Syngas tars are responsible for clogging and corrosion of pipelines and equipments due to their high condensation temperatures. At the same time, considering their energy content they cause a reduction in the energy efficiency of the conversion process. To overcome these drawbacks, the addition of a downstream reactor to perform hot catalytic gas cleaning has been proposed. However, the occurrence of significant carbon deposition and sulphide formation on the catalytically active surfaces can easily lead to early deactivation of the catalyst. Chemical looping tar reforming is based on a solid material, known as oxygen carrier, that undergoes two reactions steps: (1) reduction by interaction with the gas and tar streams; (2) regeneration by oxidation with ambient air, which also involves the combustion of any deposits on the particle surface. In this work, the first reaction step of the process is investigated in an integrated setup involving the pyrolysis or the gasification of hazelnut shells and the reaction with the oxygen carrier for tar abatement. Two reactor configurations have been considered: (1) single reactor, where the biomass and the oxygen carrier beds are loaded in series into the same reactor; (2) two reactors, where the two beds are loaded into different reactors in series. Blank tests for pyrolysis and gasification are also carried out for comparison. The results indicate that the two beds configuration enables higher tar conversion (89% wt for pyrolysis and 75% wt for steam gasification), though the presence of the oxygen carrier causes a reduction in the energy content of the syngas, especially in terms of H2 concentration, which is reduced from around 34% to 21% mol for pyrolysis and from 28% to 21% mol for steam gasification.

The potential of applying a floating PV (FPV) system in an Italian context (namely, Cecita dam and Mucone hydroelectric power plants) is studied. The additional PV energy production, as well as the effect of non-evaporated water on the productivity of the hydropower plant, is analyzed by varying the basin surface coverage. The simulations highlight that the amount of additional hydroelectricity is quite small if compared to the non-FPV system, reaching about 3.56% for 25% basin surface coverage. However, the annual PV energy production is noticeable even at low coverage values. The expected gain in electricity production in the case of 25% basin surface coverage with the FPV plant rises to 391% of that of the actual hydropower plant. This gain becomes even larger if a vertical axis tracking system is installed and the increase is about 436%. The economic analysis confirms that the production costs (USD/kWh) of FPV systems are comparable to those of land-based PV (LBPV) plants, becoming smaller in the case that a tracking system is installed. In particular, the best solution is the one with 15% coverage of the lake. In this case, the levelized cost of electricity for the LBPVs is 0.030 USD/kWh and for the FVPs, with and without tracking, it is equal to 0.032 and 0.029 USD/kWh, respectively.

The study utilizes open-access data to generate power demand curves for a hybrid automotive system, testing twelve configurations with three different energy management strategies and four values for the degree of hybridization (DOH), the latter representing the share of the total power of the vehicle powertrain supplied by the battery. The first control logic (Battery Main—BTM) uses mainly batteries to satisfy the power demand and fuel cells as backup, while in the other two controllers, fuel cells operate continuously (Fuel Cell Main—FCM) or within a fixed range (Fuel Cell Fixed—FCF) using batteries as backup. The results are assessed in terms of H2 consumption, overall system efficiency, and fuel cell predicted lifespan. The battery is heavily stressed in the BTM and FCF logics, while the FCM logic uses the battery only occasionally to cover load peaks. This is reflected in the battery’s State of Charge (SOC), indicating different battery stress levels between the BTM and FCF modes. The FCF logic has higher stress levels due to load demand, reducing battery lifetime. In the BTM and FCM modes, the fuel cell operates with variable power, while in the FCF mode, the fuel cell operates in a range between 90 and 105% of its rated power to ensure its lifetime. In the BTM and FCM modes, hydrogen consumption decreases at almost the same rate as the DOH increases, due to a decrease in battery capacity and a smaller amount of hydrogen being used to recharge it. In contrast, the FCF control logic results in a larger fuel consumption when the DOH decreases. In terms of FC durability, the FCF control logic performs better, with a predicted lifetime ranging from 1815 h for DOH = 0.5 to 2428 h for DOH = 0.1. The FCM logic has the worst performance, with a predicted lifetime of 800 to 808 h, being almost insensitive to the DOH variation. Simulations were performed on two different driving cycles, and similar trends were observed. Simulations taking into account fuel cell (FC) performance degradation showed an increase in hydrogen consumption of approximately 38% after 12 years. Overall, this study highlights the importance of optimizing control systems to improve the performance of fuel cell hybrid vehicles, also taking into account the component of performance degradation.

Introduction: In the study of neurodegenerative diseases, the possibility to follow the fate of specific cells or molecules within the whole body would be a milestone to better understand the complex evolution of disease mechanisms and to monitor the effects of therapies. The techniques available today do not allow the visualization of disease-relevant cells within the whole tridimensional biological context at high spatial resolution.Methods: Here we show the results from the first validation steps of a novel approach: by combining the conjugate nanobodies anti-glial fibrillary acidic protein (GFAP) and metal-nanoparticles (i.e. 2 nm gold NP) with X-ray phase contrast tomography (XPCT) we would be able to obtain a tridimensional visualization and identification of cells of interest together with the surrounding tissue and the vascular and neuronal networks.Results: By exploiting the X-ray attenuation properties of metal nanoparticles and the specific targeting capabilities of nanobodies, we could give XPCT the specificity it presently lacks, making it no longer a pure morphological but a molecular and targeted imaging technique. In our case, we synthesized and characterized Gold-NP/GFAP nanobody to target the astrocytes of mouse brain.Discussion: The results of the first tests presented in this paper have provided us with information on the feasibility of the approach, encouraging us to carry out further experiments in order to achieve the ultimate goal of setting up this new imaging technique.

Background:Rheumatoid arthritis (RA) is a chronic systemic autoimmune disease that affects synovial joints, leading to inflammation, joint destruction, loss of function, and disability. Although recent pharmaceutical advances have improved treatment of RA, patients with RA often inquire about dietary interventions to improve RA symptoms, as they perceive rapid changes in their symptoms after consumption of certain foods. There is evidence that some ingredients have pro- or anti-inflammatory effects. In addition, recent literature has shown a link between diet and microbiome changes. Both diet and the gut microbiome are linked to circulating metabolites that may modulate inflammation. However, evidence of the effects of an anti-inflammatory and probiotic-rich diet in patients with RA is scarce. There is also a need for biological data to support its anti-inflammatory effects. Methods:The main goal of this study is to delineate the design process for a diet tailored to our RA population. To achieve this goal, we collected information on diet, supplements, cooking methods, and intake of different ingredients for each patient. Different groups were interviewed, and their feedback was assessed to design a diet that incorporates suggested anti-inflammatory ingredients in a manner that was easy for patients to adopt based on their lifestyles and backgrounds. Results:We designed a diet that includes a high intake of potential anti-inflammatory ingredients. Feedback from highly motivated patients was critical in constructing an anti-inflammatory diet (ITIS diet) with elevated adherence. Conclusion:In order to tailor our diet, we surveyed our patients on several different parameters. We obtained important feedback on how feasible our ITIS diet is for RA patients. Using this feedback, we made minor improvements and finalized the design of the ITIS diet. This diet is being used in an on-going pilot study to determine their anti-inflammatory effect in pain and joint swelling in RA patients. Trial registration:Not applicable.

X-ray phase-contrast tomography offers a highly sensitive 3D imaging approach to investigate different disease-relevant networks at levels ranging from single cell through to intact organ. The authors present a concomitant study of the evolution of tissue damage and inflammation in different organs affected by the disease in the murine model for multiple sclerosis.

The human olfactory bulb (OB) has a laminar structure. The segregation of cell populations in the OB image poses a significant challenge because of indistinct boundaries of the layers. Standard 3D visualization tools usually have a low resolution and cannot provide the high accuracy required for morphometric analysis. X-ray phase contrast tomography (XPCT) offers sufficient resolution and contrast to identify single cells in large volumes of the brain. The numerous microanatomical structures detectable in XPCT image of the OB, however, greatly complicate the manual delineation of OB neuronal cell layers. To address the challenging problem of fully automated segmentation of XPCT images of human OB morphological layers, we propose a new pipeline for tomographic data processing. Convolutional neural networks (CNN) were used to segment XPCT image of native unstained human OB. Virtual segmentation of the whole OB and an accurate delineation of each layer in a healthy non-demented OB is mandatory as the first step for assessing OB morphological changes in smell impairment research. In this framework, we proposed an effective tool that could help to shed light on OB layer-specific degeneration in patients with olfactory disorder.

The proximal caudal vertebrae and notochord in thick-toed geckos (TG) (Chondrodactylus turneri, Gray, 1864) were investigated after a 30-day space flight onboard the biosatellite Bion-M1. This region has not been explored in previous studies. Our research focused on finding sites most affected by demineralization caused by microgravity (G0). We used X-ray phase-contrast tomography to study TG samples without invasive prior preparation to clarify our previous findings on the resistance of TG’s bones to demineralization in G0. The results of the present study confirmed that geckos are capable of preserving bone mass after flight, as neither cortical nor trabecular bone volume fraction showed statistically significant changes after flight. On the other hand, we observed a clear decrease in the mineralization of the notochordal septum and a substantial rise in intercentrum volume following the flight. To monitor TG’s mineral metabolism in G0, we propose to measure the volume of mineralized tissue in the notochordal septum. This technique holds promise as a sensitive approach to track the demineralization process in G0, given that the volume of calcification within the septum is limited, making it easy to detect even slight changes in mineral content.

Skeletal muscle is a highly adaptive organ that sustains continuous metabolic changes in response to different functional demands. Healthy skeletal muscle can adjust fuel utilization to the intensity of muscle activity, the availability of nutrients and the intrinsic characteristics of muscle fibers. This property is defined as metabolic flexibility. Importantly, impaired metabolic flexibility has been associated with, and likely contributes to the onset and progression of numerous pathologies, including sarcopenia and type 2 diabetes. Numerous studies involving genetic and pharmacological manipulations of histone deacetylases (HDACs) in vitro and in vivo have elucidated their multiple functions in regulating adult skeletal muscle metabolism and adaptation. Here, we briefly review HDAC classification and skeletal muscle metabolism in physiological conditions and upon metabolic stimuli. We then discuss HDAC functions in regulating skeletal muscle metabolism at baseline and following exercise. Finally, we give an overview of the literature regarding the activity of HDACs in skeletal muscle aging and their potential as therapeutic targets for the treatment of insulin resistance.

Glioblastoma multiforme (GBM) is one of the deadliest and most aggressive cancers, remarkably resilient to current therapeutic treatments. Here, we report preliminary in vivo studies of GBM treatments based on photo-nanotherapeutics to activate synergistic killing mechanisms. Core-shell nanoparticles have been weaponized by combining photophysical properties of a new generation PDT agent (Ir(III) complex) with the thermoplasmonic effects of resonant gold nanospheres. In order to investigate the damages induced in GBM treated with these photoactivable nanosystems, we employed X-ray phase-contrast tomography (XPCT). This high-resolution three-dimensional imaging technique highlighted a vast devascularization process by micro-vessels disruption, which is indicative of tumor elimination without relapse.

Abstract The aim of this study was the detection and quantification of the Na+ depositions in the extracellular matrix of myocardial tissue, which are suggested to be bound by negatively charged glycosaminoglycan (GAG) structures. The presented experimental results are based on high resolution X-ray fluorescence (XRF) spectromicroscopy technique used to perform a comparative analysis of sodium containment in intracellular and interstitial spaces of cardiac tissues taken from animals selected by low and high sodium intake rates. The experimental results obtained show that high sodium daily intake can result in a remarkable increase of sodium content in the myocardial interstitium.

Mitigation of greenhouse gas emissions is becoming a significant factor in all industries. Cement manufacturing is one of the industries responsible for greenhouse gas emissions, specifically carbon dioxide emissions. Pozzolanic materials have long been used as cement additives due to the pozzolanic reaction that occurs when hydrated and the formation a cementitious material similar to that of cement. In this study, shale, which is a common component found in wellbore drill cuttings, was used in various sizes and quantities to determine the effect it had on the mechanical properties of wellbore cement. The unconfined compressive strength of the cement containing shale was compared to the cement without shale to observe the effect that both the quantity and particle size had on this property. SEM–EDS microscopy was also performed to understand any notable variations in the cement microstructure or composition. The samples containing micron shale appeared to have the best results of all the samples containing shale, and some of the samples had a higher UCS than one or more of the base case samples. Utilization of cuttings as a cement additive is not just beneficial in that it minimizes the need for cuttings removal and recycling, but also in that it reduces the amount of greenhouse gas emissions associated with cement manufacturing.

Abstract The number of the Asbestos Bodies (AB), i.e. asbestos that developed an iron-protein coating during its permanence in biological tissues, is one of the most accessible markers of asbestos exposure in individuals. The approaches developed to perform AB count in biological tissues are based on the manual examination of tissue digests or histological sections by means of light or electron microscopies. Although these approaches are well established and relatively accessible, manual examination is time-consuming and can be reader-dependent. Besides, approximations are applied because of the limitations of 2D readings and to speed up manual counts. In addition, sample preparation using tissue digests require an amount of tissue that can only be obtained by invasive surgery or post-mortem sampling. In this paper, we propose a new approach to AB counting based on non-destructive 3D imaging, which has the potential to overcome most of the limitations of conventional approaches. This method allows automating the AB count and determining their morphometry distribution in bulk tissue samples (ideally non-invasive needle biopsies), with minimal sample preparation and avoiding approximations. Although the results are promising, additional testing on a larger number of AB-containing biological samples would be required to fully validate the method.

Frontotemporal dementia (FTD) is a spectrum of clinical syndromes that affects personality, behavior, language, and cognition. The current diagnostic criteria recognize three main clinical subtypes: the behavioral variant of FTD (bvFTD), the semantic variant of primary progressive aphasia (svPPA), and the non-fluent/agrammatic variant of PPA (nfvPPA). Patients with FTD display heterogeneous clinical and neuropsychological features that highly overlap with those presented by psychiatric syndromes and other types of dementia. Moreover, up to now there are no reliable disease biomarkers, which makes the diagnosis of FTD particularly challenging. To overcome this issue, different studies have adopted metrics derived from magnetic resonance imaging (MRI) to characterize structural and functional brain abnormalities. Within this field, a growing body of scientific literature has shown that graph theory analysis applied to MRI data displays unique potentialities in unveiling brain network abnormalities of FTD subtypes. Here, we provide a critical overview of studies that adopted graph theory to examine the topological changes of large-scale brain networks in FTD. Moreover, we also discuss the possible role of information arising from brain network organization in the diagnostic algorithm of FTD-spectrum disorders and in investigating the neural correlates of clinical symptoms and cognitive deficits experienced by patients.

Abstract Ancient Herculaneum papyrus scrolls, hopelessly charred in the 79 A.D. Vesuvius eruption, contain valuable writings of the Greek philosophers of the day, including works of the Epicurean Philodemus. X-ray phase contrast tomography has recently begun unlocking their secrets. However, only small portions of the text hidden inside the scroll have been recover. One of the challenging tasks in Herculaneum papyri investigation is their virtual unfolding because of their highly complicated structure and three-dimensional arrangement. Although this procedure is feasible, problems in segmentation and flattening hinder the unrolling of a large portion of papyrus. We propose a computational platform for the virtual unfolding procedure, and we show the results of its application on two Herculaneum papyrus fragments. This work paves the way to a comprehensive survey and to further interpretation of larger portions of text hidden inside the carbonized Herculaneum papyri.

Histone deacetylases (HDACs) are enzymes that regulate the deacetylation of numerous histone and non-histone proteins, thereby affecting a wide range of cellular processes. Deregulation of HDAC expression or activity is often associated with several pathologies, suggesting potential for targeting these enzymes for therapeutic purposes. For example, HDAC expression and activity are higher in dystrophic skeletal muscles. General pharmacological blockade of HDACs, by means of pan-HDAC inhibitors (HDACi), ameliorates both muscle histological abnormalities and function in preclinical studies. A phase II clinical trial of the pan-HDACi givinostat revealed partial histological improvement and functional recovery of Duchenne Muscular Dystrophy (DMD) muscles; results of an ongoing phase III clinical trial that is assessing the long-term safety and efficacy of givinostat in DMD patients are pending. Here we review the current knowledge about the HDAC functions in distinct cell types in skeletal muscle, identified by genetic and -omic approaches. We describe the signaling events that are affected by HDACs and contribute to muscular dystrophy pathogenesis by altering muscle regeneration and/or repair processes. Reviewing recent insights into HDAC cellular functions in dystrophic muscles provides new perspectives for the development of more effective therapeutic approaches based on drugs that target these critical enzymes.

Abstract Evidence supports that hereditary angioedema (HAE) may be considered as a paroxysmal permeability disorder with defective but self-limiting endothelial barrier dysfunction. A potential subclinical abnormal vascular permeability at retinal capillaries could induce damage resulting in retinopathy. We aimed at exploring for the first time the presence of microangiopathy at retinal level from a highly selective cohort of patients with HAE due to C1 esterase inhibitor protein (C1INH) deficiency (type I). We conducted a pilot, prospective, case-control study including 20 type I HAE patients and 20 age−/sex-matched healthy controls (HC). All participants underwent standard ophthalmological examination including visual fields. Superficial and deep capillary plexi in the retina were analyzed by using new optical coherence tomography angiography (OCT-A). A total of 40 eyes from 20 HAE patients and 20 eyes from HC were evaluated. Perimetric indices of visual field were slightly worse in HAE than in controls. OCT-angiograms documented in HAE patients a lower retinal capillary density in both superficial and deep scans and a higher retinal thickness compared to healthy eyes. Our findings firstly documented subclinical abnormalities in retinal microvascular network in type I HAE patients that might be associated with early subtle functional changes. This preliminary evidence supports the hypothesis of a recurrent endothelial barrier failure at retinal level in HAE patients potentially resulting in chronic damage.
