The main functions of the mammalian cecum include microbial digestion of soluble and insoluble carbohydrates and the absorption of significant amounts of water and electrolytes. The present study was carried out to investigate the gross anatomy and electron microscopy of the cecum in adult camels (Camelus dromedarius), which still needs to be clarified. The study was conducted using samples collected from camels slaughtered at an abattoir in Omdurman, Sudan. The camel cecum, which occupied the right part of the abdomen, was situated caudally to the abomasum and directed caudally to the pelvis. It was short and S-shaped and began largely at the ceco-colic junction at the level of the fifth lumbar vertebra and became smaller towards its apex. Morphometrically, camel cecum constituted 3.2% of the large intestine length, 1% of both the small intestine and large intestine length, and 0.10% of the long axis of the animal body. Ultra-structurally, the cecal mucosa showed several folds and was lined by a simple columnar epithelium with many intestinal cells (enterocytes) and goblet cells; two types of enterocytes were shown: type one contained a dark cytoplasm and type two had a light cytoplasm. In conclusion, the cecum in dromedary camels was generally similar in structure to other domestic animals; however, they have a shorter cecum compared to ruminants. [ABSTRACT FROM AUTHOR]

Background: Integrating systems biology is an approach for investigating metabolic diseases in humans. However, few studies use this approach to investigate the mechanism by which Rhizoma coptidis (RC) reduces the effect of lipids and glucose on high-fat induced obesity in rats. Methods: Twenty-four specific pathogen-free (SPF) male Sprague–Dawley rats (80 ± 10 g) were used in this study. Serum metabolomics were detected by ultra-high-performance liquid chromatography coupled with quadrupole-time-of-flight tandem mass spectrometry. Liver tissue and cecum feces were used for RNA-Seq technology and 16S rRNA gene sequencing, respectively. Results: We identified nine potential biomarkers, which are differential metabolites in the Control, Model and RC groups, including linoleic acid, eicosapentaenoic acid, arachidonic acid, stearic acid, and L-Alloisoleucine (p < 0.01). The liver tissue gene expression profile indicated the circadian rhythm pathway was significantly affected by RC (Q ≤ 0.05). A total of 149 and 39 operational taxonomic units (OTUs), which were highly associated with biochemical indicators and potential biomarkers in the cecum samples (FDR ≤ 0.05), respectively, were identified. Conclusion: This work provides information to better understand the mechanism of the effect of RC intervention on hyperlipidemia and hypoglycemic effects in obese rats. The present study demonstrates that integrating systems biology may be a powerful tool to reveal the complexity of metabolic diseases in rats intervened by traditional Chinese medicine. Resistant starch (RS) was recently approved to exert a powerful influence on gut health, but the effect of RS on the caecal barrier function in meat ducks has not been well defined. Thus, the effect of raw potato starch (RPS), a widely adopted RS material, on microbial composition and barrier function of caecum for meat ducks was determined. A total of 360 Cherry Valley male ducks of 1-d-old were randomly divided and fed diets with 0 (control), 12, or 24 % RPS for 35 d. Diets supplemented with RPS significantly elevated villus height and villus height:crypt depth ratio in the caecum. The 16S rRNA sequence analysis indicated that the diet with 12 % RPS had a higher relative abundance of Firmicutes and the butyrate-producing bacteria Faecalibacterium , Subdoligranulum , and Erysipelatoclostridium were enriched in all diets. Lactobacillus and Bifidobacterium were significantly increased in the 24 % RPS diet v. the control diet. When compared with the control diet, the diet with 12 % RPS was also found to notably increase acetate, propionate and butyrate contents and up-regulated barrier-related genes including claudin-1, zonula occludens-1, mucin-2 and proglucagon in the caecum. Furthermore, the addition of 12 % RPS significantly reduced plasma TNF- α , IL-1 β and endotoxin concentrations. These data revealed that diets supplemented with 12 % RPS partially improved caecal barrier function in meat ducks by enhancing intestinal morphology and barrier markers expression, modulating the microbiota composition and attenuating inflammatory markers. The objective of this study was to develop baseline topographical data of landmarks for locating and generating sonographic calliper measurements along with dynamic parameters of caecum and colon in 50 native adult healthy unsedated Spiti horses and Himalayan Hill mules of either sex. The wall thickness, contractility, character and echo-architecture of caecum and colon were recorded. Caecum was found from 15th to 17th inter costal space (ICS). Right dorsal colon (RDC), right ventral colon (RVC), left ventral colon (LVC) and left dorsal colon (LDC) were found from 6th to 14th, 9th to 17th, 9th to 14th and 6th to 15th ICS, respectively. Descending colon was found in the left paralumbar fossa upto tuber-coxae in both horses as well as mules. The corresponding values of horses and mules for wall thickness (in mm) was 2.1±0.05 and 1.4±0.05 for the caecum, 2.16±0.05 and 1.32±0.02 for the right dorsal colon, 2.1±0.03 and 1.32 ±0.02 for the right ventral colon, 2.12±0.03 and 1.32±0.02 for the left dorsal colon, 2.1±0.03 and 1.32±0.02 for the right dorsal colon and 2.04±0.02 for the descending colon. Prominent sacculations were observed on ventral colon and absent on dorsal colon. We report measurements of ion transport across the gastric caecum of larvae of Aedes aegypti, a vector of yellow fever that inhabits a variety of aquatic habitats ranging from freshwater to brackish water. We provide the first measurements of the effect of 5-hydroxytryptamine (5-HT) on transepithelial potential (TEP), luminal ion concentrations and electrochemical potentials, as well as basolateral membrane potential and H+, Na+ and K+ fluxes. TEP, basolateral membrane potential, and H+, K+ and Na+ fluxes across the gastric caeca declined within 3-6 min after isolation of the entire midgut from the larva. 5-HT restored both the TEP and active accumulation of H+, K+ and Na+ in the lumen. Additionally, 5-HT restored H+, K+ and Na+ fluxes across the distal caecum of freshwater larvae, and restored H+ fluxes across the distal caecum of brackish water larvae. There was no effect of 5-HT on ion fluxes across the proximal caecum.We have also shown that 5-HT restores the basolateral membrane potential in cells of the distal, but not proximal, caecum. Effects of 5-HT on TEP and basolateral membrane potential were mimicked by application of cAMP but not by a phorbol ester. We provide a working model which proposes that 5-HT and cAMP stimulate the vacuolar H+-ATPase of the distal caecum. Our results provide evidence that the gastric caecum is functionally distinct from the adjacent anterior midgut and we discuss possible roles of the gastric caecum in osmoregulation. Background and study aim Different techniques have been introduced to improve the endoscopist's view and enhance the detection of polyps. The endocuff is a polymer sleeve cap that is connected to the tip of the colonoscope in order to improve visualization of the mucosa during colonoscopy. The aim of the study was to compare adenoma detection rates (ADR) of endocuff-assisted colonoscopy and conventional colonoscopy. Patients and methods Patients 50 years or older were randomized into two groups: an endocuff-assisted colonoscopy group and a conventional colonoscopy group without the endocuff. Results A total of 337 patients were included: 174 in the endocuff group and 163 in the conventional group. The median age was 61 years (interquartile range 55 - 70 years), and 74 % were women. The ADR was higher in the endocuff group than in the conventional group (22.4 % vs. 13.5 %; P = 0.02). The mean number of adenomas was 0.30 (SD 0.25) in the endocuff group and 0.21 (SD 0.26) in the conventional group (P  = 0.02). The rate of ileal intubation was lower in the endocuff group (73 % vs. 87 %; P < 0.001). No serious adverse events occurred with the use of the endocuff. In this study we determined protein and gene expression in the caeca of newly hatched chickens inoculated with cecal contents sourced from hens of different ages. Over 250 proteins exhibited modified expression levels in response to microbiota inoculation. The most significant inductions were observed for ISG12-2, OASL, ES1, LYG2, DMBT1-L, CDD, ANGPTL6, B2M, CUZD1, IgM and Ig lambda chain. Of these, ISG12-2, ES1 and both immunoglobulins were expressed at lower levels in germ-free chickens compared to conventional chickens. In contrast, CELA2A, BRT-2, ALDH1A1, ADH1C, AKR1B1L, HEXB, ALDH2, ALDOB, CALB1 and TTR were expressed at lower levels following inoculation of microbiota. When chicks were given microbiota preparations from different age donors, the recipients mounted differential responses to the inoculation which also differed from the response profile in naturally colonised birds. For example, B2M, CUZD1 and CELA2A responded differently to the inoculation with microbiota of 4- or 40-week-old hens. The increased or decreased gene expression could be recorded 6 weeks after the inoculation of newly hatched chickens. To characterise the proteins that may directly interact with the microbiota we characterised chicken proteins that co-purified with the microbiota and identified a range of host proteins including CDD, ANGPTL6, DMBT1-L, MEP1A and Ig lambda. We propose that induction of ISG12-2 results in reduced apoptosis of host cells exposed to the colonizing commensal microbiota and that CDD, ANGPTL6, DMBT1-L, MEP1A and Ig lambda reduce contact of luminal microbiota with the gut epithelium thereby reducing the inflammatory response. The effect of mannitol on bone-related mineral absorption and retention and the mechanism was investigated in this study. Fourteen 8-week-old male Wistar rats in experiment 1 and same number and age cecectomized Wistar male rats in experiment 2 were divided into two subgroups of seven animals, respectively, fed diets containing 0 or 4% mannitol for 28 days. Mineral balance tests were determined twice during days 8-12 and days 22-26, and the rats were slaughtered on day 28 both in experiment 1 and experiment 2. The whole caecum and colon were collected with the content to analyse tissue weight, content weight, content's pH and moisture, organic acids' concentration and mineral levels. In experiment 1, Ca absorption and retention and Mg absorption were significantly increased by mannitol feeding during days 8-12. Caecal total weight, tissue weight and content weight were increased, the pH of caecum and colon was reduced, and the concentrations of caecal short-chain fatty acids ( SCFAs) were modified by mannitol feeding. In experiment 2, during days 8-12 and days 22-26, Ca absorption and retention were significantly lowered by mannitol feeding in cecectomized rats; however, mannitol feeding decreased Mg absorption during days 8-12, but did not impact Mg retention. Colonic total weight, tissue weight and content weight were significantly increased, and colonic pH was reduced by mannitol feeding. In conclusion, dietary mannitol increased the absorption of Ca and Mg and the caecum markedly contributed to this promoting effect of mannitol. [ABSTRACT FROM AUTHOR]

Pinworm nematodes of the genus Syphacia (Nemata : Oxyurida : Oxyuridae) have a global distribution, and infect the caecum of rodents. Within the Australian Bioregion, 17 species of Syphacia infect a range of rodent hosts. Pinworms are traditionally thought to have coevolutionary relationships with their hosts, but the evolution and dispersal of Australian rodents and their helminths remains unclear. This combination of factors allowed us to investigate the likely relationships of Australian Syphacia species based on phylogenetic analysis, overlaid with the ecology and relationships of host species. We conducted a phylogenetic analysis using morphological characters of the species of Syphacia from the Australian Bioregion in order to examine the relationships between species, and to investigate how host evolution and phylogeny could inform (or be informed) by parasite phylogeny. Application of the taxon pulse theory of parasite speciation by matching host species to parasites shed some light on the timing of speciation of rodent hosts. We found that species of Syphacia had reasonably close host-parasite relationships, with additional evidence for ecological fitting or host switching occurring. Evidence provided here suggests strongly that most elements of the Stockholm Paradigm are at play in structuring the relationships we observe in this pinworm-mammal system. [ABSTRACT FROM AUTHOR]

Background Segmental distribution of colorectal volume is relevant in a number of diseases, but clinical and experimental use demands robust reliability and validity. Using a novel semi-automatic magnetic resonance imaging-based technique, the aims of this study were to describe: (i) inter-individual and intra-individual variability of segmental colorectal volumes between two observations in healthy subjects and (ii) the change in segmental colorectal volume distribution before and after defecation. Methods The inter-individual and intra-individual variability of four colorectal volumes (cecum/ascending colon, transverse, descending, and rectosigmoid colon) between two observations (separated by 52 ± 10) days was assessed in 25 healthy males and the effect of defecation on segmental colorectal volumes was studied in another seven healthy males. Key Results No significant differences between the two observations were detected for any segments (All p > 0.05). Inter-individual variability varied across segments from low correlation in cecum/ascending colon (intra-class correlation coefficient [ ICC] = 0.44) to moderate correlation in the descending colon ( ICC = 0.61) and high correlation in the transverse ( ICC = 0.78), rectosigmoid ( ICC = 0.82), and total volume ( ICC = 0.85). Overall intra-individual variability was low (coefficient of variance = 9%). After defecation the volume of the rectosigmoid decreased by 44% ( p = 0.003). The change in rectosigmoid volume was associated with the true fecal volume ( p = 0.02). Conclusions & Inferences Imaging of segmental colorectal volume, morphology, and fecal accumulation is advantageous to conventional methods in its low variability, high spatial resolution, and its absence of contrast-enhancing agents and irradiation. Purpose: Characterize the contents of distal ileum and cecum in healthy adults under conditions simulating the bioavailability/bioequivelance studies of drug products in fasted and fed state. Methods: Twelve males participated in a two-phase crossover study. Phase I: subjects remained fasted overnight plus 4.5 h in the morning prior to colonoscopy. Phase II: subjects remained fasted overnight, consumed breakfast in the morning, and abstain from food until colonoscopy, 4.5 h after breakfast. Upon sampling, volume, pH and buffer capacity were measured; after ultracentrifugation, supernatant was physicochemically characterized and non-liquid particles diameter was measured. Results: In distal ileum, pH is ~8.1 and size of non-liquid particles is ~200 μm, regardless of dosing conditions; in fed state, liquid fraction was lower whereas osmolality and carbohydrate content were higher. In cecum, the environment was similar with previously characterized environment in the ascending colon; in fasted state, size of non-liquid particles is smaller than in distal ileum (~70 μm). Fluid composition in distal ileum is different from cecum, especially in fasted state. Conclusion: Differences in luminal environment between distal ileum and cecum may impact the performance of orally administered products which deliver drug during residence in lower intestine. Benign duodenocolic fistula (DCF), known as a fistula between the duodenum and colon with or without cecum of nonmalignant origin, is an unusual complication of different gastrointestinal diseases. The present paper records a case in which the patient presented with chronic diarrhea, abdominal pain, weight loss as well as having a history of gastric ulcer. Most frequently the condition presents with signs of malabsorption such as weight loss and diarrhea, but other symptoms include nausea, vomiting (sometimes with fecal), and abdominal pain. Gastrointestinal inflammatory conditions are the usual causes. The most common ones are perforated duodenal ulcer and Crohn's disease. Barium enemas are usually diagnostic. Treatment consists of excising the fistula and repairing the duodenal and colonic defects. Closure of the fistula provides quick relief. [ABSTRACT FROM AUTHOR]

The study was conducted to evaluate the performance indices, gut microbiota, short-chain fatty acid (SCFA) and lactic acid concentration and pH value in ileal and cecal digesta of 42-day-old broilers fed a diet supplemented with lignocellulose. A total of 48, 21-day-old male Ross 308 chickens were divided into 4 treatment groups with 6 replicates per treatment. Experimental diets were based on maize, wheat, triticale, soybean meal and varied in the amount of lignocellulose: 0, 0.25, 0.5 and 1.0%. There was no significant effect of lignocellulose inclusion into the diet on broiler performance indices. Ileal Lactobacillus spp. as well as ileal and cecal Bifidobacterium spp. populations increased (P<0.05) and the number of Escherichia coli and Clostridium spp. significantly decreased in birds fed diets supplemented with lignocellulose. The sum of SCFAs and lactic acid in both intestine segments as well as acetic and propionic acids in ileal and lactic acid level in cecal digesta of chickens fed a diet with 0.5% lignocellulose were significantly higher compared to the control birds. The results show that inclusion of lignocellulose in the chicken diet, especially at a dose of 0.5%, promotes the growth of Lactobacillus spp. and Bifidobacterium spp., reduces the number of E. coli and Clostridium spp. as well as enhances the concentration of SCFAs and lactic acid with no substantial effect on the pH of ileal and cecal digesta. The intensity of fermentation and substantial amounts of the lactic acid and individual SCFAs, particularly acetic and propionic acids, may suggest the prebiotic effect of lignocellulose on the broiler gastrointestinal tract. A thorough understanding of the sea urchin (Echinodermata: Echinoidea) digestive tract anatomy is a prerequisite for the correct interpretation of physiological and biomechanical analyses focusing on the gut architecture of this ecologically important group of marine invertebrates. A number of studies have addressed the general arrangement of the sea urchin digestive tract, but accessory structures such as siphons and caeca have received less attention. Two studies carried out to analyze the gut physiology of various marine invertebrates briefly mentioned the presence of a previously undescribed pouch in the posterior digestive tract of the heart urchin (Echinoidea: Spatangoida) species Brisaster latifrons. Dissections, histological, and magnetic resonance imaging data, as well as three-dimensional reconstructions corroborate these findings. The novel structure-here termed the intestinal caecum-is suspended by a thin mesentery within a coil formed by the posteriormost part of the intestine. The kidney-shaped organ constitutes a derivative of the intestine, to which it is laterally connected through a narrow canal. In contrast to the sediment-packed main digestive tract, the intestinal caecum is filled with liquid and a flocculent mass. The organ's histology is characterized by a thin connective tissue layer with only a small number of hemal lacunae and muscle fibers, as well as an inner simple columnar epithelium that contains numerous dark-brown vacuoles. The intestinal caecum is found exclusively among members of the Schizasteridae (Spatangoida: Paleopneustina). Specifically, the organ is present in selected species of the genera Abatus, Brisaster, and Tripylaster, but not in the other seven schizasterid genera analyzed. The intestinal caecum is not homologous to the sometimes equally named accessory structure present in the posterior digestive tract of other spatangoid taxa such as Echinocardium or Heterobrissus. Consequently, the previously introduced term recto-intestinal caecum is here applied for this latter organ. No correlation could be found between the absence or presence of the intestinal caecum and any known biological or morphological characteristics of schizasterid heart urchins. The distribution of the organ among schizasterids supports a close relationship of the genera Brisaster, Tripylaster, and selected species of Abatus. [ABSTRACT FROM AUTHOR]

Sieve analyses of hindgut contents of horses as well as observations in horses where plastic markers had been applied to a caecal cannula suggested that there may be a discrimination by particle size in the passage or retention of digesta. Here, we performed a similar experiment with five caecum-cannulated horses (562 ± 31 kg) fed a constant amount (6.81 kg dry matter/day) of grass hay. Passage markers representing the liquid (Co-EDTA) as well as the particulate digesta phase (Yb-undefined; Cr mordanted fibre 1-2 mm; Ce-mordanted fibre 8 mm) were given as a pulse-dose into the cannula to measure their mean retention times (MRT). The MRTs were compared by repeated-measurements analysis of variance. The MRT in the hindgut was 22.2 ± 2.4 h for Co, 25.0 ± 3.4 h for Yb, 26.2 ± 1.6 h for Cr and 26.3 ± 1.5 h for Ce. Whereas differences between the particle marker MRTs were not significant (p adj.  > 0.05), significant differences were observed between the solute marker Co and each of the particle markers Cr and Ce (p adj.  < 0.009). The results confirm the well-known significant, albeit small, difference in MRT in horses between the fluid and the particle digesta phase, and corroborate another recent study that used a combination of whole, marked hay and individual marker analysis in different particle size fractions of the faeces, which also did not detect a selective retention of any particle size class., (© 2022 The Authors. This study investigated the impact of diets containing various levels of avocado (5, 10 and 15%) on gut health in rats fed for six weeks. Avocado-fed rats had significantly higher food intakes while their body weights remained similar to the control diet-fed rats. No significant changes in intestinal bacterial populations (ileum, cecum and colon) were found in rats fed avocado diets compared to the control diet. Ileum and colon tissues of rats fed avocado diets showed significantly higher expression of genes (β-defensin 1, mucin 3 or mucin 4) and a greater number of mucin-producing goblet cells in the colon. The percentage of avocado in the diet had varying effects in altering the biomarkers, whereby diet containing 15% avocado was the more effective diet. Salmonella enterica serovar Enteritidis (SE) infection in humans is often associated with the consumption of contaminated poultry products. Binding of the bacterium to the intestinal mucosa is a major pathogenic mechanism of Salmonella in poultry. Transposon mutagenesis identified flgC as a potential binding mutant of SE. Therefore, we hypothesize FlgC which plays a significant role in the binding ability of SE to the intestinal mucosa of poultry. To test our hypothesis, we created a mutant of SE in which flgC was deleted. We then tested the in vitro and in vivo binding ability of ∆ flgC when compared to the wild-type SE strain. Our data showed a significant decrease in the binding ability of ∆ flgC to intestinal epithelial cells as well as in the small intestine and cecum of poultry. Furthermore, the decrease in binding correlated to a defect in invasion as shown by a cell culture model using intestinal epithelial cells and bacterial recovery from the livers and spleens of chickens. The study evaluated the effect of a novel commercial preparation of natural blend of essential oils from basil, caraway, laurel, lemon, oregano, sage, tea, and thyme (Tecnaroma Herbal Mix PL) on growth performance, blood biochemistry, cecal morphology, and carcass quality of broilers. Six nutritionally adequate wheat and soybean-based diets were generated by the addition of Tecnaroma Herbal Mix PL at 0, 100, 200, 300, 400, and 500 g/t of feed. The diets were fed as crumbs in the starter phase (d 0–10) and as pellets during the grower (d 10–24) and finisher (d 24–42) phases. Nine hundred sixty 1-d-old chicks were allocated to the 6 dietary treatments each having 8 replicate pens with 20 birds per pen. The data obtained were analyzed using ANOVA with a P < 0.05 level of significance. Birds fed diets supplemented with Tecnaroma Herbal Mix PL had significantly heavier BW and higher (P < 0.05) weight gain and had improved (P < 0.05) feed to gain ratio compared with the control group during grower phase and overall performance. The blood biochemistry results showed no differences (P > 0.05) between treatments. The carcass weight, breast weight, and relative percentage of breast meat increased (P < 0.05) when diets were supplemented with Tecnaroma Herbal Mix PL compared with that from birds fed the control diet. The inclusion level of 300 g of Tecnaroma Herbal Mix PL/t of feed was optimum for enhancing breast meat yield and nutrient utilization as indicated by increased (P < 0.05) cecal villus surface area. Cecal epithelial cell damage is a key factor in host injure during the development of E. tenella. The intracellular free Ca 2+ of the host cell is closely related to the invasion, development and proliferation of intracellular parasites, and cell damage. To determine the relationship between Ca 2+ and host cell damage in the schizogenic stage of E. tenella, we established a chick embryo cecal epithelial cells model of E. tenella infection. Fluorescence staining, flow cytometry, transmission electron microscopy, inhibition and blocking experiments were used to detect the damage effect and mechanism of host cells during the schizogenic stage of E. tenella. The results showed that the host cells cytoskeletal remodeling, cell and organelle structure was destroyed, and apoptosis and necrosis were increased during the schizont stage of E. tenella. Intestinal calcium (Ca2+) absorption occurs via paracellular and transcellular pathways. Although the transcellular route has been extensively studied, mechanisms mediating paracellular absorption are largely unexplored. Unlike passive diffusion, secondarily active paracellular Ca2+ uptake occurs against an electrochemical gradient with water flux providing the driving force. Water movement is dictated by concentration differences that are largely determined by Na+ fluxes. Consequently, we hypothesized that Na+ absorption mediates Ca2+ flux. NHE3 is central to intestinal Na+ absorption. NHE3 knockout mice (NHE3-/-) display impaired intestinal Na+, water, and Ca2+ absorption. However, the mechanism mediating this latter abnormality is not clear. To investigate this, we used Ussing chambers to measure net Ca2+ absorption across different segments of wild-type mouse intestine. The cecum was the only segment with net Ca2+ absorption. Quantitative RT-PCR measurements revealed cecal expression of all genes implicated in intestinal Ca2+ absorption, including NHE3. We therefore employed this segment for further studies. Inhibition of NHE3 with 100 μM 5-(N-ethyl- N-isopropyl) amiloride decreased luminal-to-serosal and increased serosal-to-luminal Ca2+ flux. NHE3-/- mice had a >60% decrease in luminal-to-serosal Ca2+ flux. Ussing chambers experiments under altered voltage clamps (-25, 0, +25 mV) showed decreased transcellular and secondarily active paracellular Ca2+ absorption in NHE3-/- mice relative to wild-type animals. Consistent with this, cecal Trpv6 expression was diminished in NHE3-/- mice. Together these results implicate NHE3 in intestinal Ca2+ absorption and support the theory that this is, at least partially, due to the role of NHE3 in Na+ and water absorption. BACKGROUND Gaps in screening quality in community practice have been well documented. The authors examined recommended indicators of screening quality in the Centers for Disease Control and Prevention's Colorectal Cancer Screening Demonstration Program (CRCSDP), which provided colorectal cancer screening and diagnostic services between 2005 and 2009 for asymptomatic, low-income, underinsured, or uninsured individuals at 5 sites around the United States. METHODS For each client screened in the CRCSDP, a standardized set of colorectal cancer clinical data elements was collected. Data regarding client age, screening history, risk level, screening test indication, results, and recommendation for the next test were analyzed. For colonoscopies, data were analyzed regarding whether the cecum was reached, bowel preparation was adequate, and identified lesions were completely removed. RESULTS Overall, 53% of the fecal occult blood tests (FOBTs) (2295 tests) distributed were completed and returned. At the 2 sites with adequate numbers of FOBTs, 77% and 97%, respectively, of clients with positive results received follow-up colonoscopies. Site-specific cecal intubation rates ranged from 90% to 98%. Adenoma detection rates were 32% for men and 21% for women. For approximately one-third of colonoscopies, the recommended interval to the next test was shorter than recommended by national guidelines. At some sites, endoscopists failed to report on the adequacy of bowel preparation and completeness of polyp removal. CONCLUSIONS Cecal intubation rates and adenoma detection rates met recommended levels. The authors identified the need for improvements in the follow-up of positive FOBTs, documentation of important elements in colonoscopy reports, and recommendations for rescreening or surveillance intervals after colonoscopy. The greater cane rat is a recently domesticated monogastric herbivore, and the importance of the caecum in the functioning of its digestive tract has frequently been mentioned. However, no data are available on caecal fermentative activity of this animal and research on the digestive physiology was only performed in adult animals. The present work was initiated to describe some parameters of the digestive tract anatomy and to study caecal fermentation of the growing cane rat. Thirty 40-day-old weaned cane rats were used in this experiment. Six of them were immediately euthanased and 12 were housed in a collective pen for gastrointestinal tract (GIT) parameters measurements. Twelve others were kept in individual cages to record growth and food intake. Captive cane rats received a pelleted diet containing 12.5% of crude protein and 16.7% of lignocellulose. Six of the 12 collectively housed animals were euthanased on day 82 and a further six on day 103. The growth rate increased with age (P < 0.01) with a linear growth curve (P < 0.001) while the feed intake (g/kg body weight) did not vary (P > 0.05). The caecum represented the largest compartment of the digestive tract constituting more than 40% of total GIT contents. Acetate was the most abundant short-chain fatty acid with more than 70% of the total, followed by propionate (less than 25%) and butyrate (around 5%). Short-chain fatty acid profiles varied with age: when age increased, the acetate proportion increased (P < 0.01), the propionate proportion decreased (P < 0.001) and the butyrate proportion remained unchanged (P > 0.05). The propionate/butyrate ratio showed a decrease with an increase in age (P = 0.05). Ammonia concentrations did not vary with age (P > 0.05). Background: Demonstration of the third duodenal segment (D3) in retroperitoneal location has been recently proposed as a method for excluding malrotation. Objective: This study was performed to determine whether a retroperitoneal third duodenal segment can reliably exclude malrotation. Materials and methods: CTs of 38 patients with proven malrotation and 100 patients without malrotation were evaluated for the location of the duodenum/proximal small bowel, and the relationship of the superior mesenteric vein (SMV) to superior mesenteric artery (SMA). Results: The D3 segment was in normal retroperitoneal location in 100% of control patients, compared to 2.5% or (1 of 38) of patients with malrotation. Nine of 11 patients (91%) with malrotation imaged prior to surgery had the proximal bowel in an abnormal location, while all 100 control patients had it in a normal location. The SMV was in normal relationship to the SMA in 11/38 patients (29%) with malrotation, compared to 79% of normal controls. In 10 controls, a branch of the SMV was partially wrapped around the SMA, potentially mimicking partial mesenteric volvulus. Conclusion: A retroperitoneal location of the D3 segment makes the diagnosis of malrotation unlikely but not impossible. Background and study aims: Independent verification of colonoscopy completion is important for quality assurance. Cecal photographs aimed at showing key landmarks, including the ileocecal valve (ICV) and appendiceal orifice are the currently recommended standard, but are often perceived as unreliable. Images of the terminal ileum demonstrating villi may provide more robust evidence of completion. We sought to prospectively evaluate the ease of routine intubation of the terminal ileum and to compare the effectiveness of terminal ileum and cecal photographs in convincing independent reviewers that total colonoscopy had been accomplished. Patients and methods: A prospective, observational study evaluated 216 consecutively completed colonoscopies performed in routine clinical practice. Cecal and terminal ileum photographs were evaluated and scored by independent reviewers. Frequency of terminal ileum intubation, time required, and safety parameters were recorded. Results: The terminal ileum was intubated and photographed in 188/216 (87 %) of cases. Median time taken to intubate, or attempt to intubate the terminal ileum was 1 min 24 s, and was achieved without complications or requirement for additional sedation. Terminal ileum images were significantly more likely to be considered convincing than cecal images (P < 0.0001 for all reviewers). There was excellent interobserver agreement amongst the opinion of reviewers regarding terminal ileum photographs (kappa = 0.91). Conclusion: Terminal ileum intubation is achievable rapidly and safely in the majority of patients undergoing colonoscopy. Terminal ileum images provide more convincing evidence of complete examination of the colon than cecal images, even when attempts to capture images specifically of the ICV and appendiceal orifice are made. Abstract: TLR signaling plays a role in Salmonella infection, but less information is available in chickens infected with Salmonella serovar Pullorum. The present study with young chickens, experimentally infected with S. Pullorum, has used real-time quantitative RT-PCR to investigate the relative expression of genes of the TLR4 signaling pathway (TLR4, MyD88, TRAF6 and NF-κB) in the spleen and caecum at 1, 3, 7 and 14 days post-infection (dpi). Three-day-old specific-pathogen-free chickens were orally infected with S. Pullorum or saline (controls). In addition to gene expression, the bacterial burden of spleen and caecum was assessed and serum concentrations and total IgG were measured. Significantly more bacteria were found in the caecum than in the spleen. Concentrations of IgG were significantly higher in infected birds at 1dpi, reached a peak at 3dpi (P <0.01), and then gradually decreased with the time. Infected young chickens had significant up-regulation of the expression of TLR4, MyD88, TRAF6 and NF-κB in the spleen at 3dpi compared to controls (P <0.05). In the caecum, only increased expression of NF-κB mRNA (P <0.05) was observed at 1dpi. The pattern of immune response to infection with S. Pullorum differed between spleen and caecum and did not directly correspond with pathogen burden. The observed changes indicate that the MyD88-dependent pathway of TLR4 signaling plays a role in young chickens infected with S. Pullorum. [Copyright &y& Elsevier]

Abstract: This study examined the effects of cholesterol on mouse intestinal microflora and on isoflavonoids in the cecum and plasma. Dietary cholesterol affects bile acid metabolism and bile acids can influence the intestinal microorganisms. Intestinal microflora appear to play an important role in isoflavone metabolism. We hypothesized that dietary cholesterol changes the metabolism of isoflavonoids and intestinal microorganisms in mice. Male mice were randomly divided into two groups, which were fed a cholesterol-daidzein (CDA) or daidzein (DA) diet (control diet) for 60d. Plasma equol and cecal equol concentrations were significantly higher in the DA group (control group) than in the CDA group. However, plasma cholesterol concentrations were significantly higher in the CDA group compared to the DA group. The composition of cecal microorganisms differed between the two dietary groups. The occupation ratios of Clostridium cluster XI, Clostridium subcluster XIVa, and Lactobacillales were significantly higher in the CDA group. The occupation ratio of Bifidobacterium was significantly lower in the CDA group. This study suggests that dietary cholesterol has the potential to affect the metabolism of equol from daidzein by altering the metabolic activity of the intestinal microorganisms and gut physiological function. The main aim of this study was to investigate the occurrence and ribotypes of Clostridium perfringens in broiler flocks reared in 2 European countries that apply European Union Regulation 1831/2003. A total of 1,532 cecum contents were collected between June 2005 and November 2006 from birds belonging to 51 intensively reared flocks produced in the Czech Republic and 41 intensive production, Organic, and freerange flocks reared in Italy. Clostridium perfringens was detected in 64.7 and 82.9% of the Czech Republic and Italian flocks, respectively, at mean loads ranging between 3.65 and 4.77 log10 cfu per gram of cecum content. More than 1 ribotype was identified among isolates belonging to the same flock in 57.1 and 76.5% of the Czech Republic and Italian flocks, respectively. Moreover, common ribotypes were identified between strains belonging to 2 up to 8 different flocks. In particular, 4 ribotypes were shared between strains isolated in the 2 European countries. The results of this study report on C. perfririgens occurrence and mean populations in broilers reared on diets devoid of antibiotic growth promoters. The benefits of bowel preparation prior to capsule endoscopy (CE) are controversial. The aim of this study was to examine whether ingesting a small amount of polyethylene glycol (PEG) during, not before, the CE procedure improves the image quality and the cecal completion rate. A prospective single-blind controlled study was conducted including 59 patients. The initial 32 patients (group A) received no preparation, and the subsequent 27 patients (group B) ingested 500 ml of PEG starting 30 min after swallowing the capsule. The capsule reached the cecum in 65.6% of the patients in group A and 88.9% of the patients in group B (P = 0.038). The use of PEG during CE examination significantly improved the image quality, and this effect was more pronounced in the distal ileum. Two groups have reported >90% of inulin digestion occurs before the cecum in pigs and argued against pigs as a proper animal model for humans in this regard. Two experiments were conducted with weanling pigs to characterize the hydrolysis profile of inulin in their digestive tracts. In Expt. 1, 12 pigs (weighing 7.7 +/- 0.2 kg) were fed a low-iron (54 mg/kg) corn-soy basal diet (BD) or BD + 4% inulin (Synergy 1, Orafti) for 6 wk. All pigs were killed at the end of the trial and digesta samples were collected from the stomach, upper and lower jejunum, cecum, and proximal, mid-, and distal colon. Inulin was detected in digesta from the first 3 segments (0.4-5.5% dry matter) but not from the large intestine of pigs fed inulin. Fructose concentrations in digesta from the stomach and jejunum were greater (P < 0.05) in pigs fed inulin than in those fed BD. To further determine whether inulin was degraded in the ileum or cecum, we conducted Expt. 2 with 12 pigs (weighing 11.2 +/- 1.1 kg) for 8 wk as in Expt.1 except that digesta samples were collected from the ileum instead of upper jejunum. Likewise, inulin was detected only in digesta from stomach, jejunum, and ileum of pigs fed inulin. Although inulin-degrading activity was detectable in digesta from the ileum, cecum, and proximal colon of both groups, the highest activity (P < 0.05) was found in the cecum digesta of pigs fed inulin. Digesta from the cecum and colon, but not from the ileum, was able to degrade added inulin in in vitro incubations. Background: A laparoscopic bowel grasper should be suitable for safely grasping the bowel in a wide variety of patients. Therefore, the inter- and intraindividual variabilities in the strength of bowel tissue to resist perforation force should be analyzed.Methods: The large and small bowels of pigs ( n = 14) and the human small bowel ( n = 7) were clamped between two hemispheres 1.5 mm in diameter. The pinch force was increased until the tissue was perforated.Results: The perforation force for the pig large bowel was higher than for the small bowel (13.5 +/- 3.7 vs 11.0 +/- 2.5 N; p = 0.014). No difference was found between the human and pig small bowel (10.3 +/- 2.9 vs 11.0 +/- 2.5 N). The intercoefficient of variation varied between 22% and 28%, and the intracoefficient of variation varied between 14% and 18%.Conclusions: The strength of the pig bowel is approximately comparable to the strength of the human bowel, and, therefore, testing of graspers on pig bowel is justified. Maize starch was acylated with acetic, propionic or butyric anhydride to produce the corresponding acylated starch. In the first experiment, butyrylated starch at a degree of substitution (DS) of 0.25 (i.e., 1 acyl unit per 4 glucosyl units) was fed to rats for 3 d. Cecal and distal colonic SCFA concentrations were 170 and 78% higher, respectively, in rats fed the butyrylated starch. However, the greatest increase was in butyrate with corresponding increases of 460 and 212%. Subsequently, acetylated, propionylated or butyrylated starches with DS of approximately 0.18 were prepared on a larger scale. Body weight gain did not differ between rats fed these acylated starches or a control starch for 14 d. Large bowel pH was significantly lower and digesta mass significantly higher throughout the large bowel in rats fed the acylated starches. Cecal + distal colonic starch averaged 12 mg in rats fed the control starch and 103, 134 and 135 (pooled SEM = 6) mg in rats fed acetylated, propionylated or butyrylated starch, respectively. Large bowel SCFA concentrations and pools were significantly higher in rats fed the three acylated starches and were disproportionately greater in the SCFA that had been esterified to the starch. In the cecum, acetate, propionate and butyrate pools were 280, 690 and 1060% higher, respectively, in rats fed the corresponding acylated starch than in those fed the control diet. In the distal colon, the corresponding increases were 320, 940 and 1370%. These data indicate that acylated starches are resistant starch (RS) and raise large bowel SCFA, apparently through bacterial release of the esterified fatty acid and fermentation of the residual starch. Recent findings indicate that in addition to its hyperlipemic effect, a high fructose diet has a pro-oxidant effect in rats compared with a starch-based diet. Oligofructose (OFS) has already been shown to decrease plasma lipids in rats. We assessed the impact of fructose on oxidative stress by supplementing a high fructose diet with OFS. Rats were fed either a high fructose diet or a starch-based diet, with or without supplementation of 10 g/100 g oligofructose for 4 wk. Regardless of the type of carbohydrate, OFS in the diet produced an enlargement of the cecum and led to a significant increase in the SCFA cecum pool. Fructose feeding was associated with significantly higher insulin plasma concentrations (+63%) in the control groups, whereas insulin plasma concentrations did not differ in rats fed the fructose diet supplemented with OFS. Plasma leptin concentration was significantly lower (approximately 50%) in the OFS-supplemented fructose group compared with the other three groups. Fructose feeding in rats also significantly increased plasma (P < 0.001) and liver (P < 0.001) triglyceride (TG) concentrations and the addition of OFS prevented the TG accumulation induced by fructose in the liver (P < 0.05) and hyperlipemia (P < 0.05). OFS consumption prevented (P < 0.05) the lower plasma vitamin E/TG ratio in rats fed the fructose diet. Control rats fed the fructose diet had high plasma TBARS values compared with rats fed the starch diet, whereas TBARS values remained unchanged when rats were supplemented with OFS. Control rats fed the fructose diet had higher TBARS urine values and higher heart tissue susceptibility to peroxidation compared with rats fed the starch diet, and this effect was significantly reduced by OFS consumption. Further studies are required to identify the mechanisms underlying the protective effect of OFS against the pro-oxidant effect of fructose. Gluconic acid reaches the large intestine to stimulate lactic acid bacteria. However, the fermentation pattern of gluconic acid has yet to be elucidated. Accordingly, we examined the fermentation properties induced by gluconic acid in the pig cecal digesta in vitro. We also tested sorbitol and glucose, substrates for which the fermentation rate and patterns are known. The gluconic acid-utilizing bacteria were further isolated from pig cecal digesta and identified to examine the effect of gluconic acid on hind gut fermentation. Gluconic acid was fermented more slowly than were the other two substrates. Gluconic acid stimulated butyrate production; the butyrate molar percentage reached 26%, which is considered a high butyrate production. The majority of gluconic acid fermenters were identified as lactic acid bacteria, such as Lactobacillus reuteri and L. mucosae, and acid-utilizing bacteria, such as Megasphaera elsdenii and Mitsuokella multiacida. The gluconic acid fermented by lactic acid bacteria, and the lactate and acetate that were produced were used to form butyrate by acid-utilizing bacteria, such as M. elsdenii. Dietary short-chain fructooligosaccharides (Sc-FOS) stimulate absorption of calcium and magnesium in the large intestine of rats. In this study, we examined whether Sc-FOS stimulate iron absorption in the large intestine by monitoring recovery from anemia in gastrectomized rats, with or without cecectomy. The rats were divided into four groups, i.e., sham-operated (Sham), gastrectomized only (GX), cecectomized only (CX) and both gastrectomized and cecectomized (GCX). Half of the rats in each group were fed a control diet (AIN-93G) and the other half were fed a Sc-FOS-containing diet (75 g/kg diet) for 28 d. Hematocrit (Ht) and hemoglobin concentration (Hb) were measured at the start, and on d 14 and 28 after the start of feeding. On the final day of the study, total blood was collected. Gastrectomy significantly decreased Ht and Hb, but cecectomy did not influence these variables. Dietary Sc-FOS prevented the decrease in Ht and Hb significantly. In the gastrectomized rats, the effectiveness of Sc-FOS in preventing postgastrectomy anemia was significantly diminished by cecectomy. These results suggest that the effect of Sc-FOS in increasing absorption of iron in gastrectomized rats takes place in part in the cecum. Maltitol is fermented in the colon due to only partial hydrolysis in the small intestine. In the present study, we examined effects of dietary maltitol on dimethylhydrazine-induced intestinal tumor in rats. In experiment 1, rats were fed a fiber-free diet or diets supplemented with 1 or 5 g/100 g maltitol for 27 wk. Each group of rats was injected with dimethylhydrazine or vehicle alone for the first 14 wk of the experimental period. Maltitol supplementation at 1 g/100 g of the diet significantly reduced tumor incidence in the cecum and the 5% supplement reduced tumor incidence in both the cecum and proximal colon in dimethylhydrazine-treated rats. In experiment 2, we investigated the effect of the 1 g/100 g maltitol diet on the short chain fatty acid concentrations in cecal contents of placebo and dimethylhydrazine-treated rats. Intake of the 1 g/100 g maltitol diet doubled (P < 0.05) the concentration of butyrate but did not affect acetate or propionate in the cecal contents. These results suggest that dietary maltitol has a protective effect against dimethylhydrazine-induced tumors in rat cecum and proximal colon and that butyrate produced by bacterial fermentation of maltitol in the cecum may be involved in the protection. Aims: Non-invasive and simultaneous recording of gastrointestinal (GI) activity during stress induction is still an unexplored field. In our previous investigation, the stress-induced alteration of the gastrointestinal tract was explored in rats. Our aims were to expand our previous rat experiment and to induce stress response in rats (Study 1) and humans (Study 2) to detect the GI tract activity, heart rate and body temperature., Materials and Methods: In the preclinical sample, acute stress was induced by immobilization in Sprague-Dawley rats (N = 10). Acute stress response was generated by the Trier Social Stress Test among healthy volunteers (N = 16). Detection of acute stress was measured by using smooth muscle electromyography, which recorded the myoelectric waves of the gastrointestinal tract (stomach, ileum and colon) simultaneously with heart rate and body temperature in rats and humans., Key Findings: The myoelectric waves of the stomach, the cecum and the ileum increased during immobilization in rats, rising in parallel with heart rate and the dermal temperature of the abdominal surface. The same alterations were found during the stress period among humans, except in the case of the colon, where no change was detected., Significance: The crucial role of the GI tract in stress response was revealed by translating the outcome of basic research into human results. The similar GI alterations during stress in rats and humans underpin the robustness of our findings. Sepsis is a major clinical challenge with unacceptably high mortality. The signal transducers and activators of transcription (STAT) family of transcription factors is known to activate critical mediators of cytokine responses, and, among this family, STAT3 is implicated to be a key transcription factor in both immunity and inflammatory pathways. We investigated whether in vivo introduction of synthetic double-stranded STAT3 decoy oligodeoxynucleotides (ODNs) can provide benefits for reducing organ injury and mortality in mice with cecal ligation and puncture (CLP)-induced polymicrobial sepsis. We found that STAT3 was rapidly activated in major end-organ tissues following CLP, which was accompanied by activation of the upstream kinase JAK2. Transfection of STAT3 decoy ODNs downregulated pro-inflammatory cytokine/chemokine overproduction in CLP mice. Moreover, STAT3 decoy ODN transfection significantly reduced the increases in tissue mRNAs and proteins of high mobility group box 1 (HMGB1) and strongly suppressed the excessive elevation in serum HMGB1 levels in CLP mice. Finally, STAT3 decoy ODN administration minimized the development of sepsis-driven major end-organ injury and led to a significant survival advantage in mice after CLP. Anorexia nervosa (AN) is a psychiatric disorder that presents with profound weight dysregulation, metabolic disturbances, and an abnormal composition of gut microbial communities. As the intestinal microbiota can influence host metabolism, the impact of enteric microbial communities from patients with AN on host weight and adiposity was investigated. Germ-free (GF) mice were colonized with fecal microbiotas from either patients with AN (n = 4) prior to inpatient treatment (AN T1, n = 50 recipient mice), the same 4 patients following clinical renourishment (AN T2, n = 53 recipient mice), or age- and sex-matched non-AN controls (n = 4 human donors; non-AN, n = 50 recipient mice). Biological and fecal microbiota data were analyzed with linear mixed-effects models. Body weight did not differ significantly between AN recipient mice (T1 and T2) and non-AN recipient mice following 4 weeks of colonization. Enteric microbiotas from recipient mice colonized with AN T1 and AN T2 fecal microbiotas were more similar to each other compared with enteric microbiotas from non-AN recipient mice. Specific bacterial families in the Actinobacteria, Bacteroidetes, and Firmicutes phyla were significantly associated with body weight, fat mass, and cecum weight irrespective of the donor group. These data suggest that body weight, fat mass, and cecum weight of colonized GF mice are associated with human fecal microbes and independent of donor AN status, although additional analyses with larger cohorts are warranted.

The gut-microbiota-brain axis is implicated in the development of behavioural disorders in mammals. As such, its potential role in disruptive feather pecking (FP) in birds cannot be ignored. Birds with a higher propensity to perform FP have distinct microbiota profiles and feed transit times compared to non-pecking counterparts. Consequently, we hypothesize that the gut microbiota is intimately linked to FP and gut motility, which presents the possibility of using probiotics to control FP behaviour. In the present study, we aim to assess the relationship between cecal motility and the probiotic Lactobacillus rhamnosus in chickens classified as peckers (P, 13 birds) and non-peckers (NP, 17 birds). We show that cecal contractions were 68% less frequent and their amplitude increased by 58% in the presence of L. rhamnosus. Furthermore, the number of FP bouts performed by P birds was positively correlated with contraction velocity and amplitude. We present the first account of gut motility measurements in birds with distinct FP phenotypes. Importantly, the present work demonstrates the clear impact of a probiotic on cecal contractions. These findings lay the foundation for identifying biological differences between P and NP birds which will support the development of FP control strategies.

Problem: Gut dysbiosis is caused by several factors, including the use of antibiotics. Since intestinal dysbiosis is associated with a wide range of immunopathological and reproductive conditions, the main goal of this study was to evaluate amoxicillin-induced gut dysbiosis and its influence on the oestrous cycle in mice., Method of Study: Mice were treated with amoxicillin or PBS, and faecal microbiota was evaluated by 16S rDNA metagenomic sequencing. The oestrous cycle was evaluated by vaginal cytology, vaginal opening and flow cytometry. After the induction of gut dysbiosis, the ovaries and the caecum were analysed to differential expression of IL-1β and IL-10 genes and histological analysis., Results: Amoxicillin-treated mice presented differing bacterial groups in the faecal microbiota when compared to the PBS-treated group indicating that amoxicillin treatment-induced gut dysbiosis and they gained weight. The vaginal cytology analysis showed that amoxicillin-induced gut dysbiosis decreased the number of cells but increased the relative number of leucocytes and altered the oestrous cycle. IL-1β was shown to be upregulated in the caecum and in the ovary of the dysbiotic mice. On the other hand, IL-10 expression was shown to be diminished in both organs of the dysbiotic mice. The oocyte area from dysbiotic group presented lower than non-dysbiotic mice with increasing thickness of the pellucid zone. The follicular teak from dysbiotic mice showed lower thickness than non-dysbiotic mice., Conclusion: The results indicate that amoxicillin induces gut dysbiosis and influences the oestrous cycle and the inflammatory status of the ovary and the caecum., (© 2020 John Wiley & Sons A/S. A study was conducted to determine effects of reducing hindgut pH through dietary inclusion of high-amylose cornstarch (HA-starch) on growth performance, organ weights relative to live body weight (BW), blood thyroid hormone levels, and glucosinolate degradation products of nursery pigs fed cold-pressed canola cake (CPCC). A total of 240 pigs (initial BW: 7.1 kg), which had been weaned at 21 d of age, were housed in 40 pens (6 pigs per pen) and fed 4 diets (10 pens per diet) in a randomized complete block design for 28 d. Four diets were a basal diet with CPCC at 0 or 40%, and with HA-starch at 0 or 40% in a 2 × 2 factorial arrangement. The diets were fed in two phases: Phase 1 from day 0 to 14 and Phase 2 from day 14 to 28 and were formulated to have the same net energy, standardized ileal digestible AA, Ca, and standardized total tract digestible P contents. Dietary inclusion of CPCC and HA-starch was achieved by a partial or complete replacement of corn, soybean meal, and soy protein. At the end of the study, one pig from each pen was euthanized to determine organ weights, blood parameters, hindgut pH, and glucosinolate degradation products. Dietary CPCC reduced (P < 0.05) overall average daily gain (ADG) by 15%; increased (P < 0.05) relative weights of liver and thyroid gland by 27% and 64%, respectively; and reduced (P < 0.05) serum tetraiodothyronine (T4) level from 30.3 to 17.8 ng/mL. Heart, kidney, and gastrointestinal tract weights; serum triiodothyronine level; and hindgut pH of pigs were unaffected by dietary CPCC. Dietary HA-starch reduced (P < 0.05) overall ADG, relative weight of thyroid gland, cecal, and colonic pH; but increased (P < 0.05) relative weight of colon; tended to increase (P = 0.062) serum T4 level. Dietary CPCC and HA-starch interacted (P = 0.024) on relative weight of thyroid gland such that dietary CPCC increased (P < 0.05) weight of thyroid gland for HA-starch-free diet (120 vs. 197 mg/kg of BW) but not for HA-starch-containing diet (104 vs. 130 mg/kg of BW). Dietary CPCC and HA-starch interacted (P = 0.001) on cecal isothiocyanate content such that dietary CPCC increased (P < 0.05) level of isothiocyanates for HA-starch-containing diet but not for HA-starch-free diet. In conclusion, dietary CPCC reduced growth performance, increased liver, size and interfered with thyroid gland functions of pigs. The intent of this study was to investigate the effects of cold stress on oxidative indexes, inflammatory factors, and microbiota in the quail cecum. A total of 192 male quails (15-day-old) were randomly divided into 12 groups (16 in each group) and were exposed to acute (up to 12 h) and chronic (up to 20 D) cold stress at 12 ± 1°C. After cold stress treatment, we examined morphological damage, oxidative stress indexes, inflammatory factors, and intestinal microbiota. Results of morphological examination showed that both acute and chronic cold stress can lead to cecal tissue injury. In addition, both acute and chronic cold stress, especially chronic cold stress can influence the activity of oxidative stress mediators. Glutathione (GSH) and glutathione peroxidase (GSH-Px) activities decreased significantly (p < 0.05), while the nitric oxide (NO) content and inducible nitric oxide synthase (iNOS) activity increased significantly (p < 0.05). Moreover, mRNA levels of inflammatory factors cyclooxygenase-2 (COX-2), prostaglandin E synthase (PTGES), and heat shock protein 70 (Hsp70) were higher in both acute and chronic cold stress groups when compared with the control group (p < 0.05). Furthermore, the intestinal microbiota was changed in both the acute and chronic cold stress groups. An experiment was conducted to evaluate the effects of age and supplemental xylanase in corn- or wheat-based diets on cecal volatile fatty acid (VFA) concentrations of Ross × Ross 708 male broilers during weekly intervals from 14 to 42 d of age. Day-old chicks (1,500) were randomly distributed into 60 floor pens (25 chicks/pen; 0.078 m2/bird) and fed 1 of 4 dietary treatments (TRT) throughout the starter (1 to 14 d of age), grower (15 to 28 d of age), and finisher (29 to 42 d of age) phases with 15 replicates per TRT. Dietary TRT consisted of a 2 × 2 factorial arrangement with 2 diet types (corn- or wheat-based) and 2 xylanase inclusions (0 or 16,000 BXU/kg) as the main factors. At 14, 21, 28, 35, and 42 d of age, cecal contents were collected (4 birds/pen) for VFA analysis. Main effects of cereal grain source (P < 0.05) affected propionic, isobutyric, butyric, isovaleric, valeric, and isocaproic acid concentrations at 14, 21, 28, 35, and 42 d of age. Broilers fed corn-based diets had higher (P < 0.05) propionic, isobutyric, isovaleric, valeric, and isocaproic concentrations than those fed wheat-based diets from 14 to 42 d of age. However, broilers fed wheat-based diets had higher (P < 0.05) butyric acid concentrations at 28, 35, and 42 d of age compared with those fed corn-based diets. Individual and total VFA concentrations increased (P < 0.05) linearly from 14 to 42 d of age. Age and cereal grain interacted (P < 0.05) to affect propionic, isobutyric, butyric, isovaleric, and valeric acid concentrations. These results indicate that broiler cecal VFA concentrations are influenced by cereal grain source and age. In contrast, supplemental xylanase inconsistently influenced broiler cecal VFA concentrations. Therefore, future research evaluating factors affecting supplemental xylanase and cecal VFA production in broilers is warranted. The aim of this study was to examine the effect of a dietary synbiotic supplement on the cecal microflora, antioxidant status, and immune response of broiler chickens under heat stress (HS). A total of 360 one-day-old male Ross 708 broiler chicks were randomly distributed among 3 dietary treatments containing a synbiotic (PoultryStar consists of Bifidobacterium animalis, Enterococcus faecium, Lactobacillus reuteri, Pediococcus acidilactici, and fructooligosaccharides) at 0 (control), 0.5 (0.5X), and 1.0 (1.0X) g/kg. Each treatment contained 8 replicates of 15 birds each housed in floor pens. Heat stimulation was at 32°C for 9 h daily from day 15 to 42. Heat stress-induced changes of cecal bacteria were detected using bacteria-specific agars, and spleen protein concentration and mRNA expression of interleukins and antioxidants were examined using ELISA and real-time PCR, respectively. Under the HS condition, synbiotic fed broilers regardless of dose had lower cecal enumerations of Escherichia coli and coliforms, and a lower heterophil/lymphocyte (H/L) ratio (P < 0.05) compared to controls. 1.0X group also had higher cecal enumerations of Bifidobacterium spp. and Lactobacillus spp., spleen glutathione peroxidase (GPx), and plasma nuclear factor erythroid 2-related factor 2 (Nrf-2), and a lower H/L ratio compared to both control and 0.5X groups (P < 0.05). However, there were no treatment effects on the levels of Enterococcus spp., the circulating monocytes, eosinophils, and basophils, Toll like receptor-4 (TLR-4), interleukin-6 (IL-6), interlukin-10 (IL-10), and their mRNA expression, as well as plasma Kelch-like ECH-associated protein 1 (Keap-1) (P > 0.05).
