/193Ir may serve as a precision control of the labeling process, allowing internal validation of the quantitative results obtained. As for SIRPα and its host cell analysis exemplified here, the Ir-Eu tag demonstrated superior figures of ICP-MS quantification with the LOD (3σ) down to 0.5 (153Eu) and 1.1 (193Ir) pM SIRPα and 220 (153Eu) and 830 (193Ir) RAW264.7 cells more than 130 times more sensitive compared with the LOD (3σ) of 65.2 pM SIRPα at 612 nm using fluorometry. Not limited to these demonstrations, we believe that the design ideas of the dual Ir-Eu tags should be applicable to various cases of bioanalysis when dual optical profiling and ICP-MS quantification are indispensable. The use of surface acoustic waves (SAW) in cell biology has gained high attention in the past years. Previous works show that SAW treatment of artificial wounds in vitro can accelerate wound healing by up to +135%. However, little is known about the mechanobiology behind these effects, and a stimulation has only been proven for continuous SAW signals so far. We here show that the stimulation efficacy observed in previous studies is preserved for pulsed stimuli applied to Madin-Darby canine kidney (MDCK-II) cells in wound healing assays on SAW chips at a resonance frequency f SAW = 160 MHz. Moreover, for a reproducible and reliable image analysis, we present the SegFormer-based deep learning algorithm Neural Cell Edge Detector for the cell edge segmentation and image binarization, that allows an automated determination of the stimulation efficacy. With these tools, we explore a wider range of applicable SAW intensities up to P in = 21 dBm (128 mW) with a maximum stimulation efficacy of E = 201%. We show that the order of magnitude of the stimulation effect is reproducible under the variation of SAW signal characteristics power P in, duty cycle D and pulse width τ, while the mean energy over time is constant. Below a distinct pulse duration limit τ = 100 ms, no direct stimulation effect was observed. From a mechanobiological point of view, these findings and time scales could narrow down the potentially triggered cellular mechanisms during the stimulation. [ABSTRACT FROM AUTHOR]

The aim of this study is to design various instructional materials to teach biological systems in a 'Systems in Our Body' unit to 6th grade students with visual impairment (SVI) and to evaluate the effectiveness of these materials. In the study, a design-based research model was used. In the first stage, individual learning needs of SVI and their understanding of biological structures in the 'Systems in Our Body' unit were determined. In the second stage, instructional materials matching the individual needs were designed and implemented. In the last stage, the usefulness of these materials and their effects on learning were explored. Simple Summary: Colostrum is the "starter kit" for every calf, containing nutrients, bioactive components, cells, and microorganisms that provide energy and protection against pathogens and contribute to the maturation and development of the gastrointestinal tract. Components such as fatty acids, whey proteins, oligosaccharides, immune cells, and microorganisms are present in colostrum and are essential for the health and well-being of the newborn calf, so its importance should not be underestimated. In this review, the variability of these components, as well as their impact on selected aspects of the newborn calf's physiology, are discussed. Colostrum contains macro- and micronutrients necessary to meet the nutritional and energy requirements of the neonatal calf, bioactive components that intervene in several physiological aspects, and cells and microorganisms that modulate the calf's immune system and gut microbiome. Colostrum is sometimes mistaken as transition milk, which, although more nutritive than whole milk, has a distinct biochemical composition. Furthermore, most research about colostrum quality and colostrum management focuses on the transfer of maternal IgG to the newborn calf. The remaining components of colostrum and transition milk have not received the same attention, despite their importance to the newborn animal. In this narrative review, a large body of literature on the components of bovine colostrum was reviewed. The variability of these components was summarized, emphasizing specific components that warrant deeper exploration. The building integrated photovoltaic applications (BIPV) are mostly affected by partial shading, which results in a significant decrease in energy efficiency and leads to the apparition of multiple power peaks on the I-V characteristic curve of photovoltaic generators. In mitigating this problem, sophisticated maximum power point tracking techniques (MPPT) are used to track the global peak among the other locals. These techniques do not take into account the overheating of the shaded cells given that, under some critical situations, shaded cell performances may degrade, thus affecting the lifespan of the photovoltaic modules. In this regard, the present work aims to investigate in which peak on the I-V characteristic curve the PV module should operate appropriately with less overheating risk. For that, the shaded cells' temperatures and currents at different operating points were monitored. The results show that the worst-case scenario occurs for small shadow ratios (in the studied case, the shadow ratio 'sr' = 25%) where the temperature of the shaded cell reached approximately three times (300%) the temperature of non-shaded cells. This temperature decreases as the operating point is moved from the short circuit current towards the open-circuit voltage; as a result, the PV module works safely with minimum thermal stress for the maximum power peak near the open-circuit voltage. Renewable energy is in very much demand in current time due to its many favorable environment effect. Solar energy is one of the frontline sources of renewable energy. Solar photovoltaic converts the solar light into electricity. The performance of the solar photovoltaic depends on various parameters and one of such parameters is shading behavior. Further, the shading pattern and the progress of the shading are also important for predicting the solar performance. The interconnections of solar cells also impact the performance of solar photovoltaic. Therefore, a 4 × 4 module with various interconnections such as series-parallel (SP), total cross tied (TCT), bridge-link (BL), honeycomb (HC) and triple tied (TT) are studied under the row and column wise shading pattern. Shading pattern on a 4 × 4 module is increased cell by cell in horizontal direction, both from left to right and right to left. In the similar fashion, the shading pattern is varied from top to bottom or vice-versa in vertical direction. This shading pattern progresses from first row to the last and first column to last column in a progressive fashion. The power-voltage and current-voltage characteristics of solar photovoltaic are investigated for the mentioned shading patterns using various reconfigurations. The power output is identical when all cells in a row or column are shaded. On the other hand, if only few cells are shaded on row or column then the power output with TCT connection is highest among all connections. Further, the power output is for complete column shading is much higher than complete row shading for all the connections. Despite significant progress in human medicine, certain diseases remain challenging to promptly diagnose and treat. Hence, the imperative lies in the development of more exhaustive criteria and tools. Tissue and cellular mechanics exhibit distinctive traits in both normal and pathological states, suggesting that "force" represents a promising and distinctive target for disease diagnosis and treatment. Atomic force microscopy (AFM) holds great promise as a prospective clinical medical device due to its capability to concurrently assess surface morphology and mechanical characteristics of biological specimens within a physiological setting. This review presents a comprehensive examination of the operational principles of AFM and diverse mechanical models, focusing on its applications in investigating tissue and cellular mechanics associated with prevalent diseases. The findings from these studies lay a solid groundwork for potential clinical implementations of AFM. Lung cancer, a prevalent and life-threatening condition, necessitates early detection for effective intervention. Considering the recent advancements in deep learning techniques, particularly in medical image analysis, which offer unparalleled accuracy and efficiency, in this paper, we propose a method for the automated identification of cancerous cells in lung tissue images. We explore various deep learning architectures with the objective of identifying the most effective one based on both quantitative and qualitative assessments. In particular, we assess qualitative outcomes by incorporating the concept of prediction explainability, enabling the visualization of areas within tissue images deemed relevant to the presence of lung cancer by the model. The experimental analysis, conducted on a dataset comprising 15,000 lung tissue images, demonstrates the effectiveness of our proposed method, yielding an accuracy rate of 0.99. Summary: Eukaryotic initiation factor 2B (eIF2B) controls translation initiation by recycling inactive eIF2-GDP to active eIF2-GTP. Under cellular stress, the integrated stress response (ISR) is activated inhibiting eIF2B activity resulting in the translation attenuation and reprogramming of gene expression to overcome the stress. The ISR can dictate cell fate wherein chronic activation has pathological outcomes. Vanishing white matter disease (VWMD) is a chronic ISR-related disorder with mutations in eIF2B targeting astrocyte and oligodendrocyte cells. Regulation of eIF2B localization (eIF2B bodies) has been implicated in the ISR. We present evidence that neuronal and glial cell types possess distinct patterns of eIF2B bodies which change in a manner correlating to acute and chronic ISR activation. We also demonstrate that while neural and glial cell types respond similarly to the acute induction of the ISR a chronic ISR exerts cell-type specific differences. These findings provide key insights into neural cell responses and adaptation to cellular stress.

Tunneling nanotubes (TNTs) that connect cells have been recognized as a pathway for long-range intercellular transport of diverse cargoes, including viruses, lysosomes or other organelles, Ca2+ and electrical signals. TNTs can initially be formed from thin finger-like actin assembly-driven protrusions or cell contacts and dislodgment. However, it remains unclear whether the mechanical properties of TNTs formed by these two mechanisms are the same. Here, we developed novel microoperation methods to investigate the mechanical properties of TNTs in HEK293 cells, in which the TNTs form from thin finger-like actin assembly-driven protrusions and C2C12 cells, in which the TNTs form through contact and cell dislodgment. We found that TNTs formed by the two mechanisms represent elastic elements with similar tensile strength. In both the HEK and C2C12 cells, the tensile strength of TNTs exhibited a distinct size dependence on their lengths and diameters. Disturbing the cytoskeleton or removing extracellular Ca2+ also changed their tensile strength. In addition, the stiffening of the extracellular matrix (ECM) enhanced the length, diameter and tensile strength of TNTs both in both HEK and C2C12 cells. Finally, a theoretical model was established to reveal the changes in the TNT's mechanical properties with its length, diameter and individual tunneling nanotubes (iTNT) number. This work not only gains insights into the properties of TNTs but also helps understand the dynamics of various cells.

In the ever-increasing quest for alternative energy sources, hydrogen emerged as a promising green option, but efficient and economical production and management have been the primary constraints. Converting wastewater into H2 and other forms of energy attracted significant attention in terms of simultaneously and sustainably managing both the wastewater and the energy generation problems. Microbial Electrolysis Cells (MEC) evolved recently as promising options for converting wastewater into H2 and electricity but with serious constraints on scalability. The current research aims to explore design and manufacturing solutions to build structurally strong and electrochemically effective electrodes that can also lead to scalable MEC. Two designs based on the interdigitated and spiral electrode architectures are proposed and evaluated. The added design freedom with additive manufacturing by selective laser melting of specific alloys of choice is effectively utilised in physically prototyping the interdigitated and spiral electrode forms designed with controlled porosity constraints. Microstructural, electrochemical, and cell performance characterisations led to the understanding that the spiral electrode configuration with polypyrrole-coated stainless steel 316L anode is a promising design option for both longitudinal and lateral scale-up of the MEC.

Summary: Executive functions, particularly visual working memory, depend on the prefrontal cortex (PFC). Phase-amplitude coupling (PAC) has been proposed as a measure of synchronized brain oscillations. To study the neural correlates of working memory in cross-frequency interactions, local field potential (LFP) recordings were made in the PFC of two macaque monkeys. PAC analysis revealed that the delta band (1–5 Hz) phase modulated the alpha-beta band (8–33 Hz) amplitude throughout task epochs, in both the pre- and post-training stages. The elevation of δ-αβ PAC in the fixation period during post-training was a signature of task learning. Interestingly, the δ-αβ PAC was not enhanced in error trials compared to correct trials, and the subject’s performance was strictly dependent on the orchestration of the delta phase. Furthermore, contrary to the dorsoventral functional specialization of PFC, spatial and shape stimuli induced the same pattern of PAC in PFC subdivisions.

Summary: The therapeutic potential of nitric oxide (NO) has been receiving increasing interest, but achieving controlled release under physiological conditions remains challenging. Herein, we report a colorimetric and fluorescence responsive naphthalimide-based amphiphilic N-nitroso-based NO donor (Nap-NO) and its NO-releasing behavior. Nap-NO was incorporated into phospholipid nanovesicles to make it biocompatible and water-soluble. Light-induced NO-releasing behavior and emission changes were monitored via UV-vis, colorimetric detection, IR (Infrared) spectroscopy studies, and Griess assay. The Nap-NO donor within the 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC)-cholesterol vesicles exhibited a slower release rate, with a significantly extended half-life as compared to the only DOPC vesicles. Incorporating the Nap-NO into alginate hydrogel beads enables a simple, visual detection of NO release through color and emission changes. Bioimaging experiments within the HCT cell line reveal the use of the new NO donor for fluorescent bio-imaging and clearly illustrate their proficiency in killing cancer cells upon NO delivery in the presence of light.

Sickle cell disease (SCD) is characterized by microvascular occlusion which leads to multiorgan damage, including left ventricular diastolic dysfunction. Left ventricular diastolic dysfunction has been shown to be an independent risk factor for death in SCD patients. Left atrial dilation (LAD) has been used as a surrogate marker for identification of left ventricular diastolic dysfunction. Investigate the association of LAD, as determined by echocardiography, with increased disease burden in SCD as reflected by increased emergency department (ED) utilization, increased hemolysis markers, and worsening anemia. A retrospective cohort study of patients from a single university hospital were selected from a national registry. Age, sickle cell phenotype, echocardiogram findings, ED utilization, baseline hemoglobin, and lab values needed for calculation of hemolytic index were recorded for each patient. Patients were then stratified into two distinct groups based on the presence or absence of LAD to compare ED utilization, baseline hemoglobin and hemolytic index between the two groups. 129 patients met the criteria for inclusion with 88 having normal left atrial volume and 41 with LAD. There was a higher percentage of high ED utilizers in the LAD group compared to the normal left atrial volume group [34% vs. 17%, p = 0.03]. Average hemoglobin was lower in the LAD group compared with the normal left atrial volume group [mean 8.57 g/dL vs. 9.47 g/dL, p = 0.011]. The mean hemolytic index was higher in the LAD group when compared with the normal left atrial volume group [0.44 vs. −0.21, p < 0.001]. For many biomedical applications, laser-assisted methods are essential to enhance the three-dimensional (3D) resolution of a light microscope. In this report, we review possibilities to improve the 3D imaging potential by axial tomography. This method allows us to rotate the object in a microscope into the best perspective required for imaging. Furthermore, images recorded under variable angles can be combined to one image with isotropic resolution. After a brief review of the technical state of the art, we show some biomedical applications, and discuss future perspectives for Deep View Microscopy and Molecular Imaging.

Abstract This study was on the use of the market definition paradigm (MDP) to track ageing and temperature effects in lithium–ion cells. This was with a view to using the technique to obtain a sequence of equilibrium voltages from readily available datasets in order to profile the effects of ageing and temperature on cells and batteries. The method employed involved using the MDP with its capability to obtain a sequence of equilibrium voltages for lithium-ion cells. This approach integrated radio incidence with radio geometry, transmission and emergence in a simplified form of the cell’s equilibrium voltage (and amperage). A standard dataset was obtained from the centre for advanced life cycle engineering repository. The data were processed and analysed using Coulomb counting, charging and discharging energy comparison methods on Python 3.8 programming tool and LibreOffice spreadsheet software. Results obtained show a close tracking of ageing and temperature phenomena in the cells studied. A respective maximum and minimum equilibrium voltages of 3.23 V and 3.10 V over two thousand (2000) cycles were similarly obtained for ageing and temperature investigations. The equilibrium voltage shows a downward trend as the battery ages and is more reliable for studies on these cells than the open circuit voltage traditionally used to track phenomena in such cells. In conclusion, typical lithium-ion cells can be classified at begin-of-life using the equilibrium voltage and useful predictions made with respect to end-of-life. This approach is relatively inexpensive, requiring fewer data points and low-cost hardware and extensible to online applications. Graphical Abstract

Recently, we have reported that biogenic silver/silver chloride nanoparticles from Asparagus racemosus (A. racemosus-AgCl-NPs) and Kaempferia rotunda (K. rotunda-Ag/AgCl-NPs) inhibited different cancer cells by inducing apoptosis and several genes alteration. Here for the first time, we assessed the effects of these two nanoparticles on human lung (A549) and hepatocellular (SMMC-7721) carcinoma cell lines. A. racemosus-AgCl-NPs and K. rotunda-Ag/AgCl-NPs inhibited A549 cell growth with IC50 values of 22.7 and 59.7 μg/ml and the calculated IC50 values for SMMC-7721 cell were 89.3 and 126.3 μg/ml, respectively. A. racemosus-AgCl-NPs exerted higher cytotoxicity against HEK293T cells than doxorubicin and K. rotunda-Ag/AgCl-NPs. Both the nanoparticles induced apoptosis in A549 and SMMC-7721 cell lines. A significant rise of early apoptotic cells and late apoptotic cells was found for A549 cells after treatment with A. racemosus-AgCl-NPs and stained with FITC-annexin V/PI. Apoptosis in A549 cells was further confirmed by monitoring the alteration of the expression level of several genes using real-time PCR and cell cycle arrest by flowcytometry after treatment with A. racemosus-AgCl-NPs. The expression of STAT-3, TNFα, and EGFR genes was decreased with the increase of caspase-8, FAS, and FADD gene expression. G2/M cell cycle phase was arrested after treatment of A549 cells with A. racemosus-AgCl-NPs.

The extracellular matrix (ECM) and cells are crucial components of natural tissue microenvironments, and they both demonstrate dynamic mechanical properties, particularly viscoelastic behaviors, when exposed to external stress or strain over time. The capacity to modify the mechanical properties of cells and ECM is crucial for gaining insight into the development, physiology, and pathophysiology of living organisms. As an illustration, researchers have developed hydrogels with diverse compositions to mimic the properties of the native ECM and use them as substrates for cell culture. The behavior of cultured cells can be regulated by modifying the viscoelasticity of hydrogels. Moreover, there is widespread interest across disciplines in accurately measuring the mechanical properties of cells and the surrounding ECM, as well as exploring the interactive relationship between these components. Nevertheless, the lack of standardized experimental methods, conditions, and other variables has hindered systematic comparisons and summaries of research findings on ECM and cell viscoelasticity. In this review, we delve into the origins of ECM and cell viscoelasticity, examine recently developed methods for measuring ECM and cell viscoelasticity, and summarize the potential interactions between cell and ECM viscoelasticity. Recent research has shown that both ECM and cell viscoelasticity experience alterations during in vivo pathogenesis, indicating the potential use of tailored viscoelastic ECM and cells in regenerative medicine.

Polarimetry is a powerful optical tool in the biomedical field, providing more comprehensive information on the sub-wavelength micro-physical structure of a sample than traditional light intensity measurement techniques. This review summarizes the concepts and techniques of polarization and its biomedical applications. Specifically, we first briefly describe the basic principles of polarized light and the Mueller matrix (MM) decomposition method, followed by some research progress of polarimetric measurement techniques in recent years. Multimodal mass spectrometry (MMS) incorporates an imaging modality with probe‐based mass spectrometry (MS) to enable precise, targeted data acquisition and provide additional biological and chemical data not available by MS alone. Two categories of MMS are covered; in the first, an imaging modality guides the MS probe to target individual cells and to reduce acquisition time by automatically defining regions of interest. In the second category, imaging and MS data are coupled in the data analysis pipeline to increase the effective spatial resolution using a higher resolution imaging method, correct for tissue deformation, and incorporate fine morphological features in an MS imaging dataset. Tumor targeting has been a great challenge for drug delivery systems. A number of nanotechnology-derived drug carriers have been developed for cancer treatment to improve efficacy and biocompatibility. Among them, the emergence of cell-nanocarriers has attracted great attention, which simulates cell function and has good biocompatibility. They can also escape the clearance of reticuloendothelial system, showing a long-cycle effect. The inherent tumor migration and tumor homing ability of cells increase their significance as tumor-targeting vectors. In this review, we focus on the combination of stem cells, immune cells, red blood cells, and cell membranes to nanocarriers, which enable chemotherapy agents to efficiently target lesion sites and improve drug distribution while being low toxic and safe. In addition, we discuss the pros and cons of these nanoparticles as well as the challenges and opportunities that lie ahead. Although research to address these limitations is still ongoing, this promising tumor-targeted drug delivery system will provide a safe and effective platform against cancer. [ABSTRACT FROM AUTHOR]

Separation of micro- and nano-sized bioparticles is essential for efficient diagnostics, chemical and biological analyses, drug development, food and chemical processing, and environmental monitoring. However, most of the currently available bio-separation techniques are based on the membrane filtration approach, whose efficiency is restricted by membrane-related disadvantages, including pore size, surface charge density, and biocompatibility, which results in a reduction in the isolation resolution. To address these issues, till now, many microfluidic devices have been developed for particle/cell profiling due to their excellent sensitivity and specificity, less sample consumption, shortened processing time, and high throughput features. Of the various microfluidic systems, the spiral inertial microfluidic technique has recently attracted attention as an innovative strategy and advanced cutting-edge technology toward bioparticle separation. Depending on the needs of the microfluidic device, the spiral inertial chip can be customized to separate bioparticles owing to their sizes and different shapes. In this review, we discuss the kinematics of microchannel particle separation mechanisms, recent developments in the inertial microfluidic device realm, and their applications for the separation of several types of bioparticles, including blood cells, stem cells, sperm cells, pathogens, and algae. Finally, we highlight challenges and economical perspectives associated with guidelines for further development of spiral inertial microfluidic devices in the future. [ABSTRACT FROM AUTHOR]

Worldwide, interest in mitochondria is constantly growing, as evidenced by scientific statistics, and studies of the functioning of these organelles are becoming more prevalent than studies of other cellular structures. In this analytical review, mitochondria are conditionally placed in a certain cellular center, which is responsible for both energy production and other non-energetic functions, without which the existence of not only the eukaryotic cell itself, but also the entire organism is impossible. Taking into account the high multifunctionality of mitochondria, such a fundamentally new scheme of cell functioning organization, including mitochondrial management of processes that determine cell survival and death, may be justified. Considering that this issue is dedicated to the memory of V. P. Skulachev, who can be called mitocentric, due to the history of his scientific activity almost entirely aimed at studying mitochondria, this work examines those aspects of mitochondrial functioning that were directly or indirectly the focus of attention of this outstanding scientist. We list all possible known mitochondrial functions, including membrane potential generation, synthesis of Fe-S clusters, steroid hormones, heme, fatty acids, and CO2. Special attention is paid to the participation of mitochondria in the formation and transport of water, as a powerful biochemical cellular and mitochondrial regulator. The history of research on reactive oxygen species that generate mitochondria is subject to significant analysis.
