This paper deals with the results of an extensive sampling of eight former channels of the Rhǒne and eight of its tributary, the Ain River, France, designed to assess the value of molluscs as 'describers'︁ of ecosystem dynamics. The sampling was stratified on the distribution of aquatic macrophytes. The distribution of molluscs species is shown to provide information on the rate of eutrophication, the groundwater supply and the succession of the abandonned river channels toward semi-aquatic and terrestrial conditions. Molluscs, together with other macroinvertebrates, not only describe the present stage of these ecosystems but also provide some predictive information on their successional pattern for assistance in the management of the alluvial plain. Abstract Background There is evidence that exposure to passive smoking in general, and in babies in particular, is an important cause of morbimortality. Passive smoking is related to an increased risk of pediatric diseases such as sudden death syndrome, acute respiratory diseases, worsening of asthma, acute-chronic middle ear disease and slowing of lung growth. The objective of this article is to describe the BIBE study protocol. The BIBE study aims to determine the effectiveness of a brief intervention within the context of Primary Care, directed to mothers and fathers that smoke, in order to reduce the exposure of babies to passive smoking (ETS). Methods/Design Cluster randomized field trial (control and intervention group), multicentric and open. Subject: Fathers and/or mothers who are smokers and their babies (under 18 months) that attend pediatric services in Primary Care in Catalonia. The measurements will be taken at three points in time, in each of the fathers and/or mothers who respond to a questionnaire regarding their baby's clinical background and characteristics of the baby's exposure, together with variables related to the parents' tobacco consumption. A hair sample of the baby will be taken at the beginning of the study and at six months after the initial visit (biological determination of nicotine). The intervention group will apply a brief intervention in passive smoking after specific training and the control group will apply the habitual care. Discussion Exposure to ETS is an avoidable factor related to infant morbimortality. Interventions to reduce exposure to ETS in babies are potentially beneficial for their health. The BIBE study evaluates an intervention to reduce exposure to ETS that takes advantage of pediatric visits. Interventions in the form of advice, conducted by pediatric professionals, are an excellent opportunity for prevention and protection of infants against the harmful effects of ETS. Group VII ethylene response factors (ERF-VII) are plant-specific transcription factors (TFs) known for their role in the activation of hypoxia-responsive genes under low oxygen stress but also in plant endogenous hypoxic niches. However, their function in the microaerophilic nitrogen-fixing nodules of legumes has not yet been investigated. We investigated regulation and the function of the two Medicago truncatula ERF-VII TFs (MtERF74 and MtERF75) in roots and nodules, MtERF74 and MtERF75 in response to hypoxia stress and during the nodulation process using an RNA interference strategy and targeted proteolysis of MtERF75. Knockdown of MtERF74 and MtERF75 partially blocked the induction of hypoxia-responsive genes in roots exposed to hypoxia stress. In addition, a significant reduction in nodulation capacity and nitrogen fixation activity was observed in mature nodules of double knockdown transgenic roots. Overall, the results indicate that MtERF74 and MtERF75 are involved in the induction of MtNR1 and Pgb1.1 expression for efficient Phytogb-nitric oxide respiration in the nodule., (© 2022 John Wiley & Sons Ltd.)

Bacillus thuringiensis (Bt) produces pore forming toxins that have been used for pest control in agriculture for many years. However, their molecular and cellular mode of action is still unclear. While a first model - referred to as the pore forming model - is the most widely accepted scenario, a second model proposed that toxins could trigger an Mg 2+ -dependent intracellular signalling pathway leading to cell death. Although Cry1Ca has been shown to form ionic pores in the plasma membrane leading to cell swelling and death, we investigated the existence of other cellular or molecular events involved in Cry1Ca toxicity. The Sf9 insect cell line, derived from Spodoptera frugiperda , is highly and specifically sensitive to Cry1Ca. Through a selection program we developed various levels of laboratory-evolved Cry1Ca-resistant Sf9 cell lines. Using a specific S. frugiperda microarray we performed a comparative transcriptomic analysis between sensitive and resistant cells and revealed genes differentially expressed in resistant cells and related to cation-dependent signalling pathways. Ion chelators protected sensitive cells from Cry1Ca toxicity suggesting the necessity of both Ca 2+ and/or Mg 2+ for toxin action. Selected cells were highly resistant to Cry1Ca while toxin binding onto their plasma membrane was not affected. This suggested a resistance mechanism different from the classical 'loss of toxin binding'. We observed a correlation between Cry1Ca cytotoxicity and the increase of intracellular cAMP levels. Indeed, Sf9 sensitive cells produced high levels of cAMP upon toxin stimulation, while Sf9 resistant cells were unable to increase their intracellular cAMP. Objectives: To assess the feasibility and efficacy of intensity-modulated radiation implemented with helical tomotherapy image-guided with daily megavoltage computed tomography for head and neck cancer., Methods: Between May 2010 and May 2013, 72 patients were treated with curative intent. The median age was 64 years, with 57% undergoing definitive and 43% postoperative radiotherapy. Primary tumour sites were oral cavity (21%), oropharynx (26%), hypopharynx (20%), larynx (22%), and others (11%). Staging included 4% stage I, 15% II, 26% III, 48% IVa, and 7% IVb. Radiotherapy was combined with chemotherapy in 64%. Primary endpoint was locoregional control, and secondary endpoints survival and toxicity., Results: Median follow-up was 20 months, with 11 locoregional recurrences. Three-year disease-free survival was 58% and overall survival 57%. In the multivariate analysis, age under 64 years, no extracapsular extension, postoperative radiotherapy, induction chemotherapy, and non-oral cavity tumour were significant favourable prognostic factors for disease-free-survival. The overall incidence of acute grade ≥3 toxicities were mucositis 32%, pain 11%, xerostomia 7%, dysphagia 53%, radiodermatitis 44%, and osteonecrosis 1%. Late grade ≥3 toxicities were fibrosis 6%, dysphagia 21%, fistula 1%, and skin necrosis 1%., Conclusions: Intensity-modulated radiation with helical tomotherapy achieved respectable locoregional control and overall survival, with acceptable toxicity, in head and neck cancer patients., (© 2018 S. Plant glutathione peroxidases (Gpx) catalyse the reduction of various peroxides, such as hydrogen peroxide (H 2 O 2 ), phospholipid hydroperoxides and peroxynitrite, but at the expense of thioredoxins rather than glutathione. A main function of plant Gpxs is the protection of biological membranes by scavenging phospholipid hydroperoxides, but some Gpxs have also been associated with H 2 O 2 sensing and redox signal transduction. Nitric oxide (NO) is not only known to induce the expression of Gpx family members, but also to inhibit Gpx activity, presumably through the S-nitrosylation of conserved cysteine residues. In the present study, the effects of NO-donors on both the activity and S-nitrosylation state of purified Medicago truncatula Gpx1 were analyzed using biochemical assay measurements and a biotin-switch/mass spectrometry approach. MtGpx1 activity was only moderately inhibited by the NO-donors diethylamine-NONOate and S-nitrosoglutathione, and the inhibition may be reversed by DTT. The three conserved Cys of MtGpx1 were found to be modified through S-nitrosylation and S-glutathionylation, although to different extents, by diethylamine-NONOate and S-nitrosoglutathione, or by a combination of diethylamine-NONOate and reduced glutathione. Legumes associate with rhizobia to form nitrogen (N 2 )-fixing nodules, which is important for plant fitness [1, 2]. Medicago truncatula controls the terminal differentiation of Sinorhizobium meliloti into N 2 -fixing bacteroids by producing defensin-like nodule-specific cysteine-rich peptides (NCRs) [3, 4]. The redox state of NCRs influences some biological activities in free-living bacteria, but the relevance of redox regulation of NCRs in planta is unknown [5, 6], although redox regulation plays a crucial role in symbiotic nitrogen fixation [7, 8]. Two thioredoxins (Trx), Trx s1 and s2, define a new type of Trx and are expressed principally in nodules [9]. Here, we show that there are four Trx s genes, two of which, Trx s1 and s3, are induced in the nodule infection zone where bacterial differentiation occurs. Trx s1 is targeted to the symbiosomes, the N 2 -fixing organelles. Trx s1 interacted with NCR247 and NCR335 and increased the cytotoxic effect of NCR335 in S. meliloti. We show that Trx s silencing impairs bacteroid growth and endoreduplication, two features of terminal bacteroid differentiation, and that the ectopic expression of Trx s1 in S. meliloti partially complements the silencing phenotype. Thus, our findings show that Trx s1 is targeted to the bacterial endosymbiont, where it controls NCR activity and bacteroid terminal differentiation. Similarly, Trxs are critical for the activation of defensins produced against infectious microbes in mammalian hosts. Anthropomorphic model observers are mathe- matical algorithms which are applied to images with the ultimate goal of predicting human signal detection and classification accuracy across varieties of backgrounds, image acquisitions and display conditions. A limitation of current channelized model observers is their inability to handle irregularly-shaped signals, which are common in clinical images, without a high number of directional channels. Here, we derive a new linear model observer based on convolution channels which we refer to as the "Filtered Channel observer" (FCO), as an extension of the channelized Hotelling observer (CHO) and the nonprewhitening with an eye filter (NPWE) observer. In analogy to the CHO, this linear model observer can take the form of a single template with an external noise term. To compare with human observers, we tested signals with irregular and asymmetrical shapes spanning the size of lesions down to those of microcalfications in 4-AFC breast tomosynthesis detection tasks, with three different contrasts for each case. Whereas humans uniformly outperformed conventional CHOs, the FCO observer outperformed humans for every signal with only one exception. Additive internal noise in the models allowed us to degrade model performance and match human performance. We could not match all the human performances with a model with a single internal noise component for all signal shape, size and contrast conditions. This suggests that either the internal noise might vary across signals or that the model cannot entirely capture the human detection strategy. However, the FCO model offers an efficient way to apprehend human observer performance for a non-symmetric signal.

The specific interaction between legumes and Rhizobium-type bacteria leads to the establishment of a symbiotic relationship characterized by the formation of new differentiated organs named nodules, which provide a niche for bacterial nitrogen (N2) fixation. In the nodules, bacteria differentiate into bacteroids with the ability to fix atmospheric N2 via nitrogenase activity. As nitrogenase is strongly inhibited by oxygen, N2 fixation is made possible by the microaerophilic conditions prevailing in the nodules. Increasing evidence has shown the presence of NO during symbiosis, from early interaction steps between the plant and the bacterial partners to N2-fixing and senescence steps in mature nodules. Both the plant and the bacterial partners participate in NO synthesis. NO was found to be required for the optimal establishment of the symbiotic interaction. Transcriptomic analysis at an early stage of the symbiosis showed that NO is potentially involved in the repression of plant defence reactions, favouring the establishment of the plant-microbe interaction. In mature nodules, NO was shown to inhibit N2 fixation, but it was also demonstrated to have a regulatory role in nitrogen metabolism, to play a beneficial metabolic function for the maintenance of the energy status under hypoxic conditions, and to trigger nodule senescence. The present review provides an overview of NO sources and multifaceted effects from the early steps of the interaction to the senescence of the nodule, and presents several approaches which appear to be particularly promising in deciphering the roles of NO in N2-fixing symbioses., (© The Author 2015. The interaction between legumes and rhizobia leads to the establishment of a symbiotic relationship characterized by the formation of new organs called nodules, in which bacteria have the ability to fix atmospheric nitrogen (N2) via the nitrogenase activity. Significant nitric oxide (NO) production was evidenced in the N2-fixing nodules suggesting that it may impact the symbiotic process. Indeed, NO was shown to be a potent inhibitor of nitrogenase activity and symbiotic N2 fixation. It has also been shown that NO production is increased in hypoxic nodules and this production was supposed to be linked - via a nitrate/NO respiration process - with improved capacity of the nodules to maintain their energy status under hypoxic conditions. Other data suggest that NO might be a developmental signal involved in the induction of nodule senescence. Hence, the questions were raised of the toxic effects versus signaling/metabolic functions of NO, and of the regulation of NO levels compatible with nitrogenase activity. The present review analyses the different roles of NO in functioning nodules, and discusses the role of plant and bacterial (flavo)hemoglobins in the control of NO level in nodules.

Synthetic yet realistic images are valuable for many applications in visual sciences and medical imaging. Typically, investigators develop algorithms and adjust their parameters to generate images that are visually similar to real images. In this study, we used a genetic algorithm and an objective, statistical similarity measure to optimize a particular texture generation algorithm, the clustered lumpy backgrounds (CLB) technique, and synthesize images mimicking real mammograms textures. We combined this approach with psychophysical experiments involving the judgment of radiologists, who were asked to qualify the visual realism of the images. Both objective and psychophysical approaches show that the optimized versions are significantly more realistic than the previous CLB model. Anatomical structures are well reproduced, and arbitrary large databases of mammographic texture with visual and statistical realism can be generated. Potential applications include detection experiments, where large amounts of statistically traceable yet realistic images are needed.

We estimated human observer linear templates underlying the detection of a realistic, spherical mass signal with mammographic backgrounds. Five trained naïve observers participated in two-alternative forced-choice (2-AFC) detection experiments with the signal superimposed on synthetic, clustered lumpy backgrounds (CLBs) in one condition and on nonstationary real mammographic backgrounds in another. Human observer linear templates were estimated using a genetic algorithm. A variety of common model observer templates were computed, and their shapes and associated performances were compared with those of the human observer. The estimated linear templates are not significantly different for stationary CLBs and real mammographic backgrounds. The estimated performance of the linear template compared with that of the human observers is within 5% in terms of percent correct (Pc) for the 2-AFC task. Channelized Hotelling models can fit human performance, but the templates differ considerably from the human linear template. Due to different local statistics, detection efficiency is significantly higher on nonstationary real backgrounds than on globally stationary synthetic CLBs. This finding emphasizes that nonstationary backgrounds need to be described by their local statistics.

Rationale and Objectives: Our project was to investigate a complete methodology for the semiautomatic assessment of digital mammograms according to their density, an indicator known to be correlated to breast cancer risk. The BI-RADS four-grade density scale is usually employed by radiologists for reporting breast density, but it allows for a certain degree of subjective input, and an objective qualification of density has therefore often been reported hard to assess. The goal of this study was to design an objective technique for determining breast BI-RADS density., Materials and Methods: The proposed semiautomatic method makes use of complementary pattern recognition techniques to describe manually selected regions of interest (ROIs) in the breast with 36 statistical features. Three different classifiers based on a linear discriminant analysis or Bayesian theories were designed and tested on a database consisting of 1408 ROIs from 88 patients, using a leave-one-ROI-out technique. Classifications in optimal feature subspaces with lower dimensionality and reduction to a two-class problem were studied as well., Results: Comparison with a reference established by the classifications of three radiologists shows excellent performance of the classifiers, even though extremely dense breasts continue to remain more difficult to classify accurately. For the two best classifiers, the exact agreement percentages are 76% and above, and weighted kappa values are 0.78 and 0.83. Furthermore, classification in lower dimensional spaces and two-class problems give excellent results., Conclusion: The proposed semiautomatic classifiers method provides an objective and reproducible method for characterizing breast density, especially for the two-class case. It represents a simple and valuable tool that could be used in screening programs, training, education, or for optimizing image processing in diagnostic tasks.

Neonatal osteoarticular infections remain rare, with an estimated incidence of 1 to 3 cases per 1000 admissions to Neonatal Intensive Care Units. It usually results from bacteraemia and may thus be induced by IV catheters. More rarely it is due to direct inoculation secondary to cutaneous damage, or extension of soft tissue infection. The particularity of bone vascularization in the newborn explains the frequency of abscess formation in the periosteum or in soft tissues. The main pathogen involved is S. aureus (3/4 of cases), followed by group B streptococci and enterobacteriacae. Infection consists mainly of localised and slowly progressing abscesses. However, multifocal and severe infection is possible, in particular when caused by an IV catheter. Ultrasonography is the best initial investigation, possibly leading to surgical care. Medical treatment must include 2 synergistic antistaphyloccocal antibiotics, possibly associated with cefotaxime. The outcome is generally favorable, but orthopaedic consequences may emerge if the growth plate is involved. Rare specific causes, such as syphilis or tuberculosis, should also be evoked, but the clinical context is generally helpful for the diagnosis.

The spread of diseases transmitted by Anopheles and Culex mosquitoes, such as malaria and West Nile fever, is a growing concern for human health. Bacillus sphaericus binary toxin (Bin) is one of the few available bioinsecticides able to control populations of these mosquitoes efficiently. We previously showed that Bin binds to Cpm1, an alpha-glucosidase located on the apical side of Culex larval midgut epithelium. We analysed the effects of Bin by expressing a construct encoding Cpm1 in the mammalian epithelial MDCK cell line. Cpm1 is targeted to the apical side of polarized MDCK, where it is anchored by glycosylphosphatidylinositol (GPI) and displays alpha-glucosidase activity. Bin bound to transfected cells and induced a non-specific current presumably related to the opening of pores. The formation of these pores may be related to the location of the toxin/receptor complex in lipid raft microdomains. Finally, Bin promoted the time-dependent appearance of intracytoplasmic vacuoles but did not drive cell lysis. Thus, the dual functionality (enzyme/toxin receptor) of Cpm1 is fully conserved in MDCK cells and Cpm1 is an essential target protein for Bin cytotoxicity in Culex mosquitoes.

Incidence of chlamydial infection depends on maternal colonization during pregnancy, which is different in each population. The transmission is not obligatory but when present, it occurs at birth through the genital tractus. Chlamydia trachomatis infection is the first cause of neonatal conjunctivitis, with no influence of eye lotion application at birth. C. trachomatis is also responsible for interstitial pneumonia with possible consequences on the lung function. The laboratory diagnosis relies on the identification of intracellular bacteria in patient samples by the mean of culture or PCR. Systemic antibiotherapy by macrolides is always necessary, with local application in the case of conjunctivitis. The key point is the detection of colonization of pregnant women with identified risk factors. In positive case, oral treatment of both parents is recommended.

The mosquitocidal activity of Bacillus sphaericus is because of a binary toxin (Bin), which binds to Culex pipiens maltase 1 (Cpm1), an alpha-glucosidase present in the midgut of Culex pipiens larvae. In this work, we studied the molecular basis of the resistance to Bin developed by a strain (GEO) of C. pipiens. Immunohistochemical and in situ hybridization experiments showed that Cpm1 was undetectable in the midgut of GEO larvae, although the gene was correctly transcribed. The sequence of the cpm1(GEO) cDNA differs from the sequence we previously reported for a susceptible strain (cpm1(IP)) by seven mutations: six missense mutations and a mutation leading to the premature termination of translation. When produced in insect cells, Cpm1(IP) was attached to the membrane by a glycosylphosphatidylinositol (GPI). In contrast, the premature termination of translation of Cpm1(GEO) resulted in the targeting of the protein to the extracellular compartment because of truncation of the GPI-anchoring site. The interaction between Bin and Cpm1(GEO) and the enzyme activity of the receptor were not affected. Thus, Bin is not toxic to GEO larvae because it cannot interact with the midgut cell membrane, even though its receptor site is unaffected. This mechanism contrasts with other known resistance mechanisms in which point mutations decrease the affinity of binding between the receptor and the toxin.

DSC1 encodes a putative voltage-sensitive sodium channel alpha subunit in Drosophila melanogaster. We generated polyclonal antibodies raised against part of the DSC1 sequence to characterize the size and the distribution of these channels in the adult fly. Immunoblotting experiments indicated that the protein has a molecular weight of about 270 kDa. We also showed that DSC1 channels are found only in the neurons of the fly. The density of channels was high in synaptic regions and in most of the axonal processes that connect the various structures of the CNS. No signal was observed in the cortical cell bodies where the para channels are mainly present. The most striking result concerns the widespread distribution of DSC1 channels in the PNS, as confirmed by experiments done with the monoclonal antibody 22C10. These results strongly suggest that DSC1 and para channels may have complementary roles, at least in the adult stage.

Knockdown resistance (kdr) to pyrethroid insecticides has been found in numerous insect species. kdr causes nerve insensitivity by altering the primary target of these insecticides, the voltage-sensitive sodium channel. In Musca domestica, cloning and sequencing of susceptible, kdr, and super-kdr alleles of the sodium channel gene (Msc) homologous to the Drosophila melanogaster para channel gene has revealed point mutations. The conservation of the nature and of the position of these mutations strongly suggests a role in the kdr mechanism. To determine if these mutations are associated with modifications of channel expression in adult flies, we investigated the localization of the Msc transcripts, and the size and the tissue distribution of the channel protein in pyrethroid-susceptible and super-kdr strains. Msc channels were mainly found in the cortical regions of the central nervous system with additional labeling in some neuronal processes and in the eyes. No qualitative or quantitative difference was observed between the strains. In immunoblotting experiments, anti-Msc antibodies bound to only one polypeptide of 260 kDa in adult brain. No differences were found in antibody staining between susceptible and pyrethroid-resistant strains. These results were correlated with those on Drosophila melanogaster, for which two sodium channel genes have been identified.

In Drosophila, the para gene has been shown to encode a functional voltage-dependent sodium channel. We used a cDNA clone to study the distribution of its transcripts by in situ mRNA hybridization on adult fly sections. These transcripts are found in cortical regions of the central nervous system and in the eyes. On immunoblots, antibodies raised against expression products of part of the gene recognize a polypeptide of M(r) approximately 270,000 in head membranes. Immunolocalization experiments indicate that anti-para antibodies bind to cortical regions of the brain and give heavy signals in the eyes. Immunohistochemistry was also performed on napts and seits1, two mutant Drosophila strains known to be defective in sodium channel activity. Only napts flies displayed a decrease in the expression of the para protein.
