Abstract Restoration of vulnerable marine habitats is becoming increasingly popular to cope with widespread habitat loss and the resulting decline in biodiversity and ecosystem services. Lately, restoration strategies have been employed to enhance the recovery of degraded meadows of the Mediterranean endemic seagrass Posidonia oceanica. Typically, habitat restoration success is evaluated by the persistence of foundation species after transplantation (e.g., plant survival and growth) on the short and long-term, although successful plant responses do not necessarily reflect the recovery of ecosystem biodiversity and functions. Recently, soundscape (the spatial, temporal and frequency attribute of ambient sound and types of sound sources characterizing it) has been related to different habitat conditions and community structures. Thus, a successful restoration action should lead to acoustic restoration and soundscape ecology could represent an important component of restoration monitoring, leading to assess successful habitat and community restoration. Here, we evaluated acoustic community and metrics in a P. oceanica restored meadow and tested whether the plant transplant effectiveness after one year was accompanied by a restored soundscape. With this goal, acoustic recordings from degraded, transplanted and reference meadows were collected in Sardinia (Italy) using passive acoustic monitoring devices. Soundscape at each meadow type was examined using both spectral analysis and classification of fish calls based on a catalogue of fish sounds from the Mediterranean Sea. Seven different fish sounds were recorded: most of them were present in the reference and transplanted meadows and were associated to Sciaena umbra and Scorpaena spp. Sound Pressure Level (SPL, in dB re: 1 μPa-rms) and Acoustic Complexity Index (ACI) were influenced by the meadow type. Particularly higher values were associated to the transplanted meadow. SPL and ACI calculated in the 200–2000 Hz frequency band were also related to high abundance of fish sounds (chorus). These results showed that meadow restoration may lead to the recovery of soundscape and the associated community, suggesting that short term acoustic monitoring can provide complementary information to evaluate seagrass restoration success.

In this study, we explored the fish acoustic community at two coralligenous sites in Sardinia (Tavolara and Santa Teresa, western Mediterranean Sea) during the summer 2023. Our goal was to understand spatial and temporal patterns of fish acoustic activity on different temporal scales to offer insights for optimizing acoustic monitoring of this crucial ecosystem. We identified seventeen distinct sound types, revealing a diverse acoustic community. Tavolara had higher acoustic richness and abundance compared to Santa Teresa, which may be attributed to site-specific factors such as habitat structure, species composition, or levels of protection. Temporal analysis revealed clear diel patterns, with certain sounds associated with nocturnal or diurnal periods, reflecting the daily rhythms of different species. The study also examined how recording duration influenced acoustic richness, finding that longer recordings (15 minutes per hour and 15 minutes per hour at night) provided a more comprehensive detection of acoustic activity. Additionally, the number of recording days required to detect species richness varied depending on the site. While extended recordings improve the likelihood of detecting rare or sporadic sounds, they also present challenges in data management and equipment maintenance. The study underscores the importance of carefully planning sampling strategies to optimize acoustic monitoring and ensure effective and sustainable ecological research in coralligenous ecosystems.

Abstract Marine protected areas (MPAs) make an essential contribution to the spatial management of critical areas, the conservation of coastal species exploited by human activities, and the sustainable use of marine resources. Within MPAs, fishing closure areas are among the most used small‐scale fishery management tools, even though their effectiveness largely remains untested or controversial. To reduce the impact of small‐scale fisheries on marine resources, a seasonal fishing closure area (SFCA) was established beginning in 2022 in autumn–winter season inside the Capo Caccia–Isola Piana MPA (Sardinia, northwestern Mediterranean Sea). Here, we assessed a posteriori whether the areas of higher habitat suitability for eight species/taxa of relevant ecological value and economic interest to small‐scale fisheries were included in the established SFCA, adequately meeting the ecological objectives of the MPA. Thus, landing data (from 2019 to 2023) were used as occurrence records to develop MaxEnt distribution models for the eight target species/taxa. The model outputs allow us to draw important insights about the spatial adequacy of the SFCA established within the MPA aimed to protect the most exploited marine resources. Furthermore, the modeling exercises were useful for understanding the local processes influencing species' habitat selection and to identify essential areas for the target species that could remain unrevealed in larger‐scale investigations.

In the framework of mapping underwater environments, Posidonia Oceanica (PO) is a fundamental indicator for environmental monitoring and related studies and analyses for climate change. The POSEIDON project aims to study, develop and test new methodologies to provide knowledge, as well as ultra-high-resolution (UHR) and beyond ultra-high-resolution (BUHR) mapping and monitoring data for the conservation of the PO and related meadows and banquettes in the Mediterranean area. The project is funded by the National Recovery and Resilience Plan and stems from the recommendation of international organizations (European Environmental Directives, Mediterranean PO Network, International Partnership for Blue Carbon, Intergovernmental Oceanographic Commission of UNESCO in the Save the Wave project). Starting from new integrated survey methods and legacy data, the PO, ichthyofauna and seagrass banquettes will be mapped to understand their evolution and extract indicators and useful trends. Furthermore, the produced information will be stored and shared in a webGIS and a dedicated app to prevent unexpected damages during sailing or other activities. POSEIDON will use imaging sensors (RGB and multi/hyperspectral cameras) to extract information. With specific lighting equipment for significant water depths, these sensors will be conducted (by scuba divers or Autonomous Underwater Vehicles) next to the object to acquire BUHR data. Finally, to monitor the fauna (fish, other species, invertebrates), terrestrial and underwater videogrammetry will be applied using mono and stereo-cameras with object detection algorithms.

Brief Introduction to the Thematic Block

Seagulls are synanthropic wild birds that can contaminate, through their droppings, beaches, urban and peri-urban environments. This concern is more serious when seagulls eliminate antimicrobial-resistant pathogenic bacteria. This study analyzed the fecal samples from 137 yellow-legged seagulls (Larus michahellis) from Central Italy. A total of 218 Escherichia coli strains were isolated and analyzed for phenotypic and genotypic antimicrobial resistance and to identify the virulence genes characterizing different pathotypes. The disk diffusion method on all isolates found relevant resistance rates to ampicillin (38.99%), tetracycline (23.85%), and enrofloxacin (21.10%). On the basis of all results obtained with this test, 62 (28.44%) isolates were classified as multidrug-resistant (MDR) and 6 (2.75%) as extensive drug-resistant (XDR). Molecular analyses conducted on the strains phenotypically resistant to carbapenems, cephalosporins, and penicillins found 9/37 (24.32%) strains positive for blaOXA-48, 52/103 (50.49%) for blaTEM, 12/103 (11.65%) for blaCMY2, 3/103 (2.91%) for blaCTX, and 1/103 (0.97%,) for blaSHV. PCR to detect virulence genes characterizing different pathotypes found that 40 (18.35%) isolates had the astA gene, indicative of the enteroaggregative (EAEC) pathotype, 2 (0.92%) had cnf1, 2 (0.92%) had cnf2, and 1 (0.46%) had cdt-IV. All five (2.29%) strains were reportable as necrotoxigenic (NTEC), while 4 (1.83%) had both eaeA and escV, reportable as enteropathogenic (EPEC). Measures to limit seagulls’ access where humans and other animals reside are pivotal to reduce the risk of infection with antimicrobial-resistant and pathogenetic E. coli strains.

Lumbar spinal stenosis (LSS) occurs when bony, ligamentous, and synovial elements of the lower axial spine degenerate and overgrow, compressing neural and vascular elements in the spinal canal. Compression can cause static back pain, radicular lower extremity pain, or neurogenic claudication. Radiological and clinical findings are needed to diagnose lumbar stenosis. In this framework, caudal epidural steroid injection (ESI) is a standard treatment. The volume injected and needle positioning are the main issues that could compromise the effectiveness of the epidural injection. However, ultrasound-guided caudal epidural injections have become more common in recent years. Since Klocke and colleagues described the ultra-sound-guided caudal block in 2003, it has grown in popularity. Multiple ethnic studies have reported ultrasound-guided caudal injection success rates of 96.9–100%. Color Doppler ultrasonography can also determine if a drug solution reaches the lumbosacral region. We enrolled 42 patients with lumbar spinal stenosis, persistent lumbosciatalgia, and neurogenic claudicatio unresponsive to painkillers who were not surgical candidates. Each patient receives four weekly injections for four weeks. If the patient responds to treatment but still has pain, monthly injections are needed to reach and maintain the benefit. Treatment will be changed if the patient does not respond after 4 caudal injections. Sterile caudal epidural injections are performed with ultrasound guidance and a spinal needle 21G. Triamcinolone 40 mg, levobupivacaine 10 mg, and physiological solution 10 ml are delivered. Each patient received an average of 4 injective treatments (4±2, Means 4, SD 2). Data analysis shows that the treatment reduced pain significantly before and after therapy, which persisted after 3 months. Caudal epidural injection is one of the most common conservative treatments for chronic low back pain with radiculopathy, and lidocaine alone or with steroids is effective. In this framework, the two main literature issues about caudal epidural injection effectiveness on lumbar pain are correct. Therefore, 10 ml is suitable and effective for treating symptoms without side effects. Pain reduction of over 50% from start to finish and three-month follow-up have shown significant results in pain control and disability improvement. Finally, caudal epidural injection for lumbar spinal stenosis symptoms is effective, safe, and provides long-term pain relief.

Birds are known to be carriers of ticks infected by tick-borne pathogens, including bacteria. However, not many studies have been carried out on avian tissues to detect these agents. The aim of the present survey was to investigate, using PCR, the presence of Anaplasma phagocytophilum, Bartonella spp., Borrelia burgdorferi sensu lato, Chlamydia psittaci, Coxiella burnetii, Ehrlichia canis, Francisella tularensis, and Rickettsia spp. in the spleens collected from 300 wild birds of different orders and species from Central Italy. A total of 53 (17.67%) samples were PCR positive for at least one investigated pathogen. One (0.33%) bird was positive for Bartonella spp., five (1.67%) birds were positive for C. burnetii, eleven (3.67%) for B. burgdorferi s.l., and thirty-six (12%) for C. psittaci. No coinfection was detected. All samples were negative for A. phagocytophilum, E. canis, F. tularensis, and Rickettsia spp. The findings showed that wild birds may harbor different zoonotic tick-borne bacteria; therefore, they can contribute to the diffusion of these agents.

Abstract Global warming is expected to have inexorable and profound effects on marine ecosystems, particularly in foundation species such as seagrasses. Identifying responses to warming and comparing populations across natural temperature gradients can inform how future warming will impact the structure and function of ecosystems. Here, we investigated how thermal environment, intra-shoot and spatial variability modulate biochemical responses of the Mediterranean seagrass Posidonia oceanica. Through a space-for-time substitution study, Fatty acid (FA) profiles on the second and fifth leaf of the shoots were quantified at eight sites in Sardinia along a natural sea surface temperature (SST) summer gradient (about 4 °C). Higher mean SST were related to a decrease in the leaf total fatty acid content (LTFA), a reduction in polyunsaturated fatty acids (PUFA), omega-3/omega-6 PUFA and PUFA/saturated fatty acids (SFA) ratios and an increase in SFA, monounsaturated fatty acids and carbon elongation index (CEI, C18:2 n-6/C16:2 n-6) ratio. Results also revealed that FA profiles were strongly influenced by leaf age, independently of SST and spatial variability within sites. Overall, this study evidenced that the sensitive response of P. oceanica FA profiles to intra-shoot and spatial variability must not be overlooked when considering their response to temperature changes.

Where sea urchin harvest has been so intense that populations have drastically regressed, concerns have arisen about the effectiveness of harvesting management. According to the theory of phase transition in shallow rocky reefs between vegetated and barren habitats, sea urchin recruitment, a key population structuring process, seems hampered by some stabilizing feedback despite an end to local human harvest of sea urchins. To shed a light on predation effects on sea urchin recruits, a 27-day field experiment was conducted using mega-predator exclusion cages (40x40x40 cm, 1 cm in mesh size) in barren and turf substrates. To facilitate this, 672 recruits (1.1 ± 0.02 cm in size) reared under control conditions were positioned in groups of 42 in each experimental unit (n = 4). Exclusion of mega-predators had a significant effect regardless the substrate, since a higher number of recruits was found under cages both in turf and barren. However, the results showed that in uncaged treatments the size of recruits that survived was larger in turf than in barren, as in the former substrate predation had reduced the abundance of the smallest recruits, highlighting that mega-predator presence affects differently the size of the recruits that had survived depending on the substrate. Overall, these results provide valuable information to address restocking actions of sea urchin populations in overharvested areas, where algal turfs are widespread, and assist studies on habitat stability mechanisms.

The harvest of the edible sea urchin Paracentrotus lividus is intensively practiced in some regions of the Western Mediterranean Sea. The removal of the largest individuals can determine an overall reduction in population size and a size class truncation that can lead to a drastic drop the self-sustenance. The aim of this study is to evaluate the variability of the population reproductive potential across 5 years in one of the main harvest hotspots of Sardinia (Western Mediterranean Sea). The breeding stock consists of commercial and under-commercial size individuals which were sampled on a monthly basis to estimate their GonadoSomatic Index (GSI) and the Individual Gamete Output (IGO). In addition, the reproductive potential of the population—Total Gamete Output (TGO)—was calculated across the 5-year period in relation with the variation of the density of the breeding stock. During the last year, the reproductive potential was also estimated in a well-conserved population of a nearby Marine Protected Area. No significant variability in GSI and IGO was found over the 5 years nor when compared with the ones of protected population in the last year. However, the intensive harvest drastically rescaled the population body-size: although density of the commercial size class remained low, density of the under-commercial size-class halved from the beginning to the end of the study. Accordingly, the proportional decrease of their gamete output contribution led to a 40% loss of the reproductive potential of the whole population in the 5-year period. Interestingly, despite the loss of reproductive potential due to the decrease of the breeding stock density, the average values of IGO slightly increased across the years leading to the highest Annual Gamete Output (AGO) during the fourth year of sampling. This positive pattern could suggest a mechanism of reproductive investments of the survivors in terms of gonad production rate or increase in spawning intensity. This work provides evidence of the direct effect of size-selective harvesting on the rapid loss of population self-sustenance. Furthermore, it lays new prospective for future research of the indirect effects of the rescaling population body-size in functional traits of the sea urchin P. lividus and that could become important for both, sustainable exploitation and ecosystem conservation management.

Background Sustainable management requires spatial mapping of both species distribution and human activities to identify potential risk of conflict. The common bottlenose dolphin (Tursiops truncatus) is a priority species of the European Union Habitat Directive, thus, to promote its conservation, the understanding of habitat use and distribution, as well as the identification and spatial trend of the human activities which may directly affect populations traits, is pivotal. Methods A MaxEnt modeling approach was applied to predict the seasonal (from April to September) habitat use of a small population of bottlenose dolphins in the north-western Sardinia (Mediterranean Sea) in relation to environmental variables and the likelihoods of boat and fishing net presence. Then, the overlapping areas between dolphin, fishing net and boat presence were identified to provide insights for the marine spatial management of this area. Results Three of the main factors influencing the seasonal distribution of bottlenose dolphins in the area are directly (boating and fishing) or indirectly (ocean warming) related to human activities. Furthermore, almost half of the most suitable area for dolphins overlapped with areas used by fishing and boating. Finally, relying on fishing distribution models, we also shed light on the potential impact of fishing on the Posidonia oceanica beds, a protected habitat, which received higher fishing efforts than other habitat types. Discussion Modelling the spatial patterns of anthropogenic activities was fundamental to understand the ecological impacts both on cetacean habitat use and protected habitats. A greater research effort is suggested to detect potential changes in dolphin habitat suitability, also in relation to ocean warming, to assess dolphin bycatch and the status of target fish species, and to evaluate sensitive habitats conditions, such as the Posidonia oceanica meadow.

Abstract Abiotic environmental conditions can significantly influence the way species interact. In particular, plant–herbivore interactions can be substantially dependent on temperature and nutrients. The overall product of these relationships is critical for the fate and stability of vegetated ecosystems like marine forests. The last few decades have seen a rapid spread of barrens on temperate rocky reefs mainly as a result of overgrazing. The ecological feedbacks that characterize the barren state involve a different set of interactions than those occurring in vegetated habitats. Reversing these trends requires a proper understanding of the novel feedbacks and the conditions under which they operate. Here, we explored the role of a secondary herbivore in reinforcing the stability of barrens formed by sea urchin overgrazing under different nutrient conditions. Combining comparative and experimental studies in two Mediterranean regions characterized by contrasting nutrient conditions, we assessed: (i) if the creation of barren areas enhances limpet abundance, (ii) the size‐specific grazing impact by limpets, and (iii) the ability of limpets alone to maintain barrens. Our results show that urchin overgrazing enhanced limpet abundance. The effects of limpet grazing varied with nutrient conditions, being up to five times more intense under oligotrophic conditions. Limpets were able to maintain barrens in the absence of sea urchins only under low‐nutrient conditions, enhancing the stability of the depauperate state. Overall, our study suggests a greater vulnerability of subtidal forests in oligotrophic regions of the Mediterranean and demonstrates the importance of environment conditions in regulating feedbacks mediated by plant–herbivore interactions.

Active Points Test on auricle was renamed Needle Contact Test (or NCT), considered an extremely effective method to select most efficient auricle acupoint for Auricular Acupuncture (or AA) therapy. Our aims were to evaluate NCT efficiency as viable diagnostic option to identify the most appropriate AA acupoints to be selected for SupraSpinatus Tendinopathy (SST) associated shoulder pain treatment and to evaluate which auricle area has greater concentration of NCT positive acupoints in subjects with SST shoulder pain, comparing results with available AA ear maps. 45 subjects with SST diagnosis were enrolled. On 30 subjects in treatment group, NCT was performed on acupoints of the auricle shoulder representation areas (i.e., Scaphoid Fossa), while in 15 subjects of control group, NCT was performed on auricle areas not shoulder associated. Statistically significant lower Numeric Pain Rating Scale scores were seen for study group over time and compared to control. A small sample showed statistically significant increase of SST shoulder maximum abduction range of Movements. NCT identified best SST shoulder pain AA acupoints in Scaphoid Fossa (or SF1 and SF2). In conclusion NCT is a viable tool to efficiently identify the best AA acupoints in SST shoulder pain treatment.

Abstract One of the most studied aspects of animal communication is the acoustic repertoire difference between populations of the same species. While numerous studies have investigated the variability of bottlenose dolphin whistles between populations, very few studies have focused on the signature whistles alone and the factors underlying differentiation of signature whistles are still poorly understood. Here we describe the signature whistles produced by six distinct geographical units of the common bottlenose dolphin (Tursiops truncatus) in the Mediterranean Sea and identify the main determinants of their variability. Particularly, the influence of the region (proxy of genetic distance), the geographic site, and the environmental (sea bottom-related) and demographical (population-related) conditions on the acoustic structure of signature whistles was evaluated. The study provides the first evidence that the genetic structure, which distinguishes the eastern and western Mediterranean bottlenose dolphin populations has no strong influence on the acoustic structure of their signature whistles, and that the geographical isolation between populations only partially affected whistle variability. The environmental conditions of the areas where the whistles developed and the demographic characteristics of the belonging populations strongly influenced signature whistles, in accordance with the “acoustic adaptation hypothesis” and the theory of signature whistle determination mediated by learning.

Mapping of coralligenous banks was carried out along the continental shelf of the northern and western margin of Sardinia Island (Italy, western Mediterranean Sea) in the context of the European Marine Strategy Framework Directive (MSFD, 2008/56/EC). Coralligenous banks are bioconstructions produced by calcareous coralline algae. Seafloor mapping was carried out through multibeam echosounder surveys and video transects, using a Remote Operating Vehicles (ROV), in areas not formerly explored. A high-resolution digital model of the seabed (DTM) was obtained from multibeam data. A total surface of 436 km2 of sparse patches of coralligenous banks was mapped in the depth range ~40-160 m. A final map of coralligenous habitat distributions along the western and northern continental shelf of Sardinia (scale 1:250,000) was produced. The base-map is formed by the shaded DTM of the seabed. Other mapped features include the edge of the continental shelf and the distribution of rocky seabed.

Abstract Comparing populations across temperature gradients can inform how global warming will impact the structure and function of ecosystems. Shoot density, morphometry and productivity of the seagrass Posidonia oceanica to temperature variation was quantified at eight locations in Sardinia (western Mediterranean Sea) along a natural sea surface temperature (SST) gradient. The locations are spanned for a narrow range of latitude (1.5°), allowing the minimization of the effect of eventual photoperiod variability. Mean SST predicted P. oceanica meadow structure, with increased temperature correlated with higher shoot density, but lower leaf and rhizome width, and rhizome biomass. Chlorophyll a (Chl-a) strongly impacted seagrass traits independent of SST. Disentangling the effects of SST and Chl-a on seagrass meadow shoot density revealed that they work independently, but in the same direction with potential synergism. Space-for-time substitution predicts that global warming will trigger denser seagrass meadows with slender shoots, fewer leaves, and strongly impact seagrass ecosystem. Future investigations should evaluate if global warming will erode the ecosystem services provided by seagrass meadows.

Global warming and Marine Heat Waves (MHWs) are having large-scale impacts on the seagrasses and their effects on the Mediterranean endemic Posidonia oceanica need to be properly defined. This research aimed to sharpen the knowledge on P. oceanica response to the warming by correlating the shoot morphology and the productivity of the seagrass to temperature conditions and MHW occurrence. Two correlative studies, along a Mediterranean temperature range (sites at the same latitude), were conducted: one explored the associations of summer 2021 Sea Surface Temperature (SST) and MHWs on P. oceanica morphology and the other used a reconstruction technique (lepidochronological analysis) to relate past temperature conditions and MHW occurrence to proxies of seagrass productivity for the corresponding periods. The results showed that the mean summer temperature affected the P. oceanica leaf necrosis and that MHWs occurrence changed the morphology of the plant by lowering the leaf area and increasing leaf necrosis. Interesting results were also found relating the past thermal environment, since rhizome biomass has been negatively affected by the number of MHWs and rhizome length by the temperature range. This research provides fundamental insights about P. oceanica changes linked to warming and MHWs, identifying the potential plant indicators assisting future experimental and modelling studies. To the best of our knowledge, this is the first time a reconstruction technique is used to evaluate the effects of past MHWs on the morphology and productivity of a plant species.

Understanding how coralline algae may acclimatize to ocean warming is important to understand their survival over the coming century. Taking advantage of natural differences in temperature conditions between coastal areas in Sardinia (Italy) and between depths, the responses in terms of biological traits to warming of the crustose coralline alga Lithophyllum stictiforme, a key bioconstructor of coralligenous reefs in the Mediterranean, were evaluated in the field by two innovative transplant experiments where translocated specimens were used as controls. Results of the first experiment (algae cross transplanted between a cold and a warm site at two depths, 23 and 34 m) showed that the marginal growth of the alga and production of conceptacles were higher in the cold site, regardless of the treatment (transplant and translocation) and depth. However, growth in thickness in algae transferred from the cold to the warm site was higher at 34 m of depth, where they had a better performance than the local (translocated) algae. Results of the second experiment (algae transplanted from 34 m to 15 m of depth under different light irradiance manipulations) evidenced that the increase in temperature of +4°C was tolerated by thalli transplanted at 15 m, but that thallus growth and conceptacles production was negatively affected by the higher light irradiance. These results suggest an overall good adaptability of L. stictiforme under warmer conditions, even those due to thermocline deepening. Overall, these results encourage consideration of the use of transplants of this bioconstructor in future restoration actions of coralligenous habitats.
