Genetic features of alcohol dependence have been extensively investigated in recent years. A large body of studies has underlined the important role of genetic variants not only in metabolic pathways but also in the neurobiology of alcohol dependence, mediated by the neuronal circuits regulating reward and craving. Serotonin transporter (5-HTT), encoded by the SLC6A4 gene (Solute carrier family 6-neurotransmitter transporter-member 4), is targeted by antidepressant drugs such as selective serotonin reuptake inhibitors (SSRIs) and plays a pivotal role in serotoninergic transmission; it has been associated with psychiatric diseases and alcohol dependence. Transcriptional regulation and expression of 5-HTT depend not only on epigenetic modifications, among which DNA methylation (CpG and non-CpG) is primarily involved, but also on sequence variations occurring in intron/exon regions and in untranslated regions in 5′ and 3′, being the first sequences important for the splicing machinery and the last for the binding of transcription factors and micro RNAs. This work intends to shed light on the role of sequence variations known to affect the expression or function of 5-HTT in alcohol-dependent individuals. We found a statistically significant difference in the allelic (p = 0.0083) and genotypic (p = 0.0151) frequencies of the tri-allelic polymorphism, with higher function alleles and genotypes more represented in the control population. Furthermore, we identified three haplotypes more frequent in subjects with AUD (p < 0.0001) and one more frequent in the control population (p < 0.0001). The results obtained for the tri-allelic polymorphism in alcohol dependence confirm what is already present in part of the literature. The role of haplotypes requires further studies to be clarified. This work highlights recent studies in epigenetic mechanisms that play a role in alcoholism, which is a complex multifactorial disorder. There is a large body of evidence showing that alcohol can modify gene expression through epigenetic processes, namely DNA methylation and nucleosomal remodeling via histone modifications. In that regard, chronic exposure to ethanol modifies DNA and histone methylation, histone acetylation, and microRNA expression. The alcohol-mediated chromatin remodeling in the brain promotes the transition from use to abuse and addiction. Unravelling the multiplex pattern of molecular modifications induced by ethanol could support the development of new therapies for alcoholism and drug addiction targeting epigenetic processes. Ce travail met en lumiere les etudes recentes des mecanismes epigenetiques qui jouent un role dans l'alcoolisme, une maladie complexe multifactorielle. De nombreuses preuves ont montre que l'alcool peut modifier l'expression genique au moyen de processus epigenetiques, a savoir la methylation d'ADN et le remodelage nucleosomique par les modifications d'histones. Dans cette veine, l'exposition chronique a l'alcool modifie l'ADN et la methylation des histones, l'acetylation des histones et l'expression de microARN. Le remodelage de la chromatine medie par l'alcool dans le cerveau favorise la transition de l'usage vers l'abus et la dependance. La resolution du patron multiplexe des modifications moleculaires induites par l'alcool implique la capacite de developper de nouvelles therapies de l'alcoolisme et de la dependance aux drogues qui ciblent les processus epigenetiques. Klinefelter syndrome (KS) is a male genetic disease caused by the presence of an extra X chromosome, causing endocrine disorders mainly responsible for a high rate of infertility and metabolic disorders in adulthood. Scientific research is interested in identifying new biomarkers that can be predictive or prognostic of alterations strictly connected to KS. Lipocalin-2 (LCN-2, also known as NGAL) is a small protein initially identified within neutrophils as a protein related to innate immunity. Serum LCN-2 estimation seems to be a useful tool in predicting the metabolic complications caused by several pathological conditions. However, little is known about its potential role in infertility conditions. The present pilot study aims to investigate the presence of LCN-2 in the serum of a group of pre-pubertal and post-pubertal children affected by KS, compared to healthy controls. We demonstrated for the first time the presence of elevated levels of LCN-2 in the serum of KS patients, compared to controls. This increase was accompanied, in pre-pubertal KS patients, by the loss of correlation with LH and HDL, which instead was present in the healthy individuals. Moreover, in all KS individuals, a positive correlation between LCN-2 and inhibin B serum concentration was found. Despite the limited size of the sample analyzed, our preliminary data encourage further studies to confirm the findings and to extend the study to KS adult patients, to verify the predictive/prognostic value of LCN-2 as new biomarker for metabolic diseases and infertility associated with the pathology. The present study was aimed at examining spatial learning and memory, in 33 men and 12 women with alcohol use disorder (AUD) undergoing ethanol detoxification, by using a virtual Morris task. As controls, we recruited 29 men and 10 women among episodic drinkers without a history of alcohol addiction or alcohol-related diseases. Elevated latency to the first movement in all trials was observed only in AUD persons; furthermore, control women had longer latencies compared with control men. Increased time spent to reach the hidden platform in the learning phase was found for women of both groups compared with men, in particular during trial 3. As predicted, AUD persons (more evident in men) spent less time in the target quadrant during the probe trial; however, AUD women had longer latencies to reach the platform in the visible condition during trials 6 and 7 that resulted in a greater distance moved. As for the probe trial, men of both groups showed increased virtual locomotion compared with the women of both groups. The present investigation confirms and extends previous studies showing (i) different gender responses in spatial learning tasks, (ii) some alterations due to alcohol addiction in virtual spatial learning, and (iii) differences between AUD men and AUD women in spatial-behaviour-related paradigms. Les presents travaux visaient a etudier a l'aide d'une tache de Morris virtuelle l'apprentissage et la memoire spatiales chez 33 hommes et 12 femmes presentant des troubles lies a la consommation d'alcool (TCA) suivant une cure de desintoxication de l'ethanol. Comme temoins, nous avons inscrit 29 hommes et 10 femmes parmi des buveurs episodiques sans antecedents d'alcoolisme ni de maladies liees a l'alcool. Nous avons observe une augmentation de la latence avant le premier mouvement uniquement dans le groupe TCA. En outre, les temoins feminins presentaient des latences plus longues que les temoins masculins. Nous avons aussi observe au cours de la phase d'apprentissage que le temps ecoule avant de rejoindre la plateforme cachee etait plus eleve chez les femmes des deux groupes que chez les hommes, en particulier au cours de l'essai 3. Comme prevu les personnes atteintes de TCA (plus evident chez les hommes) ont passe moins de temps dans le quadrant cible pendant l'essai de sonde, mais les femmes atteintes de TCA presentaient de plus longues latences avant d'atteindre la plateforme en conditions de visibilite au cours des essais 6 et 7, ce qui s'est traduit par une plus grande distance de deplacement. Quant a l'essai de sonde, les hommes des deux groupes ont presente une augmentation de la locomotion virtuelle par rapport aux femmes des deux groupes. La presente etude confirme et elargit les observations d'etudes precedentes montrant (i) des reactions differentes selon le sexe quant aux taches d'apprentissage spatial, (ii) certaines modifications causees par l'alcoolisme quant a l'apprentissage spatial virtuel, et (iii) des differences entre les hommes et les femmes atteints de TCA quant aux paradigmes lies aux comportements spatiaux. The hypothalamic pituitary adrenal axis and dopamine have a key role in transition from alcohol social use to addiction. The medial prefrontal cortex was shown to modulate dopaminergic activity and cortisol releasing factor (CRF) release in hypothalamic and extra-hypothalamic systems. The recent advancements in non-invasive neurostimulation technologies has enabled stimulation of deeper brain regions using H-coil transcranial magnetic stimulation (TMS) in humans. This randomized double-blind placebo-controlled pilot study aims to evaluate H-coil efficacy in stimulating the medial prefrontal cortex. Cortisolemia and prolactinemia were evaluated as effectiveness markers. Alcohol intake and craving were considered as secondary outcomes. Eighteen alcoholics were recruited and randomized into 2 homogeneous groups: 9 in the real stimulation group and 9 in the sham stimulation group. Repetitive TMS (rTMS) was administered through a magnetic stimulator over 10 sessions at 20 Hz, directed to the medial prefrontal cortex. rTMS significantly reduced blood cortisol levels and decreased prolactinemia, thus suggesting dopamine increase. Craving visual analogic scale (VAS) in treated patients decreased, as well as mean number of alcoholic drinks/day and drinks on days of maximum alcohol intake (DMAI). In the sham group there was no significant effect observed on cortisolemia, prolactinemia, mean number of alcoholic drinks/day, or drinks/DMAI. Le cortex prefrontal interne module l'activite dopaminergique et la liberation de corticoliberine (CRF) dans les systemes hypothalamique et extra-hypothalamique. Un progres recent sur le plan des technologies de neurostimulation non-invasive permet de stimuler ces regions profondes du cerveau a l'aide de la stimulation magnetique trans-cranienne (SMT) chez l'humain. Cet essai pilote aleatoire a double aveugle comparatif avec placebo visait a evaluer l'efficacite de stimulation du cortex prefrontal interne de la bobine solenoide. La cortisolemie et la prolactinemie ont ete evaluees en tant que marqueurs d'efficacite. La consommation d'alcool et l'etat de besoin ont ete consideres comme resultats secondaires. Dix-huit personnes alcooliques ont ete recrutees et reparties de maniere aleatoire en deux groupes homogenes, 9 personnes avec stimulation reelle et 9 personnes avec stimulation simulee. La STM repetitive (STMr) a ete appliquee au moyen d'un stimulateur magnetique en 10 sessions de 20 Hz, et dirigee sur le cortex prefrontal interne. La STMr pouvait reduire significativement le niveau de cortisol sanguin et la prolactinemie, suggerant ainsi un accroissement de la dopamine. L'echelle visuelle de l'etat de besoin chez les patients traites diminuait, de meme que le nombre moyen de consommations par jour et le nombre de consommations lors des jours de consommation d'alcool maximale. Chez le groupe simule, aucun effet significatif n'etait observe sur la cortisolemie, la prolactinemie, le nombre moyen de consommations par jour et le nombre de consommations lors des jours de consommation maximale. Le STMr profonde pourrait etre consideree comme un outil supplementaire pour traiter l'alcoolisme.
