Abstract

11[beta]-Hydroxysteroid dehydrogenase-1 (11 [beta]-HSD-1) plays a key role in the regulation of intracellular glucocorticoid concentrations. Increased message and/or activity of adipose 11[beta]-HSD-1 are characteristics of human and animal models of obesity. Hexose-6-phosphate dehydrogenase (H6PDH) is colocalized with 11 [beta]-HSD-1 and may be a critical factor in determining the oxo-reductase activity of 11 [beta]-HSD-1. This study examined the effects of sucrose solution access on body weight, body composition, and message of 11 [beta]-HSD-1 and H6PDH in mesenteric adipose and liver. Rats were assigned to 3 groups: 1) control (ad libitum intake of nonpurified diet and water only); 2) ad libitum intake of 16% sucrose solution ($16); or 3) ad libitum intake of 32% sucrose solution (S32) in addition to ad libitum intake of diet and water. The S32 group consumed more energy daily than the S16 and control groups, yet body weight did not differ among groups. Percentages of body fat did not differ between the S16 and S32 groups but were higher than in controls. Hepatic 11[beta] -HSD-1 message was suppressed by 46% in the S16 group and by 47% in the S32 group, whereas the H6PDH message nearly doubled in the S16 group compared to the control group. In mesenteric fat, 11[beta]-HSD-1 message increased 23-fold in the S16 group and 32-fold in the S32 group and the H6PDH message increased 3.5-fold in the S16 group compared to the control group. These data demonstrate that sucrose can promote increased 11[beta]-HSD-1 and H6PDH message in mesenteric fat while concomitantly decreasing 11[beta]-HSD-1 message and increasing H6PDH message in liver. Abstract

Adrenalectomy (ADX) has greater effects on weight gain and body fat accumulation in rats than does RU-486. The major role of ADX on body weight, food intake, or fat accumulation may be due to the lack of aldosterone and catecholamines. Furthermore, the dietary obesity model is sensitive enough to show the differential metabolic effects of diet composition and the differential effects of ADX and the blockade of glucocorticoid receptors by RU-486.

Abstract

Experiments for measuring alterations in energy expenditure (EE) and respiratory quotient (RQ) of corticosterone (cort) dose-treated adrenalectomy (ADX) rats to characterize their corticosterone-resistant and corticosterone-sensitive metabolic properties reveal that cort doses eliminate the inhibition of food intake and body weight by ADX and reinstate ADX-induced decrease in motoric activity to sham levels in rats. EE suppression is seen in both light and dark conditions, but RQ suppression is seen only in dark conditions. Cort replacement partially revives EE to sham levels in light conditions.

Physiol. 46): R1144-R1151, 1999.--Adult Otsuka Long-Evans Tokushima fatty (OLETF) rats lack functional cholecystokinin A (CCK-A) receptors, are diabetic, hyperphagic, and obese, and have patterns of ingestion consistent with a satiety deficit secondary to CCK insensitivity. Because dietary fat potently stimulates CCK release, we examined how dietary fat modulates feeding in adult male OLETF rats and their lean [Long-Evans Tokushima (LETO)] controls. High-fat feeding produced sustained overconsumption of high-fat diet (30% corn oil in powdered chow) over a 3-wk period in OLETF but not LETO rats. We then assessed the ability of gastric gavage (5 ml, 1-2 kcal/ml x 15 s) or duodenal preloads (1 kcal/ml, 0.44 ml/min x 10 min) of liquid carbohydrate (glucose), protein (peptone), or fat (Intralipid) to suppress subsequent 30-min 12.5% glucose intake in both strains. In OLETF rats, gastric and duodenal fat preloads were significantly less effective in suppressing subsequent intake than were equicaloric peptone or glucose. Abstract

Energy balance is achieved as the result of both central and peripheral regulators working together. Sugar intake can cause a disruption in energy balance that can increase the risk of the onset of obesity. In this review, we will discuss the relationship between sugar consumption and obesity. We have summarized the current evidence that shows how sugars, typically sugar-sweetened solutions, affect the control of energy intake. Solution consumption affects both peripheral (triglycerides, glucocorticoids, and adiposity hormone) and central (typically hypothalamus) mechanisms. We have concluded this review with an attempt to explain how sugar consumption induces changes in body weight that lead to obesity and/or associated metabolic diseases. [ABSTRACT FROM AUTHOR]

49. Dietary Self-Selection and the Zucker Rat

Abstract

Impaired protein deposition has been suggested to be a critical factor promoting the hyperphagia of the obese Zucker rat. The selection patterns of adult, male, genetically obese (fa/fa) and lean (Fa/-) Zucker rats were studied to determine if obese rats would select a diet that was higher in protein than the diet selected by lean littermates. Rats were provided ad libitum access to three macronutrient sources and were allowed to compose their own diets for 9 days. The three dietary items were: a vitamin + mineral-supplemented carbohydrate source (cornstarch), a vitamin + mineral-supplemented protein source (casein) and commercially available corn oil. Obese rats ate 43% more calories than lean littermates. Further, obese rats selected a diet that provided 12% of their total caloric intake as protein, 24% as carbohydrate and 64% as fat. Lean rats selected a diet that provided 33% of their total caloric intake as protein, 37% as carbohydrate and 30% as fat. These selection data are not consistent with the hypothesized importance of the role of dietary protein and its incorporation into lean body mass as a stimulus promoting the hyperphagia demonstrated by the genetically obese Zucker rat.
