La mise en évidence du caractère actif de la phase de la résolution de l'inflammation a ouvert la porte à de nouveaux paradigmes thérapeutiques pour les maladies inflammatoires. Des paradigmes qui reposent sur l’utilisation de médiateurs actifs ayant le potentiel de promouvoir la résolution de l’inflammation. En d’autres termes, ces médiateurs régulent la réaction inflammatoire, initient la réparation tissulaire et conduisent au retour de l’homéostasie. Parmi ces médiateurs, la résolvine D1 (RvD1) présente des propriétés anti-inflammatoires et prorésolutives remarquables et ce, dans divers fonctions et tissus de l’organisme. Bien que les effets de la RvD1 aient été bien caractérisés par de nombreuses recherches, son rôle potentiel au niveau ostéoarticulaire demeure peu documenté. Dans cette étude, nous approfondissons les connaissances sur l’effet de la RvD1 dans la protection et la préservation de l’intégrité ostéoarticulaire, en étudiant sa capacité à réguler les principaux facteurs impliqués dans la physiopathologie articulaire. Dans un premier temps, nous avons démontré que la RvD1 est bien présente dans l’environnement articulaire et que ses niveaux sont plus élevés dans le liquide synovial des genoux arthrosiques par rapport aux genoux sains. Ces genoux sont obtenus d’un modèle d’arthrose chez le chien réalisé lors d’une étude antérieure. Par la suite, nous avons étudié ses effets sur les composantes cellulaires de l’articulation. Sur les chondrocytes arthrosiques humains, nous avons démontré que la RvD1 inhibe l’expression des médiateurs inflammatoires et cataboliques induits par l'IL-1β, à savoir la COX-2, la PGE2, l’iNOS, le NO et la MMP-13. L’étude des voies de signalisation a ensuite révélé que la RvD1 s’oppose à l'activation de NF-κB / p65, p38 / MAPK et JNK1 / 2, induite par l’IL-1β. En plus de ces remarquables effets, la RvD1 confère une protection contre l’apoptose cellulaire et le stress oxydatif induits par le HNE, tel que démontré par l'inactivation des caspases, l'inhibition de la libération de la LDH, l’augmentation des taux de la Bcl-2 et de l’AKT, ainsi que la stimulation du GSH. À côté des chondrocytes, la RvD1 a montré des effets puissants sur les ostéoclastes. En effet, elle inhibe la différenciation et l'activation des ostéoclastes tel que démontré par l’inhibition de l’expression de TRAP et de la cathepsine K. Elle réduit l’expression de TNF-α, de l’IL-1β, de l’IFN-γ, de la PGE2 et de RANKL, induite par le LPS et augmente celle de l’IL- 10. De plus, elle protège contre la résorption de la matrice d’hydroxyapatite ainsi que l’érosion ii de la matrice osseuse ex vivo, induites par le RANKL et le M-CSF. Enfin, l’étude des propriétés de la RvD1 dans un modèle d’arthrite chez la souris a révélé qu’elle réduit le score clinique, l'inflammation de la patte et la destruction des os et des articulations. De surcroit, elle inhibe les médiateurs inflammatoires et diminue significativement les marqueurs sériques du remodelage osseux et cartilagineux. Nos résultats sont très prometteurs et confirment le potentiel de la RvD1 dans la résolution de l’inflammation et le maintien de l’intégrité articulaires. Ils mettent ainsi en évidence sa pertinence clinique en tant qu’agent actif dans la prise en charge thérapeutique des maladies ostéoarticulaires., The recognition of the proactive character of the resolution phase of inflammation has revealed alternative therapeutic paradigms for inflammatory conditions, based on the use of active mediators capable of promoting the resolution of inflammation. In other words, these mediators regulate the inflammatory response, initiate tissue repair and promote the return to homeostasis. Among them, resolvin D1 (RvD1) has remarkable anti-inflammatory and proresolutive properties. Although the effects of RvD1 have been well studied, its potential role in the osteoarticular tissues remains poorly documented. In this study, we expand our understanding of the potential of RvD1 in protecting and preserving joint integrity by studying its ability to regulate the major factors involved in the articular pathophysiology. First, we demonstrated that RvD1 is produced in the articular environment and that its levels are higher in the synovial fluid of osteoarthritic knees compared to healthy knees. The knees were obtained from an osteoarthritic dog model performed in a previous study. Therefore, we studied RvD1 actions on the cellular components of the joint. In human osteoarthritic chondrocytes, we demonstrated that RvD1 inhibits IL-1β-induced inflammatory and catabolic mediators, namely COX-2, PGE2, iNOS, NO, and MMP-13. This led to the study of signaling pathways, which revealed that RvD1 counter-regulates IL-1β-induced activation of NF-κB / p65, p38 / MAPK and JNK1 / 2. In addition, RvD1 confers a protection against cellular apoptosis and oxidative stress induced by HNE, as revealed by caspases inactivation, LDH inhibition, as well as increased levels of Bcl-2, AKT, and GSH. In addition to chondrocytes, RvD1 showed remarkable effects on osteoclasts. Indeed, it inhibits osteoclast differentiation and activation, as demonstrated by the inhibition of TRAP and cathepsin K expression. Moreover, it reduces LPSinduced TNF-α, IL-1β, IFN-γ, PGE2 as well as RANKL and concurrently increases IL-10 expression. Furthermore, it inhibits RANKL and M-CSF-induced hydroxyapatite matrix degradation and bone matrix erosion, ex vivo. Finally, the study of the properties of RvD1 in a mouse model of arthritis, revealed that it alleviates the clinical score, paw inflammation, as well as bone and joint destruction. Furthermore, it reduces the inflammatory mediators and significantly decreases serum markers of bone and cartilage remodeling. Our results are very promising and confirm the high potential of RvD1 in resolving inflammation and preserving joint integrity. They highlight its clinical relevance as a therapeutic agent for the management of osteoarticular diseases.

For defending against herbivores or necrotrophic microorganisms, higher plants rely on jasmonates (JAs), particularly on the hormone jasmonoyl-isoleucine (JA-Ile), to signal adaptive changes. JAs also modulate tolerance to abiotic stresses. My thesis work demonstrated the importance of basal JAs signalling, and its repression by three jasmonic acid oxidases called JAO, whose action defines a new metabolic diversion mechanism with regulatory properties. Using omics approaches, we defined the spectrum of responses activated in jao2 mutants. Downstream of JA-Ile action, two distinct catabolic pathways of JA-Ile are activated to attenuate induced signalling. Owing to the generation of new higher order mutant plant lines impaired in one, the other, or both pathways, my research demonstrated their respective roles in JA-Ile elimination in response to herbivory and infection. It particularly revealed that restrained JA-Ile catabolism results in increased defense only when negative regulators are not overstimulated. Our results provide new knowledge of JAs catabolic mechanisms regulating basal and induced signalling., Pour se défendre contre les herbivores ou les microorganismes nécrotrophes les plantes supérieures utilisent les jasmonates (JAs), en particulier l’hormone jasmonoyl-isoleucine (JA-Ile), pour activer la signalisation défensive. Les JAs régulent aussi des réponses contre des stress abiotiques. Mes travaux de thèse ont prouvé l’importance d’une signalisation JAs basale, et sa répression par trois oxydases d’acide jasmonique nommées JAO, dont l’action définit un mécanisme de détournement métabolique inédit. En aval de l'action du JA-Ile, deux voies cataboliques distinctes sont activées pour atténuer la signalisation induite. A l’aide de nouvelles lignées de plantes bloquées, pour l’une, l’autre ou les deux voies, mes recherches ont démontré leurs rôles respectifs dans l’élimination du JA-Ile en réponse à l’herbivorie ou à l’infection. Elles ont révélé qu’un catabolisme ralenti du JA-Ile limite sélectivement le gain en défense lorsque des régulateurs négatifs de la signalisation sont surstimulés. Nos résultats apportent de nouvelles connaissances sur les mécanismes cataboliques des JAs régulant les signalisations basales et induites.

Balancing rations for essential amino acids (AA) to improve efficiency of utilisation (Eff) of N requires a better knowledge of AA use by different tissues under various nutritional conditions. Effects of infusion of AA (AAinf) and glucose (Glcinf), in a 2×2 factorial arrangement, were tested using 5 cows in a 4×4 Latin Square plus one cow. At the end of each period, splanchnic, mammary and other peripheral tissue metabolisms of AA were measured. There was no AAinf×Glcinf interaction on most of the measured parameters. Milk protein yield (MPY) increased with AAinf and Glcinf. With AAinf, catabolism by portal-drained viscera (PDV) and mammary gland of group 2 AA (Ile, Leu, Lys, and Val) increased leading to a decreased Eff. In contrast, with Glcinf, liver removal of His + Met + Phe (HMP) decreased, improving their Eff.

International audience; Effects of AA and glucose infusions on efficiency of use of essential AA (EAA) were studied according to a 2 × 2 factorial using 5 multicatheterized cows in a 4 × 4 Latin square plus one cow, with 2-wk periods. The diet provided 87% of energy and 70% of metabolizable protein requirements, and the 4 treatments were abomasal infusions of (1) water, (2) an AA mixture with a casein profile (695 g/d), (3) glucose (1,454 g/d), or (4) a combination of AA and glucose infusions. Milk samples were collected on the last 6 milkings. On d 14, 6 blood samples were collected from arterial, and portal, hepatic, and mammary venous vessels. Splanchnic plasma flow was calculated by dilution of p-aminohippurate and mammary flow by the Fick principle using Phe + Tyr. The net flux of AA across tissues [splanchnic, i.e., portal-drained viscera (PDV) + liver, and mammary gland] was calculated as the efflux minus the influx across that tissue. The efficiency of EAA was calculated as the sum of exported true proteins [milk protein yield (MPY), scurf, and metabolic fecal protein] multiplied by their respective AA profile and divided by the predicted AA supply minus AA endogenous urinary loss. In addition, catabolism was estimated for each tissue: AA supply − (portal net flux + metabolic fecal protein) for the PDV; −hepatic net flux for the liver; splanchnic net flux − (−mammary net flux + scurf) for the other peripheral tissues; and −mammary net flux − milk for the mammary gland. The MIXED procedure (SAS Institute Inc., Cary, NC) was used with cow as a random effect. No AA × glucose interaction existed for most of the measured parameters. With infusions of AA and glucose, MPY increased by 17 and 14%, respectively. The decreased efficiency of EAA-N with AA infusion resulted from increased EAA-N in MPY smaller than the increased EAA-N supply and was accompanied by increased liver catabolism of His + Met + Phe (representing group 1 AA) and increased mammary and PDV catabolisms of group 2 AA-N (Ile, Leu, Lys, and Val). In contrast, the increased efficiency of EAA-N with glucose infusion, resulting from increased EAA-N in MPY with no change in EAA-N supply, was accompanied by decreased mammary catabolism of group 2 AA-N and hepatic catabolism of His + Met + Phe. No mammary catabolism of His, Met, and Phe existed in all treatments, as indicated by the mammary uptake to milk output ratio close to one for these EAA. Therefore, the mammary gland contributes significantly to variations of efficiency of group 2 AA-N through variations of AA catabolism, in response to both AA and glucose supplies, whereas additional PDV catabolism was observed with increased AA supply. Partition of AA use between tissues allows to delineate their anabolic or catabolic fate across tissues and better understand changes of efficiency of EAA in response to protein and energy supplies.

Efficient afforestation programs are crucial to limit soil degradation in various arid and semi-arid ecosystems. However, the success of these programs is dependant to the plant type selected for revegetation and the methods used for seedling production. Exotic fast-growing trees have been largely planted but their use is currently controversial because of their potential negative ecological impacts. Whereas the positive impact of arbuscular mycorrhizal (AM) fungal inoculation in nursery was demonstrated, few studies focused on the monitoring of mycorrhizal inoculation in arid and semi-arid ecosystems. In addition, the majority of studies are based on single-species inocula with non native AM fungal strains. The current study aims at evaluating the efficiency of mycorrhizal inoculation of the emblematic Mediterranean carob tree (Ceratonia siliqua) in a Moroccan degraded site, through an ecological engineering strategy based on the use of a complex native AM community (naturally associated to carob trees). Results demonstrate the high potential of this approach by improving sustainably the growth and nutrient status of carob trees in a 3-year-old plantation and also by inducing a positive soil microbial environment for nutrient cycling and environmental stress resistance. Based on the concept of nutritional programming in mammals, we tested whether a short term hyper or hypo vitamin stimulus during first-feeding could induce long-lasting changes in nutrient metabolism in rainbow trout. Trout alevins received during the 4 first weeks of exogenous feeding a diet either without supplemental vitamins (NOSUP), a diet supplemented with a vitamin premix to satisfy the minimal requirement in all the vitamins (NRC) or a diet with a vitamin premix corresponding to an optimal vitamin nutrition (OVN). Following a common rearing period on the control diet, all three groups were then evaluated in terms of metabolic marker gene expressions at the end of the feeding period (day 119). Whereas no gene modifications for proteins involved in energy and lipid metabolism were observed in whole alevins (short-term effect), some of these genes showed a long-term molecular adaptation in the muscle of juveniles (long-term effect). Indeed, muscle of juveniles subjected at an early feeding of the OVN diet displayed up-regulated expression of markers of lipid catabolism (3-hydroxyacyl-CoA dehydrogenase – HOAD - enzyme) and mitochondrial energy metabolism (Citrate synthase - cs, Ubiquitinol cytochrome c reductase core protein 2 - QCR2, cytochrome oxidase 4 - COX4, ATP synthase form 5 - ATP5A) compared to fish fed the NOSUP diet. Moreover, some key enzymes involved in glucose catabolism (Muscle Pyruvate kinase - PKM) and amino acid catabolism (Glutamate dehydrogenase - GDH3) were also up regulated in muscle of juvenile fish fed with the OVN diet at first-feeding compared to fish fed the NOSUP diet. We researched if these permanently modified gene expressions could be related to global modifications of epigenetic marks (global DNA methylation and global histone acetylation and methylation). There was no variation of the epigenetic marks in muscle. However, we found changes in hepatic DNA methylation, global H3 acetylation and H3K4 methylation, dependent on the vitamin intake at early life. In summary, our data show, for the first time in fish, that a short-term vitamin-stimulus during early life may durably influence muscle energy and lipid metabolism as well as some hepatic epigenetic marks in rainbow trout. Statement of relevance Programming at first feeding with different levels of vitamins was successfully tested in fish for the first time. These data proove one more time the importance of the early life on the fish metabolism in adults.

The widely used plasmid-free Lactococcus lactis strain MG1363 was derived from the industrial dairy starter strain NCDO712. This strain carries a 55.39 kb plasmid encoding genes for lactose catabolism and a serine proteinase involved in casein degradation. We report the DNA sequencing and annotation of pLP712, which revealed additional metabolic genes, including peptidase F, D-lactate dehydrogenase and a-keto acid dehydrogenase (E3 complex). Comparison of pLP712 with other large lactococcal lactose and/or proteinase plasmids from L. lactis subsp. cremoris SK11 (pSK11L, pSK11P) and the plant strain L. lactis NCDO1867 (pGdh442) revealed their close relationship. The plasmid appears to have evolved through a series of genetic events as a composite of pGdh442, pSK11L and pSK11P. We describe in detail a scenario by which the metabolic genes relevant to the growth of its host in a milk environment have been unified on one replicon, reflecting the evolution of L. lactis as it changed its biological niche from plants to dairy environments. The extensive structural instability of pLP712 allows easy isolation of derivative plasmids lacking genes for casein degradation and/or lactose catabolism. Plasmid pLP712 is transferable by transduction and conjugation, and both of these processes result in significant molecular rearrangements. We report the detailed molecular analysis of insertion sequence element-mediated genetic rearrangements within pLP712 and several different mechanisms, including homologous recombination and adjacent deletion. Analysis of the integration of the lactose operon into the chromosome highlights the fluidity of the MG1363 integration hotspot and the potential for frequent movement of genes between plasmids and chromosomes in Lactococcus.

International audience; Although it is usually admitted that arbuscular mycorrhizal (AM) fungi are key components in soil bio-functioning, little is known on the response of microbial functional diversity to AM inoculation. The aims of the present study were to determine the influence of Glomus intraradices inoculum densities on plant growth and soil microflora functional diversity in autoclaved soil or non-disinfected soil. Microbial diversity of soil treatments was assessed by measuring the patterns of in situ catabolic potential of microbial communities. The soil disinfection increased sorghum growth, but lowered catabolic evenness (4.8) compared to that recorded in the non-disinfected soil (6.5). G. intraradices inoculation induced a higher plant growth in the autoclaved soil than in the non-disinfected soil. This AM effect was positively related to inoculum density. Catabolic evenness and richness were positively correlated with the number of inoculated AM propagules in the autoclaved soil, but negatively correlated in the non-disinfected soil. In addition, after soil disinfection and AM inoculation, these microbial functionality indicators had higher values than in the autoclaved or in the non-disinfected soil without AM inoculation. These results are discussed in relation to the ecological influence of AM inoculation, with selected fungal strains and their associated microflora on native soil microbial activity

L’atrophie musculaire est la conséquence d’un déséquilibre du métabolisme des protéines musculaires. La plupart des états cataboliques sont caractérisés par une résistance du métabolisme protéique à l'insuline et aux acides aminés. Il en résulte une inefficacité de la prise alimentaire pour générer une balance azotée positive pendant la période post-prandiale. Dans de telles situations, les protéines à digestion rapide et riches en leucine (i.e Lactosérum (LAC)) ont été montrées pour être plus efficaces que la caséine pour optimiser la stimulation de la protéosynthèse musculaire (PS). Cependant et paradoxalement, la masse musculaire est rarement préservée par des supplémentations en LAC. Notre hypothèse est que cette stimulation ne se produit que pendant une courte période de temps à l'état nourri mais qui reste insuffisante pour se traduire par une augmentation significative de la masse musculaire au long terme. Pour valider et vérifier cette hypothèse, une mesure cinétique de la PS et de la protéolyse (PRO) au niveau musculaire est nécessaire pendant toute la période post prandiale. Matériels et Méthodes. Miniporcs ont été cathétérisés à la fois dans l'artère et la veine fémorales et infusés avec de la 13C Phe afin d'évaluer, toutes les 30 min, la PS et PRO musculaires dans la patte arrière par la méthode de différence artério-veineuse en acides aminés et en traceur. Les animaux sont étudiés avant et après la prise du repas. L'état catabolique a été induit par un traitement aux glucocorticoïdes (déxamethasone (DEX) pendant 8j) et l’effet de 2 protéines, la caséine (CAS) et le LAC a été testé sur la PS et la PRO pendant 6,5h. Les différences entre groupes ont été évaluées par ANOVA en mesures répétées à 3 facteurs (type de protéines, traitement DEX , temps post prandial) et le seuil de signification à P

Ubiquitin-dependent proteolysis is activated in skeletal muscle atrophying in response to various catabolic stimuli. Previous studies have demonstrated activation of ubiquitin conjugation. Because ubiquitination can also be regulated by deubiquitinating enzymes, we used degenerate oligonucleotides derived from conserved sequences in the ubiquitin-specific protease (UBP) family of deubiquitinating enzymes in RT-PCR with skeletal muscle RNA to amplify putative deubiquitinating enzymes. We identified USP19, a 150-kDa deubiquitinating enzyme that is widely expressed in various tissues including skeletal muscle. Expression of USP19 mRNA increased by ∼30–200% in rat skeletal muscle atrophying in response to fasting, streptozotocin-induced diabetes, dexamethasone treatment, and cancer. Increased mRNA levels during fasting returned to normal with refeeding, but 1 day later than the normalization of rates of proteolysis and coincided instead with recovery of muscle mass. Indeed, in all catabolic treatments, USP19 mRNA was inversely correlated with muscle mass and provided an index of muscle mass that may be useful in many pathological conditions, using small human muscle biopsies. The increased expression of this deubiquitinating enzyme under conditions of increased proteolysis suggests that it may play a role in regeneration of free ubiquitin either coincident with or after proteasome-mediated degradation of substrates. USP19 may also be involved in posttranslational processing of polyubiquitin produced de novo in response to induction of the polyubiquitin genes seen under these conditions. Deubiquitinating enzymes thus appear involved in muscle wasting and implicate a widening web of regulation of genes in the ubiquitin system in this process.

En situation de catabolisme musculaire, les protéines de lactosérum permettent bien de restaurer un anabolisme musculaire post prandial mais d’une durée très transitoire probablement insuffisante pour limiter la fonte musculaire

L'arthrose est une maladie articulaire dégénérative, avec une pathogenèse inconnue. Des études récentes suggèrent que l'activation du facteur de transcription du récepteur activateur de la prolifération des peroxysomes (PPAR) gamma est une cible thérapeutique pour ce maladie. Les agonistes du PPARγ inhibent l'inflammation et réduisent la synthèse des produits de dégradation du cartilage in vitro et in vivo. Cependant, des études utilisant des agonistes du PPARγ n’élucident pas les effets exacts médiés par ce gène complexe. En effet, certains de ces agonistes ont la capacité de régulariser d'autres voies de signalisation indépendantes de PPARγ, ainsi entraînant des effets secondaires graves. Afin d'obtenir une efficacité thérapeutique avec potentiellement moins de problèmes de sécurité, il est donc essentiel d'élucider, in vivo, le rôle exact de PPARγ dans la physiopathologie OA. Mon projet de thèse permettra de déterminer, pour la première fois, le rôle spécifique de PPARγ in vivo dans la physiopathologie OA. Les souris utilisées pour l’étude avaient une délétion conditionnelle du gène PPARγ dans le cartilage. Ces dernières ont été générées en employant le système LoxP/Cre. Pour tester cette hypothèse, j'ai généré deux types de souris avec une délétion au PPARγ, (a) une suppression du gène PPARγ spécifiquement dans le cartilage germinale pour l'étude de l'arthrose liée au développement et à l'âge et (b) la suppression inductible du gène PPARγ spécifiquement dans le cartilage chez la souris adulte pour les études OA. L’étude précédente dans notre laboratoire, utilisant ces souris ayant une délétion au gène PPARγ germinales, montre que ces souris présentent des anomalies du développement du cartilage. J'ai également exploré si ces souris qui présentent des défauts précoces du développement ont toutes les modifications phénotypiques dans le cartilage au cours du vieillissement. Mes résultats ont montré que les souris adultes, ayant une délétion au gène PPARγ, ont présenter un phénotype de l'arthrose spontanée associée à une dégradation du cartilage, l’hypocellularité, la fibrose synoviale. Cette étude a montré que PPARγ est un régulateur essentiel pour le cartilage, et c’est le manque (l’absence) de ce dernier qui conduit à un phénotype de l'arthrose spontanée accélérée (American Journal of Pathologie). A partir de ce but de l'étude, on n’a pas pu vérifier si ces souris présentaient l’OA spontanée en raison des défauts de développement ou à la suite de la délétion du gène PPARγ. Pour contourner les défauts de développement, j'ai généré des souris ayant une délétion du gène PPARγ spécifiquement dans le cartilage inductible avec le système Col2rTACre. Ces souris ont été soumises à modèle de la chirurgie OA (DMM: déstabilisation du ménisque médial) et les résultats révèlent que les souris PPARγ KO ont une dégradation accélérée du cartilage, une hypocellularité, une fibrose synoviale et une augmentation de l'expression des marqueurs cataboliques et des marqueurs inflammatoire. La perte de PPAR dans le cartilage articulaire est un évènement critique qui initie la dégradation de cartilage dans OA. Les études récentes suggèrent que le procès d’autophagie, une forme de survie cellulaire programmée, est altéré pendant l’OA et peut contribuer vers une protection diminuée des cellules, résultant la dégradation du cartilage. J’ai donc exploré le rôle de PPARγ dans la protection des cellules en déterminant l’effet de manque de PPARγ dans le cartilage par l’expression de mTOR (régulateur négatif principal d’autophagie) et les gènes d’autophagie durant OA. Mes résultats ont montré que les souris KO PPARγ présentent également une augmentation sur l'expression de mTOR et une diminution sur l’expression des marqueurs autophagiques en comparaison avec les chondrocytes articulaires isolés des souris contrôles OA. J'ai suggéré l'hypothèse que PPARγ contrôle la régulation de la signalisation de mTOR/autophagie, et finalement la mort des chondrocytes et l’expression des facteurs cataboliques et les facteurs inflammatoire. Pour tester cette hypothèse, j’ai fait la transfection des chondrocytes arthrosiques PPARγ-KO avec le vecteur d’expression de PPARγ pour déterminer si la restauration de l'expression de PPARγ peut sauver le phénotype des cellules PPARγ-KO OA. J'ai observé que la restauration de l'expression de PPARγ dans les cellules PPARγ-KO en présence du vecteur d'expression PPARγ, a pu considérablement régulariser négativement l'expression de mTOR et mettre en règle positivement l'expression des gènes autophagiques ainsi que le sauvetage significative de l'expression du collagène de type II et l’aggrecan et de baisser de manière significative l'expression de marqueurs cataboliques critiques et des marqueurs inflammatoires. Pour prouver que l’augmentation de la signalisation de mTOR et la diminution de l'autophagie est responsable du phénotype OA accélérée observée dans les souris PPARγ KO in vivo, j'ai généré les souris doubles KO PPARγ- mTOR inductible spécifique du cartilage en utilisant le système Col2 - rtTA -Cre et soumis ces souris à DMM modèle de l'arthrose. Mes résultants démontrent que les souris avec PPARγ- mTOR doubles KO ont été significativement protégés contre les OA DMM induites associées à une protection significative contre la destruction du cartilage, la perte de protéoglycanes et la perte de chondro-cellularité par rapport aux souris témoins. Considérant que mTOR est un répresseur majeur de l'autophagie, j'ai trouvé que l'expression de deux marqueurs de l'autophagie critiques (ULK1 et LC3B) a été significativement plus élevée dans les chondrocytes extraits les souris doubles KO PPARγ-mTOR par rapport aux souris témoins. En plus, les études de sauvetage in vitro en utilisant le vecteur d'expression PPAR et les études in vivo utilisant les souris doubles KO PPARγ- mTOR montrent que PPARγ est impliqué dans la régulation de la protéine signalant de mTOR/autophagie dans le cartilage articulaire. Ces résultats contournent PPARγ et sa signalisation en aval de mTOR/autophagie en tant que cibles thérapeutiques potentielles pour le traitement de l'arthrose., Osteoarthritis (OA) is an age related degenerative joint disease with unknown pathogenesis. Recent studies suggest that the activation of the transcription factor Peroxisome Proliferator-Activated Receptor gamma (PPARγ) is a therapeutic target for OA. Agonists of PPARγ inhibit inflammation and reduce the synthesis of cartilage degradation products both in vitro and in vivo. However, studies using agonists of PPARγ do not elucidate the exact effects mediated by this complex gene. Indeed, some of these agonists have the ability to regulate, in vivo, various other signaling pathways independent of PPARγ, resulting in serious side effects. It is therefore vital, in order to achieve therapeutic efficacy with potentially less safety concerns, to elucidate the exact in vivo role of PPARγ in OA pathophysiology. Thus, the aim of my PhD project was to determine the specific in vivo role of PPARγ in OA pathophysiology using cartilage-specific PPARγ knockout (KO) mice and subjecting these mice to surgical model of OA. I generated two separate PPARγ KO mice harboring a (a) constitutive cartilage-specific germ-line deletion of PPARγ gene for developmental and age-related OA study and (b) inducible cartilage-specific deletion of PPARγ in adult mouse specifically for OA studies using LoxP Cre system. Previous study in my laboratory using germ-line PPARγ KO mice shows that these mice exhibit cartilage developmental defects. I further explored if these mice which exhibit early developmental defects have any phenotypic changes in the articular cartilage during ageing. My results showed that adult PPARγ KO mice exhibited a spontaneous OA phenotype associated with enhanced cartilage degradation, hypocellularity, synovial fibrosis, and increased expression of catabolic and inflammatory factors. This study showed that PPARγ is a critical regulator of cartilage health, the lack of which leads to an accelerated spontaneous OA phenotype (Vasheghani et al, 2013; American Journal of Pathology). From this aim of the study, I could not ascertain if cartilage-specific germline PPARγ KO mice exhibited spontaneous OA because of developmental defects or as a result of PPARγ deficiency. To bypass the developmental defects, I then generated inducible cartilage-specific PPARγ KO mice using Col2rTACre system and subjected these mice to destabilization of medial meniscus (DMM) model of OA surgery. My results revealed that PPARγ KO mice showed accelerated cartilage degradation, hypo-cellularity, synovial fibrosis and increased expression of catabolic and inflammatory factors during OA. Loss of chondrocyte cellularity within the articular cartilage is one of the critical events that initiate the degradation of the cartilage during OA. Recent studies suggest that the process of autophagy, a form of programmed cell survival, is impaired during OA and may contribute towards decreased chondro-protection resulting in cartilage degradation. Thus, I further explored the role of PPARγ in chondro-protection by determining the effect of PPARγ deficiency in the cartilage on the expression of mTOR (master negative regulator of autophagy) and autophagy genes during OA. My results revealed that PPARγ-deficient chondrocytes exhibit significantly enhanced expression of mTOR and decreased expression of genes that initiate autophagy process compared to chondrocytes extracted from control OA mice. I then hypothesized that PPARγ controls mTOR/autophagy signaling and ultimately the fate of chondrocytes and the expression of catabolic and inflammatory factors in the articular cartilage. To test this, I transfected PPARγ KO OA chondrocytes with PPARγ expression vector to determine if restoration of PPARγ expression can rescue the phenotype of PPARγ KO OA cells. I observed that restoration of PPARγ expression in PPARγ KO cells significantly down-regulated the expression of mTOR and up-regulate the expression of autophagy genes along with significant rescue in the expression of collagen type II and aggrecan and significant down-regulation in the expression of critical catabolic and inflammatory markers. To validate our in vitro finding that enhanced mTOR signalling and resultant decrease in autophagy is responsible for accelerated OA phenotype observed in PPARγ KO mice, I generated inducible cartilage-specific PPARγ-mTOR double KO mice and subjected these mice to DMM model of OA. My results clearly demonstrate that PPARγ-mTOR double KO mice exhibit significant protection against DMM-induced OA associated with significant protection from cartilage destruction, proteoglycan loss and loss of chondro-cellularity compared with control mice. Since mTOR is a major repressor of autophagy, I found that the expression of two critical autophagy markers (ULK1 and LC3B) was significantly elevated in PPARγ-mTOR double KO mice compared to control mice. My in vitro rescue studies using PPARγ expression vector and in vivo studies using PPARγ- mTOR double KO mice clearly show that PPARγ is involved in the regulation of mTOR/autophagy signalling in the articular cartilage. Therefore, deficiency of PPARγ upregulates mTOR signalling resulting in the suppression of autophagy and decreased chondroprotection and increased catabolic activity leading to accelerated severe OA. This study for the first time provides direct evidence on the role of PPARγ in chondroprotection by modulation of mTOR/autophagy signalling in the articular cartilage. These findings outline PPARγ and its downstream signalling by mTOR/autophagy as potential therapeutic targets for the treatment of OA.

Most animal models of sepsis induced high mortality or early recovery and do not mimic the long-lasting catabolic state observed in patients. The purpose of this study is to develop a model of sepsis which reproduces these disorders, especially the long-lasting muscle wasting. This report summarizes our observations in a series of seven experiments using this model with rats to study the route of live Escherichia coli administration, dose of bacteria, reproducibility of the model, bacterial count in tissues, comparison of injection of live or dead bacteria, metabolic perturbations linked to infection, and potential role of tumor necrosis factor alpha (TNF-α) in muscle wasting. After intravenous infection, animals were anorexic and the catabolic state was long-lasting: body weight loss for 2 to 3 days followed by a chronic wasting state for several days. Liver, spleen, lung protein content, and plasma concentration of α 2 -macroglobulin were increased 2 and 6 days after infection. At 6 days, muscle protein content was substantially (−40%) reduced. The plasma TNF-α level measured 1.5 h after infection correlated with body weight loss observed 9 days later. The inhibition of TNF-α secretion by administration of pentoxifylline 1 h before infection reduced muscle wasting and activation of proteolysis at day 2 and abolished them at day 6. This septic model mimics in rats the prolonged protein metabolism alterations and muscle atrophy characteristics of infected patients and thus is useful for studying the impact of nutritional support on outcome.

However, the absence of this receptor in myofibers decreases muscle hypertrophy induced by mechanical overload and limits glucocorticoids-induced muscle atrophy. Its ablation also increases autophagy, leading to sacromeres destructuration, resulting in decreased muscle strength. Moreover, its deletion reduced the rate of glucose absorption during a glucidic overload. International audience; Carbon storage is likely to enable adaptation of trypanosomes to nutritional challenges or bottlenecks during their stage development and migration in the tsetse. Lipid droplets are candidates for this function. This report shows that feeding of T. brucei with oleate results in a 4-5 fold increase in the number of lipid droplets, as quantified by confocal fluorescence microscopy and by flow cytometry of BODIPY 493/503-stained cells. The triacylglycerol (TAG) content also increased 4-5 fold, and labeled oleate is incorporated into TAG. Fatty acid carbon can thus be stored as TAG in lipid droplets under physiological growth conditions in procyclic T. brucei. beta-oxidation has been suggested as a possible catabolic pathway for lipids in T. brucei. A single candidate gene, TFE alpha 1 with coding capacity for a subunit of the trifunctional enzyme complex was identified. TFE alpha 1 is expressed in procyclic T. brucei and present in glycosomal proteomes, Unexpectedly, a TFE alpha 1 gene knock-out mutant still expressed wild-type levels of previously reported NADP-dependent 3-hydroxyacyl-CoA dehydrogenase activity, and therefore, another gene encodes this enzymatic activity. Homozygous Delta tfe alpha 1/Delta tfe alpha 1 null mutant cells show a normal growth rate and an unchanged glycosomal proteome in procyclic T. brucei. The decay kinetics of accumulated lipid droplets upon oleate withdrawal can be fully accounted for by the dilution effect of cell division in wild-type and Delta tfe alpha 1/Delta tfe alpha 1 cells. The absence of net catabolism of stored TAG in procyclic T. brucei, even under strictly glucose-free conditions, does not formally exclude a flux through TAG, in which biosynthesis equals catabolism. Also, the possibility remains that TAG catabolism is completely repressed by other carbon sources in culture media or developmentally activated in post-procyclic stages in the tsetse.

The rapid release of intracellular enzymes due to autolysis into the cheese matrix is thought to have a positive influence on flavour development. The objective of this study was to investigate the impact of autolysis on amino acid catabolism and flavour development by using two isogenic starter strains having the same enzymatic potentials and differing only in their autolytic abilities: the highly autolytic Lactococcus lactis AM2 and its prophage cured derivative L. lactis AM2-C.

De part son profil équilibré en acides aminés essentiels (AAE), la farine de poisson (FP) est la source protéique principale utilisée dans l’alimentation des crevettes d’élevage. Cependant, compte tenu des enjeux du développement durable de la production aquacole, son utilisation doit être réduite, et remplacée par d’autres sources protéiques comme les protéines végétales (PV), qui sont souvent carencées en lysine et méthionine mais riches en cystine. Les conséquences d’un tel changement alimentaire sont peu connues chez la crevette tigrée Penaeus monodon. Pour les évaluer, nous avons utilisé des aliments semi-purifiés reflétant les carences/excès en AA des PV pour estimer les besoins en protéine, lysine et méthionine pour l’entretien et la croissance. Tout en confirmant les données antérieures sur les besoins pour la croissance des stades postlarves, nous avons pu préciser la contribution de l’apport en ces deux acides aminés pour l’entretien. Au niveau métabolique, la variation de l’apport protéique (10, 30, 50% protéine brute) et la carence en méthionine (-30% par rapport au besoin) entraînent une modification de l’activité des enzymes du catabolisme des AA, mais pas celle des voies de reméthylation et transsulfuration. En revanche, et pour la première fois chez la crevette, nos résultats démontrent une épargne de la méthionine par la cystine (et la choline), soulignant l’importance de l’apport en AA soufrés totaux (methionine + cystine). Nos résultats illustrent aussi l’importance qu’il convient d’accorder à la disponibilité des AAE dans les études de remplacement de la FP par un mélange de PV pour améliorer l’utilisation azotée chez la crevette P. monodon

Le traitement au froid en post-récolte (TPR) des ananas (Ananas comosus. (L). M) destiné à ralentir la sénescence des fruits, induit " le brunissement interne de l'ananas " (BI). Afin d'étudier des gènes susceptibles de discriminer les variétés résistantes, une comparaison entre des fruits frais ou soumis au TPR a été réalisée pour 4 variétés d'ananas différant par leur résistance au BI. D'après les résultats, en réponse au TPR l'absence de symptômes de BI est associée à une " résistance membranaire " et à une faible activité de la polyphenol oxydase. L'étude de l'activité de la phénylalanine ammonia-lyase et de l'ascorbate peroxidase a révélé que le froid induit une stimulation de l'activité de ces enzymes chez une variété sensible au BI. L'étude du catabolisme protéique a montré que la résistance des fruits au BI était liée : à la sous-expression du gène d'une protéase à cystéine, et à une sur-expression de gènes codant une cystatine et une protéase à acide aspartique, dont l'ADNc a été caractérisé et cloné pour la première fois chez l'ananas. L'expression différentielle de ces gènes indique qu'ils pourraient être utilisés pour le criblage par PCR de variétés dans les programmes d'améliorations de l'ananas pour la résistance au BI.

L’arthrose (OA) est une maladie articulaire dégénérative à l’étiologie complexe et diverse. Les travaux de ces dernières années ont démontré que l’OA est une pathologie affectant tous les tissus de l’articulation incluant le cartilage, la membrane synoviale et l’os sous-chondral. L’OA se traduit par une déstructuration et une perte de fonctionnalité de l’articulation, et est principalement caractérisée par une perte de cartilage articulaire. L’inflammation de la membrane synoviale joue un rôle déterminant dans la progression de l’OA, toutefois elle serait secondaire à la dégradation du cartilage. De plus, l’os sous-chondral est également le siège de nombreuses transformations lors de l’OA. Il est fortement suggéré que ces changements ne correspondent pas seulement à une conséquence, mais pourraient être une cause du développement de l’OA impliquant une communication entre ce tissu et le cartilage. Il est maintenant bien établi que les voies inflammatoires et cataboliques jouent un rôle crucial dans l’OA. C’est pourquoi, nous avons étudié l’implication d’une nouvelle famille de récepteurs membranaires, les PARs, et plus particulièrement le PAR-2 dans les voies physiopathologiques de l’OA. Notre hypothèse est que l’activation de PAR-2 au cours de l’OA est un phénomène majeur du développement/progression de la maladie faisant du récepteur PAR-2 un candidat privilégié pour le développement de nouvelles approches thérapeutiques ciblant non seulement le cartilage mais aussi l’os sous-chondral. Pour cette étude, nous avons travaillé in vitro avec des chondrocytes (Cr) et des ostéoblastes (Ob) OA respectivement du cartilage et de l’os sous-chondral du condyle fémoral humain. Nos résultats ont démontré que PAR-2 était plus exprimé dans les Cr et les Ob OA que dans les cellules normales. Par ailleurs, PAR-2 est régulé positivement par certains facteurs retrouvés au cours de l’OA comme l’IL-1β, le TNF-α et le TGF-β dans les Cr OA, et par l’IL-1β, le TNF-α et la PGE2 dans les Ob OA. De plus, les principaux facteurs cataboliques et inflammatoires, soit la MMP-1, la MMP-13 et la COX-2 sont produits en quantité plus élevée suite à l’activation du récepteur dans le cartilage OA. De même, l’activation de PAR-2 dans les Ob OA conduit à une production accrue de facteurs pro-résorptifs tels que RANKL, l’IL-6, la MMP-1 et la MMP-9, et à l’augmentation de l’activité pro-résorptive de ces cellules. En outre, dans les deux types tissulaires étudiés, l’activation de PAR-2 augmente l’activité de certaines protéines de la famille des MAPKinases comme Erk1/2, p38 et JNK. Finalement, nous avons conclu notre étude en employant un modèle in vivo d’OA induite chez la souris sauvage et déficiente pour le gène PAR-2. Nos résultats ont démontré que l’absence d’expression et de production de PAR-2 influençait le processus inflammatoire et les changements structuraux affectant à la fois le cartilage et l’os sous-chondral, conduisant à un ralentissement du développement de l’OA. Nos travaux de recherche ont donc permis de montrer que le récepteur PAR-2 est un élément majeur du processus OA en agissant sur les voies cataboliques et inflammatoires du cartilage, et sur le remodelage tissulaire de l’os sous-chondral. Recent studies have shown that OA is a pathology affecting all the tissues of the joint including the cartilage, the synovial membrane and the subchondral bone. OA is regarded as destruction and a loss of functionality of joint, and is mainly characterized by a loss of articular cartilage. The synovial inflammation also plays a key role in the progression of OA; however, it is believed to be secondary to cartilage degradation. In addition, there are also in the subchondral bone numerous changes which occur during the course of the disease. It is strongly suggested that these changes in subchondral bone are not just a consequence but could be a cause of the development of OA, thus involving a cross-talk between this tissue and the cartilage. It is now well established that inflammatory and catabolic pathways play a crucial role during OA. This is why we have engaged the study of the involvement of a new family of membranous receptors, the PARs, and more particularly PAR-2 during the pathophysiological process of OA. Our hypothesis is that PAR-2 activation during OA course is a major phenomenon for the development/progression of the disease, and that PAR-2 seems a suitable candidate for the development of new therapeutic approaches targeting not only the cartilage but also the subchondral bone. Thus, we have performed in vitro studies on OA chondrocyte (Cr) and osteoblasts (Ob) cells issued respectively from human femoral condyle cartilage and subchondral bone. Our results showed that PAR-2 was expressed at a higher level in the OA Cr and Ob compared to normal cells. Moreover, PAR-2 is found to be positively regulated by some factors present during the course of OA, such as IL-1β, TNF-α and TGF-β in the OA Cr, and IL-1β, TNF-α and PGE2 in the OA Ob. In addition, the main catabolic and inflammatory factors including MMP-1, MMP-13 and COX-2 are enhanced following PAR-2 receptor activation in the OA cartilage. Similarly, the activation of PAR-2 in OA Ob lead to an increased production of pro-resorptive factors such as RANKL, IL-6, MMP-1 and MMP-9, and an increased pro-resorptive activity of these cells. In addition, in both tissue types, PAR-2 activation increases the activity of certain protein MAPKinases family such as Erk1/2, p38 and JNK. Finally, we have performed an in vivo study using an OA induced model in wild-type and PAR-2 gene knock-out mice. Our results demonstrated that the absence of PAR-2 expression and production influenced the inflammatory process and structural changes affecting the cartilage and the subchondral bone, leading to a slowing down of OA development. Our research investigation have bring to light that PAR-2 receptor is a key element during OA process by acting on the cartilage catabolic and inflammatory pathways as well as the tissue remodelling of the subchondral bone. OBJECTIF: Récemment, nous avons démontré que la modification du collagène type II (Col II) par le 4-hydroxynonénal (HNE), un produit de la peroxydation lipidique, est augmentée dans le cartilage arthrosique sans qu’on sache la signification de cette augmentation dans la pathogenèse de l’arthrose. L’objectif de cette étude vise à démontrer que cette modification affecte l’interaction chondrocytes/matrice extracellulaire (MEC) et en conséquence induit des changements phénotypiques et fonctionnels de ces cellules. METHODES: Des plaques de culture ont été préalablement cotées avec du Col II puis traitées avec du HNE (0.1-2 mM) excepté le puits contrôle. Les chondrocytes ont été ensuite ensemencés puis incubés pendant 48 heures. La viabilité des cellules est évaluée par le test MTT. Le Western blot est utilisé pour mesurer l’expression des molécules d’adhésion (l’ICAM-1 et l’intégrine α1β1), de la cyclooxygenase-2 (COX-2), du Col II ainsi que la phosphorylation de la p38 MAPK, ERK1/2 et NF-κB-p65. La RT-PCR en temps réel est utilisée pour mesurer l’expression de l’ARNm de l’ICAM-1, des intégrines α1β1, de la COX-2 et de la métalloprotéinases-13 (MMP-13). La détermination de l’expression de l’ICAM-1 à la surface des cellules est réalisée par cytométrie de flux. Des kits commerciaux ont servi pour mesurer le niveau de la MMP-13, de la prostaglandine E2 (PGE2), de l’activité de la caspase-8 et de la phosphorylation de la p38 MAPK, ERK1/2 et NF-κB-p65. RESULTATS: La modification du Col II par 0.2 mM HNE induit significativement l’expression des molécules d’adhésion telles que l’ICAM-1 et l’intégrine α1β1, de la MMP-13 sans avoir un effet sur la morphologie, la survie et le phénotype cellulaires. Nos résultats montrent aussi une forte augmentation de la phosphorylation de la p38 MAPK, d’ERK1/2 et de NF-κB-p65. Cependant, la modification du Col II par 2 mM HNE affecte la morphologie et la viabilité cellulaires et induit l’activité de la caspase-8. Elle inhibe fortement l’expression des integrines α1β1 et du Col II ainsi que la phosphorylation de l’ERK1/2 et de NF-κB-p65, mais par contre, induit significativement la production de la COX-2 et son produit la PGE2 ainsi que la phosphorylation de la p38 MAPK. Fait intéressant, le prétraitement des complexes HNE/Col II par 0.1 mM de carnosine empêche les changements phénotypiques et fonctionnels des chondrocytes. CONCLUSION : Ces nouveaux résultats suggèrent le rôle important de la modification du Col II par le HNE dans l’arthrose, en affectant le phénotype et le fonctionnement cellulaires des chondrocytes. La carnosine, par sa capacité de neutraliser le HNE, a révélé d’être un agent promoteur dans le traitement de l’arthrose., OBJECTIVE: The regulation of cell phenotype and function by the surrounding environment is deeply altered by the oxidative modifications of extracellular matrix (ECM) components that modify their structural and functional properties. This modification may be one cause involved in cartilage degradation in osteoarthritis (OA). Type II collagen (Col II) was reported to be targeted for 4-hydroxynonenal (HNE) binding, a very reactive product of lipid peroxydation. In the present study, we investigated whether HNE-binding to Col II affects OA chondrocytes phenotype and function and then, we determined the protective role of carnosine treatment in preventing these changes. METHODS: Isolated human OA chondrocytes were seeded in control wells and in HNE/Col II adducts-coated plates and incubated afterwards for 48 hours. Adhesion molecules at protein and mRNA levels were determined by Western blotting, flow cytometry and real-time RT-PCR. Commercial kits were used to evaluate cell death, caspase-8 activity and levels of prostaglandin E2 (PGE2), matrix metalloproteinase-13 (MMP-13), MAPK and NF-κB-p65. Col II, cyclooxygenase-2 (COX-2), MAPK and NF-κB-p65 levels were assessed by Western blotting. RESULTS: After 48 hours of incubation, the modification of Col II by 0.2 mM HNE induced strongly the expression of ICAM-1, integrin α1β1, MMP-13 and slightly COX-2 as well as PGE2 release without affecting cell morphology and viability as well as Col II expression. However, the modification of Col II with 2 mM HNE induced shape changes of cells from typical chondrocyte-like polygon shape to round semi-detached, affecting cells viability and inducing caspase-8 activity. It inhibited the expression of ICAM-1, integrin α1β1 and Col II, but in contrast, induced strongly PGE2 release and COX-2 expression. All these effects were prevented by 0.1 mM carnosine treatment, an HNE trapping drug. Carnosine was added to HNE-modified, Col II-coated plates 1h before cell seeding. Upon examination of different signalling pathways involved in these responses, we found that modified Col II with 2 mM HNE inhibited strongly the phosphorylation of ERK1/2 and NF-κB-p65 but induced strongly p38 MAPK. In contrast, the results indicated that MAPK and NF-κB-p65 were activated when cells were incubated with modified Col II by 0.2 mM HNE. CONCLUSION: The interaction between chondrocytes and collagen-bound HNE modulates different signalling pathways via adhesion molecules regulation and consequently leads to the expression of catabolic and inflammatory factors. Carnosine was shown to be an efficient HNE trapping agent able to counteract these effects.

The hypothesis of the present study was that bacterial communities would differentiate under Eucalyptus camaldulensis and that an enhancement of arbuscular mycorrhizal (AM) density would minimize this exotic plant species effect. Treatments consisted of control plants, preplanting fertilizer application and AM inoculation. After 4 months of culture in autoclaved soil, E. camaldulensis seedlings were either harvested for growth measurement or transferred into containers filled with the same soil but not sterilized. Other containers were kept without E. camaldulensis seedlings. After 12 months, effects of fertilizer amendment and AM inoculation were measured on the growth of Eucalyptus seedlings and on soil microbial communities. The results clearly show that this plant species significantly modified the soil bacterial community. Both community structure (assessed by denaturing gradient gel electrophoresis profiles) and function (assessed by substrate-induced respiration responses including soil catabolic evenness) were significantly affected. Such changes in the bacterial structure and function were accompanied by disturbances in the composition of the herbaceous plant species layer. These results highlight the role of AM symbiosis in the processes involved in soil bio-functioning and plant coexistence and in afforestation programmes with exotic tree species that target preservation of native plant diversity.

L'épandage de déchets organiques liquides sur les sols accroît les risques d'apparition de conditions anaérobies et de mobilisation de métaux. En conditions anaérobies, les activités microbiennes sont le moteur de changements géochimiques au travers des réductions, des changements de pH, de la production nette de ligands minéraux et organiques complexant les métaux ou des altérations-néoformations des phases solides. Cette étude a pour but d'identifier les principaux processus engagés dans la réduction des métaux (Fe, Mn, Cr et Ni) sur un ferralsol de l'île de la Réunion, additionné de vinasse, déchet liquide de rhumerie. L'analyse a été menée par incubation en batch sous atmosphère de N2. Trois traitements ont été effectués: témoin sol + eau (C), sol + vinasse (+W) et sol stérile + vinasse stérile (+W+S). Durant 40 jours d'incubation, nous avons mesuré (i) les pressions partielles des gaz, (ii) les concentrations en solution des glucides totaux et protides totaux des solutés organiques et minéraux, les concentrations totales des métaux et du FeII, et (iii) le pH et EH. Les glucides totaux et protides totaux représentaient respectivement 35,7% et 6,4% du Carbone organique dissous (COD) de la vinasse. Les protides ont été très rapidement adsorbés et non détectés dans la solution du sol après 7 jours d'incubation. Les glucides, polyols et petits acides organiques ont disparu majoritairement au cours des 7 premiers jours d'incubation par des processus fermentaires et acétogénèse, aboutissant à la production d'acétate, butyrate et propionate. Après 21 jours d'incubation, l'acétate a été consommé par méthanogénèse. Les mobilisations de Fe et Mn ont augmenté durant les 21 premiers jours et légèrement diminué après cette date. A l'inverse, les mobilisations en Cr et Ni ont été accentuées par l'apport de vinasse, sans pour autant varier par la suite; ces mobilisations représentaient moins de 0,1% de la quantité maximale de Fe mobilisé. Les réductions de Fe et de Mn résulteraient principalement d'une faible production ou de l'oxydation de H2 produit par fermentation et acétogénèse; FeII en solution représente probablement une faible proportion de l'ensemble du fer réduit.

Identifier le rôle et la contribution de chaque flore au sein de l'écosystème fromager a déjà fait l'objet de nombreux travaux, mais les modes d'investigation sont restés trop souvent descriptifs par manque d'outils moléculaires. L'objectif principal de cette étude était d'élucider les voies métaboliques impliquées dans la désacidification et la production de composés soufrés chez trois levures (Debaryomyces hansenii, Kluyveromyces marxianus et Yarrowia lipolytica) et une bactérie de surface (Brevibacterium linens) par une approche transcriptomique. La conception et l'utilisation d'une puce à ADN dédiée à ces quatre micro-organismes ont permis de mettre en évidence une divergence des voies cataboliques de la L-méthionine entre les levures et la bactérie d'affinage. L'ensemble des résultats obtenus a montré que la transamination est l'étape essentielle du catabolisme de la L-méthionine chez les levures. Elle s'accompagne d'une accumulation transitoire de l'acide α-céto-γ- méthylthiobutyrique chez Y. lipolytica, et essentiellement de sa forme réduite, l'acide α- hydroxy-γ-méthylthiobutyrique, chez K. marxianus et D. hansenii. La dégradation de ces composés se traduit par une augmentation de la production de méthanethiol et des composés soufrés volatils qui en résultent. Une étude plus approfondie réalisée sur Y. lipolytica a montré que les gènes ARO8 et BAT2 jouent un rôle prépondérant dans l'étape de transamination de la L-méthionine chez cette levure. En revanche, chez la bactérie B. linens, l'enzyme clef du catabolisme de la L-méthionine est la L-méthionine γ-lyase. L'apport de L-méthionine dans le milieu de culture induit fortement l'expression du gène mgl et génère une large gamme de composés soufrés volatils. L'étude des voies de dégradation du lactose et du lactate chez les levures a également permis d'obtenir des informations sur la part fonctionnelle de chaque espèce au cours de l'affinage. Ainsi, il semblerait qu'en culture mixte, K. marxianus soit plus impliqué dans la consommation du lactose via l'induction des gènes LAC12 et LAC4, D. hansenii dans le métabolisme du pyruvate puis le catabolisme du lactate en fin de culture, et Y. lipolytica dans la dégradation de la L-méthionine via l'induction des gènes ARO8 et BAT2. De façon plus globale, l'ensemble de nos résultats permet de mettre en évidence la possibilité de complémentarités métaboliques entre les levures d'affinage.

Background & Aims : During fasting, skeletal muscle exports increased amounts of glutamine (Gln) while increasing the production of this amino acid by glutamine synthetase (GS) in order to maintain the intramuscular Gln pool. Glucocorticoid hormones are believed to be the principal mediators of GS induction during stress conditions. The aim of this study was to evaluate (1) the effect of fasting on GS activity and expression in skeletal muscle during aging and consequently, (2) the role of glucocorticoids in fasting-induced GS activity. Methods : Male Wistar rats (6-, 22-month old) were fasted for 5 days and both the activity and expression of GS were measured in tibialis anterior muscle. To better demonstrate the role of glucocorticoids in the response of GS to fasting, we suppressed their action by RU38486 administration (a potent glucocorticoid antagonist) and their production by adrenalectomy in fed and fasted rats. Results : An increase in fasting-induced GS activity was observed in skeletal muscles from both adult and aged rats. Adrenalectomy, but surprisingly not RU38486, suppressed the fasting-induced increase in GS activity and expression. Conclusion : The data clearly show that the GS responsiveness to fasting was not modified by aging in skeletal muscle.

The mechanisms leading to hypoalbuminemia in sepsis were explored by measuring plasma volume, albumin distribution, plasma albumin transcapillary escape rate (TER), and efflux (TER × albumin intravascular pool). These parameters were quantified in infected rats, injected intravenously with live Escherichia coli, and pair-fed and well-fed rats using an injection of35S-albumin and measuring plasma and whole body albumin concentrations. Animals were studied on days 1, 6, and 10 after infection. In pair-fed rats, neither albumin distribution nor exchange rate between the intra- and extravascular compartments was modified. The increase of plasma volume after infection partly explained hypoalbuminemia. Infection resulted in a reduction of the total albumin pool of the body all along the experimental period, indicating a net loss of the protein. Albumin TER (%/day) was significantly increased 1 and 6 days after infection, but the absolute efflux was increased only on day 1. Normal values were observed on day 10. Therefore, an accelerated plasma efflux contributes to hypoalbuminemia only during the early period of sepsis. During this phase, the protein was retained in the extravascular space where it was probably catabolized. Later on, other factors are probably involved.

L'épandage de déchets organiques liquides s'intensifie et permet, avec leur élimination, d'enrichir le sol en nutriments minéraux et composés organiques. Toutefois, il peut engendrer des conditions anaérobies transitoires favorables à la mobilisation des métaux. Dans ces conditions, les activités microbiennes sont le moteur des évolutions géochimiques via l'installation de conditions réductrices, le changement du pH de la solution, les productions nettes de ligands (minéraux et organiques), et l'altération de minéraux. Les modèles décrivant les processus prépondérants doivent alors distinguer explicitement activités microbiennes et réactivité géochimique abiotique. Maintenir leur opérationnalité suppose de hiérarchiser au préalable les besoins de description des grandes voies du catabolisme anaérobie des composés organiques (glucides, lipides protides). A cette fin, nous avons mené un travail expérimental basé sur des incubations en batch et en anaérobiose de boues d'un sol volcanique de la Réunion, riche en Cr et Ni, additionné ou non d'une vinasse de rhumerie. Quatre traitements ont été réalisés : (i) témoin (sol naturel), (ii) sol stérilisé + vinasse stérilisée, (iii) sol naturel + vinasse stérilisée, et (iv) sol naturel + vinasse naturelle. A quelques dates (0, 7 et 21 j), nous avons suivi les quantités totales de glucides, protides et lipides en solution. A plus petits intervalles de temps incluant ces mêmes dates, nous avons suivi des variables indicatrices des métabolismes ou des modifications du milieu (gaz, solutés organiques et minéraux dont éléments métalliques, EH, pH). La vinasse ne contenait que peu de lipides. Avec le sol, leur concentration en solution s'est très vite stabilisée. Les glucides, initialement deux fois plus abondants que les protides, ont disparu en 7 jours tandis que les protides étaient toujours abondants en fin d'incubation. La disparition des glucides a été associée à des fermentations importantes avec accumulation d'acétate pouvant complexer les métaux. La concentration de ce dernier a décru à partir de 7 j et semblait directement associée à la production de méthane. Avant 7 j d'incubation, une mobilisation des éléments métalliques majeurs (Fe et Mn) par réduction microbienne, sans phase initiale de latence, a été observée. Au delà du 7ème jour, nous avons constaté une disparition partielle de ces métaux. Ce phénomène a été attribué à la précipitation de ces éléments, favorisée par des changements de spéciation en solution suite à la disparition de l'acétate. Le comportement du Cr était très fortement corrélé aux comportements de Fe et plus particulièrement de Mn. En contraste, la durée de mobilisation initiale de Ni est apparue plus brève que Fe, Mn et Cr et la disparition finale simultanément plus précoce et brève (entre les dates 4 j et 7 j). Les concentrations maximales du Cr et du Ni en solution sont restées inférieures à 0.01% et 0.21% respectivement de la concentration maximale en Mn. Ainsi dans nos conditions expérimentales, les glucides ont été l'acteur premier de la réduction des métaux. La participation des protides et des corps microbiens morts reste à mieux cerner. Rat gastrocnemius showed increased protein degradation (+75–115%) at 48 h after traumatic injury. Injured muscle showed increased cathepsin B activity (+327%) and mRNA encoding cathepsin B (+670%), cathepsin L (+298%), cathepsin H (+159%), and cathepsin C (+268%). In in situ hybridization, cathepsin B mRNA localized to the mononuclear cell infiltrate in injured muscle, and only background levels of hybridization were observed either over muscle cells in injured tissue or in uninjured muscle. Immunogold/electron microscopy showed specific staining for cathepsin B only in lysosome-like structures in cells of the mononuclear cell infiltrate in injured muscle. Muscle cells were uniformly negative in the immunocytochemistry. Matrix metalloproteinase-9 (granulocyte-macrophage gelatinase) mRNA and activity were not present in uninjured muscle but were expressed after trauma. There was no activation of the ATP-ubiquitin-proteasome-dependent proteolytic pathway in injured muscle, by contrast to diverse forms of muscle wasting where the activity of this system and the expression of genes encoding ubiquitin and proteasome elements rise. These results suggest that proteolytic systems of the muscle cells remain unstimulated after local injury and that lysosomal enzymes of the inflammatory infiltrated cells are likely to be the major participant in protein catabolism associated with local trauma.

Plants possess both anabolic and catabolic pathways for the essential amino acid lysine (Lys). However, although the biosynthetic pathway was clearly shown to regulate Lys accumulation in plants, the functional significance of Lys catabolism has not been experimentally elucidated. To address this issue, we have isolated an Arabidopsis knockout mutant with a T-DNA inserted into exon 13 of the gene encoding Lys ketoglutarate reductase/saccharopine dehydrogenase. This bifunctional enzyme controls the first two steps of Lys catabolism. The phenotype of the LKR/SDH knockout was indistinguishable from wild-type plants under normal growth conditions, suggesting that Lys catabolism is not an essential pathway under standard growth conditions. However, mature seeds of the knockout mutant over-accumulated Lys compared with wild-type plants. This report provides the first direct evidence for the functional significance of Lys catabolism in regulating Lys accumulation in seeds. Such a knockout mutant may also provide new perspectives to improve the level of the essential amino acid Lys in plant seeds.

Plasma albumin is well known to decrease in response to inflammation. The rate of albumin synthesis from both liver and plasma was measured in vivo by use of a large dose ofl-[2H3-14C]valine in rats injected intravenously with live Escherichia coli and in pair-fed control rats during the acute-phase period (2 days postinfection). The plasma albumin concentration was reduced by 50% in infected rats compared with pair-fed animals. Infection induced a fall in both liver albumin mRNA levels and albumin synthesis relative to total liver protein synthesis. However, absolute liver albumin synthesis rate (ASR) was not affected by infection. In plasma, albumin fractional synthesis rate was increased by 50% in infected animals compared with pair-fed animals. The albumin ASR estimated in the plasma was similar in the two groups. These results suggest that hypoalbuminemia is not due to reduced albumin synthesis during sepsis. Moreover, liver and plasma albumin ASR were similar. Therefore, albumin synthesis measured in the plasma is a good indicator of liver albumin synthesis.

Not available, Revue Le catabolisme microbien des stérols passe par deux voies distinctes. L'une concerne la dégradation du noyau stéroïde, tandis que l'autre concerne le clivage de la chaîne latérale. Ce clivage est effectué par oxydation. Les actinomycètes sont généralement actifs dans le catabolisme des stérols. Le clivage sélectif de la chaîne latérale des stérols, qui fournit le noyau stéroïde est d'un intérêt économique remarquable. La cholestérol oxydase (COX), première enzyme du catabolisme des stérols, transforme la structure 5-ène-3[bêta]-ol en 4-ène-3-one. Les COXs microbiennes sont produites sous forme sécrétée et/ou sous forme cellulaire. Elles sont monomériques (54-62 kDa) et polymorphes. Le gène de l'enzyme et son opéron ont été identifiés chez une espèce de Streptomyces. Ce gène a été aussi étudié chez Brevibacterium sterolicum. Les COXs sont utilisées dans plusieurs applications. Resultats Deux isolats du sol (GK1, GK3), actifs dans le catabolisme des stérols, ont été identifiés comme appartenant au genre Rhodococcus. La souche GK1 (CIP 105335) produit la COX sous formes extracellulaire et cellulaire. La COX cellulaire est localisée à l'extérieur de la membrane cytoplasmique. Un essai calorimétrique a été mis au point pour doser l'activité de la COX. Il permet de doser l'enzyme acellulaire, et aussi l'enzyme liée aux cellules sans qu'elle soit extraite. L'induction de la COX de Rhodococcus sp. GK1 est liée à la chaîne latérale des stérols. La structure 3[bêta]-ol-5-ène ne suffit pas pour l'induire. La culture de cette souche sur stérols, comme seules sources de carbone et d'énergie, est marquée par la production de la COX cellulaire uniquement. Cette forme est extractible par traitement des cellules avec des détergents non ioniques. La croissance de GKl sur une source de carbone différente des stérols, ou sur un stérol plus un autre composé carboné, est marquée par une sécrétion importante de la COX. Les formes de la COX de GK1 ont la même spécificité de substrats. La double liaison en C-5 n'est pas nécessaire pour l'oxydation de la fonction alcool. La longueur de la chaîne latérale des stérols influence le niveau de l'activité. Ces formes possèdent un certain caractère hydrophobe. Le Triton X-100 ou le Lubrol PX (0,1- 0,2 %, p/v) stimulent leur activité. La COX de GK1 est relativement stable à des températures allant jusqu'à 50°C. La COX extracellulaire et cellulaire de cette souche ont été purifiées. Leur masse moléculaire est environ 59 kDa. La séquence en aminoacides du ségment N-terminal de la COX extracellulaire est: H2N-Ala-Pro-Pro-Val-Ala-Ser-XArg- Tyr-X-(Phe)-. Le pl de cette forme est de l'ordre de 9,0. Une cholestérol ester hydrolase (estérase) a été mise en évidence dans les cellules de GKl. Une alcool secondaire déshydrogénase NAD-dépendante a été obtenue de cette souche et caractérisée. Sa masse moléculaire apparente est 60 kDa. Elle catalyse la réduction des monocétones et des dicétones comme le 2-pentanone et le diacétyle. Elle oxyde des alcools secondaires comme l'isopropanol et le 2-octanol.

Skeletal muscle catabolism is a characteristic metabolic response to sepsis. We investigated the ability of physiological insulin (2 nM) or insulin-like growth factor I (IGF-I, 10 nM) concentrations to modify protein metabolism during incubation of epitrochlearis 2, 6, or 15 days after injection of live Escherichia coli. On days 2 and 6 postinfection, skeletal muscle exhibited an exacerbated negative protein balance resulting from both an inhibition in protein synthesis (25%) and an enhanced proteolysis (90%) compared with controls. By day 15 postinfection, protein balance in infected rats was significantly improved compared with either day 2 or 6. At this time, protein synthesis was augmented and protein degradation was decreased in infected rats relative to day 6. Insulin or IGF-I stimulated protein synthesis in muscles from septic and control rats in vitro to the same extent at each time point examined. The ability of insulin or IGF-I to limit protein degradation was severely blunted 48 h after infection. On day 6 postinfection, the effect of insulin or IGF-I to inhibit proteolysis was more pronounced than on day 2. Incubation with IGF-I limited proteolysis to a greater extent than insulin on both days in infected but not control rats. By day 15, insulin diminished proteolysis to the same extent as in controls. The results suggest that injection of bacteria causes fundamental derangements in protein metabolism that persist for days after infection.

Les Pseudomonas spp fluorescents saprophytes à oxydase positive, qui regroupent les espèces P chlororaphis, P fluorescens et P putida, sont des bactéries ubiquistes. Au sein de ces trois espèces, certaines souches présentent un intérêt pour l’agriculture car elles améliorent la croissance et l’état sanitaire des plantes. Ces effets bénéfiques sont associés à la production de métabolites secondaires (antibiotiques, sidérophores, phytohormones) dont la synthèse est influencée par le métabolisme basal. Les Pseudomonas spp fluorescents jouent aussi un rôle dans la protection de l’environnement. Leur métabolisme carboné et énergétique est en effet responsable de la dissimilation des nitrates et de la dégradation de composés xénobiotiques. Dans cet article sont présentées les voies d’assimilation des principaux oses, acides organiques et acides aminés, ainsi que les voies respiratoires couplées à ce catabolisme. L’utilisation des sucres simples se caractérise par l’oxydation extracellulaire du glucose et la prédominance de la voie d’Entner-Doudoroff. Le catabolisme des acides organiques et aminés est associé aux cycles de Krebs et du glyoxylate qui jouent un rôle majeur dans le métabolisme de ces bactéries. À l’exception de l’arginine, l’assimilation des composés étudiés n’est possible qu’en conditions aérobies ou en présence d’oxydes d’azote. La variété du métabolisme des Pseudomonas spp fluores cents confère à ces bactéries une plasticité importante dans leur adaptation à des environnements variés, tels que le sol et la rhizosphère., Saprophytic fluorescent Pseudomonas spp constitute an oxidase-positive group, including the species P chlororaphis, P fluorescens and P putida, which are common in the environment. Among these three species, some strains improve the plant growth and health and consequently are useful in agriculture. These beneficial effects are ascribed to the production of secondary metabolites (antibiotics, siderophores, phytohormones) and their synthesis is affected by the basal metabolism of the bacteria. Fluorescent pseudomonads are also implicated in the conservation of the environment. Their carbon and energy metabolism is indeed responsible for nitrate dissimilation and the degradation of xenobiotic compounds. The catabolic pathways of the main sugars, organic acids and amino acids, as well as the respiratory pathways twined to this catabolism, are presented in the present review. The use of carbohydrates is characterized by the extracellular oxidation of glucose and the prevalence of the Entner-Doudoroff pathway. The catabolism of organic and amino acids is linked to the cycles of Krebs and of the glyoxylate which play a major role in the metabolism of the bacteria. Except for arginine, the utilization of the studied compounds is only possible in the presence of oxygen or nitrogen oxides. The metabolic diversity of fluorescent Pseudomonas spp gives to these bacteria a great ability to adapt to various environments, such as the soil and the rhizosphere.

Résultats sur la variabilité des teneurs en ARN et en protéines dans le cytosol du latex en fonction des effets initiaux de la saignée, des stimulants hormonaux, de la production et de la fraction du latex prélevé au cours de la saignée. La variabilité du rARN et poly (A)+ARN est associée aux changements de l'activité glycolytique qui dépendent de la teneur en sucre, du pH du latex et de l'activité de l'invertase. Une augmentation des ARN est accompagnée par une augmentation de l'invertase ou la saccharose synthétase, durant les 1ères saignées chez des arbres déjà adaptés à l'exploitation régulière. Sur la base des corrélations établies, une implication de l'ARN et des changements protéiques dans la production du latex et la senescence des laticifères, est suggérée. L'ARN polysomal isolé à partir de latex stimule la synthèse protéique dans des systèmes acellulaires contenant du lysat de réticulocyte de lapin et code pour une large variété de polypeptides.
