This study aimed to evaluate the influence of the Er,Cr:YSGG irradiation and 980-nm diode lasers on the surface roughness (SR) and volume loss (VL) of dentin subjected to cariogenic challenge. Subsequently, 130 specimens of bovine dentin were divided into the following 13 groups: NT: no treatment; FG: fluoride gel; FV: fluoride varnish; Di: 980-nm diode; Di + FG; Di + FV; FG + D; FV + Di; Er: Er,Cr:YSGG; Er + FG; Er + FV; FG + Er and FV + Er. Er,Cr:YSGG laser parameters were as follows: 0.25 W; 5.0 Hz; 4.46 J/cm2 without water and 55% air. Furthermore, the 980-nm diode laser parameters were 2.0 W; 2.0 Hz; 21.41 J/cm2. The samples from each group were subjected to pH cycling. A confocal laser scanning microscope was used to evaluate SR and VL. Difference between the volume of the reference and treated areas + DES/RE was used to determine SR and VL. The mean values of the different groups were subjected to analysis of variance and Tukey's post-hoc test. The VL values were analyzed using the Kruskal-Wallis and Dunn post-hoc test (p < 0.05). The SR of the reference area did not show a statistically significant 1807-3107-bor-38-e025treatment and cariogenic challenge (p > 0.05). Moreover, VL in the FV + Di and FV + Er groups showed a statistically significant difference compared with areas submitted to different types of treatment and cariogenic challenge (p > 0.05). Er,Cr:YSGG and 980-nm diode lasers associated with fluoride varnishes decreased dentin VL in bovine teeth submitted to cariogenic challenge.

Purpose: To evaluate the antimicrobial activity of dentifrices based on Malva sylvestris and propolis and the effect on the acrylic resin., Methods: The inhibitory effect against Candida albicans, Enterococcus faecalis and Streptococcus mutans was determined. The specimens of resin were divided into groups: Control (brushing with distilled water); brushing with Colgate; brushing with Protex; brushing with Malvatricin, brushing with Proporalcare Dental Gel and brushing with Green Propolis Dental Gel. Brushing was carried out in a brushing machine. Roughness and color were assessed. The data were analyzed by ANOVA, followed by the Bonferroni post test (α= 0.05)., Results: The products evaluated showed antimicrobial activity against all microorganisms (P< 0.05), except Proporalcare Dental Gel, which was not effective against E. faecalis. Greater roughness (P< 0.05) was observed in the samples treated with Colgate. The samples from control and Protex groups showed the lowest color variation and the highest luminosity, with a difference in relation to the others (P< 0.05), which showed higher values, although clinically acceptable., Clinical Significance: The dentifrices based on natural products can be indicated for cleaning prostheses, since they showed antimicrobial activity and did not cause perceptible color changes of the resin tested. Alternatives have been sought to add an antimicrobial property to denture adhesives. This study evaluated the antimicrobial potential of adhesives associated with nanostructured silver vanadate decorated with silver nanoparticles (β-AgVO3). Specimens in acrylic resin were treated with the adhesives associated with β-AgVO3 (1%, 2.5%, 5% and 10%). As control, specimens treated only with Ultra Corega Cream (UCC) or Ultra Corega Powder (UCP) adhesive were used. Multispecies biofilm of Candida albicans, Candida glabrata, Streptococcus mutans and Staphylococcus aureus was evaluated by counting colony forming units per milliliter (CFU/mL), colorimetric assay and fluorescence microscopy. The data were analyzed using the two-way analysis of variance (ANOVA) and Bonferroni multiple comparisons test (α=0.05). For both adhesives, a small amount of β-AgVO3 (1%) completely inhibited S. mutans (P⟨0.05). For the other microorganisms, there was a reduction in metabolic activity and complete inhibition in the groups with intermediate or greater amounts of nanomaterial (P⟨0.05), except for C. albicans, which was reduced (P⟨0.05) but not completely inhibited in UCP. Microscopy that showed less biofilm in the groups with β-AgVO3 and in the UCC than UCP. Denture adhesives in powder and cream form with β-AgVO3 showed potential antimicrobial activity against multispecies biofilm. Background: The aim of this study was to determine if there is a correlation between fluoride release, surface hardness, and diametral tensile strength of restorative glass ionomer cements (GICs)., Material and Methods: Conventional (Riva Self Cure) and resin-modified (Riva Light Cure) GICs were used. Thirty-four samples (ø 6 x 3 mm) were prepared for each cement. The kinetics of fluoride release (n=4) was evaluated over 28 days using a fluoride-selective electrode (ISE 4010-C00). The analysis of surface hardness (n=10) was performed using a microhardness tester (Shimadzu HMV-2000, Japan) with a Knoop indenter and a load of 25 gf for 30 seconds. The diametral tensile strength test (n=10) was conducted on a universal testing machine at a speed of 0.75 mm/min. Fluoride release data were analyzed by two-way repeated measures ANOVA and Bonferroni post hoc test, while independent t-test was used for other analyses (α=0.05)., Results: Overall, the groups showed higher fluoride release until day 7 and a progressive decrease until day 28. On day 1 and day 21, Riva Self Cure showed a higher level of release than Riva Light Cure ( p =0.026). Riva Light Cure showed higher diametral tensile strength ( p <0.0001) and surface hardness ( p =0.034) than Riva Self Cure. A negative correlation was found, indicating that higher fluoride release is associated with lower surface hardness and diametral tensile strength., Conclusion: Fluoride release and mechanical performance are related properties of GICs, and these properties exhibit different values depending on the type of material. Resin-modified GIC release less fluoride but exhibit better mechanical performance compared to conventional GIC. Background: The objective this study was to evaluate the influence of preventive remineralizing techniques on surface roughness and volume loss of dentin submitted to erosive and/or abrasive challenges., Material and Methods: One hundred and eighty specimens of bovine root dentin were made; half of each was isolated (without treatment - WT) and half was subjected to the following remineralizing techniques: fluoride varnish (FV); Regenerate Boosting Serum® (RBS); Er,Cr:YSGG laser (L); fluoride varnish+laser (FV+L); Regenerate Boosting Serum®+laser (RBS+L). The specimens were submitted to erosive, abrasive and erosive followed by abrasive challenge. Erosion was carried out for 5 minutes, twice a day for 10 days. Abrasion was performed with an electric toothbrush and slurry solution for 60 seconds. The evaluation was performed by confocal laser scanning microscopy. Analysis of variance and Tukey tests were used for surface roughness; volume loss comparison was performed using the Kruskal-Wallis test and Dunn's post-hoc ( p <0.05)., Results: There was no statistically significant difference in the surface roughness of the reference area in relation to the areas submitted to different types of treatment and challenges ( p >0.05). Regarding volume loss, the untreated group submitted to erosive/abrasive challenges showed greater percentage of volume loss compared to the other groups ( p <0.05)., Conclusions: It is concluded that preventive remineralizing techniques are effective in maintaining dentin volume after erosive/abrasive challenges. Background: To evaluate the effectiveness of Er:YAG and Nd:YAG laser on dentin hypersensitivity prevention, associated or not to acidulated phosphate fluoride (APF) after erosive challenge., Material and Methods: 104 specimens were obtained from bovine dentine and divided into groups (n=13): G1: Er:YAG; G2: Er:YAG followed by application of APF; G3: application of APF followed by Er:YAG, simultaneously; G4: Nd:YAG; G5: Nd:YAG followed by application of APF; G6: application of APF followed by Nd:YAG, simultaneously; G7:application of APF; G8: untreated. The parameters for Er:YAG were:10s, distance of 4mm, water cooling flow of 2mL/min, 2Hz, 3.92J/cm2. For the Nd:YAG: 10s, distance of 1mm, without cooling, 10Hz, 70.7J/cm2. The erosive drink was a cola at 4°C, 3×/day for 1 minute, for 5 days. Roughness and wear analysis were done in confocal laser microscope. Data were statistically analyzed (α=0.05)., Results: As regards roughness, there was no statistically difference among the groups. The groups irradiated with Er:YAG had a volume loss lower. G6 showed higher values than the groups irradiated with Er:YAG and lower than the other groups. The other groups irradiated with Nd:YAG showed similar wear results to the control., Conclusions: The Er:YAG laser showed the lowest volume loss from wear analysis, suggesting the increased the acid resistance of dentin. Aim: To evaluate the biological and mechanical properties of an adhesive with nanostructured silver vanadate (AgVO 3 ). Materials & methods: Specimens in poly(methyl methacrylate) (PMMA) were treated with Ultra Corega Cream (UCCA) denture adhesive with or without AgVO 3 . Biofilms of Candida albicans , Candida glabrata and Streptococcus mutans were grown and the viable cells counted. Fluorescence microscopy was used. The viability of the VERO cell and adhesive strength were evaluated. Results: All concentrations of AgVO 3 reduced the biofilm formation and showed no cytotoxic effect. At 5 min and 24 h, UCCA with 5 and 10% AgVO 3 showed better performance, respectively. Conclusion: AgVO 3 promoted the antibiofilm activity of the adhesive, with a positive effect on the adhesive strength, and was biocompatible.

This study investigated the effect of water at high temperature on the physical and mechanical properties of polyurethane and on biofilm removal, aiming for its applicability in dental unit waterlines. The evaluations were carried out after simulating a 1-year period of daily immersion and measured changes in color, microhardness, surface roughness, and tensile strength before and after reproducing a disinfection protocol. For antibiofilm activity measurement, fragments of waterline were contaminated with Pseudomonas aeruginosa and submitted to the disinfection protocols. Relative to effects on the physical and mechanical properties, immersion in water at 60°C did not promote changes in color and tensile strength. However, lower values were observed for microhardness and increased values for surface roughness. Regarding antibiofilm action, water at 60°C significantly reduced the microbial load and promoted substantial changes in cells morphology. In conclusion, disinfection with water at 60°C demonstrated possible application in controlling cross-contamination in dentistry.

Purpose: To evaluate the effect of different forms of denture adhesives on the formation of biofilm and on adhesive strength, as well as the effectiveness of hygiene protocols for their removal., Materials and Methods: Samples of the heat-cured polymethyl methacrylate denture base resin were made and divided into four groups: control (no adhesive), ultra Corega cream, Corega strip adhesive, and ultra Corega powder (GlaxoSmithKline). Biofilm formation was evaluated by counting colony-forming units and fluorescence microscopy. To evaluate the effectiveness of the hygiene protocols, the samples were divided into five subgroups: brushing with distilled water; brushing with Protex soap; brushing with Colgate toothpaste; immersion in Corega Tabs; and immersion in Corega Tabs followed by brushing with the solution itself. The remaining adhesive was quantified with ImageJ software. The adhesive strength was tested at different time intervals after application. After verifying the data distribution using Shapiro-Wilk test, parametric or nonparametric analysis was applied (α = .05)., Results: Candida albicans formed more biofilm in strip (P = .007) and powder (P = .001), Pseudomonas aeruginosa in cream (P < .001) and powder (P < .001), and Staphylococcus aureus in strip (P < .001). All forms of the adhesives promoted higher biofilm formation when compared to control (P = .003). Brushing with Colgate and Protex was most effective for removing the adhesives (P < .05). Independently, Powder had the highest adhesive strength (P < .05). Only Strip showed a change in adhesive strength, with higher values after 3 hours of application (P = .004)., Conclusion: Daily treatments with mechanical cleaning of the prosthesis are important for removing the adhesives, since the presence of this material can favor biofilm accumulation. The adhesive strength may vary depending on the commercial type, but all forms can be effective in retaining prostheses for a satisfactory period of time.

Context: Artificial teeth are widely used in oral rehabilitation. Despite the benefits, they are more susceptible to colour changes, causing aesthetic problems., Aims: To evaluate the effect of conventional cigarette and straw smoke on the colour of artificial teeth and the effectiveness of hygiene protocols in removing pigmentation., Material and Methods: Acrylic resin incisors were divided into two groups (n = 50): Exposed to conventional cigarette and straw smoke. Regarding the effectiveness of hygiene protocols, the teeth were divided into ten subgroups with predetermined immersion times. The colour was measured with a colorimeter. The CIE values L* a* b* were recorded before and after exposure to smoke and after hygiene protocols. Statistical analysis used T-test of independent samples and two-way ANOVA with Bonferroni post-test (α = 0.05)., Results: Conventional (16.16 ± 1.65) and straw (16.29 ± 1.95) cigarettes provided clinically unacceptable ΔE values, with no significant difference between them (P = 0.719). Conventional cigarettes promoted less luminosity (ΔL = -12.68 ± 1.28) (P < 0.001) and straw greater tendency to yellow (Δb = 11.00 ± 1.46) (P < 0.001). The hygiene protocols influenced the ΔE, ΔL, and Δb of the samples, depending on the type of smoke (P < 0.05)., Conclusions: The conventional and rolled cigarette smoke promote an unacceptable colour change in artificial teeth. Hygiene protocols with the use of brushing, in isolation or in association with chemical solutions are more effective in removing pigmentation caused by both types of cigarettes compared to only the chemical solution., Competing Interests: None

The presence of microplastics in the aquatic ecosystem represents a major issue for the environment and human health. The capacity of organic pollutants to adsorb onto microplastic particles raises additional concerns, as it creates a new route for toxic compounds to enter the food web. Current knowledge on the impact of pristine and/or contaminated microplastics on aquatic organisms remains insufficient, and we provide here new insights by evaluating their biological effects in zebrafish (Danio rerio). Zebrafish larvae were raised in ZEB316 stand-alone housing systems and chronically exposed throughout their development to polyethylene particles of 20-27 μm, pristine (MP) or spiked with benzo[α]pyrene (MP-BaP), supplemented at 1% w/w in the fish diet. While they had no effect at 30 days post-fertilization (dpf), MP and MP-BaP affected growth parameters at 90 and 360 dpf. Relative fecundity, egg morphology, and yolk area were also impaired in zebrafish fed MP-BaP. Zebrafish exposed to experimental diets exhibited an increased incidence of skeletal deformities at 30 dpf as well as an impaired development of caudal fin/scales, and a decreased bone quality at 90 dpf. An intergenerational bone formation impairment was also observed in the offspring of parents exposed to MP or MP-BaP through a reduction of the opercular bone in 6 dpf larvae. Beside a clear effect on bone development, histological analysis of the gut revealed a reduced number of goblet cells in zebrafish fed MP-BaP diet, a sign of intestinal inflammation. Finally, exposure of larvae to MP-BaP up-regulated the expression of genes associated with the BaP response pathway, while negatively impacting the expression of genes involved in oxidative stress. Altogether, these data suggest that long-term exposure to pristine/contaminated microplastics not only jeopardizes fish growth, reproduction performance, and skeletal health, but also causes intergenerational effects., (Copyright © 2022 Elsevier Ltd. Context: A promising option for the prevention of dental caries is the use of laser irradiation., Aims: Evaluate the effects of Er:YAG, Nd:YAG, and CO 2 laser irradiation, associated or not to 2% sodium fluoride (2% NaF), on root caries prevention., Material and Methods: One hundred and four human root dentin samples were divided in eight groups (n = 13). A 9-mm 2 -area on each dentin sample was delimited and treated as follows: G1: no treatment (control); G2: 2% NaF; G3: Er:YAG; G4: 2% NaF + Er:YAG; G5: Nd:YAG; G6: 2% NaF + Nd:YAG; G7: CO 2 ; G8: 2% NaF + CO 2 . When used, the 2% NaF was applied before irradiation for 4 min. The samples were subjected to a 2-week cariogenic challenge, consisted of daily immersion in de-remineralizing solutions for 6 h and 18 h, respectively. Knoop hardness (KHN) were evaluated (10 g and 20 s) at different depths from the dentin surface. The samples (n = 3) were prepared for scanning electron microscopy (SEM). Microhardness data were analysed by one-way analysis of variance (ANOVA) and Fisher's test (α = 5%)., Results: The Er:YAG laser group (KHN = 41.30) promoted an increase in acid resistance of the dentin (P < 0.05) when compared to all groups. There was no synergism between laser irradiation and 2% NaF application. Morphological changes were observed after irradiation with all lasers; carbonization and cracks were also observed, except when Er:YAG were used., Conclusions: Er:YAG laser irradiation can safely increase the acid resistance of the dentin surface of the root, since it promoted a significant increase in surface hardness. Objective: This study evaluated the effect of different glucose concentration on biofilm formation of Candida albicans and Candida glabrata strains isolated from diabetic and non-diabetic individuals., Methods: The study was divided into two stages: (I) selection and identification of 48 C. albicans and C. glabrata strains by polymerase chain reaction followed by restriction fragment length polymorphism analysis (PCR/RFLP); (II) evaluation of biofilm formation by means of viability rates (colony-forming units), biofilm dry matter (mg) and biofilm-covered areas (μm 2 ). Statistical comparisons were performed through nonparametric analysis of longitudinal data in factorial experiments with pairwise comparisons using Friedman Conover's test (α = 0.05)., Results: All the Candida spp. had their identifications confirmed by PCR/RFLP. C. albicans biofilm of strains from diabetic individuals cultivated in different glucose concentration showed higher viability rates than strains from non-diabetic individuals. No difference was observed on viability of C. glabrata biofilm. Regarding biofilm dry matter, C. albicans biofilm of strains from diabetic individuals cultivated in different glucose concentration showed lower amount in weight than strains from non-diabetic individuals. In C. glabrata strains, this result was only observed in biofilms cultivated with no glucose supplementation. With regard to biofilm-covered areas, only glucose supplementation and non-diabetic condition showed a positive effect on C. albicans biofilm development, and no condition affected C. glabrata biofilm formation., Conclusion: The strain type (C. albicans and C. glabrata) isolated from diabetic and non-diabetic individuals influenced on biofilm formation, but glucose supplementation did not., (Copyright © 2021 Elsevier Ltd. This study aimed to evaluate two methods of the incorporation of nanostructured silver vanadate decorated with silver nanoparticles (AgVO 3 ) into acrylic resin and characterize the profile of early and late microbial communities in class and family taxonomic level by pyrosequencing. The specimens were made by adding different concentrations of AgVO 3 (1, 2.5, and 5%) to the heat-activated acrylic resin by two methods: vacuum spatulation (VS) and polymeric film (PF). A control group (0%) without AgVO 3 was also obtained for both methods. After 24 h and 7 days of incubation in human saliva, biofilm samples were collected, DNA was extracted, and 16S rRNA genes were sequenced by the 454-Roche sequencing platform. Seventeen classes and 51 families of bacteria were identified. The abundance of Bacteroidia, Bacilli, Negativicutes, Fusobacteria and Betaproteobacteria classes decreased after 7 days of incubation, and Clostridia, Gammaproteobacteria, and unclassified bacteria increased. The Negativicutes and Betaproteobacteria classes were more abundant when the PF method was used, and Gammaproteobacteria was more abundant when VS was used. The incorporation of 5% AgVO 3 promoted a reduction in the prevalence of Bacilli, Clostridia, Negativicutes, Betaproteobacteria, and unclassified bacteria, and increased Gammaproteobacteria. The addition of AgVO 3 to acrylic resin altered the early and mature microbiome formed on the specimen surface, and the PF method presented a more favorable microbial profile than the VS method.

This study evaluated how Er,Cr:YSGG laser, associated or not with 5% fluoride varnish, influences the surface roughness and volume loss of bovine root dentin submitted to erosive and/or abrasive wear. One hundred and twenty dentin specimens were divided into the groups: without preventive treatment (WPT), 5% fluoride varnish (FV); Er,Cr:YSGG laser irradiation (L), and varnish combined with laser (FV + L). The specimens (n = 10) were subdivided into: 1 = erosion (E); 2 = abrasion (A); and 3 = erosion followed by abrasion (E + A). The erosive solution used was a soft-drink (pH = 2.42 at 4ºC) applied in 5-min cycles twice a day for 10d. Abrasive wear involved brushing for 60s with an electric brush (1,600-oscillations/s) at a load of 2.0N. Surface roughness and volume loss were evaluated using a laser scanning confocal microscope. Roughness data were submitted to one-way ANOVA and Tukey post-hoc test. For volume loss, the Kruskal-Wallis and Dunn's post-hoc tests were used (α = 5%). The lowest values of roughness were found in the control areas of all subgroups (p > 0.05). In the experimental area, the [(WPT) + (E+A)] subgroup had a significantly higher roughness (5.712 ± 0.163 μm 2 ) than the other subgroups (p < 0.05). The L and (FV + L) groups had statistically similar roughness, regardless of the type of wear. The (FV + L) group had the lowest volume loss, regardless of the type of wear performed: [(FV + L) + (E)] = 7.5%, [(FV + L) + (A) = 7.3%, and [(FV + L) + (E + A)] = 8.1%. The subgroup [(WPT) + (E + A)] had the highest volume loss (52.3%). The proposed treatments were effective in controlling dentin roughness. Laser irradiation can be an effective method to increase root dentin resistance after challenges and limit problems related to non-carious lesions.

The modification of endodontic sealers with nanoparticles to confer antimicrobial activity allow greater effect, with interaction at a molecular level. The nanostructured silver vanadate decorated with silver nanoparticles (AgVO 3 ) is a nanomaterial unprecedented in dentistry for this application. This study incorporated the AgVO 3 into three endodontic sealers of different compositions and evaluate the cytotoxicity and release of compounds. The groups of commercially available AH Plus, Sealer 26, and Endomethasone N and groups of the same sealers with incorporated AgVO 3 (at concentrations 2.5, 5, 10%) were prepared, and extracts of the specimens were obtained for 24 h. The cell viability (cytotoxicity) of human gingival fibroblasts (HGF) was assessed after 24 h, 7 and 14 days. Silver (Ag + ) and vanadium (V 4+ /V 5+ ) ion release was quantified after 24 h by ICP-MS. Data were analyzed by Kruskal-Wallis and Dunn's post-hoc (α = 0.05). The cell viability was inversely proportional to treatment time. The Sealer 26 and Endomethasone N groups were cytotoxic for HGF cells, regardless of the incorporation of the AgVO 3 (p > 0.05), and the incorporation reduced cell viability of AH Plus (p < 0.05). The release of ions was proportional to the concentration of AgVO 3 . AH Plus released more Ag + ions, and Sealer 26 and Endomethasone N releases more V 4+ /V 5+ ions. In conclusion, it was not possible to confirm the influence of AgVO 3 on HGF cell viability to Sealer 26 and Endomethasone N, however, nanomaterial influenced cell-viability to AH Plus, so the commercial sealers can be cytotoxic in synergy with the nanomaterial. The release of Ag + and V 4+ /V 5+ was proportional to the AgVO 3 incorporated.

Purpose: To evaluate the surface roughness and hardness of thermopolymerized acrylic resin incorporated with nanostructured silver vanadate (AgVO3) subjected to saliva and beverages., Methods: The 128 specimens (5×5×2 mm) were prepared in thermopolymerized acrylic resin, according to the AgVO3 concentrations (n=32): 0%, 2.5%, 5% and 10%. The roughness and hardness were analyzed before and after immersion in saliva, Coca-Cola, orange juice and red wine, for 12 and 24 days. 2-way ANOVA and Bonferroni test (α= 0.05) were performed., Results: After 12 days, Coca-Cola caused the highest roughness increase in the 2.5% group. The 10% group with saliva presented a higher roughness increase (P= 0.009). The control presented a decrease in roughness when in beverages (P< 0.05). After 24 days, orange juice and Coca-Cola produced a higher decrease in roughness in the control group (P< 0.05). After 12 days, saliva and wine produced a higher decrease in hardness of the 2.5% group (P< 0.05). Coca-Cola produced a decrease in hardness and wine an increase in hardness in the group with 10% AgVO3 concentration (P< 0.05). After 24 days, the group with 2.5% presented the highest decrease in hardness (P< 0.05). The immersions produced decreased hardness in the acrylic resin. Aims: To evaluate the effects of different cleansing solutions on the physical-mechanical properties (roughness, surface hardness, and fatigue resistance) of three polymeric materials used to manufacture retentive attachments for overdentures., Materials and Methods: The roughness and surface hardness analyses each employed 150 specimens measuring . 9 mm × 2 mm in thickness (polyacetal, polytetrafluoroethylene [PTFE], and polyethylene terephthalate [PET]; n = 50 each). For fatigue resistance analysis, 180 retentive attachments measuring 4 mm × 3 mm in height (n = 60 each) were used. The properties were evaluated before and after immersion in different cleansing solutions: distilled water, alkaline peroxide, 0.5% sodium hypochlorite (NaOCl), and Listerine. After the data distribution was verified using Shapiro-Wilk test, parametric or nonparametric analysis was applied (α = .05)., Results: The use of NaOCl caused a significant alteration in the roughness of the materials (P = .011), with a reduction in roughness in polyacetal and an increase in PTFE. The type of solution also influenced the surface hardness (P = .036); with the exception of distilled water, the other solutions promoted increased hardness. During the 24-month period, immersion in water, peroxide, and NaOCl (0.5%) caused increased fatigue resistance (P < .05) of the PET attachments. The immersion protocol resulted in greater fatigue resistance (P < .05) in polyacetal, while PTFE was not affected (P > .05)., Conclusion: Regarding the physical-mechanical properties evaluated, the polymers PET, polyacetal, and PTFE were susceptible to cleansing solutions.

Purpose: To evaluate the influence of the Er,Cr:YSGG laser with or without the 5% fluoride varnish on the acid resistance of dentin after erosive challenge., Methods: 36 incisors were selected and sectioned, obtaining 72 specimens of 4 mm × 4 mm and randomly divided into eight groups (n = 9). In G1: application of Er,Cr:YSGG (0.1W; 5Hz, air 55%); G2: laser (0.25W; 5Hz, air 55%); G3: fluoride varnish + laser (0.1W; 5Hz, air 55%); G4: fluoride varnish + laser (0.25W, 5Hz, air 55%); G5: fluoride varnish + laser (0.1W; 5Hz, without air); G6: fluoride varnish + laser (0.25W, 5Hz, without air); G7: fluoride varnish and G8: no treatment. When used, the laser was irradiated without water cooling, scanning mode during 10 seconds. The surface roughness data were subjected to ANOVA. For wear profile, we used Kruskal-Wallis test and Dunn post-hoc, all with α= 0.05., Results: The results showed no statistically significant difference when comparing the groups as regards to the surface roughness (P> 0.05). Regarding the percentage of lost volume, the G5 and G6 groups presented the best results (G5 = 7.8% and G6 = 8.5%), with the lowest loss of dentin volume compared to other groups (P< 0.05). Purpose: To evaluate the antimicrobial activity and physico-mechanical properties of an irreversible hydrocolloid in which nanostructured silver vanadate decorated with silver nanoparticles (AgVO 3 ) was added at various concentrations (0% - control, 2.5%, 5%, and 10% by weight)., Materials and Methods: The agar diffusion method (n = 10) was used to evaluate the inhibitory effect on the following species: Streptococcus mutans, Staphyloccocus aureus, Pseudomonas aeruginosa, Escherichia coli, and Candida albicans. The gelation time, flow capacity and plastic deformation were verified (n = 10). The data were analyzed using the Kruskal-Wallis test followed by the Dunn post-test, or via one-way ANOVA with multiple comparisons with a Bonferroni adjustment depending on the distribution (α = 0.05)., Results: All percentages of the nanomaterial were able to promote the antimicrobial activity of a hydrocolloid, with the formation of an inhibition zone (p < 0.05). In general, there was a dose-dependent effect on antimicrobial activity: higher concentrations of the nanomaterial promoted greater action except in the cases of P. aeruginosa (p < 0.001; F = 51.74) and S. aureus (p < 0.001), where the highest inhibition was for the 2.5% group. No difference was found in the gelation time when the control was compared with the groups with AgVO 3 (p > 0.05). The difference was between the 5% and 10% groups (p = 0.007), and the latter promoted an increase in time. The flow capacity of the hydrocolloid with 5% of AgVO 3 was significantly lower when compared with the control (p = 0.034). The AgVO 3 influenced the plastic deformation (p < 0.001) in such a way that concentrations of 5% (p = 0.010) and 10% (p < 0.001) promoted an increase in this property when compared with the control., Conclusions: AgVO 3 can be incorporated into an irreversible hydrocolloid as an antimicrobial agent without promoting adverse effects on physical-mechanical properties when used in concentrations of 2.5%., (© 2019 by the American College of Prosthodontists.)

This in vitro study evaluated the influence of the Er,Cr:YSGG laser, associated or not to desensitizing agents, in the prevention of acid erosion in bovine root dentin. Eighty dentin specimens were selected and divided into eight groups (n = 10): G1: negative control; G2: positive control (5% fluoride varnish-FV); G3: Er,Cr:YSGG laser; G4: FV + laser; G5: 3% potassium oxalate; G6: 3% potassium oxalate + laser; G7: biphasic calcium silicate/phosphate gel (gel); G8: gel + laser. Laser parameters: 0.5 W, 6.25 J/cm 2 at 1-mm distance. The erosive drink used was a cola soft-drink (pH = 2.42 at 4 °C), lasting 5 min, twice a day, with 6-h intervals between the challenges, during 14 days. Kolmogorov-Smirnov and Levene's tests were satisfied. The surface roughness data were submitted to ANOVA and Tukey post hoc tests. For the wear profile, Kruskal-Wallis and Dunn post hoc tests were used. Afterwards, the Spearman correlation test was performed. All statistical tests assumed a significance level of 5% (α = 0.05). G1 presented the highest surface roughness value after the erosive challenge (3.586 μm 2  ± 0.205 μm 2 ) and the G7 presented the lowest surface roughness value after the erosive challenge (1.071 μm 2  ± 0.180 μm 2 ). For the lost volume, G4 presented the lowest percentage (9.7% ± 0.9%), while G1 had the highest percentage (41.8% ± 2.5%), both with p < 0.05. There was a weak correlation between the response variables (ρ = 0.33). All groups presented lower values of surface roughness and loss of volume when compared to the negative control group. For the surface roughness, the biphasic calcium silicate/phosphate gel presented the best result. For volume loss, the 5% fluoride varnish + Er,Cr:YSGG laser showed the best results compared to the other groups.

The purpose of thisin vitrostudy was to evaluate the influence of a new proposal of implant design and surgical protocol on primary stability in different bone densities. Four groups were tested (n=9): G1 - tapered, cone morse, Ø 4.3 mm x 10 mm in length (Alvim CM); G2 - experimental tapered; G3 - cylindrical, cone morse, Ø 4.0 mm x 11 mm in length (Titamax CM) and G4 - experimental cylindrical. The experimental implants were obtained from a design change in the respective commercial models. The insertion was performed in polyurethane (PU) blocks 0.24 g/cm3(20 pcf) and 0.64 g/cm3(40 pcf), according to different surgical protocols. The primary stability was measured by means of insertion torque (IT) and pullout test. Data were analyzed by ANOVA, Tukey's test (α=0.05) and Pearson's correlation. For IT and pullout, conventional and experimental implants showed no difference between them when inserted in the 20 pcf PU (p>0.05). In the 40 pcf PU, the modified implants exhibited greater IT (p<0.05) and lower pullout (p<0.05) compared to the respective conventional models. The implant design tested associated with the surgical protocol, positively influenced primary stability in higher density bones.

Objectives: This study characterized the microbial diversity of formed biofilm on the surface of acrylic resins modified with nanostructured silver vanadate decorated with silver nanoparticles (AgVO 3 ) after incubation in human saliva., Design: Resin specimens prepared with AgVO 3 at concentrations 0%, 1%, 2.5%, and 5% by either vacuum mixing or polymer solubilization were characterized by scanning electron microscopy (SEM) and X-ray diffraction (XRD). After 24 h and 7 days of saliva incubation, biofilm samples were collected from the surface of the specimens. The 16S rDNA genes were amplified, sequenced with the 454-Roche next-generation sequencing platform and analyzed to identify the Operational Taxonomic Units at the genus or higher level., Results: Significant differences in the dispersion pattern of the nanoparticles were observed among the two different methods of AgVO 3 incorporation. In the microbiological analysis, a total of 103 genera and 7 more inclusive taxa, representing the phyla Bacteroidetes, Firmicutes and Proteobacteria were identified colonizing resin surfaces. The incorporation method of the AgVO 3 had little to no significant effect on the microbiota of samples. Significant time and concentration-dependent responses to AgVO 3 caused changes in the taxonomic profile at the phylum and genus level., Conclusions: The results show differences in relation to the microbial diversity of modified resins during the initial phase of biofilm maturation. This study analyzed nanostructured silver vanadate (AgVO3) films on certain substrate surfaces found in dental implant devices in order to see if these films promote antimicrobial activity. Enamel and glaze vehicles were used to apply the nanomaterial (0.0% [control], 2.5%, 5.0%, and 10.0% concentrations) on 3 substrates (280 specimens per substrate): polytetrafluoroethylene (PTFE), polyacetal, and acrylic resin. An agar diffusion method was performed in triplicate to determine the antimicrobial activity against 8 microorganisms. Physical behavior and morphologic properties were also analyzed. The growth of Pseudomonas aeruginosa, Escherichia coli, Streptococcus mutans, Enterococcus faecalis, and Staphylococcus aureus was inhibited with the application of AgVO3. For Candida albicans, Porphyromonas gingivalis, and Prevotella intermedia, no antimicrobial activity was observed. Except for S mutans, the type of substrate did not influence the microbiologic results. The glaze showed greater antimicrobial efficacy against S aureus, E faecalis, and S mutans than did the enamel vehicle (P < 0.05). Films with 10% AgVO3 showed greater antimicrobial activity (P < 0.05). The choice of substrate or vehicle did not influence the surface roughness (P > 0.05); however, the application of 10% AgVO3 caused greater surface change (P < 0.05). The application of the nanomaterial did not influence the surface hardness of substrates when compared with the control group (P > 0.05). The use of the glaze as a vehicle provided higher hardness values (P < 0.05).

Objectives: The objective of the study is to incorporate the nanostructured silver vanadate (AgVO 3 ) decorated with silver nanoparticles at endodontic sealers AH Plus, Sealapex, Sealer 26, and Endofill, at concentrations of 2.5%, 5%, and 10%, and to evaluate physical-chemical properties., Materials and Methods: The study was to evaluate the radiopacity using digital radiography ( n = 5), the tooth color change in 48 incisors ( n = 3) for 7, 30, 90, and 180 days in a spectrophotometer, and the topographic distribution in a confocal laser ( n = 5). The radiopacity was analyzed by Kruskal-Wallis test and the permutation (α = 0.05) and the color and topographical distribution by descriptive statistical analysis., Results: The AgVO 3 had no effect on the radiopacity of Endofill and Sealapex ( P > 0.05) and at 2.5% concentrations increased the radiopacity of Sealer 26 and AH Plus ( P < 0.05). The Endofill 10% showed less color change, and the major changes were the modified groups in 180 days. The AgVO 3 showed a circular topographic distribution in areas of the sealers., Conclusion: It was found that the addition of AgVO 3 did not affect the radiopacity of Endofill and Sealapex however, increased the radipacity of Sealer 26 and AH Plus. For modified groups, the greatest color change was promoted after 180 days, except for Endofill with 10%. Topographic distribution of nanomaterial affected the color change of theevaluated sealers., Competing Interests: There are no conflicts of interest.

Objectives: This study evaluated the release of ions and the cytotoxicity of acrylic resins incorporated with silver vanadate decorated with silver nanoparticles (AgVO 3 )., Background: The inhibition of the accumulation of microorganisms on the resins is critical in preventing diseases. However, the hypothesis is that the release of ions from the incorporation of AgVO 3 may be important in biocompatibility., Materials and Methods: Specimens of autopolymerising (AP) and heat-polymerising resin (HP) with AgVO 3 were prepared and immersed in culture medium. The release of silver ions (Ag) and vanadium (V) was evaluated by mass spectrometry with inductively coupled plasma (ICP-MS) (n=9) and the cell viability of fibroblasts L929 by MTT (3-[4,5-dimethylthiazol- 2yl]-2,5-diphenyltetrazolium bromide) (n=12). The results were evaluated with analysis of variance (ANOVA), Tukey and Pearson correlation test (α=.05)., Results: The groups containing AgVO 3 presented a difference in relation to the control (0%) regarding the release of Ag and V (P<.0001). All groups showed a reduction in L929 viability when compared with the cellular control (100%) (P<.0001). In comparison with the control resins for HP, a reduction in the metabolism of cells occurred starting at 2.5% and for AP at 5% (P<.0001). A positive correlation was found between the concentration of AgVO 3 and the ion release, and a negative between the ion release and the cell viability., Conclusions: Significant numbers of Ag and V ions were released from resins with higher concentrations of AgVO 3 , presenting cytotoxicity for cells, suggesting that the use of low concentrations is indicated to avoid risks to patients., (© 2017 John Wiley & Sons A/S and The Gerodontology Association. The purpose of this study was to compare, through photoelastic analysis, the stress distribution around conventional and modified external hexagon (EH) and morse taper (MT) dental implant connections. Four photoelastic models were prepared (n=1): Model 1 - conventional EH cylindrical implant (Ø 4.0mm×11mm - Neodent®), Model 2 - modified EH cylindrical implant, Model 3 - conventional MT Conical implant (Ø 4.3mm×10mm - Neodent®) and Model 4 - modified MT conical implant. 100 and 150N axial and oblique loads (30° tilt) were applied in the devices coupled to the implants. A plane transmission polariscope was used in the analysis of fringes and each position of interest was recorded by a digital camera. The Tardy method was used to quantify the fringe order (n), that calculates the maximum shear stress (τ) value in each selected point. The results showed lower stress concentration in the modified cylindrical implant (EH) compared to the conventional model, with application of 150N axial and 100N oblique loads. Lower stress was observed for the modified conical (MT) implant with the application of 100 and 150N oblique loads, which was not observed for the conventional implant model. The comparative analysis of the models showed that the new design proposal generates good stress distribution, especially in the cervical third, suggesting the preservation of bone tissue in the bone crest region., (Copyright © 2017 Elsevier B.V. Objective: This preliminary study aimed to evaluate the antimicrobial activity, flow and radiopacity of endodontic sealers with nanostructured silver vanadate decorated with silver nanoparticles (AgVO3)., Methods: The minimum inhibitory concentration (MIC) of AgVO3 was evaluated against Enterococcus faecalis, Pseudomonas aeruginosa and Escherichia coli . Specimens were prepared from the following endodontic sealers: AH Plus (DENTSPLY DeTrey GmbH, Konstanz, Germany), Sealapex (Sybron Endo, Orange, CA, USA), Sealer 26 (DENTSPLY, Petrópolis, Brazil) and Endofill (DENTSPLY, Petrópolis, Brazil), with concentrations of 0%, 2.5%, 5% and 10% of AgVO3. Agar diffusion was used to evaluate the materials after 48 hours and 7 days (n=6). Flow (n=6) and radiopacity (n=9) were evaluated. The data were analysed by analysis of variance (ANOVA) and the Tukey honestly significant difference (HSD) (α=0.05)., Results: The MIC of AgVO3 was 500 μg/mL for E. faecalis and 31.25 μg/mL for P. aeruginosa and E. coli . The AgVO3 did not influence the antimicrobial activity of AH Plus against E. faecalis (P>0.05) but did promote this activity for Sealapex (P<0.01). Moreover, this activity increased for Endofill from 2.5% and for Sealer 26 from 5% (P<0.05). Against P. aeruginosa , only AH Plus and Endofill 10% inhibited zone formation (P<0.01). The antimicrobial activity of Endofill increased from 2.5% against E. coli (P<0.01). Sealapex 5% and 10% (P<0.01), Sealer 26 10% and AH Plus promoted antimicrobial activity against E. coli. An increase in the zone of inhibition occurred between 48 hours and 7 days in the Sealapex 10% and Endofill 5% groups against E. coli. The flow of AH Plus and Endofill decreased with the increase of AgVO3 (P<0.05), and the flow of Sealer 26 and Sealapex was not affected (P>0.05). The radiopacity of AH Plus increased with AgVO3 (P<0.05). Endofill 5% and 10% did not differ from the control Endofill (P>0.05). The incorporation of AgVO3 did not influence the radiopacity of Sealer 26 (P>0.05). Statement of Problem: The marginal adaptation of prosthetic crowns is still a significant clinical problem., Purpose: The purpose of this in vitro study was to evaluate the marginal deficiency and misfit of Ni-Cr alloys with and without beryllium under different casting conditions., Material and Methods: Four casting conditions were selected: flame-torch, induction/argon, induction/vacuum, and induction/air; and 2 alloys were used, Ni-Cr-Be and Ni-Cr. For each group, 10 metal specimens were prepared. Silicone indirect impressions and analysis of the degree of rounding were used to evaluate the marginal deficiencies of metal copings, and a standardized device for the setting pressure associated with optical microscopy was used to analyze the marginal misfit. Results were evaluated with 2-way ANOVA (α=.05), followed by the Tukey honest significant difference post hoc test, and the Pearson correlation test (α=.05)., Results: Alloy (P<.001) and casting technique (P<.001) were shown to affect marginal deficiencies. The Ni-Cr cast using the torch technique showed the highest marginal deficiency, and the Ni-Cr-Be cast in a controlled argon atmosphere showed the lowest (P<.001). Alloy (P=.472) and casting techniques (P=.206) did not affect the marginal misfit, but significant differences were found in the interaction (P=.001); the lowest misfit was achieved using the Ni-Cr-Be, and the highest misfit occurred with the molten Ni-Cr, using the cast torch technique. No correlation was found between deficiency and marginal misfit (r=.04, P=.69)., Conclusions: The interactions demonstrated that the alloy containing beryllium that was cast in an argon atmosphere led to reduced marginal deficiency. Objective: The purpose of this study was evaluate, for the first time, the impact of incorporation of nanostructured silver vanadate (β-AgVO3) in antibiofilm and mechanical properties of dental acrylic resins (poly(methyl methacrylate), PMMA)., Design: The β-AgVO3 was synthesized and characterized by X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy, and microanalysis (SEM/EDS). Resins specimens were prepared with 0-10% wt.% β-AgVO3 and characterized by SEM, XRD and optical microscopy. The antibiofim activity of the samples against Candida albicans and Streptococcus mutans was investigated by XTT reduction test, colony-forming units (CFUs), and confocal laser scanning microscopy (CLSM). The flexural strength, hardness, and surface roughness of the samples containing β-AgVO3 were compared with the pure PMMA matrix., Results: The incorporation of 10% β-AgVO3 significantly reduced the metabolic activity of C. albicans and S. mutans (p<0.05). There was a reduction in microbial load (CFU/mL) of microorganisms for the different concentrations used (p<0.05), which was confirmed by confocal microscopy. The addition of β-AgVO3 did not change the mechanical properties of hardness and surface roughness of the resins (p>0.05). However, flexural strength decreased with the addition of amounts greater than 1% (p<0.05)., Conclusions: β-AgVO3 additions in dental acrylic resin may have an impact on inhibition of biofilm of main microorganisms associated with dental prostheses. The aim of this study was to compare the compressive strength of a silorane-based composite resin (Filtek P90) to that of conventional composite resins (Charisma, Filtek Z250, Fill Magic, and NT Premium) before and after accelerated artificial aging (AAA). For each composite resin, 16 cylindrical specimens were prepared and divided into 2 groups. One group underwent analysis of compressive strength in a universal testing machine 24 hours after preparation, and the other was subjected first to 192 hours of AAA and then the compressive strength test. Data were analyzed by analysis of variance, followed by the Tukey HSD post hoc test (α = 0.05). Some statistically significant differences in compressive strength were found among the commercial brands (P < 0.001). The conventional composite resin Fill Magic presented the best performance before (P < 0.05) and after AAA (P < 0.05). Values for compressive strength of the silorane-based composite were among the lowest obtained, both before and after aging. Comparison of each material before and after AAA revealed that the aging process did not influence the compressive strength of the tested resins (P = 0.785).

Background: The macrogeometry of dental implants strongly influences the primary stability and hence the osseointegration process., Purpose: Compare the performance of conventional and modified implant models in terms of primary stability., Materials and Methods: A total of 36 implants (Neodent®) with two different formats (n = 18): Alvim CM (Conical CM, Ø 4.3 mm × 10 mm in length) and Titamax Ti (Cylindrical HE, Ø 4.0 mm × 11 mm in length) were inserted into artificial bone blocks. Nine implants from each set were selected to undergo external geometry changes. The primary stability was quantified by insertion torque and resonance frequency using an Osstell device and the pullout test. One-way analysis of variance and Tukey's test were used for statistical evaluation., Results: The comparative analysis of the implants showed a significant increase of the insertion torque for the modified Conical CM implants (p = 0.000) and Cylindrical HE (p = 0.043); for the resonance frequency the modified Cylindrical HE showed a lower statistical mean (p = 0.002) when compared to the conventional model, and in the pullout test both modified implants showed significant reduction (p = 0.000)., Conclusions: Within the limitations of this study, the proposed modification showed good stability levels and advantages when compared to the conventional implants., (© 2015 Wiley Periodicals, Inc.)

Statement of Problem: The accumulation of bacteria on the surface of dental prostheses can lead to systemic disease., Purpose: The purpose of this in vitro study was to evaluate the growth of Staphylococcus aureus and Pseudomonas aeruginosa on the surface of autopolymerizing (AP) and heat-polymerizing (HP) acrylic resins incorporated with nanostructured silver vanadate (β-AgVO3) and its impact strength., Material and Methods: For each resin, 216 circular specimens (9 × 2 mm) were prepared for microbiologic analysis and 60 rectangular specimens (65 × 10 × 3.3 mm) for mechanical analysis, according to the percentage of β-AgVO3: 0%, control group; 0.5%; 1%; 2.5%; 5%; and 10%. After a biofilm had formed, the metabolic activity of the bacteria was measured using the XTT reduction assay (2,3-bis-(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide) (n=8), and the number of viable cells was determined by counting colony forming units per milliliter (CFU/mL) (n=8). Confocal laser scanning microscopy (CLSM) was used to complement the analyses (n=2). The mechanical behavior was evaluated by impact strength assays (n=10). Data were analyzed by 2-way ANOVA, followed by the Tukey honestly significant difference (HSD) post hoc test (α=.05)., Results: The addition of 5% and 10% β-AgVO3 significantly decreased the metabolic activity of P. aeruginosa for both resins (P<.05). The HP resin promoted a greater reduction in metabolic activity than the AP resin (P<.05). No difference was found in the metabolic activity of S. aureus according to the XTT (P>.05). The number of CFU/mL for S. aureus and P. aeruginosa decreased significantly when 5% and 10% β-AgVO3 were added (P<.001). These concentrations significantly reduced the impact strength of the resins (P<.001) because the system was weakened by the presence of clusters of β-AgVO3., Conclusion: The addition of β-AgVO3 can provide acrylic resins with antibacterial activity but reduces their impact strength. Natural products can be a source of biomolecules with antioxidant activity which are able to prevent oxidative stress-induced diseases and show antitumor activity, making them important sources of new anticancer drug prototypes. In this context, this study aimed to analyze the chemical composition of an ethanol extract of Senna velutina leaves and to assess its antioxidant and cytotoxic activities in leukemic cells. The antioxidant properties were evaluated using a DPPH free radical scavenging assay and by examining the extract's inhibition of AAPH-induced lipid peroxidation in human erythrocytes. Its cytotoxicity and possible mechanisms of action were assessed in Jurkat and K562 leukemic cell lines. The ethanol extract contained flavonoids, such as epigallocatechin, epicatechin, kaempferol heteroside, rutin, and dimeric and trimeric proanthocyanidin derivatives. The extract exhibited antioxidant activity by scavenging free radicals and antihemolytic action, and it decreased malondialdehyde content in human erythrocytes. Furthermore, the extract also induced leukemic cell death by activating intracellular calcium and caspase-3, decreasing mitochondrial membrane potential, and arresting the cell cycle in S and G2 phases. Hence, S. velutina leaf extract contains antioxidant and antileukemic biomolecules with potential applications in diseases associated with oxidative stress and in the inhibition of tumor cell proliferation.

Objectives: The purpose of this study was to analyze the influence of the shape of various implants and the density of substrate on primary stability using a combination of methods., Materials and Methods: Fifty-four Neodent® brand cylindrical and conical implants with different prosthetic platforms were used. Implants were inserted into a pork rib bone and polyurethane blocks. Primary stability was assessed by insertion torque (IT), resonance frequency analysis (RFA), and pullout strength. Screws were also analyzed by scanning electron microscopy (SEM) before insertion and after removal to justify their use for inserting in different substrates., Results: The conical cone morse implant had the highest average for all of the assays performed and was significantly different (p < 0.05) from the cylindrical implants for IT in the bone, pullout strength in the 40 per cubic foot (PCF) polyurethane, and the bone. The internal hex cylindrical implant had the lowest averages, which were significantly different (p < 0.05) from the conical implants for IT and RFA in the bone, pullout strength in the 40 PCF polyurethane, and the bone. The IT, RFA, and pullout strength assays were moderately correlated, and the photomicrographs did not reveal changes in the implants., Conclusions: The analysis of different implants showed a better primary stability of tapered implants; the density of the substrate influences the primary stability and the 15 PCF polyurethane was not adequate to evaluate primary stability; correlation was obtained between the different methodologies of analysis of primary stability., Clinical Relevance: The study shows the influence of different implant macro-geometries and densities of substrates on primary stability.

Unlabelled: The adhesion of biofilm on dental prostheses is a prerequisite for the occurrence of oral diseases., Objective: To assess the antimicrobial activity and the mechanical properties of an acrylic resin embedded with nanostructured silver vanadate (β-AgVO3)., Material and Methods: The minimum inhibitory concentration (MIC) of β-AgVO3 was studied in relation to the species Staphylococcus aureus ATCC 25923, Streptococcus mutans ATCC 25175, Pseudomonas aeruginosa ATCC 27853, and Candida albicans ATCC 10231. The halo zone of inhibition method was performed in triplicate to determine the inhibitory effect of the modified self-curing acrylic resin Dencor Lay-Clássico. The surface hardness and compressive strength were examined. The specimens were prepared according to the percentage of β-AgVO3 (0%-control, 0.5%, 1%, 2.5%, 5%, and 10%), with a sample size of 9x2 mm for surface hardness and antimicrobial activity tests, and 8x4 mm for the compression test. The values of the microbiologic analysis were compared and evaluated using the Kruskal-Wallis test (α=0.05); the mechanical analysis used the Shapiro-Wilk's tests, Levene's test, ANOVA (one-way), and Tukey's test (α=0.05)., Results: The addition of 10% β-AgVO3 promoted antimicrobial activity against all strains. The antimicrobial effect was observed at a minimum concentration of 1% for P. aeruginosa, 2.5% for S. aureus, 5% for C. albicans, and 10% for S. mutans. Surface hardness and compressive strength increased significantly with the addition of 0.5% β-AgVO3 (p<0.05). Higher rates of the nanomaterial did not alter the mechanical properties of the resin in comparison with the control group (p>0.05)., Conclusions: The incorporation of β-AgVO3 has the potential to promote antimicrobial activity in the acrylic resin. At reduced rates, it improves the mechanical properties, and, at higher rates, it does not promote changes in the control.

The aim of this study was to investigate the influence of accelerated artificial aging (AAA) on the microstructure and mechanical properties of the Filtek Z250, Filtek Supreme, 4 Seasons, Herculite, P60, Tetric Ceram, Charisma and Filtek Z100. composite resins. The composites were characterized by Fourier-transform Infrared spectroscopy (FTIR) and thermal analyses (Differential Scanning Calorimetry - DSC and Thermogravimetry - TG). The microstructure of the materials was examined by scanning electron microscopy. Surface hardness and compressive strength data of the resins were recorded and the mean values were analyzed statistically by ANOVA and Tukey's test (α=0.05). The results showed significant differences among the commercial brands for surface hardness (F=86.74, p<0.0001) and compressive strength (F=40.31, p<0.0001), but AAA did not affect the properties (surface hardness: F=0.39, p=0.53; compressive strength: F=2.82, p=0.09) of any of the composite resins. FTIR, DSC and TG analyses showed that resin polymerization was complete, and there were no differences between the spectra and thermal curve profiles of the materials obtained before and after AAA. TG confirmed the absence of volatile compounds and evidenced good thermal stability up to 200 °C, and similar amounts of residues were found in all resins evaluated before and after AAA. The AAA treatment did not significantly affect resin surface. Therefore, regardless of the resin brand, AAA did not influence the microstructure or the mechanical properties.

Global DNA methylation of peripheral blood leukocytes has been recently proposed as a potential biomarker for disease risk. However, the amplitude of the changes in DNA methylation associated with normal aging and the impacts of environmental changes on this variation are still unclear. In this context, we evaluated the association of global DNA methylation with nutritional habits, tobacco smoking, body mass index (BMI), clinical laboratory parameters, polymorphism C677T MTHFR, functional cognition and the daily practice of physical activity in a cancer-free older population. Leukocyte global DNA methylation from 126 older individuals was quantified using a high-throughput ELISA-based method. Global DNA hypomethylation was observed in older individuals when compared to a younger population (p = 0.0469), confirming changes in DNA methylation in the aging process. Furthermore, the methylation profile of elders was correlated with the daily ingestion of carbohydrates (p = 0.0494), lipids (p = 0.0494), vitamin B6 (p = 0.0421), magnesium (p = 0.0302), and also to the serum levels of total protein (p = 0.0004), alpha 2 globulin (p = 0.0013) and albumin (p = 0.0015). No statistically significant difference was observed when global DNA methylation were stratified according to C677T MTHFR genotypes (p = 0.7200), BMI (p = 0.1170), smoking habit (p = 0.4382), physical activity in daily life (p = 0.8492), scored cognitive function (p = 0.7229) or depression state (p = 0.8301). Our data indicate that age-related variations in the global DNA methylation profile of leukocytes might be modulated by the daily intake of carbohydrates, lipids, vitamin B6, and magnesium and be associated with serum protein levels, however it is independent of C677T MTHFR genotype and not correlated with BMI, smoking habit, cognitive function or the routine physical activities.
